Nowadays, self-assembled polymeric micelles from amphiphilic polymer conjugates are urgently needed to enhance drug targeting and release efficiency. On account of the excellent properties of alginate and the redox-responsiveness of disulfide bonds , a redox-sensitive alginate–SS–ibuprofen derivative (LSA–SS–IBU) based anti-tumor target drug delivery system was created by attaching the hydrophobic ibuprofen (IBU) onto the hydrophilic alginate molecular skeleton through disulfide linkages for glutathione (GSH) triggered delivery of doxorubicin hydrochloride (DOX). The structure and self-assembly behavior of the synthesized LSA–SS–IBU was systematically evaluated by FT-IR spectrometer, 1 H NMR spectrometer, fluorescence spectrophotometer (FM), transmission electron microscope (TEM) and dynamic light scattering (DLS). Furthermore, the in vitro release of drugs from the LSA–SS–IBU micelles loaded with DOX was examined, along with the assessment of their cytotoxicity to HeLa cells. The resultant LSA–SS–IBU had a critical micelle concentration (CMC) value of 0.19 mg/mL that was able to self-assemble into the spherical micelles with the particle size and zeta potential of 148.2 ± 4.3 nm (PDI = 0.25) and −47.2 ± 0.4 mV, respectively. At a mass ratio of 3:10 for DOX/LSA–SS–IBU, the LSA–SS–IBU micelles achieved drug loading ratio (DLR) and encapsulation efficiency (EE) values of 12.2 % and 46.3 %, respectively. Furthermore, the in vitro drug release study indicated that the total drug release percentage of the drug-encapsulated LSA–SS–IBU micelles in PBS with the presence of 10 mmol/L GSH for 12 h exceeded 67.0 %, demonstrating a substantial increase compared to that in GSH-free PBS. The in vitro cytotoxicity findings indicated that LSA–SS–IBU had good biocompatibility , while the IC 50 value and the cell viability of DOX-loaded LSA–SS–IBU micelles cultured with HeLa cells for 48 h was 2.2 μg/mL and less than 20.1 %, respectively, which could significantly inhibit the growth of HeLa cells. Therefore, redox-responsive LSA–SS–IBU micelles had potential applications in cancer therapy for hydrophobic anti-tumor drug delivery.

The research goal of the present study was to develop a carrier for loading and controlled -release of the hydrophobic drug with the combined use of organo-montmorillonite (OMMT) and alginate. The OMMT was synthesized through the intercalation modification of sodium montmorillonite (Na-MMT) with cationic cetyltrimethylammonium bromide (CTAB), nonionic nonylphenol polyoxyethylene ether (NPE) and the mixture of them via simple and convenient wet ball-milling method. Furthermore, the organo-montmorillonite/alginate (OMMT/Alg) composite hydrogel beads with slow and controlled release properties were constructed by using alginate as a coating material under the exogenous cross-linking of calcium ions. The physical and chemical properties of OMMT were comparatively evaluated by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), transmission electron microscopy (TEM), dynamic light scattering (DLS), thermogravimetric analyzer (TGA), BET-specific surface area measurements, and drug adsorption experiments. Experimental results showed that the presence of CTAB was able to facilitate the intercalation of CTAB/NPE into Na-MMT through the cation exchange reaction. And the cationic CTAB and nonionic NPE were adsorbed or intercalated into the MMT lamellar structure through the wet ball-milling process, which could change the hydrophilic nature of Na-MMT and improve its affinity to the hydrophobic drug molecules. In addition, the OMMT/Alg composite hydrogel beads displayed superior sustained-release properties than Na-MMT/Alg, mainly ascribed to the good affinity of OMMT to hydrophobic drug that retarded the drug diffusion. In particular, CTA/NPE-MMT/Alg with the highest loading capacity (LC) and encapsulation efficiency (EE) revealed the optimal controlled performance for the release of hydrophobic ibuprofen. The release followed the Korsmeyer–Peppas model suggested non-Fickian diffusion release mechanism. Based on the high drug loading capacity and excellent controlled drug release properties, the CTA/NPE-MMT/Alg incorporating hydrophobic drugs into hydrophilic matrices could be a highly promising material for use in hydrophobic drug delivery.

BACKGROUND Stimuli-responsive pesticide controlled release system provides a new strategy for the development of high-efficiency pesticides formulation. RESULTS In this article, we report a novel polydopamine surface modified MXene-Ti 3 C 2 T x nanocarrier for pesticide delivery and plant protection. Polydopamine modified Ti 3 C 2 T x (PDA@Ti 3 C 2 T x ) nanocarrier was prepared by biomimetic self-polymerization of dopamine on the surface of Ti 3 C 2 T x . A typical pesticide, emamectin benzoate (EB), was loaded on PDA@Ti 3 C 2 T x through physisorption process, with a high pesticide loading rate of 45.37%. PDA@Ti 3 C 2 T x exhibited excellent photothermal conversion effect ( η = 34.5%). Under the irradiation of near-infrared (NIR) laser, EB would sustained release from PDA@Ti 3 C 2 T x nanocarrier to surrounding medium. Compared with free EB, EB@PDA@Ti 3 C 2 T x exhibited prolonged persistence period, which can keep antipest activity at 14 days post spraying. In addition, PDA@Ti 3 C 2 T x nanocarrier and EB@PDA@Ti 3 C 2 T x nanoformulation are of good safety, showing no side effect to the seed germination and seedling growth. CONCLUSION This research developed a novel nanocarrier for water-insoluble pesticide delivery, realizing NIR-responsive controlled release and sustained antipest activity.

Bio-based coated controlled-release fertilizers (BCRFs) can significantly improve nutrient utilization and reduce pollution. However, challenges such as spray gun clogging, adhesion and tearing between fertilizers during the actual production of BCRFs pose a severe challenge to production chain development. In addition, the poor hydrophobicity and cross-linking properties of bio-based coatings severely affect the controlled release properties of BCRFs. Although some measures have been taken to ameliorate the above problems, the cumbersome and costly modification process is challenging to be applied on a large scale. Therefore, the primary goal of this work is to search for a more straightforward and efficient method of production and modification to address the aforementioned unresolved challenges in the field of CRFs. In this study, bio-based polyester polyol (BPP) was used as the base for polyurethane reaction, and low-viscosity castor oil (CO) was used as the modifier to reduce the viscosity of BPP. This led to the preparation of Inter-Modified Coated Controlled-Release Fertilizers (ICRFs) to prevent spray gun clogging. Building upon this, hydrophobic and dense wax-based coated fertilizer (WBCF) was prepared by modifying ICRF with Ceresin wax to alleviate the challenges associated with adhesion and tearing between fertilizer particles during the coating process. The WBCF exhibited remarkable controlled-release characteristics, with a nitrogen (N) release longevity of nearly 180 days, requiring only 3 % of the coating materials. This work provides a simple and efficient strategy for overcoming the key constraints in the development and production of CRFs and provides insights into developing green, low-cost fertilizers with excellent controlled-release properties.

Polymer-based drug-controlled release systems offer greater efficacy and potency than conventional therapies. However, prominent drug side effects, lower circulation, and low drug loading capabilities limit their application range. In this work, the combination of Simvastatin (SIV) and Carvacrol (CAV) into PEG-PLGA microspheres (SIV-CAV-PP-MS) was achieved via an emulsification-solvent evaporation technique, resulting in microspheres characterized by high encapsulation efficiency and reduced particle size. In vitro studies demonstrated that the cumulative drug release increased with higher SIV and CAV levels in the release medium, reaching 88.91% and 89.35% at 25 days. Pharmacokinetic analysis revealed that the concentrations of SIV and CAV reached their maximum levels at approximately seven days in the SIV-CAV-PP-MS group, which indicates that using PEG-PLGA as a carrier significantly delays drug release. In vivo, evaluation demonstrated that the SIV-CAV-PP-MS high-dose group and positive drug control group showed reductions in low-density lipoprotein cholesterol levels by 0.39-fold and 0.36-fold compared to the Hyperlipidemia model group, and the addition of CAV significantly enhanced the lipid-lowering effects of SIV. Histological examinations indicated that the SIV-CAV-PP-MS medium-dose group displayed histological features more closely resembling those of normal mice compared to the Simvastatin control group, with a well-organized hepatocyte structure, a significant reduction in lipids, and improved liver health. The prepared polymeric microsphere utilizing SIV and SAV will be a promising dosage form for hyperlipidemia disease patients, with superior lipid-lowering efficacy and improved patient compliance.

This study developed a stable nanoparticle (CUR-SPIH/SSPS) using soybean protein isolate hydrolysate (SPIH) and soybean polysaccharides (SPSS) to protect curcumin (CUR) from degradation during storage and exposure to light and heat conditions, achieving controlled release. The SPIH to SPSS mass ratio of 5:1 gave the CUR-SPIH/SPSS nanoparticles with the highest CUR encapsulation efficiency (95.60 ± 3.00 %) and the strongest antioxidant capacity (90.26 ± 2.42 % and 66.78 ± 1.89 % for ABTS •+ and DPPH radical scavenging ability, respectively), and CUR was successfully encapsulated within the CUR-SPIH/SPSS as evidenced by X-ray diffraction. FTIR and fluorescence spectroscopy analysis confirmed that the interactions in CUR-SPIH/SPSS are primarily driven by electrostatic, hydrogen bonding, and hydrophobic interactions. Moreover, the CUR-SPIH/SPSS nanoparticles significantly enhanced CUR's thermal and UV light stability. The UV degradation kinetics showed that the half-life of CUR-SPIH/SPSS (247.55 min) was 1.61 times longer than that of free CUR (154.03 min). The release rate of CUR incorporated into CUR-SPIH/SPSS was significantly delayed during in vitro gastrointestinal digestion. This study introduces an innovative nanoparticle strategy for the stable delivery of lipophilic compounds.

Spinal cord injury (SCI) is one of the most devastating acute diseases. Zn 2+ has been verified plays a crucial role in SCI repair, however, how to continuously release zinc ions at the lesion site is still a cutting-edge challenge. Therefore, it is urgent to design a novel nanoplatform for promoting SCI repair with multifunction of controlled release of Zn 2+ and regulating ROS microenvironment of mitochondrion. Herein, we propose a protocol of preparing NIR sensitive ZnO quantum dots (QDs) decorated MXene hydrogel for SCI repair. TEM and EDS mapping showed that the ZnO QDs were uniformly distributed on the Mxene nanosheets. Catalase (CAT) analysis showed that the ZnO-Ti 3 C 2 nano enzymes could effectively scavenge ROS. Molecular biology studies showed that ZnO-Ti 3 C 2 @H repaired mitochondrial function by scavenging ROS with a clearance rate of up to 80 %. Neuronal apoptosis was subsequently reduced, and nerves and blood vessels were regenerated. NIR irradiation warmed the injury site to 42.9 °C, which enabled rapid enrichment of Zn 2+ at the lesion site. Footprinting and pathology experiments have shown that it improves functional recovery after injury and results in near-normal BMS scores. These results suggest that this multifunctional type of material incorporating NIR controlled release of Zn 2+ provides a promising strategy for promoting SCI repair.

The objective of this study was to investigate the controlled release effect of various polar functional groups in acrylate pressure-sensitive adhesives (PSA) on permeation enhancers (PE), as well as to elucidate the disparities in their enhancing effects under such circumstances. The synthesis of acrylate PSA with hydroxyl group (AAOH), amide group (AACONH 2 ), hydroxyphenyl group (AAPhOH), and carboxyl group (AACOOH) was achieved via a solution polymerization process initiated by free radical. Gel permeation chromatography, molecular dynamic simulation and thermal analysis were conducted to characterize the properties of PSA. In vitro release study, Raman imaging and correlation analysis were performed to research the controlled release effect on the permeation enhancer azone (AZ). These were evaluated using a novel parameter called the control release factor ( F cr ). The more negative the F cr value, the stronger the control release effect . In vitro skin permeation study and pharmacokinetic study reflected the enhancing effects of AZ on drug ketoprofen (KET). Fourier-transform infrared spectroscopy, 1 H nuclear magnetic resonance ( 1 H NMR), molecular docking, rheology study, Raman spectra and solid-state nuclear magnetic resonance (ssNMR) characterized mechanisms of functional groups’ controlled release effect and the changes in skin barrier function. The results demonstrated that among the four PSA, AACOOH exhibited the most pronounced controlled release effect ( F cr = -45.91). This can be attributed to the formation of a robust ionic bond between AACOOH and AZ, which effectively restricted the release of AZ into the skin, resulting in minimal impact on disordering functionality of the skin barrier and enhancing drug permeation. The order of controlled release for the other three PSA on AZ was as follows: AAPhOH ( F cr = -43.60) > AACONH 2 ( F cr = -40.26) > AAOH ( F cr = -38.15), resulting the best enhancing effect attributed to AZ that released from AAOH into the skin. Besides, the interaction between the above three PSA and AZ was hydrogen bond, only the interaction of AAPhOH-AZ was a special hydrogen bond: ionic hydrogen bond. In summary, the type and strength of the interaction between PSA and PE determined the amount of PE entering the skin, thereby determining the extent to which PE disorder skin barrier function.

Polysaccharide-based active packaging materials have extensively explored in the field of food safety, whereas the highly hydrophilic traditional biopolymer was susceptible to be broken under moisture conditions, leading to the invalid food preservation. Herein, a multi-mechanism cross-linking densification strategy was employed to design and construct novel carboxymethyl chitosan (CMCS)/sodium alginate-dopamine complex (SD)/oxidized starch (OS)/tea polyphenol (TP) composite film (COSDT) composite. Multiple cross-linking reduces the spacing between polysaccharide chains to make the structure denser, hindering the adsorption and penetration of water molecules. The optimized COSDT-2 film maintains intact structures without any dissolution after immersing in water for 24 h and demonstrated relatively stable water contact angle of 71.97° at 1 s and 68.48° after 60 s. The water vapor permeability and oxygen transmission rate were reduced from 5.10 × 10 −7 g m −1 s −1 Pa −1 and 8.28 cm 3 μm (m 2 day KPa) −1 for COT sample to 1.03 × 10 −7 g·m −1 ·s −1 ·Pa −1 and 2.70 cm 3 μm (m 2 day KPa) −1 for COSDT-2 film. Moreover, COSDT films exhibited stable wet mechanical properties in high-humidity environments (95 % RH) of 30.76 MPa. COSDT-2 accelerated the release of tea polyphenols by structural loosening due to enhanced interchain repulsion under alkaline conditions, resulting in a significant reduction of volatile alkaline nitrogen production by 61.58 % in chicken breast meat and prolonged shelf life. This study provided a novel perspective in designing and constructing water-resistant active food packaging biofilms through modulating the interchain spacing of polymers.

In this study, dual functionalized mesoporous silica nanoparticle (Dual-MSN) with functions of carboxyl modification and chirality was successfully developed and its special contribution in delivering doxorubicin hydrochloride (DOX) in vitro was mainly studied. Characteristics of Dual-MSN and its application as DOX carrier were intensively explored by comparing with naked non-functionalized MSN (Naked MSN). The results indicated that both Naked MSN and Dual-MSN significantly controlled DOX release due to the release hindrance caused by mesopores. As expected, Dual-MSN exhibited obvious enhanced pH-response because of its negative charges of carboxyl groups. DOX loaded Naked MSN and DOX loaded Dual-MSN presented better cytotoxicity than DOX due to carrier-mediated endocytosis and the favorable intercalation of DOX into DNA in the nuclei. The cytotoxicity of DOX loaded Dual-MSN was better than DOX loaded Naked MSN owing to its enhanced cellular uptake induced by chirality of Dual-MSN, demonstrating that double functions of Dual-MSN had unique advantages in improving antitumor effect of DOX towards MCF-7 cells and thus confirming its special contribution in DOX delivery.

Biodegradable hemostatic gauze used for surgical hemostasis has attracted great interest due to its excellent compliance and local anti-inflammatory and therapeutic effects when combined with drugs. Herein, we demonstrate the successful fabrication of water-soluble absorbed cellulose hemostatic material by introducing a biocompatible hydroxyethyl cellulose (HEC) hemostasis gauze into doxorubicin-loaded in situ gel (GEL(DOX)) for the prevention of breast cancer recurrence after surgical tumor resection. The present results show that HEC has a shorter metabolic period, no anaphylaxis and peripheral nerve toxicity, and possesses more advantages than oxidative regenerated cellulose hemostasis gauze, a commercially available product in market. HEC is of the physical hemostasis in mechanism, which does not induce physiological hemostasis and hemolysis. In addition, the combination of HEC with GEL(DOX) not only stops the bleeding efficiently, but also effectively reduces the proliferation of tumor with no cardiac toxic and bone marrow suppression. After treatment, the tumor inhibition rate is up to 90%, resulting in prolonged survival time to 58 days. In conclusion, HEC hemostatic gauze has a broad prospect in clinical application due to its perfect biocompatibility, and we envision that it is a new strategy for the prevention of breast cancer to implant HEC hemostatic gauze containing GEL(DOX) at the postoperative site after surgery.
