Immobilization of fungal cells on positively charged cyclodextrin-based carriers: Enhanced PhAC degradation and multi-omics implications

Yizhou Tu,Yizhou Wu,Yang Pan,Xiaolu Ding,Xinyi Wang,Xingqi Zhu,Jun Li,Xianchuan Xie,Yong Chen,Wentao Li,Chendong Shuang,Qing Zhou,Aimin Li

The rational design of cellular interfaces is essential for advancing biocatalytic systems. White rot fungi are promising biodegraders of pharmaceutically active compounds (PhACs), but they are readily inactivated in wastewater treatment, while conventional carriers cannot simultaneously immobilize cells and retain extracellular enzymes. In this study, a stable fungus-enzyme synergistic interface was constructed by assembling Trametes versicolor with a positively charged porous thienyl cyclodextrin polymer (Th-CDP) to form Tv@Th-CDP. This design simultaneously immobilized fungal cells and retained their secreted laccases within the carrier, enhancing PhAC removal in continuous-flow reactors treating secondary municipal effluent to 93%. The system demonstrated sustained 30-d performance and reduced effluent toxicity by 58%. This performance was associated with the high positive charge density of Th-CDP, which enhanced mycelial attachment and laccase retention (48.2 U kg –1 ). Furthermore, the hydrophobic cavities of cyclodextrin, coupled with electrostatic interactions, enhanced PhAC adsorption and enriched Trametes sp. (46%). This immobilization strategy improved the fungus-enzyme interactions, with enhanced laccase-related catalytic currents observed in electrochemical assays. Metaproteomics analysis and transformation pathway identification supported a degradation model mediated by electrostatic interactions between the fungus and the retained enzymes. A total of 305 proteins associated with oxidoreductase activity were enriched, while the key enzymes aldehyde dehydrogenase (ALDH) and epoxide hydrolase (EPHX2) were upregulated. These changes coincided with enhanced C–N bond cleavage, indicating activation of associated metabolic pathways. Overall, this study provides mechanistic understanding of immobilization mediated by electrostatic interactions and demonstrates its potential to improve the biodegradation of emerging contaminants in engineered biocatalytic systems.

Integration of Medium Engineering, Immobilization, and Rotating Bed Reactor for Intensifying Enzymatic Reduction of Halogenated Aryl Ketones

Shuling Zhang,Jiajing Yan,Quan Yuan,Yanjun Jiang,Lihui Wang,Yunting Liu

Enzymatic reduction of halogenated aryl ketones (HAKs) is an important reaction for the synthesis of pharmaceutical-related enantiopure halohydrins, but its preparative application remains restricted due to low substrate solubility and poor mass transfer in aqueous media. Herein, a multiengineering approach involving medium optimization, enzyme immobilization, and a rotating bed reactor was applied for intensifying the enzymatic reduction of HAKs. A deep eutectic solvent (DES) composed of betaine, glucose, and water (Bet-Glc-H2O) was demonstrated to be effective in improving enzymatic activity and stability, enabling efficient reduction of high-concentration HAKs (1.0 M, 85% conversion). In addition, enzyme immobilization was conducted using DES-modified resin supports, markedly enhancing the operational stability and reusability of the biocatalysts. Finally, a SpinChem rotating bed reactor (RBR) was implemented for further intensifying the enzymatic process, leading to dramatically shorter reaction times and increased overall productivity.

Linker Peptide Engineering Combined With SA-PVA Immobilization in Fed-Batch Biocatalysis for High-Efficiency Carcinine Synthesis

Man Zhao,Mengying Yu,Huiru Yuan,Yiting Shen,Zhiqiang Liu,Yuguo Zheng

Carcinine, a valuable imidazole dipeptide with antioxidant and therapeutic properties, faces biosynthesis challenges due to enzyme aggregation and substrate inhibition. In this study, an integrated strategy combining linker peptide engineering and immobilization was applied to address these challenges and enhance carcinine production. Rational design of linker peptides (D 5 , L 2 , L 3 ) in the sfp-Ebony fusion protein enabled its highest soluble expression in WSL 2 E strain, achieving 93.1% conversion efficiency—3.5-fold higher catalytic efficiency than WSGE strain. Response surface methodology optimized sodium alginate-polyvinyl alcohol (SA-PVA) immobilization parameters (5% PVA, 3% SA, 2.3% CaCl₂), yielding excellent immobilized WSL 2 E@SA-PVA cells with 95.93% activity recovery. Structural characterization by scanning electron microscopy (SEM), Fourier-transform infrared spectroscopy (FT-IR), and X-ray diffraction (XRD) confirmed the formation of a porous SA-PVA matrix that protected cells from harsh conditions. The immobilized biocatalyst exhibited superior operational stability (retaining > 80% activity after 7 cycles) and storage stability (maintaining 44.89% activity after 14 days at 4°C). Fed-batch scale-up (50 mL) achieved a record carcinine titer of 71.13 mM, mitigating the inhibitory effect of high substrate concentrations through phased substrate feeding. This study provides a scalable biocatalytic platform for industrial carcinine production, effectively addressing key bottlenecks in biocatalyst stability and substrate tolerance.

Preparation of Streptavidin-Coated Magnetic Nanoparticles for Specific Immobilization of Enzymes with High Activity and Enhanced Stability

Jie Long,Xingfei Li,Xiaoxiao Liu,Zhengyu Jin,Zhengjun Xie,Xueming Xu,Cheng Lu

In this study, streptavidin-functionalized magnetic nanoparticles were developed as a support for enzyme immobilization based on the specific recognition between biotin and streptavidin. The enzyme-immobilizing ability was evaluated using pullulanase as a model enzyme. Magnetic poly(2-hydroxyethyl methacrylate-co-glycidyl methacrylate) nanoparticles were prepared and functionalized with streptavidin using cyanuric chloride. The immobilized enzymes were characterized using UV–vis spectra, Fourier-transform infrared spectroscopy, and transmission electron microscopy. The immobilized pullulanase retained high levels of activity (85.3%) and exhibited significantly improved pH and thermal stability as compared to the free enzyme. At pH 5.5, the relative activity of the immobilized enzyme (75.2%) was significantly greater than that of the free enzyme (15.8%; p < 0.01). After incubation for 360 min at 60 °C, the residual activity of the free enzyme was only 21.5%, while the immobilized enzyme retained more than 70.6% of its residual activity. Moreover, the immobilized pullulanase also exhibited excellent recyclability, retaining more than 74.2% of its initial activity after 8 consecutive reuses. These results indicated that streptavidin-coated magnetic nanoparticles have great potential for use as a support for the immobilization of the various enzymes required for continuous biotechnological applications.

Synergistic effects of lattice incorporation and spatial confinement for radionuclides immobilization in sodalite derived from 4A zeolite

Junyi Feng,Shiyu Wang,Jiwei Zhang,Mengyi Tang,Xiaodan Zhang,Xuesong Feng,Kaifa Du,Wei Li,Huayi Yin,Bowen Deng,Dihua Wang

The efficient immobilization of radionuclides such as Cs, Sr, Ba, and Ce remains a critical challenge in the safe treatment of radioactive waste salts. Here we report a synergistic lattice incorporation and spatial confinement strategy that enables one-step immobilization of multiple radionuclides within a sodalite framework derived from topotactic transformation of 4 A zeolite. Mechanical blending followed by solid-state sintering at 850℃ for 2 h produces a dense sodalite solidified matrix exhibiting exceptional thermal stability, radiation resistance, acid-alkali tolerance and mechanical integrity. The normalized leaching rates of Cs, Sr, Ba, and Ce were all below 5 × 10 −5 g/(cm 2 ·d) after 28 days. Multiscale characterization combined with density functional theory (DFT) reveals that chemical adsorption, ion exchange, lattice incorporation, and covalent coordination collectively stabilize radionuclides through coupled spatial confinement and lattice immobilization. A prospective life cycle assessment (LCA) confirms that this sodalite operates with markedly reduced carbon emissions and energy demand compared to conventional vitrification and ceramic process. These findings provide a promising low-energy, high-safety pathway for the immobilization and disposal of irradiated nuclear fuels.

Immobilization of trypsin onto multifunctional meso-/macroporous core-shell microspheres: A new platform for rapid enzymatic digestion

Gong Cheng,Ping Chen,Zhi-Gang Wang,Xiao-Jing Sui,Ji-Lin Zhang,Jia-Zuan Ni

A simple, fast, efficient, and reusable microwave-assisted tryptic digestion system which was constructed by immobilization of trypsin onto porous core-shell Fe 3 O 4 @fTiO 2 microspheres has been developed. The nanostructure with magnetic core and titania shell has multiple pore sizes (2.4 and 15.0   nm), high pore volume (0.25   cm 3   g −1 ), and large surface area (50.45   m 2   g −1 ). For the proteins, the system can realize fast and efficient microwave-assisted tryptic digestion. Various standard proteins (e.g., cytochrome c (cyt-c), myoglobin (MYO), β-lactoglobulin (β-LG), and bovine serum albumin (BSA)) used can be digested in 45   s under microwave radiation, and they can be confidently identified by mass spectrometry (MS) analysis; even the concentration of substrate is as low as 5   ng μL −1 . Furthermore, the system for the 45   s microwave-assisted tryptic digestion is still effective after the trypsin-immobilized microspheres have been reused for 5 times. Importantly, 1715 unique proteins from 10   μg mouse brain proteins can be identified with high confidence after treatment of 45   s microwave-assisted tryptic digestion.

In situ immobilization of layered double hydroxides onto cotton fiber for solid phase extraction of fluoroquinolone drugs

Xuemei Wang,Wei Zhou,Chenlu Wang,Zilin Chen

Cotton fiber was proposed as a biodegradable support material. The properties like high specific area, hygroscopicity and adjustable shape make it be an idea substrate for solid phase extraction (SPE). In this work, inorganic layered double hydroxides (LDHs) were in situ immobilized onto cotton fiber through the linker of polydopamine as stationary phase. Uniform NiAl-LDHs crystal with nanostructure was observed on cotton fiber after one-step synthesis through urea hydrolytic method. The cotton fiber can not only increase the specific area and mass transfer rate, but also prevent aggregation and blocking of the freestanding LDHs crystal. Due to the electrostatic interaction and anion exchange features of LDHs, the prepared LDHs modified cotton fiber showed good extraction efficiency and selectivity towards fluoroquinolone drugs including norfloxacin , ciprofloxacin and enrofloxacin . After elution by 1 M NaOH, a simple acid-base neutralization method was proposed, so the natural elution can be directly analyzed by HPLC system. After optimizing the extraction parameters including adsorbent amount, sampling rate, sample volume, sample pH and eluent types and concentration, excellent extraction efficiency towards three fluoroquinolone drugs was obtained with high enrichment factors of 37–47. The adsorption capacity of LDHs modified cotton fiber was 21.18, 21.85 and 20.54 mg g −1 for norfloxacin, ciprofloxacin and enrofloxacin, which was almost twice as much as that of freestanding LDHs. The established method based on SPE-HPLC-FLC (fluorescence detector) technique exhibited good linearity (R 2 ≥ 0.995), low limits of detection (0.01 µg L −1 ) and good reproducibility (intra-day RSDs ≤ 3.89%). Moreover, this method has also been applied in analyzing fluoroquinolone drugs in chicken eggs and pork liver.

Spatial localization immobilization of metals and enzymes at Pickering emulsion droplet interfaces for chemo-enzymatic cascade catalysis

Geling Kuang,Jinchao Hou,Zichen Wang,Lixue Zhao,Yuxiao Feng,Hongwu Wang,Jiandong Cui

Developing chemo-enzymatic cascade catalysis in catalytic asymmetric synthesis is of utmost significance, but such processes often suffer from low reaction efficiency because of incompatible reaction environments and suppressed intermediate transportation. Herein, a novel compartmentalised metal–enzyme microcapsule nanohybrid, Candida antarctica lipase B@Pd(II)-loaded hydrophobic mesoporous silica nanoparticles (CALB@Pd(II)/HB-MSNs), was fabricated by spatially localizing Pd(II) active sites and CALB at the outer and inner interfacial layers of Pickering emulsion droplets, respectively. Benefiting from the localization of two catalysts in their preferred reaction microenvironments, nanoscale proximity of each other, and interfacial activation of lipase, the CALB@Pd(II)/HB-MSNs exhibited 60-fold higher activity than traditional random co-immobilized metal-enzyme catalysts (Pd(II)/HB-MSNs/CALB). Furthermore, the conversion of rac-1-phenylethanol catalyzed by CALB@Pd(II)/HB-MSNs reached 47.92 %, compared to only 4 % for the randomly co-immobilized catalyst Pd(II)/HB-MSNs/CALB, with a product enantiomeric excess of≥99 % in the cascade reaction of 4-pentynoic acid cycloisomerisation and racemic 1-phenylethanol chiral resolution. Our study offers a platform for the design of efficient chemoenzymatic catalytic cascades for the synthesis of fine chemicals.

Green synthesis and immobilization of AgNPs by lumpy corn stalk as interlayer filling material for durable antibacterial

Qimeng Jiang,Zhaoyun Lin,Bin Gu,Chunsheng Pang,Xiaoying Wang

Lumpy corn stalk is conventionally used as interlayer filling material in plywood, while high temperature and humidity in tropical and subtropical areas are easy to cause bacteria growth on plywood. Silver nanoparticles (AgNPs) are one of efficient antibacterial metal nanoparticles due to their excellent broad-spectrum antibacterial properties. But most of these AgNPs are normally in solution and easy to aggregate, which limits their utilization. Besides, the synthesis of AgNPs always needs chemical reducing agents, which can cause environmental and cost problems. In this study, lumpy corn stalk with honeycomb-like structure acted as green reducing agent and stabilizer to in situ synthesize AgNPs, the obtained antibacterial lumpy corn stalk (CS@AgNPs) could be used as interlayer filling material to protect plywood from bacteria erosion. Durable antibacterial study showed that the bacteriostatic properties of lumpy CS@AgNPs against E. coli and S. aureus were stronger and more durable than commercial Ag + spray and hydrogel. The antibacterial mechanism may be that the controlled-released AgNPs destroyed the morphological structure of bacteria by penetrating into the bacterial cell membrane, following by leaking cellular content. Notably, the final release amount of AgNPs from lumpy CS@AgNPs after 12 days in buffer solution was only 0.011 mg/L, which was 9 times lower than the domestic drinking water limit presented by United States Environmental Protection Agency (USEPA). The plywood with lump CS@AgNPs as interlayer filling material could keep efficient and durable bacteria killing activity in 21 days, which could be used safely in food packaging, furniture and flooring fields.

The immobilization on various concentrations of iodine in silver-coated silica gel via B2O3–Bi2O3 based material

Xiao Wang,Bingsheng Li,Zhi Chen,Xiaoyan Shu,Zhentao Zhang,Jingjun Yang,Guilin Wei,Yi Liu,Shunzhang Chen,Yi Xie,Xirui Lu

The treatment of iodine wastes has been widely concerned. In this work, B 2 O 3 –Bi 2 O 3 based material was used to immobilize the silver coated silica gel with different iodine loadings. Based on XRD results analyses, the sample with 90 mol% B 2 O 3 was selected for iodine waste immobilization. XRD results represented that the sintering temperature of 500 °C was suitable to immobile iodine waste. The micro-structure of the sintered matrix mainly consists of the stretching vibration of [SiO 4 ]. SEM-EDS results showed that the elements of Ag and I exist element aggregation in some regions, which indicates that part of AgI was immobilized in the glassy phase while the other was physically enclosed in the sintered matrix. This study provides certain valuable reference and experimental basis for the subsequent curing of iodine-containing silver coated silica gel with multicomponent glass.

Study on the Immobilization of Horseradish Peroxidase on a Multi-Level Composite Carrier SiO2@MnO2@MAF-7

Mengjie Huang,Baihui Zhang,Xiangyu Jiang,Maojie Jiang,Peng Yin,Xuan Fang,Yanna Lin,Fuqiang Ma

This study addresses the issues of poor stability and difficulty in recovery of free horseradish peroxidase (HRP) by developing a multi-level composite immobilized carrier that combines high loading capacity with long-term stability. The SiO2@MnO2@MAF-7 core–shell structured carrier was prepared via a solvothermal self-assembly method. Three immobilization strategies—adsorption, covalent cross-linking, and encapsulation—were systematically compared for their immobilization efficacy on HRP. The material structure was analyzed using techniques such as specific surface area analysis (BET), X-ray diffraction (XRD), and Fourier transform infrared spectroscopy (FTIR) to characterize the material structure. Enzyme kinetic parameter determination experiments were conducted to systematically evaluate the performance advantages of the immobilized enzyme. BET analysis showed that SiO2@MnO2@MAF-7 had a specific surface area of 251.99 m2/g and a mesoporous area of 12.47 nm, and its HRP loading was 50.37 U/mg (immobilization efficiency 85.03%). Compared with free HRP, the Km value of the immobilized enzyme was decreased by 42%, the activity retention rate was increased by 35–50% at 80 °C and pH 4–9, and the activity was maintained by 65% after five repeated uses. In this study, MAF-7 was combined with MnO2/SiO2for HRP immobilization for the first time, and the triple effect of rigid support-catalytic synergy-confined protection synergistically improved the stability of the enzyme, providing a new strategy for the industrial application of oxidoreductases.

Fabrication of ultrafine Gd2O3 nanoparticles/carbon aerogel composite as immobilization host for cathode for lithium-sulfur batteries

Mangmang Shi,Yinglin Yan,Yiqi Wei,Yiming Zou,Qijiu Deng,Juan Wang,Rong Yang,Yunhua Xu,Tong Han

Summary Carbon aerogel (CA), possessing abundant pore structures and excellent electrical conductivity, have been utilized as conductive sulfur hosts for lithium-sulfur (Li-S) batteries. However, a serious shuttle effect resulted from polysulfide ions has not been effectively suppressed yet due to the weak absorption nature of CA, resulting in rapid decay of capacity as the cycle number increases. Herein, ultrafine (~3 nm) gadolinium oxide (Gd 2 O 3 ) nanoparticles (with upper redox potential of ~ 1.58 V versus Li + /Li) are uniformly in-situ integrated with CA through directly sol-gel polymerization and high-temperature carbonization. The Gd 2 O 3 modified CA composites (named as Gdx-CA, where x means molar ratio of Gd 2 O 3 nanoparticles to carbon) are incorporated with S. Then, the products (S/Gdx-CA) are acted as sulfur host materials for Li-S batteries. The results demonstrate that adding ultrafine Gd 2 O 3 nanoparticles can dramatically improve the electrochemical properties of the composite cathodes. The S/Gd2-CA electrode (loading with 58.9 wt% of S) possesses the best electrochemical properties, including a high initial capacity of 1210 mAh g −1 and a relatively high capacity of 555 mAh g −1 after 50 cycles at 0.1 C. It is noteworthy that the performance of long-term cycle (350 cycles) for the S/Gd2-CA (317 mAh g −1 after 100 cycles and 233 mAh g −1 after 350 cycles at 1 C) is improved significantly than that of S/CA (150 mAh g −1 after 150 cycles at 1 C). Overall, the enhancement of electrochemical performances can be due to the strong polar nature of the ultrafine Gd 2 O 3 nanoparticles, which provide strong adsorption sites to immobilize S and polysulfide. Furthermore, the Gd 2 O 3 nanoparticles present a catalytic effect. Our research suggests that adding Gd 2 O 3 nanoparticles into S/CA composite cathode is an effective and novelty method for improving the electrochemical performances of Li-S batteries.
