Naphthalimide-functionalized pH-responsive amphiphilic block copolymer nanoparticles

Fan Kong,Fei Fang

An amphiphilic block copolymer, poly(ethylene glycol)- b -poly( N -vinyl carbazole), functionalized with a 1,8-naphthalimide fluorescent signaling moiety containing a pH-responsive piperazinyl recognition unit (PEG- b -PVK- co -PNA), was synthesized via reversible addition-fragmentation chain transfer polymerization. The block copolymer demonstrates a low molar mass dispersity and spontaneously self-assembles into uniform nanoparticles in aqueous media, serving as a fluorescent sensor for detecting hydrogen ion. The emission from the PVK block is effectively quenched, whereas the 1,8-naphthalimide moiety exhibits a pronounced fluorescence enhancement at 505 nm due to Förster resonance energy transfer between the PVK donor and the 1,8-naphthalimide acceptor in the copolymer nanoparticles. The photoluminescence intensity at 505 nm displays a linear correlation with decreasing pH values in the range 3.0–6.5 for the copolymer nanoparticles in acidic medium, highlighting the block copolymer's potential for quantitative pH sensing in acidic environments. © 2025 Society of Chemical Industry.

pH-responsive alliin delivery system: sustained intestinal release

Ying Zhu,Yapeng Chen,Yijia Yao,Xinkuan Li,Meiyi Wang

Alliin, a natural bioactive compound derived from garlic, exhibits potent antibacterial and antioxidant properties. However, its application in food and medicine is limited by degradation in the acidic gastric environment. In this study, we developed a novel pH-responsive delivery system based on hollow mesoporous silica nanoparticles (HMSNs) encapsulated within sodium alginate-starch (SA/ST) composite gel beads. The HMSNs were synthesized via a soft-template method and loaded with alliin, achieving an encapsulation efficiency of 31.5% and a drug loading capacity of 48.4%. The composite gel beads demonstrated excellent pH-sensitive behavior, with minimal alliin release (9%) in simulated gastric fluid (pH 1.2) and sustained release (91% over 36 h) in simulated intestinal fluid (pH 7). The system exhibited significant antioxidant activity, with DPPH and hydroxyl radical scavenging rates of 80.2% and 57.3%, respectively, comparable to free alliin. Furthermore, the beads showed strong antibacterial effects against Staphylococcus aureus and Escherichia coli , with inhibition rates of 80.65% and 72.73%, respectively, under intestinal conditions. This study highlights the potential of the alliin@HMSNs@SA/ST system as a smart delivery platform for functional foods and nutraceuticals, offering targeted protection and controlled release of bioactive compounds in the gastrointestinal tract.

pH Switchable Emulsions Based on Dynamic Covalent Surfactants

Gaihuan Ren,Lei Wang,Qianqian Chen,Zhenghe Xu,Jian Xu,Dejun Sun

Dynamic covalent surfactants were designed to prepare pH switchable emulsions. A dynamic covalent bond between nonamphiphilic building blocks (polyethylenimine (PEI) and benzaldehyde (B)) was introduced to form the dynamic covalent surfactant PEI-B. The dynamic nature of covalent bond in PEI-B was confirmed by 1H NMR and fluorescence probe analysis. Stable emulsions were successfully prepared with interfacial active PEI-B at pH 7.8 with various water/paraffin oil ratios under sonication. When lowering the pH to 3.5, a complete phase separation was observed as a result of breaking dynamic covalent bond in the interfacial active PEI-B. After tuning the pH back to 7.8, stable emulsion was obtained again due to the reformation of the dynamic covalent bond and hence interfacial active PEI-B. The emulsification and demulsification were dependent on the formation and breaking of dynamic covalent bond in PEI-B. Such pH-triggered emulsification and demulsification can be switched at least three times. Application of dynamic covalent surfactants will open up a novel route for preparing responsive emulsions.

Carbon nanotube field-effect transistor based pH sensors

Kemin Wang,Xiaofeng Liu,Zijun Zhao,Luyao Li,Jiajun Tong,Qian Shang,Yiwei Liu,Zhiyong Zhang

Field-effect transistor (FET) biosensor has received widespread attention due to their unique high-sensitivity properties. In conventional FET biosensors, the complicated interaction between the sample solution and the semiconductor channel always results in poor stability. Especially for FET-based pH sensors, the interface failure caused by the harsh acid-base working environment makes accurate, continuous, and low hysteresis pH measurement quite challenging. Here, we report a carbon nanotube (CNT)-FET pH sensor with enhanced environmental stability by introducing HfO 2 film into the gate insulator. The proposed devices show desired immunity to environmental interference including various chemical and physical factors. More importantly, we verified the feasibility of this CNT-FET device for continuous pH sensing, which demonstrates a sensitivity of 67.62 mV/pH, a wide pH range of 1.34–12.68, and low hysteresis of 500 pA with pH switching loops of 4.22–10.24. This work not only realizes a high-performance pH sensor but also paves the way for the development of FET biosensors with high environmental stability, which is of great significance for FET biosensors to maintain normal working performance in complex systems.

pH-sensitive polyion nanocomplexes for antimicrobial peptide delivery

Shuting Wang,Yan Yu,Heyang Li,Yan Huang,Jing Wang,Qiao Jin,Jian Ji

As an alternative to antibiotics, antimicrobial peptides (AMPs) are attracting more and more attention for non-antibiotic antibacterial therapy. However, there are still many issues of AMPs for clinical applications. For example, AMPs are generally positively charged and can be easily cleared during blood circulation. In addition, the cationic AMPs show strong cytotoxicity, which brings potential biosafety risks. In order to address these issues, pH-sensitive polyion nanocomplex is designed for the delivery of AMPs by electrostatic self-assembly of positively charged AMPs Magainin-I and negatively charged 2,3-dimethyl maleic anhydride (DA) modified ε-polylysine (EPL) (EPL-DA). The size and surface charge of the polyion nanocomplex is fully investigated by dynamic laser scattering (DLS) and transmission electron microscope (TEM). Magainin-I loaded polyion nanocomplex is stable in physiological environment (pH 7.4), which can effectively reduce the cytotoxicity of AMPs. In the slightly acidic environment (pH 5.0–6.0) in bacteria infected tissue, the nanocomplex would be disintegrated to positively charged EPL and Magainin-I, thereby restoring antibacterial properties. The excellent antibacterial activity of Magainin-I loaded polyion nanocomplexes is confirmed by a series of in vitro antibacterial experiments. The fabrication of polyion nanocomplex provides an innovative way for the delivery of AMPs. Graphical

pH-Responsive Liquid Marbles Based on Dihydroxystearic Acid

Jianxin Zhang,Yao Gu,Jianzhong Jiang,Raojun Zheng

Herein, we report pH-responsive liquid marbles stabilized by 9,10-dihydroxystearic acid (DHSA). The particle morphology and the pH-responsive behavior of the liquid marbles were investigated. The rolling time during the preparation of liquid marbles has a great influence on the thickness of powder adsorption and the stability of the marbles. Compared with the liquid marbles stabilized by other fatty acids (e.g., stearic acid and docosoic acid), the liquid marbles prepared by DHSA have a much higher mechanical robustness. The increase in the number of hydroxyl groups on the carbon chain of fatty acids improves the mechanical robustness of the liquid marbles. Such liquid marbles immediately disintegrated on the surface of an alkaline solution or after exposure to NH3 gas, which extends their applications in the NH3 sensor and chemical reactions.

Multicyclic topology-enhanced micelle stability and pH-sensitivity

Wei Ma,Shuang Li,Li-Wei Ma,Cui-Yun Yu,Hua Wei

Cyclic polymers have been reported to show unique properties and often better performance than linear analogues due to the absence of polymer chain ends, therefore great efforts have been devoted to the development of various cyclic polymers for biomedical applications. However, the effect of multicyclic topology on the stimuli-responsiveness of polymers has not been elucidated so far, to our knowledge, likely due to the lack of an efficient and powerful strategy for the controlled preparation of multicyclic polymers with well-defined structures and compositions. Clarification of this structure–property relationship will not only contribute to the rational design and development of novel cyclic polymer-derived materials with better properties for biomedical applications, but also promote greatly the clinical translations of these cyclic topology-based materials. A panel of amphiphilic pH-responsive multicyclic polymers (MCPs) were prepared via reversible addition-fragmentation chain-transfer (RAFT) copolymerization of a cyclic macromonomer, poly(oligoethylene glycol methacrylate) (M− c POEGMA) and N,N -dimethyl aminoethyl methacrylate (DMAEMA). The effect of multicyclic topology on the colloidal stability and stimuli-responsiveness of the resulting self-assembled micelles was investigated in detail. Interestingly, MCP1 micelles with a polymer composition of P(DMAEMA 50 - st -(M− c POEGMA 7 ) 3 ) showed much greater stability with a CMC value as low as 130.5 nM and more complete acidic pH-triggered disassembly than the single cyclic polymers (SCPs)-based micelle counterparts, which further endows the doxorubicin (DOX)-loaded MCP1 micelles with an approximately twofold lower IC 50 value (greater in vitro cytotoxicity) than that of the SCP-based analogues. This study therefore highlights for the first time the incorporation of multicyclic topology as a useful and efficient alternative to the simultaneously enhanced colloidal stability and stimuli-responsiveness of polymeric self-assemblies for controlled release application.

New fluorescent pH indicators for characterizing cement carbonation

Ning Du,Ruifang Guan,Pengkun Hou

Fluorescent indicators of a wider labeling pH range of 7.0–12.5 were prepared to characterize the pH evolution of the accelerated/natural carbonation of cement and the corrosion of steel bar. Compared with conventional indicators (phenolphthalein, pH range of 8.2–8.5), these indicators indicate multiple pH-induced switches of different fluorescence emissions for the fluorescence resonance energy transfer between donor–acceptor. The results showed that the newly-developed indicators are sensitive and applicable for labeling both natural and accelerated carbonation of cement. Investigations by Fourier Transform infrared spectroscopy, X-ray diffraction, scanning electron microscopy, and thermogravimetric analysis demonstrated the reliability of these indicators, that is, in the “uncarbonated area” shown by phenolphthalein, carbonization actually occurs as exhibited by fluorescent indicators. Furthermore, electrochemical impedance spectroscopy, as well as dynamic potential polarization studies showed that rusting/corrosion of reinforcing steel started when the pH dropped to 11.5 in the simulation study. This work promoted the contribution of fluorescent indicators in cement carbonation characterization, with a wide pH response.

pH-Regulated supramolecular chirality of phenylalanine-based hydrogels

A. Kousar,J. Liu,N. Mehwish,F. Wang,A.Y. Dang-i,C. Feng

To achieve desired physicochemical and biochemical activities of optically active molecules at targetted sites in biological systems, tuning of chiral sense by varying internal conditions (e.g., pH) is of utmost importance. However, it has been a challenge to gain the balance of non-covalent interactions required for chiral transformations merely by deviations in biochemical environment. Herein, pH-controlled chirality of supramolecular nanostructures obtained from coassembly of phenylalanine-derived gelator and achiral cationic polyacrylamide has successfully been achieved. Specifically, any changes in the pH remarkably influence the chirality of these nanostructures. For instance, coassembled hydrogels possessing l -enantiomer provide right-handed nanostructures at pH < 7; whereas at pH ≥ 7, handedness of nanostructures switches to the left-handed nanostructures. Moreover, viscoelastic properties and twist pitch were also regulated under the influence of pH change. This study may provide a methodology to control chirality by simply varying biochemical conditions, which points toward its potential to be used in biological systems as a key to gain preferential chiral sense in different sites of living systems.

Dual thermal- and pH-responsive polypeptide-based hydrogels

Meng Fan-dong,Ni Yun-xia,Ji Si-fan,Fu Xiao-hui,Wei Yu-han,Sun Jing,Li Zhi-bo

Smart hydrogels have received increasing attention for their great potential for the applications in many fields. Herein, we report a facile approach to prepare a class of dual-responsive hydrogels assembled from synthetic statistical/block thermal-responsive copoly(L-glutamate)s copolymerized with poly(ethylene glycol), which were prepared by ring-opening polymerization (ROP) and post-modification strategy. The incorporation of oligo(ethylene glycol) (OEG) and glutamic acid residues offers the gels with thermal- and pH-responsive properties simultaneously. We have systematically studied the influence of both temperature and pH on the gelation behaviors of these copolymers. It is found that the increase of glutamic acid content and solution pH values can significantly suppress the gelation ability of the samples. Circular dichroism (CD) results show that the α -helix conformation appears to be the dominant secondary conformation. More interestingly, the gelation property of the block copolymer with statistical thermal-responsive copoly(L-glutamate)s shows greater dependence on pH as compared to that with block segments due to the distinct morphology of the self-assemblies. The obtained hydrogels exhibit pH-dependent and thermal-responsive gelation behaviors, which enable them as an ideal smart hydrogel system for biomedical applications.

A light-sensitive protein-based wearable pH biometer

Wenxue Li,Siyuan Chen,Shiwang Xie,Xianmao Lu,Zhou Li,Yujia Lv,Dan Luo

Bacteriorhodopsin is a biological material with excellent photosensitivity properties. It can directly convert optical signals into electrical signals and is widely used in various biosensors. Here, we present a bR-based wearable pH biometer that can be used to monitor wound infection. The mechanism of the pH-sensitive effect of the bR electrode is explained, which generates a transient photovoltage under light irradiation and a negative photovoltage when the lamp is turned off. Since the photoelectric signal of bR is affected by different pH values, the photovoltage is changed by adjusting the pH value. The ratio (Vn/Vp) of negative photovoltage (Vn) to positive photovoltage (Vp) has a good linear relationship (R2 = 0.9911) in the pH range of 4.0–10.0. In vitro experiments using rats as a model confirmed that this wearable pH biometer can monitor pH changes that occur in wound infection.

Reversible conductivity modulation of PEDOT:PSS based on pH

Feng-Fei Pang,Sen Li,Wen-Quan Sun,Guo-Zhi Han

We report the reversible conductivity and morphology modulation of PEDOT:PSS composites based on pH. Through studying effects of pH on the morphology and conductivity of PEDOT:PSS composites, it was found that different pH of PEDOT:PSS composites solution will result in different morphology of PEDOT:PSS composites film . At the same time, conductivity of the film also changed accordingly. With the increase of pH from 3 to 11, the microstructure of PEDOT:PSS film reversibly changed from a continuous network to non-continuous clusters, and the corresponding conductivity decreased from 0.62 S/cm to 0.42 S/cm. Furthermore, the mechanism was innovatively investigated by Raman spectra , along with UV–Vis spectra and SEM .
