The solvent effect modulates the formation of homogeneous polyphenol composite hydrogels with improved transparency and mechanical strength for antibacterial delayed sternal closure films

Jiahui Xie,Dawei Jin,Jiaqi Qiu,Jingyuan Cui,Meng Yin,Xue Qu

The usage of delayed sternal closure films after thoracotomy surgery helps doctors deal with emergency conveniently. There is a growing demand to develop suturable, antibacterial and transparent films for delayed sternal closure. Although polyphenol incorporated hydrogels provide good suture ability, they lose transparency because of the heterogeneous distribution of polyphenols during the post-immersion process. Here, a solvent exchange method is proposed to fabricate homogeneous polyphenol composite hydrogels in a bottom-up manner, which utilizes the distinct solvent effect of DMSO and H2O to modulate the association and disassociation between polyphenols and the polymer backbones on demand. DMSO first provides a protective environment to turn off the intermolecular interactions and allows tannic acid (TA) to be dispersed into the polymer network PEG–lysozyme (PEG–LZM) homogeneously. The following water rehydration turns on the intermolecular interactions between titanic acid and PEG–lysozymes, and results in a homogeneous titanic acid toughened composite hydrogel (PEG–LZM–TA (DH)), which has an improved transparency and mechanical properties than those of the materials prepared by the post-immersion method. In addition, the TA integration provides antibacterial function to the hydrogels. We establish a rabbit delayed sternal closure model to demonstrate that PEG–LZM–TA (DH) films can be sutured to temporarily close the thoracic cavity of rabbits, provide a transparent window to inspect the wound at any time, and control the bacterial contamination efficiently. We further explore the solvent exchange method to other polyphenols and polymeric hydrogel composites. The results suggest that the solvent exchange method provides generic opportunities to fabricate homogeneous polyphenol strengthened hydrogel systems with high performance.

Multomics Analysis of the Characteristic Changes in Polyphenol Accumulation and Cell Wall Polysaccharide Remodelling During the Development of Zingiber mioga Roscoe Flower Buds

Chenglin Tang,Cheng Zhang,Xingyu Chen,Luolin Bao,Jiao Xie

Background/Objectives : At present, there are only a few studies on characteristic changes in polyphenols and cell wall polysaccharides and their correlations in Z. mioga flower buds during development. Methods : Polyphenols were analysed using ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). Content and enzyme activities of cell wall components were examined using a microplate reader. Expression of genes related to these components was detected using de novo-Seq. Results : Most polyphenols accumulated significantly, with the highest levels being found in cyanidin-3- O -glucoside and epicatechin. PCA results show that changes in polyphenols were largely dependent on the germination and ripening stage, which might represent its specific period. Additionally, the increased flavonoid and anthocyanin fractions might be due to the up-regulated expression of DFR 1/2, ANS and BZl . During development, PME, PG and β -galactosidase synergistically break down protopectin to soluble pectin; PME coordinates with cellulase in cellulose degradation, while xylanase dominates hemicellulose degradation. Lac collaborated with PME, PG and cellulase to regulate lignin synthesis. Key upregulated genes driving cell wall polysaccharide alterations include PME 35, PG and GAUT 7 for pectin metabolism, CESA2/3 for cellulose synthesis, and Lac 25, POD 6/7/47/52 and CCR 6 for lignin synthesis. Correlation analysis revealed that the synergistic effects of p -coumaric acid, chlorogenic acid, epicatechin, cyanidin-3- O -glucoside, peonidin-3- O -glucoside, protopectin, pectin, lignin and cellulose might be responsible for the sensory quality formation in Z. mioga . Conclusions : This study further investigates the binding mode of polyphenols and cell wall polysaccharides, providing a theoretical basis for understanding the development of sensory qualities in Z. mioga flower buds during growth and maturation.

Optimization of hollow ZIF-8/GO composite imprinted membrane synthesis by polyphenol etching technique and study of its selective separation performance for ribavirin

Yilin Wu,Yanke Xu,Faguang Ma,Jingjing Zhen,Yue Li,Fanlei Kong,Ming Yan

In this study, modified hollow e-ZIF-8 nanoparticles with a hollow structure and hydroxyl-rich surface were ingeniously synthesized through a one-step etching method, thereby increasing the contact area and addressing the issue of stacking imprinting sites, fundamentally enhancing selectivity. Among these, the hollow structure of the e-ZIF-8 nanoparticles not only increased the interlayer spacing of the graphene oxide (GO) nanosheets, providing additional pathways for water molecules, but also, through π-π interactions, cross-linked the phenol-modified nanoporous metal-organic frameworks (MOFs) with the GO nanosheets, significantly enhancing the structural stability and anti-fouling capability of the composite membrane. The effects of different e-ZIF-8 addition amounts on membrane flux and hydrophilicity were investigated, constructing a GO-based imprinting structure on the stacked e-ZIF-8/GO nanosheet surfaces, offering a novel strategy to improve the selectivity and stability of the membrane. The maximum adsorption capacity of e-MG-IMs reached 38.35 mg g −1 , demonstrating good selective separation factors ( α RBV/ACV  = 3.24, α RBV/AZT = 2.41 and α RBV/LAM  = 4.21), as well as excellent permeation selectivity ( β ACV/RBV  = 4.75, β AZT/RBV  = 4.42 and β LAM/RBV  = 5.92). In this study, the optimized regulation of GO nanosheets extended the molecular transport channels, facilitating the construction of more molecular recognition sites, thus positively impacting the separation performance of the imprinting membrane.

Low MWCO nanofiltration membranes prepared by stepwise depositing polyphenol and aminosilane coupling agent with the assistance of ethanol-regulated oxidation for antibiotic removal

Runkai Su,Guoying Feng,Zhiguo Yan,Man Xu,Cunwen Wang,Yanbo Li

Mussel-inspired catechol amine chemistry has emerged as a novel strategy to fabricate nanofiltration (NF) membranes. However, the preparation takes hours, and the molecular weight cut-off (MWCO) of the resulting membranes is usually not low enough to reject small molecules. This work described a new method to prepare dense NF membranes using tannic acid (TA) and aminosilane coupling agent (KH792 (N-(2-aminoethyl)-3-aminopropyltrimethoxysilane)) as materials, with the assistance of ethanol-regulated oxidation. The NF membranes were made by stepwise depositing oxidized-TA and KH792 on PSF substrates. The oxidation of TA was regulated by adjusting the solvent composition or the dosage of NaIO 4 . The effect of NaIO 4 on the chemistry of TA was explored by various characterizations. The selective layer of an NF membrane, with a flux of 26.7 L·m −2 ·h −1 ·bar −1 and a MWCO of 200 Da, formed only within 10 min. The membrane had high rejections of naproxen, bezafibrate, and ibuprofen (>90 %), and the rejection of NaCl was lower than 20 %, exhibiting a good selectivity for antibiotics toward monovalent ions. Compared with literature-reported mussel-inspired membranes, the proposed method resulted in a membrane with much higher permeance and a lower MWCO, indicating the improvement and advantages among the Mussel-inspired methods for the synthesis of NF membranes.

Study on the stability of sodium alginate modified zein/adzuki bean seed coat polyphenol composite nanoparticles and the loading of curcumin in emulsion

Xu Zhang,Xiaozhuo Sun,Yelinxin Chen,Fangzhu Zheng,He Ban,Jingsheng Liu,Dan Cai,Lin Xiu

Curcumin, a bioactive compound characterized by its limited solubility in water and low bioaccessibility, which requires an effective delivery system to enhance stability and absorption. This research focused on creating Pickering emulsions stabilized by zein-adzuki bean seed coat polyphenol-sodium alginate composite nanoparticles (ZAS) to improve curcumin delivery. The effects of different concentrations of sodium alginate (SA) on the properties of ZAS were investigated. Fourier transform infrared spectroscopy (FTIR) showed that there were hydrophobic and hydrogen bonds interactions between SA and nanoparticles, and Differential Scanning Calorimetry demonstrated an improvement in thermal stability. Accelerated oxidation experiments showed that ZAS stabilized Pickering emulsions had good anti-oxidant capacity. In comparison to the control group, the addition of SA increased the loading rate of curcumin to 81.65 ± 1.56 % with improved bioaccessibility (27.99 ± 0.26 %) and in vitro digestion stability (55.72 ± 0.22 %). The findings indicate that ZAS nanoparticles modified with SA markedly enhance the stability of Pickering emulsions, while also attaining a high loading efficiency of 81.65 % and bioaccessibility of 27.99 %. This suggests their potential utility in functional food applications.

Structural evolution and prebiotic-like potential of insoluble dietary fiber-polyphenol complexes from Rosa roxburghii Tratt during in vitro digestion and colonic fermentation

Xiaoli Zhou,Yan Chen,Shengyan Zhang,ShuangQin Zhang,Tailan Liu,Nian Liu,Yanxin Yang

Rosa roxburghi Tratt. (RRT) pomace is rich in insoluble dietary fiber (IDF) and polyphenols, but free polyphenols are susceptible to premature loss during digestion. This study constructed an IDF–polyphenol complex (IDF-PP) from RRT pomace via non-covalent adsorption and evaluated its gastrointestinal fate and in vitro colonic fermentation behavior. IDF-PP reduced premature phenolic loss during simulated upper gastrointestinal digestion and enabled a more gradual release during fermentation. Structural analyses showed fermentation-induced remodeling, generating a porous residual matrix while retaining part of the polysaccharide framework. Compared with free polyphenols and IDF, IDF-PP increased short-chain fatty acid production, especially butyrate, and was associated with higher relative abundance of Prevotella -centered Bacteroidetes taxa and Faecalibacterium , together with lower relative abundance of Proteobacteria . Fermentation products also showed enhanced antioxidant and α-glucosidase inhibitory activities. These findings suggest that RRT pomace-derived IDF-PP is a promising fermentation-responsive functional ingredient with potential to modulate gut microbial fermentation.

Plant-Derived Polyphenol and LL-37 Peptide-Modified Nanofibrous Scaffolds for Promotion of Antibacterial Activity, Anti-Inflammation, and Type-H Vascularized Bone Regeneration

Shue Jin,Renli Yang,Chen Hu,Shiqi Xiao,Yi Zuo,Yi Man,Yubao Li,Jidong Li

The regeneration of oral tissues is a challenging clinical problem because of the complex microbial and biological stress environments. Electrospun fibrous scaffolds have attracted significant interest as effective barrier membranes for guided bone regeneration (GBR); however, no mature strategy yet exists for the surface modification of fibers to provide versatility to satisfy clinical requirements. This study demonstrated a practical biosafety strategy: the combined use of plant polyphenols and LL-37 peptides to modify the fiber surface to endow the fibrous scaffold with antimicrobial activity, immunoregulation, and vascularized bone regeneration. We confirmed that the LL-37 peptides interacted with tannic acid (TA) through noncovalent bonds through experiments and molecular docking simulation analysis. In vitro experiments showed that the TA coating imparted strong antibacterial properties to the fibrous scaffold, but it also caused cytotoxicity. The grafting of LL-37 peptide promoted the spreading, migration, and osteogenic differentiation of mesenchymal stem cells and was also conducive to the M2 polarization of RAW264.7 cells. In vivo experiments further verified that the LL-37 peptide-grafted fibrous scaffold significantly enhanced angiogenesis, anti-inflammatory effects, and type-H vascularized bone regeneration. Overall, the fibrous scaffold modified by the LL-37 peptide through TA grafting has significant potential for GBR applications.

Tea polyphenol modified, photothermal responsive and ROS generative black phosphorus quantum dots as nanoplatforms for promoting MRSA infected wounds healing in diabetic rats

Xu Shibo,Chang Linna,Hu Yanan,Zhao Xingjun,Huang Shuocheng,Chen Zhenhua,Ren Xiuli,Mei Xifan

Background Healing of MRSA (methicillin-resistant Staphylococcus aureus) infected deep burn wounds (MIDBW) in diabetic patients remains an obstacle but is a cutting-edge research problem in clinical science. Surgical debridement and continuous antibiotic use remain the primary clinical treatment for MIDBW. However, suboptimal pharmacokinetics and high doses of antibiotics often cause serious side effects such as fatal complications of drug-resistant bacterial infections. MRSA, which causes wound infection, is currently a bacterium of concern in diabetic wound healing. In more severe cases, it can even lead to amputation of the patient's limb. The development of bioactive nanomaterials that can promote infected wound healing is significant. Results The present work proposed a strategy of using EGCG (Epigallocatechin gallate) modified black phosphorus quantum dots (BPQDs) as therapeutic nanoplatforms for MIDBW to achieve the synergistic functions of NIR (near-infrared)-response, ROS-generation, sterilization, and promoting wound healing. The electron spin resonance results revealed that EGCG-BPQDs@H had a more vital photocatalytic ability to produce singlet oxygen than BPQDs@H. The inhibition results indicated an effective bactericidal rate of 88.6% against MRSA. Molecular biology analysis demonstrated that EGCG-BPQDs significantly upregulated CD31 nearly fourfold and basic fibroblast growth factor (bFGF) nearly twofold, which were beneficial for promoting the proliferation of vascular endothelial cells and skin epidermal cells. Under NIR irradiation, EGCG-BPQDs hydrogel (EGCG-BPQDs@H) treated MIDBW area could rapidly raise temperature up to 55 °C for sterilization. The MIBDW closure rate of rats after 21 days of treatment was 92.4%, much better than that of 61.1% of the control group. The engineered EGCG-BPQDs@H were found to promote MIDBW healing by triggering the PI3K/AKT and ERK1/2 signaling pathways, which could enhance cell proliferation and differentiation. In addition, intravenous circulation experiment showed good biocompatibility of EGCG-BPQDs@H. No significant damage to major organs was observed in rats. Conclusion sThe obtained results demonstrated that EGCG-BPQDs@H achieved the synergistic functions of photocatalytic property, photothermal effects and promoted wound healing, and are promising multifunctional nanoplatforms for MIDBW healing in diabetics.Graphical Abstract

Nanoization of Technical Pesticides: Facile and Smart Pesticide Nanocapsules Directly Encapsulated through “On Site” Metal–Polyphenol Coordination Assembly for Improved Efficacy and Biosafety

Yuxia Chen,Hang Li,Yuhui Peng,Tongtong Li,Xiaona Li,Chen Wang,Ruixi Xiao,Jiangtao Dong,Xuezhong Du

Facile pesticide nanocapsules were successfully prepared by directly encapsulating the antisolvent precipitation of pesticides through instantaneous “on site” coordination assembly of tannic acid and Fe3+, avoiding tedious preparation, time consumption, and large amounts of organic solvents. The pesticide nanocapsules showed excellent resistance to ultraviolet photolysis and rainwater washing owing to the nanocapsule walls. The smart pesticide nanocapsules exhibited the controlled release of pesticides under multidimensional stimuli, such as acidic/alkaline pH, glutathione, H2O2, phytic acid, laccase, tannase, and sunlight, which were related to the physiological and natural environments of crops, pests, and pathogens. The tebuconazole nanocapsules not only enhanced the fungicidal activity against Fusarium graminearum and effective control efficacy in wheat powdery mildew through foliar spray and seed coating, but also improved the biosafety of target plant growth and nontarget organisms. The facile, smart, efficient, safe, and green pesticide nanocapsules using the universal strategy have broad application prospects in ecoagriculture.

Efficient polyphenol extraction from Moringa oleifera Lam. leaves using natural deep eutectic solvents: COSMO-RS screening, ANN-GA optimization and antioxidant activity evaluation

Weilong Peng,Lijun Wang,Xiaoguang Wang,Yaya Wang,Weimei Wang,Junjie Huang,Ruigang Zhou,Chaoyang Chen,Ruonan Bo,Mingjiang Liu,Jingui Li

A novel method for extracting polyphenols from Moringa oleifera Lam. ( M. oleifera ) using natural deep eutectic solvents (NADES) has been developed, with extraction conditions optimized via an artificial neural network-genetic algorithm (ANN-GA) to maximize efficiency. Traditional extraction methods for polyphenols from M. oleifera leaves, such as ethanol extraction, are often associated with low efficiency, environmental concerns, and high operational costs. Furthermore, the empirical selection of solvents and extraction conditions is time-consuming and lacks precision. In this study, 28 NADES were employed to predict the solubility of polyphenol molecules through a conductor-like screening model for realistic solvents (COSMO-RS) model. Based on the results, eight highly effective NADES combinations were selected and experimentally verified. The betaine-maleic acid (Bet-MA) combination with the ultrasonic-assisted extraction showed the highest extraction rate of M. oleifera leaves polyphenol (MOLP), which was 1.56 times that of ultrasonic-assisted ethanol solvent. Simplex centroid mixture design (SCMD) and two-level factor experiments were conducted to optimize the ratio of betaine, maleic acid, and water and identify the most influential extraction factors on MOLP yield. The ANN-GA model was further applied to refine the extraction conditions and analyze their interactions on MOLP content. Additionally, antioxidant assays demonstrated that MOLP extracted with the Bet-MA solvent displayed significant antioxidant activity. In conclusion, this study demonstrates that COSMO-RS and ANN-GA may serve as powerful tools for solvent selection and extraction process optimization, offering a green and efficient approach for polyphenol extraction from M. oleifera leaves. The findings highlight the potential of NADES-based extraction methods for broader applications in the extraction of various bioactive substances, contributing to the development of sustainable and environmentally friendly extraction technologies.

Effect of Surface Crosslinking on the Wear and Friction Behavior of Tea Polyphenol Stabilized Ultra-High Molecular Weight Polyethylene for Total Joint Replacements

Shah Nouman Ali,Ahmad Adnan,Gul Rizwan M.,Kamran M. Ali,Ishaq M. Aaqib,Shakir Namra,Xu Jia-Zhuang,Li Zhong-Ming,Sağbaş Binnur

Highly crosslinked ultra-high molecular weight polyethylene (UHMWPE) with vitamin E (VE) as an antioxidant is widely used for joint implants. VE helps resist oxidation but raises concerns about reduced crosslink density and wear performance. This research hypothesized that tea polyphenols (tPPs), such as lipid-soluble epigallocatechin gallate (lsEGCG) and epigallocatechin gallate (EGCG), could prevent the usual decrease in crosslink density caused by VE, thereby enhancing wear performance. The antioxidants were blended with UHMWPE at 0.2 wt% and surface chemically crosslinked using di-cumyl peroxide. The surface properties, including crosslink density, roughness, coefficient of friction, and wear performance, were evaluated in detail. The results showed that lsEGCG and EGCG blended UHMWPE had significantly higher crosslink density compared to VE stabilized UHMWPE, which was 17% lower than virgin UHMWPE. The coefficient of friction increased after crosslinking and was higher in tPPs blended UHMWPE, indicating a highly crosslinked network structure. The wear resistance of surface crosslinked tPPs stabilized UHMWPE was significantly higher than VE-stabilized UHMWPE. Additionally, a substantial number of scratches, furrows, and flakes were observed on the surface of VE-stabilized UHMWPE compared to tPPs stabilized UHMWPE. It was concluded that tPPs are promising alternatives to VE for improving the performance and longevity of UHMWPE-based implants.

Investigating the effects of probiotic fermentation on the pancreatic lipase inhibitory activity of sea buckthorn polyphenol extracts by using untargeted metabolomics and multispectral analysis

Yueling Shang,Jinhui Yi,Ying Fan,Shihaoran Xu,Xin Zhao,Xujia Dai,Xia Zhao,Yanli Liu,Jinbin Liu,Xiaohong Yu

In this study, sea buckthorn juice phenolics, free, bound, and conjugated fractions, were investigated for pancreatic lipase (PL) inhibition, and conjugated phenolics (CPs) were found to exhibit the highest inhibitory activity (IC50: 253.9 μg/mL pre-fermentation, 185.2 μg/mL post-fermentation). Fermentation modified the conjugated phenolic composition, upregulating 31 compounds and downregulating 18 compounds, notably protocatechuic acid and isorhamnetin derivatives. Importantly, kinetic analysis demonstrated a fermentation-induced shift in the inhibition mode from mixed-type (unfermented CPs [CCP]) to uncompetitive (fermented CPs [FCP]), suggesting its potential application as an adjunctive therapy. Spectroscopic studies indicated that CP-PL interactions occurred through static quenching and hydrophobic forces, with FCP inducing secondary structural perturbations in PL. Notably, FCP demonstrated synergistic effects with orlistat, suggesting its potential for combined therapeutic applications. These results highlight the value of sea buckthorn as a source of anti-obesity polyphenols and provide a mechanistic basis for the development of functional ingredients that target PL activity regulation. These findings suggest that fermented sea buckthorn phenolics are promising candidates for metabolic disorder interventions owing to their dual inhibition mechanism and structural modulation of PL.
