Quality Characteristics and Anthocyanin Profiles of Different Vitis amurensis Grape Cultivars and Hybrids from Chinese Germplasm

Lei Zhu,Xinyue Li,Xixi Hu,Xin Wu,Yunqing Liu,Yiming Yang,Yanqing Zang,Huacheng Tang,Changyuan Wang,Jingyu Xu

To evaluate the importantVitis amurensisgermplasm, the quality characteristics and anthocyanin profiles of the ripe berries of 20V. amurensisgrapes and 11 interspecific hybrids in two consecutive years were analysed. Compared with theV. viniferagrapes,V. amurensisgrapes had small berries with low total soluble solids and high titratable acids, and were richer in phenolic compounds except for flanan-3-ols in their skins but had lower phenolic contents in their seeds and showed lower antioxidant activities. An outstanding feature of theV. amurensisgrapes was their abundant anthocyanin contents, which was 8.18-fold higher than the three wine grapes ofV. vinifera. The anthocyanin composition ofV. amurensiswas characterized by an extremely high proportion of diglucoside anthocyanins (91.71%) and low acylated anthocyanins (0.04%). Interestingly, a new type of speculated 3,5,7-O-triglucoside anthocyanins was first identified and only detected inV. amurensisgrapes and hybrids. Based on the total phenolic and anthocyanin characteristics,V. amurensisgrapes were set apart fromV. viniferacultivars and the interspecific hybrids, for the same qualities, fell between them, as assessed by principal component analysis. Keywords:Vitis amurensis;anthocyanin composition and content;total phenolic content;general characteristic;antioxidant activity

Synthesis, characterization, in vitro antioxidant and hypoglycemic activities of selenium nanoparticles decorated with polysaccharides of Gracilaria lemaneiformis

Li Tang,Xiaomin Luo,Meiyuan Wang,Zhong Wang,Juan Guo,Fansheng Kong,Yongguang Bi

Under a simple redox system of selenite and ascorbic acid , we used Gracilaria lemaneiformis polysaccharides (GLPs) as a stabilizer and dispersing agent to generate well-dispersed and stable selenium nanoparticles (SeNPs). The size, stability, morphology and physicochemical properties of GLPs-SeNPs were characterized by dynamic light scattering (DLS), scanning electron microscopy (SEM), transmission electron microscopy (TEM), Ultraviolet-visible spectroscopy (UV–vis), Fourier transform infrared (FT-IR) spectra, energy dispersive X-ray (EDX), X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), Thermogravimetric (TG). The results showed that orange-red, amorphous, zero-valent and spherical GLPs-SeNPs with mean diameter of approximately 92.5 nm were successfully prepared, which exhibited good storage stability at 4 °C and remaining highly stable at different ion strengths and pH. The 2,2-diphenyl-1-pycrylhydrazyl (DPPH), 2,2′-Azinobis-(3-ethylbenzthiazoline-6-sulphonate) (ABTS) and superoxide anion radical (O 2 •− ) radical scavenging ability of GLPs-SeNPs were higher than those of bare SeNPs, GLPs and sodium selenite (Na 2 SeO 3 ), and could reach 103.41%, 94.23%, 86% at a concentration of 1.5 mg/mL, respectively. Besides, GLPs-SeNPs also showed higher inhibitory effects on α-amylase and α-glucosidase. In vitro cytotoxicity assay and hemolysis activity examinations indicated that GLPs-SeNPs have excellent biocompatibility. Therefore, the GLPs-SeNPs might be used as a potential antioxidant agent and antidiabetic agent for food and medical applications.

Target Functionalized Carbon Dot Nanozymes with Dual-Model Photoacoustic and Fluorescence Imaging for Visual Therapy in Atherosclerosis

Qiao Chen,Xinmei Duan,Yao Yu,Rongrong Ni,Guojing Song,Xu Yang,Li Zhu,Yuan Zhong,Kun Zhang,Kai Qu,Xian Qin,Wei Wu

Multifunctional nanomedicines have been used in atherosclerosis theranostics. Herein, phosphatidylserine-specific peptide CLIKKPF-functionalized carbon-dots nanozymes (pep-CDs) are reported for specific and efficient noninvasive theranostic of atherosclerosis. Surprisingly, pep-CDs are discovered to not only inherit the inherent properties of carbon dots (CDs), including deep-red fluorescence emission, photoacoustic response, and superoxide dismutase-like antioxidant, and anti-inflammatory activities but also possess the ability to target recognition on foam cells and target localization on plaques due to the specific interaction of CLIKKPF with phosphatidylserine on the membrane outer surface of foam cells. Furthermore, the target localization effect of pep-CDs vastly promotes the efficient accumulation of CDs in plaque, thus maximizing AS theranostic of CDs. Interestingly, pep-CDs could be developed to image plaque for monitoring atherosclerosis pathological progression in real-time resulting from the different content of foam cells. This work on the one hand proposes a simple and feasible strategy to construct theranostic nanoplatform employing only a single functional unit (i.e., multifunctional CDs) to simplify the fabrication procedure, on the other hand, highlights the advantages of the active target auxiliary mode for atherosclerosis theranostic applications.

Enhanced stabilization of multifunctional phenolic acids-grafted chitin nanofibers for Pickering emulsions

Mengxia Duan,Cailing Tong,Chen Huang,Zhisheng Tong,Jingting Xu,Danjie Li,Jie Pang,Zijian Zhi,Chunhua Wu

The objective of this study was to develop novel functional stabilizers for Pickering emulsions using phenolic acids-grafted chitin nanofibers (phenolic acids-g-ChNF), which were fabricated by grafting ferulic acid (FA), sinapic acid (SA) and caffeic acid (CA) onto ChNF via free radical-mediated method. The Fourier transform infrared spectrum and Proton nuclear magnetic resonance showed that graft copolymerization occurred between the amino groups of ChNF and the carbonyl of the phenolic acids. Further, it was revealed that CA-g-ChNF and SA-g-ChNF possessed stronger antioxidant and antibacterial properties than the original ChNF and FA-g-ChNF. Additionally, we applied phenolic acids-g-ChNF to develop Pickering emulsions and found that SA-g-ChNF- and CA-g-ChNF-stabilized emulsions displayed reduced droplet sizes compared to FA, the main reason for which was that SA and CA had a rather close bonding relationship with ChNF. Taken together, SA-g-ChNF and CA-g-ChNF as novel multi-functional particles can be employed for facilitating the stability of Pickering emulsions.

Antioxidant Activity of Auricularia auricula Polysaccharides with Different Molecular Weights and Cytotoxicity Difference of Polysaccharides Regulated CaOx to HK-2 Cells

Bao-Li Heng,Fan-Yu Wu,Jing-Hong Liu,Jian-Ming Ouyang

Objective. This study aimed to investigate the growth of calcium oxalate (CaOx) crystals regulated by Auricularia auricular polysaccharides (AAPs) with different viscosity-average molecular weights (), the toxicity of AAP-regulated CaOx crystals toward HK-2 cells, and the prevention and treatment capabilities of AAPs for CaOx stones. Methods. The scavenging capability and reducing capacity of four kinds of AAPs ( of 31.52, 11.82, 5.86, and 3.34 kDa) on hydroxyl, ABTS, and DPPH free radicals and their capability to chelate divalent iron ions were detected. AAP-regulated CaOx crystals were evaluated by using zeta potential, thermogravimetric analysis, X-ray diffraction, and scanning electron microscopy. The cytotoxicity of AAP-regulated crystals was evaluated through examination of cell viability, cell death, malondialdehyde (MDA) content, and cell surface hyaluronic acid (HA) expression. Results. The in vitro antioxidant activities of the four AAPs were observed in the following order: AAP0 < AAP1 < AAP2 < AAP3. Thus, AAP3, which had the smallest , had the strongest antioxidant activity. AAPs can inhibit the growth of CaOx monohydrate (COM), induce the formation of CaOx dihydrate (COD), and reduce the degree of crystal aggregation, with AAP3 exhibiting the strongest capability. Cell experiments showed the lowest cytotoxicity in AAP3-regulated CaOx crystals, along with the lowest MDA content, HA expression, and cell mortality. In addition, COD presented less cytotoxicity than COM. Meanwhile, the cytotoxicity of blunt crystals was less than that of sharp crystals. Conclusion. AAPs, particularly AAP3, showed an excellent antioxidative capability in vitro, and AAP3-regulated CaOx crystals presented minimal cytotoxicity.

Exploring Antioxidant and α-Glucosidase Inhibitory Activities in Mulberry Leaves (Morus alba L.) across Growth Stages: A Comprehensive Metabolomic Analysis with Chemometrics

Wenjie Li,Shenghui Hao,Hengyang Li,Qi An,Lina Yang,Bing Guo,Zijing Xue,Yongli Liu,Long Guo,Yuguang Zheng,Dan Zhang

Metabolic product accumulation exhibited variations among mulberry (Morus albaL.) leaves (MLs) at distinct growth stages, and this assessment was conducted using a combination of analytical techniques including high-performance liquid chromatography (HPLC), gas chromatography–mass spectrometry (GC-MS), and liquid chromatography–mass spectrometry (LC-MS). Multivariate analysis was applied to the data, and the findings were correlated with antioxidant activity and α-glucosidase inhibitory effects in vitro. Statistical analyses divided the 27 batches of MLs at different growth stages into three distinct groups. In vitro assays for antioxidant activity and α-glucosidase inhibition revealed that IC50values were highest at the Y23 stage, which corresponds to the ‘Frost Descends’ solar term. In summary, the results of this study indicate that MLs at different growth stages throughout the year can be categorized into three primary growth stages using traditional Chinese solar terms as reference points, based on the observed variations in metabolite content. Keywords:Morus albaL.;primary metabolites;secondary metabolites;different growth stages;antioxidant activity

Secondary metabolite profiles and bioactivities of red raspberry juice during fermentation with Wickerhamomyces anomalus

Yin Qin,You Luo,Shuyi Qiu,Qunying Zhang,Le Yang

Raspberry contains a broad array of valuable phytochemicals and therefore has positive effects on health. Fermentation can transform these components. The variations in the secondary metabolite profiles and bioactivities of red raspberry juice during Wickerhamomyces anomalus fermentation remain unknown. Herein, the effects of W. anomalus on the secondary metabolites, other constituents (total sugars, reducing sugars, and total acids) and bioactivities of raspberry juice were investigated. The fermentation significantly decreased the level of total sugars, reducing sugars, total acids and anthocyanins, but greatly increased the content of polyphenols and flavonoids. The antioxidant capacity and α-glucosidase inhibitory activity of raspberry juice were markedly enhanced after fermentation, whereas the cholesterol esterase inhibitory activity was reduced significantly. Moreover, 721 secondary metabolites were identified, wherein 437 secondary metabolites (240 upregulated, 197 downregulated) were selected as differential metabolites via targeted metabolomics. A total of 89 secondary metabolites were annotated in the Kyoto Encyclopedia of Genes and Genomes pathways, most of which were matched to biosynthesis of secondary metabolites and metabolic pathways. Phenolic acids were mainly transformed via phenylalanine metabolism and tyrosine metabolism. These results can provide reference for the development of functional raspberry beverages.

Designed Fabrication of Phloretin-Loaded Propylene Glycol Binary Ethosomes: Stability, Skin Permeability and Antioxidant Activity

Meng Zhang,Xue Zhuang,Siqi Li,Yansong Wang,Xiangyu Zhang,Jinlian Li,Dongmei Wu

Binary ethosome vesicles have been developed as flexible lipid vesicles for the enhanced physicochemical stability and skin delivery of drugs. This work aimed to prepare phloretin-loaded propylene glycol ethosomes (PHL-PGEs) to improve their stability, skin permeability and antioxidant activity. PHL-PGEs were prepared via the ethanol injection method and optimized using different weight ratios of ethanol to propylene glycol (PG). When the ethanol/PG mass ratio changed from 10:0 to 0:10, the encapsulation efficiency and stability of ethosomes increased. At a PHL concentration of 1mg/mL, the EE% was 89.42 ± 2.42 and the DL% was 4.21 ± 0.04, which exhibited their highest values. The encapsulation of the PHL in the PHL-PGEs was strengthened via XRD analysis and FTIR analysis. The results of the in vitro percutaneous permeability test demonstrated that the combined use of ethanol and PG exhibited a notable enhancement in skin permeability, and the skin retention of PHL-PGEs was 1.06 times that of PHL-ethosomes (PHL-Es) and 2.24 times that of the PHL solution. An in vitro antioxidant activity study indicated that solubility and antioxidant activity was potentiated via the nanoencapsulation of phloretin. Therefore, these results confirm the potential of this nanocarrier to enhance physicochemical stability, skin permeability and antioxidant activity. Keywords:binary ethosomes;propylene glycol;skin permeability;antioxidant activity

Pterostilbene binding to whey protein: Multi-spectroscopy and the antioxidant activity

Xiuping Liang,Long Chen,Zipei Zhang,Ruojie Zhang,David Julian McClements,Zhenlin Xu,Man Meng,Zhengyu Jin

In this study, Pterostilbene (Pte) was incorporated into whey protein isolate (WPI) to form a whey protein isolate-pterostilbene (WPI-Pte) complex. The interaction mechanism between these two components and its impact on the antioxidant performance of Pte were investigated through multispectral techniques and molecular docking. UV and fluorescence analyses revealed a distinct fluorescence static quenching mechanism of WPI by Pte, resulting in the formation of a complex with a 1:1 M ratio. Thermodynamic parameter assessments indicated that the interaction between Pte and WPI is a spontaneous non-covalent binding process, primarily driven by hydrophobic interactions, followed by hydrogen bonding. Molecular docking confirmed that Pte molecules become stably embedded within hydrophobic pockets formed by amino acid residues of the protein after interacting with protein and are surrounded by these residues. DPPH and ABTS free radical scavenging assays demonstrated that the binding of WPI and Pte enhances the antioxidant capacity by more than fourfold compared to free Pte. This study provides insights into the interaction, modification, and development of novel polyphenol antioxidant systems, thereby effectively harnessing its physiological benefits.

Qingqing Wang,Yinzi Chang,Qinjie Yuan,Fenghua Wu,Hao Hu,Jiaojiao Zhang,Zhiping He

This study establishes an enzymatic membrane-coupled production process for antioxidant peptides fromCarya cathayensiscake by comparing the effects of not feeding, water feeding, substrate feeding, and gradient dilution feeding supplementary material modes, to assess their impact on production efficiency. The optimal operational conditions were determined as follows: pH 10.5, temperature 50 °C, and enzyme-to-substrate ratio of 10% (w/w). The continuous production using the gradient dilution supplementary material mode resulted in Chinese pecan antioxidant peptides with 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging rate of 0.044 mg/mL, 2, 2-azinobis-(3-ethylbenzthiazoline-6-sulfonate) (ABTS) radical scavenging rate of 0.518 mg/mL, and ferrous ion chelating ability (IC50) of 0.252 mg/mL. Compared with traditional enzymatic hydrolysis processes, the gradient dilution supplementary material enzymatic membrane-coupled production process increased peptide yield, peptide production, and unit enzyme-peptide production by 14.36%, 11.35%, and 235.63%, respectively. This continuous production method facilitates scalability, enabling the production of high-yield and high-activityCarya cathayensiscake peptides, making better use of byproducts after oil extraction, and laying a solid foundation for the comprehensive development and deep processing of Chinese pecan, thus enhancing its economic value. Keywords:enzyme membrane reactor;Carya cathayensiscake;enzyme membrane coupling process;antioxidant peptides

Piezocatalytic strategy facilitates diabetic bone regeneration through high-performance anti-oxidative recycling

Jiwei Sun,Junyuan Zhang,Lina Yang,Cencen Zhang,Yifan Wang,Haoqi Lei,Keqi Wo,Wenjie Fan,Baoying Zhao,Jinyu Wang,Yunsong Shi,Zhiqiang Luo,Bin Su,Jinlin Song,Yingying Chu,Lili Chen

Anti-oxidative therapy for resistance of oxidative stress-induced damage has attracted substantial attention for bone regeneration under diabetic conditions. However, most anti-oxidants suffer from low efficiency due to their one-off reaction with oxidative products and rapid loss of activity. Herein, the piezoelectric nanocomposite with built-in electric field was fabricated as a cyclic anti-oxidative device for improved reactive oxygen species (ROS) scavenging and pro-osteogenic outcome. Under ultrasound (US) irradiation, the polydopamine (PDA) -functionalized generator with piezocatalytic effect exhibits high-performance recycling of redox catalytic reaction via continuous electron supplementation for cyclic group switch from quinone to phenol. The piezocatalytic method substantially boosts ROS elimination and reverses inflammatory status, contributing to strengthened osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs) and M2 polarization of macrophages. In vivo results further validated superior bone healing in rats with diabetes mellitus (DM). Our study therefore provides fundamental insights into the potential value of piezocatalytic effect-triggered cyclic anti-oxidative function for optimized osteogenesis under DM condition, and lays a basis for application of piezocatalytic effect in regenerative medicine.
