Cholesterol derivatives play a vital role in various fields, including medicine, biology, and smart materials. The Langmuir-Blodgett (LB) technology uses molecular self-assembly to create interfacially organized thin film materials with unique characteristics. This work primarily used two cholesterol derivatives (CCA and CCH) coupled with various dyes to create multilayer composite LB films. The SERS test determined the enhancement factor EF=1.07×10 5 for the CCA/ST composite film and EF=1.12×10 5 for the CCH/ST multilayer composite films, with relative standard deviation RSD=12.58% and 3.86%, respectively. The result demonstrates the superior signal responsiveness and uniformity of the resultant LB films, offering a novel option for active substrate materials in SERS detection of trace compounds. The photoelectric response signal of the CCA/dye and CCH/dye composite LB films was demonstrated to be sensitive by the photoelectric conversion test, and the films' superior reusability was demonstrated by the persistence of the same pattern over several recurring cycles. It offers a novel idea for investigating the use of cholesterol derivatives in optoelectronic devices in more detail.

Antibiotics are the primary drugs utilized to combat bacterial infections; however, their overuse and misuse have caused significant harm to human health. Rapid and efficient detection of antibiotics is crucial for food safety and environmental monitoring. Surface-enhanced Raman scattering (SERS) technology offers ultra-high sensitivity for molecular detection. In this study, we introduce a SERS substrate AuNRs-SH-BN (ASBN), which integrates semiconductor boron nitride with noble metal gold nanorods (AuNRs) for the trace detection of antibiotics. The prepared porous boron nitride features a large specific surface area with numerous pores and adsorption sites, facilitating the adsorption of antibiotic molecules. Coupling porous boron nitride with AuNRs generates numerous “hot spots,” thereby enhancing the Raman signal. The SERS activity of the ASBN substrate was evaluated using signaling molecules, demonstrating a high enhancement factor (7.13 × 10 6 ), excellent uniformity (RSD = 4.45%), and good stability (retaining 87.22% of the initial intensity after 28 days). The linear detection range for tetracycline hydrochloride is in the range 10 −11 –10 −4 M. Additionally, significant recoveries of 82.11%‒97.10% were achieved for tetracycline hydrochloride in real samples. Graphical

Background Biorhythmic played an important role in physical health. Biorhythmic disorders led to various health issue, including cardiovascular diseases and metabolic disorders. Early diagnosis of biorhythmic disorders was crucial in slowing the progression of disease. Therefore, it was important to develop technologies for detecting biorhythmic and their disorder markers. This study selected HMGB1(High mobility group box 1), BMAL1 (Brain and Muscle Aryl hydrocarbon receptor nuclear translocator-like 1), and MICU (mitochondrial calcium uptake 1) mRNA for detection. Results CRISPR/Cas12a/SERS (surface-enhanced Raman spectroscopy) ultrasensitive biosensing platform was developed for the detection of biorhythmic markers. The biosensor was partly an ultrabright Raman signal reporter (on) of a novel satellite structure formed by gold/silver bimetallic nanoparticles. This novel satellite structure utilizes the potential difference between gold and silver nanoparticles to promote the transfer of electrons from the core structure to the AuNPs and MBA (4-mercaptobenzoic acid) molecules, while enhancing the electromagnetic field distribution. Compared with the Raman signal of the satellite structure composed of Au/Au bimetallic, the Raman signal of the satellite structure in this study is increased by about 10 times. CRISPR/Cas12a recognized the target molecule and activated trans -cleavage activity, cleaving the DNA/RNA hairpin structure and releasing RNA 2∗ . RNA 2∗ caused the satellite structure to disperse, significantly attenuating the ultra-strong Raman signal (off), thereby enabling quantitative detection of the target nucleic acid. The LOD for three targets were of less than 1 fM. Significance In summary, the CRISPR/Cas12a/SERS biosensing platform enables mRNA detection without preamplification. This biosensing platform can detect target molecules at the fM level, achieving ultra-sensitive detection. In addition, the satellite structure exhibits long-term stability, providing a theoretical basis for the practical application of this biosensing platform.

This study presents the development of a multifunctional microfluidic chip designed for the rapid detection and inactivation of hazardous bacterial contaminants in complex environmental and industrial settings. The chip integrates nanozyme cascade colorimetric reactions, surface-enhanced Raman spectroscopy (SERS), and photothermal sterilization, enabling multimodal bacterial detection and decontamination. Constructed using Au@Fe/ZIF nanomaterials, the platform exhibits robust glucose oxidase (GOx)-like and peroxidase (HRP)-like activities, facilitating quantitative bacterial analysis through enzyme cascade reactions and highly sensitive species identification via SERS. For both Brucella melitensis and Bacillus cereus , the system achieves a detection limit as low as 10 CFU/mL both by SERS detection and enzyme cascade colorimetric analysis, with strong correlation (R 2 = 0.988) to standard plate counting. Additionally, Au@Fe/ZIF demonstrates high photothermal conversion efficiency (45.2 %), resulting in an inactivation rate exceeding 98 % under near-infrared irradiation. The platform's reusability and high detection accuracy make it a promising tool for monitoring bacterial hazards in contaminated water, industrial waste, and biothreat scenarios, offering a novel approach to biosafety and hazardous material management.

Surface-enhanced Raman scattering (SERS) with ultrahigh sensitivity has garnered significant attention for quantitative analysis and chemically specific detection. However, conventional SERS platforms, typically structured by depositing plasmonic micro/nanoparticles onto rigid substrates, face limitations in further advancement and applications. In this study, we present a novel method to prepare a flexible SERS film substrate composed of graphene overlayer (G) atop a self-assembled array of silver micropopcorns (Ag MPs) and a polymethyl methacrylate (PMMA) membrane. We show that adding an overlayer of graphene improves substantially overall SERS performance. This flexible G/Ag MPs/PMMA film substrate achieves a detection limit as low as 100 picomolar for rhodamine 6G (Rh6G) molecules and identifies mixtures containing Rh6G (10 −11 M) and crystal violet (CV, 10 −6 M) molecules. Furthermore, the flexible SERS structure enables quantitative detection of thiram, achieving a detection limit of 0.005 ppm, well below the allowable limit in food. This study provides a robust SERS platform that advances fundamental research and facilitates the development of practical applications in quantitative analysis, detection, and biological sensing. Combining graphene and Ag MPs on a flexible PMMA substrate offers a versatile and scalable approach to enhancing SERS performance, paving the way for future innovations.

Plasmonic nanostructures are a promising class of materials for solar-chemical conversion and trace substance detection. However, the low effective electron density, poor charge separation efficiency and the inability to tandem solar-chemical conversion and trace detection capabilities limit their further application. Here, we report a multicomponent plasmonic hybrid consisting of in situ reduced gold nanoparticles anchored at the hetero-interface of reduced graphene oxide/polyethyleneimine/graphitic carbon nitride electrostatically assembled layers as the plasmonic photocatalysts and surface-enhanced Raman scattering (SERS) hotspots. Experiments and theoretical calculations prove that the hybrid simultaneously optimizes three keys to photocatalysis: i) the generation of electrons via multiple pathways and the increase of the effective electron density, ii) the formation of a hetero-interface to lower the Schottky barrier and the synergistic plasma resonance to form an electric field to promote the charge separation, iii) the promotion of the substrate molecule adsorption and the reconfiguration of the gold nanoparticles. Thus, the hybrid achieves an H 2 O 2 production rate of 2.196 mmol g −1 h −1 in air and under acidic conditions. We further show that the complexes have synergistic effects in applications such as SERS detection of fruit and vegetable residues and photodegradation and antimicrobial preservation. This work may be useful for rationally designing plasmonic nanostructures and their application to photocatalysis in conjunction with SERS.

Chiral molecules are crucial in the field of optical devices, molecular recognition, and other novel functional materials due to their unique spatially asymmetric configuration and optical activity. In this study, a chiral molecule, Cholest-3-yl (E)-3-(4-carbamoylphenyl)acrylate (CCA), was combined with dyes containing large conjugated structures, tetramethylporphyrin tetrasulfonic acid (TPPS), and Nickel(II) phthalocyanine-tetrasulfonic acid tetrasodium salt (TsNiPc), and composite LB films of CCA/TPPS and CCA/TsNiPc were successfully prepared by using Langmuir-Blodgett (LB) technology. The circular dichroism (CD) test proved that the CCA/TPPS composite film had a strong CD signal at 300–400 nm, and the composite film showed chirality. This significant optical activity provides a new idea and option for the application of LB films in chiral sensors. In the Surface Enhanced Raman Spectroscopy (SERS) test, the CCA/TPPS composite film was sensitive to signal sensing, in which the enhancement factorEF= 2.28 × 105, indicating that a large number of effective signal response regions were formed on the surface of the film, and the relative standard deviation (RSD) = 12.08%, which demonstrated that the film had excellent uniformity and reproducibility. The high sensitivity and low signal fluctuation make the CCA/TPPS composite LB film a promising SERS substrate material.

Background Methamphetamine (MAMP) poses a significant public health and safety risk in communities. Its use causes behavioral changes in a person that can hinder their ability to undertake tasks such as driving and work. Therefore, the development of a rapid and reliable detection method for MAMP is critical for both employers and law enforcement agencies. Surface-enhanced Raman scattering (SERS) can reliably distinguish MAMP by identifying its unique spectral fingerprints. Hydrogels can act as scaffolds for plasmonic nanomaterials to enable effective SERS detection of a wide range of analytes. Results In this study, we investigated the fabrication of polyvinyl alcohol (PVA) hydrogels integrated with two-dimensional (2D) plate-like silver nanoplates (AgNPls) for ultrasensitive SERS analysis of MAMP. The resulting PVA hydrogel containing silver nanoprisms (AgNP-Hydrogel) was further modified through halide ion-induced etching and salting-out using the Hofmeister effect, significantly enhancing its SERS performance. The PVA matrix not only provides a scaffolding effect, preventing the aggregation of silver nanoparticles, but also enables the effective tuning of their morphologies. Its porous network structure facilitated efficient analyte diffusion and exchange. The salting-out process induces the AgNP-Hydrogel to shrink, generating "hot spots" that significantly enhance SERS sensitivity. By treating the AgNP-Hydrogel with KCl and subsequently salting it out using Na 2 SO 4 , an optimal SERS hydrogel (SERS-Hydrogel-I) was developed, capable of detecting Rhodamine 6G (R6G) at concentrations as low as 10 −12 M. Significance This promising SERS-Hydrogel-I was successfully applied to detection of MAMP, with a minimum detectable concentration of 0.01 ppm. The integration of PVA hydrogel and silver nanoplates, with the assistance of etching with halide ions and salting-out with sulfate ions, provides a convenient and effective platform for SERS-based detection, demonstrating strong potential for practical applications.

Surface-enhanced Raman scattering (SERS) has gained widespread attention as an effective method for trace molecular detection. Herein, a bowl-structured Ag NPs@Ag-PDMS substrate was developed via a multi-step assembly strategy, where silver nanoparticles (Ag NPs) were partially embedded into the bowl-structured surface of a polydimethylsiloxane (PDMS) matrix while additional Ag NPs were incorporated into the interior. This synergistic architecture significantly amplified the local electromagnetic field, as confirmed by experiments and finite element simulations. Using rhodamine 6G (R6G) as a probe and a fiber Raman spectrometer, the optimized substrate enabled detection at 5 nM with an enhancement factor of 1.22 × 10 6 . The substrate also exhibited good signal uniformity with a relative standard deviation of 6.99 % and maintained performance after five rinsing–reuse cycles. Furthermore, it successfully detected thiram residues on apple peels, showing a linear response down to 3 ng/cm 2 . These findings highlight the high sensitivity, reproducibility, and reusability of the substrate, indicating strong potential for real-world SERS applications in food inspection, environmental analysis, and public health screening.

Nitrobenzene is currently the most widely used explosive substance, and is known for its high toxicity and mutagenicity. It can cause severe environmental and water pollution, posing a risk to public health. Among various explosives analysis methods, surface-enhanced Raman spectroscopy (SERS) has the advantages of fast analysis speed, low detection cost, and easy operation, and has become one of the most promising analytical detection methods. Here, we present a portable and reliable sol-based SERS method for the detection of trace amounts of 2,4,6-trinitrotoluene (TNT) in different water bodies. The Meisenheimer complex formed by nitrobenzene and hydrazine hydrate can assemble on unmodified Au nanoparticles in a sol via Au–N bonds, enabling rapid detection of TNT in seawater, lake water, and tap water using a portable Raman spectrometer. Experimental results show that this SERS method can complete the detection within a few minutes and the detection sensitivity can reach 0.01 mg L−1, which is far lower than China's national standard of no more than 0.5 mg L−1. Furthermore, this method was also successfully applied to detect trace 2,4-dinitrotoluene (2,4-DNT) and picric acid (2,4,6-trinitrophenol) in water, demonstrating its strong applicability for on-site detection of nitrobenzene explosives.

Microcystin-LR (MC-LR), a potent cyanotoxin in freshwater, poses a risk of severe liver damage and other health issues, making its detection vital. However, the detection capabilities of conventional antibodies are constrained, which limited their use in immunoassays. In this work, we designed a new bifunctional nanobody, named A2.3-SBP (comprised of nanobody and streptavidin binding peptide), capable of binding with MC-LR and streptavidin. Based on A2.3-SBP and Fe 3 O 4 @Au-Pt nanozyme, we introduced an enzyme-free immunosensor that operated in microplate with colorimetric and surface-enhanced Raman scattering (SERS) detection modes. The dual-mode assay showed color changes and SERS intensity directly correlating to MC-LR concentrations with a range from 1.0 to 500 ng/mL and a limit of detection of 0.26 and 0.032 ng/mL, respectively. This strategy eliminated the need for complex enzymatic reactions and realized dual-signal detection of MC-LR in 96 water samples (0.03 μg/kg) within 30 min, suggesting its potential in drinking water detection.

In this study, the novel method combining molecularly imprinted polymers and surface-enhanced Raman spectroscopy for selectively extracting and detecting nitenpyram in fruits was proposed. The molecular imprinted polymers were prepared by precipitation polymerization method. The obtained imprinted polymers presented uniform spherical particles, and the average diameter (about 2 μm) was obviously larger than non-imprinted polymers. Dynamic and static adsorption experiments validated that imprinted polymers possessed fast adsorption rate (only 10 min), large adsorption capacity (5187.06 µg/g) and good specific recognition ability towards nitenpyram. Under optimum solid phase extraction conditions (80 mg dosage, pH at 5, H 2 O as sample solvent, 5 % methanol-H 2 O as washing solvent, 2 mL 10 % formic acid-ethyl acetate as elution solvent), the molecularly imprinted polymers coupled surface-enhanced Raman spectroscopy method showed good linear relationship (1–25 μg/g) with R 2 > 0.9736. The lower limits of detection (0.14–0.74 µg/g) in different fruits (pear, peach, apple, tomato) were achieved. The satisfactory spiked recovery (71.3–103.89 %) was also obtained. This study provided a rapid, sensitive, specific and practical method for analyzing nitenpyram based on synergistic effect of specific recognition of imprinted polymers and high sensitivity of surface-enhanced Raman spectroscopy. More importantly, it presented useful strategy for removing interference for SERS system, promoting SERS into actual application for rapid detection of trace pesticide in foods.
