Construction of lignin–metal polyphenol networks and their application in smart paper-based active packaging: Multifunctional materials with both antioxidant and UV-shielding functions

Lirong Lu,Jiatian Zhu,Jingpeng Zhou,Xi Cheng,Baojie Liu,Chengrong Qin,Chen Liang,Caoxing Huang,Shuangquan Yao

Facing the urgent need for sustainable and functional food packaging, developing high-performance materials from natural polymers is crucial. Herein, we propose a green strategy inspired by crustacean exoskeletons to construct lignin–metal polyphenol networks (L-MPNs) via proton-based ionic liquid pretreatment and metal-ion coordination. This forms a stable 3D network utilizing lignin's phenolic hydroxyl groups, which significantly enhances antioxidant activity, achieves >99% UV shielding, and exhibits 92.5% DPPH radical scavenging capacity. Applied to paper packaging, Ferric-ion-coordinated lignin-metal polyphenol networks (FL-MPNs, 100 g/m 2 ) impart excellent barrier and antioxidant properties alongside maintained mechanical strength, cutting water vapor transmission by ∼40%. In freshness tests, they reduced mass loss to 3.52% and pH change to 0.17, effectively inhibiting food deterioration. Thus, FL-MPNs advance lignin valorization for next-generation sustainable smart packaging to ensure safer, more stable food preservation.

Halting Mitochondrial Domino Effect in Acute Liver Injury: Polyphenol-Sr Nanodrugs Attenuate Cell Death and Sterile Inflammation by Combating ROS Burst and Calcium Overload

Min Liu,Shuya Wang,Ruishi Li,Weimin Qi,Tingli Xiong,Xiaojing Shi,Wensheng Chen,Feixiang Ding,Chenglong Wei,Junyan Liu,Niansheng Li,Qiong Huang,Zhaoqian Liu,Kelong Ai

Acute liver injury (ALI) is a significant clinical cause of liver failure, potentially occurring at any stage of liver disease and posing a considerable health burden. One prevalent model of ALI stems from acetaminophen (APAP) overdose, where a vicious cycle of “mitochondrial damage-inflammation amplification” and limited availability of small molecular drugs hinder current therapeutic approaches. Herein, a novel tannic acid (TA)-strontium (Sr) nanodrug (SrTA) is proposed. With liver-accumulating and mitochondria-targeting capabilities, SrTA effectively harnesses the broad-spectrum antioxidant properties of TA along with the calcium homeostasis-regulating function of Sr 2+ . And its therapeutic efficacy surpassed that of an equivalent dose of N-acetylcysteine (NAC). Mechanistically, SrTA directly scavenges mitochondrial reactive oxygen species (ROS), protects mitochondrial integrity, and alleviates endoplasmic reticulum (ER) stress and intracellular oxidative damage. Additionally, SrTA antagonizes calcium signaling, reduces the formation of mitochondria-ER contacts, and inhibits mitochondrial calcium overload. By safeguarding mitochondrial function and preventing the aberrant opening of the mitochondrial permeability transition pore (mPTP), SrTA significantly curtail hepatocyte death and mitigates mtDNA-induced sterile inflammation, effectively halting the injury cascade. In conclusion, this study presents a novel therapeutic strategy for ALI that targets mitochondria and synergistically regulates ROS bursts and calcium overload, achieving multifaceted therapeutic effects.

Long-term oxidative stability and tribological performance of tea polyphenol-stabilized ultra-high molecular weight polyethylene after 7 years of real-time shelf aging

Yi-Zi Wang,Shao-Peng Zhao,Han-Qiao Zhang,Tao Chen,Jiang-Yu Li,Peiqi Yu,Yu Han,Yue Ren,Chuntai Liu,Changyu Shen

Although antioxidants such as vitamin E (VE) and tea polyphenols have been employed to enhance the oxidation resistance of ultra-high molecular weight polyethylene (UHMWPE) for artificial joint applications, the long-term oxidative behavior of tea polyphenol-stabilized, crosslinked UHMWPE under real-time shelf aging remains insufficiently understood. In this study, the performance evolution of crosslinked UHMWPE containing VE, epigallocatechin gallate (EGCG), and lipid-soluble EGCG ( ls EGCG) was investigated after 7 years of real-time shelf storage. Notably, the EGCG-stabilized UHMWPE exhibited a markedly higher crosslink density (≥ 40% increase compared with the VE group) and superior tribological performance, with the coefficient of friction reduced by 16‒35%. Furthermore, it reduced the fraction of submicron wear particles (≤ 1 µm), lowering the potential risk of osteolytic inflammation. The enhanced stabilization efficacy of tea polyphenols, particularly EGCG, originates from their polyphenolic hydroxyl groups, which could effectively quench free radicals and preserve crosslink network during long-term storage. Overall, these findings demonstrate that tea polyphenols represent a durable and effective antioxidant strategy for UHMWPE, with significant potential to extend the clinical service life of joint implants.

Lutein-loaded Pickering high internal phase emulsions stabilized by protein-polyphenol-polysaccharide self-assembled particles: Interfacial behavior, in vitro/in vivo stability and release

Guangfan Qu,Yanxiang Gao,Shuguo Sun

QPI-TA-HMP particles formed a highly viscoelastic interfacial film. • Lutein-loaded Pickering HIPEs possessed controllable rheological properties. • Pickering HIPEs improved the encapsulation efficiency and bioavailability of lutein. • Pickering HIPEs enhanced the in vivo biodistribution of lutein. This study aimed to encapsulate lutein in high internal phase emulsions (HIPEs) stabilized by quinoa protein isolate (QPI), tannic acid (TA), and high-methoxy pectin (HMP) particles at varying concentrations to address its low delivery efficiency and bioavailability. High concentrations (3%–4%) of QPI-TA-HMP particles demonstrated strong interfacial adsorption, forming thick viscoelastic films around oil droplets. These interfacial properties imparted controllable rheological behaviors, textural characteristics, and stable 3D-printing scaffolds to the lutein-loaded HIPEs, achieving an encapsulation efficiency of 81.65 ± 2.36%. In vitro tests indicated that HIPEs enhanced lutein's resistance to storage, heat, and UV exposure while facilitating sustained intestinal release, resulting in a lutein bioaccessibility of 43.73 ± 1.44%. In vivo experiments further demonstrated that the HIPEs delivery system maintained high lutein concentrations in the small intestine, cecum, and colon, thereby significantly enhancing lutein accumulation in systemic circulation. These findings provide new insights into enhancing lutein's stability, delivery performance, and bioavailability. Download: Download high-res image (202KB) Download: Download full-size image

en This link goes to a English section zh This link goes to a Chinese section Cisplatin-related acute kidney injury (CP-AKI) is a serious complication of cisplatin (CP) treated patients with cancer with high morbidity and mortality rates. It is directly related to the abnormal accumulation of CP in renal tubular epithelial cells, causing metabolic toxicity, DNA damage, and oxidative stress. Herein, we developed a polyphenol nanozyme (SrEG) by the coordination of strontium ions (Sr 2+ ) and epigallocatechin gallate. It was demonstrated that SrEG, combined with NIR irradiation, exhibited excellent capabilities of scavenging various types of reactive oxygen species (ROS). It was also determined that SrEG + NIR efficiently eliminated intracellular ROS levels, inhibited inflammation related cytokines expression, and induced macrophage M2 directional polarization, thus preventing cellular damage. By intravenous (IV) administration, SrEG was efficiently accumulated in the kidney and gradually degraded. Accordingly, the administration of SrEG + NIR offered the perfect therapeutic effects in CP-AKI rats by inhibiting the PI3K/AKT pathway, thus accelerating kidney function repair. Overall, the strategy of SrEG + NIR showed substantial clinical promise as a potential therapeutic strategy in ROS-related diseases. 摘要 en This link goes to a English section zh This link goes to a Chinese section 顺铂相关的急性肾损伤(CP-AKI)是癌症患者进行顺铂(CP)治疗时的严重并发症,具有较高的致病率和死亡率。

Effects of Tea Polyphenol and Its Combination with Other Antioxidants Added during the Extraction Process on Oxidative Stability of Antarctic Krill (Euphausia superba) Oil

Ziwei Wang,Fujun Liu,Ying Luo,Xiangbo Zeng,Xuechen Pei,Guanhua Zhao,Min Zhang,Dayong Zhou,Fawen Yin

Antarctic krill (Euphausia superba) oil contains high levels of marine omega-3 long-chain polyunsaturated fatty acids (n-3 LC-PUFA), including eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). In industrial production, krill oil is usually extracted from krill meals by using ethanol as a solvent. Water in the krill meal can be easily extracted by using ethanol as an extraction solvent. During the extraction process, the EPA and DHA are more easily oxidized and degraded when water exists in the ethanol extract of krill oil. Based on the analysis of peroxide value (POV), thiobarbituric acid-reactive substances (TBARS), fatty acid composition, and lipid class composition, the present study indicated that the composite antioxidants (TP-TPP) consist of tea polyphenol (TP) and tea polyphenol palmitate (TPP) had an excellent antioxidant effect. By contrast, adding TP-TPP into ethanol solvent during the extraction process is more effective than adding TP-TPP into krill oil after the extraction process. Keywords:antarctic krill;oil;antioxidant;tea polyphenol;composite antioxidants;ethanol

Metal–Polyphenol Coordination at the Aqueous Contra-diffusion “Interface”: A Green Way to High-Performance Iron(III)/Tannic Acid Thin-Film-Composite Nanofiltration Membranes

Yi-Xin Wang,Cheng-Ye Zhu,Feng Lu,Zi-Feng Yu,Hao-Cheng Yang,Ming Xue,Zhi-Kang Xu

Thin-film-composite (TFC) nanofiltration membranes have found wide uses in environment remediation and industrial separation. There is a growing trend to avoid the use of organic solvents and toxic chemicals during membrane fabrication. Therefore, the aqueous fabrication of TFC membranes receives considerable interest as a green and sustainable process. However, it remains challenging to construct a defect-free and ultrathin film in a homogeneous aqueous phase without the assistance of an interface. The contra-diffusion process provides a special “interface” to confine the film formation within a narrow space by regulating the competition between precursor diffusion and interfacial reactions. Herein, Fe3+/tannic acid (TA) TFC membranes were fabricated by a contra-diffusion process. The effects of fabrication parameters on the Fe3+/TA TFC membrane microstructure and performance were also investigated. The negatively charged membrane performs a competitive Na2SO4 rejection of 95.6% with a permeation flux of 44.3 L m–2 h–1 under 0.6 MPa as well as more than 99.5% rejection to several anionic dyes. The as-prepared membranes perform superior nanofiltration performance compared to other reported Fe3+/TA-based membranes, owing to the thin and defect-free selective layers by self-regulation. Moreover, the membranes exhibit stable rejection during a long-term nanofiltration test.

In-situ coordination assembly of polyphenol with cage-like Prussian blue as an ecofriendly nanocarrier for site-specific pesticide delivery and sustained pest control

Lihong Zhang,Min Fang,Chunlin Yang,Fengrui Zhang,Zaiyu Lu,Haiqun Cao

In sustainable agricultural production, an efficient and environmental-friendly nanoplatform for precise pesticide delivery is considerably desirable to enhance efficacy and minimize ecological side effects. In this study, a pH-responsive nanohybrid named EB@HMPB@TA was designed using hollow mesoporous Prussian blue (HMPB) coordinated with tannin (TA) for targeted delivery of emamectin benzoate (EB). Results demonstrated that the cage-like HMPB nanoparticles possessed a remarkable loading capacity for EB, reaching up to 63.7%. Through a coordination assembly process, an in-situ Fe 3+ -chelated TA coating was formed on the surface of EB@HMPB to obtain EB@HMPB@TA. Notably, EB@HMPB@TA appreciably improved the foliar affinity of EB. Moreover, under UV-light irradiation, the degradation rate of EB loaded in the HMPB@TA nanocarriers decreased by 47% compared to EB alone, thus reducing the loss of EB. Additionally, the release ratio of EB@HMPB@TA at pH = 8.5 was 9.47-fold higher than that at neutral pH over 48 h. This pH-dependent release feature held great potential for targeting lepidopteran pests, which own a unique alkaline gut environment. Importantly, compared to commercial EB suspension concentrate, EB@HMPB@TA exhibited potent insecticidal activity against three target pests. Safety evaluations conducted on both HK-2 cells in vitro and maize crops demonstrated the relative safety of EB@HMPB@TA. Consequently, EB@HMPB@TA offered a new approach for accurate pesticide delivery, facilitating efficient and eco-friendly pest control practices and holding tremendous potential for sustainable agricultural production, which aligns with the principles of cleaner production in agriculture.

Inhibitory effect of bound polyphenol from foxtail millet bran on miR-149 methylation increases the chemosensitivity of human colorectal cancer HCT-8/Fu cells

Shan Shuhua,Lu Yang,Zhang Xiaoli,Shi Jiangying,Li Hanqing,Li Zhuoyu

Nature polyphenols widely present in plants and foods are promising candidates in cancer chemotherapy. Emerging evidence has shown that plant polyphenols regulate the expression of miRNAs to exert the anti-Multidrug resistance (MDR) activity, which partly attributes to their regulation on miRNAs methylation. Our previous study found that bound polyphenol from foxtail millet bran (BPIS) had potential as an anti-MDR agent for colorectal cancer (CRC), but its mechanism remains unclear. The present findings demonstrated that BPIS upregulated the expression of miR-149 by reducing the methylation of its CpG islands, which subsequently induced the cell cycle arrest in G2/M phase, resulting in enhancing the chemo-sensitivity of HCT-8/Fu cells. Mechanically, BPIS and its active components (FA and p-CA) reduced miR-149 methylation by inhibiting the expression levels of DNA methyltransferases, promoting a remarkable increase of miR-149 expression. Further, the increased miR-149 induced cell cycle arrest in G 2 /M phase by inhibiting the expression of Akt, Cyclin B1 and CDK1, thus increasing the chemosensitivity of HCT-8/Fu cells. Additionally, a strong inducer of DNA de-methylation (5-aza-dc) treatment markedly increased the chemosensitivity of CRC through elevating miR-149 expression, which indicates the hypermethylation of miR-149 may be the key cause of drug resistance in CRC. The study indicates that the enhanced chemosensitivity of BPIS on CRC is mainly attributed to the increase of miR-149 expression induced by methylation inhibition.

An Amphiphilic Conjugate Approach toward the Design and Synthesis of Betulinic Acid–Polyphenol Conjugates as Inhibitors of the HIV-1 gp41 Fusion Core Formation

Yan Liu,Zhuofeng Ke,Kwok Yiu Wu,Shuwen Liu,Wen-Hua Chen,Shibo Jiang,Zhi-Hong Jiang

Graphical Stopping HIV in its tracks! A series of betulinic acid–polyphenol conjugates were synthesized, with the aid of rational design by docking simulation, as novel HIV-1 fusion/entry inhibitors targeting gp41. These amphiphilic conjugates cooperatively interact with hydrophobic and hydrogen-bonding areas in the groove of the gp41 NHR trimeric coiled coil, demonstrating a promising strategy for the discovery of HIV-1 fusion/entry inhibitor binding to these two crucial pockets. Exploration of potent inhibitors of the HIV-1 gp41 fusion core formation is a promising strategy to discover small-molecule HIV-1 entry inhibitors for the treatment of HIV-1 infection. In this paper, a series of novel betulinic acid–polyphenol conjugates was designed, guided by molecular modeling of the binding of betulinic acid (BA) and phenolic galloyl/caffeoyl groups in the groove on the gp41 N-terminal heptad repeat (NHR) trimeric coiled coil. These conjugates were synthesized via conjugation of galloyl and caffeoyl groups with BA at the C-28 position. Their inhibitory activities of HIV gp41 six-helix bundle (6-HB) formation between the NHR peptide N36 and the C-terminal heptad repeat (CHR) peptide C34 were evaluated with size-exclusion HPLC. Conjugates bearing a galloyl group were found to exhibit four to sixfold higher inhibitory activities than that of parent compound BA, suggesting that they may be exploitable as HIV-1 fusion/entry inhibitors targeting gp41. The docking study on BA and its derivatives suggests that hydrophobic and hydrogen-bonding pockets exist in the groove of the gp41 NHR trimeric coiled coil and that a potent inhibitor should have amphiphilic structures to cooperatively interact with both pockets. This possibility was explored by incorporating both lipophilic and hydrophilic groups into the conjugates in a well-defined orientation to bind with both pockets in the gp41 NHR-trimer.

Tough and adhesive conductive hydrogels with fast gelation from a polyphenol–aluminium ion dual self-catalysis system for wearable strain sensors and triboelectric nanogenerators

Maolin Yu,Yuecong Luo,Qiannian Yang,Tengfei Duan,Zengmin Tang,Lijian Xu,Na Li,Jianxiong Xu

Hydrogels have been widely used as flexible electrodes for the construction of strain sensors and triboelectric nanogenerators (TENGs) with high performance owing to their attractive flexibility and conductivity. However, traditional fabrication methods of hydrogels involve time-consuming synthesis and/or use of external stimuli (i.e., heat and light). Herein, a tough and adhesive conductive double network hydrogel (PVA/PHEAA–TA–Al3+ gel) was prepared via rapid in situ room temperature gelation processes (25 °C, 215 s) in a tannic acid–aluminium ion (TA–Al3+) dual self-catalysis system. This involved the collaborative use of TA–Al3+ to induce the decomposition of ammonium persulfate (APS), which generated abundant free radicals to trigger the polymerization of the HEAA monomer within a polyvinyl alcohol/N-(2-hydroxyethyl)acrylamide/tannic acid (PVA/HEAA/TA) aqueous solution. The obtained hydrogel showed excellent mechanical properties (tensile stress/strain of 240 kPa/920%), adhesion, and self-healing ability. Benefitting from the ultra-wide sensing range (1–600%), high sensing sensitivity (GF = 2.7) and long-term stability (500 cycles), the PVA/PHEAA–TA–Al3+ gel was used to construct a strain sensor, which can accurately identify and distinguish the changes in human expression and joint movement. Furthermore, the PVA/PHEAA–TA–Al3+ gel was used to fabricate TENGs (named PT-TENGs). PT-TENGs with an area of 2 × 2 cm2 exhibited attractive electrical output properties (VOC = 109 V, ISC = 1.3 μA, and QSC = 35 nC at a fixed frequency of 2.0 Hz), which can power 22 LED arrays. This TA–Al3+ dual self-catalysis system is expected to provide a new way for the fabrication of tough and adhesive conductive hydrogels toward health monitoring sensors and energy supply.

Mechanism and Functional Activities of Dietary Fiber-Bound Polyphenol Released from Pomegranate Pomace in Mixed Solid-State Fermentation with Aspergillus niger and Rhizopus oryzae

Changding Gao,Mei Yang,Yihan Liu,Yuhan Wang,Ligong Zhai,Jinlong Zhao,Yijun Liu,Baoshi Wang

The insoluble dietary fiber (IDF) of pomegranate peel residue was treated by mixed solid-state fermentation (SSF) of Aspergillus niger and Rhizopus oryzae. The content of bound polyphenols (BP) reached a maximum of 26.50 mg GAE/g DW after 5 days of fermentation, which was 3.22-fold higher than that of the unfermented group. The BP release during fermentation was strongly correlated with the activities of CMCase, xylanase and β-glucosidase. A total of 35 polyphenolic compounds and catabolic metabolites were identified, and potential biotransformation pathways were proposed. Structural characterization of IDF before and after fermentation via SEM, XRD, and TGA revealed that the degradation of cellulose and disruption of hemicellulose and hemicellulose–lignin complexes played a key role in BP release. Moreover, the postfermentation BP in vitro showed enhanced antioxidant capacity and pronounced inhibition of α-amylase/α-glucosidase. Collectively, the mixed SSF significantly promote the BP release, facilitating pomegranate peel valorization.
