Crystal-Seeded Growth of pH-Responsive Metal–Organic Frameworks for Enhancing Encapsulation, Stability, and Bioactivity of Hydrophobicity Compounds

Zexun Liu,Qiao Wu,Jie He,Frank Vriesekoop,Hao Liang

Zeolitic imidazolate framework-L (ZIF-L) could effectively improve the stability, controlled release, and anticancer activity of natural hydrophobicity drugs in drug delivery systems (DDSs). A simple and universal strategy was developed to prepare the curcumin-loaded ZIF-L (CCM@ZIF-L) by the antisolvent coprecipitation method, which was different from the traditional approaches. The microcrystal molecules of curcumin were used as the core of ZIF-L growth to form CCM@ZIF-L, which has a very high drug encapsulation efficiency of 98.21% and a regular leaf or cruciate flower-like structure. The formation of CCM@ZIF-L with a distinct composite structure was supported by scanning electron microscopy, transmission electron microscopy, Fourier-transform infrared, powder X-ray diffraction, and zeta-potential. Because of the protective effect of ZIF-L, CCM@ZIF-L exhibited excellent stability and about a 5-fold increase in temperature stability over free curcumin. CCM@ZIF-L exhibited controlled drug release behavior in simulated in vitro tumor microenvironments (almost 81.2% drug release over a period of 72 h). Furthermore, confocal laser scanning microscopy results and cytotoxicity experiments confirmed that the encapsulated curcumin showed a significant improvement in cellular uptake and anticancer activity against A549 cancer cells. Moreover, the curcumin encapsulated in ZIF-L exhibited remarkable cellular antioxidant activity based on MGC-803 cell models. This work presents a novel approach to solve the drug loading problem by employing ZIF-L and exhibits enormous potential of ZIF-L as an effective DDS in cancer treatments.

Phloretin as both a substrate and inhibitor of tyrosinase: Inhibitory activity and mechanism

Jianmin Chen,Qinglian Li,Yaling Ye,Ziyao Huang,Zhipeng Ruan,Nan Jin

Tyrosinase is the rate-limiting enzyme for controlling the production of melanin in the human body, and overproduction of melanin can lead to a variety of skin disorders. In this paper, the inhibitory kinetics of phloretin on tyrosinase and their binding mechanism were determined using spectroscopy, molecular docking, antioxidant assays and chromatography. The spectroscopic results indicate that phloretin reversibly inhibits tyrosinase in a mix-type manner through a multiphase kinetic process with the IC 50 of 169.36 μmol/L. It is shown that phloretin has a strong ability to quench the intrinsic fluorescence of tyrosinase mainly through a static quenching procedure, suggesting that a stable phloretin-tyrosinase complex is generated. Molecular docking results suggest that the dominant conformation of phloretin binds to the gate of the active site of tyrosinase. Moreover, the antioxidant assays demonstrate that phloretin has powerful antioxidant capacity and has the ability to reduce o -dopaquinone to l -dopa just like ascorbic acid . Interestingly, the results of spectroscopy and chromatography indicate that phloretin is a substrate of tyrosinase but also an inhibitor. The possible inhibitory mechanism is proposed, which will be helpful to design and search for tyrosinase inhibitors.

Pomegranate seed oil stabilized with ovalbumin glycated by inulin: Physicochemical stability and oxidative stability

Bing Hu,Kangping Wang,Lingyu Han,Bin Zhou,Jixin Yang,Shugang Li

Pomegranate seed oil is rich of conjugated fatty acids which are highly appealing for a variety of applications in food industry. In this research, ovalbumin (OVA) and ovalbumin-inulin glycoconjugates with different Maillard reaction times were used to stabilize pomegranate seed oil emulsions and their impact on physicochemical stability and oxidative stability of the products was investigated. The OVA-inulin glycoconjugate produced on 10th day of Maillard reaction has exhibited significantly higher conjugation efficiency, lower surface hydrophobicity and lower surface tension than other glycoconjugates . The secondary conformation of OVA and conjugates determined by far-UV circular dichroism spectroscopy has remarkably changed. The reduction in intensity of Trp-fluorescence observed in glycated proteins with inulin indicated that the glycation affected partially the side chains of protein in tertiary structure through the Maillard reaction without great disruption of native structure. The emulsion stabilized by OVA-inulin glycoconjugate obtained by 10 days Maillard reaction has shown the best physicochemical stability. Compared with the OVA emulsion, the oxidative stability of the glycated OVA emulsion system was significantly improved (p < 0.05). Fatty acid profile results also confirmed that OVA-inulin glycoconjugates were able to prevent the pomegranate seed oil from oxidation . It is suggested that the inulin attached to OVA by glycation played a vital role in physicochemical stability and oxidative stability of pomegranate seed oil emulsions.

Effects of sulfated, phosphorylated and carboxymethylated modifications on the antioxidant activities in-vitro of polysaccharides sequentially extracted from Amana edulis

Yu-Yao Cao,Yu-Hang Ji,Ai-Mei Liao,Ji-Hong Huang,Kiran Thakur,Xiao-Li Li,Fei Hu,Jian-Guo Zhang,Zhao-Jun Wei

In this study, four sequentially extracted polysaccharides (AEPs) from Amana edulis were modified by sulphation, phosphorylation, and carboxylation modifications (S-AEPs, P-AEPs, C-AEPs), and compared for their anti-oxidant activities. After modification, sugar and protein contents were decreased and uronic acid content was increased in comparison to native AEPs. UV absorption showed similar maximum absorption peaks of modified derivatives which indicated their homogeneous nature. FTIR spectra confirmed the conversion of hydroxyl groups to O S, COO, and P OH bonds, respectively. The phosphorylated derivatives (P-AEPs) displayed the highest DPPH, hydroxyl radical, and ferrous ions radical scavenging abilities. Sulfated polysaccharides (S-AEPs) were observed with high reducing ability. The C-AEPs maintained the stable antioxidant properties after carboxylation modification. Our results indicated that the chemical modification of different polysaccharide components has significantly affected their antioxidant potential for their use in food industry and human health.

Structural characterization and antioxidant activity of polysaccharide from four auriculariales

Yue Su,Liang Li

The structural characterization and antioxidant activity of four Auricularia polysaccharides (A.cornea ACP)、A.auricula(AAP)、A.polytricha (APP) and M.fungus(MFP)) were studied in this paper. The results shown: polysaccharides of four Auricularia were mainly composed of mannose and galactose, all polysaccharides contained uronic acid and pyran ring structure with spectroscopy and NMR analysis. There was a significant difference in the total antioxidant capacity and APP was significantly higher than the other polysaccharides. The ability of APP to scavenge DPPH radicals and hydroxyl radicals was significantly higher than that of other polysaccharides, respectively. The molecular weight was significantly positively correlated with DPPH radicals, superoxide anion radicals and hydroxyl radicals. Total antioxidant capacity was significantly negatively correlated with fucose and galactose. The result indicated that fucose and galactose jointly determine total antioxidant capacity. The polysaccharide from four Auricularia had good oxidation resistance and could be used as natural antioxidants.

Construction of cellulose-based Pickering stabilizer as a novel interfacial antioxidant: A bioinspired oxygen protection strategy

Qi Li,Pan Chen,Yan Li,Bin Li,Shilin Liu

Oxygen protection/isolation is imperative to prevent the lipid oxidation since oxygen molecule is an ultimate quencher in photon conversion process. Inspired by the structural buildup of seeds from oil crops, a sustainable solid particle stabilizer with novel antioxidant activity was prepared by using cellulose and polyphenol. In this work, bacterial cellulose (BC) nanofibrils modified by tea polyphenols (TPs) was prepared and used as Pickering emulsifier for the O/W emulsion. BC nanofibirls exhibited excellent adsorption capacity up to 55 μg/mg, and the adsorption kinetics between BC and TPs were further investigated. After modification, the interfacial diffusion rate constant of BC was significantly increased to from 0.43 to 1.21 mN m −1  s −0.5 . Moreover, the obtained O/W interfacial modulus of the dilatational elasticity was increased from 58 to 130 mN/m. Furthermore, the emulsions exhibited excellent free-radical scavenging activity at oil-water interface, suggesting a potential application in usage to extend the lifespan of the food containing polyunsaturated fats.

Effect of Methyl Jasmonate on Phenolic Accumulation in Wounded Broccoli

Yuge Guan,Wenzhong Hu,Aili Jiang,Yongping Xu,Rengaowa Sa,Ke Feng,Manru Zhao,Jiaoxue Yu,Yaru Ji,Mengyang Hou,Xiaozhe Yang

In order to find an efficient way for broccoli to increase the phenolic content, this study intended primarily to elucidate the effect of methyl jasmonate (MeJA) treatment on the phenolic accumulation in broccoli. The optimum concentration of MeJA was studied first, and 10 μM MeJA was chosen as the most effective concentration to improve the phenolic content in wounded broccoli. Furthermore, in order to elucidate the effect of methyl jasmonate (MeJA) treatment on phenolic biosynthesis in broccoli, the key enzyme activities of phenylpropanoid metabolism, the total phenolic content (TPC), individual phenolic compounds (PC), antioxidant activity (AOX) and antioxidant metabolism-associated enzyme activities were investigated. Results show that MeJA treatment stimulated phenylalanine ammonia-lyase (PAL), cinnamate 4-hydroxylase (C4H), and 4-coumarin coenzyme A ligase (4CL) enzymes activities in phenylpropanoid metabolism, and inhibited the activity of polyphenol oxidase (PPO), and further accelerated the accumulation of the wound-induced rutin, caffeic acid, and cinnamic acid accumulation, which contributed to the result of the total phenolic content increasing by 34.8% and ferric reducing antioxidant power increasing by 154.9% in broccoli. These results demonstrate that MeJA in combination with wounding stress can induce phenylpropanoid metabolism for the wound-induced phenolic accumulation in broccoli. Keywords:wounded broccoli;methyl jasmonate;phenolic accumulation;phenylpropanoid metabolism;antioxidant capacity

Structural characterization and antioxidant activity of a new polysaccharide from Bletilla striata fibrous roots

Ziyan Chen,Yan Zhao,Mengke Zhang,Xiufang Yang,Pengxiang Yue,Dikang Tang,Xinlin Wei

A new polysaccharide (pFSP) was first isolated from the originally discarded fibrous roots part of Chinese traditional herb, Bletilla striata. pFSP was composed of D-glucose, D-galactose and D-mannose in a molar ratio of 1: 2.03: 3.45 with molecular weight of 9.1 × 10 4 Da. It could effectively scavenge DPPH and superoxide radicals with inhibition rate of 64.47% and 72.27% at 5.0 mg/ml, higher than that of polysaccharide from Bletilla striata tuber. Structural investigations of the periodate oxidation studies and Smith-degradation as well as the FT-IR spectroscopy were performed, and combined with 1D and 2D NMR spectroscopy, the repeating unit of pFSP contained (1→4)-linked-α-D-Glcp, (1→4)-linked-β-D-Manp and (1→3,6)-linked-β-D-Manp units, together with the branches of (1→6)-linked-β-D-Galp and terminated with (1→)-linked-β-D-Manp residue.

Natural Small Biological Molecule Based Supramolecular Bioadhesives with Innate Photothermal Antibacterial Capability for Nonpressing Hemostasis and Effective Wound Healing

Xiang Ke,Shuxian Tang,Hao Wang,Yusong Cai,Zhiyun Dong,Mingjing Li,Jiaojiao Yang,Xinyuan Xu,Jun Luo,Jianshu Li

Bioadhesives with immediate wound closure, efficient hemostasis, and antibacterial properties that can well integrate with tissue are urgently needed in wound management. Natural small biological molecule based bioadhesives hold great promise for manipulating wound healing by taking advantage of integrated functionalities, synthetic simplification, and accuracy, cost efficiency and biosafety. Herein, a natural small biological molecule based bioadhesive, composed of natural small biological molecules (α-lipoic acid and tannic acid) and a small amount of ferric chloride, was prepared via an extremely simple and green route for wound management. In this system, covalent and noncovalent interactions between each component resulted in the self-healing supramolecular bioadhesive. It possessed appropriate wet-tissue adhesion, efficient nonpressing hemostasis and free radical scavenging abilities. More importantly, the interaction between tannic acid and Fe3+ endowed the bioadhesive with innate and steady photothermal activity, which showed excellent photothermal bactericidal activity to both E. coli and S. aureus. The bioadhesive promoted wound healing for linear and circular wounds in vivo, especially for infectious wounds under near-infrared (NIR) irradiation. This bioadhesive will have promising value as a safe and effective antimicrobial adhesive for infectious wound management.

A high-protein retained PES hemodialysis membrane with tannic acid as a multifunctional modifier

Qianyu Wei,Shuman Feng,Zezhen Zhang,Lulu Liu,Lili Wu

A high protein retention polyethersulfone (PES) membrane was prepared by nonsolvent-induced phase separation and surface coating , which exhibited enhanced hemocompatibility and antioxidant stress performance. The cross-linked network was constructed by tannic acid (TA) and alpha-lipoic acid (α-LA) on the surface of the membrane, which controlled the pores to a reasonable size. The enrichment of heparin-like groups on the membrane surface, implemented by “hydrophobic interaction” and “click reaction”, confers anticoagulant properties; the presence of a large number of phenolic hydroxyl groups from TA and the introduction of α-LA allows the modified membranes to intervene in oxidative stress . The hemocompatibility characterizations included plasma recalcification time (PRT), activated partial thromboplastin time (APTT), prothrombin time (PT), thrombin time (TT) and hemolysis rate (HR). Additionally, the DPPH ABTS radical scavenging capacity was tested to evaluate the antioxidant performance. The results show that the modified membrane presents an outstanding protein retention rate (99.3%) along with permeability. In addition, the PRT is prolonged to 341.7 s, and the DPPH• scavenging ability reaches 0.74 µmol•cm -2 . The membranes can be easily prepared and present excellent comprehensive performance. This work provides a simple and facile strategy for the fabrication of hemodialysis membranes with controllable pore sizes.

Oxidative Stress Amelioration of Novel Peptides Extracted from Enzymatic Hydrolysates of Chinese Pecan Cake

Jiaojiao Zhang,Shaozhen Wu,Qingqing Wang,Qinjie Yuan,Yane Li,Patricia Reboredo-Rodríguez,Alfonso Varela-López,Zhiping He,Fenghua Wu,Hao Hu,Xingquan Liu

Pecan (Carya cathayensis) is an important economic crop, and its hydrolyzed peptides have been evidenced to reduce the effect of oxidative stress due to their antioxidant capacity. Hence, the protocols of ultrafiltration and gel filtration chromatography were established to obtain bioactive peptides from by-products ofC. cathayensis(pecan cake). As measured by DPPH/ABTS radical scavenging, the peptides with less molecular weight (MW) possess higher antioxidant capacity. PCPH-III (MW < 3 kDa) presented higher radical scavenging capacity than PCPH-II (3 kDa < MW < 10 kDa) and PCPH-I (MW > 10 kDa) measured by DPPH (IC50: 111.0 μg/ mL) and measured by ABTs (IC50: 402.9 μg/mL). The secondary structure and amino acid composition varied by their MW, in which PCPH-II contained more α-helices (26.71%) and β-sheets (36.96%), PCPH-III contained higher ratios of β-turns (36.87%), while the composition of different secondary of PCPH-I was even 25 ± 5.76%. The variation trend of α-helix and random experienced slightly varied from PCPH-I to PCPH-II, while significantly decreased from PCPH-II to PCPH-III. The increasing antioxidant capacity is followed by the content of proline, and PCPH-III had the highest composition (8.03%). With regard to the six peptides identified by LC-MS/MS, two of them (VYGYADK and VLFSNY) showed stronger antioxidant capacity than others. In silico molecular docking demonstrated their combining abilities with a transcription factor Kelch-like ECH-associated protein 1 (Keap1) and speculated that they inhibit oxidative stress through activating the Keap1-Nrf2-ARE pathway. Meanwhile, increased activity of SOD and CAT—antioxidant markers—were found in H2O2-induced cells. The residue of tyrosine was demonstrated to contribute the most antioxidant capacity of VYGYADK and its position affected less. This study provided a novel peptide screening and by-product utilization process that can be applied in natural product developments. Keywords:Chinese pecan by-products;antioxidant peptides;proline content;molecular docking;Keap1-Nrf2-ARE pathway

Structural Characterization, Antioxidant and Antitumor Activities of the Two Novel Exopolysaccharides Produced by Debaryomyces hansenii DH-1

Yajing Yang,Guoqiang Chen,Xiaoqi Zhao,Xiaohe Cao,Lei Wang,Jingjiu Mu,Fenghui Qi,Lijuan Liu,Haibo Zhang

Exopolysaccharides produced by edible microorganisms exhibit excellent constructive physicochemical and significant biological activity, which provide advantages for the food or pharmaceutical industries. Two novel exopolysaccharides produced byDebaryomyces hanseniiDH-1 were characterized, named S1 and S2, respectively. S1, with a molecular weight of 34.594 kDa, primarily consisted of mannose and glucose in a molar ratio of 12.19:1.00, which contained a backbone fragment of α-D-Manp-(1→4)-α-D-Manp-(1→2)-α-D-Glcp-(1→3)-α-D-Manp-(1→3)-β-D-Glcp-(1→4)-β-D-Manp-(1→. S2, with a molecular weight of 24.657 kDa, was mainly composed of mannose and galactose in a molar ratio of 4.00:1.00, which had a backbone fragment of α-D-Manp-(1→6)-β-D-Manp-(1→2)-α-D-Manp-(1→4)-α-D-Galp-(1→3)-β-D-Manp-(1→6)-α-D-Manp-(1→. Both S1 and S2 exhibited good thermal stability and potent hydroxyl radical scavenging activity, with ~98%. Moreover, S1 possessed an additional strong iron-reducing capacity. In vitro antitumor assays showed that S1 and S2 significantly inhibited the proliferation of Hela, HepG2, and PC-9 cancer cells. Moreover, PC-9 was more sensitive to S1 compared with S2. The above results indicate that S1 and S2 have great potential to be utilized as natural antioxidants and candidates for cancer treatment in the food and pharmaceutical industries. Keywords:Debaryomyces hansenii;exopolysaccharides;structure analysis;antioxidant activity;antitumor activity;thermal property
