Earlier studies have shown that selenomethionine (SM) supplements in broiler breeders had higher deposition in eggs, further reduced the mortality of chicken embryos, and exerted a stronger antioxidant ability in offspring than sodium selenite (SS). Since previous studies also confirmed that Se deposition in eggs was positively correlated with maternal supplementation, this study aimed to directly investigate the antioxidant activities and underlying mechanisms of SS and SM on the chicken hepatocellular carcinoma cell line (LMH). The cytotoxicity results showed that the safe concentration of SM was up to 1000 ng/mL, while SS was 100 ng/mL. In Se treatments, both SS and SM significantly elevated mRNA stability and the protein synthesis rate of glutathione peroxidase (GPx) and thioredoxin reductase (TrxR), two Se-containing antioxidant enzymes. Furthermore, SM exerted protective effects in the H2O2-induced oxidant stress model by reducing free radicals (including ROS, MDA, and NO) and elevating the activities of antioxidative enzymes, which performed better than SS. Furthermore, the results showed that cotreatment with SM significantly induced apoptosis induced by H2O2on elevating the content of Bcl-2 and decreasing caspase-3. Moreover, investigations of the mechanism revealed that SM might exert antioxidant effects on H2O2-induced LMHs by activating the Nrf2 pathway and enhancing the activities of major antioxidant selenoenzymes downstream. These findings provide evidence for the effectiveness of SM on ameliorating H2O2-induced oxidative impairment and suggest SM has the potential to be used in the prevention or adjuvant treatment of oxidative-related impairment in poultry feeds. Keywords:selenomethionine;sodium selenite;antioxidants;glutathione peroxidase;Nrf2

A H 2 O 2 etching strategy was adopted to introduce coordinatively unsaturated sites (CUS) on MoS 2 -based catalysts for dibenzothiophene (DBT) hydrodesulfurization (HDS). The CUS concentrations on MoS 2 slabs were finely regulated by changing the concentrations of H 2 O 2 solution. With the increasing H 2 O 2 concentrations (0.1–0.3 mol/L), The CUS concentrations on MoS 2 slabs increased gradually. However, the high-concentration H 2 O 2 etching (0.5 mol/L) increased the MoO x S y and MoO 3 contents on MoS 2 slabs compared to etching with the H 2 O 2 concentration of 0.3 mol/L, which led to the less CUS concentration in the sulfided Mo–H-0.5 catalyst than in the sulfided Mo–H-0.3 catalyst. A microstructure-activity correlation indicated that the CUS introduced by H 2 O 2 etching on MoS 2 slabs significantly enhanced DBT HDS. Different Co loadings were further introduced into Mo–H-0.3, which had the most CUS concentration, and the corresponding 0.2-CoMo catalyst with the highest CoMoS content (3.853 wt%) exhibited the highest reaction rate constant of 6.95 × 10 −6 mol g −1 s −1 among these CoMo catalysts.

Electrochemical hydrogen peroxide (H 2 O 2 ) synthesis from water and air via two-electron (2e − ) water oxidation or oxygen reduction reactions could provide a sustainable route for on-site H 2 O 2 production. Unfortunately, the electrochemical H 2 O-to-H 2 O 2 process usually suffers from limited selectivity under high current density. Here, we present an atomic Ni-doped TiO 2 electrocatalyst that enables H 2 O 2 production through 2e − water oxidation with high Faradaic efficiency of 70% at ∼300 mA cm −2 . Furthermore, when paired with the 2e − oxygen reduction reaction using the oxidized carbon nanotube cathode, as high as 146% Faradaic efficiency for H 2 O 2 is achieved at 240 mA in a full flow cell device. Moreover, this device could be operated for modular H 2 O 2 synthesis with a production rate of 109.12 μmol min −1 (i.e., 3.7 mg min −1 ). This work demonstrates the atomically controlled metal oxide electrocatalyst for harsh anodic H 2 O to H 2 O 2 and the practical applicability for the on-site H 2 O 2 production from earth-abundant resources.

Resveratrol performs a variety of biological activities, including the potential regulation of autophagy. However, it is unclear whether resveratrol protects against luteal dysfunction and whether autophagy involves the regulation of resveratrol. This study aims to investigate whether resveratrol can regulate autophagy to resist H2O2-induced luteinized granulosa cell dysfunction in vitro. Our results showed that resveratrol can enhance cell viability, stimulate the secretion of progesterone and estradiol, and resist cell apoptosis in H2O2-induced luteinized granulosa cell dysfunction. Resveratrol can activate autophagy by stimulating the expression of autophagy-related genes at the transcriptional and translational levels and increasing the formation of autophagosomes and autophagolysosomes. Rapamycin, 3-methyladenine, and bafilomycin A1 regulated the levels of autophagy-related genes in H2O2-induced luteinized granulosa cell dysfunction and further confirmed the protective role of autophagy activated by resveratrol. In conclusion, resveratrol activates autophagy to resist H2O2-induced oxidative dysfunction, which is crucial for stabilizing the secretory function of luteinized granulosa cells and inhibiting apoptosis. This study may contribute to revealing the protective effects of resveratrol on resisting luteal dysfunction from the perspective of regulating autophagy. Keywords:autophagy;resveratrol;corpus luteum;luteinized granulosa cell

Diabetic retinopathy (DR) is a prevalent microvascular complication of diabetes and the leading cause of blindness and severe visual impairment in adults. The high levels of glucose trigger multiple intracellular oxidative stress pathways, such as POLDIP2, resulting in excessive reactive oxygen species (ROS) production and increased expression of VCAM-1, HIF-1α, and VEGF, causing microvascular dysfunction. Dihydromyricetin (DMY) is a natural flavonoid small molecule antioxidant. However, it exhibits poor solubility in physiological environments, has a short half-life in vivo, and has low oral bioavailability. In this study, we present, for the first time, the synthesis of ultra-small Fe-DMY nano-coordinated polymer particles (Fe-DMY NCPs), formed by combining DMY with low-toxicity iron ions. In vitro and in vivo experiments confirm that Fe-DMY NCPs alleviate oxidative stress-induced damage to vascular endothelial cells by high glucose, scavenge excess ROS, and improve pathological features of DR, such as retinal vascular leakage and neovascularization. Mechanistic validation indicates that Fe-DMY NCPs can inhibit the activation of the Poldip2-Nox4-H 2 O 2 signaling pathway and downregulate vital vascular function indicators such as VCAM-1, HIF-1α, and VEGF. These findings suggest that Fe-DMY NCPs could serve as a safe and effective antioxidant and microangio-protective agent, with the potential as a novel multimeric drug for DR therapy.

Background Osteoarthritis (OA) is a chronic degenerative joint disease characterized by cartilage degeneration and intra-articular inflammation. Daurisoline (DAS) is an isoquinoline alkaloid isolated from Rhizoma Menispermi, whose antitumor and anti-inflammatory pharmacological effects have been demonstrated, but the effects of DAS on OA have rarely been researched. In this study, we aimed to explore the potential role of DAS in OA and its partial mechanism.
Materials and methods The cytotoxicity of H 2 O 2 and DAS toward chondrocytes was detected by the Cell Counting Kit-8 assay. Safranin O staining was used to detect chondrocyte phenotype changes. Cell apoptosis was measured by both flow cytometry and quantitative analysis of the protein levels of the apoptosis-related factors Bax, Bcl-2 and cleaved caspase-3 by western blot. Western blotting and immunofluorescence were used to assess the expression of the autophagy-related proteins LC3, Beclin-1 and p62. In addition, key signal pathway targets and matrix-degrading indicators were measured by western blot. Results Our results indicated that H 2 O 2 induced human chondrocyte apoptosis and activated autophagy in a dose-dependent manner. DAS treatment dose-dependently reversed the expression of apoptosis-related proteins (Bax, Bcl-2 and cleaved caspase3) and the apoptosis rate induced by H 2 O 2 . Western blot and immunofluorescence analyses showed that DAS decreased the H 2 O 2 -induced upregulation of the autophagy marker Beclin-1 and the LC3 II/LC3 I ratio and upregulated the p62 protein level. Mechanistically, DAS inhibited autophagy through the activation of the classical PI3K/AKT/mTOR signaling pathway and protected chondrocytes from apoptosis. In addition, DAS alleviated the H 2 O 2 -induced degradation of type II collagen and the high expression of matrix metalloproteinase 3 (MMP3) and MMP13. Conclusion Our research demonstrated that DAS alleviated chondrocyte autophagy caused by H 2 O 2 through activation of the PI3K/AKT/mTOR signaling pathway and protected chondrocytes from apoptosis and matrix degradation. In conclusion, these findings suggest that DAS may serve as a promising therapeutic strategy for OA.

Solar-powered photocatalytic H2O2 production is promising for its advantages of being safe, portable, green and energy efficient but achieving high efficiency is challenging, especially under pure water and air conditions. Herein, a titanium dioxide/polyethylenimine/Ag nanoparticle (abbreviated as TiO2/PEI/AgNP) photocatalyst is prepared in a facile manner by electrostatic assembly and in situ polymer reduction, which combines local surface plasmon resonance and photocatalysis to exhibit ultra-fast H2O2 production activity in pure water. The obtained photocatalysts exhibit an unexpectedly high H2O2 production activity (1605 μmol g−1 h−1 under simulated sunlight) in air and pure water, which is 24-fold higher than that of pristine rutile TiO2. The TiO2/PEI/AgNP composites simultaneously optimize three parts of photocatalytic H2O2 production: (i) improved charge separation, (ii) captured H+ faster and (iii) blocked direct contact of H2O2 and the catalyst. Experimental data and density functional theory calculations prove that the AgNP component can rapidly conduct high-energy electrons generated by themselves and photogenerated electrons generated by TiO2 to reactants. The PEI component can capture H+ from pure water faster to promote the superoxide radical to generate H2O2 through the proton coupling reaction and block direct contact of H2O2 and the TiO2 component. This work establishes a paradigm for the rational design of efficient plasmonic photocatalysts via self-assembly and in situ reduction technologies toward H2O2 production in pure water and air.

Enzyme mimetics have been widely applied on H 2 O 2 assay, but it is still challenging and interesting to realize the sensitive detection for ultra-trace H 2 O 2 . Here, an ultrasensitive Raman assay method based on novel WO 3 @IP6-Fe 3+ enzyme mimetics with peroxidase-like activity was established. WO 3 microspheres (MSs) were found to have weak peroxidase-like activity, and the combination of IP6-Fe 3+ and WO 3 can produce stronger activity. WO 3 @IP6-Fe 3+ MSs showed polyhedron-like structure, uniform size, and smooth surface. Although WO 3 @IP6-Fe 3+ enzyme mimetics have low catalytic efficiency and high absorbance background, the proposed Raman method can bypass the above problems. In Raman method, high concentration of WO 3 @IP6-Fe 3+ can be used to overcome low catalytic efficiency without high absorbance background. Moreover, 3,3′,5,5′-tetramethylbenzidine oxide has prominent characteristic Raman peak at 1608 cm −1 , greatly improving the sensitivity and eliminating interference of impurities. Due to the high sensitivity and low background, Raman assay showed the ultra-low limit of detection (5.49 × 10 −15 M), which was 4–7 orders of magnitude lower than other detection methods. The ultrasensitive Raman assay not only provided the possibility for the enzyme mimetics-based detection of ultra-trace H 2 O 2 , but also enable the enzyme mimetics with low activity to be applied.

The low temperature solvothermal method synthesized Cr 2 O 3 NPs has not only peroxidase activity, but also oxidase activity. Then, the oxidase activity of Cr 2 O 3 NPs is effectively shielded by nanogel immobilization using three monomers acrylamide, NIPAAM (N-isopropylacrylamide) and MBA (N,N′-methylene bisacrylamide) in HEPES (4-(2-hydroxyerhyl)piperazine-1-erhanesulfonic acid) buffer. Ultimately, the enzymatic activity of Cr 2 O 3 @nanogel/Au is significantly enhanced after doping Au NPs by SERS (Surface Enhanced Raman Spectroscopy) evaluation. A SERS strategy was proposed for the detection of H 2 O 2 by Cr 2 O 3 @nanogel/Au. The linear range was 10 −8 mol·L −1 -10 −1 mol·L −1 .

In the present study, carbon nanofibe (CNFs) with a highly defective structure were prepared via microwave pyrolysis of H 2 O 2 activated alder wood. The nickel oxide nanoparticles (NiO NPs) were anchored on the surface of the carbon nanofibers with the aid of microwave hydrothermal treatment . The electrochemical reactivity of the carbon nanofiber/nickel oxide electrode was evaluated for nonenzymatic H 2 O 2 detection and exhibited a fast response speed (within 1 s), high sensitivity (304.2 µA cm −2 mM −1 ), wide linear working range (0.1 µM to 3489.9 µM), low detection limit (0.57 µM), and good reproducibility, anti-interference and stability. For tap water, the prepared electrode was shown the recovery and RSD of H 2 O 2 between 94.7 and 98.9 % and 2.6–5.4 %, respectively as well the recovery and RSD of H 2 O 2 in river water samples reached 101.2–104 % and 2.8–4.7 %, respectively. The catalytic mechanism of hydrogen peroxide by using the prepared carbon nanofibers/nickel oxide was analyzed.

Photoacoustic (PA) imaging with much deeper tissue penetration and better spatial resolution had been widely employed for the prevention and diagnosis of many diseases. In this study, a new type of hydrogen peroxide (H 2 O 2 ) -activated photoacoustic nanoprobe [Mn-AH nanoscale coordination polymer nanodots (NCPs)] was successfully synthesized by a simple one-step method in water phase containing 2,2 ′ -azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), horse radish peroxidase (HRP), and manganese ion (Mn 2 + ) . After modification by polyethylene glycol (PEG), Mn-AH NCPs exhibited excellent stability and biocompatibility for in vivo H 2 O 2 -responsive chromogenic assay with great specificity and sensitivity. In the presence of H 2 O 2 , colorless ABTS would be converted by HRP into the oxidized form with strong near-infrared (NIR) absorbance, enabling photoacoustic detection of endogenous H 2 O 2 . Using H 2 O 2 -activated Mn-AH NCPs, we have successfully performed PA imaging and H 2 O 2 detection of subcutaneous murine colon CT26 tumor and deep-seated orthotopic bladder tumor. Due to the inherent Mn element existence inside the Mn-AH, this nanoprobe also serves as a good T1-weighted magnetic resonance imaging (MRI) contrast agent simultaneously. Lastly, after accomplishing its imaging functions, the Mn-AH NCPs could be cleared out from the body without any long-term toxicity, providing a new opportunity for cancer diagnosis and treatment.

Kinetic models and mechanism of isomaltooligosaccharides (IMOs) by hydroxyl ( OH) radicals-driven degradation in ultraviolet/hydrogen peroxide system were investigated. Electron paramagnetic resonance spectra indicated that UV radiation played an important role in the steady generation of OH radicals. The OH radicals could effectively decrease the molecular weight of IMOs and significantly oxidize hydroxy group into carbonyl and carboxy groups . Main components of IMOs were separated and identified by HPAEC-PAD and UHPLC-MS/MS methods. Degradation behaviors of IMOs were well fitted to pseudo first-order kinetics. The hydrogen abstraction by OH radicals from different C H sites at pyranose ring began a cascade reaction leading to cleavage of α-glycosidic linkage or C C bonds with formation of stable uronic/aldonic acids. Degradation rate was closely influenced by the degree of polymerization (DP) of IMOs, the initial concentrations of IMOs and H 2 O 2 . The results would pave the way for free radicals-driven degradation of polysaccharides .
