Pursuing a selective and stable optical sensor for the reliable detection of perfluorooctane sulfonate (PFOS) is critical to controlling and reducing the risk of exposure to human. Here, we elaborately prepared an atomic copper(I)-carbon nitride (Cu-CN) peroxidase-mimic catalyst and therefore developed a reliable and accessible PFOS detection method. Well-distributed Cu(I) atoms are exclusively anchored within CN nanosized matrix as identified Cu-CN single-site active centers. Benefiting from the high peroxidase-mimic catalytic capability and colloidal stability, Cu-CN exhibits outstanding detection of PFOS selectively in the range of 0.07 to 3 μM coupled with the detection limit of 14.9 nM. The occupation of atomic Cu(I) specific catalytic sites caused by strong PFOS coordination and the fluorophilic interaction between PFOS should be responsible for the damping of TMB oxidation. Moreover, this detection platform was also used successfully to detect PFOS in three real water samples with RSD ≤ 8.37%. This well-established colorimetric sensing system based on peroxidase-mimic capability of single-atom catalyst is promising for the detection of other hazards in aquatic environment.

Fast and convenient synthesis of zero-dimensional (0D) cesium-copper(I) halide (Cs3Cu2X5 (X = Cl, I)) single crystals (SCs) is critical to achieve practical radiography. In this study, rapid growth of high-quality Cs3Cu2X5 (X = Cl, I) SCs with sizes reaching centimeter scale were achieved within 24 hours using scalable solution-phased methods. These crystals exhibited highly effective emission with large Stokes shifts, excellent PL linearity, and PLQYs exceeding 86%. Additionally, post-treated Cs3Cu2X5–PMMA films demonstrated outstanding scintillation performance, including excellent stabilities, light yield surpassing 36 000 photons per MeV, and spatial resolution up to 6.5 lp per mm in X-ray imaging tests. The high scintillation efficiency can be attributed to their compact 0D crystalline structure, resulting in a small defect density and strong exciton localization.

Our previous work demonstrated the tendon-derived extracellular matrix (ECM) extracts as vital niches to specifically direct mesenchymal stem cells towards tenogenic differentiation. This study aims to further define the effective ECM molecules capable of teno-lineage induction on human adipose-derived stem cells (hASCs) and test their function for tendon engineering. By detecting the teno-markers expression levels in hASCs exposed to various substrate coatings, collagen I (COL1) and fibromodulin (FMOD) were identified to be the key molecules as a combination and further employed to the modification of poly(L-lactide- co -ε-caprolactone) electrospun nanoyarns, which showed advantages in inducting seeded hASCs for teno-lineage specific differentiation. Under dynamic mechanical loading, modified scaffold seeded with hASCs formed neo-tendon in vitro at the histological level and formed better tendon tissue in vivo with mature histology and enhanced mechanical properties. Primary mechanistic investigation with RNA sequencing demonstrated that the inductive mechanism of these two molecules for hASCs tenogenic differentiation was directly correlated with positive regulation of peptidase activity, regulation of cell-substrate adhesion and regulation of cytoskeletal organization. These biological processes were potentially affected by LOC101929398/has-miR-197-3p/TENM4 ceRNA regulation axis. In summary, COL1 and FMOD in combination are the major bioactive molecules in tendon ECM for likely directing tenogenic phenotype of hASCs and certainly valuable for hASCs-based tendon engineering.

There has been an increase in the incidence of meniscus injuries worldwide, as well as growing needs for injectable hydrogels with the potentials to promote in situ meniscal tissue repair. This work developed poly(N-isopropylacrylamide)-collagen I composite hydrogels, with a series of different collagen I contents. The hydrogels were injectable under room temperature and started to solidify around 32°C due to their thermosensitivities, meeting the requirements of the intended applications. The addition of collagen I to the poly(N-isopropylacrylamide) provided physical reinforcement to the hydrogels. Rheological characterizations showed that, under room temperature, the complex viscosity, the storage modulus, and the loss modulus, respectively, increased from Pa•s to Pa•s, from Pa to Pa, and from Pa to Pa, as the collagen I content in the hydrogel increased from 0% to 1%. Cell proliferation tests and high-density culture also revealed that collagen I promoted bioactivity of the hydrogels and induced fibrochondrocyte phenotype of the chondrocytes. The expressions of collagen I protein and collagen I a1 gene in the PNIPAm-0.5Coll group were 3.50-fold and 3.64-fold of those in the PNIPAm group. In contrast, the expressions of collagen II protein and collagen II a1 gene were less prominent (respectively, 2.36-fold and 3.34-fold in the PNIPAm-0.5Coll group compared with the PNIPAm group). This phenotyping was believed to be conducive to the regeneration of meniscal tissues. The findings of this study have provided an important basis for future in vivo studies and clinical applications of this type of composite hydrogels.

This paper proposes a selective strategy for quantitative detecting cardiac troponin I (cTnI) in clinical serum samples by using Raman spectroscopy combined with magnetic molecularly imprinted polymers (MMIPs). The signature peptide was selected as a template for molecular imprinting due to its chemical stability and small molecular mass. MMIPs were synthetized with the signature peptide as template and methylacrylic acid as monomer in presence of vinyl-modified magnetic particles. MMIPs with high magnetic character and adsorption properties were able to be stained by Coomassie Brilliant Blue G-250 after the target template was specifically captured, which showing a characteristic absorption peak at Raman shift 1610 cm −1 . The Raman signal intensity at 1610 cm −1 showed a linear relationship in the 0.01 to 100 ng·mL −1 concentration range. The recovery of cTnI in serum were 92.7–111.2 % with relative standard deviation in the range of 2.51 % to 8.52 %. This paper has important practical application for preliminary rapid detection of cTnI in clinical serum samples.

Chronic cerebral hypoperfusion (CCH) is the leading cause of chronic cerebral dysfunction syndrome with its complex pathological mechanisms involving cortical and hippocampal neuronal loss, white matter lesions, and neuroinflammation. I-C-F-6 is a septapeptide, which has anti-inflammatory and anti-fibrotic effects. This study aimed to evaluate the neuroprotective effect of I-C-F-6 in chronic cerebral hypoperfusion (CCH)-induced neurological injury. C57BL/6 J mice were subjected to bilateral common carotid artery stenosis (BCAS), and BV2 microglia cells were induced with oxygen-glucose deprivation (OGD). In vivo, mice were divided randomly into four groups: Sham, BCAS, GBE (30 mg/kg), and I-C-F-6 (0.5 mg/kg). In vitro, microglia were divided randomly into four groups: control, OGD, I-C-F-6 (25 μg/mL), and Shikonin (800 nmol/L). Through LFB, TUNEL, and NeuN staining, we found that I-C-F-6 was able to mitigate myelin pathology and reduce the number of apoptotic neurons. Furthermore, immunofluorescence staining revealed that I-C-F-6 was able to reduce microglia clustering and downregulate NF-κB p65. We also observed a significant downregulation of M1 phenotype microglia signature genes, such as TNF-α, iNOS, and upregulation of anti-inflammatory cytokines, such as Arg-1 and IL-10, indicating that I-C-F-6 may mainly reduce polarization towards the M1 phenotype in microglia. Notably, I-C-F-6 downregulated the expression of NF-κB signaling pathway-related proteins IKK-β and NF-κB p65, as well as pro-inflammatory cytokines IL-1β and iNOS. In conclusion, I-C-F-6 can improve neurological damage, alleviate neuroinflammation, and inhibit microglia polarization to the M1 phenotype via the NF-κB signaling pathway.

Antimicrobial resistance remains an utmost concern in human and veterinary medicine, impacting humans, animals, and the environment while significantly influencing the principles of One Health. While Riemerella anatipestifer ( R. anatipestifer ) is recognized as a waterfowl pathogen with multidrug-resistant properties, the specifics of its lincosamide resistance mechanism are inadequately understood. In this study, we identified a novel lincosamide resistance gene, lnu (I), in R. anatipestifer RCAD0121, and investigated its potential origin, transfer mechanisms, and dissemination status through genomic epidemiology. This exhibited 74.80 % amino acid identity with a previously reported gene, lnu (H). PCR analysis revealed lnu (I) prevalence in at least 44 R. anatipestifer isolates collected from multiple provinces in China. Furthermore, genomic mining unveiled 56 lnu (I) sequences within publicly available databases, primarily originating from environmental sources. In addition, members of the family Flavobacteriaceae were the dominant (16/56, 28.57 %) bacteria carrying the lnu (I) gene, with Flavobacterium exhibiting a similar GC content as lnu (I). Notably, specific instances of the lnu (I) gene were linked to mobile genetic elements within human and animal pathogenic bacteria. These findings suggest that Flavobacterium species within the environment could serve as potential ancestral sources of the novel lnu (I) gene, which has undergone mobilization events toward pathogenic bacteria.

By providing the spatial separation of the active sites and retaining high oxidative and reducing capacity, the direct Z-scheme heterostructure is considered the most potential structure for yielding photo-electric response. However, challenges still exist in the directional transfer of charge carriers between two semiconductors in direct Z-scheme structures. In this regard, by constructing the Vzn defect and p–n junction, a direct Z-scheme ZnxCd1−xS@ZnS-NiS heterostructure was obtained for the regulated electronic structure, which ensured high-yield hydrogen properties. The Zn vacancy in the partially-coated ZnS shell led to the Vzn energy level, and the addition of NiS led to the p–n structure, which caused a drastic downshift of the band edge potentials in comparison to that of pristine CdS. This variation gave rise to a staggered band edge alignment between ZnxCd1−xS and NiS, resulting in the variation of charge transfer kinetics from type-I to direct Z-scheme. Through careful characterization, it was found that the optimal photocatalytic hydrogen precipitation activity reached 16 683.6 μmol g−1 h−1, which was 70 times that of CdS, and this improvement was considered to form a spatial barrier, providing a clear direction and path for carrier transmission.

A new three-dimensional (3D) supramolecular compound, [Ag(dppe)(CF 3 COO)] n ( 1 ), was prepared by using Ag(I) as the silver source and dppe as the organic ligand through the self-assembly approach. The bridging dppe ligand makes compound 1 appear as a one-dimensional (1D) wavy chain consolidated by the intramolecular hydrogen bonds. Two adjacent 1D chains are expanded to a two-dimensional (2D) layer passing through the intermolecular C H···π interaction between the dppe ligands, and the 2D layers further weave into a 3D supramolecular structure via another intermolecular C H···π interactions between the dppe ligands. The precise structure of 1 was determined by single crystal X-ray diffractometer (SCXRD), and it was well-characterized by Fourier transform infrared spectroscopy (FT-IR), nuclear magnetic resonance (NMR) spectroscopy, powder X-ray diffraction (PXRD), thermogravimetric analysis (TG) and differential thermogravimetry analysis (DTG), and UV–vis diffuse reflectance spectrum. In addition, the photocurrent response, electrochemistry characteristic and photodegradation of 1 were investigated. The degradation efficiency (82 %) of 1 as catalyst to degrade methylene blue (MB) was significantly higher than that without catalyst.

As newly developed synthetic enzymes with exceptional catalytic capabilities and outstanding stability, nanozymes have drawn considerable interest in the realm of sensing. Using a simple hydrothermal process, iodine and copper-doped carbon dots (Cu,I-CDs) with simulated enzymes were fabricated in the current investigation. Cu,I-CDs demonstrate peroxidase-mimicking function together with high catalytic effectiveness due to aforementioned features. This led to generation of a colorimetric sensor for quick and accurate quantitative assessment of morphine (MOR). The outcomes showed the method's usefulness for the colorimetric detection of MOR. The linear range for MOR detection is 0.25–25 μg/mL having a reduced detection limit of 64 ng/mL. This sensor's successful use in the analysis of MOR in biological material is more noteworthy.

Photodynamic therapy (PDT) is a promising treatment regime for cancer therapy with some advantages such as minimal invasiveness, high spatiotemporal selectivity, and negligible drug resistance. Developing Type I photosensitizer (PS) is a viable strategy to overcome the limitations of conventional O 2 -dependent Type Ⅱ PS, whereas there are still significant challenges. Herein, we report the synthesis of β -cyclodextrin-based fluorescent hyperbranched polymer ( β -CD-HPG-DEP) through a one-step anionic ring-opening polymerization between β -cyclodextrin ( β -CD), glycidol and a fluorescent molecule DEP. The potential utilization of β -CD-HPG-DEP as the agent for cancer phototheranostics was also investigated in detail. The results demonstrated that β -CD-HPG-DEP possesses excellent water dispersibility, good biocompatibility, high reactive oxygen species (ROS) generation efficiency mainly via Type I mechanism, and extraordinary fluorescence imaging capability. Meanwhile, β -CD-HPG-DEP has great potential to serve as drug delivery vehicle since the cyclodextrin unit has many hydroxyl groups, thus providing a promising strategy for designing and fabrication of multifunctional theranostics. Considering the simple synthesis route, ready designability and multifunctional potential of anionic ring-opening polymerization, as well as good biocompatibility of resultant polymer, we believe the strategy developed in this work could also be a rational route to synthesize multifunctional composites for various biomedical applications.

With the increasing number of reports on aristolochic acid I (AAI), more and more toxic and side effects have been discovered successively. The main recognized carcinogenic mechanism is that AAI is metabolized into aristololactam I (AAT) in the body by nitroreductases , ultimately forming AAT-DNA adducts that cause disease. However, the carcinogenic mechanism is still not well understood by currently reported indirect method, there has always been a great demand to develop a direct method for real-time monitoring such process. In this work, surface-enhanced Raman spectroscopy (SERS) was used for the first time to monitor the process of AAI under the action of reducing agent sodium borohydride and catalyst Raney nickel to form AAT. We first found the abundant intermediate product-amino derivative of AAI, which was never reported before by other methods. The AAT was then obtained by a one-step dehydration reaction from the amino derivative of AAI under such reduction conditions. The product of amino derivative of AAI and AAT were further verified by thin-layer chromatography, H nuclear magnetic resonance spectra , mass spectrometry, and ultra-high performance liquid chromatography. Furthermore, a density functional theory-supported in-depth vibrational characterization of AAI and AAT was performed. The monitoring of the AAI reduction process by SERS can be of great significance for further exploration of its pathogenic mechanism, prevention, and monitoring of “nephropathy” and other diseases caused by AAI.
