The residues of crystal violet (CV) in shrimp were easily absorbed by biological tissues and caused serious damage to human health. In this study, doped TiO 2 SERS substrates were fabricated with three doping metals including Fe, Cu, and Co by sol-hydrothermal method. The SERS intensities reached the optimum at a Fe 3+ doping ratio of 5% with probe molecules of 4-MBA and CV. The 5% Fe-TiO 2 SERS substrates for monitoring CV residues obtained the best performance with LOD of 0.016 ppm and 0.03 ppm for standard solutions and shrimp protein extract solutions, respectively. The results indicated suitable doping metals could effectively modulate the energy bands of the semiconductor and improve the charge transfer between the substrate and analyte for a stronger SERS signal. The study would enrich the SERS technique for the fast test of CV in the actual samples.

Osteosarcoma (OS) calls for early diagnosis to significantly improve patient survival rates. Exosomes hold significant potential as noninvasive biomarkers for the early diagnosis of cancer. Here, we design a microfluidic device to purify and analyze plasma-derived exosomes by label-free surface-enhanced Raman spectroscopy (SERS) profiling for OS diagnosis. Exosomes were isolated, purified, and enriched using a size-dependent microfluidic chip with tangential flow filtration, achieving a high recovery rate of 82 %. The isolated exosomes were then analyzed by label-free SERS using a nanoarray chip with self-assembly monolayers of gold nanoparticles (GNPs). Exosomes originating from different OS cell types were differentiated based on the intrinsic SERS signals. Our approach was further employed to analyze the plasma-derived exosomes from healthy donors and OS patients without the need for specific biomarker labeling. A machine learning-based diagnostic model for OS was constructed, achieving an accuracy of 93 %. The findings indicate that our method is valuable for noninvasive and precise diagnosis of OS and could be generalized to other diseases in the future.

Noble metal aggregates have received increasing interest in environmental monitoring and biological analysis as SERS substrates due to their high sensitivity. Depositing nanoparticles onto oxide support presents an effective approach for assembling noble metals. In this study, Au@Fe 2 O 3 composite substrates were utilized for SERS detection. Low detection limits for 4-MBA (LOD 4-MBA = 4.79 × 10 −9 M) and MB (LOD MB = 3.36 × 10 −9 M) demonstrate this SERS technique has high sensitivity. The target molecule signal did not show a significant decline, indicating good temporal stability of the substrate. The synergistic effects arising from the electromagnetic enhancement from closely spaced Au NPs, chemical enhancement from Fe 2 O 3 , and enrichment by an external magnetic field endow the Au@Fe 2 O 3 composite substrate with outstanding SERS performance. Moreover, the Au@Fe 2 O 3 composite substrates have been successfully used to detect 4-ATP and 4-NTP in water, which has exhibited outstanding detection capability. (LOD 4-ATP = 7.25 × 10 −9 M, LOD 4-NTP = 1.85 × 10 −9 M).

Perovskite/metallic heterojunction-based surface enhanced Raman scattering (SERS) substrates have been proven to be capable of providing Raman enhancement. However, the inherent water instability and poor dispersibility of perovskite/metallic nanocomposites-based SERS substrates pose significant challenges to their application in aqueous environments. Herein, polydopamine (PDA)-encapsulated cesium lead bromide (CsPbBr 3 ) adsorbing gold nanoparticles (AuNPs), termed as CsPbBr 3 @PDA@AuNPs, is prepared as SERS substrate, which exhibits excellent water stability and SERS activity. Dopamine as organic ligand not only passivates surface defects during the growth of perovskite nanocrystals, but also forms porous PDA protective layer, effectively preventing degradation of perovskite in aqueous medium. Meanwhile, PDA with abundant functional groups and conjugated π structure will adsorb AuNPs and promote electron flow between CsPbBr 3 and AuNPs, resulting in strong SERS activity. Based on the results, a SERS aptasensor has been fabricated by conjugation between CsPbBr 3 @PDA@AuNPs and double-stranded DNA (dsDNA), which is composed of neuron-specific enolase (NSE) aptamer and partial complementary signal-stranded DNA (ssDNA). The working strategy of as-fabricated SERS aptasensor is based on the conformational change (of ssDNA)-triggered Raman response for the detection of NSE. Upon the addition of NSE, the specific binding of NSE aptamers to NSE can convert rigid dsDNA into a flexible ssDNA, and the Cy5 signal molecule modified at the end of ssDNA will close to CsPbBr 3 @PDA@AuNPs SERS substrate, generating significant Raman signals with the lower limit of detection (1.02 pg/mL) of NSE. The SERS aptasensor has broad application prospect in the field of life/medicine science fields (e.g. early diagnosis and screening of disease).

As one of the most extensively used antibiotics in aquaculture in China, enrofloxacin (ENR) tends to accumulate in human body and endanger people’s health. Rapid, sensitive, and facile assays for enrofloxacin are urgently needed. In this study, based on the solid-phase substrate preparation strategy combined with microsphere etching and magnetron sputtering, Ag@Au nanoparticle (Ag@Au NP) solid array (Ag@Au NSA) with high surface-enhanced Raman spectroscopy (SERS) enhancement factor (EF≈8.3 × 10 7 ) was developed. Finite-difference time-domain (FDTD) simulations revealed that the Ag@Au NSA achieved maximum electromagnetic enhancement (EM) under the following optimized conditions: 633 nm excitation wavelength, 7.9 nm Au shell thickness, and 30.5 nm Ag core size. Additionally, SERS mapping results confirmed both the excellent enhancement performance and signal reproducibility of Ag@Au NSA, due to the expected micro-region enhancement with long-term effects accompanied by a larger range of high-density “hot spots.” The proposed SERS sensing for ENR based on Ag@Au NSA had a limit of detection (LOD) of 0.076 ng/mL ( R 2 ≈ 0. 9991) and was further applied to real-sample detection of ENR in crayfish tail meat with good recoveries ranging from 91.36 to 112.60%, indicating its great potential in practice. Determination of antipsychotic drugs in humans is meaningful for both individualized therapy and therapeutic drug monitoring. Quetiapine (QTP), as one of the newly developed antipsychotic drugs, may pose risks to human health if used improperly. In this work, we synthesized a three-dimensional (3D) Au nanorods/MXene (Ti 3 C 2 T x )/nickel foams (named as AuNRs/MXene/NFs) composite material for detecting the antipsychotic drugs QTP. The prepared negatively charged MXene was first assembled on the surface of the NFs through electrostatic interaction to form MXene/NFs, and then the prepared AuNRs were anchored on the MXene/NFs to obtain the final 3D AuNRs/MXene/NFs. By integrating the porous structure and magnetic properties of NFs, large specific surface area of MXene, and the plasmonic hot spots of Au nanorods, the AuNRs/MXene/NFs-based SERS platform can easily and sensitively detect QTP in human urine with the limit of detection of 1.71 × 10 −9 mol/L. The linearity was distinguished over the concentration range of QTP from 1 × 10 −7 to 1 × 10 −4 mol/L (calculated at 1030 cm −1 ), with correlation coefficients (R 2 ) of 0.9955. The presented AuNRs/MXene/NFs-based SERS strategy realizes QTP detection using SERS technology and provides a novel protocol for the therapeutic drug monitoring.

Alternariol (AOH) is a secondary toxin produced by the metabolism of Alternaria, which poses a significant food safety risk due to its widespread presence in fruits and vegetables. In this study, a dual-mode sensor based on AOH aptamer was developed, which combined with surface-enhanced Raman scattering (SERS) and fluorescence (FL) detection technology to achieve sensitive and specific detection of AOH. The sensor significantly improves the reliability of detection through the synergistic change and mutual verification of SERS and FL signals. The linear detection range of SERS mode was 0.5–100 ng/mL ( R 2 = 0.9927), and the detection limit was as low as 0.064 ng/mL. The linear range of FL mode was 1–100 ng/mL ( R 2 = 0.9792), and the detection limit was 0.14 ng/mL. Experiments showed that the sensor could effectively distinguish AOH from other mycotoxins, and showed good recovery (SERS: 84.09 % -101.40 %, FL: 4.69 % -105.57 %) and precision (RSD < 6.3 % and 8.9 %, respectively) in complex matrices such as kiwifruit, tomato and citrus. The results were consistent with those of the traditional LC-MS method, which verified its reliability. The sensing platform constructed in this study not only provides a reliable technical means for the trace detection of AOH in agricultural products, but also opens up a new way for the rapid screening and quantitative analysis of other mycotoxins in food matrices.

Deoxynivalenol (DON) is a secondary metabolite produced by fungi that contaminates food during the process of production and storage, and can potentially cause acute or chronic poisoning upon ingestion. This study employs surface-enhanced Raman spectroscopy (SERS) combined with density functional theory (DFT) to detect the pure DON sample. The investigation focuses on the adsorption mechanism of DON molecules on gold nanoparticles (AuNPs) and indicates that the O6 atoms in the DON molecule act as sensitive sites for nucleophilic reactions by using DFT calculations. Experimental results demonstrate that pure DON molecules exhibit a strong SERS response, displaying excellent linear and non-linear relationships across concentration ranges of 0.01–0.2 μg/mL (R 2 = 0.992) and 0.2–1 μg/mL (R 2 = 0.997), respectively. The limit of detection (LOD) and quantification (LOQ) are 0.0085 μg/mL and 0.0256 μg/mL, respectively, which show excellent detection sensitivity. These findings can provide a theoretical foundation for monitoring DON toxins in foodstuffs.

Digital microfluidics (DMF) is a powerful platform for a broad range of applications, especially immunoassays having multiple steps, due to the advantages of low reagent consumption and high automatization. Surface enhanced Raman scattering (SERS) has been proven as an attractive method for highly sensitive and multiplex detection, because of its remarkable signal amplification and excellent spatial resolution. Here we propose a SERS-based immunoassay with DMF for rapid, automated, and sensitive detection of disease biomarkers. SERS tags labeled with Raman reporter 4-mercaptobenzoic acid (4-MBA) were synthesized with a core@shell nanostructure and showed strong signals, good uniformity, and high stability. A sandwich immunoassay was designed, in which magnetic beads coated with antibodies were used as solid support to capture antigens from samples to form a beads–antibody–antigen immunocomplex. By labeling the immunocomplex with a detection antibody-functionalized SERS tag, antigen can be sensitively detected through the strong SERS signal. The automation capability of DMF can greatly simplify the assay procedure while reducing the risk of exposure to hazardous samples. Quantitative detection of avian influenza virus H5N1 in buffer and human serum was implemented to demonstrate the utility of the DMF-SERS method. The DMF-SERS method shows excellent sensitivity (LOD of 74 pg/mL) and selectivity for H5N1 detection with less assay time (<1 h) and lower reagent consumption (∼30 μL) compared to the standard ELISA method. Therefore, this DMF-SERS method holds great potentials for automated and sensitive detection of a variety of infectious diseases.

Flexible wearable biosensors enable dynamic tracking of biomarkers in sweat through noninvasive sampling. Surface-enhanced Raman spectroscopy (SERS) utilizes the “fingerprint” vibrations of molecular vibration spectra to simultaneously analyze multiple substances, facilitating the identification of complex analytes. In this study, we integrated a Janus fabric with efficient wearable moisture management properties with silver (Ag) nanoparticles to develop an SERS sensor for human sweat detection. The hydrophobic side of the Janus fabric was designed to contact the skin, while hydrophobic patterns were printed on the opposite side to enable the efficient enrichment of target molecules. Rapid sweat transport was achieved through a combination of a hydrophilic–hydrophobic interfacial design. The moisture management capability and SERS enhancement mechanism of the platform were further explained through simulation. Moreover, the platform demonstrated the successful detection of real human sweat samples. This work is expected not only to provide a novel approach for the fabrication and preparation of the SERS active fabric with excellent performance but also to promote the theoretical basis and technical support for the research of textile on rapid detection in highly diluted biochemical samples.

The overused quinolone antibiotics in animal husbandry and clinical medicine pose a growing threat for global health as they enter ecosystem via agricultural discharge and medical wastewater. Consequently, risk assessment for environmental and human exposure is highly demanded, which calls for an accurate, reliable and rapid qualitative and quantitative analysis of trace antibiotics in different matrices. Surface-enhanced Raman spectroscopy (SERS), providing fingerprint chemical information with near-single-molecule sensitivity, has received considerable attention. Herein, considering the common physicochemical properties of various quinolone antibiotics, we first developed sample pretreatment strategy consisting of solid-phase extraction (SPE), liquid–liquid extraction (LLE), salting-out assisted liquid–liquid extraction (SALLE), and NPs purification (NPs (p)). The modular SPE–LLE–SALLE–NPs (p) coupling with spectral separate network (SSNet) (a deep-learning based spectral unmixing algorithm) enabled a rapid (<8 min per sample), sensitive (μg L–1), on-site, and intelligent SERS analysis of 19 quinolone antibiotics in food, biological, and environmental matrices. Besides, the identification of each QN in a five-target mixture sample enabled SPE–LLE–SALLE–NPs (p)–SSNet–SERS to be a highly promising rapid detection strategy in blind sample screening qualitatively and quantitatively. The integrated SPE–LLE–SALLE–NPs (p)–SSNet–SERS offers a cross-media, full-process, and big data-enhanced rapid monitoring solution for antibiotic pollution within the “source-pathway-sink” framework.

In the quality control of braised meat products, the content of spice extract in braised soup is often used as one of the important indexes to evaluate its flavor and quality. However, the existing research methods mostly rely on chromatography-mass spectrometry for quantitative analysis, which has some problems such as complex operation, long period, high requirements for equipment and professional technology. To enable rapid detection of spice extracts while reducing analytical costs and improving efficiency, this study took the main extracts of star anise, Sichuan pepper and chili peppers, namely shikimic acid xanthoxylin and capsaicin, as the research objects. By preparing AuNPs as the substrate material, a rapid classification and recognition method for SERS spectra of the three different spice extracts was constructed. The results showed that the Raman signals of three spice extracts could be obviously enhanced by using AuNPs as substrate, with good reproducibility ( RSD < 5.5 %). SERE spectrum could clearly distinguish the differences of SERS peaks of shikimic acid, xanthoxylin and capsaicin. Principal component analysis (PCA) and linear discriminant analysis (LDA) further confirmed clear visual classification of the spice extracts. Finally, based on the SERS spectra of different concentrations, the digital quantitative equations of SERS spectra of three spice extracts were constructed. This work provides a theoretical foundation and technical framework for rapid detection of spice extracts based on SERS technology.
