The healing process of a diabetic wound (DW) is often impeded by a series of interrelated factors, including severe infection, persistent inflammation, and excessive oxidative stress. Therefore, it is particularly crucial to develop a medical dressing that can address these issues simultaneously. To this end, different ratios ofBletilla striatapolysaccharide (BSP) and berberine (BER) were physically blended with Carbomer 940 (CBM940) to develop a composite hydrogel as a medical dressing. The BSP/BER hydrogel was characterized using SEM, FTIR, rheological testing and other techniques. The anti-inflammatory, antioxidant, and antibacterial properties of the hydrogel were evaluated using cell and bacterial models in vitro. A DW model of ICR mice was established to evaluate the effect of the hydrogel on DW healing in vivo. The hydrogel exhibited excellent biocompatibility and remarkable antibacterial, anti-inflammatory, and antioxidant properties. In addition, animal experiments showed that the BSP/BER hydrogel significantly accelerated wound healing in DW mice. Among the different formulations, the LBSP/BER hydrogel (2% BSP, mBER:mBSP= 1:40) demonstrated the most remarkable efficacy. In conclusion, the BSP/BER hydrogel developed exhibited immense properties and great potential as a medical dressing for the repair of DW, addressing a crucial need in clinical practice. Keywords:diabetic wound;Bletilla striatapolysaccharide;berberine;hydrogel

The research on the activity of selenium (Se)-enriched agricultural products is receiving increasing attention since Se was recognized for its antioxidant activities and for its enhancement of immunity in trace elements. In this study, antioxidant Se-containing peptides, namely, Se-TAPepI-1 and Se-TAPepI-2, were optimally separated and prepared from Se-enriched tea protein hydrolysates by ultrafiltration and Sephadex G-25 purification, and subsequently, their physicochemical properties, oligopeptide sequence, and potential antioxidant mechanism were analyzed. Through the optimization of enzymatic hydrolysis conditions, the Se-enriched tea protein hydrolyzed by papain exhibited a better free radical scavenging activity. After separation and purification of hydrolysates, the two peptide fractions obtained showed significant differences in selenium content, amino acid composition, apparent morphology, peptide sequence, and free radical scavenging activity. Therein, two peptides from Se-TAPepI-1 included LPMFG (563.27 Da) and YPQSFIR (909.47 Da), and three peptides from Se-TAPepI-2 included GVNVPYK (775.42 Da), KGGPGG (552.24 Da), and GDEPPIVK (853.45 Da). Se-TAPepI-1 and Se-TAPepI-2 could ameliorate the cell peroxidation damage and inflammation by regulating NRF2/ARE pathway expression. Comparably, Se-TAPepI-1 showed a better regulatory effect than Se-TAPepI-2 due to their higher Se content, typical amino acid composition and sequence, higher surface roughness, and a looser arrangement in their apparent morphology. These results expanded the functional activities of tea peptide and provided the theoretical basis for the development of Se-containing peptides from Se-enriched tea as a potential natural source of antioxidant dietary supplements. Keywords:selenium;tea protein;tea peptide;identification;antioxidant activity

Alcalase time-specifically dictated hydrolysis property, peptide profiles and multifunction. • Antioxidant and ACE inhibition positively correlated with structural unfolding related hydrolysis efficacy promotion. • DPP-IV inhibition relied more on characteristics of specific peptide motifs. • Activity-guided multivariate and differential peptide profile analysis revealed candidate peptides. Endpoint-based studies failed to clarify time-resolved hydrolysis properties, bioactivities, peptide profiles, and structural characteristics. Current study aimed to explore the correlations of these characteristics during Alcalase hydrolysis of cricket protein (CP) for 30–300 min. Prolonged hydrolysis increased degree of hydrolysis (DH) to 20.3%, with low-molecular-weight peptides (< 2 kDa) accounting for 88.0%–94.0%. Cricket protein hydrolysates (CPHs) showed strongest antioxidant and angiotensin I-converting enzyme (ACE) inhibitory activities at 30 and 300 min, respectively, whereas dipeptidyl peptidase-IV (DPP-IV) inhibition exhibited a bimodal pattern. Structural analyses demonstrated hydrolysis disrupted ordered CP structure, with α -helix+ β -sheet decreasing by 17.4%–26.5%, alongside lowered surface hydrophobicity and increased sulfhydryl exposure. Antioxidant and ACE inhibition were positively associated with structural unfolding and hydrolysis property promotion, whereas DPP-IV inhibition appeared to relate more to peptide-release characteristics. Activity-guided peptidomics and multivariate analyses prioritized LYPL/VYGPL, SGLFDK/PAALGL, and SGPRLH/FAGPS as candidate peptides potentially associated with antioxidant, ACE inhibitory, and DPP-IV inhibitory activities, respectively. Alzheimer's disease (AD) possesses a complex pathogenetic mechanism. Nowadays, multitarget agents are considered to have potential in effectively treating AD via triggering molecules in functionally complementary pathways at the same time. Here, based on the screening (∼1400 compounds) against neuroinflammation , an imidazolylacetophenone oxime ether ( IOE ) was discovered as a novel hit. In order to obtain SARs , a series of imidazolylacetophenone oxime derivatives were constructed, and their C=N bonds were confirmed as the Z configuration by single crystals . These derivatives exhibited potential multifunctional neuroprotective effects including anti-neuroinflammatory, antioxidative damage, metal-chelating, inhibition of acetylcholinesterase (AChE) properties. Among these derivatives, compound 12i displayed the most potent inhibitory activity against nitric oxide (NO) production with EC 50 value of 0.57 μM 12i can dose-dependently suppress the expression of iNOS and COX-2 but not change the expression of HO-1 protein. Moreover, 12i exhibited evidently neuroprotective effects on H 2 O 2 -induced PC12 cells damage and ferroptosis without cytotoxicity at 10 μM, as well as selectively metal chelating properties via chelating Cu 2+ . In addition, 12i showed a mixed-type inhibitory effect on AChE in vitro . The structure-activity relationships (SARs) analysis indicated that dioxolane groups on benzene ring and rigid oxime ester can improve the activity. Parallel artificial membrane permeation assay (PAMPA) also verified that 12i can overcome the blood-brain barrier (BBB). Overall, this is the first report on imidazolylacetophenone oxime-based multifunctional neuroprotective effects, suggesting that this type of compounds might be novel multifunctional agents against AD.

High susceptibility of wet Spirulina platensis to post-harvest degradation causes significant nutritional and functional quality losses. Fermentation is a proven strategy to improve bioactive properties but the advancement of this process through integration of predictive modeling analysis remains uninvestigated. Therefore, this study combined response surface methodology (RSM) with Multilayer perceptron artificial neural network (MLP-ANN) modeling to improve the accumulation of phenolic compounds in wet Spirulina beverage via sequential fermentation with Kluyveromyces marxianus and Lactiplantibacillus plantarum . Predicted fermentation conditions (yeast-to-bacteria inoculum ratio of 3.75:1.25, glucose concentration of 8.5%, and Spirulina concentration of 9.5%) suggested the production of 60.56 mg gallic acid equivalents (GAE)/L of total phenolic content (TPC),which was experimentally confirmed at 60.86 mg GAE/L. Both RSM and MLP-ANN models demonstrated strong predictive performance (R 2 > 0.99), but RSM outperformed MLP-ANN by achieving 0.49% of relative error as compared to 0.74% for MLP-ANN. Experimental validation of predictive condition confirmed 76.2%, 119.6%, and 96.69% increase in TPC, total flavonoid content (TFC), and DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity respectively. UPLC-MS (ultra-performance liquid chromatography-mass spectrometry) analysis of TPC revealed substantial increases in caffeic acid, kaempferol, and apigenin contents in the fermented beverage. TPC improvement could potentially be attributed to the possible activation of complementary microbial biotransformation pathways including aromatic amino acid metabolism and flavonoid precursor synthesis. An improvement in sensory acceptability and reduction in detectable biogenic amines has also been observed under predicted sequential fermentation conditions. These findings underscore the feasibility of predictive models to improve the functional quality of Spirulina -based fermented beverages.

Heteroatom doping except oxygen is the main method to improve the performance of carbon quantum dots (CQDs), the application is also limited by unpredictable biological negative effects and environmental risks from heteroatoms (N/P/S, etc.). To maximize the role of oxygen while avoiding the negative effects of other heteroatoms, herein, we rationally designed and synthesized a new type of carbon-oxygen quantum dots (COQDs) with high fluorescent performance aim to realize pH sensing and reactive oxygen species (ROS) scavenging. The toxicity test results from four levels of bacterias, Hela cells , Daphnia Magna, and zebrafish indicate that the designed COQDs possess high biocompatibility and low environmental risk. Based on the opposing response of COQDs and Rhodamine 6G (Rh 6G) fluorescent signal to pH changes in the range of 6.0–9.0, a COQDs-Rh 6G complex was constructed by combining Rh 6G for ratiometric pH sensing, and a scanning colorimetric analysis model was further developed for the rapid pH judgment of commercially available waters. The proposed COQDs and pH sensing system were also successfully applied for bioimaging and intracellular pH analysis, respectively. More importantly, the designed COQDs also exhibit excellent ROS scavenging ability, which can be used as an effective antioxidant with good biocompatibility .

Hemodialysis membranes play an important role in the treatment of renal failure, but their anticoagulant and antioxidant properties need to be improved to prevent membrane fouling and oxidative stress in dialysis. To these ends, a heparinoid gel incorporating carbon dots (CDs) was applied to modify polyethersulfone hemodialysis membranes. The CDs were mixed with ammonium persulfate , Sodium p-styrene sulfonate, and N, N - Methylenebisacrylamide monomers , and modified to surfaces of hemodialysis membranes through the in-situ polymerization of these monomers. As prepared hybrid hydrogel featured a high free radical scavenging rate and anticoagulant time for the hemodialysis membrane. The scavenging rates of the modified membrane for 2, 2-diphenyl-1-picrylhydrazyl (DPPH) and 2, 2′-azinobis-(3-ethylbenzthiazoline-6-sulphonate (ABTS + ) free radicals are as high as 74.01% and 97.78%. The activated partial thromboplastin time (APTT), prothrombin time (PT) and thrombin time (TT) of the dialysis membrane are 600 s,283 s and 348 s, respectively.

Date palm ( Phoenix dactylifera L.) is an important commercial crop extensively consumed as a staple food and has been applied in many ethnomedical systems. Fruit vinegar is a preservative, condiment, and beverage with a spectrum of health benefits. Studies about the preparation, chemical profiles, and bioactivities of date fruit vinegar (DFV) are fundamental requirements for industrialization production. This study focused on the lab-scaled producing conditions, chemical profiles of DFV, and its bioactivities in vitro. According to the results, a date wine containing 9.75% methanol was obtained by yeast fermenting the enzyme-hydrolyzed date juice with 23.11% ± 0.39% of total sugar content. The optimized acidic fermentation conditions were an inoculation amount of 0.02%, a fermentation temperature of 31.14°C, and an initial alcohol content of 7.78%. Total acidity and total phenolic contents of the DFV were 7.74% ± 0.29% and 1.44 mg gallic acid equivalent/mL, respectively. In total, 32 organic acids were quantitated in the unaged DFV, with acetic, L -malic, and oxoglutaric acids as the predominant compounds. A total of 930 volatiles were identified in the DFV, including 186 esters, 177 terpenoids, and 148 heterocyclic compounds. There are 18 phenolic acids presented in the DFV. In addition, 42 flavonoids were quantitated in the DFV, including catechin, taxifolin, and cynaroside. The results of free radical scavenging activities and reducing power demonstrated that the DFV displayed good antioxidant properties. The DFV also acted well on angiotensin-converting enzyme 2 inhibition. These results suggest that the DFV can be industrially developed as a dietary supplement.

A circular sustainable bioeconomy model, utilizing renewable resources like agricultural waste biomass, can address environmental issues caused by the massive utilization of fossil resources. The study aims at exploring the potential utilization of underutilized cottonseed hulls (CSHs) as a source of bioactive proanthocyanidins (PAs), and investigate their applicability in the production of biodegradable films. FT-IR, HPLC-QTOF/MS, MALDI-TOF/MS analyses served for characterizing the structure of PAs extracted from CSHs, and thiolysis reaction revealed that they primarily consisted of (epi)catechins with a few (epi)gallocatechins and (epi)afzelechin units, forming oligomeric proanthocyanidins of which the mean degree of polymerization (mDP) was 2.99. Three monomers, ten dimers, one tetramer, one pentamer, and one hexamer were theoretically identified in CSHs for the first time. The unique structure of CSHs sets them apart from most plants. The extracts displayed high in vitro antioxidant activity with IC 50 value of 0.074 in DPPH assay and 0.018 mg/mL in ABTS assay. Additionally, the CSH extracts were incorporated as antioxidants into chitosan (CS)-based biodegradable films. The addition of 0.25% CSH extracts significantly improved film thickness and water vapor resistance, and also reduced the water solubility and visible light transmission. Furthermore, the films enhanced the blueberry quality and extended the shelf-life. This comprehensive study not only addresses waste management issues but also offers avenues for innovative applications in sustainable agriculture and packaging.

Polydopamine (PDA) as a melanin-like biomimetic material with excellent biocompatibility, full spectrum light absorption capacity and antioxidation property has been extensively applied in the biomedical field. Based on the high reactivity of dopamine (DA), exploiting new strategies to fabricate novel PDA-based nano-biomaterials with controllable size and improved performance is valuable and desirable. Herein, we reported a facile way to synthesize pyrrole-doped polydopamine-pyrrole nanoparticles (PDA-nPY NPs) with tunable size and enhanced near-infrared (NIR) absorption capacity through self-oxidative polymerization of DA with PY in an alkaline ethanol/H2O/NH4OH solution. The PDA-nPY NPs maintain excellent biocompatibility and surface reactivity as PDA. By regulating the volume of added PY, PDA-150PY NPs with a smaller size (<100 nm) and four-fold higher absorption intensity at 808 nm than that of PDA can be successfully fabricated. In vitro and in vivo experiments effectively further demonstrate that PDA-150PY NPs can effectively inhibit tumor growth and completely thermally ablate a tumor. It is believed that these PY doped PDA-nPY NPs can be a potential photothermal (PT) agent in biomedical application.Keywords:polydopamine;pyrrole;self-oxidative polymerization;photothermal treatment;cancer therapy

Quercetin (QCT) is a promising dose-dependent nutraceutical that usually suffers from poor water solubility and low bioavailability issues. In this work, a novel QCT-loaded nanoscale delivery system was constructed based on the oxidative self-polymerization of melanin (Q@MNPs). The FT-IR, XRD, and Zeta potential analyses confirmed that QCT was successfully absorbed on the melanin nanoparticles (MNPs) via Π−Π and hydrogen bonding interactions. The encapsulation efficiency and particle size of Q@MNPs were 43.78% and 26.68 nm, respectively. Q@MNPs improved the thermal stability of QCT and the antioxidant properties in comparison to MNPs. Meanwhile, Q@MNPs presented fantastic photothermal conversion capacity and stability triggered by the NIR laser, which significantly enhanced the antibacterial capability with a sterilization rate of more than 98% againstE. coliandS. aureus. More importantly, Q@MNPs exhibited NIR/pH dual-responsive drug release behavior and good biocompatibility (at concentrations of < 100 μg/mL). Thus, Q@MNPs show promising prospects for flavonoid delivery. Keywords:melanin nanoparticles;quercetin;antioxidant activity;photothermal antimicrobial;stimuli drug release

Natural polyphenol-rich plant resources, such as agricultural waste, were proven to diminish insulin resistance and weight gain in rats on a high-fat diet. To test whether date seed polyphenol pills (DSPPs) might lower adipose tissue accumulation by precisely affecting adipocytes, we explored the impacts of DSPPs on cell proliferation, differentiation, and lipolysis in 3T3-L1 cells. We utilized tablets made commercially from date seed polyphenols that were mostly composed of epicatechin (45.9 g/kg). The total polyphenol and antioxidant capacities of the digested and non-digested DSPPs were also evaluated. DSPPs at doses of 25, 50, and 100 µg/mL hindered the proliferation of both pre-confluent preadipocytes and mature post-confluent adipocytes. DSPPs decreased the quantity of viable cells in completely developed adipocytes. Treatment with 100 µg/mL of DSPPs decreased the basal lipolysis of completely differentiated adipocytes but modestly boosted epinephrine-induced lipolysis. A significant transcription factor for the adipogenic gene, the peroxisome proliferator-activated receptor (PPAR), was repressed by DSPPs, which significantly decreased lipid buildup. The total polyphenol and antioxidant capacities were also increased after digestion with a good bubble Pearson correlation between both. DSPPs may have anti-obesity and anti-diabetic characteristics by inhibiting adipocyte development and basal lipolysis, which could be commercially industrialized. Keywords:agricultural waste;date seed polyphenols;industrial-scale pills;3T3-L1 cells;obesity
