l-Lysine effects on myosin carbonylation in metal-catalyzed oxidation: Implications for thermal gelation

Yuling Liu,Yuqi Wu,Ge Han,Jiaxin He,Yawen Wang,Jinfeng Pan,Shengjie Li

This study investigated the impact of α-amino adipic semialdehyde (AAS)-mediated lysine carbonylation on the thermal gelation of porcine myosin. Purified myosin (2 mg/mL) was pre-treated with N-Ethylmaleimide to block the sulfhydryl groups, then oxidized in a metal-catalyzed oxidation system (10 μM FeCl₃, 100 μM ascorbic acid, and 5 mM H₂O₂) with varying l -lysine concentrations (0–60 mM) for 24 h at 4 °C. Physiochemical attributes, structural conformation, and rheological properties ( G' ) of myosin were systematically monitored. Following heat-induced gelation (20–80 °C), the resulting gels were evaluated for water-holding capacity (WHC), microstructure, and intermolecular forces. Results showed that oxidation significantly induced myosin carboxylation, evidenced by a substantial increase in both total carbonyl and AAS contents. This oxidative stress triggered a decrease in protein solubility, increased turbidity and particle size, and the disruption of α-helix and tertiary structures. Consequently, the WHC and G' of the resulting gel declined markedly, characterized by a more porous and disordered microstructural network. Conversely, the addition of l -lysine effectively attenuated AAS formation in a dose-dependent manner, thereby mitigating the oxidative impairment of myosin's physiochemical and structural properties and preserving its gelation functionalities. These results indicate that AAS-mediated lysine carbonylation plays a detrimental role in myosin structural integrity and gelation performance. Furthermore, the incorporation of exogenous l -lysine offers a potential strategy to mitigate oxidative deterioration in muscle protein systems.

Study on Direct Grafting Maleic Anhydride onto Poly(D,L-lactide-co-glycolide) Chains

Shan Bai,Zhenzhong Ouyang

Direct graft copolymerization of maleic anhydride (MAH) onto poly(D,L-lactide-co-glycolide) (PLGA) was performed by use of benzoyl peroxide (BPO) as initiator. Fourier transform infrared spectra, nuclear magnetic resonance, titration analysis of maleation extent and water contact angle were employed to characterize the obtained MPLGA. The effects of monomer concentration, initiator concentration and reaction temperature on the degree of maleation were investigated. The MPLGA scaffold was obtained by using NaCl as the porogen. The results showed that maleic anhydride was successfully grafted onto the PLGA chains. The monomer and initiator concentrations played an important role in the graft copolymerization; the extent of maleation first increased with increasing content of MAH and BPO and then declined. The extent of maleation continuously increased with increasing temperature to 100°C, and then the rate of increase decreased. The extent of maleation could be controlled in the range from 0.15 to 0.71% by adjusting the graft copolymerization conditions. The introduction of anhydride enhanced the water contact angle of PLGA. The MPLGA scaffold showed a network of interconnected pores ranging from 50 to 300 μm.

In-vitro Degradation Behaviors of Poly(L-lactide-co-glycolide-co-ε-caprolactone) Microspheres

Meiling Yi,Zhihua Zhou,Wenjuan Liu,Tianlong Huang,Yanmin Zhao,Ping Chen,Ziwei Zhou,Dan Wang,Chao Zhang,Jianjun Fang

Among drug delivery systems, polymer microspheres have been proved to be the most promising carriers during the past several years. In this paper we describe the preparation of poly(L-lactide-co-glycolide-co-ε-caprolactone) (PLLGC) random terpolymer microspheres; their in-vitro degradation behaviors, including the changes of pH value, mass loss, molecular mass and surface morphology, were investigated during various degradation periods and compared with poly(lactic-co-glycolic acid) (PLGA) microspheres. The results showed that the introduction of the ε-caprolactone (ε-CL) unit reduced the degradation rate of the PLGA polymers. Therefore, we suggest the PLLGC microspheres with controllable degradation rate could supply a wider application future in drug delivery field than pure PLGA microspheres.

Structural characterization and antioxidant activities of polysaccharides from okra (Abelmoschus esculentus (L.) Moench) pericarp

Lu-Yao Ma,Rou Xu,Hai-Feng Lin,Ming-Yong Xie,Shao-Ping Nie,Jun-Yi Yin

In this study, okra ( Abelmoschus esculentus  (Linn.) Moench) pods (pericarp) obtained after removing seeds and flesh were applied for polysaccharide extraction. The 60 °C water extract polysaccharide (OPW-60) was obtained from okra peel at 60 °C, and then 100 °C water extract polysaccharide (OPW-100) was obtained from the residue at 100 °C. Acid extraction at 60 °C was finally used to obtain the polysaccharide by acid leaching at 60 °C (OPA-60). Then, structural characteristics and antioxidant activities investigations were performed on the extracted polysaccharides. The results showed that monosaccharide composition of three polysaccharides was different. OPW-60 was mainly type I rhamnogalacturonan (RG-I) pectin, while OPW-100 and OPA-60 consist of RG-I and homogalacturoan (HG) pectins. 1 H NMR analysis confirmed the existence of rhamnose and acetyl group among them. The structure diversity resulted in the difference of viscosity, molecular weight, surface morphology and antioxidant activities. OPW-60 exhibited the highest apparent viscosity, which might be attributed to the differences in molecular weight distribution and monosaccharide composition. OPW-60 was most effective in scavenging 2,2-Diphenyl-1-picrylhydrazyl radicals and chelating ferrous ion ability, while OPW-100 exhibited the highest hydroxyl radical scavenging activity. The results showed that polysaccharides with different structure and antioxidant ability could be obtained from okra under different extraction conditions.

Jinyan Gu,Haihui Zhang,Hui Yao,Jie Zhou,Yuqing Duan,Haile Ma

To investigate and compare the preliminary structural characteristics and biological activity in vitro of polysaccharides from Sagittaria sagittifolia L. (SSs) by different extration methods, three polysaccharides (SSW, SSU, and SSP) were obtained with hot water, ultrasound-assisted, and subcritical water extraction. Their structural features were elucidated using High Performance Liquid Chromatography (HPLC), Gas Chromatography (GC), Scanning Electron Microscopy (SEM), Infrared Spectroscopy (IR), Atomic Force Microscopy (AFM), Zeta Potential and Congo red methods. Furthermore, the antioxidant activity and immunostimulatory effects were investigated in vitro . Molecular weight and monosaccharide composition analysis exhibited that SSW (2275.0 kDa), SSU (148.7 kDa), and SSP (1984.0 kDa) were heteropolysaccharide with dramatically different monosaccharide species and mole ratios. In addition, SSP exhibited stronger antioxidant activity in vitro and more potent immunomodulatory activity than SSW and SSU. SSP has greater potential to be explored as biologicalagents for use in complementary medicine or functional foods.

Photoionization Study of L-Valine in the Gas Phase by Vacuum Ultraviolet Synchrotron Radiation

Shaohui Zhou,Genbai Chu,Lanlan Cao,Xiaobin Shan,Fuyi Liu,Ju-Guang Han,Liusi Sheng

The photoionization and photodissociation of L-valine are studied by tunable synchrotron vacuum ultraviolet photoionization mass spectrometry at the photon energy of 13 eV. The ionization energy of L-valine and the appearance energies of major fragments are measured by the photoionization efficiency spectrum in the photon energy range of 8–11 eV. Possible formation pathways of the major fragments, NH2CHC(OH)2+ (m/z = 75), NH2(CH3)2(CH)2+ (m/z = 72) and NH2CHCO+ (m/z = 57), are discussed in detail with the theoretical calculations at the B3LYP/6-31++G (d, p) level. Hydrogen migration is considered as the key way for the formation of NH2CHC(OH)2+ (m/z = 75) and NH2CHCO+ (m/z = 57). Further more, other fragments, NH2CHCOOH+ (m/z = 74), (C H3)2(C H)2+ (m/z = 56), C4H7+ (m/z = 55), NH2CHOH+ (m/z = 46), NH2CH2+ (m/z = 30) and m/z = 18, species are also briefly described.

Multi-enzyme cascade strategy for efficient biosynthesis of 5-hydroxypipecolic acid from L-lysine

Huamei Zhong,Xiaotong Li,Tongqing Tang,Hongyu Zhu,Xuean Song,Shewei Hu,Feifei Chen,Alei Zhang,Kequan Chen

Multi-enzyme cascade designed for 5-hydroxypipecolic acid synthesis from low-cost substrate. • Self-sufficient cofactor recycling achieved without expensive exogenous coenzymes. • Under glucose-controlled feeding, the system achieved a product titer of 23.65 g L -1 . 5-Hydroxypipecolic acid (5-HPA) is a chiral building block widely employed in pharmaceutical synthesis. Conventional chemical synthesis of 5-HPA is limited by poor stereoselectivity and environmental burdens. Herein, we developed a biosynthetic method for 5-HPA production from L-lysine (L-Lys) through a multi-enzyme cascade. Firstly, a lysine 6-aminotransferase SkL6AT from Stenotrophomonas koreensis and an imine reductase HeP5C from Halomonas elongata were screened and introduced into recombinant strains capable of converting L-Lys to L-pipecolic acid (L-PA). After optimization of plasmid copy number and ribosome-binding site strength, as well as establishment of the NADH regeneration system, the L-PA titer reached 7.50 g L⁻¹. Subsequently, a hydroxylase XdPH from Xenorhabdus doucetiae was introduced to construct a 5-HPA-producing strain. Fermentation conditions were optimized in shake flasks and subsequently scaled up to a 5 L bioreactor. Under a fed-batch strategy, the final engineered strain produced 23.65 g L -1 5-HPA with a yield of 0.587 g g⁻¹ from L-Lys. Multi-omics reveals mechanism of combined L-cysteine and GABA in loquat postharvest quality

Zhang Huifen,Pu Jing,Wang Miao,Du Ying,Tang Xiaofu,Niu Jie,Wang Hongxia,Luo Xian,Wang Yongqing,Deng Qunxian

Loquat is highly susceptible to postharvest deterioration, including browning and lignification, which limits its storage life. This study investigated the effects of combined L-cysteine (L-Cys) and γ-aminobutyric acid (GABA) treatment on postharvest quality maintenance of loquat fruit and elucidated the underlying molecular mechanisms through integrated transcriptomic and metabolomic analyses. Physiological assessments revealed that the combined treatment significantly delayed fruit softening, reduced weight loss and browning, suppressed decay incidence, and maintained higher levels of soluble solids, titratable acidity, and total phenolics. Integrated multi-omics analysis demonstrated that L-Cys+GABA treatment primarily modulated the phenylpropanoid metabolic pathway, leading to suppressed lignin biosynthesis through coordinated downregulation of key biosynthetic genes ( COMT , CCR , LAC ) and reduced accumulation of corresponding metabolites. These findings reveal that L-Cys+GABA combined treatment delays postharvest senescence in loquat fruit by targeting the phenylpropanoid pathway to inhibit lignification, providing a physiological and molecular basis for its application as an effective preservation strategy.

Continuously scaled-up preparation of poly(l-lactic acid) microspheres using high gravity RPB

Chao-min Cheng,Xiang-cheng Li,Tian-wei Shen,De-xu Liu,Guang-wen Chu,Yuan Le,Jian-feng Chen

High-throughput preparation of the specific size (20–50 μm) microspheres with narrowly distributed is of significant interest to the medical aesthetics market. This study developed a novel method for preparing this kind of poly(l-lactic acid) (PLLA) microspheres using high-gravity rotating packed bed (RPB) reactor. Investigated the effects of operating parameters, initial liquid distribution and wire mesh packing wettability on microspheres size distribution. The RPB system was configured with a composite jet for the initial liquid distribution and a modified wire mesh packing featuring a lotus-leaf-like self-cleaning surface to intensify liquid dispersion. The prepared PLLA microspheres is of D 50  = 31.27 μm, with a distribution span of 0.78. Employing a dimensionless group analysis, correlations were established to predict the average particle size and uniformity of the PLLA microspheres. The predicted values showed good agreement with experimental data, typically within a ± 10% deviation. Based on the obtained results, a large scale test was carried out, achieving a production capacity of 2 kg/h. The PLLA microspheres prepared on a large scale have a D 50  = 41.29 μm, with a distribution span of 0.70. High-gravity RPB, with its superior liquid–liquid heterogeneous mixing capability, should be a promising candidate for the large-scale production of polymeric microspheres.

Plasma-Assisted Extraction of Polysaccharides from Siegesbeckia orientalis L.: Optimization, Purification, and Structural Characterization

Yong-Hua Li,Li-Jie Zeng,Jin-Yun Wu,Jun Meng,Meng-Na Li,Jia-Yi Huang,Yan-Yan Huang,Feng-Song Liu

Natural polysaccharides fromSiegesbeckia orientalisL. have been reported to exhibit promising bioactivities. To enhance extraction efficiency, low-temperature plasma-assisted extraction was optimized forS. orientalisL. polysaccharides using single-factor experiments and response surface methodology. Column chromatography purified a homogeneous SIE-III fraction, followed by structural characterization. Optimal parameters were 80 kV discharge voltage, 153 Hz frequency, and 109 s treatment time, under which the polysaccharide yield reached 15.68%, significantly higher than that of the conventional hot water extraction method. Plasma treatment loosened the raw material’s surface, potentially facilitating polysaccharide release. SIE-III had a molecular weight of 20.831 kDa and comprised mainly galactose (51.7%), rhamnose (19.1%), arabinose (11.3%), and galacturonic acid (9.9%). It featured typical rhamnogalacturonan-I (RG-I) domains and a triple-helix conformation. Fourier transform infrared spectroscopy and nuclear magnetic resonance confirmed both α- and β- glycosidic linkages, and methylation analysis revealed a highly branched →3,4)-Galp-(1→ structure. This study provides an effective extraction method for plant polysaccharides and valuable insights into their potential applications in the food and other industries.

Molecularly imprinted polymer-based electrochemical sensor for L-tyrosine competition-mediated T cell exhaustion

Zhen Peng,Yihao Long,Zhihao Tu,Muhammad Tahir,Liang He

Nutrition competition exists in the tumor microenvironment (TME), which is considered as an important factor of T cell exhaustion. Various amino acids are the targets for both tumor cells and T cells, including L-Tyr, which plays an essential role in normal function of T cells. Herein, a novel sensing interface of MIP/Fc/PEDOT:PSS-PPy/graphite/Au established on a PCB substrate, was proposed for L-Tyr detection. This sensor was systematically characterized and showed excellent repeatability, stability, reproducibility, and specificity. Meanwhile, the sensor exhibited good detection capability, with a linear range of 1 nmol/L to 1 mmol/L, a limit of detection of 1.62 × 10 −11  mol/L, and a sensitivity of 97.14 μA/(mol/L). An in-vitro co-culture model of T cells and tumor cells was established to mimic the TME, and its culture medium was used as real samples for L-Tyr determination via the proposed sensor. The results demonstrated that the failure of L-Tyr competition by T cells in the TME was a vital cause of their exhaustion, which was further validated by the IL-2 expression of T cells within the TME using ELISA. Collectively, our study is expected to provide a new strategy for tumor treatment, by supplementing exogenous amino acids to alleviate T cell exhaustion.

Enhanced Lethal Effects of Combined P-tert-Butylcatechol and L-Lysine on Microcystis aeruginosa

Heyun Jiao,Gangwei Jiao,Ruitong Jiang,Yifei Shen,Peimin He,Liu Shao

Simple SummaryHarmful algal blooms caused byMicrocystis aeruginosathreaten aquatic ecosystems and human health. This study explores the combined effects of two natural allelochemicals, P-tert-butylcatechol (TBC) and L-lysine, in controllingM. aeruginosa. When combined at low concentrations (0.04 mg/L TBC + 1 mg/L L-lysine), they achieved over 90% growth inhibition within 96 h. The treatment damaged algal cell membranes, reduced photosynthetic efficiency, and triggered oxidative stress. Furthermore, intracellular and extracellular microcystin levels decreased significantly compared to untreated algae, minimizing ecological risks. These findings highlight the potential of the combination of TBC and L-lysine as a safe and cost-effective strategy for mitigating harmful algal blooms.Allelochemicals are recognized as promising algaecides due to their environmental safety. Para-tert-butylcatechol (TBC) and L-lysine exhibit significant potential in suppressing harmful algal blooms (HABs); however, their combined effects and algae inhibition mechanisms remain unelucidated. Therefore, this study systematically investigated the growth inhibition ofMicrocystis aeruginosaby TBC and L-lysine individually and in combination, while simultaneously examining their combined effects on algal growth, cell membrane integrity, photosynthetic activity, antioxidant responses, and microcystin production. The results revealed a significant interactive effect between TBC (0.04 mg/L) and L-lysine (1 mg/L), achieving over 90% growth inhibition within 96 h. The combined treatment significantly inhibitedM. aeruginosagrowth through impaired photosynthetic efficiency and elevated oxidative stress. Compared to the control group, the treatment group exhibited a continuous decline in chlorophyll-a content, phycobiliprotein levels,Fv/Fm, YII, α, and rETRmax, while phosphoenolpyruvate carboxylase (PEPC) activity decreased by 96.48% by day 8. And antioxidant enzymes, including superoxide dismutase (SOD) and reduced glutathione (GSH), showed a progressive increase in activity. In addition, the structure and integrity of the cell membrane ofM. aeruginosawere damaged after treatment, and the conductivity of the treatment groups increased continuously from 2.32 to 4.63 μs/cm. In addition, under combined treatment, intra- and extracellular microcystin levels initially increased (peaking at day 2) but sharply declined thereafter, becoming significantly lower than controls by day 8. These findings highlight the potential of combining TBC and L-lysine as an eco-friendly and cost-effective strategy for mitigatingM. aeruginosa-dominated harmful algal blooms.
