A combination of photothermal therapy (PTT) and chemotherapy is an emerging therapeutic strategy with promising clinical prospects in cancer treatment. Despite the huge progress achieved in the past years, a number of obstacles still hamper the therapeutic efficacy of this synergistic modality such as uneven heat distribution, lack of targetability of anti-cancer agents and dosage-related side effects. Thus, developing a nanoplatform for targeted drug delivery against cancer is of great necessity. Herein, a lipid–polymer hybrid nanosystem (LP/ID) based on polyethyleneimine (PEI)-lecithin-polyethylene glycol (PEG) was fabricated to co-load indocyanine green (ICG) and dichloroacetate (DCA) for combined photothermal/chemotherapy. DCA and ICG were linked to the PEI backbone to form a dense hydrophobic core through amide bonds and electrostatic interactions, which increased the payload of DCA and ICG as well as achieved enzyme-responsive drug release because of the overexpressed amidase in tumor cells. Lecithin and DSPE-PEG2000 self-assembled around the hydrophobic complexes to obtain prolonged blood circulation and attenuated systemic toxicity of the hybrid nanosystem. The prepared LP/ID exhibited favourable stability in a physiological environment, good tumor imaging properties, and satisfactory photothermal/chemotherapeutic performance. Moreover, LP/ID could also enhance the cellular uptake and tumor retention capacity in comparison with free drug administration. Notably, by co-loading two therapeutic agents with different anti-cancer mechanisms, an obvious inhibitory effect on tumor growth was observed with negligible damage to normal tissues and organs because of the synergistic photothermal/chemotherapy effect, indicating the great potential of LP/ID as a robust nanoplatform for cancer treatment.

Composite nanoparticles consisting upconversion nanoparticles (β-NaYF 4 :Yb/Er) and nitrogen doped TiO 2 nanoparticles were constructed via a chemical assembly method. Efficient energy transfer was enabled since the upconverted energy matched up with the band gap of N-TiO 2 . This enables the resulting composite nanoparticles to possess excellent Rhodamine B (RhB) degradation activity under near infrared (NIR) light irradiation.

Patients with bone defects suffer from a high rate of disability and deformity. Poor contact of grafts with defective bones and insufficient osteogenic activities lead to increased loose risks and unsatisfied repair efficacy. Although self-expanding scaffolds were developed to enhance bone integration, the limitations on the high transition temperature and the unsatisfied bioactivity hindered greatly their clinical application. Herein, we report a near-infrared-responsive and tight-contacting scaffold that comprises of shape memory polyurethane (SMPU) as the thermal-responsive matrix and magnesium (Mg) as the photothermal and bioactive component, which fabricated by the low temperature rapid prototyping (LT-RP) 3D printing technology. As designed, due to synergistic effects of the components and the fabrication approach, the composite scaffold possesses a homogeneously porous structure, significantly improved mechanical properties and stable photothermal effects. The programmed scaffold can be heated to recover under near infrared irradiation in 60s. With 4 wt% Mg, the scaffold has the balanced shape fixity ratio of 93.6% and shape recovery ratio of 95.4%. The compressed composite scaffold could lift a 100 g weight under NIR light, which was more than 1700 times of its own weight. The results of the push-out tests and the finite element analysis (FEA) confirmed the tight-contacting ability of the SMPU/4 wt%Mg scaffold, which had a signficant enhancement compared to the scaffold without shape memory effects. Furthermore, The osteopromotive function of the scaffold has been demonstrated through a series of in vitro and in vivo studies. We envision this scaffold can be a clinically effective strategy for robust bone regeneration.

Recently, the near-infrared-II (NIR-II, 1000–1350 nm) region has been extensively applied in deep-tissue photothermal therapy (PTT) on account of it having stronger tissue penetration and a higher maximum permissible exposure (MPE) than the near-infrared-I (NIR-I, 650–950 nm) region. In this study, we developed a rapid and convenient in situ polymerization strategy to fabricate polypyrrole nanosheets (PPy NSs) within a few minutes using manganese dioxide nanosheets (MnO2NSs) as both the oxidant and the self-sacrificed template. The fabricated PPy NSs exhibited excellent NIR-II absorption, which conferred its high photothermal conversion efficiency (66.01%) at 1064 nm and its photoacoustic (PA) imaging capability. Both in vivo and in vitro studies have shown that that PPy NSs possess good biological safety and excellent PTT efficacy and PA imaging performances. Thus, the as-synthesized PPy NSs could effectively achieve PA imaging-guided photothermal tumor ablation under 1064 nm excitation. Our work provides a novel and promising method for the rapid preparation of PPy NSs without the addition of exogenous oxidants and subsequent template removal, which could be regarded as potential photothermal agents (PTAs) to integrate the diagnosis and treatment of cancer. Keywords:polypyrrole nanosheets;in situ polymerization;near-infrared-II region;photoacoustic imaging;photothermal therapy

Photodetectors are widely used in optoelectronic applications such as remote controlling, optical communication, and intelligent manufacturing. Multi-wavelength response capability can expand the application scenarios of a photodetector. However, responding to and recognizing multiple bands is still challenging for a photodetector. Here, we develop a dual-mode photodetector that can respond to and recognize near-infrared (NIR) and ultraviolet (UV) light with the assistance of a machine-learning model. A hybrid laser processing method was proposed to fabricate this photodetector based on the polyimide substrate. This method uses femtosecond laser and continuous wave laser to induce the formation of carbon and zinc oxide composite material in different forms. The femtosecond laser-processed area and the continuous wave laser-processed area realized stable NIR light detection and UV light detection, respectively. Recognition of the type of incident light was realized with the assistance of a machine-learning classification model. The proposed hybrid laser processing method combines the characteristics of two kinds of lasers and provides new insights into fabricating dual-mode photodetector. This photodetector could advance the development of dual-mode photodetection and has application potential in many fields like wearable devices and smart clothing.

The second near-infrared biological window two (NIR-II, 1000–1700 nm) imaging has made remarkable achievements in fluorescence imaging of diseases. Here, this work takes advantage of the rich spectral properties of rare-earth doped nanoparticles (RENPs) and their capability to present multiple emission peaks in different luminescence regions simultaneously; novel RENPs are synthesized that have emission peaks in the NIR-II 1000nm (1000–1300 nm) region and enhanced emission in the NIR-IIb (1500–1700 nm) window and are named as NEU-RENPs. These RENPs with new compositions enable a fair comparison to evaluate their imaging performance in NIR-II 1000nm and NIR-IIb regions under the same conditions. The imaging abilities of the resulted nanoparticles in NIR-II 1000 nm and NIR-IIb regions are investigated systematically for local blood vessels, tumor vessels, acute inflammatory vessels, arterial thrombosis, cerebrovascular inflammation, and brain injury models. The results indicate that imaging of NIR-IIb shows higher resolution and sensitivity than that of NIR-II 1000nm , and it has a stronger ability to accurately distinguish normal and pathological tissues. Interestingly, the fuzzy image of NIR-II 1000nm reveals more information about the background structure of the target, which is difficult to be achieved in NIR-IIb imaging. The results prove NEU-RENPs based NIR-IIb imaging is preferable for biomedical applications.

The effectiveness of phototheranostics induced immunotherapy is still hampered by limited light penetration depth, the complex immunosuppressive tumor microenvironment (TME) and the low efficiency of immunomodulator drug delivery. Herein, self-delivery and TME responsive NIR-II phototheranostic nanoadjuvants (NAs) were fabricated to suppress the growth and metastasis of melanoma through the integration of photothermal-chemodynamic therapy (PTT-CDT) and immune remodeling. The NAs were constructed by the self-assembly of ultrasmall NIR-II semiconducting polymer dots and the toll-like receptor agonist resiquimod (R848) utilizing manganese ions (Mn 2+ ) as coordination nodes. Under acidic TME, the NAs responsively disintegrated and released therapeutic components, which enable NIR-II fluorescence/photoacoustic/magnetic resonance imaging-guided tumor PTT-CDT. Moreover, the synergistic treatment of PTT-CDT could induce significant tumor immunogenic cell death and evoke highly efficacious cancer immunosurveillance . The released R848 stimulated the maturation of dendritic cells, which both amplified the antitumor immune response by modulating and remodeling the TME. The NAs present a promising integration strategy of polymer dot-metal ion coordination and immune adjuvants for precise diagnosis and amplified anti-tumor immunotherapy against deep-seated tumors. Statement of significance The efficiency of phototheranostics induced immunotherapy is still limited by insufficient light penetration depth, low immune response and the complex immunosuppressive tumor microenvironment (TME). In order to improve the efficacy of immunotherapy, self-delivery NIR-II phototheranostic nanoadjuvants (PMR NAs) were successfully fabricated via the facile coordination self-assembly of ultra-small NIR-II semiconducting polymer dots and toll-like receptor agonist resiquimod (R848) utilizing manganese ions (Mn 2+ ) as coordination nodes. PMR NAs not only enable TME responsive cargo release and NIR-II fluorescence/photoacoustic/magnetic resonance imaging mediated precise localization of tumors, but also achieve synergistic photothermal-chemodynamic therapy, evoking an effective anti-tumor immune response by ICD effect. The responsively released R848 could further amplify the efficiency of immunotherapy by reversing and remodeling the immunosuppressive tumor microenvironment, thereby effectively inhibiting tumor growth and lung metastasis.

Hepatic alveolar echinococcosis (HAE) is caused by the metacestode of Echinococcus multilocularis, which shows characteristics of malignant tumors with high mortality. However, traditional diagnostic imaging methods are still not sufficient for the recognition of HAE microlesions in the early stages. Near-infrared-II (900–1700 nm, NIR-II) fluorescence microscopic imaging (NIR-II-FMI) has shown great potential for biomedical detection. A novel type of negative target imaging method based on NIR-II-FMI with the assistance of indocyanine green (ICG) was explored. Then, NIR-II-FMI was applied to the early detection of HAE for the first time. The negative targeting NIR-II fluorescence imaging of HAE-infected mice at different stages with the assistance of ICG under 808 nm of laser irradiation was obtained. Especially, HAE microlesions at the early stage were detected clearly. Moreover, clear intrahepatic angiography was achieved under the same NIR-II-FMI system.

Surgical excision, chemotherapy, and radiotherapy are the main approaches used for treating melanoma. Unfortunately, surgical excision usually inevitably causes large area skin defects. In addition, chemotherapy and radiotherapy are often accompanied by adverse reactions and multi-drug resistance. To overcome these limitations, a near-infrared (NIR)- and pH-responsive injectable nanocomposite hydrogel was developed using sodium alginate- graft -dopamine (SD) and biomimetic polydopamine-Fe(III)-doxorubicin nanoparticles (PFD NPs) for treating melanoma and promoting skin regeneration. Firstly, the SD/PFD hydrogel can precisely deliver anti-cancer agents to the tumor site to reduce its loss and off-target toxicity. Then, PFD can convert light into heat energy under NIR irradiation to kill cancer cells. Meanwhile, doxorubicin can be administered continuously and controllably by NIR- and pH-responsive. Additionally, the SD/PFD hydrogel can also relieve tumor hypoxia by decomposing endogenous hydrogen peroxide (H 2 O 2 ) into oxygen (O 2 ). Therefore, photothermal, chemotherapy, and nanozyme synergetic therapy resulted in the tumor suppression. Specifically, the SA-based hydrogel can kill bacteria, scavenge reactive oxygen species , promote the proliferation and migration of cells, and significantly accelerate skin regeneration. Therefore, this study provides a safe and effective strategy for melanoma treatment and wound repair.

Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by joint inflammation. Macrophages accelerate the pathological process of RA through the continuous cell accumulation and secretion of cytokines. Therefore, multiscale visualization of macrophages is of great significance for the early diagnosis of RA. In this work, folate receptor-targeted Cptnc-4F nanoprobes, named FA-CF-NPs, are developed for dual-model molecular imaging of macrophages in RA mouse models. FA-CF-NPs exhibit complementary imaging advantages with high fluorescence sensitivity and resolution in the second near-infrared window (NIR-II) and promising photoacoustic (PA) imaging contrast with deep tissue penetration. The high affinity of the folate ligand on the surface of FA-CF-NPs to macrophages with overexpressed folate receptors ensures the specific targeting function of the nanoprobes for imaging in vivo. FA-CF-NPs are employed to achieve NIR-II fluorescence imaging to accurately localize macrophages in the joint, obtaining a high signal-to-background ratio (SBR) of 15.5. Meanwhile, PA imaging is also performed to observe macrophage distribution in the longitudinal section of the whole joint. Moreover, macrophage infiltration is detected in the joint of RA mice with the absence of RA pathologies such as synovitis and cartilage loss. The results provide an opportunity for in vivo macrophage tracking and early diagnosis of RA.

Harvesting heat or solar energy to directly generate affordable and sustainable electricity holds great promise toward both fundamental science and practical applications in self-powered wearable electronics. However, solar thermoelectric (STE) devices remain challenging in establishing a stable and significant temperature gradient (Δ T ) across thermoelectric materials for cost-effective electricity generation. Herein, a wearable STE generator with all-weather and high-performance electricity generation was elaborately designed and synthesized sandwiched by a thermogalvanic cell and a large-area nanofiber membrane of Cs 0.32 WO 3 (CWO) nanoparticles with strong near-infrared (NIR) absorption. These prepared CWO membranes, which were prepared via an electrostatic spinning technique, present a high photothermal conversion efficiency of 42.7 % with no degradation after running for 10 cycles. Notably, a significant Δ T of 31.5 K across the thermoelectric modular was achieved for STE generation via the thermal concentration of these photothermal membranes under natural sunlight. Owing to the high thermopower of 2.87 mV K −1 for each p-n pair, STE generation demonstrates a maximum output voltage of 200 mV under natural solar irradiance during the time period of noon. This work explores a new strategy to achieve efficient heat management of STE devices for high electricity generation under all-weather conditions, which demonstrates great potential for self-powered wearable electronics.

MoS 2 nanosheets have received a lot of favor in biomedical applications due to their outstanding near-infrared (NIR) responsiveness. However, the poor aqueous dispersibility of MoS 2 limits their biomedical application. In this paper, in-situ growth method was applied to integrate gold nanorods (AuNRs) with MoS 2 , and then MoS 2 @AuNRs/HAP/PDA (denoted as MAHP) was prepared by introducing hydroxyapatite (HAP) and polydopamine (PDA) into MoS 2 @AuNRs (MA) via self-assembly approach. In-situ grown MA inhibited the aggregation of MoS 2 due to the insertion of AuNRs between MoS 2 nanolayers. Therefore, MA revealed excellent aqueous dispersibility and synergistically enhanced NIR photothermal responsiveness (42.16% of photothermal conversion efficiency). In addition, MAHP exhibited outstanding DOX loading capacity (80%) and distinct pH-/NIR- dual responsive drug release properties, owing to the structural collapse of HAP in acidic medium and the excellent NIR responsiveness of MA. In vitro cell viability results revealed that MAHP could effectively kill MCF-7 cells under the dual effect of NIR laser irradiation and acidic medium. This paper presents a feasible process to prepare MoS 2 -based nanoparticles with NIR/pH dual-responsive release properties, which shows great potential in controlled photothermal therapy field.
