In situ covalent modification of soy protein isolate during alkaline extraction: effects of polyphenol structure and concentration on protein yield, conformation, functional properties, and emulsifying performance

Xiaofan Lv,Hao Liu,Yingtao Yu,Jinshui Wang,Benguo Liu

This study first investigated in situ polyphenol covalent modification during alkaline extraction to form SPI-polyphenol conjugates. The effects of polyphenol type (gallic acid, GA; epigallocatechin gallate, EGCG; tannic acid, TA) and concentration (0.5–5 %, w /w) on yield, structure, functionality, and emulsifying properties were evaluated. The results demonstrated that polyphenol structure significantly influenced the covalent binding efficiency and protein yield, with EGCG at 5 % showing the highest binding (89.78 mg/g) and yield (43.35 %). SDS-PAGE, spectroscopy, and amino acid analysis confirmed covalent bonding and conformational changes, with secondary structure shifting from β-sheet to α-helix, decreased fluorescence, and increased UV absorption. Conjugates exhibited improved antioxidant activity, with EGCG at 5 % reaching 99.5 % ABTS scavenging and highest reducing power. Nanoemulsions stabilized by conjugates (0.4 % w /w, oil-to-water ratio 5:95  v /v) showed 40-day stability, inhibited oil-phase peroxide formation, and enhanced lutein protection. This provides a green SPI modification strategy and insights into polyphenol-protein conjugation design.

Harnessing metal-polyphenol networks (MPNs) to regulate the digestive kinetics and enhance gel properties of whey protein gel: The influence on intermolecular interaction force and microstructure

Yafei Zhang,Zhen Zhang,Zenghui Xia,Yao Lu,Yuning Zhang,Yujia Luo,Sijia Song,Heng Quan,Yingying Lin,Yongjian Ai,Huiyuan Guo

Whey protein (WP) has significant applications in the fields of food and nutrition. However, WP has the characteristics of fast digestion rate and a wide range of different gel properties depending on processing history, which limits its application in industrial production. Therefore, in this study, proanthocyanidins and calcium ions were introduced as model polyphenols and metal ions to form a versatile novel material, metal-polyphenol networks (MPNs). MPNs with strong adhesive properties were added to whey protein isolate gel (GWPI) to form MPNs-enhanced GWPI (GWPI@MPNs). This experiment first elucidated the mechanisms by which proanthocyanidins and calcium ions form MPNs and the molecular interactions between MPNs and WP. Then, the effects of adding different proportions of MPNs on the digestion kinetics and gel properties of GWPI@MPNs were determined. The indicators of digestion rate included degree of hydrolysis, digital image of the digestion process, SDS-page, and distribution of polypeptide molecular weight. Gel properties encompassed texture properties, rheological properties, denaturation behaviours, low-field nuclear magnetic resonance, and water-holding capacity. It was found that adding an appropriate amount of MPNs was an effective method to control the digestion rate and enhance the gel properties of GWPI. The mechanism might be that MPNs could enhance the intermolecular forces of WP and make the GWPI form a dense and uniform microstructure. The novel method developed in this research has the potential to contribute to overcoming the inherent limitations of WP and adding a new digestion related functional value for its application in the food industry.

Mussel-inspired triple bionic adsorbent: Facile preparation of layered double hydroxide@polydopamine@metal-polyphenol networks and their selective adsorption of dyes in single and binary systems

Mingkun Gao,Donghui Xu,Yuhang Gao,Ge Chen,Rongqi Zhai,Xiaodong Huang,Xiaomin Xu,Jing Wang,Xin Yang,Guangyang Liu

To effectively address the serious human health challenges and ecological damage caused by organic dyes in wastewater, we developed a novel bionic adsorbent ( [email protected] @MPNs) for the selective adsorption and removal of malachite green (MG) and crystalline violet (CV). The adsorbent was prepared using a facile two-step method based on mussel-inspired chemistry and metal complexation. The physicochemical structure, surface morphology, and composition of the [email protected] @MPNs were characterized by scanning electron microscopy, Fourier-transform infrared spectrometry, X-ray photoelectron spectroscopy, and thermogravimetric analysis . Adsorption of MG and CV with the [email protected] @MPNs was evaluated. Under optimal conditions, the maximum adsorption of MG and CV by the adsorbent was 89.608 and 40.481 mg/g, respectively. The adsorption kinetics showed that the experimental data were in good agreement with the pseudo-second-order kinetic model, and the equilibrium adsorption isotherm data fitted well with the Freundlich model. The thermodynamic results indicated that the adsorption of the dyes on [email protected] @MPNs was a spontaneous endothermic process. Importantly, the bionic adsorbent not only shows high removal efficiency by easy regeneration with low-cost reagents but also exhibits high selectivity for dyes in both single and binary systems. Therefore, [email protected] @MPNs have the potential to adsorb and remove dyes from complex wastewater solutions.

Core-shell structured magnetic mesoporous carbon nanospheres derived from metal-polyphenol coordination polymer-coated Fe3O4 and its application in the enrichment of phthalates from water samples

Shan Xue,Yiyu Wang,Dan Wu,Jiwei Shen,Yinmao Wei,Chaozhan Wang

In this work, core-shell structured magnetic mesoporous carbon nanospheres were fabricated from the carbonization of metal-polyphenol coordination polymer-coated Fe 3 O 4 nanoparticles. The preparation method is simple, fast, versatile, and easy to scale up. Magnetic mesoporous carbon nanospheres exhibit a high specific surface area, high superparamagnetism, and high adsorption efficiencies for phthalates. Four phthalates were extracted from aqueous solutions by using magnetic mesoporous carbon nanospheres via magnetic solid phase extraction. Subsequent analysis was performed by using high-performance liquid chromatography with ultraviolet detection. The analytical method has good linearity in the concentration range of 1–200 ng/mL for diethyl phthalate, diisobutyl phthalate, and dicyclohexyl phthalate, and 3–200 ng/mL for dipropyl phthalate. The limits of detection were in the range of 0.10–0.62 ng/mL. Compared with previous methods, this method has a lower detection limit, wider linearity range, and faster adsorption and desorption rates. The results indicate that magnetic mesoporous carbon nanospheres are suitable for the enrichment of hydrophobic substances from aqueous solutions.

Exploring the potential mechanism of Rubus corchorifolius L. fruit polyphenol-rich extract in mitigating non-alcoholic fatty liver disease by integration of metabolomics and transcriptomics profiling

Yin Qin,Ruyan Fan,Yingxin Liu,Shuyi Qiu,Lu Wang

Nonalcoholic fatty liver disease (NAFLD), as the commonest chronic liver disease, is accompanied by liver oxidative stress and inflammatory responses. Herein, the extract obtained from Rubus corchorifolius fruits was purified and characterized for its polyphenol composition. The liver protective effect of the purified R. corchorifolius fruit extract (RCE) on mice with high-fat-diet (HFD)-induced NAFLD were investigated, and the potential mechanisms were explored through the integration of transcriptomics and metabolomics. Results showed that the polyphenolic compounds in RCE mainly included (−)-epigallocatechin, procyanidin B2, keracyanin, vanillin, dihydromyricetin, and ellagic acid. In addition, RCE intervention ameliorated liver and mitochondrial damage, which was evidenced by decreased indices of oxidative stress, liver function markers, and lipid profile levels. The liver metabonomics research revealed that RCE intervention affected the metabolic pathways of metabolites, including linoleic acid metabolism, galactose metabolism, alanine, aspartate and glutamate metabolism, retinol metabolism, glycine, serine and threonine metabolism, tryptophan metabolism, aminoacyl-tRNA biosynthesis, riboflavin metabolism, starch and sucrose metabolism, and arachidonic acid metabolism. Additionally, liver transcriptomics research indicated that pathways like fatty acid degradation, circadian rhythm, valine, leucine and isoleucine degradation, primary bile acid biosynthesis, cytokine–cytokine receptor interaction, adipocytokine signaling pathway, glutathione metabolism, lipid and atherosclerosis were significantly enriched. The transcriptomics and metabolomics analysis demonstrated that RCE intervention had significant modulatory effects on the metabolic pathways associated with glycolipid metabolism. Moreover, RT-PCR results verified that RCE intervention regulated liver mRNA levels associated with the inflammatory response. Therefore, our findings suggest that the intake of RCE might be an effective strategy to alleviate liver damage.

Investigating the induction of polyphenol biosynthesis in the cultured Cycolocarya paliurus cells and the stimulatory mechanism of co-induction with 5-aminolevulinic acid and salicylic acid

Ling Li-Juan,Wang Meng,Pan Chuan-Qing,Tang Dao-Bang,Yuan En,Zhang Yuan-Yuan,Chen Ji-Guang,Peng Da-Yong,Yin Zhong-Ping

Background: Plant cell culture technology is a potential way to produce polyphenols, however, this way is still trapped in the dilemma of low content and yield. Elicitation is regarded as one of the most effective ways to improve the output of the secondary metabolites, and therefore has attracted extensive attention. Methods: Five elicitors including 5-aminolevulinic acid (5-ALA), salicylic acid (SA), methyl jasmonate (MeJA), sodium nitroprusside (SNP) and Rhizopus Oryzae Elicitor (ROE) were used to improve the content and yield of polyphenols in the cultured Cyclocarya paliurus (C. paliurus) cells, and a co-induction technology of 5-ALA and SA was developed as a result. Meanwhile, the integrated analysis of transcriptome and metabolome was adopted to interpret the stimulation mechanism of co-induction with 5-ALA and SA.Results: Under the co-induction of 50 μM 5-ALA and SA, the content and yield of total polyphenols of the cultured cells reached 8.0 mg/g and 147.12 mg/L, respectively. The yields of cyanidin-3-O-galactoside, procyanidin B1 and catechin reached 28.83, 4.33 and 2.88 times that of the control group, respectively. It was found that expressions of TFs such as CpERF105, CpMYB10 and CpWRKY28 increased significantly, while CpMYB44 and CpTGA2 decreased. These great changes might further make the expression of CpF3′H (flavonoid 3′-monooxygenase), CpFLS (flavonol synthase), CpLAR (leucoanthocyanidin reductase), CpANS (anthocyanidin synthase) and Cp4CL (4-coumarate coenzyme A ligase) increase while CpANR (anthocyanidin reductase) and CpF3′5′H (flavonoid 3′, 5′-hydroxylase) reduce, ultimately enhancing the polyphenols accumulationConclusion: The co-induction of 5-ALA and SA can significantly promote polyphenol biosynthesis in the cultured C. paliurus cells by regulating the expression of key transcription factors and structural genes associated with polyphenol synthesis, and thus has a promising application.

Polyphenol mediated α-FeOOH/g-C3N4 heterojunction membrane for fast purification of surfactant-stabilized O/W emulsions with super-wetting and Photo-Fenton self-cleaning functions

Hongshan Jia,Yingqing Zhan,Fei Zhu,Ximin Chen,Yajie Lei,Xinyue Duan,Jie Liu

In the practical purification of oily wastewater, the application of polymer-based membrane is greatly restricted due to the oil adhesion and complicated fouling problems. Herein, to conquer the aforementioned challenges, we prepared the polyphenol mediated Photo-Fenton self-cleaning and superhydrophilic/underwater superoleophobic α-FeOOH/g-C 3 N 4 @PVDF composite membrane via simple vacuum-assisted self-assembly technology. The g-C 3 N 4 nanosheets modified by tannic acid were introduced into the surface of PVDF ultrafiltration membrane, in which the intercalation structure was regulated by the random interpenetration of α-FeOOH nanorods . The abundant hydroxyl and amino groups and the rough micro-nano structure formed by intercalation structure were favorable for the superhydrophilic feature (WCA = 0°, UOCA>150°), thus resulting in ultra-low oil adhesion. Specially, the intercalation structure improved the water permeation channel, further promoting the water/oil separation flux. Therefore, the composite membrane exhibited superior separation flux (452.35–650.21 L m −2  h −1 ) and separation efficiency (>99.06 %) for various surfactant-stabilized emulsions. Moreover, thanks to the Photo-Fenton reaction, the composite membrane showed excellent degradation efficiency on simulated pollutants (various dyes) within 60 min (MeB: 95.55 %, CR: 90.83 %, MO: 98.90 %, CV: 95.11 %), showing satisfactory photocatalytic self-cleaning performance. This work provides a strategy for the potential application of polymer-based membranes in the treatment of complex surfactant-stabilized emulsion wastewater from petrochemical industry.

Self-assembly of tea polyphenol onto the high strength chitosan film fabricated via a new way to enhance the antioxidant and antibacterial activity for meat preservation

Yuntao Wang,Xiuqin Guo,Zhilong Qin,Rongqin Liu,Shengjie Li,Manting Du,Si Qiu,Yanhong Bai

Chitosan (CS) films traditionally prepared from acid have poor mechanical properties, thus limiting their application as packaging materials. In this study, high strength chitosan-tea polyphenol (CS-TP) films were fabricated via a new way. Firstly, high strength CS film was prepared by regeneration of CS dissolved in alkali from hot water, then TP was incorporated into CS film through the one step self-assembly method. It was found that large amount of TP was adsorbed onto CS film due to the strong hydrogen bonding and hydrophobic interaction between CS and TP confirmed by results of adsorption tests, FTIR and XRD. Moreover, the mechanical, anti-UV, antibacterial and anti-oxidant properties of the CS-TP films gradually enhanced with increasing content of TP. The water vapor permeability rate of CS-TP films slightly increased with increasing content of TP. Pork perseveration results indicated that the CS-TP films lowered the pH, slowed the growth of bacterial and change of color, decreased the lipid oxidation products content of fresh pork compared with CS film or the control sample, thus prolonging the shelf life of fresh pork for 2 days. Therefore, this study proposed a new strategy for preparing high strength muti-functional CS-TP films as food packaging materials.

Oxidized sodium alginate/carboxymethyl chitosan/γ-polyglutamic acid injectable hydrogels enabling pH-responsive synergistic delivery of antimicrobial agents and metal-polyphenol nanoparticles for diabetic foot ulcer healing

Guixi Wang,Sihang Shen,LiDong Wang,YiChenTian,Chao Li,Junnan He,Yuzhu Song

Chronic diabetic wounds have become a major global health challenge due to the vicious cycle driven by infection, oxidative stress, and inflammation. To address this issue, researchers developed a pH-responsive injectable hydrogel (SCP@QCNPs&DOX). The hydrogel is composed of oxidized sodium alginate, dihydrazide-modified γ-polyglutamic acid, and carboxymethyl chitosan, and is co-loaded with quercetin nanoparticles (QCNPs) and doxycycline. The quercetin nanoparticles are prepared via coordination between quercetin and cerium, which not only overcomes the inherent poor water solubility of quercetin but also endows them with antioxidant, anti-inflammatory, and enzyme-mimetic catalytic activities. The SCP@QCNPs&DOX hydrogel exhibits excellent biodegradability, biocompatibility, and stable rheological properties, enabling controlled release of quercetin nanoparticles and doxycycline. The hydrogel alleviates oxidative stress by scavenging reactive oxygen species (ROS) and maintaining mitochondrial membrane potential, inhibits inflammation by modulating the NF-κB pathway and inducing M2 macrophage polarization, and demonstrates potent antibacterial activity against Staphylococcus aureus and methicillin-resistant S. aureus (MRSA). In a diabetic rat model with MRSA infection, the hydrogel significantly suppresses inflammation, exerts antibacterial effects, promotes angiogenesis, and thereby accelerates wound healing. In summary, the SCP@QCNPs&DOX hydrogel represents a promising multifunctional platform for the treatment of infected diabetic wounds. Binary probiotic fermentation promotes signal (cyclic AMP) exchange to increases the number of viable probiotics, anthocyanins and polyphenol content, and the odor scores of wolfberry fermented beverages

Xiankang Fan,Mingzhen Liu,Zihang Shi,Tao Zhang,Lihui Du,Zhen Wu,Xiaoqun Zeng,Xiang Wu,Daodong Pan

The effects and underlying molecular mechanisms of binary probiotics ( Lactiplantibacillus plantarum subsp. plantarum CGMCC 1.5953 and Lacticaseibacillus casei CGMCC 1.5956) on the quality of wolfberry fermented beverages (WFB) were investigated. The results indicated that binary probiotics increased the number of probiotics, anthocyanin (89.92 ± 1.64 mg/L), polyphenol content (283.04 ± 3.81 µg/mL), and odor score (24.19) in WFB. Metabolomics found that they could enhance signal exchange (cyclic AMP) between binary probiotics and improve the utilization of citrulline, d -proline, d -glucose, and d -galactose through galactose metabolism and amino acid biosynthesis pathway to promote probiotics growth. Furthermore, HS-SPME-GC–MS and GS-IMS revealed that the improvement in flavor was mainly due to an increase in the content of the aromatic flavor substances 3-heptanol, glutaraldehyde, and 2-heptanone, and a decrease in the content of the off-flavor substances methyl isobutyl ketone-D and 2-undecanone. This is strategically important for the development of WFB with high probiotic content and unique flavor.

N-doped graphitic carbon shell-encapsulated FeCo alloy derived from metal–polyphenol network and melamine sponge for oxygen reduction, oxygen evolution, and hydrogen evolution reactions in alkaline media

Hu Liu,Dong-Hui Yang,Xu-Yun Wang,Jiangwei Zhang,Bao-Hang Han

Exploiting low cost and durable electrocatalysts with high efficiency for oxygen reduction reaction (ORR), oxygen evolution reaction (OER), and hydrogen evolution reaction (HER) is of great significance for energy conversion and storage applications. Herein, a hybrid electrocatalyst of FeCo alloy nanoparticles embedded in a porous N -doped carbon was prepared via a pyrolysis process of low-cost melamine sponge and mass-produced metal–polyphenol network. Benefting from the metal coordination of metal–polyphenol network and abundant N source of melamine sponge, the metal–N moiety and FeCo alloy nanoparticles (wtih a diameter around 50 nm) encapsulated in a N -doped graphene-like carbon layer were formed in-situ. Such intimate integration of graphene-like carbon-encapsulated FeCo alloys, metal–N active species, and porous structure is conducive to improve the catalytic activity and increase the catalytic durability in alkaline media. As a consequence, the as-prepared electrocatalyst exhibits the pronounced activity toward ORR, OER, and HER simultaneously under alkaline condition, particularly on the performances of potential, stability, and methanol tolerance.

Effects of alkaline water-assisted tea polyphenol soaking on the physicochemical, biochemical, bacterial structure, and free amino acid composition of grass carp fillets during storage at 4 °C

Yilin Lin,Xie Xuan,Kabore Manegdebwaoga Arthur Fabrice,Yigang Yu,Lihua Huang,Yehui Zhang

High concentrations of tea polyphenols (TP) negatively affect the color and texture of fish. This study aimed to investigate the effects of alkaline electrolyzed water (AEW) pretreatment combined with low-concentration TP immersion on the physicochemical, biochemical, bacterial structure, and free amino acid composition of grass carp fillets. The results showed that AWE-treated with 5 min (W) and immersion in 0.1 mg/mL TP (T) significantly increased hardness, springiness, and water-holding capacity (WHC), and the initial total viable count (TVC) decreased by 1.11 lg CFU/g and 0.36 lg CFU/g, respectively. Compared with the individual treatments (W or T), the combined treatment of W and T (WT) effectively reduced lipid oxidation and maintained the TVC (5.92 lg CFU/g) and total volatile basic nitrogen (15.56 mg/100 g) within acceptable limits for up to storage for 12 d. The W treatment significantly increased protein solubility and surface hydrophobicity, which is beneficial for promoting interactions between proteins and TP, improving the thermal stability and storage stability of fillets. An increase of Lys, Arg, and Phe concentration in WT treatment could promote a more compact microstructure of MPs. The WT treatment reduced the synergistic effects of Pseudomonas and Acinetobacter and delayed the spoilage of fish fillets. Furthermore, the WT treatment effectively inhibited the abundance of Brochothrix and Serratia , delaying MPs degradation and maintaining the freshness of fillets.
