Background: Carbohydrate antigen 15-3 (CA15-3), a specific breast cancer-related biomarker in serum, can provide direct information to monitor a patient's postoperative status and can predict recurrence and metastasis of breast cancer. The conventional Enzyme-Linked Immunosorbent Assay
(ELISA) is an effective approach for CA15-3 detection, but there is a high limit of detection which is more than 1 U mL-1. Therefore, a convenient, highly sensitive and specific CA15-3 detection method is needed for earlier prognoses of breast cancer. Objective: In this work, we aim to
develop a novel sensitive and selective immunofluorescence assay for the highly effective detection of CA15-3. Method: We pre-coat the antibody I onto the solid phase to capture antigen CA15-3. The A10254- labeled antibody II identified by electrophoresis and fluorescence spectra recognized
the antigen CA15-3 and formed the sandwich format. The fluorescence signal of A10254 directly relating to the amount of antigen could be detected and quantified by the microplate reader excited at 440nm. Results: The obtained method for CA15-3 detection showed lower limits of detection
(0.11 and 0.18 U mL-1) in a linear range of 0.25-14 U mL-1 in the buffer and 2% serum, respectively. This assay exhibited a good specificity and reproducibility in 2% serum with lower relative standard deviations in different tested concentrations. Conclusion: The developed immunofluorescence
assay enhanced the performance of the conventional CA15-3 ELISA kit. Moreover, the acceptable accuracy and good reproducibility in diluted serum indicated the assay has great potential for the diagnosis of breast cancer in clinical practice.

A novel oxalate-based piezoelectric crystal, NaBaAl(C2O4)3·5H2O, was successfully synthesized and structurally characterized. The compound crystallizes in the polar space group Pca21, forming a robust three-dimensional framework constructed from [Al(C2O4)3]3– units stabilized by Na+, Ba2+, and coordinated water molecules. Optical diffuse reflectance spectroscopy combined with Tauc’s analysis indicates a direct bandgap of 4.53 eV, while polarized light microscopy measurements yield an experimental moderate birefringence of 0.044, confirming significant optical anisotropy. More notably, piezoelectric measurements reveal a relatively large coefficient of 67.9 pC/N at room temperature, representing an order-of-magnitude enhancement compared with previously reported Al-based piezoelectric crystals. Furthermore, variable-temperature measurements demonstrate the evolution of the piezoelectric response with heating. In addition, PFM measurements confirm polarization switching under applied bias, supporting the piezoelectric assignment. Collectively, these results expand the family of Al-based piezoelectrics and highlight the potential of ligand-engineered frameworks as a viable strategy for developing high-performance piezoelectric materials.

3-Hydroxy-2-butanone (3H2B) is a key gaseous metabolite of Listeria monocytogenes, a potential lung cancer biomarker, and a widely used food flavoring agent, underscoring the demand for real-time monitoring solutions. In this study, we designed a salicylaldehyde-functionalized 1,8-naphthalimide derivative (NI-OH-CHO) and reacted it with 1,3,5-tri(4-aminophenyl)benzene (TAPB) at a liquid/liquid interface to synthesize fluorescent NI-TAPB nanofilms. Upon deprotonation, the nanofilms (NI-TAPB@F) exhibit a reversible and unprecedented fluorescence response to 3H2B. Leveraging this discovery, we developed a fluorescent 3H2B sensor with exceptional performance: ultrahigh sensitivity (detection limit: ∼ 1.8 ppb), rapid response (<8 s), outstanding selectivity, and remarkable stability at room temperature. These advantages stem from the rational design of binding sites and the porous structure of the nanofilms. Furthermore, visual 3H2B detection was achieved using the deprotonated films. Notably, the nanofilms are self-standing, self-adhesive, and flexible, enabling substrate-independent detection by eliminating interference from background color or emission. These properties also facilitate on-site qualitative and quantitative analysis. We anticipate this work will pave the way for the development of portable 3H2B detection devices.

Pyroptosis is a newly discovered form of cell death. Peroxiredoxin 3 (PRX3) plays a crucial role in scavenging reactive oxygen species (ROS), but its hepatoprotective capacity in acetaminophen (APAP)-induced liver disease remains unclear. The aim of this study was to assess the role of PRX3 in the regulation of pyroptosis during APAP-mediated hepatotoxicity. We demonstrated that pyroptosis occurs in APAP-induced liver injury accompanied by intense oxidative stress and inflammation, and liver specific PRX3 silencing aggravated the initiation of pyroptosis and liver injury after APAP intervention. Notably, excessive mitochondrial ROS (mtROS) was observed to trigger pyroptosis by activating the NLRP3 inflammasome, which was ameliorated by Mito-TEMPO treatment, indicating that the anti-pyroptotic role of PRX3 relies on its powerful ability to regulate mtROS. Overall, PRX3 regulates NLRP3-dependent pyroptosis in APAP-induced liver injury by targeting mitochondrial oxidative stress.

Background Osteoarthritis (OA) is a degenerative disease characterized by chronic inflammation. Indole-3-aldehyde (3-IAld) is a tryptophan metabolite secreted by intestinal flora, which can exert anti-inflammatory effects in multiple inflammatory diseases. However, the potential therapeutic role of 3-IAld in OA and the underlying mechanism remain to be explored. Methods IL-1β was utilized to induce chondrocytes inflammation. Then, cell counting kit-8 was carried out to assess the cytotoxicity of 3-IAld on rat chondrocytes viability. Meanwhile, RT-qPCR, Western blot, and immunofluorescence were performed to evaluate the expression of inflammatory factors, matrix-degrading enzymes and matrix synthesis protein, and the NF-κB pathway in chondrocytes treated with IL-1β alone, with 3-IAld or with siRNA-AhR. Results Our results showed that 3-IAld did not affect cellular viability at concentrations up to 50 μM. 3-IAld significantly inhibited the expression of pro-inflammatory cytokines (IL-6, iNOS and COX-2), and matrix-degrading enzymes (MMP3, MMP13 and ADAMTS5), upregulated the expression of matrix synthesis protein (aggrecan and collagen-II), and inactivated the NF-κB pathway in IL-1β-treated chondrocytes. However, AhR knockdown could totally abolish the aforementioned therapeutic capabilities and the inactivation of the NF-κB pathway induced by 3-IAld. Conclusions 3-IAld reduced inflammation through the AhR-NF-κB signalling pathway in IL-1β-induced chondrocytes, which is expected to provide a new therapeutic strategy for OA.

The temperature dependent photoluminescence of temperature sensitive paint (TSP) makes it an attractive candidate for infrared (IR) thermal imaging, but its low luminescence intensity and poor photostability hinder its extensive use in infrared detectors . Herein, a novel complex luminophore of AgNPs/[Ru(phen) 3 ]Cl 2 /PVDF was designed to fabricate a thermal imaging device with improved sensitivity and signal-to-noise ratio. It is found that the localized surface plasmon resonance (LSPR) of silver nanoparticles (AgNPs) could enhance the local intensity of the excitation light , consequently an increase in the luminescence intensity of neighboring luminophore can be achieved. Thus, not only the fluorescence intensity of the obtained sensor array is enhanced, but the temperature sensitivity is also improved to − 4.5%/K at 20 °C, which increased by 25% compared to that of the blank film without AgNPs addition. Furthermore, the detector array is used for IR thermal imaging, which can successfully obtain the thermal images of observation object at room temperature . The minimal detectable temperature is as small as about 313 K.

Introduction Numerous options for treatment of glioblastoma have been explored; however, single-drug therapies and poor targeting have failed to provide effective drugs. Chemotherapy has significant antitumor effect, but the efficacy of single-drug therapies in the clinic is limited over a long period of time. Thus, novel therapeutic approaches are necessary to address these critical issues. Objectives The present study, we investigated a tumor-specific metal-tea polyphenol-based cascade nanoreactor for chemodynamic therapy-enhanced chemotherapy. Methods HA-EGCG was synthesized for the first time by introducing epigallocatechin-3-gallate (EGCG) into the skeleton of hyaluronic acid (HA) with reducible disulfide bonds. A rapid and green method was developed to fabricate the metal-tea polyphenol networks (MTP) with an HA-EGCG coating ( [email protected] /HA-EGCG) based on Fe 3+ and EGCG for targeted delivery of doxorubicin hydrochloride (DOX). GL261 cells were used to evaluate the antitumor efficacy of the [email protected] /HA-EGCG nanoreactor in vitro and in vivo . Results [email protected] /HA-EGCG nanoreactors were able to disassemble, resulting in escape of their components from lysosomes and precise release of DOX, Fe 3+ , and EGCG in the tumor cells. HA-EGCG depleted glutathione to amplify oxidative stress and enhance chemodynamic therapy. The results of in vivo experiments suggested that [email protected] /HA-EGCG specifically accumulates at the CD44-overexpressing GL261 tumor sites and that sustained release of DOX and Fe 3+ induced a distinct therapeutic outcome. Conclusions The findings suggested the developed nanoreactor has promising potential as a future GL261 glioblastoma therapy.

Numerous therapeutic agents and strategies were designed targeting the therapies of Alzheimer’s disease, but many have been suspended due to their severe clinical side effects (such as encephalopathy) on patients. The attractiveness for small molecules with good biocompatibility is therefore restarted. Epigallocatechin-3-gallate (EGCG), extracted from green tea, is expected to be a promising small-molecule drug candidate, which can remodel the structure of preformed β-sheet-rich oligomers/fibrils and then effectively interfere with neurodegenerative processes. However, as the structure of non-fibrillary aggregates cannot be directly characterized, the atomic details of the underlying inhibitory and destructive mechanisms still remain elusive to date. Here, all-atom molecular dynamics simulations and experiments were carried out to elucidate the EGCG-induced remodeling mechanism of amyloid β (Aβ) fibrils. We showed that EGCG was indeed an effective Aβ fibril inhibitor. EGCG was capable of mediating conformational rearrangement of Aβ1–42 fibrils (from a β-sheet to a random coil structure) and triggering the disintegration of fibrils in a dose-dependent manner. EGCG redirected the structure of Aβ by breaking the β-sheet structure and hydrogen bonds between peptide chains within the Aβ protofibrils, especially the parallel β-strand (L17VFFAEDVGS26). Moreover, reduced solvent exposure and multisite binding patterns all tended to induce the conformation conversion of Aβ17–42 pentameric protofibrils, destroying pre-formed fibrils and inhibiting continued fibril growth. Detailed data analysis revealed that structural features of EGCG with abundant benzene ring and phenolic hydroxyl moieties preferentially interact with the parallel β-strands to effectually hinder the interaction of the interpeptide chain and the growth of the ordered β-sheet structure. Furthermore, experimental studies confirmed that EGCG was able to disaggregate the preformed fibrils and alter the protein structure. This study will enable a deeper understanding of fundamental principles for design of structural-based inhibitors.

Four novel betulinic acid (BA) derivatives modified at the C-3 position were prepared by the Mitsunobu reaction and subsequently evaluated for antitumor activity against four common cancer cell lines in vitro. The results from the MTT assay revealed that the four tested compounds showed remarkable cytotoxic activity against four cancer cell lines, much better than BA. Western blotting results indicated that the four compounds, especially compound 2c , caused a significant up-regulation expression of the caspase-12 in CT26 cell. Our results showed that configuration inversions of chiral C-3 of BA could also produce BA derivatives with potent antitumor activity.

In this paper, LiTi 2 (PO 4 ) 3 @N-doped carbon anode has been synthesized by in situ carbon coating approach. The well-proportioned N-doped carbon layer and loose nanoporous structure was obtained by using urea as nitrogen source and pore former. LiTi 2 (PO 4 ) 3 @N-doped carbon as anode demonstrates much better rate capability than LiTi 2 (PO 4 ) 3 @carbon in ALIBs. The optimized anode delivers the discharge capacity of 93.7 mAh g −1 and 74.2 mAh g −1 at rates of 10C and 20C, 22.5 mAh g −1 and 50.0 mAh g −1 larger than that of LiTi 2 (PO 4 ) 3 @carbon. Moreover, LiTi 2 (PO 4 ) 3 @N-doped carbon exhibits excellent cycling performance with capacity retention of 84.3% at 5C after 1000 cycles. As verified, the well-proportioned N-doped carbon layer could reduce charge transfer resistance and improve electrical conductivity. The loose nanoporous structure could shorten pathway and facilitate diffusion for Li ion. Therefore, LiTi 2 (PO 4 ) 3 @N-doped carbon gets the superior electrochemical properties benefiting from those two characteristics.

To synthesize a series of 3-, 4-, and/or 11-trihydroxy modified bergenin derivatives and evaluated their cytotoxic activity in vitro . The phenolic hydroxyl groups of bergenin were protected by benzyl groups with benzyl bromide. Treatment of dibenzyl bergenin with the corresponding acid in the presence of EDC·HCl and DMAP in CH 2 Cl 2 , followed by hydrogenation over Pd/C catalysts, afforded derivatives of bergenin esters. All of the target compounds were identified by IR, MS, and 1 H NMR. Twenty-six novel and three known derivatives of bergenin esters were synthesized. Their cytotoxicity values were evaluated by the MTT assay on the inhibition of DU-145 and BGC-823 cells in vitro . Several triply-substituted ( 3a, 4a, 5a, 6a, 7a ) and doubly-substituted ( 8b, 9b ) bergenin derivatives exhibited higher cytotoxic activity than bergenin. The result showed that the size of substituents and the lipophilicity of the bergenin esters displayed an important role on their cytotoxic activity.

Omega-3 polyunsaturated fatty acids (n-3 FAs) are essential nutrients and are considered effective in improving human health. Recent studies highlight the importance of the combination of n-3 FAs and polyphenols for limiting the oxidation of n-3 FAs and exhibiting synergistic beneficial effects. Herein, we developed a novel formulation technology to prepare oleogels that could be used for the codelivery of n-3 FAs and polyphenols with high loading efficacy and oxidative stability. These oleogels are made from algal oil with polyphenol-enriched whey protein microgel (WPM) particles as gelling agents via simple and scalable ball milling technology. The oxidative status, fatty acid composition, and volatiles of protein oleogels during accelerated storage were systematically assessed by stoichiometry and gas chromatography–mass spectrometry. These results showed that protein oleogels could overcome several challenges associated with the formulation of n-3 oils, including long-term oxidative stability and improved sensory and textural properties. The protein oleogel system could provide an excellent convenience for formulating multiple nutrients and nutraceuticals with integrating health effects, which are expected to be used in the care of highly vulnerable populations, including children, the elderly, and patients.
