pH-responsive collagen nanocomposite films reinforced by curcumin-loaded Laponite nanoplatelets for dynamic visualization of shrimp freshness

Jiabo Shi,Li Sheng,Meng Chen,Xiaoyi Wu,Xiaojie Li,Yan Qin Tan

Sustainable and intelligent packaging systems, utilizing biomass-derived matrix and bioactive indicators represent promising approaches for real-time detection and monitoring of freshness of food, as well as prolonging the shelf-life and improving the quality and safety of food products. Herein, we reported pH-responsive collagen nanocomposite films reinforced by curcumin-loaded Laponite nanoplatelets which could act as smart indicators for monitoring the freshness of shrimps. Caffeic acid, as a bio-based modifier, was covalently grafted onto the surfaces of the nanoplatelets loaded with curcumin . The collagen nanocomposite films were fabricated by the incorporation of functionalized nanoplatelets through all-aqueous casting-evaporation approach. The triple-helical structure of collagen molecules was stabilized by the functionalized nanoplatelets mainly through hydrogen bonding and electrostatic interactions with uniform distribution into the film matrix. Moreover, the resultant nanocomposite films not only demonstrated favorable hydrophobicity and water vapor barrier properties, but also exhibited effective antioxidant activity and anticipated color responsiveness to pH changes and NH 3 presence. The consistent changes in color of the nanocomposite film with the shrimps following days of storage supported its feasibility and potential for monitoring the freshness of shrimps. Therefore, we envision that the developed collagen-based nanocomposite films showcase great potential in the application of sustainable and intelligent packaging system for dynamic visualization of seafood freshness.

pH-responsive hydrogel containing curcumin-loaded lipid nanocapsules for oral curcumin’s stability, bioaccessibility and intestinal absorption improvement

Runliang Feng,Mingzhu Wang,Zhimei Song,Yunjing Jia,Yuli Li,Qingmin Liu,Fuyou Wang,Guangxi Zhai

A liquid formulation with suitable consistency and sustained release characteristics can improve compliance in users with dysphagia. In the study, we prepared curcumin-loaded lipid nanocapsules (Cur-LNCs) and incorporated it in a mixture of sodium alginate, dextrin, calcium chloride and sodium citrate to form a pH-responsive pre-hydrogel solution (SA-DEX:Cur-LNCs). The SA-DEX:Cur-LNCs exhibited a pH-responsive solution-gel conversion: solubilized at near neutral (pH = 7) and rapidly gelatinized to a strong elastic gel at stomach acid (pH = 2.5). The SA-DEX:Cur-LNCs could effectively improve the thermal and pH stability of Cur, and maintain its antioxidant effect. Interestingly, the bioaccessibility of SA-DEX:Cur-LNCs in gastrointestinal was significantly increased to 37.00% when compared with free Cur. The SA-DEX:Cur-LNCs also significantly improved the small intestinal absorption of Cur, being 16 times higher than that of free Cur. Therefore, incorporating lipid nanocapsule in pH-responsive pre-hydrogel provided a new oral administration platform of hydrophobic bioactive compounds for compliance improvement of users with dysphagia.

Photo-crosslinking methacrylated-amylopectin/polyacrylamide hydrogels loading curcumin for applications as degradable, injectable, and antibacterial wound dressings

Fang Zhu,Yanru Hu,Lihui Meng,Wenchao Li,Bin Xie,Zilin Zhou,Shuojie Cui,Meng Wang,Youfa Wang,Zebin Chen,Qingzhi Wu

The preparation of biodegradable and antibacterial hydrogels has important clinical value. In this work, a novel strategy has been developed to prepare degradable hydrogel dressings without chemical crosslinking agent using methacrylate anhydride (MA)-modified amylopectin (APMA) and polyacrylamide (PAM). After introducing C C bonds, APMA/PAM hydrogels can be formed under light irradiation. This strategy improves the gelling ability of AP and degradation properties of the hydrogel by avoiding the addition of crosslinking agent. The degradation rate of APMA/PAM hydrogel is 74.04 ± 0.69 % within 12 weeks, while that of APMA/PAM hydrogel containing crosslinking agent is only 38.5 ± 0.1 %. The APMA/PAM hydrogel loading curcumin (Cur) (APMA/PAM-Cur) exhibits high antibacterial efficiency of 98.29 ± 0.41 % and 97.18 ± 0.81 % against S. aureus and E. coli , respectively, with light irradiation. Animal experiments show that the APMA/PAM-Cur hydrogel reduces the infiltration of inflammatory factors, increases the density of collagen, and makes the newly formed granulation tissue thicker and tighter. This study not only proves the promising potential of the APMA/PAM-Cur hydrogel as degradable and antibacterial wound dressing for clinical treatment, but also provides a new strategy for developing low-cost, degradable, and antibacterial wound dressings and reducing antibiotic abuse and environmental pollution caused by medical waste.

Spectral analysis of the impact of various polysaccharides on the entrapment of curcumin by whey protein isolate

Yuhan Yan,Rui Tang,Feng Li,Liurong Huang,Yu Chen,Ronghai He

This study employed spectroscopic methods to investigate the interactions between whey protein isolate/polysaccharide (WPI/PS) complexes and curcumin. The UV–visible absorption spectra and fluorescence spectra indicated effective binding of curcumin to WPI and its complexes with chitosan (CS), carrageenan (CAR) and carboxymethylcellulose (CMC). This binding significantly increased the UV–visible absorption intensity of curcumin, with its maximum fluorescence emission peak shifting from 552 nm to 510 nm. Fluorescence kinetics analysis suggested that the binding constant between curcumin and WPI/CS complexes reached a maximum of 4.0794 × 10 5  L/mol. Circular dichroism and infrared spectroscopy indicated that the higher binding constant was attributed to the binding of CS, which reduced the α-helix structure and exposed hydrophobic groups of WPI. Curcumin entrapped by WPI/CS complexes showed the highest DPPH radical scavenging activities, UV and thermal stability. In summary, the WPI/CS complexes effectively protect curcumin against various environmental stresses and maintain its antioxidant properties.

Uncovering Cocrystal Formation and Competition Mechanism of Polyhydroxy Natural Products: Cases of Quercetin, Hesperidin, Resveratrol, and Curcumin

Detao Li,Wenqian Chen,Limin Zhou,Li Xu,Feiqiang He,Jerry Y Y Heng,Hamza Shehzad,Jinbo Ouyang

Understanding drug cocrystal formation and its competitive mechanism is important for improving the physicochemical properties of drugs. In this article, the cocrystal formation of polyhydroxy natural products and their competitive mechanisms are discussed through experimental screening and theoretical calculations. The formation conditions and types of cocrystals were determined based on efficient screening experiments of polyhydroxy natural product cocrystals. To investigate the difficulty and stability of cocrystal formation, competitive cocrystal experiments of polyhydroxy natural products were carried out simultaneously. By simulation of the electrostatic potential surface of molecules and the Hirshfeld surface of crystals, the mode of hydrogen bonding within the crystals was determined. The strength of hydrogen bonding was assessed based on the intermolecular interactions and energy framework within the crystal structure. A “substitution model” was constructed based on experimental results and theoretical calculations, which is of great significance for the prediction and design of natural product cocrystals and provides a feasible solution for improving the physicochemical properties of drugs.

Characterization of zein / hyaluronic acid complex colloidal nanoparticles stabilized Pickering emulsions as a delivery vehicle of curcumin

Di Wu,Yang Lu

In this work complex nanoparticles composed of zein and hyaluronic acid (HA), referred to as ZHPs, were fabricated through the combined effects of hydrogen bonding and electrostatic attractions. Compared to zein particles, ZHPs exhibited a larger mean particle size and a reversal in zeta potential from positive to negative. The three-phase contact angle ( θ o/w ) of ZHPs with a 10:3 ratio was measured at 88.9°, indicating their suitability for stabilizing Pickering emulsions. Both droplet diameter and the emulsified phase volume fraction of the emulsions were significantly affected by the concentration of ZHPs and the oil phase ratio. Confocal laser scanning microscopy revealed that ZHPs adsorbed on the oil-water interface formed a compact interfacial layer. Rheological analysis showed that emulsions displayed shear-thinning behavior and exhibited characteristics of an elastic gel network. Moreover, the Pickering emulsions stabilized by ZHPs markedly suppressed curcumin degradation under UV exposure compared to both bulk oil and conventional emulsions. Results from vitro digestion indicated that the release of free fatty acids (FFAs) and the bioaccessibility of curcumin were enhanced in ZHP-stabilized emulsions relative to bulk oil. Overall, the findings support the potential of ZHP-stabilized Pickering emulsions as efficient delivery vehicles for food and pharmaceutical applications.

Design of a self-assembled curcumin nanoemulsion and construction of a micropellet delivery system for enhanced stability

Li Wang,Jianlu Qu,Zixuan Yan,Cunhao Li,Xingchu Gong,Wenlong Li

Curcumin (CUR) exhibits broad pharmacological activities, including anti-inflammatory, antibacterial, and antioxidant effects, and shows promising therapeutic potential in diseases such as diabetes and liver injury. However, its poor water solubility and low stability significantly limit clinical application. Conventional nanoemulsion solidification techniques, such as spray-drying and lyophilization, often involve high energy input, drug degradation, and poor redispersibility, making it challenging to achieve both stability and performance. In this study, we developed a novel strategy combining design-of-experiments (DoE) optimized curcumin nanoemulsions (CUR-NE) with fluidized bed coating to systematically enhance solubility and stability. Plackett-Burman design was used to screen key variables, and Box-Behnken design further optimized these factors, revealing that formulation parameters exert a more pronounced effect on particle size and polydispersity index (PDI) than process variables. The optimized CUR-NE exhibited an extremely small droplet size (Z-Ave = 13.48 ± 0.20 nanometers) and uniform distribution (PDI = 0.064 ± 0.013), and maintained a low redispersibility index (109.95 ± 1.18 percent) when coated onto pellets (CUR-Ps). Structural characterization using FT-IR and XRD confirmed negligible drug and excipient interactions and partial amorphization of CUR. Unlike conventional solidification methods, fluidized bed coating significantly improved the storage stability of the nanoemulsion while preserving its excellent solubility. This approach not only provides a new technical pathway for the development of curcumin delivery systems but also offers an innovative reference for the industrial production and application of poorly soluble drugs.

Stable emulsion produced by thermal modified coconut (Cocos nucifera L.) globulins-xanthan gum for protection of curcumin

Jingrong Ma,Rongrong He,Yuan Chai,Xiaoshan Long,WenZheng Shi,Haiming Chen,Chuang Pan,Yongqiang Zhao

In order to explore the potential applications of thermal modified (90 and 120 °C, 20 min) coconut globulins (CG) in encapsulation systems, the long-term emulsion stability was improved by adding xanthan gum (XG). Subsequently, the protection effect of thermal modified CG-XG emulsion on curcumin (Cur) was demonstrated. The results showed that the XG decreased the interfacial adsorption of heated-CG. When 0.1 % XG was added, the K r value and interfacial viscoelasticity of 90 °C-CG were significantly increased. However, when the concentration of XG increased to 0.2 %, the K r value and interfacial viscoelasticity showed significant decrease. The interfacial viscoelasticity of 120 °C-CG decreased with the increase of XG. However, the emulsion prepared by 120 °C-CG-XG had a higher viscosity and a more compacted three-dimensional gel network structure, which led to the higher long-term stability. The heated-CG-XG emulsion system showed an excellent encapsulation effect on Cur, especially for the 120 °C-CG-0.2XG encapsulation system, which could improve the bioaccessibility of Cur from 18.21 % to 36.23 % ( p  < 0.05). Our work indicated that the 120 °C-CG emulsion system have the potential use in Cur encapsulation delivery, which is of great significance for the commercial application of thermal modified CG.

Polyvinylpyrrolidone-curcumin nanoparticles with immune regulatory and metabolism regulatory effects for the treatment of experimental autoimmune uveitis

Fan Cao,Kun Liang,Wei-Wei Tang,Qin-Yu Ni,Zhi-Yu Ji,Chen-Kai Zha,Ya-Kun Wang,Zheng-Xuan Jiang,Shengping Hou,Li-Ming Tao,Xianwen Wang

Uveitis comprises a cluster of intraocular inflammatory disorders characterized by uncontrolled autoimmune responses and excessive oxidative stress leading to vision loss worldwide. In the present study, curcumin (CUR) was conjugated with polyvinylpyrrolidone (PVP) to form PVP-CUR nanoparticles with significantly elevated solubility and outstanding multiple radical scavenging abilities. In vitro studies revealed that PVP-CUR nanoparticles markedly mitigated oxidative stress and reduced apoptosis in a H 2 O 2 -induced human retinal pigment epithelial cell line (ARPE-19) and promoted phenotypic polarization from M1 to M2 in an LPS-induced human microglial cell line (HMC3). Further in vivo studies demonstrated the prominent therapeutic effects of PVP-CUR nanoparticles on experimental autoimmune uveitis (EAU), which relieved clinical and pathological progression, improved perfusion and tomographic manifestations of retinal vessels, and reduced blood–retinal barrier (BRB) leakage; these effects may be mediated by mitigating oxidative stress and attenuating macrophage/microglia-elicited inflammation. Notably, treatment with PVP-CUR nanoparticles was shown to regulate metabolite alterations in EAU rats, providing novel insights into the underlying mechanisms involved. Additionally, the PVP-CUR nanoparticles showed great biocompatibility in vivo . In summary, our study revealed that PVP-CUR nanoparticles may serve as effective and safe nanodrugs for treating uveitis and other oxidative stress- and inflammation-related diseases.

Phenylboronic acid functionalized dextran loading curcumin as nano-therapeutics for promoting the bacteria-infected diabetic wound healing

Sheng Ni,Kun Zhang,Xiong Zhao,Shuai Wu,Meng Yan,Da Sun,Li Zhu,Wei Wu

Chronic bacterial infections, excessive inflammation, and oxidative stress significantly hinder diabetic wound healing by prolonging the inflammatory phase and complicating the healing process. In this study, phenylboronic acid functionalized dextran (PODP) was developed to encapsulate curcumin , referred to as PODP@Cur. Experimental results indicate that PODP significantly improves the water solubility of curcumin and exhibits synergistic biological activity both in vitro and in vivo . PODP@Cur is capable of accelerating drug release under the pathological microenvironment with ROS accumulation. Furthermore, phenylboronic acid (PBA) has demonstrated potential for targeted bacterial drug delivery, enhancing antibacterial efficacy and trapping free LPS/PGN from dead bacteria to reduce undesirable inflammation. In a diabetic mouse model, PODP@Cur exhibits an excellent antibacterial, anti-inflammatory and antioxidant activities to ultimately promote the efficient and safe wound healing. Due to the specific interaction between PBA and LPS , PODP@Cur could enhance antibacterial activity against bacteria, reduce toxic side effects on normal cells, and alleviate the LPS-mediated pro-inflammatory pathological microenvironment. Therefore, PODP@Cur is capable of being exploited as an efficient and safe candidate for promoting the bacteria-infected diabetic wound healing.

pH-responsive Aminated mesoporous silica microspheres modified with soybean hull polysaccharides for curcumin encapsulation and controlled release

Shumin Wang,Shengnan Wang,Lina Yang,Peng Wang,Hong Song,He Liu

Mesoporous silica microspheres (MSMs) possess poor biocompatibility . This study focuses on integrating MSMs with polymers to obtain hybrid materials with superior performance compared to the individual components and responsive release in specific environments. The synthesized MSMs were aminated, and subsequently, soybean hull polysaccharide (SHPs) was modified onto MSMs-NH 2 to produce MSMs-NH 2 @SHPs nanoparticles . The encapsulation rate, loading rate of curcumin (Cur), and in vitro release behavior were investigated. Results indicated that the encapsulation efficiency of Cur by MSMs-NH 2 @SHPs nanoparticles reached 75.58%, 6.95 times that of MSMs-NH 2 with a load capacity of 35.12%. It is noteworthy that these nanoparticles exhibit pH-responsive release capacity in vitro. The cumulative release rate of the three nanoparticles at pH 5.0 was higher than that at pH 7.4. MSMs-NH 2 @SHPs had a cumulative release rate of 56.55% at pH 7.4, increasing to 76.21% at pH 5.0. In vitro experiments have shown that MSMs-based nanoparticles have high delivery efficiency and can achieve pH-sensitive drug release, with a high release rate in a slightly acidic acid, highlighting the potential for controlled release of Cur.

Advancing the pH-driven encapsulation technique of curcumin: Molecular interaction shifts due to structural and charge variations

Hanyu Li,Minghui Zhao,Jinghong Li,Jiaqi Wang,Huajiang Zhang,Jing Wang,Ning Xia,Zhongjiang Wang,Ahmed M. Rayan

The pH-driven method, recognized for its material safety and avoidance of organic solvents, is considered a promising approach for the encapsulation of hydrophobic active substances. However, the interaction mode and mechanism between carriers and receptors in two stages (alkalization and neutralization) remain underexplored. In this study, the hydrophilic globular protein ovalbumin (OVA) was utilized as a carrier to load curcumin (Cur) by the pH-driven method, with an alkalization pH of 8–12. The results indicated that the solubility of Cur and the degree of protein unfolding increased with increasing alkalization pH, with a transformation in the secondary structure observed at pH 12. During the alkalization stage at pH 11 and 12, the loading efficiency of Cur exceeded 90%, with binding constants reaching as high as 5.79 × 10 5  M −1 and 1.44 × 10 5  M −1 , respectively. The interactions transitioned through three phases in the alkalization process: electrostatic dominance to hydrophobic dominance, electrostatic assistance to hydrophobic dominance, and electrostatic repulsion hindrance. Notably, the generation of electrostatic repulsion and the potential loss of binding sites resulted in a decrease in the binding affinity between OVA and Cur at pH 12. In the neutralization stage, a stable structure dominated by hydrogen bonds and van der Waals forces was observed, with Cur encapsulated within the hydrophobic core of OVA. The mechanism proposed in this study elucidates the factors affecting the efficacy of the pH-driven method and helps to refine the application in hydrophobic active substance delivery systems.
