VIS-NIR radiation properties of starchy and protein foods and their impacts on radiant heating effects

Wei Yang,Linshuang Long,Yanjun Sun,Fan Yang,Luo Zhang,Kai Xu,Hong Ye

Infrared heating has been widely used in food processing; however, there are few studies on the VIS-NIR radiation properties of food. We measured the radiation properties of starchy foods (potato, Chinese yam and sweet potato) and protein foods (beef tenderloin, pork tenderloin and chicken breast) and dispersions of starches and proteins before and after heat treatment in the 0.38–2.5 μm band. The results show that the radiation penetration depths (RPDs) of all tested foods are relatively high in the 0.6–1.3 μm band, with maximum values in the range of 4–8 mm. After heat treatment, the RPDs of starchy foods slightly increase, while those of protein foods decrease to approximately 1/2 of the original values. Comparative heating experiments of potato, Chinese yam and sweet potato heated by a halogen lamp (1.04 μm) and a carbon fiber lamp (1.76 μm) with peak wavelengths in and out of the 0.6–1.3 μm band, respectively, show that the foods heated by the halogen lamp have faster heating rates and more uniform temperature distributions. Because starchy and protein foods have high RPDs in the 0.6–1.3 μm band, the VIS-NIR radiation containing this band is expected to achieve rapid heating.

NIR-triggered thermosensitive polymer brush coating modified intraocular lens for smart prevention of posterior capsular opacification

Qin Chen,Wen Shimin,Fei Fan,Han Yuemei,Wang Haiting,Chen Hao,Lin Quankui

Posterior capsule opacification (PCO) is the most common complication after cataract surgery. Drug-eluting intraocular lens (IOLs) is a promising concept of PCO treatment in modern cataract surgery. However, the large dose of drugs in IOL leads to uncontrollable and unpredictable drug release, which inevitably brings risks of overtreatment and ocular toxicity. Herein, a low-power NIR-triggered thermosensitive IOL named IDG@P(NIPAM- co -AA)-IOL is proposed to improve security and prevent PCO by synergetic controlled drug therapy and simultaneous photo-therapy. Thermosensitive polymer brushes Poly(N-isopropylacrylamide- co -Acrylic acid) (P(NIPAM- co -AA)) is prepared on IOL via surface-initiated reversible addition-fragmentation chain transfer (SI-RAFT) polymerization. Then, Doxorubicin (DOX) and Indocyanine green (ICG) co-loaded Gelatin NPs (IDG NPs) are loaded in P(NIPAM- co -AA) by temperature control. The IDG NPs perform in suit photodynamic & photothermal therapy (PTT&PDT), and the produced heat also provides a trigger for controllable drug therapy with a cascade effect. Such functional IOL shows excellent synergistic drug-phototherapy effect and NIR-triggered drug release behavior. And there is no obvious PCO occurrence in IDG@P(NIPAM- co -AA) IOL under NIR irradiation compared with control group. This proposed IDG@P(NIPAM- co -AA)-IOL serves as a promising platform that combines phototherapy and drug-therapy to enhance the therapeutic potential and medication safety for future clinical application of PCO treatment.

Rational design of high performance nanotheranostics for NIR-II fluorescence/magnetic resonance imaging guided enhanced phototherapy

Qi Wang,Jie Cai,Xinrui Niu,Jing Wang,Jiawei Liu,Chen Xie,Wei Huang,Quli Fan

Nanotheranostics, which can provide great insight into cancer therapy, has been deemed as a promising technology to settle the unmet medical needs. The rational design of high performance nanotheranostics with multiple complementary imaging features and satisfactory therapeutic efficacy is particularly valuable. Herein, versatile nanotheranostic agents DPPB-Gd-I NPs were fabricated by using gadolinium-diethylenetriaminepentaacetic acid chelates and an iodine-decorated copolymer as encapsulation matrixes to encapsulate a polymer DPPB through one-step nanoprecipitation. We have demonstrated that such nanoagents are able to efficiently damage tumors under single dose injection and NIR laser illumination conditions due to the enhanced photodynamic therapy and enhanced photothermal therapy (the tumor inhibition rate was as high as 94.5%). Moreover, these nanoagents can be utilized as dual-modal NIR-II fluorescence/magnetic resonance imaging probes for tumor diagnosis with high sensitivity, deep tissue penetration, and excellent spatial resolution. Overall, this work offers a powerful tactic to fabricate high performance nanotheranostics for clinical application.

Enhanced UV–Vis–NIR composite photocatalysis of NaBiF4:Yb3+, Tm3+ upconversion nanoparticles loaded on Bi2WO6 microspheres

Chunyan Zhang,Zhendong Fu,Feng Hong,Jingzhi Dou,Tengri Dong,Yunxiao Zhang,Dan Li,Guixia Liu,Xiangting Dong,Jinxian Wang

NIR light accounts for 50% of total solar energy, but only a small amount of the energy is applied to photocatalytic reactions. The incorporation of upconversion nanoparticles with semiconductor materials is an effective tactics to exploit NIR-driven photocatalyst . In this work, we report a composite material that can convert NIR light into UV–Vis photons. The light conversion material is composed of NaBiF 4 : Yb 3+ , Tm 3+ up-conversion nanocrystals . The petal-like Bi 2 WO 6 is used as a semiconductor photocatalyst to directly degrade model pollutants, such as RhB. By controlling the morphology of Bi 2 WO 6 , the contact area between the semiconductor photocatalyst and the pollutant molecules is increased. The preparation of “double bismuth” composite material NaBiF 4 :Yb 3+ , Tm 3+ /Bi 2 WO 6 provides the possibility to make full use of sunlight. Under the excitation of simulated sunlight, the photocatalyst of NaBiF 4 :Yb 3+ , Tm 3+ /Bi 2 WO 6 reaches the highest degradation efficiency of 98% in 27 min. Compared with Bi 2 WO 6 , the prepared composite photocatalyst shows better photocatalytic performance, which is attributed to the effective energy transfer between NaBiF 4 :Yb 3+ ,Tm 3+ and Bi 2 WO 6 . The capture tests were used to detect active species used in the degradation process. After four continuous photocatalytic cycles, about 95% of RhB solution is degraded. Under the simulated solar light illumination, the photocatalytic mechanism of the composite photocatalyst is discussed. The results propose a promising system to improve the photocatalytic activity through the efficient use of sunlight.

NIR molecule induced self-assembled nanoparticles for synergistic in vivo chemo-photothermal therapy of bladder cancer

Guanchen Zhu,Qingfeng Zhang,Xiaozhi Zhao,Qing Zhang,Wei Chen,Linfeng Xu,Sheng Zhao,Kaikai Wang,Tianqing Liu,Hongqian Guo

Bladder cancer (BC) is one of the commonest malignancies in the urinary system. Bladder cancer is divided into non-muscle invasive bladder cancer (NMIBC) and muscle invasive bladder cancer (MIBC) according to the depth of invasion. Besides, the prognosis of MIBC remains poor. Surgical resection combined with radiotherapy or chemotherapy is the standard treatment for MIBC. However, the major obstacle that hinders successful chemotherapy is its lack of tumor targeting. Here, we fabricated nanoparticles that respond to near-infrared laser irradiation in order to increase the drug accumulation at the tumor sites and combine chemotherapy with photothermal therapy to overcome challenges of bladder cancer treatment. IR780 and Doxorubicin (DOX) were loaded into albumin nanoparticles (IR780-DOX@Albumin NPs). In the process of IR780-DOX@Albumin NPs synthesis, the near-infrared molecule IR780 was used as the assembly molecular bridge. Under irradiation, the nanoparticles were decomposed due to the degradation of IR780 while the release of DOX increased. Nanoparticles can be ingested by tumor cells in a short time. The IR780-DOX@Albumin NPs were sensitive to near-infrared laser irradiation. Near-infrared laser irradiation can promote the release of the drugs from the nanoparticles and induce a photothermal effect, thus destroying the tumor cells. Given the excellent tumor-targeting ability and negligible toxicity to normal tissue, IR780-DOX@Albumin NPs can greatly increase the concentration of chemotherapeutic drugs in tumor cells. This study combines photothermal therapy and chemotherapy to treat MIBC, so as to avoid chemotherapy resistance, reduce the toxicity to normal cells, and achieve the purpose of improving the treatment of MIBC.

Hybrid Hydrogels for Synergistic Periodontal Antibacterial Treatment with Sustained Drug Release and NIR-Responsive Photothermal Effect

Jian Lin,Zhiqi He,Fen Liu,Jie Feng,Chengyi Huang,Xueli Sun,Hui Deng

Background Periodontal pathogenic bacteria promote the destruction of periodontal tissues and cause loosening and loss of teeth in adults. However, complete removal of periodontal pathogenic bacteria, at both the bottom of the periodontal pocket and the root bifurcation area, remains challenging. In this work, we explored a synergistic antibiotic and photothermal treatment, which is considered an alternative strategy for highly efficient periodontal antibacterial therapy. Methods Mesoporous silica (MSNs) on the surface of Au nanobipyramids (Au NBPs) were designed to achieve the sustained release of the drug and photothermal antibacterials. The mesoporous silica-coated Au NBPs (Au NBPs@SiO 2 ) were mixed with gelatin methacrylate (GelMA-Au NBPs@SiO 2 ). Au NBPs@SiO 2 and GelMA-Au NBPs@SiO 2 hybrid hydrogels were characterized, and the drug content and photothermal properties in terms of the release profile, bacterial inhibition, and cell growth were investigated. Results The GelMA-Au NBPs@SiO 2 hybrid hydrogels showed controllable minocycline delivery, and the drug release rates increased under 808 nm near-infrared (NIR) light irradiation. The hydrogels also exhibited excellent antibacterial properties, and the antibacterial efficacy of the antibiotic and photothermal treatment was as high as 90% and 66.7% against Porphyromonas gingivalis ( P. gingivalis ), respectively. Moreover, regardless of NIR irradiation, cell viability was over 80% and the concentration of Au NBPs@SiO 2 in the hybrid hydrogels was as high as 100 µg/mL. Conclusion We designed a new near-infrared light (NIR)-activated hybrid hydrogel that offers both sustained release of antibacterial drugs and photothermal treatment. Such sustained release pattern yields the potential to rapidly eliminate periodontal pathogens in the periodontal pocket, and the photothermal treatment maintains low bacterial retention after the drug treatment.

Magnetic Silica Nanosystems With NIR-Responsive and Redox Reaction Capacity for Drug Delivery and Tumor Therapy

Jia Chengzheng,Wu Hang,Luo Keyi,Hao Weiju,Wang Shige,Huang Mingxian

In recent years, more and more researches have focused on tumor photothermal therapy and chemodynamic therapy. In this study, we prepared a multifunctional nanomaterial with potential applications in the above area. The Fe3O4 nanoparticles were synthesized with suitable size and uniformity and then coated with mesoporous silica and polydopamine. The unique core-shell structure not only improves the drug loading of the magnetic nanomaterials, but also produces high photothermal conversion efficiency. Furthermore, the reducibility of polydopamine was found to be able to reduce Fe3+ to Fe2+ and thus promote the production of hydroxyl radicals that can kill the tumor cells based on the Fenton reaction. The magnetic nanomaterials are capable of simultaneously combining photothermal and chemodynamic therapy and permit the efficient treatment for tumors in the future.

Upconversion Nanoparticle-Based Organosilica–Micellar Hybrid Nanoplatforms for Redox-Responsive Chemotherapy and NIR-Mediated Photodynamic Therapy

Zhenyang Wei,Xiaohang Liu,Dechao Niu,Limei Qin,Yongsheng Li

Upconversion nanoparticles (UCNPs) can convert near-infrared light (NIR, 980 or 808 nm) to ultraviolet (UV) or visible light, which can be widely used to improve tissue penetration depth in photodynamic therapy (PDT). Herein, we develop a kind of UCNP-based organosilica–micellar hybrid nanoplatform for redox-responsive chemotherapy and NIR-mediated PDT. The nanoplatform was constructed by the self-assembly of block copolymers polystyrene-b-poly (acrylic acid) and oil-soluble UCNPs in the oil/water system and the subsequent organosilica coating with 3-mercaptopropyltrimethoxysilane molecules. To endow the nanosystem with more stability in biological media, polyethylene glycol molecules were further modified via the Michael addition reaction. As a promising nanocarrier, chlorin e6 (Ce6) and doxorubicin (DOX) molecules were loaded into the hydrophobic core and the disulfide-doped organosilica shell, respectively. With endocytosis by SMMC-7721 tumor cells, the Ce6 and DOX coloaded nanosystem was activated by UCNPs through luminescence resonance energy transfer under the irradiation of 808 nm laser, thus generating cytotoxic 1O2 for NIR-mediated PDT. Meanwhile, DOX was selectively released because of the redox-responsive biodegradation of the disulfide-doped organosilica shell in the glutathione over-expressed SMMC-7721 tumor cells. Based on these, the chemotherapy/PDT combination toxic feature of the multifunctional nanosystem was further demonstrated in the DOX-resistant MCF-7 tumor cells. On the other hand, the Ce6 and DOX coloaded nanosystem exhibited negligible toxicity to the normal 3T3 cell because of the protective effects of organosilica coating. We envision that the resultant hybrid nanoplatform provides us a promising nanocarrier for the combination therapy of redox-responsive safe chemotherapy and efficient NIR-mediated PDT.

NIR-driven intracellular photocatalytic oxygen-supply on metallic molybdenum carbide@N-carbon for hypoxic tumor therapy

Huaying Hou,Zhongxu Wang,Yajie Ma,Kai Yu,Jingxiang Zhao,Huiming Lin,Fengyu Qu

The intracellular O 2 -supply not only can relieve tumor hypoxia but also enhance the effects of photodynamic therapy (PDT). In this work, metallic Mo 2 [email protected] N [email protected] nanoparticles were constructed to reveal the near infrared (NIR)-photocatalytic O 2 generation and promote photodynamic therapy (PDT). Here, (NH 4 ) 6 Mo 7 O 24 ·4H 2 O nanorods and urea were adopted as resources that were calcined to obtain Mo 2 [email protected] N -carbon nanoparticles (20 nm). All samples displayed high NIR absorption as well as photothermal conversion efficiency of up to 52.7 % (Mo 2 [email protected] N [email protected] ). The density functional theory calculations demonstrated the metallic characteristic of Mo 2 C and that the consecutive interband/intraband charge-transition was responsible for the high NIR harvest and redox ability of electron-hole pairs, making the NIR-photocatalytic O 2 and reactive oxygen species (ROS) generation. In comparison with the pure Mo 2 C, the heterostructure displayed twice the performance due to the enhanced charge-segregation between Mo 2 C and N -carbon. Given the high X-ray absorption coefficient and photothermal ability, the nanocomposite could be used in novel computer tomography and photothermal imaging contrast. Furthermore, the novel biodegradation and metabolism behaviors of nanocomposites were investigated, which were reflected as elimination from the body (mouse) via feces and urine within 14 days. The as-synthesized Mo 2 [email protected] N [email protected] nanocomposites integrated the dual-model imaging, intracellular O 2 -supply, and phototherapy into one nanoplatform, revealing its potential for anti-cancer therapy.

NIR-II-driven and glutathione depletion-enhanced hypoxia-irrelevant free radical nanogenerator for combined cancer therapy

Zhang Li,Fan Yadi,Yang Zhe,Yang Mo,Wong Chun-Yuen

Background Though the combination of photodynamic therapy (PDT) and chemodynamic therapy (CDT) appears to be very attractive in cancer treatment, hypoxia and overproduced glutathione (GSH) in the tumor microenvironment (TME) limit their efficacy for further application. Results In this work, a smart hypoxia-irrelevant free radical nanogenerator (AIPH/PDA@CuS/ZIF-8, denoted as APCZ) was synthesized in situ via coating copper sulphide (CuS)-embedded zeolitic imidazolate framework-8 (ZIF-8) on the free radical initiator 2,2′-azobis[2-(2-imidazolin-2-yl)propane]-dihydrochloride (AIPH)-loaded polydopamine (PDA). APCZ showed promising GSH-depleting ability and near-infrared (NIR)-II photothermal performance for combined cancer therapy. Once internalized by 4T1 cells, the outer ZIF-8 was rapidly degraded to trigger the release of CuS nanoparticles (NPs), which could react with local GSH and sequentially hydrogen peroxide (H2O2) to form hydroxyl radical (·OH) for CDT. More importantly, the hyperthermia generated by APCZ upon 1064 nm laser excitation not only permitted NIR-II photothermal therapy (PTT) and promoted CDT, but also triggered the decomposition of AIPH to give toxic alkyl radical (·R) for oxygen-independent PDT. Besides, the PDA together with CuS greatly decreased the GSH level and resulted in significantly enhanced PDT/CDT in both normoxic and hypoxic conditions. The tumors could be completely eradicated after 14 days of treatment due to the prominent therapeutic effects of PTT/PDT/CDT. Additionally, the feasibility of APCZ as a photoacoustic (PA) imaging contrast agent was also demonstrated. Conclusion sThe novel APCZ could realize the cooperative amplification effect of free radicals-based therapies by NIR-II light excitation and GSH consumption, and act as a contrast agent to improve PA imaging, holding tremendous potential for efficient diagnosis and treatment of deep-seated and hypoxic tumors.Graphic abstract

Ruiyi Dai,Xiangrong Peng,Bi Lin,Danyang Xu,Ruichan Lv

In this research, a NIR II luminescence imaging and enhanced chemo-/photothermal therapy system of CuS–DOX–Nd/FA NPs for breast cancer and lymph node tracing under single 808 nm irradiation is proposed. Nd–DTPA molecular cluster with the NIR II imaging effect as the carrier was designed to load the ultrasmall CuS nanoparticles and chemotherapeutic drug doxorubicin hydrochloride (DOX). The composite probe is used for tumor lesion imaging and tracking the breast cancer sentinel lymph nodes with simultaneous chemo-/photothermal therapy (PTT) for breast cancer under the single 808 nm laser. This designed probe not only has high permeability and retention (EPR) targeting effect but also can respond to the tumor microenvironment (TME), realizing more precise and efficient release of DOX at the cancer focus. At the same time, CuS as a drug carrier has a good photothermal therapy effect (photothermal conversion efficiency: 27.9%). The serialized released chemotherapy DOX and synergistic PTT effect can be used to the treat the in situ breast cancer land and simultaneously kill the metastasis cancer. The system made the combined molecular clusters Nd–DTPA achieve NIR II imaging of tumor lesions of breast cancer and lymph node to obtain the integration of diagnosis of the transferred disease for better prognosis. The feasibility of the system had obvious tumor growth inhibition effect with NIR II imaging guided is verified by a series of in vitro and in vivo experiments.

Polymer encapsulated clinical ICG nanoparticles for enhanced photothermal therapy and NIR fluorescence imaging in cervical cancer

Shuang Chen,Lijun Zhu,Zhong Du,Rong Ma,Ting Yan,Gulinigaer Alimu,Xueliang Zhang,Nuernisha Alifu,Cailing Ma

Photothermal therapy (PTT) is a popular tumor therapy method, which is based on efficient photothermal nanoagents (PTNs). Clinical Indocyanine Green (ICG), as a Food and Drug Administration (FDA) approved agent, is an often-used PTN, meanwhile it is also a good near-infrared (NIR) fluorescence contrast agent. However, the further applications of ICG in biomedical fields are limited due to its poor stability. In this study, ICG was encapsulated by the amphiphilic polymer poly(styrene-co-maleic anhydride) (PSMA) to form ICG@PSMA nanoparticles. Furthermore, optical and thermal characteristics of ICG@PSMA nanoparticles were studied in detail. Strong NIR fluorescence and excellent photothermal properties of ICG@PSMA nanoparticles under 808 nm laser irradiation were measured. Besides, favorable biocompatibility of ICG@PSMA nanoparticles was demonstrated on a human cervical cancer cell line (HeLa) via cell viability studies. Hence, ICG@PSMA nanoparticles were further applied to enhanced PTT of living HeLa cells under 808 nm excitation, and a high PTT efficiency of ∼70% was obtained. The novel ICG nanoparticles as a promising PTT nanoplatform could offer an opportunity for further tumour treatments.
