An Insulin-Modified pH-Responsive Nanopipette Based on Ion Current Rectification

Xu-Fan Wang,Yi-Fan Duan,Yue-Qian Zhu,Zi-Jing Liu,Yu-Chen Wu,Tian-Hao Liu,Ling Zhang,Jian-Feng Wei,Guo-Chang Liu

The properties of nanopipettes largely rely on the materials introduced onto their inner walls, which allow for a vast extension of their sensing capabilities. The challenge of simultaneously enhancing the sensitivity and selectivity of nanopipettes for pH sensing remains, hindering their practical applications. Herein, we report insulin-modified nanopipettes with excellent pH response performances, which were prepared by introducing insulin onto their inner walls via a two-step reaction involving silanization and amidation. The pH response intensity based on ion current rectification was significantly enhanced by approximately 4.29 times when utilizing insulin-modified nanopipettes compared with bare ones, demonstrating a linear response within the pH range of 2.50 to 7.80. In addition, insulin-modified nanopipettes featured good reversibility and selectivity. The modification processes were monitored using the I-V curves, and the relevant mechanisms were discussed. The effects of solution pH and insulin concentration on the modification results were investigated to achieve optimal insulin introduction. This study showed that the pH response behavior of nanopipettes can be greatly improved by introducing versatile molecules onto the inner walls, thereby contributing to the development and utilization of pH-responsive nanopipettes.

A capillary-aided microfiber Bragg grating pH sensor for hydrovoltaic technology

Yongkang Zhang,Heyi Xia,Qiaochu Yang,Zhiyuan Xu,Wenbo Wang,Ziyu Yuan,Zesen Li,Shifang Cao,Bai-Ou Guan,Ling Qiu,Yang Ran

Hydrovoltaic is an emerging technology that aims to harvest energy from water flow and evaporation, in which the plasmonic hydrogen ions are generated by the interaction between water and hydrovoltaic device. However, the volume of the water sample for the interaction is usually ultra-small due to the compact size of hydrovoltaic device, making the quantification and characterization of the hydrogen ions in such water sample an elusive goal. To address this issue, a miniature fiber-optic pH probe is proposed using a unilaterally tapered-microfiber Bragg grating. The microfiber Bragg grating has an intrinsic Bragg reflection signal with a narrow linewidth. The fiber probe is functionalized by coating the sodium alginate, which can respond to the variation of pH mediated by the alteration of the hydrophilicity. The rigidity and robustness of microfiber Bragg grating facilitates the encapsulation of the sensor into a sampling capillary, allowing for the detection of trace aqueous sample less than 2 μL. The pH sensitivity of the tapered-μFBG-based sensor is 62.8 p.m./pH (R 2  = 0.995) with a limit resolution of 0.096 pH. The sensor performed a practical application in the monitoring and characterization of the hydrovoltaic microdevice, which can generate microcurrent as soaked in the water. This work demonstrates a promising technology in the fields of materials, energy, biology and medicine, in which the detection of the microsamples is inevitable.

Zwitterionic pH-responsive hyaluronic acid polymer micelles for delivery of doxorubicin

Qin-Qin Gao,Chuan-Min Zhang,En-Xia Zhang,Hui-Ying Chen,Yu-Hong Zhen,Shu-Biao Zhang,Shu-Fen Zhang

As zwitterionic polymers show great promise in drug delivery, hyaluronic acid (HA) was deacetylated and grafted with dodecylamine to prepare a pH-sensitive zwitterionic polymer dHAD used as a carrier for antitumor drugs. The polymer was negatively charged at pH 7.4 and became positive at pH 6.2. In vitro delivery of DOX against MCF-7 cells showed that the blank micelle dHAD had low cytotoxicity and the dHAD-DOX micelles could greatly prohibit the growth of the MCF-7 cells. In addition, the dHAD-DOX micelles had higher cellular uptake, indicating that the micelles were rapidly internalized into the cells via CD44 receptor-mediated endocytosis. The in vivo delivery of DOX to tumor-bearing mice confirmed that the dHAD-DOX micelles greatly inhibited the tumor growth and significantly reduced systemic toxicity of DOX. These results demonstrated that biocompatible pH-responsive zwitterionic dHAD micelles are promising carriers for the delivery of DOX.

Facet-Selective hydrogen evolution on Rh2P electrocatalysts in pH-Universal media

Sufang Yang,Xiaobo Yang,Qiang Wang,Xueqin Cui,Houbing Zou,Xili Tong,Nianjun Yang

Surface atomic structures of transition metal phosphides determine the electrocatalytic activities toward hydrogen evolution reaction (HER). In this work, we developed a new facet-guided phosphorization strategy to synthesize Rh 2 P electrocatalysts with tunable facet exposure and further unveil their intrinsic facet-dependent HER activity in pH-universal media. The Rh 2 P nanoparticles feature {1   1   1} and {2   0   0} facets when the precursors of Rh nanoctahedron and nanocube are used, respectively. The Rh 2 P{2   0   0} catalyst showed a remarkable HER performance with the overpotential of 23, 22 and 54 mV at a current density of 10 mA cm −2 in 1.0 M KOH, 0.5 M H 2 SO 4 and 1.0 M phosphate buffer, respectively. The mass activity and specific activity of Rh 2 P{2   0   0} were 3~5-times higher than those of Rh 2 P{1   1   1} and Pt/C catalysts. As elucidated using theoretical calculations, the overlapping of Rh-4d and P-3p of Rh 2 P{2   0   0} and Rh 2 P{1   1   1} results in distinct electronic states of surface Rh atoms, leading to varied free energies for water dissociation and hydrogen adsorption/desorption.

Metal-polyphenol network coated photothermal nanocarriers for pH-activated drug delivery

Qiong Wang,Jing Chen,Junhong Ling,Han Zhao,Xiao-kun Ouyang,Nan Wang

The combination of photothermal and chemotherapy is a promising strategy for the synergistic inhibition of tumor cells. By exploiting the tumor microenvironment, chemotherapeutic drugs can target the tumor site, thereby enhancing their efficacy. This study focused on the development of nanodrugs, aiming to harness the synergistic photothermal and chemotherapy capabilities for enhanced anti-tumor effects. The core material was made of copper sulfide (CuS), which possesses photothermal conversion properties, while doxorubicin (DOX) was loaded on mesoporous polydopamine (MPDA). The drug-loaded nanoparticles were then encapsulated in acid-sensitive tannic acid and an Fe complex (TA-Fe). The photothermal conversion properties, drug loading, and release of DOX from CuS-MPDA-DOX@TA-Fe were then investigated. The results showed that DOX had an encapsulation ratio of 82.2 % and a release ratio of 43.49 % in an acidic environment (pH 5.2). In vitro studies showed that CuS-MPDA-DOX@TA-Fe triggered by NIR effectively inhibited tumor cell proliferation and that the tumor cells showed suitable levels of nanoparticle uptake. The results thus show that CuS-MPDA-DOX@TA-Fe nanodrugs could be utilized as a promising therapeutic strategy for cancer treatment with synergistic photothermal/chemotherapeutic anti-tumor effects.

Multifunctional Copper Nanoclusters for the Detection of pH and Berberine Hydrochloride

Chen De Xing,Long Yun Fei

The copper nanoclusters (CuNCs) have been garnered significant attention owing to their low cost raw materials, outstanding optical properties, environmental friendliness and good biocompatibility. In this study, copper nanoclusters stabilized by Acid Chromic Blue K (ACBK-CuNCs) were synthesized. The optimal excitation wavelength of the synthesized ACBK-CuNCs is 344 nm and the optimal emission wavelength is 460 nm. Its fluorescence intensity can be retained for more than two weeks, which indicated that the antioxidant capacity of the synthesized ACBK-CuNCs is strong. It was noted that the fluorescence intensities of the synthesized ACBK-CuNCs significantly increased with the decrease of pH. There are good linear correlation between the fluorescence intensity ratio of the system to the fluorescence intensity of the original solution (F/F 0 ) and the pH of the medium. Based on this finding, a method for fluorescence analysis to detect the pH of solution was developed. Additionally, it was discovered that the fluorescence intensity of CuNCs could be quenched efficiently by Berberine hydrochloride (BH), and there is a favorable linear relationship between the change value of fluorescence intensity of CuNCs ((F 0 - F) / F 0 ) and the concentration of BH. This gave rise to the establishment of a fluorescence analysis method for determining the concentration of BH. Simultaneously, visual quantitative detection for BH and pH was achieved based on smartphones and colorimetric analysis software using ACBK-CuNCs as the fluorescent probes. Graphical

Oppositely Sensitive pH-Responsive Surfactants Derived from Polymer-Grafted Carbon Dots

Tianhao Zhang,Junjie Fan,Wenxiu Qiu,Xinyu Yuan,Jianwen Hu,Haotian Gao,Wenqing Xie,Chunling Li,Shuangqing Sun,Songqing Hu

This study reports the development of two pH-responsive Pickering emulsifiers utilizing carbon dots (CDs) as the core component. Both emulsifiers were fabricated via a post-synthetic method, involving the grafting of poly[2-(dimethylamino)ethyl methacrylate] (PDMAEMA) onto the surface of CDs to create CD-based emulsifier. Notably, despite employing an identical synthetic route, the two CD-based emulsifiers exhibit markedly distinct pH-responsive behaviors. The findings demonstrate that both CD-based emulsifiers effectively stabilize Pickering emulsions, and their emulsification and demulsification behaviors can be precisely controlled by adjusting the pH, displaying excellent reproducibility in their pH-responsive performance. To elucidate the underlying reason for this differing pH-responsiveness, the concept of hydrophilic-lipophilic balance (HLB) was introduced. The analysis of the HLB values of the two CD-based emulsifiers revealed that their hydrophilic-lipophilic properties exhibit opposite trends under differing pH conditions, contributing to their contrasting pH-responsive behaviors. This study not only lays a fundamental basis foundation and offers guidance for further research and applications of CD-based smart-responsive emulsifiers but also presents novel insights into design and development of more intelligent and functional emulsifying agents. Moreover, owing to the enhanced fluorescence characteristics of CDs, these pH-responsive emulsifiers hold significant potential for interdisciplinary applications, such as oil and gas exploration and fluorescent labelling.

pH-Sensitive oligopeptide magnetic mesoporous silica beads for deoxyribonucleic acid extraction

Sihua Qian,Yiting Wang,Junjie Fan,Tong Kong,Yuhui Wang,Kaizhe Wang,Yufeng Liao,Li Wang,Jianping Zheng

Exploring novel synthesis strategies for magnetic beads to extract nucleic acids is of great significance in the field of in vitro diagnostics. In the present research, monodisperse magnetic mesoporous silica beads were synthesized via the thermolysis reaction of Fe(acac)3 by using large-pore dendritic silica colloids as templates, and were further functionalized with a highly pH-sensitive histidine-glutamate co-oligopeptide for deoxyribonucleic acid extraction. The large-pore dendritic silica colloid scaffolds were utilized for high-density incorporation of superparamagnetic iron oxide nanoparticles within the vertical channels. The morphology and properties of the as-prepared pH-sensitive oligopeptide magnetic mesoporous silica beads were evaluated by transmission electron microscopy, scanning electron microscopy, vibrating sample magnetometry, X-ray photoelectron spectroscopy, X-ray diffraction testing and so on. The average size of the obtained magnetic beads was 370 nm in diameter with a narrow size distribution. The saturation magnetization and magnetic content of the resultant magnetic beads were 25 emu g−1 and 59%, respectively. Moreover, the magnetic mesoporous silica beads exhibited an obvious pH-responsive behavior. Due to these remarkable features, successful deoxyribonucleic acid capture using the as-prepared pH-sensitive oligopeptide magnetic mesoporous silica beads was achieved.

pH triggered polycarvacrol-based antibacterial microspheres: Integrated offensive and defensive platform

Xiangbin Sun,Xiaobing Ma,Yufeng He,Pengfei Song,Rongmin Wang

The natural plant essential oil carvacrol exhibits substantial antimicrobial properties. However, its efficacy is significantly compromised following functionalization, thereby limiting its optimal application in polymer science. In this study, a quaternary ammonium carvacrol ester ionic liquid monomer was synthesized utilizing the ionic liquid technique. Subsequently, a series of cyclodextrin-grafted poly(quaternary ammonium carvacrol ester) polymer nanospheres were prepared via reversible addition-fragmentation chain transfer (RAFT) polymerization. The controllability offered by RAFT polymerization enables precise loading of carvacrol small molecules, representing a significant breakthrough for the incorporation of carvacrol into polymers. Particularly noteworthy is the CD-DM x formulation, which demonstrates exceptional antibacterial efficacy against both E. coli and S. aureus . This is attributed to the synergistic interaction between carvacrol and cationic components, resulting in a 100% antibacterial ratio. Additionally, the labile nature of the betaine ester bonds with carvacrol and cations facilitates the controlled release of carvacrol from the CD-DM x matrix while preventing its deactivation during functionalization. This property facilitates the transition of CD-DM x from exhibiting antibacterial properties (offense mode) to demonstrating anti-protein adhesion properties (defense mode). Furthermore, cytotoxicity assessments and electrospinning studies have demonstrated the biocompatibility and potential applications of CD-DM x in medical protective equipment. In summary, we have not only proposed a strategy for the efficient utilization of natural carvacrol essential oil but has also developed a green, environmentally friendly cationic essential oil-based antibacterial agent that is well-suited for medical applications.

PH-activated off/on nanosystem for combined photothermal/chemodynamic antibacterial application

Chengyang Ban,Haimeng Liu,Yaoyu Zhang,Qiong Ding,Changzhi Xu,Jibin Miao,Ru Xia,Jiasheng Qian,Ming Cao

Photothermal therapy (PTT) holds great promise for the treatment of bacterial infections, but the traditional “always-on” photothermal agents severely restrict further clinical applications. Here, a pH-activated off/on PTT/CDT nanosystem for synergistic antibacterial/antibiofilm application was reported. Using mesoporous silica as a nanocarrier, such activatable photoagent was achieved via specific redox reactions between 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and Fe(III), which could switch PTT/CDT from “off” to “on” in response to pH. Under neutral conditions, the nanosystem was “off state” due to the isolation of Fe(III) and ABTS. Whereas under acidic conditions, the nanosystem selectively activated to “on state”. The released Fe(III) can oxidize ABTS to photothermal reagent of ABTS •+ , and itself was reduced to Fe(III)/Fe(II) pairs. The Fe(II)/Fe(III) pairs can catalyze the H 2 O 2 to hydroxyl radicals (•OH) via Fenton reactions for chemodynamic therapy (CDT). The in vitro results showed that the pH-activated off/on PTT/CDT nanosystem have an excellent antibacterial efficiency (˃90 %) and enhanced antibiofilm activity for Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ).

Sprayable Hydrogel for pH-Responsive Nanozyme-Derived Bacteria-Infected Wound Healing

Furong Chao,Chengliang Cao,Yin Xu,Yunjie Gu,Xinyu Qu,Qian Wang,Lu−Lu Qu,Yuxin Guo,Xiaochen Dong

Long-term inflammation and persistent bacterial infection are primary contributors to unhealed chronic wounds. The use of conventional antibiotics often leads to bacteria drug resistance, diminishing wound healing effectiveness. Nanozymes have become a promising alternative to antimicrobial materials due to their low cost, easy synthesis, and good stability. Herein, we develop a novel sprayable hydrogel based on carboxymethyl chitosan (CMCS) and oxidized hyaluronic acid (OHA), incorporating Au nanoparticle-carbon nitride (AuNPs-C3N4) nanozyme, glucose, and Mn2+ for bacteria-infected wound healing. The hydrogel forms rapidly in situ upon spraying and gradually degrades on the wound area, releasing the AuNPs-C3N4 nanozyme, which exhibits robust glucose oxidase-like (GOx-like) activity, initiating a comprehensive catalytic cascade through a Mn2+-mediated Fenton-like reaction that generates hydroxyl radicals (•OH) to eliminate Staphylococcus aureus (S. aureus) and Methicillin-resistant S. aureus (MRSA). Computational results indicate that interactions between AuNPs and g-C3N4 maximize their synergistic effects in a heterojunction, improving O2 adsorption and facilitating electron-O2 interactions to optimize catalytic activity. Further experiments demonstrate that the hydrogel can rapidly cover wounds in situ, while CMCS promotes collagen production and fibroblast proliferation, offering a viable strategy for the healing of bacteria-infected wounds.

Zwitterionic Polycatechols for Covalent Conjugation With Bortezomib and pH-Triggered Release

Naikuan Fu,Ao Li,Jing Zhang,Peng Zhang,Hong Zhang,Shicheng Yang,Jianhua Zhang

The therapeutic efficacy of Bortezomib (BTZ) is severely limited by its low solubility, poor stability in vivo and nonspecific toxicity. PEGylated nanocarriers can improve drug delivery efficiency, but their applications often suffer from low drug loading, premature leakage and accelerated blood clearance phenomenon. Herein a kind of catechol-functionalized and sulfobetaine-based zwitterionic block copolymer (PGMAD-PSBMA) is prepared by RAFT copolymerization and an epoxy-amino click reaction. And then PGMAD-PSBMA is readily used to conjugate with BTZ by the formation of dynamic boronate bonds to obtain zwitterionic BTZ prodrug (PGMAD@BTZ-PSBMA) and PGMAD@BTZ-PSBMA micelles. The structure and morphology, physicochemical characteristics, drug loading, pH-triggered drug release as well as in vitro cytotoxicity of PGMAD@BTZ-PSBMA micelles are investigated in detail. The results demonstrate that PGMAD@BTZ-PSBMA micelles can not only possess high drug loading (12.9%) and stable dispersion in physiological pH condition (pH 7.4), but also respond to the tumor acid microenvironment and achieve pH-responsive BTZ release. The nanocarriers designed here readily combine the desirable functions of polycatechols for stable conjugation and acid-triggered release and polysulfobetaines for long circulation in blood, which have great potential to enhance therapeutic efficacy and reduce toxic side effects of BTZ and other boronic acid-containing drugs, such as Ixazomib and Steboronine.
