As a prerequisite of gene delivery in living cells, DNA condensation has attracted more and more attention. In order to improve the efficiencies of polyamine-β-cyclodextrin-based cationic polyrotaxanes (PR-EDA and PR-DETA) as DNA condensation materials, we have designed and prepared two novel scutellarin-grafted cationic polyrotaxanes (PR-EDA-SCU and PR-DETA-SCU), in which scutellarins (SCU), the planar molecules, were conjugated on the cyclodextrin molecules of PR-EDA and PR-DETA. These materials were characterized by 1D and 2D NMR, XRD, TG and DSC. The electrophoresis assays showed that pDNA condensation efficiencies of PR-EDA and PR-DETA were better than that of PR-EDA and PR-DETA. The complexes of PR-EDA, PR-DETA, PR-EDA-SCU and PR-DETA-SCU with pDNA were further investigated by zeta potential and atomic force microscopy analysis. The results indicated that the planar structure of SCU played an important role in improvement of pDNA condensation efficiencies of PR-EDA-SCU and PR-DETA-SCU. The satisfactory pDNA condensation abilities of PR-EDA-SCU and PR-DETA-SCU could be helpful in designing non-viral gene delivery vectors to control gene expression and delivery.

β-Glucosidases are enzymes that hydrolyze β-glycosidic bonds to release non-reducing terminal glucosyl residues from glycosides and oligosaccharides and thus have significant application potential in industries. However, most β-glucosidases are feedback inhibited by the glucose product, which restricts their application. Remarkably, some β-glucosidases of the glycoside hydrolase (GH) 1 family are tolerant to or even stimulated by glucose. Elucidation of the mechanisms of glucose tolerance and stimulation of the GH1 β-glucosidases will be crucial to improve their application through enzyme engineering. In this study, by comparing the primary and tertiary structures of two GH1 β-glucosidases with distinct glucose dependence, some putative glucose-dependence relevant sites were mutated to investigate their exact roles. Both biochemical and structural characterization of the mutants suggested that some sites at the entrance and middle of the substrate channel regulate the effects of glucose and the relative binding affinity/preference of these sites to glucose modulates the glucose dependence. A mechanism was therefore proposed to interpret the glucose dependence of GH1 β-glucosidases. This research provides fresh insight into our current understanding of the properties and mechanisms of GH1 β-glycosidases and related enzymes that modulate their activity via feedback control mechanism.

In order to improve the carbonation properties of steel slag, the carbonation reaction process of steel slag and the formation of carbonation products were regulated by the interaction between multiple hydroxyl groups within β-cyclodextrin (β-CD) molecule and calcium ions leached from steel slag. Through a meticulous experimental design, varying contents of β-CD, from 0.0 % to 5.0 %, were mixed with steel slag to investigate their impact on carbonation kinetics, mechanical strength, and microstructural evolution. The results indicate that β-CD significantly accelerates the carbonation process of steel slag, with a particular boost to the carbonation of β-dicalcium silicate (β-C 2 S), a key mineral contributing to the carbonation activity. At an optimal β-CD concentration of 2.5 %, the compressive strength of the treated slag reached a peak of 138.5 MPa, marking a 13.4 % improvement over the control sample. Similarly, the CO 2 uptake was optimized at 17.9 %, increasing by 21.8 %. Mechanism analysis reveals that β-CD not only facilitates carbonation but also refines the resulting carbonation products, leading to a denser microstructure and enhanced mechanical properties. However, at a β-CD content of 5.0 %, the carbonation process was hindered by excessive viscosity. These pivotal findings propose an innovative strategy to augment the efficiency of carbonation and the inherent material properties of steel slag, thereby fostering a more sustainable approach to the utilization of industrial byproducts.

As the main active compound of Glycyrrhiza glabra L., glabridin (GLD) has been shown to have multiple bioactivities, whereas the clinical application of GLD is restricted by its low water solubility. In this study, GLD was encapsulated into a sulfobutylether-β-cyclodextrin (SBE-β-CD)-based inclusion complex (SBE-β-CD/GLD) by the freeze-drying method. The materials characterization, antibacterial activity, stimulated cellular behavior and in vivo full-thickness diabetic wound healing ability of the hydrogels were assessed and analyzed. The successful encapsulation of the inclusion complex was confirmed by ultraviolet (UV) visible spectroscopy, Fourier transform infrared (FT-IR), X-ray diffractometer (XRD), scanning electron microscope (SEM), and nuclear magnetic resonance (NMR). SBE-β-CD as an excipient significantly enhances the water solubility of GLD, and SBE-β-CD/GLD showed excellent biocompatibility on human vascular endothelial cells (HUVECs) and erythrocytes. The SBE-β-CD/GLD inclusion complex exerted a pronounced antibacterial activity on Staphylococcus aureus and Escherichia coli in vitro. The SBE-β-CD/GLD inclusion complex markedly enhanced the antioxidant activity compared with free GLD. The SBE-β-CD/GLD inclusion complex potently accelerates the healing of full-thickness skin defects by inhibiting inflammation. The outcomes suggest that SBE-β-CD could be used as a promising drug delivery system for the clinical application of GLD.

Lactose can cause health problems in patients with lactose intolerance. β-Galactosidase is commonly used to catalyze the hydrolysis of lactose, thereby producing low-lactose or lactose-free products. However, most existing methods employ free enzymes, which significantly increases the economic cost. In this study, the polyurethane sponge (PUS) was selected as the carrier, and the cross-linked enzyme aggregates (CLEAs) were loaded onto the carrier after the sponge was modified. The reaction products were characterized by scanning electron microscopy, X-ray photoelectron spectroscopy, Fourier infrared transform spectroscopy, and simultaneous thermal analysis, respectively. The loading amount of β-galactosidase under the optimal conditions was 63 U/g support . The prepared PUS@β-galactosidase-CLEAs was successfully used for the hydrolysis of lactose in milk and could hydrolyze 228–304 μg of lactose in 30 min with good stability (RSD = 4.4%) and reusability. The method proposed in this study effectively combined the advantages of sponge and cross-linking enzyme aggregates, successfully achieving the decomposition of lactose in milk and rapid separation of the product. It is worth mentioning that this study provided a more effective and convenient industrial option for the production of lactose-free milk.

Type 1 diabetes (T1D) is an autoimmune disease characterized by hyperglycemia caused by the destruction of insulin-producing β cells. Viral infection is an important environmental factor which is associated with the islet autoimmunity in genetically susceptible individuals. Loss of β-cells and triggering of insulitis following viral infection could result from several non-exclusive mechanisms. Despite a significant increase in ISG15 levels following viral infection, the specific role of ISG15 in the impairment of insulin-producing β-cells is unclear. To address this issue at the clinical level, we conducted this experimental work, and found elevated levels of ISG15 in the peripheral blood of T1D patients, suggesting a potential link between ISG15 and T1D. In the T1D animal model, we discovered that both ISG15 levels and cellular apoptosis were increased in pancreatic islet tissue. To investigate at the cellular level, we cultured MIN6 cells in the presence of supernatants derived from iBMDM cells transfected with poly(I:C) (PIC), a viral mimic. This exposure led to an upregulation of ISG15 expression in MIN6 cells, which was accompanied by the suppression of their functional capabilities and viability. Intriguingly, the direct transfection of MIN6 cells with PIC increased the expression of ISG15. We further found that elevated levels of ISG15 had a direct inhibitory effect on insulin secretion and it also contributed to β-cell apoptosis in a TNF-α-dependent manner. In conclusion, our study revealed a potential underlying mechanism through which ISG15 increases the apoptosis of β-cells, providing valuable insights that could facilitate the development of T1D treatment strategies.

As the improvement of consciousness of environment, a considerable effort has been made to shift from petroleum-based plastics to biodegradable alternatives, especially in the food packaging industry. In this work, we develop an innovative food packaging film by encapsulating quercetin (Que) within β -cyclodextrin ( β -CD) to create β -CD-Que inclusion complexes, which are then dispersed in a matrix of tragacanth gum (TG) and carboxymethyl chitosan (CMCS) using a solution casting technique. β -CD-Que inclusion complexes were characterized using various methods, including 1 H NMR, 2D ROESY and phase solubility testing, to elucidate the interaction between Que and β -CD. ATR–IR, X-ray diffraction, and SEM were employed to confirm the uniform dispersion of β -CD-Que within the macromolecular substrate. The incorporation of β -CD-Que considerably improves the mechanical properties of films, increasing the elongation at break from 13.68 % to 25.74 %. Rheological tests demonstrate that the viscosity of the film-forming complex solution became larger. The films were demonstrated to reduced water vapor and oxygen permeability, possessing remarkable antioxidant capability evidenced by DPPH and ABTS radical scavenging rates of 78.01 % and 99.59 %, respectively. The TG/CMCS/ β -CD-Que composite film shows excellent potential for food packaging and preservation applications.

β- Cyclodextrin ( β -CD) exhibits exceptional chiral recognition ability, and covalent organic frameworks (COFs) have been reported as highly promising chiral carriers. Therefore, incorporating cyclodextrin into COFs may result in chiral covalent organic frameworks (CCOFs) suitable for chiral applications. In this work, a solvothermal method was employed to synthesize a classical porous covalent organic framework (COF-V), while β -CD underwent a two-step modification process, resulting in the synthesis of mercapto- β -cyclodextrin ( β -CD-SH). Subsequently, a β -CD-based covalent organic framework (COF-S-CD) was synthesized through a Thiol-Ene “click” reaction. The synthesized COF-S-CD was successfully applied in enantioselective liquid-liquid extraction for five racemic arylcarboxylic acids, including 2-phenylpropionic acid, 2-(4-methylphenyl) propionic acid, 2-(3-chlorophenyl) propionic acid, 2-(4-nitrophenyl) propionic acid and α -cyclohexylmandelic acid. Under optimized conditions, enantioseparation factors for the five racemates reached 1.78, 2.20, 1.84, 1.76 and 1.96, respectively, which indicated a significant enhancement in chiral recognition ability upon the incorporation of cyclodextrin into the COF material. This new application suggested that the synthesis of covalent organic framework materials incorporating β -CD, with the ability to uniformly disperse in the aqueous phase, holds promising potential for highly effective enantioselective liquid-liquid extraction.

Background Macrophages play an important role in the pathogenesis of ulcerative colitis (UC). We will explore the effects of sodium butyrate (SB) on macrophage function. Methods The targets of butyric acid were identified using SwissTargetPrediction database and surface plasmon resonance (SPR). Limited proteolysis mass spectrometry (Lip-MS) was used to further investigate the binding sites of butyric acid with its targets and molecular docking was employed to simulate their binding modes. Macrophage polarization model was established with lipopolysaccharide (LPS) in vitro. Takeda G protein-coupled receptor 5 (TGR5) and β-arrestin2 expression and macrophage polarization markers were detected with or without SB.Results TGR5 was identified as the target of butyric acid. Moreover, the amino acid regions 275–286 and 321–330 of TGR5 (GPBAR1 [275–286] and GPBAR1 [321–330]) were the potential binding regions for butyric acid. Based on molecular docking analysis, butyric acid formed effective hydrogen-bonding interactions with ASP-284 and TYR-287 of TGR5. In cell experiments, LPS inhibited the expression of TGR5, β-arrestin2, IL-10, ARG1, and CD206 and increased the expression of IL-1β, iNOS, and CD86, while SB reversed the effect of LPS. SBI-115, a TGR5 antagonist, and knockdown of β-arrestin2 inhibited the effect of sodium butyrate. INT-777, a TGR5 agonist, reversed the inhibitory effect of knockdown of β-arrestin2.Conclusion SB inhibited M1-like polarization and promoted M2-like polarization induced by LPS via TGR5/β-arrestin2 in RAW264.7 cells and TGR5 was the target of SB.

Inhibition of gut β-glucuronidase (GUS) protects the gastrointestinal tract by reducing local exposure to toxic aglycones from inactive glucuronides. Development of highly safe GUS inhibitors has attracted extensive attentions. The current study demonstrates that natural and synthetic vitamin E (VE) analogues all display potent inhibition toward GUS. dl-α-tocopherol acts as the most potent inhibitor with the Ki value of 0.14 μM, while the other analogues have Ki values below 15.1 μM. Docking analysis indicates that the amino residues in the active pocket of GUS form various hydrophobic interactions with both the chromanol ring and side chain of VE, as well as hydrogen bonds with the hydroxyl group. It is predicted that GUS inhibition readily occurs owing to the daily intake of common plant foods. The current study offers highly safe inhibitors for GUS, as well demonstrates a novel nonantioxidant target for VE providing new insights into the vitamin’s gastrointestinal protection.

Near-infrared (NIR) luminescent materials based on Fe 3+ are growing interest in recent years because of the low-toxicity, high-efficiency, and adjustable emission. In this work, a broadband NIR emitting β-Mg 3 Ga 2 GeO 8 : Fe 3+ with [Ga/GeO 4 ] tetrahedron for the replacement of Fe 3+ ion is synthesized in air. X-ray diffraction patterns are used to confirm the crystal structure and phase purity. The excitation spectrum is observed in the region from 210 to 550 nm due to the O 2− - Fe 3+ charge transfer and the d – d electron transitions in 3 d 5 orbit of Fe 3+ . A broadband NIR emission of β-Mg 3 Ga 2 GeO 8 : Fe 3+ with full width at half maximum (∼108 nm) in the region from 610 to 880 nm is assigned to the transition 4 T 1 ( 4 G) → 6 A 1 ( 6 S) of Fe 3+ ion. Photoluminescence (PL) spectra under different excitation wavelengths, concentration dependent PL spectra, time-resolved PL spectra, temperature dependent PL spectra, and lifetimes of β-Mg 3 Ga 2 GeO 8 : Fe 3+ are investigated. We confirm the luminous center (Fe 3+ ), the optimal Fe 3+ concentration (0.8 mol%), and the good thermal stability of β-Mg 3 Ga 2 GeO 8 : Fe 3+ . The luminescence mechanism is analyzed theoretically by the Tanabe-Sugano and energy level diagrams of Fe 3+ in β-Mg 3 Ga 2 GeO 8 : Fe 3+ , and the concentration and thermal quenching mechanisms are explained. The research results will be conducive to the development of new Fe 3+ -doped luminescence materials.

TEMPO (2,2,6,6-tetramethylpiperidine-1-oxyl radical)-mediated 6-carboxy β-chitin derivatives (T-chitin) with different carboxylate content were successfully synthesized by controlling the addition level of NaClO as the primary oxidant. The structural and biochemical properties of the derivatives were investigated. The carboxylate contents of the derivatives calculated by electrical conductivity titration were 1.33, 1.68, 1.80, and 2.08 mmol/g, respectively. The yield of T-chitin with carboxylate content of 2.08 mmol/g reached 74.55%. T-chitin exhibited stronger bile acid binding capacities than that of β-chitin. The scavenging ability of T-chitin against hydroxyl radicals improved with increasing concentration, and EC 50 values were below 1.2 mg/mL. All T-chitin exhibited a strong ferrous ion chelating effect. At 8 mg/mL, the chelating effects of T-chitin with carboxylate content of 0.81 mmol/g reached 80.15%. These results showed that T-chitin had good bile acid binding capacity and antioxidant activities and it may be a potential antioxidant in vitro .
