Construction of asymmetric dual-layer polysaccharide-based porous structure on multiple sources for potential application in biomedicine

Huimin Li,Jundan Feng,Kejing Yu,Shuiping Liu,Hongbo Wang,Jiajia Fu

Biomedical materials can produce high efficiency and special behavior with an integrated internal structure. It is possible that changing the structure of biomedical materials could extend and promote the application of eco-friendly and multifunctional biomaterials. However, the instantaneous formation of complex structures between tannic acid (TA) and polysaccharides is disrupted, and the reconstruction of the new porous structure becomes a key issue. Here, we present an innovative one-step forming method for an asymmetric dual-layer porous structure of carboxymethyl chitosan (CC)/sodium alginate (SA)/TA, which can be utilized in various biomedical applications. Even after 6 months of storage, it still demonstrates a range of desirable properties including tailorable performance, efficient antibacterial activity , ultrarapid antioxidant activity , low differential blood clotting index and cytotoxicity. This suggests its potential for regulating and controlling wound bleeding, providing flexible possibilities for potential applications in biomedicine .

Preparation and characterization of bioactive binary composite film based on gelatin–dextran incorporated with citronellal and α-tocopherol

Chenlinrui Yue,Fengjiao Fan,Yinji Chen,Yong Fang,Hongkai Xie

Biodegradable active packaging films with antimicrobial and antioxidant functions have received much attention due to the food safety and plastic pollution issues. This study prepared and characterized a novel bioactive binary composite film based on gelatin (Ge) and dextran (Dx) incorporated with various concentrations (0%, 0.3%, 0.6%, 0.9%, 1.2%, w/w) of citronellal (Cit) and α-tocopherol (Toc). The thickness, L *, b *, total color difference, water contact angle, opacity, and crystallinity of film significantly increased, whereas the film solubility, swelling degree, water vapor permeability, tensile strength, elongation at break, and a * of film significantly decreased with the increase of Cit and Toc concentrations. However, the addition of Cit and Toc had no significant effect on the film's moisture content and thermal stability. Fourier transforms infrared spectral pattern indicated that physical interactions (H-bonding) between biopolymer matrix and active fillers were formed. Scanning electron microscope images showed that the film added with active fillers had a rough surface and a spongelike cross-section structure compared to control film. Moreover, the release curves of Cit and Toc showed that the film in 95% ethanol solution had the longest release time. Furthermore, the film added with Cit and Toc effectively inhibited the growth of pathogenic and spoilage bacteria ( Shewanella putrefaciens and Escherichia coli ) and quenched the free radicals (2,2-diphenyl-1-picrylhydrazyl and 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)). Overall, it can be inferred that the Ge–Dx-based composite film incorporated with Cit and Toc is a potential antibacterial and antioxidant biodegradable material for food packaging applications to maintain food quality.

Asymmetric Janus fiber membrane for preventing cerebrospinal fluid leakage and promoting bone regeneration

Xiaomei Xia,Ziyang Liu,Hongshui Wang,Xianrui Zou,Lei Zhang,Kun Qin,Jiantao Zheng,Huan Zhou,Lei Yang,Chunyong Liang

Repairing skull base defects caused by pituitary tumor surgery remains challenging as it is difficult to restore its structural integrity and avoid cerebrospinal fluid leakage. To address this issue, this study aimed to prepare a Janus bilayer membrane with asymmetric wetting properties (hydrophobicity/super hydrophilicity) using polycaprolactone (PCL), gelatin (Gel), and polydopamine (PDA) through a layer-by-layer electrospinning technique. Randomly stacked hydrophobic PCL fibers were designed as the upper layer to control leakage of cerebrospinal fluid. Meanwhile, the bottom layer composed of hydrophilic PG-PDA nanofibers was utilized to promote the skull base bone repair. The experimental results showed that under a pressure of 200 mm water column, the Janus bilayer membrane could efficiently prevent leakage for several days. The prepared Janus PCL/PG-PDA membrane revealed excellent mechanical properties to mimic bone ECM and showed surface bioactivity to accelerate mineralization. The results of in vitro cell experiments indicated that the Janus membrane prepared in this study displays exceptional biocompatibility, with the PCL/PG-0.5%PDA fiber membrane demonstrating the highest capacity to promote osteogenic differentiation. These observations suggest that the asymmetric Janus fiber membrane (PCL/PG-PDA) has the potential to serve as a promising candidate for skull base repair.

Stability and antioxidant activity of 10 isoflavones and anthocyanidins during in vitro digestion

Yaoyao Qi,Leyi Xie,Zeyuan Deng,Bing Zhang,Hongyan Li

The stability and antioxidant activity changes of 10 phytochemicals (isoflavones: genistein, genistin, daidzein, daidzin, formononetin, and anthocyanidins: delphinidin, cyanidin, pelargonidin, malvidin, petunidin) and food representatives (soybeans and blueberries) during in vitro digestion were investigated. Results indicated that the stability of isoflavones and anthocyanidins decreased with digestion. Among the isoflavones, the loss rate of formononetin was the largest, reaching 98.24%, and that of daidzein was the smallest (22.78%). And after intestinal digestion, anthocyanidins degraded into phenolic acids and other compounds, and the loss rate was above 95%. The loss of phenolic compounds in food extract was smaller than the standards, and the loss rate of phenolic compounds in hydrolyzed blueberry extracts (90.66%–96.04%) was higher than that of soybean extract (31.22%–59.74%). Moreover, the antioxidant activity of isoflavones and anthocyanidins decreased after the oral phase. During the gastrointestinal phase, the ABTS, DPPH and FRAP of isoflavones increased, while the ORAC and CAA decreased. The antioxidant activity of anthocyanidins increased in the gastric digestion and decreased in the intestinal phase. The glucoside linkage in the A ring and the hydroxyl methoxylation in the B ring weakened the antioxidant activity of isoflavones, while the hydroxylation and methoxylation of B rings of anthocyanidins could enhance thier antioxidant activity. The changes in antioxidant activity of the two food extracts were consistent with their contents.

Demethylation of ethanol organosolv lignin by Na2SO3 for enhancing antioxidant performance

Ying Han,Zihao Ma,Xing Wang,Yuanyuan Sheng,Yu Liu

Natural polyphenol antioxidants have excellent antioxidant activity and degradability which are considered as one of the possible alternatives to chemical synthetic antioxidants. Here, lignin-based polyphenols were prepared from poplar lignin, pine lignin and rice husk lignin by demethylation, and no toxic reagents were used in the preparation process. The phenolic hydroxyl groups of all three lignin were increased and the methoxyl content was significantly reduced after the demethylation without degradation or structural changes in the lignin as shown by GPC and 2D HSQC analyses. Furthermore, the antioxidant activity of the modified lignin was significantly increased. In this paper, a green and efficient strategy was developed to prepare lignin-based polyphenols with high phenolic hydroxyl content, low molecular weight , strong antioxidant activity and low cytotoxicity, which have a promising future in the field of antioxidant applications.

Nano Self-Assemblies of Caffeic Acid–Fibronectin Mimic a Peptide Conjugate for the Treatment of Corneal Epithelial Injury

Yiping Wu,Lulu Du,Xiaoning Xu,Yuhan Hu,Jia Liu,Jingwei Zhang,Lei Lei,Wenfang He,Zihao Sheng,Yuanao Ni,Jia Qu,Xingyi Li,Jun Jiang

Rapid corneal re-epithelialization is important for corneal wound healing. Corneal epithelial cell motility and oxidative stress are important targets for therapeutic intervention. In this study, we covalently conjugated the antioxidant caffeic acid (CA) with a bioactive peptide sequence (PHSRN) to generate a CA–PHSRN amphiphile, which was formulated into nanoparticular eye drops with an average size of 43.21 ± 16 nm. CA–PHSRN caused minimal cytotoxicity against human corneal epithelial cells (HCECs) and RAW264.7 cells, exhibited an excellent free radical scavenging ability, and remarkably attenuated reactive oxygen species (ROS) levels in H2O2-stimulated HCECs. The antioxidant and anti-inflammatory activities of CA–PHSRN were assessed in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. The results show that CA–PHSRN treatment effectively prevented LPS-induced DNA damage and significantly reduced the levels of LPS-induced pro-inflammatory cytochemokines (i.e., iNOS, NO, TNF-α, IL-6, and COX-2) in a dose-dependent manner. Moreover, using a rabbit corneal epithelial ex vivo migration assay, we demonstrated that the proposed CA–PHSRN accelerated corneal epithelial cell migration and exhibited high ocular tolerance and ocular bioavailability after topical instillation. Taken together, the proposed CA–PHSRN nanoparticular eye drops are a promising therapeutic formulation for the treatment of corneal epithelial injury.

Physicochemical properties, antioxidant activities and comprehensive phenolic profiles of tea-macerated Chardonnay wine and model wine

Zijian Liang,Pangzhen Zhang,Wen Ma,Xin-An Zeng,Zhongxiang Fang

A new type of flavored wine was produced by macerating either green tea or black tea into Chardonnay wine and model wine respectively, where the physicochemical properties (pH, titratable acidity, color) were modulated. Significant ( p  < 0.05) increases of total phenolic content and antioxidant activity (assessed by DPPH, FRAP and ABTS assays) were also observed in the tea macerated wines. A total of 160 phenolic and non-phenolic compounds were identified by HPLC-DAD-ESI-QTOF-MS/MS, where 55 phenolics were newly found in the tea macerated Chardonnay wine. The interaction between wine and tea phenolics led to additional 29 phenolic compounds and 4 non-phenolic compounds that were not found in either Chardonnay wine or tea. Catechin and epigallocatechin gallate were the most abundant phenolic compounds and contributed to the improved antioxidant activities. This study provided a promising prospect of tea as a novel additive in the production of flavored wine with enhanced functionalities.

Crosslinked fish scale gelatin/alginate dialdehyde functional films incorporated with carbon dots derived from pomelo peel waste for active food packaging

Yongshi Li,Junxian Yang,Linping Sun,Bingzhen Liu,Hui Li,Lincai Peng

A multifunctional and environmentally friendly composite film was developed by incorporating pomelo peel-derived carbon dots (PCDs) into a fish scale gelatin (FSG)/alginate dialdehyde (ADA) biopolymer matrix. ADA was used to reinforce the physicomechanical properties of the FSG film via Schiff base crosslinking. PCDs with strong antioxidant and antimicrobial activities were synthesized via a hydrothermal method . The effect of various PCDs content on the surface morphological, physicochemical, and functional characteristics of the composite films was investigated. The results showed that the introduction of PCDs into the FSG/ADA matrix effectively reinforced the mechanical performance, enhanced the water vapor and water resistance, increased UV-light blocking, conferred fluorescence properties, and improved the thermal properties of the composite films. Under 3 wt% PCDs content, the FSG/ADA/PCDs-3 % composite film not only presented significant antioxidant capacity with a radical scavenging rate of 91.71 % for DPPH and approximately 100 % for ABTS , but also exhibited excellent antimicrobial ability against bacteria and fungi. Results of a preservation experiment showed that the prepared FSG/ADA/PCDs-3 % film preserved the physiological qualities of strawberries post-harvest and extended their shelf-life to 7 days at room temperature. Overall, the fabricated FSG/ADA/PCDs composite films are promising for use in eco-friendly active food packaging.

Sustainable ultrasound-assisted extraction of Polygonatum sibiricum saponins using ionic strength-responsive natural deep eutectic solvents

Hongli Zhang,Xinpeng Li,Miao Kang,Zhanrong Li,Xiaowen Wang,Xu Jing,Jiajun Han

The sustainable extraction of saponins was investigated using natural deep eutectic solvents (NADESs) combined with ultrasound-assisted extraction. A novel NADES (butyric acid-urea) that was responsive to ionic strength was designed and used as the extractant. Ultrasound treatment and a catalyst ferric chloride with plant cell wall breaking function were applied to improve the extraction efficiency.   Since the solubility of the NADES varied significantly with ionic strength, 95% of NADES was readily separated from the water phase after the addition of sodium chloride, while saponins remained in the water phase for easy collection. The reuse capacity of NADES, the eco-friendliness of the extraction method, and the antioxidant activity of the extract were further evaluated.   NADES was continuously recovered and used to extract Polygonatum sibiricum powder: the yield of saponins did not decrease after five cycles of recovery and re-extraction. The penalty point on the “Eco-scale” suggested that the extraction method was “green” ( i.e . eco-friendly).   Compared with ethanol extracts, the NADES extracts showed a higher saponin concentration and antioxidant activity.   The study can contribute to the sustainable and green extraction of hydrophilic active substances in the food and pharmaceutical industries.

Response surface methodology-optimized extraction of flavonoids from pomelo peels and isolation of naringin with antioxidant activities by Sephadex LH20 gel chromatography

Yuchen Shangguan,Jing Ni,Lili Jiang,Yang Hu,Chuanbo He,Ying Ma,Guohong Wu,Hejian Xiong

In this study, flavonoids were extracted from pomelo peels and naringin was isolated from the flavonoid extract. The effects of extraction parameters, namely, ethanol concentration, solid-to-liquid ratio, and extraction time, on the yield of flavonoids extracted from pomelo peels were analyzed according to the Box-Behnken design of response surface methodology. The experimental conditions for flavonoid extraction were optimized, and naringin was separated from the extracted flavonoids using Sephadex LH-20 column chromatography. Experimental results showed that the influence of factors on the extraction rate of flavonoids from pomelo peels was in the order of ethanol concentration > solid-to-liquid ratio > extraction time, and the optimal extraction parameters were 85% ethanol concentration, 1:20 solid-to-liquid ratio, and 4-h extraction time for extracting flavonoids from pomelo peels. Under these conditions, the yield of flavonoids was 6.07 ± 0.06 mg/g. After three times of extraction, the flavonoid extraction rate reached 96.55%, and the residual naringin in the pomelo peels was 0.017 mg/g, at which point the bitterness in the pomelo peels disappeared. Two components, namely, PF1 and PF2, were separated from the crude flavonoid of pomelo peels through Sephadex LH20 column chromatography. PF2 was identified as naringin by high-performance liquid chromatography tandem mass spectrometry, with a purity of 95.7 ± 0.23%. Both flavonoids and PF2 exhibited good in vitro radicals scavenging activities on DPPH, ABTS, superoxide anion and hydroxyl.

Potential folate-producing strains and their applications in biofortification of fermented Moringa oleifera leaves powder

Huan Du,Yingyun Hu,Kaixiang Lu,Tao Li,Yang Tian,Yongjin Hu

Folate is an essential vitamin for the human body and must be obtained exogenously. It is recognized that some microorganisms are capable of producing folates during growth, revealing possibilities for innovations of natural folate biofortified foods. The aim of this study was to identify potential folate-producing strains to enhance folate content in fermented Moringa oleifera leaves powder (MOLP). Although most of the tested strains exhibited folate production, there were significant differences among them in terms of both quantity and distribution, whether intracellular or extracellular. 10 strains with potential for folate production were chosen for enhancing the folate content of MOLP. The two most promising strains, Lactiplantibacillus sp. LP1 and Saccharomyces cerevisiae Y-J-1, demonstrated high folate levels up to 1940 and 2115 μg/100 g dry matter, respectively. These levels were 1.6 and 1.8-fold the initial value in MOLP. Additionally, the fermentation of MOLP not only enhanced its nutritional quality but also preserved its significant bioactivity. This study provides the initial understanding of how selected microorganisms can naturally enrich folates in fermented MOLP, presenting an alternative solution to address the widespread issue of folate deficiency while promoting human health.

A Smart Stimulation–Deadhesion and Antimicrobial Hydrogel for Repairing Diabetic Wounds Infected with Methicillin-Resistant Staphylococcus aureus

Zhongwu Bei,Linghong Zhang,Jianan Li,Qi Tong,Kun Shi,Wen Chen,Yan Yu,Ao Sun,Yang Xu,Jie Liu,Zhiyong Qian

The healing of chronic diabetic wounds is a common and significant challenge in the medical field. Despite extensive efforts, the development of hydrogel dressings with satisfactory functionality remains an ongoing concern. In this study, a multifunctional hydrogel wound dressing (PAN/Ag-PLG) with adhesion, antibacterial, hemostatic, and other properties, which can effectively repair diabetic wounds infected with methicillin-resistant Staphylococcus aureus (MRSA), is presented. The hydrogel dressing is composed of gallic acid (GA)-functionalized polylysine (PL)-reduced silver nanoparticles (Ag-PLG), oxidized hyaluronic acid (OHA), and cross-linked polyacrylic acid grafted with N -hydrosuccinimide ester. Notably, compared to most conventional wound dressing that lack adhesion or are difficult to remove, the prepared hydrogels exhibit excellent adhesion and mild stimulation-triggered detachment. In vitro and in vivo experiments reveal that the PAN/Ag-PLG hydrogel exhibits outstanding biocompatibility and antibacterial properties and promotes diabetic wound repair by reducing oxidative damage and promoting cell migration and angiogenesis. The smart PAN/Ag-PLG hydrogel reported in this study provides an approach for the potential clinical development of painless antibacterial dressings.
