Perturbation of energy metabolism is an essential feature during infection and inflammation. TANK-binding kinase 1 (TBK1) is crucial for initiating the innate immune response against viral infection, although aberrant and ongoing TBK1 activation induces excessive production of type I interferons (IFN-I). Nonetheless, the mechanisms whereby energy metabolism controls TBK1 activation remain unclear. Here, we elucidate a mechanism linking energy metabolism to the inhibition of TBK1-induced IFN-I responses via the immune response gene 1 (IRG1)-itaconic acid axis. Mechanistically, itaconic acid and its derivatives alkylated TBK1 at Cys605, thereby disrupting TBK1 dimerization and rapid activation. IRG1, the enzyme that catalyzes itaconic acid production, is upregulated during late-phase viral infection and acts as a feedback regulator to restrain TBK1 activity. We developed itaconic acid-based compounds ITA-5/ITA-9 as alternative TBK1 inhibitors. ITA-5/ITA-9 effectively limited excess IFN-I-mediated hyperinflammation. These findings provide a promising therapeutic strategy for treating diseases mediated by aberrant TBK1 activation.

In this study, the copper-nickel (Cu–Ni) bimetallic electrocatalysts for electrochemical CO 2 reduction reaction(CO 2 RR) are fabricated by taking the finely designed poly(ionic liquids) (PIL) containing abundant Salen and imidazolium chelating sites as the surficial layer, wherein Cu–Ni, PIL–Cu and PIL–Ni interaction can be readily regulated by different synthetic scheme. As a proof of concept, Cu@Salen-PIL@Ni(NO 3 ) 2 and Cu@Salen-PIL(Ni) hybrids differ significantly in the types and distribution of Ni species and Cu species at the surface, thereby delivering distinct Cu–Ni cooperation fashion for the CO 2 RR. Remarkably, Cu@Salen-PIL@Ni(NO 3 ) 2 provides a C2+ faradaic efficiency (FE C2+ ) of 80.9% with partial current density ( j C 2+ ) of 262.9 mA cm −2 at −0.80 V (versus reversible hydrogen electrode, RHE) in 1 m KOH in a flow cell, while Cu@Salen-PIL(Ni) delivers the optimal FE C2+ of 63.8% at j C2+ of 146.7 mA cm −2 at −0.78 V. Mechanistic studies indicates that the presence of Cu–Ni interfaces in Cu@Salen-PIL@Ni(NO 3 ) 2 accounts for the preserve of high-valence Cu(I) species under CO 2 RR conditions. It results in a high activity of both CO 2 -to-CO conversion and C–C coupling while inhibition of the competitive HER.

Cu(I)-based halide scintillators are promising candidates for X-ray detection due to excellent scintillation and low-cost solution processability. However, the rational design of Cu(I)-based halide scintillators remains challenging due to insufficient theoretical frameworks elucidating their structure-property correlations. In this work, two Cu(I)-based hybrid metal halides, Cu 2 I 4 -IC (Ionic compound) are designed and Cu 4 I 4 -CC (Coordination compound) via bonding mode control engineering. The Cu 2 I 4 -IC exhibits blue emission, and the Cu 4 I 4 -CC exhibits yellow emission, and the photoluminescence quantum yield (PLQY) of Cu 2 I 4 -IC and Cu 2 I 4 -IC are 91% and 100% respectively. The non-radiative transitions are reduced due to the high rigidity of Cu 4 I 4 , resulting in a light yield up to 67 500 photons MeV −1 and a detection limit as low as 47.3 nGy s −1 of Cu 4 I 4 -CC. Additionally, a high-performance scintillator hydrogel based on Cu 4 I 4 -CC with polyvinyl alcohol (PVA),H 2 O and dimethyl sulfoxide (DMSO) is innovatively developed. Owing to the high transmittance, the large-area scintillator hydrogel film (10 × 10 cm 2 ) achieves an X-ray imaging resolution of 14 lp mm −1 . Furthermore, the synergistic effects of hydrogen bonding and coordination bonding endow the hydrogel scintillator with excellent plasticity and flexible stretchability. The Cu 4 I 4 -CC@PVA hydrogel proves promising for X-ray imaging with excellent stability in harsh environments, matching state-of-the-art scintillators.

Hybrid luminescent metal halides has emerged as the state-of-the-art scintillators, while remaining challenges for enhancing light yield and X-ray imaging capability. Herein, halogen engineering is reported to optimize Cu(I)-based hybrid halides by enhancing the X-ray absorption cross-section, resulting in an increase of light yield by 188% to 79,770 photons MeV −1 from (C 12 H 30 N 2 ) 2 Cu 6 Cl 10 to optimized (C 6 H 15 N) 2 CuI 3 . The transparent glassy scintillation screen with optical transparency beyond 82% demonstrates good thermal stability and high mechanical flexibility. More importantly, the high X-ray imaging resolution of 20.5 lp mm −1 is achieved based on the Cu-I-based glassy scintillation screen. The combined merits of enhancing scintillation performance and X-ray imaging resolution provide a platform for developing halide glass scintillators and enriching their applications in radiology and security screening.

Single-atom catalysts (SACs) catalysts have unique electronic properties and geometric structures, offering maximum atom-utilization efficiency. Noble metal-free SACs anchored within metal–organic frameworks (MOFs) enable precise tuning of electronic structures and significantly enhance catalytic activity beyond conventional porous materials. Herein, we synthesized the synthesis of Cu(I)/Ce-MOF-808-EA by anchoring atomically dispersed Cu(II) species onto the open Ce 6 nodes of the Ce-MOF-808 framework, followed by in situ reduction to Cu(I) via ethanol treatment and thermal activation processes. The atomic dispersion and valence transformation of Cu sites on Ce-MOF-808 were confirmed by X-ray Absorption Spectroscopy (XAS), high-angle annular dark-field scanning transmission electron microscopy (HAADF-STEM), and X-ray photoelectron spectroscopy (XPS). The resulting Cu(I)/Ce-MOF-808-EA exhibited outstanding catalytic performance for the hydrogenation of Dicyclopentadiene (DCPD), achieving 100 % conversion, 91.54 % selectivity toward Dihydrodicyclopentadiene (DHDCPD), and 8.45 % selectivity toward Tetrahydrodicyclopentadiene (THDCPD) at 100 °C, 2 MPa. Notably, Cu(I)/Ce-MOF-808-EA outperformed other transition metal catalysts (Co/Ce-MOF-808-EA, Ni/Ce-MOF-808-EA and Zn/Ce-MOF-808-EA). For the first time, Density Functional Theory (DFT) calculations, combined with experimental investigations, revealed a low-energy reaction pathway for DCPD hydrogenation, wherein Cu(I) sites, in cooperation with adjacent oxygen atoms, act as bi-active centers to heterolytically cleave H 2 into proton and hydride species (*H δ+ -H δ- ) for subsequent hydrogenation. This study underscores the potential of MOFs as versatile platforms for engineering single-atom catalysts with enhanced catalytic properties.

Hybrid copper(I)–based halides are important materials in radiation detection and anticounterfeiting owing to their excellent luminescence efficiency and low toxicity. This study employed a chiral racemic strategy to synthesize ( Rac -MPZ) 2 Cu 2 I 6 (where MPZ represents 2-methylpiperazine) single crystals with excitation-dependent dual-color emission. ( Rac -MPZ) 2 Cu 2 I 6 has a zero-dimensional crystal structure, which is different from the two-dimensional structure of chiral ( R / S -MPZ) 2 Cu 2 I 6 . Interestingly, ( Rac -MPZ) 2 Cu 2 I 6 exhibits excitation-dependent cyan and red emissions at room temperature (298 K), with the photoluminescence quantum yield of the cyan emission peak approaching 90 %. Its luminescence mechanism is proposed through density functional theory calculations and theoretical experiments. ( Rac -MPZ) 2 Cu 2 I 6 exhibits excellent X-ray absorption and responsivity, with a light yield of up to 34,500 photons/MeV. X-ray imaging screens developed with ( Rac -MPZ) 2 Cu 2 I 6 show an excellent resolution of >10 lp/mm. Therefore, the synthesized material has great potential for practical applications as a scintillator. In addition, its unique excitation-dependent dual-color emission property at room temperature and temperature-dependent thermochromicity have excellent potential in anticounterfeiting and information encryption, which can realize multimode and multilevel anticounterfeiting or encryption effects. This study also provides new prospects for developing multifield applications of copper(I)-based halides.

Aristolochic acids (AAs) are a family of nitrophenanthrene carboxylic acids. Some AAs-containing herbs are used as raw materials in medicines, health foods, cosmetics, and dietary supplements. However, aristolochic acid I (AAI) as the main active component of AAs, is nephrotoxic, mutagenic, and carcinogenic to humans. Thus, it is essential to develop effective techniques for determining AAI in relevant products. In this study, an anti-AAI monoclonal antibody (mAb) with high sensitivity was generated. Based on the mAb, a chemiluminescence enzyme immunoassay (CLEIA) was developed, exhibiting an IC 50 of 20 ng/mL and a linear range from 2.5 ng/mL to 250 ng/mL. The limit of detection (LOD) of the CLEIA was 1.96 ng/mL. The recovery rates of AAI in spiked patent medicines and cosmetics ranged from 79.7 % to 121.7 % with coefficient of variations (CVs) ranging from 1.2 % to 13.6 %. A good correlation coefficient of 0.9591 was observed between CLEIA and high-performance liquid chromatography (HPLC) results, indicating that the developed CLEIA is accurate and reliable for the determination of AAI in patent medicines and cosmetics.

Perovskite's surface defects trigger deep level traps and energy misalignment, resulting in substantial interface recombination and energy loss in perovskite solar cells (PSCs). Herein, 9-fluoreneacetic acid (FAA), a self-assembled molecule (SAM), is employed to passivate the interface defects and modulate energy alignment. SAM modification reduces the defect density from 6.37 × 10 15 to 3.11 × 10 15 cm −3 and produces a p-type surface with an upward band bending, thus constructing a well-defined n-i-p heterojunction for efficient charge separation. Accordingly, the target PSC realizes 24.75% power conversion efficiency (PCE) and retains 92% for 1100 h during maximum power point tracking (MPPT) at room temperature. Furthermore, over 80% of initial PCE has been reserved after 2500 h aging in 25–30% relative humidity (RH). This SAM strategy is expected to enhance the efficiency and stability for n-i-p PSCs.

Bioactive synthetic scaffolds with 3D porous structure are the most attractive materials among various guided bone regeneration membranes. However, its osteogenic potential is still insufficient. This study was aimed to develop a Calcium surface-anchored Collagen I-PLGA/PCL scaffolds (PP/COL I-pDA-Ca) with enhanced osteogenicity. PP/COL I-pDA-Ca was electrospun from a collagen I-blended PLGA/PCL matrix and then modified by the chelation of Ca-ions via mussel-inspired polydopamine coating. PLGA/PCL, PLGA/PCL-polydopamine, PLGA/PCL-pDA chelated by Ca-ions were used as controls. Osteogenic effects of the scaffolds were examined using MC3T3-E1 cell culture. PP/COL I-pDA-Ca maintained 3D porous architecture with interconnected pores formed by randomly-oriented filamentous fibers and MC3T3-E1 cells cultured on it for 12 h or 24 h were more stretched and spread than those on the controls. PP/COL I-pDA-Ca significantly upregulated α10, α11 and β1 integrin expression after 48 h culture. ALP activity, OCN, OSX, BMP2 and RUNX2 expression of MC3T3-E1 cells cultured on PP/COL I-pDA-Ca scaffolds for 7 and 14 days were substantially enhanced when compared to controls ( p < 0.05). The pDA-based Ca chelation, COL I incorporation and 3D bionic structure promoted cell adhesion and osteogenic differentiation of MC3T3-E1 cells. This novel PP/COL I-pDA-Ca scaffolds is an attractive alternative for guided bone regeneration.

The authors describe a silver(I) mediated fluorescent assay for glutathione (GSH). An allosteric oligonucleotide strand containing a G-rich sequence is used to produce a G-quadruplex, and N-methylmesoporphyrin IX (NMM) is chosen as the fluorescent probe. In the absence of Ag(I), the DNA strand is partially intramolecularly hybridized to form a hairpin structure wherein the G-rich sequence is partially caged. On addition of Ag(I), the hairpin is disrupted by forming C-Ag(I)-C base pairs. As a result, the G-rich sequence is released and folds into a G-quadruplex structure, which is able to bind NMM to generate strong fluorescence at 612 nm. However, in the presence of GSH, due to the strong binding ability between GSH and Ag(I), the C-Ag(I)-C structure is not formed. Hence, the DNA probe reverts back to its original structure and fluorescence is not increased. Based on these findings, a method was worked out that has a detection limit as low as 3.5 nM. Due to the inherent selectivity of the interaction between GSH and Ag(I), the method is highly selective over common potential interfering species. It was successfully applied to the fluorometric determination of GSH in cell extracts.

Herein, a dual-signal electrochemical biosensor has been developed by self-assembly of pH-activatable i-motif probes on magnetic microparticles (MMPs) coupled with DNA walker for signal amplification. In this study, the cytosine (C)-rich single-stranded DNAs are hybridized with DNA initiators to obtain the long-nicked duplexes with repeated units, which are further captured on MMPs to form the magnetic i-motif containers. The resulting duplexes contain abundant G–C base pairs, thus providing extensive binding sites for doxorubicin (DOX). At acidic pH, the C-rich sequences are folded into i-motif structure, resulting in the release of DOX and walker initiators. In this case, the liberated DOX is adsorbed on graphene quantum dots-modified glassy carbon electrode via π–π interaction, while the walker initiators as a moving part can catalyze the hybridization between MB-modified fueler DNA and tracker DNA on electrode, contributing to the generation of dual electrochemical signals induced by MB and DOX. Importantly, the magnetic separation can effectively reduce the background, achieving sensitive biosensing of pH ranging from 4.0 to 7.4 with excellent stability. Moreover, the proposed dual-signal electrochemical biosensor has been successfully applied for accurate monitoring of pH in human serum, which holds great potential in pH-dependent bioassays, especially in ultra-micro analysis for clinical applications.

Plants have evolved intricate strategies to cope with various abiotic stresses. Ubiquitin-mediated protein degradation plays a key role in plant development as well as abiotic stress tolerance. In this study, we identify LAO1, an F-box protein with unknown function, as a negative regulator of plant fitness during nitrogen starvation. DOMINANT SUPPRESSOR OF KAR 2 (DSK2) interacts with and mediates the autophagic degradation of LAO1 protein during nitrogen starvation. The loss of LAO1 improves the fitness of an autophagy-deficient mutant, atg5-1, under nitrogen starvation. Intriguingly, mutations in DSK2 facilitate rather than reduce plant growth after nitrogen starvation. This unexpected effect of DSK2 knockout led us to discover that DSK2 also interacts with and degrades a group of class I TCP transcription factors. Phenotypic observations demonstrate that class I TCPs are crucial for plant adaptation to nitrogen starvation. Moreover, genetic analyses indicate that class I TCPs function downstream of LAO1 and counteract its negative effects. Collectively, our findings unveil a previously undescribed regulatory network governing plant fitness during nitrogen starvation.
