A hybrid nanomaterial with NIR-induced heat and associated hydroxyl radical generation for synergistic tumor therapy

Cheng Zhang,Wen-Long Liu,Xue-Feng Bai,Si-Xue Cheng,Zhen-Lin Zhong,Xian-Zheng Zhang

Although photothermal therapy (PTT) and photodynamic therapy (PDT) are widely commended for tumor treatment recently, they still suffer severe challenges due to the non-specificity of photothermal agents (PTAs)/photosensitizers (PSs) and hypoxic tumor microenvironment . Here, an oxygen independent biomimetic nanoplatform based on carbon sphere dotted with cerium oxide and coated by cell membrane (MCSCe) was designed and synthesized with good biocompatibility , homologous targeting ability, and improved photophysical activity. Notably, MCSCe could realize accumulation of hydrogen peroxide (H 2 O 2 ) in tumor cells and hyperthermia under single laser (808 nm) irradiation, which were simultaneously utilized by itself to produce more toxic hydroxyl radical ( OH). Resultantly, the synergistic therapeutic effect against tumor cells was obtained under near infrared (NIR) laser irradiation.

A hematoporphyrin and indocyanine green co-delivery system with NIR triggered-controllable photoactivities for photodynamic therapy

Ziying Li,Tingting Lv,Yingying Zhang,Liang Xu,Lu Zhang,Xiuying Wang,Haijun Chen,Yu Gao

Photodynamic therapy (PDT) was hindered by the poor water solubility and long-enduring photoactivities of photosensitizers . Here, we developed a novel controllable photodynamic agent CPHI based on β-cyclodextrin modified polyamidoamine (PAMAM) dendrimer (CP) co-delivery of indocyanine green (ICG) and hematoporphyrin (HP). The CPHI possessed appropriate particle sizes and zeta potentials , considerable photostability, and NIR-triggered release property. The fluorescence of HP could be quenched by ICG when HP and ICG were co-loaded inside the CP, and the quenching efficiency was related with the weight ratio of HP and ICG. When the feeding weight proportion of HP and ICG is 1/10, the fluorescence of HP was quenched by 87.3% in CPHI. Under 808 nm laser irradiation , the ICG in CP was degraded, and the fluorescence of HP could recover. The recovered HP was capable of generating reactive oxygen species (ROS) for PDT with 606 nm laser irradiation. The cytotoxicity and cell apoptosis studies showed that CPHI could effectively induce MCF-7 cells apoptosis with PDT effects under 808/660 nm laser. These results suggested that CPHI could be an efficient cancer therapeutic agent for controllable PDT with low dark toxicity.

DOX/ICG Coencapsulated Liposome-Coated Thermosensitive Nanogels for NIR-Triggered Simultaneous Drug Release and Photothermal Effect

Lixia Yu,Anjie Dong,Ruiwei Guo,Muyang Yang,Liandong Deng,Jianhua Zhang

Chemo-photothermal therapy has shown enormous potential in treating cancer. To achieve the chemo-photothermal synergistic effect, an efficient nanoparticulate system with the ability for simultaneous codelivery of chemotherapeutic drug and photothermal agent as well as photothermal-triggered drug release is highly desirable. Herein, an in situ polymerization within liposome template was designed to prepare liposome-coated poly(N-isopropylacrylamide-co-acrylamide) (P(NIPAM-co-AAM)) nanogels, which can efficiently coencapsulate a NIR dye indocyanine green (ICG) and high amount of doxorubicin hydrochloride (DOX). The DOX/ICG coloaded hybrid nanogels, denoted as DI-NGs@lipo, integrated the desirable functions of PEGylated liposomes and thermosensitive nanogels. The PEGylated liposome shell provided excellent storage stability, hemodynamic stability, and fluorescence stability. Meanwhile, the thermosensitive P(NIPAM-co-AAM) nanogels core endowed DI-NGs@lipo with volume phase transition temperature (VPTT) at about 40 °C, allowing for thermo-controlled transformation and drug release. The significant photothermal effect of DI-NGs@lipo and the simultaneous hyperthermia-triggered DOX release were observed under NIR light irradiation. The DI-NGs@lipo was demonstrated to be uptaken by 4T1 murine breast cancer cells via endocytosis, enhancing the distribution of DOX in the cell nucleus. Compared with chemo or photothermal treatment alone, the combination treatment of DI-NGs@lipo with NIR light irradiation induced significantly higher cytotoxicity to 4T1 cells, demonstrating the chemo-photothermal synergistic therapeutic effects on tumor cells. In a word, the strategy provided here offers a facile approach to develop a multifunctional nanoplatform for codelivery of DOX and ICG, which can synergistically improve the cancer-cell-killing efficiency, demonstrating great potential in chemo-photothermal therapy.

Cancer cell membrane-coated magnetic nanoparticles for MR/NIR fluorescence dual-modal imaging and photodynamic therapy

Jiong Li,Xuandong Wang,Dongye Zheng,Xinyi Lin,Zuwu Wei,Da Zhang,Zhuanfang Li,Yun Zhang,Ming Wu,Xiaolong Liu

Theranostic nanoprobes integrated with dual-modal imaging and therapeutic functions, such as photodynamic therapy (PDT), have exhibited significant potency in cancer treatments due to their high imaging accuracy and non-invasive advantages for cancer elimination. However, biocompatibility and highly efficient accumulation of these nanoprobes in tumor are still unsatisfactory for clinical application. In this study, a photosensitizer -loaded magnetic nanobead with surface further coated with a layer of cancer cell membrane (SSAP-Ce6@CCM) was designed to improve the biocompatibility and cellular uptake and ultimately achieve enhanced MR/NIR fluorescence imaging and PDT efficacy. Compared with similar nanobeads without CCM coating, SSAP-Ce6@CCM showed significantly enhanced cellular uptake, as evidenced by Prussian blue staining, confocal laser scanning microscopy (CLSM) and flow cytometric analysis. Consequently, SSAP-Ce6@CCM displayed a more distinct MR/NIR imaging ability and more obvious photo-cytotoxicity towards cancer cells under 670 nm laser irradiation. Furthermore, the enhanced PDT effect benefited from the surface coating of cancer cell membrane was demonstrated in SMMC-7721 tumor-bearing mice through tumor growth observation and tumor tissue pathological examination. Therefore, this CCM-disguised nanobead that integrated the abilities of MR/NIR fluorescence dual-modal imaging and photodynamic therapy might be a promising theranostic platform for tumor treatment.

Highly luminescent NIR-emitting CuFeS2/ZnS core/shell quantum dots for optical imaging of inflamed tissue

Tong Yan,Yunyan Li,Xiaoxiao Song,Jie Wang,Zhuoying Xie,Dawei Deng

The cytotoxicities of the elements of current quantum dots (QDs) greatly restrict their applications in biomedical fields. Herein, we firstly proposed and synthesized oil-soluble heavier-element-free (or Cd-free) CuFeS2/ZnS QDs, and focused on the synthetic controls over the core/shell structure. It was found that although the as-prepared CuFeS2 QDs are nonemissive, the in situ ZnS-overcoating could make them exhibit bright near-infrared (NIR) photoluminescence (PL) (PL peak at 840 nm; PL quantum yield (QY) up to 52%). We also analyzed their shape, structure and elemental composition in detail. Next, by the interaction between oil-soluble QDs and liposomes, the aqueous QD-laden liposomes (named as LipQDs) were fabricated, allowing further exploration of bio-application. Finally, after further decorating with isolated macrophage membrane, the resulting LipQDs@M nanocomposites were observed to target actively postoperative tumor inflamed tissue (or other inflamed tissues), using an in vivo small animal IR optical imaging system. Our results indicate that these NIR-emitting CuFeS2/ZnS QDs should be a new promising fluorescent marker for biomedical optical imaging, as an alternative for Cd-, Hg-, and Pb-based ones.

Highly Stable and NIR Luminescent Ru–LPMSN Hybrid Materials for Sensitive Detection of Cu2+ in Vivo

Fangman Chen,Fangnan Xiao,Weibing Zhang,Chentao Lin,Yunkun Wu

Herein, new near-infrared (NIR) luminescent ruthenium complexes were prepared for detecting Cu2+ ions. Then, ruthenium complex hybrid nanomaterials (Ru–LPMSNs) were fabricated successfully by imbedding the ruthenium complex into mesoporous silica nanoparticles. Benefiting from the novel large-pore mesoporous structure and good adsorbility of LPMSNs, Ru–LPMSN hybrid materials showed a significantly enhanced fluorescence intensity and stability. NIR fluorescence of Ru–LPMSNs was rapidly quenched by Cu2+ ions. Ru–LPMSNs also showed high Cu2+ ion selectivity and sensitivity as a sensor. The detection limit of Cu2+ ions was 10 nM with a wide linear relationship between the fluorescence intensity of Ru–LPMSNs and the concentration of Cu2+ ions. The mechanism of fluorescence quenching might be that the combination of the ruthenium complex and Cu2+ ions constrained the photoinduced electron-transfer process. Furthermore, Ru–LPMSNs dramatically increased the fluorescence signals in cells and achieved Cu2+-ion detection. Ru–LPMSNs had different tissue affinities and could monitor distribution of exogenous Cu2+ ions in vivo. Moreover, Ru–LPMSNs realized direct and rapid detection of Cu2+-ion content in serum. These results indicated the potential applications of the prepared nanomaterials as Cu2+ detection agents.

TiO2–x Based Nanoplatform for Bimodal Cancer Imaging and NIR-Triggered Chem/Photodynamic/Photothermal Combination Therapy

Wei Guo,Fei Wang,Dandan Ding,Chuanqi Song,Chongshen Guo,Shaoqin Liu

Integration of cancer diagnosis and treatment, namely theranostics, is an important issue in the biomedical field. Benefiting from an excellent photothermal effect, ROS generation ability, and the desired mesoporous structure of the TiO2–x matrix, we strategically designed and fabricated a TiO2–x based theranostic system for realizing fluorescence/photoacoustic tomography (PAT) bimodal imaging guided triple therapy for photothemal/photodynamic/chemotherapy in this work. Nonstoichiometric TiO2–x nanospheres are excellent near-infrared absorptive material, which takes on both photosensitizer and photothermal agent roles in implementing PDT/PTT combination therapy and PAT imaging. Moreover, the mesoporous structure of TiO2–x also allowed drug loading, and the polydopamine sealing layer enabled it to induce NIR/pH-triggered drug controlled release. Resultantly, both the in vitro and in vivo experiment manifested the remarkable tumor inhibition and tumor imaging effects by the TiO2–x based theranostic system. The antitumor mechanism was attributable to a synergistic therapeutic effect (combination index = 0.318) of DOX-induced DNA damage, and PDT/PTT caused mitochondrial dysfunction and a change in the cell membrane permeability. Innovatively, the B-mode ultrasonography was adopted to monitor the rehabilitation process at the solid tumor site after treatment, which observed a liquefaction necrosis process.

Biomimetic gold nano-modulator for deep-tumor NIR-II photothermal immunotherapy via gaseous microenvironment remodeling strategy

Honglin Huang,Zhengxin Xie,Ningxi Li,Li Zeng,Qianyi Zeng,Ziman Yang,Jinyang Shen,Hong Yang,Yiyao Liu,Chunhui Wu

Introduction Effective immunotherapeutic treatment of solid tumors has been greatly challenged by the complex hostile tumor immunosuppressive microenvironment (TIME), which typically involves hypoxia and immunosuppression. Methods Herein, a multifunctional biomimetic gold nano-modulator (denoted as GNR-SNO@MMT ) was developed to realize the efficient second near-infrared (NIR-II) photothermal immunotherapy via tumor targeting and deep penetration, vascular normalization and immune reprogramming. NIR-II photothermal agent gold nanorods (GNR) were grafted with thermosensitive S-nitrosothiol (SNO) donors and camouflaged with the tumor-penetrating peptide tLyp-1-modified macrophage membrane (MM) to yield GNR-SNO@MMT . Results The engineered membrane coating increased the capacity for tumor inflammatory tropism and deep penetration, which aided GNR-SNO@MMT in ablating tumors together with NIR-II laser irradiation. Moreover, hyperthermia-stimulated nitric oxide (NO) release in situ acted as a gas immunomodulator to effectively enhance blood perfusion and reprogram the TIME via multiple functions (e.g., decreasing PD-L1, repolarizing tumor-associated macrophages, and revitalizing cytotoxic T cells). Ultimately, the inhibition rate against 4T1 mouse mammary tumor model mediated by GNR-SNO@MMT plus NIR-II laser was 94.7% together with 2.4-fold CD8 + T cells infiltrated into tumors than that of the untreated counterpart. Conclusions The engineered biomimetic nano-modulator of GNR-SNO@MMT provides an effective and novel photoimmunotherapy candidate against deep-sited solid tumors through immune reconfiguration via NO-involved nanomedicine and external NIR-II laser assistance. Biomimetic NIR-II fluorescent proteins created from chemogenic protein-seeking dyes for multicolor deep-tissue bioimaging

Xu Jiajun,Zhu Ningning,Du Yijing,Han Tianyang,Zheng Xue,Li Jia,Zhu Shoujun

Near-infrared-I/II fluorescent proteins (NIR-I/II FPs) are crucial for in vivo imaging, yet the current NIR-I/II FPs face challenges including scarcity, the requirement for chromophore maturation, and limited emission wavelengths (typically < 800 nm). Here, we utilize synthetic protein-seeking NIR-II dyes as chromophores, which covalently bind to tag proteins (e.g., human serum albumin, HSA) through a site-specific nucleophilic substitution reaction, thereby creating proof-of-concept biomimetic NIR-II FPs. This chemogenic protein-seeking strategy can be accomplished under gentle physiological conditions without catalysis. Proteomics analysis identifies specific binding site (Cys 477 on DIII). NIR-II FPs significantly enhance chromophore brightness and photostability, while improving biocompatibility, allowing for high-performance NIR-II lymphography and angiography. This strategy is universal and applicable in creating a wide range of spectrally separated NIR-I/II FPs for real-time visualization of multiple biological events. Overall, this straightforward biomimetic approach holds the potential to transform fluorescent protein-based bioimaging and enables in-situ albumin targeting to create NIR-I/II FPs for deep-tissue imaging in live organisms.

Highly Efficient White-Light Emission and UV–Visible/NIR Luminescence Sensing of Lanthanide Metal–Organic Frameworks

Xinyu Wang,Pengfei Yan,Yuxin Li,Guanghui An,Xu Yao,Guangming Li

A series of new isostructural lanthanide metal–organic frameworks (Ln-MOFs), [Ln2(L)2(DMAC)2]·nH2O [Ln = La (1), Pr (2), Nd (3), Sm (4), Eu (5), Gd (6), Tb (7), and Tm (8); H4L = 5-(bis(4-carboxybenzyl)amino)-isophthalic acid, DMAC = N,N′-dimethylacetamide], have been isolated. Single-crystal X-ray diffraction analysis reveals that all complexes exhibit a rare (4,8)-connected alb-4,8-P topology with binuclear [Eu2(COO)8]n secondary building units as 8-connected nodes and H4L ligands as 4-connected nodes. Two mixed-lanthanide analogues of single-lanthanide MOFs were designed and prepared [Ln = La0.93Eu0.02Tb0.05 (9) and Tm0.47Eu0.18Tb0.35 (10)] by way of careful regulation of the relative concentration of lanthanide ions which are able to emit pure white light. Luminescent sensing of complexes 3 and 5 has been investigated. Strikingly, complex 5 exhibits an excellent luminescent sensing to TNP with a high Ksv value of 3.58 × 104 M–1 and a low detection limit of 4.66 × 10–4 mM. Complex 3 reveals high selectivity and sensitivity toward benzaldehyde (Ksv of 4.9 × 104 M–1; detection limit of 3.4 × 10–4 mM). It represents the first example of an NIR luminescent MOF for sensing of benzaldehyde.

NIR-triggered high-efficient photodynamic and chemo-cascade therapy using caspase-3 responsive functionalized upconversion nanoparticles

Na Zhao,Baoyan Wu,Xianglong Hu,Da Xing

Stimuli-responsive nanoparticles with multiple therapeutic/diagnostic functions are highly desirable for effective tumor treatment. Herein novel caspase-3 responsive functionalized upconversion nanoparticles (CFUNs) were fabricated with three-in-one functional integration: near-infrared (NIR) triggered photodynamic damage along with caspase-3 activation, subsequent caspase-3 responsive drug release, and cascade chemotherapeutic activation. CFUNs were formulated from the self-assembly of caspase-3 responsive doxorubicin (DOX) prodrug tethered with DEVD peptide (DEVD-DOX), upconversion nanoparticles (UCNP), a photosensitizer (pyropheophorbide-a methyl ester, MPPa), and tumor-targeting cRGD-PEG-DSPE to afford multifunctional CFUNs, MPPa/UCNP-DEVD-DOX/cRGD. Upon cellular uptake and NIR irradiation, the visible light emission of UCNP could excite MPPa to produce reactive oxygen species for photodynamic therapy (PDT) along with the activation of caspase-3, which further cleaved DEVD peptide to release DOX within tumor cells, thus accomplishing NIR-triggered PDT and cascade chemotherapy. CFUNs presented silent therapeutic potency and negligible cytotoxicity in the dark, whereas in vitro and in vivo experiments demonstrated the NIR-triggered cascade therapeutic activation and tumor inhibition due to consecutive PDT and chemotherapy. Current NIR-activated cascade tumor therapy with two distinct mechanisms is significantly favorable to overcome multidrug resistance and tumor heterogeneity for persistent tumor treatment.

Albumin-Mediated Biomineralization of Paramagnetic NIR Ag2S QDs for Tiny Tumor Bimodal Targeted Imaging in Vivo

Jing Zhang,Guangyu Hao,Chenfei Yao,Jiani Yu,Jun Wang,Weitao Yang,Chunhong Hu,Bingbo Zhang

Bimodal imaging has captured increasing interests due to its complementary characteristics of two kinds of imaging modalities. Among the various dual-modal imaging techniques, MR/fluorescence imaging has been widely studied owing to its high 3D resolution and sensitivity. There is, however, still a strong demand to construct biocompatible MR/fluorescence contrast agents with near-infrared (NIR) fluorescent emissions and high relaxivities. In this study, BSA-DTPAGd derived from bovine serum albumin (BSA) as a novel kind of biotemplate is employed for biomineralization of paramagnetic NIR Ag2S quantum dots (denoted as Ag2S@BSA-DTPAGd pQDs). This synthetic strategy is found to be bioinspired, environmentally benign, and straightforward. The obtained Ag2S@BSA-DTPAGd pQDs have fine sizes (ca. 6 nm) and good colloidal stability. They exhibit unabated NIR fluorescent emission (ca. 790 nm) as well as high longitudinal relaxivity (r1 = 12.6 mM–1 s–1) compared to that of commercial Magnevist (r1 = 3.13 mM–1 s–1). In vivo tumor-bearing MR and fluorescence imaging both demonstrate that Ag2S@BSA-DTPAGd pQDs have pronounced tiny tumor targeting capability. In vitro and in vivo toxicity study show Ag2S@BSA-DTPAGd pQDs are biocompatible. Also, biodistribution analysis indicates they can be cleared from body mainly via liver metabolism. This protein-mediated biomineralized Ag2S@BSA-DTPAGd pQDs presents great potential as a novel bimodal imaging contrast agent for tiny tumor diagnosis.
