Hierarchical AuNPs@BN@AgNR SERS sensor for ultrasensitive detection of bilirubin and biliverdin in saliva

Tianxin Zuo,Hengchang Zhang,Kaidi Sun,Hairong Chen,Caiqin Han

AuNPs@BN@AgNR SERS sensor for ultrasensitive salivary BR/BV detection. • Ultra-low LODs of 5 × 10 −8  M for BR and 1 × 10 −7  M for BV in spiked saliva. • FDTD simulations confirm 5.54-fold electric field enhancement via LSPR coupling. • BN layer enables target enrichment and suppressed non-specific adsorption. Bilirubin (BR) and biliverdin (BV) are non-invasive biomarkers for hepatic function assessment. Their detection in saliva is limited by sub-micromolar concentrations and matrix interference. Conventional methods exhibit limits of detection (LODs) ≥ 10 −6  M. Herein, an AuNPs@BN@AgNR SERS sensor is fabricated via oblique angle deposition, wet chemical etching, and chemical reduction. FDTD simulations demonstrate a 5.54-fold local electric field enhancement in the x–z plane relative to the BN@AgNR bilayer, resulting from LSPR coupling between AuNPs and AgNRs. Experimentally, the substrate achieves enhancement factors up to 4.06 × 10 9 with a spatial relative standard deviation (RSD) of ≤7.00%. The sensor yields LODs of 1 × 10 −12  M (BPE), 2.5 × 10 −8  M (R6G), and 1 × 10 −6  M (CV). In spiked saliva, the sensor exhibits quantifiable concentration ranges of 5 × 10 −8  M to 1 × 10 −4  M for BR and 1 × 10 −7  M to 1 × 10 −4  M for BV, with corresponding LODs of 5 × 10 −8  M and 1 × 10 −7  M. This performance is attributed to BN-mediated electrostatic target enrichment and the suppression of non-specific protein adsorption. This study provides an analytical platform for the non-invasive trace analysis of hepatic biomarkers. Download: Download high-res image (112KB) Download: Download full-size image

Dual-mode analysis of ischemic stroke based on urine SERS spectra and carotid B-ultrasound

Wenrou Yu,Zhaoxian Chen,Yao Liu,Yan Du,Hui Wang,Fengjie Tang,Xin Xie,Yingzhou Huang

Achieving noninvasive high-frequency monitoring of ischemic stroke (IS) remains a major clinical challenge for timely intervention and precise secondary prevention. Establishing precise correlations between patients’ systemic microscopic molecular fingerprints and localized macroscopic organ pathological events is essential to overcome the limitations of single-modal detection and enhance the efficacy of clinical risk assessment. However, because of the complexity of heterogeneous data, effectively integrating the cross-dimensional “molecular imaging” data remains a critical bottleneck in achieving this goal. Here, we present a proof-of-concept method to distinguish patients with confirmed IS from healthy controls (HCs) that used machine learning (ML)–based methods to surface-enhanced Raman spectroscopy (SERS) of urine (one-dimensional) and carotid artery B-ultrasound imaging (CBI) (two-dimensional). In an exploratory cohort of 101 participants, this approach analyzed 10,100 SERS spectra and 481 CBI images, achieving 92% classification accuracy and an area under the curve (AUC) of 0.95. Furthermore, by combined SERS spectra and liquid chromatography–mass spectrometry technology, this study preliminarily explored the urinary biomarker differences between HC/IS groups. The multidimensional data fusion strategy proposed in this study effectively bridges the information gap between traditional molecular detection and clinical phenotypes by systematically correlating microfluidic biomarkers with macro-organ imaging features. This approach provides a previously unexplored, noninvasive, and highly accurate tool for risk stratification and clinical decision-making in classifying HC/IS groups.

mSiO2-CQDs-Au@Ag nanocomposites for sensitive SERS detection of amisulpride: performance and mechanistic insights

Yun Qin,Li Zhang,Yu Ren,Yihao Wu,Yiping Wu,Ying Wen,Xinling Liu,Xiaoyu Guo,Haifeng Yang

Rational design of a multifunctional mSiO 2 -CQDs-Au@Ag nanocomposite. • Au@Ag and CQDs synergistically strengthen the SERS response. • The substrate achieves an enhancement factor of 1.0 × 10 9 . • Amisulpride in seawater is detected at 6.1 × 10 −9 mol·L −1 . • Charge transfer assisted enhancement is revealed experimentally. Pharmaceutically active compounds (PhACs) are receiving increasing attention because of their widespread use and continuous release into aquatic environments. Among them, amisulpride (AMI), a widely prescribed antipsychotic drug, is a plausible aquatic contaminant due to its release through wastewater and incomplete metabolic removal. Herein, Au@Ag core–shell nanoparticles were prepared using β-cyclodextrin as both the reducing agent and stabilizer, and further assembled with carbon quantum dots (CQDs) and mesoporous silica (mSiO 2 ) to construct a composite Surface-enhanced Raman scattering (SERS) substrate, mSiO 2 -CQDs-Au@Ag, for sensitive AMI detection. The substrate exhibited strong enhancement capability, with an enhancement factor (EF) as high as 1.0 × 10 9 . When applied to the detection of AMI in seawater, the proposed platform achieves a low limit of detection (LOD) of 6.1 × 10 −9 mol·L −1 , together with a good linearity (R 2  = 0.993). Combined theoretical and experimental analyses indicated that CQDs not only participate in interfacial charge transfer, but also contribute to analyte adsorption. This work offers a rational strategy for constructing semiconductor-integrated SERS platforms for sensitive detection and demonstrates its promising potential for environmental monitoring of emerging organic pollutants. Regulating Rare-Earth Single-Atom Nanozyme for Colorimetric-SERS Dual-Modal Sensing of Intracellular Glutathione

Huifang Zhao,Yi Chen,Lili Liu,Fujuan Peng,Bowei Guan,Jinze Li,Qian Wang,Luliang Liao,Baozhu Chi,Ruiping Zhang

Rare-earth single-atom nanozymes (RESAzymes) have emerged as promising catalysts due to their maximized atom utilization efficiency and tunable enzymatic activities. Herein, a colorimetric-Surface-enhanced Raman scattering (SERS) dual-modal sensing platform is constructed to introduce 4f electron configurations of single-atom ytterbium active site decorated on a nitrogen-doped porous carbon (Yb-N/C) by a facile hierarchical cascade-anchoring protocol. Specifically, this unique coordination environment can promote substrate molecule adsorption and activation energy barriers, possessing excellent peroxidase (POD)-mimicking activity and SERS enhancement performance in TMB-H 2 O 2 system. In the presence of glutathione (GSH), Yb-N/C catalyst is inhibited to result in reducing oxTMB for returning its colorless state, consequently diminishing both the colorimetric readout and the SERS intensity. Interestingly, this reaction mechanism from Yb-N/C is systematically illustrated through the steady-state kinetic, electron paramagnetic resonance (EPR), and density functional theory (DFT) calculations. Notably, the limited of detection (LOD) of GSH with colorimetric and SERS dual-mode was as low as 0.30 and 0.11 µM, respectively. Our established dual-modal analysis for intracellular GSH exhibits the favorable consistent with the standard kit in HepG2, KYSE150, and MCF-7 cells, respectively. This work will offer an in-depth understanding of RESAzymes-mimicking activity of colorimetric-SERS biosensors by adjusting electronic structures in carbon-based carrier.

SERS Chemical Enhancement Mechanism of Phenthoate on Gold Nanoparticles Revealed by DFT and TD-DFT

Qi Wang,Chao Song,Zhuang Bo,Haotong Yang,Xun Gao,Jingquan Lin

This study employed density functional theory (DFT) and time-dependent density functional theory (TD-DFT) to investigate the interaction mechanism between Phenthoate and Au20 nanoclusters, and the solvent environment effect was examined under the polarizable continuum model (PCM). By comparing and analyzing theoretical Raman active spectra and experimental Raman spectra, the phenomenon of selective enhancement of specific vibrational modes in surface-enhanced Raman spectroscopy (SERS) was explained. By analyzing the molecular structure, frontier molecular orbitals (FMOs), ultraviolet–visible absorption spectra (UV–vis), noncovalent interactions (NCI), reduced density gradients (RDG), electron localization function (ELF), and localized orbital locator (LOL), the adsorption mechanism and the spectral characteristics induced by interfacial charge transfer were comprehensively revealed at the electronic level. Phenthoate chemically adsorbed onto the Au20 surface through the S atom, forming a stable complex. Meanwhile, this charge transfer mechanism effectively modulated the polarization rate of the molecule. This study provides a theoretical basis for elucidating the adsorption behavior and spectral response mechanism of organophosphorus pesticides on metal nanostructures, offering valuable insights for the design of highly sensitive SERS sensors.

Flexible PDMS-based SERS substrates fabricated by replication and dual laser structuring of Ti templates

Junhao He,Duoduo Yang,Kaishen Chen,Yiping Cheng,Xuanling Luo,Xingyu Chen,Chengyun Zhang

Dual fs laser structuring creates hierarchical PDMS SERS substrates. • Secondary laser processing increases nanoscale roughness and hotspots. • Detection limitation down to 10 −10 M with high linearity. • Excellent uniformity (RSD 2.57%) and reusability (<4% decay). • Flexible substrate enables adhesive sampling on curved surfaces. Flexible and sensitive surface-enhanced Raman scattering (SERS) substrates are essential for real-world applications such as wearable sensors and on-site molecular detection. We developed two polydimethylsiloxane (PDMS)-based flexible SERS substrates via the replication of femtosecond (fs) laser-nanostructured titanium (Ti) templates: a single-processed Ti-Fs-PDMS substrate and a dual-processed Dual-Fs-PDMS substrate, the latter incorporating an additional fs laser treatment to enrich nanoscale surface features. Gold was thermally deposited onto the structured PDMS to introduce plasmonic enhancement. The dual-structured substrate exhibits higher roughness and hydrophobicity (water contact angle >130°). SERS performance was evaluated using Rhodamine 6G (R6G) and malachite green (MG), revealing high sensitivity with a detection limit as low as 10 −10 M for Dual-Fs-PDMS and excellent signal linearity (R 2  > 0.99). Furthermore, the Dual-Fs-PDMS substrate exhibits superior signal uniformity (RSD = 3.203%), reusability, mechanical robustness, and good long-term stability over 180 days, as well as the capability for in situ detection on curved surfaces. Three-dimensional electromagnetic field simulations based on AFM-acquired topography indicated that the Dual-Fs-PDMS substrate exhibits a stronger local electric field and a higher density of “hot spots”, correlating with its enhanced SERS performance. These results highlight the potential of Dual-Fs-PDMS as a flexible and high-performance SERS platform for practical applications. Interlaced 2D cellulose networks with molecular enrichment capability for sensitive SERS detection of sweat biomarkers

Lin Luyao,Wang Haonan,Lei Xinlin,Zhuang Xin,Chen Yiqing,Lai Qiyue,Li Bo,Fang Yulin,Xu Yunli,You Ruiyun,Lu Yudong

Efficient localization of analytes near plasmonic hotspots remains a major challenge for surface-enhanced Raman scattering (SERS) detection in complex liquid-phase systems. Herein, an interlaced cellulose-based porous membrane composed of dissolving pulp fibers (DPFs), mechanically ground nanofibers (MGNFs), sodium alginate (SA), and Ag nanoparticles was developed as a molecular enrichment-assisted SERS platform for sweat biomarker determination. The hierarchical porous architecture provided interconnected transport channels and abundant interfacial adsorption sites, facilitating analyte retention and localized enrichment within the plasmonic region. Benefiting from the synergistic effects of porous confinement and uniformly distributed Ag nanoparticles, the optimized substrate exhibited sensitive and reproducible SERS performance with an enhancement factor of 4.36 × 10⁷, a relative standard deviation of 8.04%, and a detection limit down to 1.0 × 10⁻⁸ mol/L for Rhodamine 6G (R6G). Systematic adsorption experiments using molecules with different charge properties further demonstrated the broad molecular enrichment capability of the structured cellulose network. The developed platform enabled quantitative detection of lactate and urea within physiologically relevant sweat concentration ranges and showed satisfactory analytical performance in spiked sweat samples. More importantly, this work demonstrates a structure-engineered porous membrane strategy for integrating molecular enrichment with plasmonic sensing, providing new insight into cellulose-based SERS platforms for complex bioanalytical applications. Superhydrophobicity-driven interfacial enrichment and SERS detection of PFOA on nano-Cu modified Ag foam

Xiaoya Yuan,Lei Ouyang,Zhou Song,Xuelin Dong,Zhenli Zhu,Qin Shuai,Shenghong Hu

Perfluoroalkyl and polyfluoroalkyl substances (PFAS) present a serious risk to ecosystems and human health, underscoring the need for highly sensitive detection methods. Existing analytical approaches often lack the portability and rapid response required for on-site screening, and conventional surface-enhanced Raman spectroscopy (SERS) substrates exhibit limited affinity toward hydrophobic PFAS molecules. To overcome these limitations, we designed a superhydrophobic nano-Cu modified Ag foam that integrates hydrophobic enrichment with specific metal-carboxylate coordination for effective perfluorooctanoic acid (PFOA) capture and detection. The critical role of Cu 2+ as a bridging site for selective PFOA adsorption was elucidated. This design achieves a remarkably low detection limit of 0.1 μg L −1 for PFOA and enables discrimination between different PFAS categories. Moreover, it maintains excellent SERS activity after two months of storage, effectively addressing the issue of facile oxidation of Cu. The substrate was successfully applied to detect PFOA in various real water samples, demonstrating its potential as a practical and powerful platform for environmental PFAS monitoring.

3D RGO@MoS2/Ag NPs based SERS platform for label-free detection of IL-6

Qinqin Qian,Haocheng He,Songyin Fu,Jia Xu,Ho-Pui Ho,Shuwen Zeng,Tao Jiang,Sheqin Lv,Chenjie Gu

The efficient and sensitive monitoring of the interleukin-6 (IL-6) is crucial for the clinical tracking and diagnosis of chronic obstructive pulmonary disease (COPD). Herein, a novel surface-enhanced Raman scattering substrate (RGO@MoS 2 /Ag NPs), comprising an efficient charge-transfer and adsorption layer of reduced graphene oxide (GO) encapsulated in molybdenum disulfide (MoS 2 ) and an electromagnetic enhancement layer of Ag nanoparticles, was developed for the highly sensitive, label-free detection of IL-6. Experimentally, a precursor film consisting of (NH 4 ) 2 MoS 4 and GO was first deposited onto a rough g -C 3 N 4 substrate via spin-coating process. Subsequently, the above precursor film was subjected to a two-step high-temperature annealing process for preparing the RGO@MoS 2 charge transfer layer. Finally, Ag NPs were sputtered onto the RGO@MoS 2 film to induce the localized surface plasmons for the further enhancement of the SERS performance. Leveraging the high specific surface area and exceptional charge transfer properties of RGO@MoS 2 , together with the high-density “hot spots” generated by surface Ag NPs, the fabricated SERS substrate exhibited remarkable electromagnetic and chemical enhancement effects. By using the rhodamine 6G (R6G) as the Raman reporter, it was determined that the lowest detection limit could be as low as 10 −9  M on the optimal SERS substrate, with an enhancement factor of 2.2 × 10 9 . Meanwhile, the substrate also exhibited good uniformity, with a relative standard deviation of 9.84%. Finally, the prepared SERS substrate was used to detect the IL-6, it showed good linear response in the range of 5.0-1000 pg/mL, with a detection limit as low as 0.41 pg/mL.

Hybrid Ag/Au nanocavity arrays for ultrasensitive SERS discrimination of molecular isomers via machine learning

Zhiming Chen,Jiabin Chen,Haoming Bao,Kaitao Liu,Pan Zeng,Fan Yang,Wei Cao,Dongqi Yang,An Cao,Tao Zhang,Yue Li

Surface-enhanced Raman scattering (SERS) has been one of the efficient strategies for trace molecular analysis, but the traditional substrates with sharp tips cannot effectively detecting the molecular isomers. Here, we develop a facile way to fabricate large-area, reproducible Ag/Au nanocavity arrays by the combination of microsphere-templated soft lithography and self-assembly. Compared to the flat and convex substrate, the Ag/Au units arranged on the concave surface exhibit a high-density of hotspots via spatial light confinement and multi-reflection-enhanced electromagnetic coupling, which enables highly sensitive detection of probe molecules down to10 −12 M. Moreover, by integrating machine learning algorithms, the test set shows a coefficient of determination (R 2 ) of 0.9896, achieving advanced training performance for classifying four structurally analogous quinolone antibiotics (ciprofloxacin, enrofloxacin, danofloxacin, and enoxacin) with a quantitative prediction of 10 −10  M. This work demonstrates a robust pathway for the controllable fabrication of high-performance SERS substrates for the development of intelligent, quantitative, and on-site sensing platforms in complex practical scenarios. Sensitive SERS detection of antibiotics using Ag-coated nanostructures fabricated by femtosecond laser cross-scanning

Jianjun Cao,Haoyue Yang,Taohua Zhou,Huaxin Zhu,Chaoqun Ma,Yuanlin Zheng,Wenjie Wan,Quanzhong Zhao

The excessive use of antibiotics in medicine, agriculture, and aquaculture has led to widespread environmental contamination and public health concerns, underscoring the urgent need for rapid, sensitive, and cost-effective detection methods. Here, we report a scalable surface-enhanced Raman scattering (SERS) platform based on silver-coated cross-scanned laser-induced periodic surface structures (CS-LIPSS) fabricated via femtosecond laser processing. By employing cylindrical lens-assisted beam shaping followed by orthogonal double-pass scanning, we generated periodic nanostructures enriched with dense nanoparticles. Subsequent Ag deposition produce a dense distribution of plasmonic hotspots, enabling strong electromagnetic enhancement. Compared with conventional mesh and single-scanned LIPSS substrates, the CS-LIPSS demonstrate superior SERS activity, with an enhancement factor of 6 . 4 × 1 0 7 and a detection limit down to 1 0 − 1 1 M for crystal violet. More importantly, the platform enable reliable identification of representative antibiotics including amoxicillin, tetracycline, and neomycin sulfate at concentrations as low as 1 ppm, highlighting its sensitivity and molecular specificity. The fabrication process is chemical-free, template-free, and compatible with wafer-scale production, offering a cost-effective route to reproducible, high-performance SERS substrates

Ag/UiO-66-Decorated 3D-Printed Slippery Liquid-Infused Porous Surfaces for Ultrasensitive SERS Detection

Yao Zhang,Hui Zhang,Ziyao Jia,Chunya Li,Lianqing Chen,Shiwei Jin

Surface-enhanced Raman scattering (SERS) holds significant potential for rapid food safety screening; however, its application is often constrained by substrate sensitivity, stability, and cost. Here, we developed a multifunctional SERS substrate by integrating a 3D-printed microstructured template with a slippery liquid-infused porous surface (SLIPS) and Ag/UiO-66 metal–organic framework (MOF) nanoparticles for food safety monitoring. The nearly pinning-free SLIPS interface enabled evaporation-driven enrichment of nanoparticles at microcone tips, generating dense, spatially confined hotspots with an enhancement factor (EF) of 1.44 × 108. The Ag/UiO-66@SLIPS substrate demonstrated ultrasensitive SERS detection of illicit sildenafil (SD) and food-dye additives (using rhodamine 6G as a model). The substrate achieved limits of detection (LOD) for SD of 100 pg/mL (2.11 × 10–10 M) in ethanol and 1 ng/mL (2.11 × 10–9 M) in health wine using only 10 μL samples. For rhodamine 6G (R6G), an ultralow LOD of 1 fg/mL (2.09 × 10–15 M) was obtained. Furthermore, for photocatalytically degradable dyes, the inherent catalytic activity of Ag/UiO-66 provided a self-cleaning pathway to mitigate signal carryover and enabled repeated measurements. The substrate maintained 83.5% signal intensity over 150 days and can be refreshed for continued use. This work combines biomimetic structures with functionalized nanomaterials, enabling the simple, cost-effective development of robust, uniform SERS substrates for multiscenario trace molecular sensing in food safety.
