Multifunctional active packaging of poly(butylene adipate-co-terephthalate)/cellulose acetate electrospun film with curcumin and tannic acid

Qian Han,Yuqing Zhou,Fangfang Liu,Yang Guo,Xing Zhang,Jinda Peng,Chenzhuo Wei,Nanfeng Zhu,Juqing Cui

Functional nanofiber films were successfully fabricated through an electrospinning process by incorporating curcumin and tannic acid as active components. • The resulting PBAT/CA@Cur/TA films exhibited remarkable and sustained antioxidant and antibacterial performance. • These PBAT/CA films effectively protected strawberries from postharvest quality deterioration, thereby significantly extending their shelf life. Poly(butylene adipate- co -terephthalate) (PBAT) is a biodegradable polyester that has attracted considerable attention in food packaging applications due to its favorable mechanical performance and processability. However, PBAT films are not ideal for the loading and controlled release of active compounds, and their relatively high cost further limits their practical application. This study reports the fabrication of PBAT/cellulose acetate (CA) films by electrospinning, with the incorporation of curcumin (Cur) and tannic acid (TA) as natural bioactive agents to confer antimicrobial and antioxidant functionalities. The incorporation of Cur improved the mechanical performance of PBAT/CA films and imparted approximately 90% DPPH radical scavenging activity, while TA contributed antibacterial properties. Strawberry preservation experiments demonstrated that PBAT/CA@Cur/TA films, particularly those containing 12% Cur and TA, effectively prolonged fruit shelf life. At 4% and 8%, the scavenging efficiency reached approximately 91%. This preservation effect is attributed to the antioxidant action of Cur and the antimicrobial properties of TA. Based on the obtained results, the PBAT/CA films can be considered potential candidates for multifunctional active packaging. Chicken head gelatin/curcumin composite active packaging film: effects on quality and shelf life of refrigerated chicken breast

Mingyue Wang,Yana Liu,Shaolin Deng,Xinyi Li,Siming Lu,Xia Wang,Yun Bai,Xinglian Xu,Minyi Han

Chicken head gelatin enabled high-value utilization of poultry by-products. • Curcumin improved multiple properties of gelatin-based composite films. • Curcumin exerted antibacterial activity via release and contact dual mechanisms. • The optimized film inhibited quality deterioration of refrigerated chicken breast. • Selected films prolonged shelf life from 4 days to 6 and 8 days versus the control. To address the spoilage susceptibility of poultry products during refrigerated storage, as well as the environmental limitations of conventional non-degradable packaging films and the single functionality of existing gelatin films with insufficient antioxidant and antibacterial activities, this study utilized chicken heads, a by-product of poultry processing, for gelatin extraction. Curcumin was incorporated as a natural active ingredient to develop an active packaging film for extending the shelf life of refrigerated chicken breast. The effects of different curcumin loadings (0, 5, 10, 15, and 20 g/kg) on the packaging and functional properties of gelatin-based composite films were investigated to determine the optimal concentration. The results indicated that films with 20 g/kg curcumin loading exhibited optimal performance, with significantly enhanced antioxidant (DPPH and ABTS radical scavenging rates were 87.95% and 95.21%, respectively) and antibacterial activities (bactericidal rates against Escherichia coli and Staphylococcus aureus were 74.43% and 75.35%, respectively). Under storage at 4 °C, films at this concentration effectively inhibited microbial growth, delayed protein decomposition and lipid oxidation, and extended the shelf life from 4 to 8 d. This study achieves high-value utilization of poultry processing by-products and provides a novel, green and efficient packaging solution for the refrigerated preservation of chicken breast.

Development of a Curcumin-Loaded Nanomicelles-Injectable Sustained-Release Hydrogel System for Modulating Oxidative Stress to Alleviate Tendinopathy

Shuang Wang,Keyi Wu,Meiqi Sun,Xinrui Wang,Jingying Li,Guorong Zhang,Zhidong Qiu

Tendinopathy is a common musculoskeletal disorder that increases the risk of tendon rupture if not properly treated. Current local injection therapies require frequent administration, and no fully effective drug is yet available. Curcumin (Cur) exhibits excellent anti-inflammatory and antioxidant effects, but its poor water solubility and low stability limit its clinical application. To overcome these challenges, this study encapsulated Cur into pluronic F127-based nanomicelles (Cur-F127) to improve its aqueous solubility and stability. Subsequently, the micelles were incorporated into a hydrogel network (Cur-F127&gel) formed by oxidized hyaluronic acid (oxi-HA) and adipic acid dihydrazide (ADH) to achieve sustained release. The resulting Cur-F127 micelles had a particle size of 20.14 ± 0.287 nm, an encapsulation efficiency (EE%) of 89.95 ± 0.60%, and a drug loading (DL%) of 5.57 ± 0.05%. The composite hydrogel possessed a loose, porous three-dimensional network, excellent biocompatibility, and favorable degradation behavior. The system enabled sustained release of Cur for over 20 days without an initial burst. In a rat model of tendinopathy, Cur-F127&gel significantly promoted tendon repair, as evidenced by reduced inflammatory cell infiltration, improved collagen fiber alignment, restored expression of key mitochondrial-related proteins (Ndufs3, Uqcrq, Uqcr10, Atp5mc3), and alleviated oxidative stress damage demonstrated by increased SOD activity and decreased MDA content in tendon tissue, thereby suppressing disease progression. This injectable sustained-release hydrogel system for poorly soluble drugs provides an effective approach for the local, long-acting delivery of Cur and long-term repair of tendinopathy, highlighting its potential value for clinical application.

Anthocyanin/curcumin intelligent films with high pH sensitivity for freshness maintenance and real-time monitoring of dried tofu

Ning Zhang,Lina Yang,Fuming Zhang,Huaying Pan,He Liu

Intelligent packaging materials offer innovative solutions for the freshness monitoring and quality maintenance of high-protein foods during their preservation and distribution. However, most existing intelligent packaging films rely on single natural pigments or synthetic indicators, which suffer from narrow color-changing ranges or safety controversies, while their biodegradability is largely overlooked. To this end, the present study prepared composite films comprising soy hull nanocellulose, polyvinyl alcohol, and pullulan, into which purple cabbage anthocyanin (PCA), curcumin (CU), and their blends at different ratios (PCA:CU = 8:2, 5:5, and 2:8 v/v) were incorporated. Results indicated that PCA and CU formed a hydrogen bond network with the film matrix via phenolic hydroxyl groups, significantly enhancing the mechanical properties of the composite films (P < 0.05). The incorporation of PCA and CU also improved thermal stability and hydrophobicity while reducing water vapor permeability. Among the formulations, PCA/CU-3 (8:2 v/v) exhibited optimal overall performance, showing the most distinct color response across pH 4–12, an antibacterial rate of 93.87% against Pseudomonas fragi , and effective delay of microbial growth and total volatile basic nitrogen (TVB-N) accumulation in dried tofu, extending its shelf life to 12 days. The film color gradually changed from deep red to deep purple during spoilage, enabling visual monitoring. Additionally, PCA/CU-3 degraded noticeably within 25 days under soil-burial conditions. Overall, this study demonstrates a novel degradable food-packaging material integrating antibacterial preservation and real-time spoilage monitoring functions.

Highly stable Pickering emulsions stabilized by gelatin–pectin complexes: Interfacial architecture, rheological behavior, and curcumin loading and protection

Biao Liang,Xinying Guo,Yuanyuan Wang,Shumin Yi,Jianrong Li,Beibei Ye,Yanwei Liu,Jinxiang Wang,Cikun Liu,Xuepeng Li

At 9000 s, G i ′ increased by approximately six orders of magnitude relative to C1. • The 0.5% pectin emulsion showed a dense interfacial layer and a 3D network. • At 0.5% pectin, the emulsified phase volume fraction remained at 100% over 180 d. • C1–P0.5 resisted freeze–thaw, thermal, pH, and salt stresses. • Curcumin loading efficiency peaked at 0.3% pectin, while retention peaked at 0.5%. Designing highly stable Pickering emulsions with common biopolymers and expanding their applications to the effective loading of lipophilic bioactives remain formidable challenges. In this study, we successfully fabricated a series of Pickering emulsions stabilized by gelatin–pectin complexes containing gelatin (1.0%, w/w) and pectin (0.1%, 0.3%, and 0.5%, w/w). Pectin incorporation significantly reduced the mean droplet size and increased the zeta-potential magnitude, with C1–P0.5 reaching < 6 μm and > 35 mV, respectively. Interfacial and bulk rheological measurements, together with cryo-SEM and LF-NMR, indicated that pectin incorporation strengthened both the interfacial layer and the continuous-phase structure, forming a multiscale reinforcing architecture. This architecture comprised a dense, viscoelastic interfacial layer and an interconnected 3D honeycomb-like scaffold, which collectively provided robust electrosteric stabilization and geometric confinement. Consequently, the high-pectin group (C1–P0.5) achieved high stability across a broad range of environmental stressors, including 180-day room temperature storage, freeze–thaw cycles, thermal treatment (60–100 °C), and wide fluctuations in pH and ionic strength (NaCl/MgCl 2 ). Furthermore, the C1–P0.3 formulation achieved a curcumin loading efficiency of > 95%, and pectin incorporation enhanced curcumin retention under UV exposure and thermal treatment. These findings provide a mechanistic basis for using simple biocomposites to develop highly stable Pickering systems for the loading and protection of sensitive lipophilic bioactives.

Engineering a Multifunctional Sprayable Curcumin-Modified Polyitaconate-Based Hydrogel to Accelerate Methicillin-Resistant Staphylococcus aureus-Infected Wound Healing

Min Wang,Guoqiang Wang,Sihua Li,Jiachen Wang,Jing Tian,Yanzi Zhao,Hao Yang

Hydrogel dressings that can simultaneously curb infection and dampen inflammation are particularly attractive for accelerating multidrug-resistant bacteria-infected wound healing. However, clinical translation of many reported hydrogels is often constrained by multi-component formulations, complicated preparation procedures, and insufficient sprayability, which limit conformal coverage on irregular wound beds. Polyitaconate possesses intrinsic antibacterial and anti-inflammatory activities, yet its weak antioxidative properties and lack of thermogelling properties limit its utility in infected wound healing. In this study, we develop a curcumin-modified polyitaconate-based hydrogel (FPIC) that integrates antibacterial, anti-inflammatory, and antioxidant properties. FPIC hydrogel exhibits thermoresponsive gelation, injectability, sprayability, and shear-thinning properties, allowing convenient close contact with complex wound geometries. FPIC hydrogel inhibits bacterial growth, moderates excessive inflammatory responses, and eliminates reactive oxygen species while promoting angiogenesis, thereby facilitating the shift from the inflammatory phase to the proliferative phase of healing. Moreover, FPIC hydrogel enhances collagen deposition, speeds re-epithelialization, and supports balanced tissue remodeling, enabling improved skin regeneration with a lower propensity for fibrosis. These findings establish FPIC hydrogel as a multifunctional and highly bioactive polyitaconate-based hydrogel dressing with strong promise for treating infection- and inflammation-associated wounds and advancing toward clinical use.

Rational design of curcumin-borneol hybrids for rheumatoid arthritis pathological intervention via AKT/mTOR/HIF-1α pathway modulation

Yue Peng,Cuiman Tang,Mingcong Niu,Tianyi Xu,QingYu Meng,Xinran Li,Rongheng Ma,Huimin Yu

A traditional medicine-inspired strategy provides a logical framework for constructing carrier-free hybrids. • Precise structural modifications successfully circumvent the inherent bioaccessibility limitations of natural products. • The covalent integration of diverse functional modules enables synergistic therapeutic efficacy against arthritis. • The therapeutic action is closely associated with the modulation of protein kinase B and hypoxia-inducible factor signaling. Curcumin (Cur) possesses potent anti-inflammatory activity but faces clinical challenges due to poor aqueous dispersibility, insufficient membrane affinity, and low bioavailability. Borneol (Bor), a classic “envoy” component in Traditional Chinese Medicine (TCM), exhibits prominent membrane-penetrating properties. Inspired by the “monarch-envoy” compatibility theory, we rationally designed carrier-free hybrids by covalently conjugating Cur (“monarch”) with Bor (“envoy”). Systematic characterization confirmed the successful synthesis and controllable optimization of key properties, including enhanced dispersibility, improved cellular uptake, and favorable biocompatibility. Using rheumatoid arthritis (RA) as a disease model, we identified SCBMS as the optimal formulation owing to its superior dispersibility and uptake, which directly translated into the most potent anti-RA efficacy. The hybrids exerted synergistic multifunctional effects—including robust anti-inflammatory and antioxidant activities—effectively reducing the arthritis index and ameliorating synovial hyperplasia and joint destruction in vivo . Mechanistically, our results indicate a close correlation between the therapeutic effects and the downregulation of the AKT/mTOR/HIF-1α signaling pathway, supporting the involvement of this pathway in the observed anti-RA activity. This study provides a generalizable framework for developing functional hybrids guided by TCM principles and validates their translational potential. Electrospun curcumin-loaded polylactic acid/microcrystalline cellulose nanofiber membranes with high mechanical and antibacterial properties for wound dressings

Rui Tie,Xueqing Chai,Xingzhuo Zhang,Zhaocan Li,Zhiyan Wang,Shuyi Zhou,Simin Wu,Yao Zhang,Yujuan Jin,Huafeng Tian,Yan Jia

The synergistic optimization of antibacterial activity and mechanical properties has emerged as a priority in the development of medical dressings for chronic wounds. In this study, curcumin (Cur)-loaded polylactic acid/microcrystalline cellulose (PLA/MCC) nanofiber membranes were fabricated via electrospinning technology. It was found that the incorporation of 3% Cur significantly enhanced the mechanical properties of the electrospun nanofiber membrane compared with neat PLA. Its tensile strength and elongation at break increased by 240.2% and 181.2%, respectively, which meets the mechanical property requirements for clinical applications (1–40 MPa). The antibacterial activity is a core indicator that directly determines the practical application value of wound dressings. 3% Cur-loaded PLA/MCC nanofiber membranes show excellent antibacterial and antioxidant properties. Compared with PLA/MCC, the inhibition rate was increased by 20.1% against Escherichia coli and 72.2% against Staphylococcus aureus , with antioxidant activity exceeding 68.0%. In addition, cytotoxicity and hemocompatibility tests verified that all nanofiber membranes complied with the safety standards of biomedical materials. In conclusion, the PLA/MCC/Cur nanofiber membranes hold great promise as a novel, efficient, and safe dressing candidate for the clinical management of chronic wounds. Mechanically robust and highly hydrated curcumin-loaded lysine-crosslinked carboxymethyl cellulose hydrogels for steroid-free atopic dermatitis treatment

Zhang Xinyan,Zhou Jingqiu,Han Xue,Xing Xu,Yang Kun,Wu Tao,Li Hao,Hao Xujia,Du Qiang,Liu Xinwei,Gong Tianxing,Leng Hongrui,Wei Shibo

Carboxymethyl cellulose (CMC) exhibits excellent biocompatibility and water retention, yet its high hydrophilicity often leads to excessive swelling and poor mechanical stability in physiological environments, limiting its utility in topical skin therapies. In this study, we developed a robust, chemically crosslinked CMC-based hydrogel loaded with curcumin (Cur@CL) using natural lysine as a non-toxic crosslinker. This green crosslinking strategy significantly reinforced the hydrogel network, achieving a swelling ratio of 3074 ± 226% and a compressive stress of 20.06 ± 1.0 kPa. The Cur@CL hydrogel demonstrated a sustained curcumin release profile of 83.98 ± 5.68% over 144 h, ensuring prolonged local bioavailability. In vitro, Cur@CL exhibited excellent cytocompatibility with L929 fibroblasts, accelerated fibroblast-associated wound closure, and reduced intracellular reactive oxygen species (ROS) levels by 62.97%. Furthermore, in a 2,4-dinitrochlorobenzene (DNCB)-induced atopic dermatitis (AD) mouse model, topical application of Cur@CL significantly alleviated clinical symptoms, reducing epidermal thickening by 71.5% compared with the DNCB-challenged model group, with no detectable systemic toxicity. This study presents a simple, tunable, and eco-friendly cellulose-based hydrogel platform for the effective, non-steroidal management of AD.

Antibacterial and preservation efficiency of photodynamic ice glaze driven by curcumin/β-cyclodextrin complex against Pacific white shrimp

Xiaowen Huang,Lu Chen,Huajian Ou,Yanyan Huang,Zhiyun Peng,Langhong Wang,Konglong Feng,Qiaohui Zeng,Jing Jing Wang

A novel photodynamic ice glaze with high antibacterial ability based on curcumin (Cur)/β-cyclodextrin (β-CD) inclusion complex was developed, and its active cold-chain preservation efficiency for Pacific white shrimp was investigated. Results showed that β-CD/Cur inclusion complex significantly enhanced the aqueous solubility and photostability of curcumin by facilitating the formation of a novel crystalline phase, which enabled efficient reactive oxygen species (ROS) generation in its frozen aqueous state. Under blue LED irradiation during frozen storage, the photodynamic ice exhibited broad-spectrum antibacterial ability against foodborne pathogens, notably with a reduction of >5.98 Log CFU/mL for Gram-positive Staphylococcus aureus and an undetectable level for Gram-negative Vibrio parahaemolyticus . Mechanistic studies confirmed that the generated ROS disrupted cellular structural integrity, and inflicted severe oxidative damage to bacterial DNA and proteins. During 15 days of frozen storage of shrimp, the photodynamic ice glaze slowed microbial proliferation (<5.20 Log CFU/g), inactivated endogenous proteases, retarded water migration, and maintained muscle quality. Therefore, the photodynamic ice glaze is a green and feasible strategy for improving the microbial safety and quality of aquatic products.

Encapsulation of Curcumin using Curdlan-whey Protein Isolate Composite Gels: Gel Properties, Stability and in vitro Release Performance

Hu Wenmei,Qin Xianglin,Huang Kai,Ren Erfang,Miao Yifan,Liao Siru,Zhang Siqi,Chen Shan

This research primarily investigates the encapsulation of curcumin within curdlan-whey protein isolate(CUR-WPI) composite gels and the subsequent release of the encapsulated compound. Results demonstrated that the composite gel could effectively encapsulate curcumin, with the highest encapsulation rate reaching 83.6%. The main force for the gel to encapsulate curcumin was electrostatic interaction and hydrogen bond. Fluorescence spectroscopy showed that curcumin can bind to WPI through hydrophobic interactions and suppress WPI's natural fluorescence. The composite gel exhibits excellent water-holding capacity (WHC) and the favorable texture profile analysis (TPA), which improved the photostability (2.57 times) and storage stability of curcumin (6.09 times). Moreover, the release rate of curcumin from the composite gel in intestinal fluid was higher than that in the stomach, which was related to the non-digestible characteristics of the curdlan and the dense network structure of the gel. The release kinetics results indicated that the main release mechanism of curcumin in the composite gel during the simulated digestion process was the diffusion mechanism. In conclusion, CUR-WPI composite gel is a good delivery carrier for curcumin, indicating its potential value for application in functional dairy products. Graphical

Design and Characterization of Curcumin-Modified Polyurethane Material with Good Mechanical, Shape-Memory, pH-Responsive, and Biocompatible Properties

Man Wang,Hongying Liu,Wei Zhao,Huafen Wang,Yuwei Zhuang,Jie Yang,Zhaohui Liu,Jing Zhu,Sichong Chen,Jinghui Cheng

In the context of critical challenges in curcumin-modified polyurethane synthesis—including limited curcumin bioavailability and suboptimal biodegradability/biocompatibility—a novel polyurethane material (Cur-PU) with good mechanical, shape memory, pH-responsive, and biocompatibility was synthesized via a one-pot, two-step synthetic protocol in which HO-PCL-OH served as the soft segment and curcumin was employed as the chain extender. The experimental results demonstrate that with the increase in Cur units, the crystallinity of the Cur-PU material decreases from 32.6% to 5.3% and that the intensities of the diffraction peaks at 2θ = 21.36°, 21.97°, and 23.72° in the XRD pattern gradually diminish. Concomitantly, tensile strength decreased from 35.5 MPa to 19.3 MPa, and Shore A hardness declined from 88 HA to 65 HA. These observations indicate that the sterically hindered benzene ring structure of Cur imposes restrictions on HO-PCL-OH crystallization, leading to lower crystallinity and retarded crystallization kinetics in Cur-PU. As a consequence, the material’s tensile strength and hardness are diminished. Except for the Cur-PU-3 sample, all other variants exhibited exceptional shape-memory functionality, with Rfand Rrexceeding 95%, as determined by three-point bending method. Analogous to pure curcumin solutions, Cur-PU solutions demonstrated pH-responsive chromatic transitions: upon addition of hydroxide ion (OH−) solutions at increasing concentrations, the solutions shifted from yellow-green to dark green and finally to orange-yellow, enabling sensitive pH detection across alkaline gradients. Hydrolytic degradation studies conducted over 15 weeks in air, UPW, and pH 6.0/8.0 phosphate buffer solutions revealed mass loss <2% for Cur-PU films. Surface morphological analysis showed progressive etching with the formation of micro-to-nano-scale pores, indicative of a surface-erosion degradation mechanism consistent with pure PCL. Biocompatibility assessments via L929 mouse fibroblast co-culture experiments demonstrated ≥90% cell viability after 72 h, while relative red blood cell hemolysis rates remained below 5%. Collectively, these findings establish Cur-PU as a biocompatible material with tunable mechanical properties, and pH responsiveness, underscoring its translational potential for biomedical applications such as drug delivery systems and tissue engineering scaffolds.
