Microbioreactors with multioptical sensors have become increasingly important because of their small working volumes, high degree of parallelization and the available robotics. A novel hydrophobic luminescent copolymer P(Pt-TPP-TFEMA) along with reference P(Pt-TPP-EMA) containing the pendant group of 5,10,15,20-tetraphenylporphyrin (TPP) moiety as low-cost dissolved oxygen (DO) chemosensor film for high-throughput microbioreactors is designed. Its sensor film exhibits fast response to DO with good stability and fatigue resistance, being capable of applying as a low-cost DO indicator for high-throughput bioprocess measurement. Results show that the quenching response of DO increases with the enhancement in the copolymeric hydrophobicity using the presented hybrid fluorinated ethyl methacrylate. Furthermore, the long emission band at 650 nm of chromophore TPP with large Stoke's shift about 250 nm brings several advantages, such as low scattering, deep penetration, and minimal interferences of absorption and fluorescence from the fermentation system, which shows high-promising application in bioprocess monitoring. © 2013 American Institute of Chemical Engineers AIChE J , 59: 2743–2752, 2013

Magnetic targeting that utilizes a magnetic field to specifically delivery theranostic agents to targeted tumor regions can greatly improve the cancer treatment efficiency. Herein, we load chlorin e6 (Ce6), a widely used PS molecule in PDT , on polyethylene glycol (PEG) functionalized iron oxide nanoclusters (IONCs), obtaining IONC–PEG–Ce6 as a theranostic agent for dual-mode imaging guided and magnetic-targeting enhanced in vivo PDT. Interestingly, after being loaded on PEGylated IONCs, the absorbance/excitation peak of Ce6 shows an obvious red-shift from ∼650 nm to ∼700 nm, which locates in the NIR region with improved tissue penetration. Without noticeable dark toxicity, Ce6 loaded IONC–PEG (IONC–PEG–Ce6) exhibits significantly accelerated cellular uptake compared with free Ce6, and thus offers greatly improved in vitro photodynamic cancer cell killing efficiency under a low-power light exposure. After demonstrating the magnetic field (MF) enhanced PDT using IONC–PEG–Ce6, we then further test this concept in animal experiments. Owing to the strong magnetism of IONCs and the long blood-circulation time offered by the condensed PEG coating, IONC–PEG–Ce6 shows strong MF-induced tumor homing ability, as evidenced by in vivo dual modal optical and magnetic resonance (MR) imaging. In vivo PDT experiment based magnetic tumor targeting using IONC–PEG–Ce6 is finally carried out, achieving high therapeutic efficacy with dramatically delayed tumor growth after just a single injection and the MF-enhanced photodynamic treatment. Considering the biodegradability and non-toxicity of iron oxide, our IONC–PEG–Ce6 presented in this work may be a useful multifunctional agent promising in photodynamic cancer treatment under magnetic targeting.

A polymer-encapsulated colorful Al pigment (PCAP) for fabric coatings was successfully prepared by a combined sol–gel spray drying assisted in situ polymerization method, and the near-infrared (NIR) reflectance and ultraviolet protection factor (UPF) of the coated fabrics were investigated. TG-DSC, SEM, and antisolvent test results proved that the reactive dyes were chemically attached to KH-550 modified Al pigment via a covalent bond. The PCAP was stable in strong alkaline (pH 12) or acidic (pH 1) conditions. The NIR reflectance and ultraviolet protection factor (UPF) of PCAP coated cotton fabrics were 80.05% and 290.76, respectively, when the mass ratio of monomer to colorful Al pigment was 1:10 with the coating thickness of 50 μm. PCAP coated fabrics showed excellent color strength, and good washing and rubbing fastness with a relatively good handle.

A near-infrared (NIR) fluorescent “turn-on” probe is presented for dissolved carbon dioxide (dCO 2 ) detection in aqueous medium. This probe is based on perylene bisimide-functionalized hyperbranched polyethylenimine (PBI-HPEI) polymer, which exhibits typical NIR fluorescence due to the excimer emission of π–π stacked PBI fluorophores. Both UV–vis absorption and fluorescent spectroscopy studies reveal that the PBI-HPEI polymer shows significantly dCO 2 -sensitive NIR emission via a known photoinduced electron transfer (PET) mechanism. Importantly, this sensory system is characterized with facile synthesis, fast response, excellent water-solubility and wide linear detection concentration range. It is thus capable of detecting dCO 2 even in real-world carbonated beverages and drinking water samples. Additionally, in view of the modular design principle of the present fluorescent polymer with typical NIR excimer emission of π–π stacked PBI molecules, the proposed strategy is expected to be applicable to the fabrication of novel versatile NIR-emitting sensory materials.

Near-infrared (NIR) light has great potential in biomedical applications due to its advantages of deep penetration depth and low photodamage to biological tissues. In this paper, we constructed a novel core-shell structured drug nanocarrier, Fe 3 O 4 @Au@SiO 2 , for the controlled delivery of etoposide (VP16), a chemotherapeutic drug for cancer patients. The novel core-shell structured drug delivery platform is composed of a mesoporous silica shell and a magnetic Fe 3 O 4 core using Au nanoparticles (AuNPs) as the interlayer, which is characterized by atomic force microscopy (AFM), X-ray diffraction (XRD), Fourier-transform infrared (FTIR) spectroscopy, N 2 adsorption/desorption isotherms and the magnetic measurements with vibrating-sample magnetometer (VSM). The synergistic effects of AuNPs, mesoporous silica and Fe 3 O 4 make the core-shell structured nanocomposites an excellent candidate for targeted and NIR light irradiation-controlled drug delivery. For the proposed nanocarrier of VP16, the mesopores in silica can enhance the encapsulation capacity of the nanocarrier and the AuNPs can effectively convert the NIR light into heat to speed up the drug deliver; meanwhile, the incorporation of Fe 3 O 4 with high magnetization to the drug delivery platform realize drug targeting under an applied external magnetic field.

Gold nanorods (AuNRs) are important photothermal therapeutic agents; however, a single therapy does not achieve satisfactory outcomes, and the synthesis process often leads to the adsorption of cetyltrimethylammonium bromide on the surface of AuNRs, which reduces its biocompatibility. Natural polyphenols are abundant in natural plants and have good biocompatibility. The metal-polyphenol network is formed by the coordination of metal ions and polyphenols, which has good drug loading, surface adhesion, and biocompatibility. In this study, the metal-polyphenol network structure formed by a transition metal (iron) and natural polyphenol tannic acid was used to modify the surface of gold nanorods (AuNRs@TF). Additionally, the surfaces of AuNRs were modified using the targeted functional molecule mercapto folic acid (AuNRs@TFF). The constructed composite nanomaterials AuNRs@TFF has good biocompatibility and tumor targeting ability. Tannic acid‑iron degrades in the tumor microenvironment and releases iron ions that catalyze the Fenton reaction, thereby facilitating chemodynamic therapy. The good photo-thermal ability of AuNRs generate good photoacoustic signals to facilitate photoacoustic imaging mediation and enhances photothermal and chemodynamic therapy performance. This study expands on the application of AuNRs in the field of nanomedicine. The simple and effective design of AuNRs@TFF provides a strategy for the development of synergistic therapeutic agents for photothermal therapy and chemodynamic therapy.

Hollow natural polysaccharide microcapsules have broad applications in drug delivery field due to their excellent biocompatibility and drug loading efficiency. In this paper, pH/near-infrared (NIR) dual-responsive microcapsules composed of hyaluronic acid (HA), chitosan (CS) and hollow CuS (HA/CS/HA@CuS) had been fabricated via a layer-by-layer (LbL) approach. The negative charge, rough surface and hollow structure of microcapsules are very favorable for loading positively charged DOX. As a result, hollow microcapsules display a high drug loading efficiency of 91.15 %. The variation in the degree of amino ionization at different pH values leads to the changes in the electrostatic force between CS/HA multilayers, resulting in the structural change in microcapsules. Therefore, microcapsules exhibit significant pH-responsive drug release properties. In addition, hollow CuS nanoparticles with excellent photothermal conversion ability are capped on the multilayer surface, enabling microcapsules to exhibit excellent NIR-responsive drug delivery properties. Overall, hyaluronic acid/chitosan-based hollow microcapsules with notable pH/NIR dual-responsiveness have been prepared, which can be used as a potential drug carrier for controlled drug delivery and photothermal chemical combination therapy.

Cancer which causes high mortality globally threatens public health seriously. There is an urgent need to develop tumor-specific near-infrared (NIR) imaging agents to achieve precise diagnosis and guide effective treatment. In recent years, imaging probes that respond to acidic environments such as endosomes, lysosomes, or acidic tumor microenvironments (TMEs) are being developed. However, because of their nonspecific internalization by both normal and tumor cells, resulting in a poor signal-to-noise ratio in diagnosis, these pH-sensitive probes fail to be applied to in vivo tumor imaging. To address this issue, a cholecystokinin-2 receptor (CCK2R)-targeted TME-sensitive NIR fluorescent probe R2SM was synthesized by coupling pH-sensitive heptamethine cyanine with a CCK2R ligand, minigastrin analogue 11 (MG11) for in vivo imaging, in which MG11 would target overexpressed CCK2Rs in gastrointestinal stromal tumors (GISTs). Cell uptake assay demonstrated that R2SM exhibited a high affinity for CCK2R, leading to receptor-mediated internalization and making probes finally accumulated in the lysosomes of tumor cells, which suggested in the tumor tissues, the probes were distributed in the extracellular acidic TME and intracellular lysosomes. With a pKa of 6.83, R2SM can be activated at the acidic TME (pH = 6.5–6.8) and lysosomes (pH = 4.5–5.0), exhibiting an apparent pH-dependent behavior and generating more intense fluorescence in these acidic environments. In vivo imaging showed that coupling of MG11 with a pH-sensitive NIR probe facilitated the accumulation of probe and enhanced the fluorescence in CCK2R-overexpressed HT-29 tumor cells. A high signal was observed in the tumor region within 0.5 h postinjection, indicating its potential application in intraoperative imaging. Fluorescence imaging of R2SM exhibited higher tumor-to-liver and tumor-to-kidney ratios (2.1:1 and 2.3:1, respectively), compared separately with the probes that are lipophilic, pH-insensitive, or MG11-free. In vitro and in vivo studies demonstrated that the synergistic effect of tumor targeting with pH sensitivity plays a vital role in the high signal-to-noise ratio of the NIR imaging probe. Moreover, different kinds of tumor-targeting vectors could be conjugated simultaneously with the NIR dye, which would further improve the receptor affinity and targeting efficiency.

Developing deep-red to near infrared (NIR) materials with multi-mode luminescence is challenging, which has promising utilization in various applications such as stress sensing and dynamic signature anti-counterfeiting. Different to the mainstream non-centrosymmetric mechanoluminescence (ML), a centrosymmetric perovskite compound (MgGeO 3 : Mn 2+ ) ML material is reported. Deep-red to NIR light (600–800 nm) from Mn 2+ ion is confirmed, which is attributed to the strong crystal field within the [MgO 6 ] polyhedron. The stiffness tetrahedral units and large defect accommodating space within the host are favorable for both ML and persistent luminescence (PersL). Mn 2+ ion is the only luminescent center, which is confirmed by spectral characterizations. Based on diffuse reflectance spectra, thermoluminescence analysis and density functional theory calculations, the inherent relationship between PersL, ML and PL is further clarified in detail. Due to the formation of Mn Mg defects, shallow trap significantly become shallower after adding Mn 2+ ions. This study reports a Mn 2+ -doped deep-red to NIR ML phosphor, and further describes the PersL and ML mechanisms via defect engineering regulation. In general, this study may also open up a new approach for the development of efficient NIR ML materials, and show various potential applications such as in anti-counterfeiting and information encryption fields.

Planar 1D photonic crystals (1DPhCs), owing to their photonic bandgaps (PBGs) formed by unique structural interference, are widely utilized in light protection applications. Multifunctional coatings that integrate various light management functions are highly desired. In this study, we present the fabrication of dual-PBG 1DPhCs with high reflectance in both the blue and near-infrared (NIR) regions. The 1DPhCs were constructed through the alternating stacking of hollow SiO2 (HS) nanoparticles hybridized with poly(vinyl alcohol) (PVA), referred to as PHS, and TiO2 layers. The 1DPhC with 7 bilayers demonstrated remarkable reflectance of 98.2% and 93.4% in the blue and NIR spectra, respectively, with shielding efficiencies of 93.9% and 85.6%. Compared to bare PC, the temperature was observed to be 10.6 °C lower, achieving a thermal insulation efficiency of 44.8% in the 900 nm near-infrared range. Additionally, spectral fitting using the transfer matrix method (TMM) and optimization of process parameters, including PHS concentration and spin-coating speed, enabled the fabrication of 1DPhCs with tunable optical thickness ratios. The fabricated films demonstrated a smooth surface morphology with flat and uniform interfaces, as well as excellent environmental stability. These features make the 1DPhCs highly promising for applications in light protection and thermal management. This study provides a novel approach for the development of dual-PBG 1DPhCs with enhanced protective capabilities.

Acute kidney injury (AKI) is a prevalent critical condition in hospitalized patients with an elevated risk of death. Challenges in AKI management persist due to the absence of efficient early diagnostic modalities and the lack of effective therapeutic interventions, imposing significant burdens on medical resources for prolonged treatment. Here, we proposed a second near-infrared (NIR-II) photoacoustic imaging (PAI) guided triggered on-demand release of (nitric oxide) NO from the injury zone to monitor and treat AKI through the anti-inflammatory, anti-apoptotic, and oxidative stress-damage-reducing cell- or organ-protective effects of NO. We engineered hollow copper sulfide nanoparticles (Cu 9 S 8 NPs) loaded with platelet membrane-encapsulated NO-releasing precursors (S-Nitroso-N-acetylpenicillamine, SNAP), as a nanoplatform for early AKI diagnosis and precise spatiotemporal NO release Besides acting as a drug carrier, the inherent PAI compatible Cu 9 S 8 also enables targeted renal injury localization, exploiting the natural propensity of platelets to accumulate at vascular injury sites, facilitating real-time AKI monitoring in murine models. Experiments demonstrated that nitric oxide delivery nanoparticles can specifically target and accumulate in the kidney, reduce inflammation, and promote damage repair. Overall, this gas therapy strategy represents a new therapeutic pathway for the clinical AKI management.

In this study, the mesoporous Fe3O4 nanodrug carriers containing disulfide bonds (CHO-SMNPs) were successfully synthesized and characterized. Doxorubicin (DOX) was loaded onto the CHO-SMNPs as a model drug and gatekeeper through the formation of imine bonds with the aldehyde groups on the surface of the mesoporous materials. This drug carrier demonstrates effective drug release triggered by pH, glutathione (GSH), and near-infrared (NIR) light, along with satisfactory photothermal conversion efficiency under NIR irradiation at 808 nm. Furthermore, CHO-SMNPs exhibit excellent blood compatibility and biodegradability. They also show good biocompatibility and efficient cellular uptake in HeLa and MCF-7 cancer cells. Most importantly, the CHO-SMNPs/DOX has shown significant effectiveness in killing both HeLa and MCF-7 cancer cells. Consequently, CHO-SMNPs/DOX presents substantial potential as a magnetic-targeted, pH/GSH/NIR triple-triggered drug delivery system for synergistic chemo-photothermal therapy in tumor treatment.
