Study of 8 Types of Glutathione Peroxidase Mimics Based on β-Cyclodextrin

Liwu Wang,Xiaonan Qu,Ying Xie,Shaowu Lv

Glutathione peroxidase is key for the removal of H2O2and other hydroperoxides and therefore, it has an important role in the maintenance of the reactive oxygen species (ROS) metabolic balance in vivo. The native enzymes of the glutathione peroxidase family (GPx) have many defects, such as instability in vitro and poor availability. GPx mimetics has become a topic of considerable interest in artificial enzyme research. Many forms of GPx mimics have been synthesized, by including selenium and tellurium (double-bridged and single-bridged, 2-substituted and 6-substituted) in a mother molecule but differences the GPx mimics enzymatic activity have rarely been compared. We designed and synthesized eight cyclodextrin derivatives and used two types of enzyme assays to determine their activities. The results show that: (a) tellurium-containing GPx mimics have higher activity than that of selenium-containing GPx mimics; (b) dual-bridged mimics have higher activity than bis-bridged mimics; and (c) 2-position modified cyclodextrin has higher activity than 6-position modified cyclodextrin. Keywords:glutathione peroxidase;mimic enzyme;cyclodextrin derivatives;enzyme activity;kinetics

Preparation and Antibacterial Activities of Porous Silver-doped β-Tricalcium Phosphate Bioceramics

Shen Liu,Cunyi Fan,Fangchun Jin,Li Zhao,Kerong Dai,Jianxi Lu

Infection is still a major concern in bone implants, especially in the implants with porous structures. As silver shows superior and broad-spectrum antibacterial activity, porous silver-doped β-tricalcium phosphate (β-TCP) bioceramics are prepared with 5% and 10% nanometer silver. The bioceramics show similar porous macrostructure with pure β-TCP bioceramic, except slightly color change. They have almost identical microstructure to its pure β-TCP counterpart under field emission scanning electron microscope. Their physical, chemical, and mechanical properties were investigated with X-ray diffraction, Fourier transforming infrared spectrometer, and AG-5kN, and no significant difference has been found between silver-doped β-TCP bioceramics and pure β-TCP bioceramic. Bactericidal concentration of silver ions was detected in the solution soaked with the bioceramics. They can efficiently inhibit the growth of Staphylococcus epidermidis and Styphylococcus aureus , but show no cytotoxicity to L929 cells. It suggests that silver-doped β-TCP bioceramics can be developed into new type of bone substitutes with anti-infection properties.

Interaction poses, intermolecular forces, dynamic preferences between flavonoids and maltosyl-β-cyclodextrin

Ruihong Wang,Botian Ding,Guizhao Liang

Cyclodextrins (CDs) can improve the solubility and stability of flavonoids by forming supramolecular complexes. However, the detailed inclusion mechanism has not been fully elucidated. In this study, the binding ability of eight flavonoids with four common CDs (α-, β-, methyl-β-, γ-CD) was investigated. Then, the interaction characteristics of two flavonoids (Dihydromyricetin (DMY) and Naringenin) with M-β-CD were further explored by using phase solubility, ultraviolet, infrared, X-ray diffraction, differential scanning calorimetry, molecular docking and ONIOM calculations. The results indicated that the molecular volume of flavonoids and the cavity size of CDs have distinct effects on the host-guest interaction. DMY and Naringenin form stable supramolecular complexes with M-β-CD mainly through hydrophobic interactions and Van der Waals forces. In addition, the stability of the Naringenin/m-β-CD is higher than that of DMY/M-β-CD, which is consistent with the experimental stability constant. Both of the above flavonoids entered the large hole of the hydrophobic cavity of M-β-CD through one end of the B-ring, which has a significant contribution to the formation of the host-guest hydrogen bonds . Taken together, this study is helpful to further understand the interaction mechanism between CDs and guest molecules, expand the application of CDs, and promote the utilization of natural active compounds.

Expression and application of α/β hydrolase from Kurthia gibsonii TYL-A1

Ye Wang,Heshi Tian,Qiannan Li,Zhenhua Ma,Yunna Jia,Xiaojun Liang,Xiuzhen Yu,Xiuyun Jiang,Yunhang Gao

Tylosin (TYL), a widely used macrolide antibiotic in livestock and poultry production, has attracted considerable attention owing to its strong adsorption capacity, persistence, and ecological risks in aquatic and soil environments. Because of their high efficiency and substrate specificity, microbial degradation enzymes are among the most promising agents for removing TYL from the environment. In this study, α/β hydrolase (Rcmd-2829) was expressed and purified from Kurthia gibsonii TYL-A1. The results showed that the optimum reaction conditions for Rcmd-2829 were pH 9.0 and 30°C. Molecular docking analyses, molecular dynamics simulations, and site-directed mutagenesis confirmed the key role of the Ser-102 residue in the catalytic degradation of TYL by Rcmd-2829. Rcmd-2829 retained its high catalytic activity in aquaculture wastewater, removing 83 % of 150 mg/L of TYL within 5 d. This study is the first to elucidate the Ser-102-dependent molecular mechanism underlying TYL degradation by the α/β hydrolase Rcmd-2829, providing a foundation for the development of efficient enzymatic-remediation technologies.

Efficient Oral Insulin Delivery Through Thiolated Trimethyl Chitosan-Grafted β-Cyclodextrin Nanoparticles

Lizhen Yu,Fengge Wang,Shuyun Bao,Yue Zhang,Xuebin Shen,Desheng Wang,Zhisheng Liu,Xinyi Liu,Lihua Li,Renmin Gong

Background: Oral insulin improves compliance and convenience in patients with diabetes who require regular needle injections. However, the clinical application of oral insulin preparations has been limited due to instability and inefficient permeation through the gastrointestinal tract. In this study, a novel cationic polysaccharide nanodrug delivery platform was designed for efficient oral insulin delivery. Methods: The innovative thiolated trimethyl chitosan-grafted β-cyclodextrin (NCT) was synthesized by utilizing N-trimethyl chitosan (TMC) as the polymer backbone. This involved modifying TMC with thiol group-containing N-acetylcysteine and carboxymethyl-β-cyclodextrin that possesses hydrophobic cavities via an amide condensation reaction. Subsequently, this polymer was employed to construct the NCT nanoparticle system using an ionic cross-linking method. The physicochemical properties of the NCT nanoparticles were systematically analyzed, and their therapeutic efficacy was comprehensively evaluated in streptozotocin (STZ)-induced animal models. Results: The NCT nanoparticles demonstrated mucus adhesion, permeability, and pH sensitivity, which facilitated a slow and controlled release within the gastrointestinal microenvironment due to both ionic electrostatic interactions and disulfide bonding interactions. The experiments revealed in vivo that insulin/NCT nanoparticles extended the retention time of insulin in the small intestine. Blood glucose levels decreased to approximately 39% of the initial level at 5 h post-administration while exhibiting smooth hypoglycemic efficacy. Simultaneously, insulin bioavailability increased to 12.58%.Conclusions: The NCT nanoparticles effectively protect insulin from degradation in the gastrointestinal microenvironment while overcoming intestinal barriers, thereby providing a promising approach to oral biomolecule delivery.

Extracellular Matrix Inspired Molecular Polarization in Hydroxypropyl-β-Cyclodextrin Organic Dielectric Films

Huang Yuxuan,Lu Hongwei,Yang Shijia,Su Weitao

Inspired by the extracellular matrix (ECM), which integrates a fibrous collagen scaffold with a hydrated polysaccharide matrix, we developed a PVDF-based ternary dielectric energy storage film by introducing hydroxypropyl-β-cyclodextrin (HP-β-CD) within a fluorinated methacrylate crosslinked network. The crosslinked network serves as a compact insulating scaffold analogous to collagen fibers, while HP-β-CD mimics the hydrated polysaccharide components. The abundant hydroxyl groups of HP-β-CD form stable hydrogen bonds with PVDF chains, strengthening interfacial adhesion and tuning local chain conformation. Under an external electric field, the polar hydroxyl and hydroxypropyl groups of HP-β-CD contribute to orientational polarization, increasing the dielectric constant to 9.7. In addition, the hydroxyl-rich exterior and cavity-bearing structure of HP-β-CD may provide charge-trapping sites that help suppress charge accumulation and conduction losses, thereby retarding electrical breakdown. The fluorinated crosslinked network enhances breakdown strength by restricting PVDF segmental motion and improving film compactness and structural stability. Consequently, the leakage current density decreases to 5.25 × 10 –7 A/cm 2 , approximately one order of magnitude lower than that of neat PVDF (2.02 × 10 –6 A/cm 2 ). Benefiting from this ECM-inspired molecular-regulation approach, the optimized composite achieves a discharged energy density of 9.03 J/cm 3 at 560 MV/m, while the residual PVDF-related hysteresis indicates that further efficiency optimization remains necessary for practical high-efficiency capacitor applications.

Protocol for tumor detection in mice using β-glucuronidase targeted breath analysis

Guochao Wu,Tianran Si,Han Jin

Early tumor detection is critical for improving cure rates. Here, we present a protocol for tumor detection in mice using a targeted breath-analysis strategy in which phenyl-β-D-glucuronide specifically targets β-glucuronidase, which is highly expressed in the tumor microenvironment. We describe steps for drug screening, enzymatic reaction, drug stability and toxicity evaluation, and construction of the mouse exhaled-breath extraction device. We then detail procedures for animal-model breath analysis, enabling highly sensitive early tumor detection. Mechanism of bile salt-promoted β-lactoglobulin digestion: a structural demasking perspective

Zhiyue Zhang,Timothy V. Kirk,Xiao Dong Chen

CD/FTIR showed limited secondary-structure rearrangement after BS exposure. • ANS fluorescence indicated increased hydrophobic exposure after BS addition. • Bile salts increase the accessibility of cleavage sites near hydrophobic cores. • LC–MS revealed BS-dependent changes in β-Lg peptide profiles. • Bile salts reroute proteolysis via site-specific demasking. The structural stability of the β-lactoglobulin (β-Lg) lyx confers high resistance to gastric proteolysis. While bile salts are known to promote its intestinal digestion, the specific conformational mechanisms and their impact on the resulting peptide cleavage patterns are not fully understood. In this study, we investigated the influence of bovine bile salts on the structural reorganization and digestibility of β-Lg during simulated gastrointestinal digestion. Conformational analyses indicated that bile salts induced limited structural perturbation within an overall retained secondary-structure framework, accompanied by increased exposure of hydrophobic regions. These changes were consistent with increased accessibility of selected cleavage regions. LC–MS peptide mapping showed that bile salts altered the peptide profile and promoted the degradation of regions that were more resistant under BS-free conditions. Overall, the findings support a demasking-related interpretation of how bile salt–protein interactions may influence β-Lg proteolysis under simplified in vitro conditions. Green β-cyclodextrin-based molecularly imprinted polymer for selective adsorption of sulfamethoxazole

Hui Feng,Jiaxin Xu,Jieyu Xu,Hua Zou

Sulfamethoxazole (SMX), a widely detected antibiotic residue, threatens human health and ecosystems. For selective SMX removal, a green molecularly imprinted polymer (CD-MIP) was synthesized using β-cyclodextrin (β-CD) as functional monomer via a facile route, with comparative evaluation against conventional methacrylic acid-based MIP (MAA-MIP). Molecular conformation modeling and spectroscopic analysis revealed stronger SMX-β-CD binding, guiding polymer design. CD-MIP achieved both higher sorption capacity (8.02 mg/g) and imprinting factor (2.34) within 60 min compared to MAA-MIP (7.21 mg/g; 1.10). CD-MIP demonstrated eco-friendliness with minimal desorption, strong matrix interference resistance, and high SMX selectivity. Practical tests confirmed its high selectivity, underscoring its potential for safeguarding water supply security. Adsorption was driven by physisorption via hydrogen bonding, π-π and hydrophobic interactions, with imprinted sites playing a crucial role. The host-guest recognition capability of β-CD was preserved in CD-MIP, with diverse functional groups enabling efficient imprinting, and non-imprinted polymer controls could reduce template use. A comprehensive sustainability assessment incorporating green metrics, life cycle analysis and economic evaluation confirmed the environmental and economic viability. This work establishes β-CD-based molecular imprinting as a sustainable strategy that effectively combines inherent molecular recognition with environmental adaptability, offering a cost-effective solution for safeguarding water supply.

Controllable Hydrothermal Synthesis of 1D β-Ga2O3 for Solar-Blind Ultraviolet Photodetection

Lingfeng Mao,Xiaoxuan Wang,Chaoyang Huang,Yi Ma,Feifei Qin,Wendong Lu,Gangyi Zhu,Zengliang Shi,Qiannan Cui,Chunxiang Xu

Gallium oxide (Ga2O3), an ultrawide bandgap semiconductor, is an ideal material for solar-blind photodetectors, but challenges such as low responsivity and response speed persist. In this paper, one-dimensional (1D) Ga2O3nanorods were designed to achieve high photodetection performance due to their effective light absorption and light field confinement. Through modulating source concentration, pH value, temperature, and reaction time, 1D β-Ga2O3nanorods were controllably fabricated using a cost-effective hydrothermal method, followed by post-annealing. The nanorods had a diameter of ~500 nm, length from 0.5 to 3 μm, and structure from nanorods to spindles, indicating that different β-Ga2O3nanorods can be utilized controllably through tuning reaction parameters. The 1D β-Ga2O3nanorods with a high length-to-diameter ratio were chosen to construct metal-semiconductor-metal type photodetectors. These devices exhibited a high responsivity of 8.0 × 10−4A/W and detectivity of 4.58 × 109Jones under 254 nm light irradiation. The findings highlighted the potential of 1D Ga2O3nanostructures for high-performance solar-blind ultraviolet photodetectors, paving the way for future integrable deep ultraviolet optoelectronic devices.

Structuring β-Ga2O3 Photonic Crystal Photocatalyst for Efficient Degradation of Organic Pollutants

Xiaofang Li,Xiuzheng Zhen,Sugang Meng,Jiangjun Xian,Yu Shao,Xianzhi Fu,Danzhen Li

Coupling photocatalysts with photonic crystals structure is based on the unique property of photonic crystals in confining, controlling, and manipulating the incident photons. This combination enhances the light absorption in photocatalysts and thus greatly improves their photocatalytic performance. In this study, Ga2O3 photonic crystals with well-arranged skeleton structures were prepared via a dip-coating infiltration method. The positions of the electronic band absorption for Ga2O3 photonic crystals could be made to locate on the red edge, on the blue edge, and away from the edge of their photonic band gaps by changing the pore sizes of the samples, respectively. Particularly, the electronic band absorption of the Ga2O3 photonic crystal with a pore size of 135 nm was enhanced more than other samples by making it locate on the red edge of its photonic band gap, which was confirmed by the higher instantaneous photocurrent and photocatalytic activity for the degradation of various organic pollutants under ultraviolet light irradiation. Furthermore, the degradation mechanism over Ga2O3 photonic crystals was discussed. The design of Ga2O3 photonic crystals presents a prospective application of photonic crystals in photocatalysis to address light harvesting and quantum efficiency problems through manipulating photons or constructing photonic crystal structure as groundwork.

Asymmetric Hydrosilylation of β-Silyl Styrenes Catalyzed by a Chiral Palladium Complex

Yi-Fan Wang,Yu-Han He,Yan Su,Yang Ji,Rui Li

A palladium complex coordinated with a chiral SIPHOS ligand was evaluated as an efficient catalyst for asymmetric hydrosilylation of β-silyl styrenes with trichlorosilane and 23 1,2-bis(silyl) chiral compounds were produced. Good to excellent enantioselectivities were observed with 1-aryl-2-silyl ethanols, where the trichlorosilyl groups of the hydrosilylation products were selectively converted into a hydroxyl group in the presence of pre-installed trialkylsilyl groups. Asymmetric hydrosilylation of β-silyl styrenes followed by methylation of the trichlorosilyl group gave stable 1,2-bis(silyl) chiral compounds 4 with excellent yields. DFT calculations of hydridopalladium B coordinated with a SIPHOS ligand, an intermediate of the hydrosilylation reaction, established the optical structures to be energy minima, and the structural information could well illustrate the enantioselectivity for the hydrosilylation reaction.
