Osteoarthritis (OA), a common degenerative disease, is characterized by high disability and imposes substantial economic impacts on individuals and society. Current clinical treatments remain inadequate for effectively managing OA. Organoids, miniature 3D tissue structures from directed differentiation of stem or progenitor cells, mimic native organ structures and functions. They are useful for drug testing and serve as active grafts for organ repair. However, organoid construction requires extracellular matrix-like 3D scaffolds for cellular growth. Hydrogel microspheres, with tunable physical and chemical properties, show promise in cartilage tissue engineering by replicating the natural microenvironment. Building on prior work on SF-DNA dual-network hydrogels for cartilage regeneration, we developed a novel RGD-SF-DNA hydrogel microsphere (RSD-MS) via a microfluidic system by integrating photopolymerization with self-assembly techniques and then modified with Pep-RGDfKA. The RSD-MSs exhibited uniform size, porous surface, and optimal swelling and degradation properties. In vitro studies demonstrated that RSD-MSs enhanced bone marrow mesenchymal stem cells (BMSCs) proliferation, adhesion, and chondrogenic differentiation. Transcriptomic analysis showed RSD-MSs induced chondrogenesis mainly through integrin-mediated adhesion pathways and glycosaminoglycan biosynthesis. Moreover, in vivo studies showed that seeding BMSCs onto RSD-MSs to create cartilage organoid precursors (COPs) significantly enhanced cartilage regeneration. In conclusion, RSD-MS was an ideal candidate for the construction and long-term cultivation of cartilage organoids, offering an innovative strategy and material choice for cartilage regeneration and tissue engineering.

In order to exploit the applications of polypyrrole (PPy) derivatives in biosensors and bioelectronics, the different immobilization mechanisms of biomolecules onto differently functionalized conducting PPy films are investigated. Pyrrole and pyrrole derivatives with carboxyl and amino groups were copolymerized with ω -( N -pyrrolyl)-octylthiol self-assembled on Au surface by the method of the chemical polymerization to form a layer of the copolymer film, i.e. , poly[pyrrole- co -( N -pyrrolyl)-caproic acid] (poly(Py- co -PyCA)) and poly[pyrrole- co -( N -pyrrolyl)-hexylamine] (poly(Py- co -PyHA)), in which the carboxyl groups in poly(Py- co -PyCA) were activated to the ester groups. Based on the structure characteristics, the immobilization/hybridization of DNA molecules on PPy, poly(Py- co -PyCA) and poly(Py- co -PyHA) were surveyed by cyclic voltammograms measurements. For differently functionalized copolymers, the immobilization mechanisms of DNA are various. Besides the electrochemical properties of the composite electrodes of PPy and its copolymers being detected before and after bovine serum albumin (BSA) adsorption, the kinetic process of protein binding was determined by surface plasmon resonance of spectroscopy. Since few BSA molecules could anchor onto the PPy and its copolymers surfaces, it suggests this kind of conducting polymers can be applied as the protein-resistant material.

We have synthesized three novel organobismuth(V) complexes Ph 3 Bi(OOCC 6 H 3 F 2 ) 2 1 , Ph 3 Bi(OOCC 6 H 4 CF 3 ) 2 2 and Ph 3 Bi(OOCC 4 H 3 S) 2 3 , and their structures were characterized by IR spectroscopy, elemental analysis, 1 H NMR and single crystal X-ray diffraction. X-ray crystallography showed that all of the bismuth atoms adopt distorted trigonal–bipyramidal geometries. The complex molecules lead to one-dimensional chain structure and two-dimensional network structure through C–H…X (X = O, F, C) interactions. The interaction of the complex 1 with calf-thymus DNA (CT-DNA) was investigated by UV absorption spectroscopy, fluorescence emission spectroscopy and viscosity. All results revealed that the complex 1 binds to DNA via a intercalative mode. Furthermore the proliferation inhibitory activities of the three complexes on MDA-MB-231 breast cancer cells were investigated. The results indicated that all of the three complexes have superior inhibition of cellular proliferation.

The binding of benzoyl peroxide (BPO), a flour brightener, with calf thymus DNA (ctDNA) was predicted by molecular simulation, and this were confirmed using multi-spectroscopic techniques and a chemometrics algorithm. The molecular docking result showed that BPO could insert into the base pairs of ctDNA, and the adenine bases were the preferential binding sites which were validated by the analysis of Fourier transform infrared spectra. The mode of binding of BPO with ctDNA was an intercalation as supported by the results from ctDNA melting and viscosity measurements, iodide quenching effects and competitive binding investigations. The circular dichroism and DNA cleavage assays indicated that BPO induced a conformational change from B-like DNA structure towards to A-like form, but did not lead to significant damage in the DNA. The complexation was driven mainly by hydrogen bonds and hydrophobic interactions. Moreover, the ultraviolet–visible (UV–vis) spectroscopic data matrix was resolved by a multivariate curve resolution–alternating least–squares algorithm. The equilibrium concentration profiles for the components (BPO, ctDNA and BPO–ctDNA complex) were extracted from the highly overlapping composite response to quantitatively monitor the BPO–ctDNA interaction. This study has provided insights into the mechanism of the interaction of BPO with ctDNA and potential hazards of the food additive.

The interaction between graphene oxide (GO) and DNA is very sensitive to the environment. For example, under acidic conditions, the affinity of GO for DNA is enhanced, weakening the capability of GO to distinguish DNAs with different conformations. This effect has impeded the development of sensitive pH biosensors based on GO–DNA nanosystems. In this work, we systematically studied the affinity between GO and i-motif forming oligonucleotides (IFOs) at different pH values and developed a herring sperm DNA (HSD) treatment method. Using this method, HSD occupies the surface of GO, compromising the attractive force of GO that is significantly enhanced under acidic conditions. As a result, the ability of GO to distinguish between “open” and “closed” IFOs is successfully generalized to a wider pH range. Finally, a pH-sensitive GO–IFO nanosystem was fabricated that showed excellent sensing ability both in vitro and for intracellular pH detection. Because the interaction between GO and DNA is the basis for constructing GO–DNA biosensors, the strategy developed in this work shows great potential to be applied in a variety of other GO–DNA sensing systems.

DNA adsorption and recognition are selective for carbon nanotubes (CNTs). In this work, we reported a novel method for control of electrochemiluminescence (ECL) enhancement through selective assembly of CNTs with DNA on indium tin oxide (ITO) electrode. The successful assembly of CNTs with well-preserved electrochemical conductivity was confirmed by scanning electron microscope and electrochemical impedance spectroscopy measurements. In comparison with the bare ITO electrode, the presence of CNTs catalyzes electrochemical reaction of an ECL coreactant, tri- n -propylamine, present in the solution, producing significantly enhanced ECL signal and ensuring substantial signal amplification. It is demonstrated that the efficiency of ECL enhancement is inversely correlated with the DNA recognition reaction, offering a promising ECL sensing platform for label-free and universal bioassay. The proposed strategy definitely differs from those works previously reported the introduction of CNTs for ECL enhancement, allowing the rise of ECL enhancement to be designed based on a biological recognition process. Just instead of the capture probe sequences, it is confirmed that the assembly of DNA with CNTs enables quantitative analysis of the different types of targets, including nucleic acid, Hg 2+ and thrombin.

In this work, a highly effective biosensor supported on morpholino-functionalized magnetic microspheres was investigated for the sequence-specific detection of DNA. Briefly, morpholino was first modified on the surface of magnetic microspheres through amido bonds, and then hybridized with target DNA in the ensuing step. Biotin-labeled signal probe DNA was designed to hybridize with the DNA nearby the 5′ end of sequence, and the biotin was used to bind with streptavidin-alkaline phosphatase (SA-ALP). L-Ascorbic acid-2-phosphate (AAP) was hydrolyzed by ALP to generate L-ascorbic acid, which could reduce resazurin to resorufin, resulting in a turn-on fluorescence signal. UV-vis absorbance spectroscopy was applied to prove the conjugation of ALP with signal probe DNA. Under optimal conditions, this biosensor displayed a good linear relationship between the fluorescence intensity and logarithm of single-stranded DNA concentrations in the range of 0.1 pM to 0.1 nM with a low detection limit of 9.32 fM. Moreover, fully complementary versus single-base mismatched, three-base mismatched and non-complementary DNA could be effectively distinguished. In addition, this novel approach rendered satisfactory analytical results for the determination of DNA in serum, thus exhibiting practical significance. These results demonstrated that this assay method displays excellent sensitivity and specificity for DNA detection and has great potential for practical applications.

Many planar photosensitizers tend to self-aggregate via van der Waals interactions between π-conjugated systems. The self-aggregation of the photosensitizer may reduce the efficiency of the photosensitizer to generate singlet oxygen, thereby diminishing its photodynamic activity. Efforts have been made to improve the photodynamic activity of bis-( o -diiminobenzosemiquinonato)platinum(II) which has planar geometry by the introduction of the sterically hindered triphenylamine moiety into the ligand. Herein we report the photoinduced DNA damage and cytotoxicity by a triphenylamine-modified platinum-diimine complex in red light studied by fluorescence spectra , agarose gel assay and cell viability assay. The results suggest that the triphenylamine-modified platinum-diimine complex has better capability to generate singlet oxygen than bis-( o -diiminobenzosemiquinonato)platinum(II), and it can induce DNA damage in red light, causing high photocytotoxicity in HepG-2 cells in vitro.

The binding interaction of tetrabromobisphenol A with Calf thymus DNA was studied by multi-spectroscopic and molecular modeling methods. The UV–vis study revealed that an obvious interaction between tetrabromobisphenol A and Calf thymus DNA happened. The π – π ∗ transitions and the electron cloud of tetrabromobisphenol A might be changed by entering the groove of Calf thymus DNA. From the fluorescence spectral and thermodynamics studies, it was concluded that the hydrogen bonds and hydrophobic force played a major role in the binding of tetrabromobisphenol A to Calf thymus DNA. The molecular modeling study showed that the possible sites of tetrabromobisphenol A in the groove of DNA. Circular dichroism study also depicted that tetrabromobisphenol A bond to DNA. These above results would further advance our knowledge on the molecular mechanism of the binding interactions of brominated flame-retardants with nucleic acid.

We demonstrated an easy and efficient approach to produce amino-group-functionalized graphene (G-NH2) for highly sensitive DNA biosensing through intermolecular interaction. The nucleotide molecules prefer to anchor onto the as-prepared G-NH2 surface rather than be immobilized on pristine graphene. The electrostatic interaction between the positive charges of the ionized amino on G-NH2 and the negative charges of phosphate groups on nucleotide chains enabled label-free probe DNA to be immobilized on the G-NH2 surface. The electrochemical performance and quality variations during the preparation of G-NH2 and nucleotide immobilization processes were determined by electrochemical measurements and quartz crystal microbalance in situ, respectively. Results showed that complementary target DNA could be hybridized with probe DNA within the concentration range of 0.1 nM to 200 nM, and the detection limit was 0.8 nM. Therefore, this kind of G-NH2 could be an alternative for DNA biosensing.

By utilizing multispectrosopic techniques, the toxic interaction of 2-aminoanthraquinone (2-AAQ) with calf thymus deoxyribonucleic acid (ctDNA) was investigated in vitro under simulated physiological conditions. The experimental results proved that 2-AAQ has a toxic interaction with ctDNA. The binding capacity of DNA with 2-AAQ is diminishing as the pH value of system increasing in the optimization of experimental condition. Moreover we selected pH 7.4, which is nearly physiological condition to enhance the practical significance. According to the Stern–Volmer equation, the quenching was the static quenching process. And the quenching constant can be derived from the fluorescence quenching spectrogram. Ultraviolet absorption spectra and the change in the fluorescence intensity at different ionic strengths further indicated that there was electrostatic binding between 2-AAQ and ctDNA. The circular dichroism experiment showed that the DNA conformation varied from B to A conformation. The basic group enhanced after 2-AAQ embedding. The double helix is more compact, and the DNA conformation changes. © 2013 Wiley Periodicals, Inc. J BiochemMol Toxicol 27:272-278, 2013; View this article online at wileyonlinelibrary.com . A photoelectrochemical (PEC) sensing platform was designed for catechol determination using CdS quantum dots (QDs), DNA and pristine graphene (PGR) nanocomposite film-modified electrode. In such a composite film-based PEC sensor, CdS QDs served as the photoelectric conversion element which produced photocurrent signal under visible light illumination; and DNA was designed as the biological recognition element which improved the PEC response of sensor toward catechol due to the interaction between DNA and catechol. To improve the electron transfer of the composite film, PGR with high electrical conductivity was doped, which significantly amplified the photocurrent signal of sensor. Based on such a CdS-DNA-PGR composite film-modified electrode, catechol showed a linear PEC response proportional to its concentration from 1.0 × 10 −8 to 1.0 × 10 −6 mol L −1 . The detection limit (3S/N) was estimated to be 4.9 × 10 −9 mol L −1 . The developed PEC sensor was successfully applied to determine trace catechol in water samples.
