The superior optical properties of lead-halide perovskites (LHPs) inspired significant research in cell imaging applications; many encapsulating processes have improved perovskite stabilities with comparable biosafety. Herein, facile solvent evaporation encapsulation based on an oil-in-fluorine emulsion for aqueous-stable and extremely nontoxic LHP microcapsules is described. Perfluorooctane dispersed the emulsifier fluorocarbon surfactant to form a continuous fluorine phase, while LHPs and polymethylmethacrylate (PMMA) were dispersed in 1,2-dichloroethane, then emulsified in the fluorine phase to form an oil-in-fluorine emulsion. CsPbBr3microcapsules with a dense PMMA shell that protect fragile CsPbBr3from the external environment and inhibit lead ion release were obtained after solvent evaporation. The CsPbBr3microcapsules not only retained 91% of fluorescence intensity after exposure to water for 30 d but also possess extremely low cytotoxicity for MCF-7 cells. After exposure to 2 mg/mL of CsPbBr3microcapsules for 48 h, the cell viability remained >90%. The intracellular uptake of CsPbBr3microcapsules indicates its potential use in cell imaging.Keywords:lead-halide perovskites;microencapsulation;oil-in-fluorine emulsion;cell imaging

Chlorinated polyfluorinated ether sulfonate (F–53 B) is a distinct substitute for perfluorooctane sulphonate. It has been reported to be biologically toxic to mammals, causing enteric toxicity, liver toxicity and neurotoxicity. However, studies about the effects of F–53 B on patients with gastrointestinal diseases such as inflammatory bowel disease are very limited. In this study, whether the toxic impacts of F–53 B on the gut and liver can be exacerbated in mice with colitis was explored. The sensitivity of mice with acute colitis caused by dextran sulfate sodium salt (DSS) to F–53 B was compared with that of healthy mice. The mice were administered water containing F–53 B at doses of 10 and 100 μg/L sequentially for two weeks, respectively. F–53 B exposure exacerbated DSS-induced colonic inflammation, including inducing shortening of colon length, inflammatory cell infiltration and more severe histopathological symptoms. In addition, F–53 B administration significantly increased the levels of inflammatory cytokines, including interleukin (IL)-1, IL-6 and tumour necrosis factor-α, in the plasma of mice with enteritis compared with control group. F–53 B impaired intestinal integrity of mice with colitis by downregulating Claudin-1 and antimicrobial peptide-related genes while elevating serum lipopolysaccharide levels. In addition, in mice with colitis, F–53 B increased the levels of serum total cholesterol, triglyceride, low-density lipoprotein cholesterol, aspartate aminotransferase, and alanine aminotransferase, resulted in more severe liver inflammation and increased the level of genes related to the Gasdermin D-mediated pyrolysis . Conclusively, our results indicated that F–53 B delayed the self-healing of ulcerative colitis (UC) and caused liver inflammation in mice. This study provided some new insights into the health risks of F–53 B and raises concerns about the health of individuals with UC.

Bisphenol AP (BPAP), a structural analog of bisphenol A (BPA), has been widely detected in environment and biota. BPAP was reported to interfere with hormone and metabolism, while limited data were available about its effects on neurobehavior, especially exposure to it during early-life time. A mouse model of early-life BPAP exposure was established to evaluate the long-term neurobehaviors in offspring . Collectively, early-life BPAP exposure caused anxiety-like behaviors and impaired learning and memory in adult offspring. Through brain bulk RNA-sequencing (RNA-seq), we found differential expressed genes were enriched in pathways related to behaviors and neurodevelopment, which were consistent with the observed phenotype. Besides, single-nucleus RNA-sequencing (snRNA-seq) showed BPAP exposure altered the transcriptome of microglia in hippocampus. Mechanistically, BPAP exposure induced inflammations in hippocampus through upregulating Iba-1 and activating the microglia. In addition, we observed that BPAP exposure could activate peripheral immunity and promote proportion of macrophages and activation of dendritic cells in the offspring. In conclusion, early-life exposure to BPAP impaired neurobehaviors in adult offspring accompanied with excessive activation of hippocampal microglia. Our findings provide new clues to the underlying mechanisms of BPAP's neurotoxic effects and therefore more cautions should be taken about BPAP.

Surface charging in electron spectroscopy for chemical analysis (ESCA) detrimentally affects the analytical accuracy, and the common charge neutralization techniques struggle to handle the heterogeneous surface charging which plagues ESCA studies of many high-impact composites having different electrically resistive constituents. To facilitate the research of such composites, this report articulates the scientific causes of heterogeneous surface charging plaguing such research and discloses technical details in mitigating the problems by an in-situ sub-nanometer gold coating technique. Briefly, the report describes a method of translating the common in-situ sputter-cleaning facility in an X-ray photoelectron spectroscopy (XPS) system to a means for the precise deposition of a few gold monolayers onto a sample. Such an ultrathin gold overlayer guarantees the presence of a uniform surface potential that is virtually equal to spectrometer-ground and thereby relieves any heterogeneous surface charging problems in XPS. The Au 4f 7/2 peak of the overlayer is also a more accurate ESCA/XPS reference. Further, the report clarifies that the in-situ deposition of gold and the XPS monitoring of the deposition is essential. An arbitrary thick gold overlayer may block almost all XPS signals from the sample, and too little gold deposition fails the purpose of mitigating heterogeneous surface charging. In short, the method supports particularly studies of composites with heterogeneities in electrical conductivity .

Self-sensitized degradation is a typical fate of azo dyes in aquatic systems but its kinetics is far too slow to meet the requirement for a thorough decolorization. Small diketones were found being able to enhance the degradation of dyes under UV irradiation and the introduction of an electron-denoting group at the α -position of the simplest β -diketone (acetylacetone, AA) had a further enhancement effect on the degradation. However, it is unclear whether such enhancement effects exist in the solar-irradiated systems. To clarify this question, the effects of three AA derivatives were investigated with Acid Orange 7 (AO7) as a representative azo dye. Compared with the AA derivatives with a methyl or an ethyl group at the central carbon (AA-CH 3 and AA-CH 2 CH 3 ), 3,5-diacetylheptane-2,6-dione (AA-CH 2 -AA, which is a condensed combination of AA and AA-CH 3 ) showed a >2-fold stronger enhancement effect. The nonstoichiometric enhancement effect of AA-CH 2 -AA on AO7 photodegradation disclosed a novel pathway for dye degradation: AA-CH 2 -AA could enhance the degradation of AO7 by reacting with the photo-sensitized 1 O 2 , whereas AA led to an inhibitory effect on the self-sensitized degradation of AO7 by quenching the excited state of the azo dye. These results demonstrate that small diketones could be designed to regulate the photodegradation of azo dyes in multiple pathways.

l -glutamate ( l -Glu) is the basic unit for the synthesis of protein, metabolic substances and chlorophyll, and plays an important role in the carbon and nitrogen metabolism in plants. The in vivo monitoring of l -Glu level is of great significance to study the growth and development of plants. Although a variety of methods have been reported for the in vitro determination of l -Glu, there are very limited research focused on the determination of l -Glu in plants. Here we firstly report a simple method to construct a minitype glutamate sensor for in vivo monitoring of l -glutamate in cucumber. The minitype graphite rod electrode (GRE) allows its application for in vivo monitoring, and modification of Pt nanoparticles (PtNPs) and poly ( m -phenylenediamine) film (PMPD) significantly improves the sensitivity and anti-interference ability, and immobilization of the glutamate oxidase (GluOx) could effectively catalyze the l -Glu into a detectable H 2 O 2 . The resulting l -Glu sensor exhibits a good performance in terms of high sensitivity, good selectivity and excellent stability with a linear range of 2–550 μM and limit of detection (LOD) as low as 0.536 μM. More importantly, the as-developed sensor was successfully applied for the determination of l -Glu in cucumber juice and the in vivo monitoring of l -Glu level in cucumber fruit. We envision that the constructed sensor will show great potentials for studying the growth mechanisms of plants.

Designing active and stable electrocatalysts with economic efficiency for acidic oxygen evolution reaction is essential for developing proton exchange membrane water electrolyzers. Herein, we report on a cobalt oxide incorporated with iridium single atoms (Ir-Co 3 O 4 ), prepared by a mechanochemical approach. Operando X-ray absorption spectroscopy reveals that Ir atoms are partially oxidized to active Ir >4+ during the reaction, meanwhile Ir and Co atoms with their bridged electrophilic O ligands acting as active sites, are jointly responsible for the enhanced performance. Theoretical calculations further disclose the isolated Ir atoms can effectively boost the electronic conductivity and optimize the energy barrier. As a result, Ir-Co 3 O 4 exhibits significantly higher mass activity and turnover frequency than those of benchmark IrO 2 in acidic conditions. Moreover, the catalyst preparation can be easily scaled up to gram-level per batch. The present approach highlights the concept of constructing single noble metal atoms incorporated cost-effective metal oxides catalysts for practical applications.

Background The adipokine chemerin regulates adipogenesis and the metabolic function of both adipocytes and liver. Chemerin is elevated in preeclamptic women, and overexpression of chemerin in placental trophoblasts induces preeclampsia-like symptoms in mice. Preeclampsia is known to be accompanied by dyslipidemia, albeit via unknown mechanisms. Here, we hypothesized that chemerin might be a contributor to dyslipidemia. Methods Serum lipid fractions as well as lipid-related genes and proteins were determined in pregnant mice with chemerin overexpression in placental trophoblasts and chemerin-overexpressing human trophoblasts. In addition, a phospholipidomics analysis was performed in chemerin-overexpressing trophoblasts. Results Overexpression of chemerin in trophoblasts increased the circulating and placental levels of cholesterol rather than triglycerides. It also increased the serum levels of lysophosphatidic acid, high-density lipoprotein cholesterol (HDL-C), and and low-density lipoprotein cholesterol (LDL-C), and induced placental lipid accumulation. Mechanistically, chemerin upregulated the levels of peroxisome proliferator-activated receptor g, fatty acid-binding protein 4, adiponectin, sterol regulatory element-binding protein 1 and 2, and the ratio of phosphorylated extracellular signal-regulated protein kinase (ERK)1/2 / total ERK1/2 in the placenta of mice and human trophoblasts. Furthermore, chemerin overexpression in human trophoblasts increased the production of lysophospholipids and phospholipids, particularly lysophosphatidylethanolamine. Conclusions Overexpression of placental chemerin production disrupts trophoblast lipid metabolism, thereby potentially contributing to dyslipidemia in preeclampsia.

Natural biopolymers can be controllably in situ synthesized in organisms and play important roles in biological activities. Inspired by this, the manipulation of in situ biosynthesis of functional polymers in vivo will be an important way to obtain materials for meeting biological requirements. Herein, in situ biosynthesis of functional conjugated polymer at the tumor site was achieved via the utilization of specific tumor microenvironment (TME) characteristics for the first time. Specially, a water-soluble aniline dimer derivative (N-(3-sulfopropyl) p-aminodiphenylamine, SPA) was artfully in situ polymerized into polySPA (PSPA) nanoparticles at the tumor site, which was activated via the catalysis of hydrogen peroxide (H2O2) overexpressed in TME to produce hydroxyl radical (•OH) by coinjected horseradish peroxidase (HRP). Benefiting from outstanding near-infrared (NIR)-II absorption of PSPA, the in situ polymerization process can be validly monitored by photoacoustic (PA) signal at the NIR-II region. Meanwhile, in situ polymerization would induce the size of polymeric materials from small to large, improving the distribution and retention of PSPA at the tumor site. On the combination of NIR-II absorption of PSPA and the size variation induced by polymerization, such polymerization can be applied for tumor-specific NIR-II light mediated PA image and photothermal inhibition of tumors, enhancing the precision and efficacy of tumor phototheranostics. Therefore, the present work opens the way to manipulate TME-activated in situ biosynthesis of functional conjugated polymer at the tumor site for overcoming formidable challenges in tumor theranostics.

Pulmonary hypertension (PH) is a life-threatening cardiopulmonary disease characterized by pulmonary vascular remodeling. Excessive growth and migration of pulmonary artery smooth muscle cells (PASMCs) are believed to be major contributors to pulmonary vascular remodeling. Ubiquitin-specific protease 15 (USP15) is a vital deubiquitinase that has been shown to be critically involved in many pathologies. However, the effect of USP15 on PH has not yet been explored. In this study, the upregulation of USP15 was identified in the lungs of PH patients, mice with SU5416/hypoxia (SuHx)-induced PH and rats with monocrotaline (MCT)-induced PH. Moreover, adeno-associated virus-mediated functional loss of USP15 markedly alleviated PH exacerbation in SuHx-induced mice and MCT-induced rats. In addition, the abnormal upregulation and nuclear translocation of YAP1/TAZ was validated after PH modeling. Human pulmonary artery smooth muscle cells (hPASMCs) were exposed to hypoxia to mimic PH in vitro, and USP15 knockdown significantly inhibited cell proliferation, migration, and YAP1/TAZ signaling in hypoxic hPASMCs. Rescue assays further suggested that USP15 promoted hPASMC proliferation and migration in a YAP1/TAZ-dependent manner. Coimmunoprecipitation assays indicated that USP15 could interact with YAP1, while TAZ bound to USP15 after hypoxia treatment. We further determined that USP15 stabilized YAP1 by inhibiting the K48-linked ubiquitination of YAP1. In summary, our findings reveal the regulatory role of USP15 in PH progression and provide novel insights into the pathogenesis of PH.

Sulfur bonds, especially trisulfide bond, have been found to ameliorate the self-assembly stability of homodimeric prodrug nanoassemblies and could trigger the sensitive reduction-responsive release of active drugs. However, the antitumor efficacy of homodimeric prodrug nanoassemblies with single reduction-responsivity may be restricted due to the heterogeneous tumor redox microenvironment. Herein, we replace the middle sulfur atom of trisulfide bond with an oxidizing tellurium atom or selenium atom to construct redox dual-responsive sulfur-tellurium-sulfur and sulfur-selenium-sulfur hybrid chalcogen bonds. The hybrid chalcogen bonds, especially the sulfur-tellurium-sulfur bond, exhibit ultrahigh dual-responsivity to both oxidation and reduction conditions, which could effectively address the heterogeneous tumor microenvironment. Moreover, the hybrid sulfur-tellurium-sulfur bond promotes the self-assembly of homodimeric prodrugs by providing strong intermolecular forces and sufficient steric hindrance. The above advantages of sulfur-tellurium-sulfur bridged homodimeric prodrug nanoassemblies result in the improved antitumor efficacy of docetaxel with satisfactory safety. The exploration of hybrid chalcogen bonds in drug delivery deepened insight into the development of prodrug-based chemotherapy to address tumor redox heterogeneity, thus enriching the design theory of prodrug-based nanomedicines.

The formation of hydrate increases the uncertainty of the flow system in subsea multiphase transportation pipelines. Waxes and asphaltenes have been found to influence hydrate formation, but the detailed mechanism of their interaction remains unclear. A lab-scale high-pressure stirring reactor was used to analyze the influence of asphaltenes on CO2 hydrate evolution behavior in waxy water-in-oil (W/O) emulsions. Results demonstrated that waxes inhibited CO2 hydrate nucleation. The addition of asphaltenes promoted the inhibition effect of waxes, and the enhancement effect was nonmonotonic with the asphaltene contents increased. Based on microscopic observation and wax precipitation characteristics, this behavior could be interpreted as the competitive effect of increased adsorption of wax crystals on the surface of the droplets and reduced amount of wax precipitation. Moreover, asphaltenes were found to facilitate the decomposition process of CO2 hydrates in waxy systems by decreasing the wax precipitation amount and stabilizing the emulsion. This work can provide potential engineering guidance for flow assurance applications.
