Organic–Inorganic Hybrid One-Dimensional Photonic Crystals with Blue and NIR Dual Bandgaps for Light Protection

Minglu Zhang,Qi Yuan,Yanyan Guo,Saijin Song,Dongyu Wang,Junli Zhang,Hao-Yang Mi,Jian Han,Chuntai Liu,Changyu Shen

Planar 1D photonic crystals (1DPhCs), owing to their photonic bandgaps (PBGs) formed by unique structural interference, are widely utilized in light protection applications. Multifunctional coatings that integrate various light management functions are highly desired. In this study, we present the fabrication of dual-PBG 1DPhCs with high reflectance in both the blue and near-infrared (NIR) regions. The 1DPhCs were constructed through the alternating stacking of hollow SiO2 (HS) nanoparticles hybridized with poly(vinyl alcohol) (PVA), referred to as PHS, and TiO2 layers. The 1DPhC with 7 bilayers demonstrated remarkable reflectance of 98.2% and 93.4% in the blue and NIR spectra, respectively, with shielding efficiencies of 93.9% and 85.6%. Compared to bare PC, the temperature was observed to be 10.6 °C lower, achieving a thermal insulation efficiency of 44.8% in the 900 nm near-infrared range. Additionally, spectral fitting using the transfer matrix method (TMM) and optimization of process parameters, including PHS concentration and spin-coating speed, enabled the fabrication of 1DPhCs with tunable optical thickness ratios. The fabricated films demonstrated a smooth surface morphology with flat and uniform interfaces, as well as excellent environmental stability. These features make the 1DPhCs highly promising for applications in light protection and thermal management. This study provides a novel approach for the development of dual-PBG 1DPhCs with enhanced protective capabilities.

A hematoporphyrin and indocyanine green co-delivery system with NIR triggered-controllable photoactivities for photodynamic therapy

Ziying Li,Tingting Lv,Yingying Zhang,Liang Xu,Lu Zhang,Xiuying Wang,Haijun Chen,Yu Gao

Photodynamic therapy (PDT) was hindered by the poor water solubility and long-enduring photoactivities of photosensitizers . Here, we developed a novel controllable photodynamic agent CPHI based on β-cyclodextrin modified polyamidoamine (PAMAM) dendrimer (CP) co-delivery of indocyanine green (ICG) and hematoporphyrin (HP). The CPHI possessed appropriate particle sizes and zeta potentials , considerable photostability, and NIR-triggered release property. The fluorescence of HP could be quenched by ICG when HP and ICG were co-loaded inside the CP, and the quenching efficiency was related with the weight ratio of HP and ICG. When the feeding weight proportion of HP and ICG is 1/10, the fluorescence of HP was quenched by 87.3% in CPHI. Under 808 nm laser irradiation , the ICG in CP was degraded, and the fluorescence of HP could recover. The recovered HP was capable of generating reactive oxygen species (ROS) for PDT with 606 nm laser irradiation. The cytotoxicity and cell apoptosis studies showed that CPHI could effectively induce MCF-7 cells apoptosis with PDT effects under 808/660 nm laser. These results suggested that CPHI could be an efficient cancer therapeutic agent for controllable PDT with low dark toxicity.

Acidity-Triggered “Sticky Spotlight”: CCK2R-Targeted TME-Sensitive NIR Fluorescent Probes for Tumor Imaging In Vivo

Ruiqi Sun,Yuxin Wang,Wenhui Shi,Hongfu Zhang,Jianhua Liu,Weina He

Cancer which causes high mortality globally threatens public health seriously. There is an urgent need to develop tumor-specific near-infrared (NIR) imaging agents to achieve precise diagnosis and guide effective treatment. In recent years, imaging probes that respond to acidic environments such as endosomes, lysosomes, or acidic tumor microenvironments (TMEs) are being developed. However, because of their nonspecific internalization by both normal and tumor cells, resulting in a poor signal-to-noise ratio in diagnosis, these pH-sensitive probes fail to be applied to in vivo tumor imaging. To address this issue, a cholecystokinin-2 receptor (CCK2R)-targeted TME-sensitive NIR fluorescent probe R2SM was synthesized by coupling pH-sensitive heptamethine cyanine with a CCK2R ligand, minigastrin analogue 11 (MG11) for in vivo imaging, in which MG11 would target overexpressed CCK2Rs in gastrointestinal stromal tumors (GISTs). Cell uptake assay demonstrated that R2SM exhibited a high affinity for CCK2R, leading to receptor-mediated internalization and making probes finally accumulated in the lysosomes of tumor cells, which suggested in the tumor tissues, the probes were distributed in the extracellular acidic TME and intracellular lysosomes. With a pKa of 6.83, R2SM can be activated at the acidic TME (pH = 6.5–6.8) and lysosomes (pH = 4.5–5.0), exhibiting an apparent pH-dependent behavior and generating more intense fluorescence in these acidic environments. In vivo imaging showed that coupling of MG11 with a pH-sensitive NIR probe facilitated the accumulation of probe and enhanced the fluorescence in CCK2R-overexpressed HT-29 tumor cells. A high signal was observed in the tumor region within 0.5 h postinjection, indicating its potential application in intraoperative imaging. Fluorescence imaging of R2SM exhibited higher tumor-to-liver and tumor-to-kidney ratios (2.1:1 and 2.3:1, respectively), compared separately with the probes that are lipophilic, pH-insensitive, or MG11-free. In vitro and in vivo studies demonstrated that the synergistic effect of tumor targeting with pH sensitivity plays a vital role in the high signal-to-noise ratio of the NIR imaging probe. Moreover, different kinds of tumor-targeting vectors could be conjugated simultaneously with the NIR dye, which would further improve the receptor affinity and targeting efficiency.

Biomimetic gold nano-modulator for deep-tumor NIR-II photothermal immunotherapy via gaseous microenvironment remodeling strategy

Honglin Huang,Zhengxin Xie,Ningxi Li,Li Zeng,Qianyi Zeng,Ziman Yang,Jinyang Shen,Hong Yang,Yiyao Liu,Chunhui Wu

Introduction Effective immunotherapeutic treatment of solid tumors has been greatly challenged by the complex hostile tumor immunosuppressive microenvironment (TIME), which typically involves hypoxia and immunosuppression. Methods Herein, a multifunctional biomimetic gold nano-modulator (denoted as GNR-SNO@MMT ) was developed to realize the efficient second near-infrared (NIR-II) photothermal immunotherapy via tumor targeting and deep penetration, vascular normalization and immune reprogramming. NIR-II photothermal agent gold nanorods (GNR) were grafted with thermosensitive S-nitrosothiol (SNO) donors and camouflaged with the tumor-penetrating peptide tLyp-1-modified macrophage membrane (MM) to yield GNR-SNO@MMT . Results The engineered membrane coating increased the capacity for tumor inflammatory tropism and deep penetration, which aided GNR-SNO@MMT in ablating tumors together with NIR-II laser irradiation. Moreover, hyperthermia-stimulated nitric oxide (NO) release in situ acted as a gas immunomodulator to effectively enhance blood perfusion and reprogram the TIME via multiple functions (e.g., decreasing PD-L1, repolarizing tumor-associated macrophages, and revitalizing cytotoxic T cells). Ultimately, the inhibition rate against 4T1 mouse mammary tumor model mediated by GNR-SNO@MMT plus NIR-II laser was 94.7% together with 2.4-fold CD8 + T cells infiltrated into tumors than that of the untreated counterpart. Conclusions The engineered biomimetic nano-modulator of GNR-SNO@MMT provides an effective and novel photoimmunotherapy candidate against deep-sited solid tumors through immune reconfiguration via NO-involved nanomedicine and external NIR-II laser assistance. Biomimetic NIR-II fluorescent proteins created from chemogenic protein-seeking dyes for multicolor deep-tissue bioimaging

Xu Jiajun,Zhu Ningning,Du Yijing,Han Tianyang,Zheng Xue,Li Jia,Zhu Shoujun

Near-infrared-I/II fluorescent proteins (NIR-I/II FPs) are crucial for in vivo imaging, yet the current NIR-I/II FPs face challenges including scarcity, the requirement for chromophore maturation, and limited emission wavelengths (typically < 800 nm). Here, we utilize synthetic protein-seeking NIR-II dyes as chromophores, which covalently bind to tag proteins (e.g., human serum albumin, HSA) through a site-specific nucleophilic substitution reaction, thereby creating proof-of-concept biomimetic NIR-II FPs. This chemogenic protein-seeking strategy can be accomplished under gentle physiological conditions without catalysis. Proteomics analysis identifies specific binding site (Cys 477 on DIII). NIR-II FPs significantly enhance chromophore brightness and photostability, while improving biocompatibility, allowing for high-performance NIR-II lymphography and angiography. This strategy is universal and applicable in creating a wide range of spectrally separated NIR-I/II FPs for real-time visualization of multiple biological events. Overall, this straightforward biomimetic approach holds the potential to transform fluorescent protein-based bioimaging and enables in-situ albumin targeting to create NIR-I/II FPs for deep-tissue imaging in live organisms.

Switching between upconversion luminescence imaging and therapy in vitro enabled by NIR excitation modulation of nanocomposites

Yuan Jiang,Yale Hong,Yuan-Yuan Liu,Yue Guan,Junxun Zhou,Haifang Wang,Lining Sun

Herein, we designed and synthesized a theranostics-integrated nanocomposite capable of switching between upconversion luminescence imaging and photothermal therapy (PTT)/photodynamic therapy (PDT)/NO gas therapy by changing the wavelength of near-infrared light irradiation. L-Arginine (L-Arg) and indocyanine green (ICG) were encapsulated within mesoporous polydopamine layers coated on upconversion nanoparticles (UCNPs) to obtain the final nanocomposite UCNPs@mPDA/ICG@L-Arg (named UPIL). Under 980 nm irradiation, UPIL exhibits excellent performance in upconversion luminescence imaging. Meanwhile, upon 808 nm irradiation, UPIL not only produces thermal energy for PTT but also directly generates reactive oxygen species for effective PDT, achieving a synergistic effect of PTT/PDT. Furthermore, ICG in UPIL can serve as an oxidant, reacting with L-Arg to produce NO for gas therapy, which can also improve the PDT effect through the generated NO gas. Notably, the simple and effective switching between imaging (excited at 980 nm) and therapy (irradiated at 808 nm) can be achieved by altering the exciting wavelength of the nanocomposite. This orthogonal mode minimizes damage to normal tissue cells. Hence, this versatile nanocomposite platform, designed for near-infrared light-driven upconversion luminescence imaging and tri-modal synergistic therapy, holds the potential for imaging-mediated synergistic therapy, showcasing promising prospects for tumor treatment.

Targeted Theranostic Nanoprobes Assisted In Vivo NIR-II Fluorescence Imaging-Guided Surgery Therapy for Alveolar Echinococcosis

Chi Zhang,Li Jun,Yuxiang Gao,Le Guo,Jiayi Ding,Lijun Zhu,Zhong Du,Jiabao Xiong,Xueliang Zhang,Wenbao Zhang,Biao Dong,Nuernisha Alifu

Alveolar echinococcosis (AE) is a serious parasitic infectious disease that is highly invasive and destructive to the liver and has a high mortality rate. However, currently, there is no effective targeted imaging and treatment method for the precise detection and therapy of AE. We proposed a new two-step targeting strategy (TSTS) for AE based on poly(lactic-co-glycolic acid) (PLGA). We designed and constructed a novel type of ICG@PLGA@Lips nanoprobe with integrated imaging and treatment properties. First, we used the characteristics of PLGA gluconeogenic raw material to target the liver during blood circulation. Then, we utilized the characteristics of PLGA specifically penetrating the AE shell to achieve specific identification of AE in the liver. Under 808 nm laser excitation, ICG@PLGA@Lips effectively achieved accurate imaging of AE based on near-infrared II (NIR-II, 900–1700 nm) fluorescence imaging methods and achieved AE treatment through PDT effects. PLGA improved the optical properties of ICG, while liposomes further improved the biocompatibility of the nanoprobe. As ICG@PLGA@Lips showed strong NIR-II fluorescence emission and good biocompatibility, ICG@PLGA@Lips showed advantages in the specific fluorescence navigation of AE surgical resection lesions. Thus, with the assistance of ICG@PLGA@Lips, we achieved precise targeted and real-time NIR-II fluorescence imaging of AE for the first time. We successfully obtained in vivo NIR-II fluorescence imaging-guided photodynamic/surgical therapy of AE. This TSTS-based AE imaging and treatment exploration provided a new strategy for accurate imaging and treatment of early AE, which is expected to significantly improve the prognosis of patients.

Reprogrammable soft actuators based on a photochromic organic–inorganic hybrid membrane with modulatable NIR photothermal conversion

Xingchen Cui,Zhezhi Li,Zhihui Yi,Bing Zhang,Xing Gao,Fuqiang Fan,Yu Fu,Junbiao Wu,Tieqiang Wang

Shape-transformation soft actuators that can quickly respond to external stimuli have been potential candidates in biomimetic soft robots and flexible driving devices. However, the lack of shape reprogrammability restricts their complexity of deformation as well as reusability in practical application. Herein, we present a unique reprogrammable actuator based on asymmetric bilayer compounding with UV-modulated photochromic complex (NEU20) through a sequential polymerization process. The UV-induced electron transfer photochromic behavior synergistic with NIR photothermal effect of NEU20 fillers endows the soft actuator with rapid (∼90°/s) and customizable actuation under external NIR stimuli. Through the joint modulation of UV exposure region and duration, the UV-induced photothermal conversion efficiency is also adjustable at each local, facilitating the spatio-temporally programmable actuation process of the hybrid bilayer actuator. Significantly, the programmed actuation process of the actuator also features thermal-erasing and UV-reprogramming capacities, highlighting the unique recyclability of the hybrid actuator. In addition, the proposed reprogrammable actuator also demonstrates good universality in combination with other thermal-responsive active material systems. This work offers insight into the design and fabrication of reprogrammable soft actuators, shining enlightenment for potential application in bioinspired soft robots and smart mechanical devices.

Sialic Acid-Modified NIR-II Fluorophore with Enhanced Brightness and Photoconversion Capability for Targeted Lymphoma Phototheranostics

Fang Nan,Zhongxin Zhou,Qian Bai,Kai Chen,Yu Liu,Shuqi Wu

Lymphoma is a malignant cancer characterized by a rapidly increasing incidence, complex etiology, and lack of obvious early symptoms. Efficient theranostics of lymphoma is of great significance in improving patient outcomes, empowering informed decision-making, and driving medical innovation. Herein, we developed a multifunctional nanoplatform for precise optical imaging and therapy of lymphoma based on a new photosensitizer (1-oxo-1H-benzoo[de]anthracene-2,3-dicarbonitrile-triphenylamine (OBADC-TPA)). OBADC-TPA is a donor–acceptor (D–A) molecule characterized by a novel small coplanar and strong electron-withdrawing acceptor skeleton, while the OBADC moiety facilitates strong intramolecular charge transfer. OBADC-TPA-based nanoparticles (NPs) were prepared through encapsulation with an amphiphilic polymer and subsequent modification with sialic acid (SA). Both in vitro and in vivo studies demonstrated that NPs-SA possessed good biocompatibility, effective tumor accumulation, high photoacoustic (PA) contrast, bright second near-infrared (NIR-II) fluorescence emission, and efficient photothermal/photodynamic conversion capabilities, which can serve as a multifunctional nanocomposite for targeted PA/NIR-II fluorescence imaging-guided synergistic type I/II photodynamic and photothermal therapy (PDT/PTT) of lymphoma. This work not only provides a new NIR-II fluorophore with a novel acceptor moiety but also offers a new, accurate, and effective approach for the targeted diagnosis and treatment of lymphoma, holding promising prospects for clinical application.

UV- and X-ray-activated broadband NIR garnet-type Ca3Ga2Sn3O12:Fe3+ phosphors with efficient persistent luminescence

Yongjie Wang,Qingzhou Bu,Dongshuo Li,Shuai Yang,Li Li,Guotao Xiang,Sha Jiang,Ying Chang,Chuan Jing,Xianju Zhou,Lev-Ivan Bulyk,Andrzej Suchocki

Near-infrared phosphor-converted light-emitting diodes (NIR pc-LEDs) are compact light sources of great interest for NIR spectroscopy applications. Beyond typical Cr 3+ -activated NIR-emitting phosphors, there exists a strong demand for Cr 3+ -free alternatives with high efficiency and broadband emission to rich the landscape of NIR luminescent materials and extend their range of application fields. Here, we report a series of Fe 3+ -activated Ca 3 Ga 2 Sn 3 O 12 garnet-type phosphors exhibiting broadband NIR emission in the 650–1000 nm range attributed to 4 T 1 (G) → 6 A 1 (S) transition, with a maximum at 754 nm and a FWHM of 89 nm upon UV excitation. The spectroscopic results were analyzed according to the Tanabe-Sugano theory from which the crystal field parameter D q and Racah parameters B and C were obtained for the octahedrally coordinated Fe 3+ ion. Notably, the NIR persistent luminescence lasting over 1 h was detected following UV or X-ray irradiation. The possible mechanism involving electron traps was proposed to explain the observed persistent luminescence. Furthermore, a NIR pc-LED was fabricated by coating synthesized phosphor on a UV chip, and its performance was evaluated to assess its potential suitability as a NIR light source. Our discovery of novel type of nontoxic Fe 3+ -activated broadband NIR luminescence phosphors with efficient NIR persistent luminescence paves the way for discovering Cr 3+ -free multifunctional NIR luminescence materials, thereby expanding their application possibilities.

Ultrasensitive NIR-II Surface-Enhanced Resonance Raman Scattering Nanoprobes with Nonlinear Photothermal Effect for Optimized Phototheranostics

Laicui Song,Jin Li

Surface-enhanced resonance Raman scattering (SERRS) in the second near-infrared (NIR-II) window has great potential for improved phototheranostics, but lacks nonfluorescent, resonant and high-affinity Raman dyes. Herein, it is designed and synthesize a multi-sulfur Raman reporter, NF1064, whose maximum absorption of 1064 nm rigidly resonates with NIR-II excitation laser while possessing absolutely nonfluorescent backgrounds. Ultrafast spectroscopy suggests that the fluorescence quenching mechanism of NF1064 originates from twisted intramolecular charge transfer (TICT) in the excited state. Gold nanorods (AuNRs) decorated with such nonfluorescent NF1064 (AuNR@NF1064) show remarkable SERRS performances, including zero-fluorescence background, femtomolar-level sensitivity as well as superb photostability without fluorescence photobleaching. More importantly, AuNR@NF1064 exhibits a nonlinear photothermal effect upon plasmonic fields of AuNRs by amplifying the non-radiative decay of nonfluorescent NF1064, thus achieving a high photothermal conversion of 68.5% in NIR-II window with potential for further augmentation. With remarkable SERRS and photothermal properties, the NIR-II nanoprobes allow for high-precision intraoperative guided tumor resection within 8 min, and high-efficient hyperthermia combating of drug-resistant bacterial infection within living mouse body. This work not only unlocks the potential of nonfluorescent resonant dyes for NIR-II Raman imaging, but also opens up a new method for boosting photothermal conversion efficiency of nanomaterials.

Breaking shackles of molecular weight and emission for NIR-II fluorophores by regulating Columb attraction interaction

Ye Miantai,Wang Xiaoyu,Zou Jingwen,Sun Wei,Chi Weijie,Mao Zhiqiang,Liu Zhihong

The second near-infrared (NIR-II) dyes provide advantages for in vivo imaging, but challenges persist. A primary issue is the lack of practicable strategies to balance emission wavelength and molecular weight, particularly for low-molecular-weight (<500 Da) NIR-II (λ em  > 1000 nm) dyes. Here, we propose a strategy that tunes NIR-II emissions by reducing Coulomb attraction interaction, contrasting with traditional approaches that redshift absorption wavelengths through energy gap reduction. Leveraging this concept, we extend the emission wavelength of GFP chromophore-based dyes LS1-12 from the visible range into the NIR-II region, achieving a maximum emission wavelength exceeding 1200 nm with molecular weights between 226 and 449 Da. Further, the optimized NIR-II dye LS7 selectively binds Aβ 42 fibrils, yielding a 22.7-fold fluorescence increase in vitro and enabling real-time imaging of deposited Aβ proteins in the brains of living mice with Alzheimer’s disease. This study introduces a distinct design strategy for low-molecular-weight NIR-II dyes and addresses a longstanding bottleneck in this field.
