Discovery of the High-Affinity Aptamer for Candidalysin Using a Dual-Mode Colorimetric–SERS Platform

Yige Sun,Canlan Zheng,Yuxuan Shi,Mingyuan Sun,Chao Wang,Lin Han,Yu Zhang,Tiezhou Hou,Le Qiang

Candida albicansposes significant health risks through its virulent peptide toxin Candidalysin. As no existing therapeutics specifically target this toxin, developing high-affinity aptamers for its efficient and safe removal is urgently needed. In response, we developed a dual-mode biosensor based on gold nanoparticles (AuNPs) and aptamers for screening high-affinity aptamers for Candidalysin. This biosensor leverages the localized surface plasmon resonance (LSPR) phenomenon and surface-enhanced Raman scattering (SERS) of AuNPs to detect changes in color and Raman signals, respectively, indicative of high-affinity aptamer for Candidalysin presence. This dual-mode capability reduces false-negative signals and enhances detection accuracy. Our findings reveal a specific aptamer with high affinity for Candidalysin, presenting a significant advancement in candidiasis treatment. This work sets the stage for the development of effective therapeutic strategies against Candida infections.

Tautomers of 2-aminothiazole molecules in aqueous solutions explored by Raman, SERS and DFT methods

Xue Chen,Yongjun Hu,Jiao Gao

2-Aminothiazole, as a typical heterocyclic amine, may exist amino-imino tautomeric equilibrium in nature. In present report, the optimized structural parameters and vibrational wavenumbers of amino and imino tautomers of the molecule have been predicted by density functional theoretical (DFT) calculation. Comparing the experimental and the calculated Raman spectra, it indicates that the amino tautomer would be the main configuration in the 2-aminothiazole saturated solution. In the concentration-dependent SERS spectra of 2-aminothiazole adsorbed on gold nanoparticles , it is found that the tautomeric equilibrium shifts toward to the amino tautomer when the concentrations is decreased. While the amino tautomer is end-on the surface through the nitrogen atom of thiazole ring at the higher concentrations, the molecule becomes to stand on the metal surface by interacting simultaneously with two nitrogen atoms of the molecule in the diluted solution.

Highly sensitive SERS sensing of metabolic modulators by COF@Ag nanocomposite for food safety monitoring

Yueyue Ma,Yu Zeng,Ruxi Lin,Liping Chen,Mingzhu Shen,Xiaojun Luo,Bing Shao

Letrozole, anastrozole and trimetazidine are all included in the prohibited list of the World Anti-Doping Agency (WADA). At present, there is still a lack of a systematic Surface-Enhanced Raman Spectroscopy (SERS) spectral database for these drugs, and traditional detection methods suffer from limitations such as insufficient sensitivity and cumbersome operation procedures. In this study, a novel COF@Ag composite was utilized as a SERS-enhanced substrate in combination with a portable Raman spectrometer, allowing for accurate identification and effective differentiation of the characteristic peaks of the three prohibited drugs. Moreover, this method exhibited excellent performance in detecting actual milk matrix samples, with the spiked recovery rate ranging from 98.9% to 101.1%, thereby directly meeting the practical demands of on-site rapid screening. This work offers a reliable and convenient strategy for both sports food safety monitoring and pharmaceutical analysis.

Machine learning-assisted SERS detection of pyrethroid pesticides in edible fungi using a magnetic nanosensor

Liu Bo,Han Qianxi,Wang Wenxuan,He Lijing,Wang Wenjie,Cheng Yanfen,Yun Shaojun,Cheng Feier,Feng Cuiping,Cao Jinling

Pyrethroid pesticide residues pose a significant global public health challenge, particularly in complex edible fungus matrices where trace, structurally similar pesticides are difficult to distinguish and detect. To address this critical gap, a novel surface-enhanced Raman spectroscopy (SERS) platform is presented integrating “magnetic enrichment, covalent organic framework (COF)-mediated capture, silver shell enhancement, and machine learning (ML)-driven intelligent recognition”. The core innovation lies in the tailored Fe₃O₄@COF@Ag nanocomposite architecture where the magnetic Fe₃O₄ core enables rapid sample separation, the COF intermediate layer provides specific molecular capture, and the Ag shell generates dense SERS “hotspots”, synergistically enhanced by ML algorithms to enable precise discrimination of structurally analogous pesticides. The developed Fe₃O₄@COF@Ag substrate demonstrated exceptional performance, achieving detection limits as low as 5.21 µg/kg for deltamethrin (CF), 6.08 µg/kg for fenvalerate (FV), and 11.6 µg/kg for lambda-cyhalothrin (LCF). The method exhibited outstanding linearity (R² > 0.99) for quantitative analysis and anti-interference capability, with recoveries of 94.36-106.89% in real samples. By integrating ML algorithms, the platform achieved 100% classification accuracy for distinguishing structurally similar pyrethroids. Principal component analysis (PCA) and support vector machine (SVM) models effectively extracted discriminative spectral features, enabling precise identification even at trace concentrations. This work provides a rapid, sensitive, and field-deployable solution for pesticide monitoring, offering significant potential to enhance food safety and public health protection. Graphical

Silver-nanoparticles/graphene hybrids for effective enrichment and sensitive SERS detection of polycyclic aromatic hydrocarbons

Xiujuan Wang,Qiaoling Xu,Xiaoye Hu,Fangming Han,Chuhong Zhu

Polycyclic aromatic hydrocarbons (PAHs) are one of the most widespread and dangerous group of pollutants existing in the environment. Trace detection of PAHs is essential and important. Surface-enhanced Raman scattering (SERS) is a powerful analytical tool for ultrasensitive chemical analysis. However, the direct detection of PAHs by SERS is difficult due to poor affinity of PAHs to metal surfaces . In this work, we present a SERS platform based on the Ag-nanoparticles/graphene hybrid for the direct detection of PAHs with graphene as PAHs assemblies. The target PAHs are captured by the graphene through π-π electronic stacking, and brought close to the hot spots generated by dense Ag-nanoparticles decorated on the graphene. Sensitive detection of PAHs has been realized using this SERS substrate without further surface modification. The limit of detection for the three typical PAHs including pyrene, anthracene and phenanthrene was as low as 0.73 ppb, 1.1 ppb and 0.57 ppb, respectively. Our results indicate that the immobilization of PAHs on graphene is a process that can be applied in the design of sensitive sensors for these aromatic pollutants. This functional SERS sensor shows a great potential application in food safety inspection and environment pollutants monitoring .

Nanostructured gold microelectrodes for SERS and EIS measurements by incorporating ZnO nanorod growth with electroplating

Zong Xianli,Zhu Rong,Guo Xiaoliang

In this paper, a fine gold nanostructure synthesized on selective planar microelectrodes in micro-chip is realized by using an advanced hybrid fabrication approach incorporating growth of nanorods (NRs) with gold electroplating. By this developed nanostructure, integration of in-situ surface-enhanced Raman spectroscopy (SERS) detection with electrochemical impedance spectroscopy (EIS) measurement for label-free, nondestructive, real-time and rapid monitoring on a single cell has been achieved. Moreover, parameters of Au nanostructures such as size of nanoholes/nanogaps can be controllably adjusted in the fabrication. We have demonstrated a SERS enhancement factor of up to ~2.24 × 10 6 and double-layer impedance decrease ratio of 90% ~ 95% at low frequency range below 200 kHz by using nanostructured microelectrodes. SERS detection and in-situ EIS measurement of a trapped single cell by using planar microelectrodes are realized to demonstrate the compatibility, multi-functions, high-sensitivity and simplicity of the micro-chip system. This dual function platform integrating SERS and EIS is of great significance in biological, biochemical and biomedical applications.

Depositing a flexible substrate of triangular silver nanoplates onto cotton fabrics for sensitive SERS detection

Deshan Cheng,Mantang He,Jianhua Ran,Guangming Cai,Jihong Wu,Xin Wang

A flexible and effective substrate for surface-enhanced Raman spectroscopy (SERS) detection has been developed by dip coating of triangular silver nanoplates (TSNPs) onto cotton fabrics. The morphology, chemical composition and crystalline structure of the coated cotton fabrics were characterized by scanning electron microscope (SEM), ultraviolet-visible spectroscopy (UV-vis), X-ray photoelectron spectroscopic (XPS) and X-ray diffraction (XRD), respectively. These results confirmed the presence and well dispersion of the TSNPs on the surface of cotton fabrics. p -aminothiophenol (PATP) has been used as the probe molecule to evaluate the performance of the coated cotton fabrics as a flexible SERS substrate, and the PATP in diluted solutions can be detected with the concentration as low as 10 −8  M. The results indicate that the TSNPs-coated fabrics exhibit sensitive SERS signals with excellent reproducibility and stability. The flexible SERS substrate has been demonstrated to rapidly detect carbaryl pesticides on fruits.

3D RGO@MoS2/Ag NPs based SERS platform for label-free detection of IL-6

Qinqin Qian,Haocheng He,Songyin Fu,Jia Xu,Ho-Pui Ho,Shuwen Zeng,Tao Jiang,Sheqin Lv,Chenjie Gu

The efficient and sensitive monitoring of the interleukin-6 (IL-6) is crucial for the clinical tracking and diagnosis of chronic obstructive pulmonary disease (COPD). Herein, a novel surface-enhanced Raman scattering substrate (RGO@MoS 2 /Ag NPs), comprising an efficient charge-transfer and adsorption layer of reduced graphene oxide (GO) encapsulated in molybdenum disulfide (MoS 2 ) and an electromagnetic enhancement layer of Ag nanoparticles, was developed for the highly sensitive, label-free detection of IL-6. Experimentally, a precursor film consisting of (NH 4 ) 2 MoS 4 and GO was first deposited onto a rough g -C 3 N 4 substrate via spin-coating process. Subsequently, the above precursor film was subjected to a two-step high-temperature annealing process for preparing the RGO@MoS 2 charge transfer layer. Finally, Ag NPs were sputtered onto the RGO@MoS 2 film to induce the localized surface plasmons for the further enhancement of the SERS performance. Leveraging the high specific surface area and exceptional charge transfer properties of RGO@MoS 2 , together with the high-density “hot spots” generated by surface Ag NPs, the fabricated SERS substrate exhibited remarkable electromagnetic and chemical enhancement effects. By using the rhodamine 6G (R6G) as the Raman reporter, it was determined that the lowest detection limit could be as low as 10 −9  M on the optimal SERS substrate, with an enhancement factor of 2.2 × 10 9 . Meanwhile, the substrate also exhibited good uniformity, with a relative standard deviation of 9.84%. Finally, the prepared SERS substrate was used to detect the IL-6, it showed good linear response in the range of 5.0-1000 pg/mL, with a detection limit as low as 0.41 pg/mL.

SERS Chemical Enhancement Mechanism of Phenthoate on Gold Nanoparticles Revealed by DFT and TD-DFT

Qi Wang,Chao Song,Zhuang Bo,Haotong Yang,Xun Gao,Jingquan Lin

This study employed density functional theory (DFT) and time-dependent density functional theory (TD-DFT) to investigate the interaction mechanism between Phenthoate and Au20 nanoclusters, and the solvent environment effect was examined under the polarizable continuum model (PCM). By comparing and analyzing theoretical Raman active spectra and experimental Raman spectra, the phenomenon of selective enhancement of specific vibrational modes in surface-enhanced Raman spectroscopy (SERS) was explained. By analyzing the molecular structure, frontier molecular orbitals (FMOs), ultraviolet–visible absorption spectra (UV–vis), noncovalent interactions (NCI), reduced density gradients (RDG), electron localization function (ELF), and localized orbital locator (LOL), the adsorption mechanism and the spectral characteristics induced by interfacial charge transfer were comprehensively revealed at the electronic level. Phenthoate chemically adsorbed onto the Au20 surface through the S atom, forming a stable complex. Meanwhile, this charge transfer mechanism effectively modulated the polarization rate of the molecule. This study provides a theoretical basis for elucidating the adsorption behavior and spectral response mechanism of organophosphorus pesticides on metal nanostructures, offering valuable insights for the design of highly sensitive SERS sensors.

Regulating Rare-Earth Single-Atom Nanozyme for Colorimetric-SERS Dual-Modal Sensing of Intracellular Glutathione

Huifang Zhao,Yi Chen,Lili Liu,Fujuan Peng,Bowei Guan,Jinze Li,Qian Wang,Luliang Liao,Baozhu Chi,Ruiping Zhang

Rare-earth single-atom nanozymes (RESAzymes) have emerged as promising catalysts due to their maximized atom utilization efficiency and tunable enzymatic activities. Herein, a colorimetric-Surface-enhanced Raman scattering (SERS) dual-modal sensing platform is constructed to introduce 4f electron configurations of single-atom ytterbium active site decorated on a nitrogen-doped porous carbon (Yb-N/C) by a facile hierarchical cascade-anchoring protocol. Specifically, this unique coordination environment can promote substrate molecule adsorption and activation energy barriers, possessing excellent peroxidase (POD)-mimicking activity and SERS enhancement performance in TMB-H 2 O 2 system. In the presence of glutathione (GSH), Yb-N/C catalyst is inhibited to result in reducing oxTMB for returning its colorless state, consequently diminishing both the colorimetric readout and the SERS intensity. Interestingly, this reaction mechanism from Yb-N/C is systematically illustrated through the steady-state kinetic, electron paramagnetic resonance (EPR), and density functional theory (DFT) calculations. Notably, the limited of detection (LOD) of GSH with colorimetric and SERS dual-mode was as low as 0.30 and 0.11 µM, respectively. Our established dual-modal analysis for intracellular GSH exhibits the favorable consistent with the standard kit in HepG2, KYSE150, and MCF-7 cells, respectively. This work will offer an in-depth understanding of RESAzymes-mimicking activity of colorimetric-SERS biosensors by adjusting electronic structures in carbon-based carrier.

Fluid Domain-Limited Ag Nanoparticles on Janus Fabric for SERS-Based Label-Free Sweat Detection

Jiaji Hou,Fengyan Ge,Haixia Jiang,Weiguang Liu,Chaofan Li,Xin Yan

Flexible wearable biosensors enable dynamic tracking of biomarkers in sweat through noninvasive sampling. Surface-enhanced Raman spectroscopy (SERS) utilizes the “fingerprint” vibrations of molecular vibration spectra to simultaneously analyze multiple substances, facilitating the identification of complex analytes. In this study, we integrated a Janus fabric with efficient wearable moisture management properties with silver (Ag) nanoparticles to develop an SERS sensor for human sweat detection. The hydrophobic side of the Janus fabric was designed to contact the skin, while hydrophobic patterns were printed on the opposite side to enable the efficient enrichment of target molecules. Rapid sweat transport was achieved through a combination of a hydrophilic–hydrophobic interfacial design. The moisture management capability and SERS enhancement mechanism of the platform were further explained through simulation. Moreover, the platform demonstrated the successful detection of real human sweat samples. This work is expected not only to provide a novel approach for the fabrication and preparation of the SERS active fabric with excellent performance but also to promote the theoretical basis and technical support for the research of textile on rapid detection in highly diluted biochemical samples.

Graphene-Ag nanoparticles-cicada wings hybrid system for obvious SERS performance and DNA molecular detection

Wen Yang,Zhen Li,Zhengyi Lu,Jing Yu,Yanyan Huo,Baoyuan Man,Jie Pan,Haipeng Si,Shouzhen Jiang,Chao Zhang

In recent years, biomaterials have increasingly attracted attention on surface-enhanced Raman spectroscopy (SERS) due to their well Raman performance while metal particles are combined with biological substrates. Therefore, we propose an environmentally friendly substrate based on silver-plated cicada wings with seamless graphene layer (Gr-AgNPs-C.w.), which can be prepared with a simple and inexpensive method. Compared with AgNPs-C.w., Gr-AgNPs-C.w. hybrids show better SERS performance with high sensitivity, good uniformity and good stability with R6G detection. The minimum detected concentration can reach 10−15 M, and the value of R2 can reach 0.996, respectively. Theoretical simulation demonstrates the situation of electromagnetic field through COMSOL software. In addition, due to the affinity of graphene for biomolecules, we can successfully detect the DNA biomolecules through a simple process. Therefore, this cheap and efficient natural SERS substrate has great potential for a considerable number of biochemical SERS applications and can broaden the way in which multiple SERS platforms derived from other natural materials are prepared.
