New Type Annihilator of π-Expanded Diketopyrrolopyrrole for Robust Photostable NIR-Excitable Triplet–Triplet Annihilation Upconversion

Fang Qi,Hong-Juan Feng,Yi Peng,Lin-Han Jiang,Le Zeng,Ling Huang

Near-infrared light excitable triplet–triplet annihilation upconversion (NIR TTA-UC) materials have attracted interest in a variety of emerging applications such as photoredox catalysis, optogenetics, and stereoscopic 3D printing. Currently, the practical application of NIR TTA-UC materials requires substantial improvement in photostability. Here, we found that the new annihilator of π-expanded diketopyrrolopyrrole (π-DPP) cannot activate oxygen to generate superoxide anion via photoinduced electron transfer, and its electron-deficient characteristics prevent the singlet oxygen-mediated [2 + 2] cycloaddition reaction; thus, π-DPP exhibited superior resistance to photobleaching. In conjunction with the NIR photosensitizer PdTNP, the upconversion efficiency of π-DPP is as high as 8.9%, which is eight times of the previously reported PdPc/Furan-DPP. Importantly, after polystyrene film encapsulation, less than 10% photobleaching was observed for this PdTNP/π-DPP-based NIR TTA-UC material after four hours of intensive NIR light exposure. These findings provide a type of annihilator with extraordinary photostability, facilitating the development of NIR TTA-UC materials for practical photonics.

NIR triggered polydopamine coated cerium dioxide nanozyme for ameliorating acute lung injury via enhanced ROS scavenging

Yin Mingjing,Lei Doudou,Liu Yalan,Qin Tao,Gao Huyang,Lv Wenquan,Liu Qianyue,Qin Lian,Jin Weiqian,Chen Yin,Liang Hao,Wang Bailei,Gao Ming,Zhang Jianfeng,Lu Junyu

Acute lung injury (ALI) is a life threatening disease in critically ill patients, and characterized by excessive reactive oxygen species (ROS) and inflammatory factors levels in the lung. Multiple evidences suggest that nanozyme with diversified catalytic capabilities plays a vital role in this fatal lung injury. At present, we developed a novel class of polydopamine (PDA) coated cerium dioxide (CeO 2 ) nanozyme (Ce@P) that acts as the potent ROS scavenger for scavenging intracellular ROS and suppressing inflammatory responses against ALI. Herein, we aimed to identify that Ce@P combining with NIR irradiation could further strengthen its ROS scavenging capacity. Specifically, NIR triggered Ce@P exhibited the most potent antioxidant and anti-inflammatory behaviors in lipopolysaccharide (LPS) induced macrophages through decreasing the intracellular ROS levels, down-regulating the levels of TNF-α, IL-1β and IL-6, up-regulating the level of antioxidant cytokine (SOD-2), inducing M2 directional polarization (CD206 up-regulation), and increasing the expression level of HSP70. Besides, we performed intravenous (IV) injection of Ce@P in LPS induced ALI rat model, and found that it significantly accumulated in the lung tissue for 6 h after injection. It was also observed that Ce@P + NIR presented the superior behaviors of decreasing lung inflammation, alleviating diffuse alveolar damage, as well as promoting lung tissue repair. All in all, it has developed the strategy of using Ce@P combining with NIR irradiation for the synergistic enhanced treatment of ALI, which can serve as a promising therapeutic strategy for the clinical treatment of ROS derived diseases as well.

NIR-Actuated Botanicals/Nanozymes Nanofibrous Membranes for Antibiotic-Free Triple-Synergistic Therapy Against Polymicrobial Wound Infections

Kai Dong,Wenbo Lin,Tianyu Zhu,Shuwen Sun,Chenhao Zhang,Chen Xu,Fangfang Cao,Xiaoyuan Chen

Polymicrobial wound infection represents a significant contributor to infectious disease-related morbidity and mortality globally. Traditionally, the management of such infection relies heavily on high doses of antibiotics, often resulting in systemic side effects and exacerbating antibiotic resistance. Consequently, there is an urgent need to develop a safe and effective platform for antibiotic-free treatment of polymicrobial wound infections. In this study, a near-infrared (NIR) light-actuated nanofibrous membrane incorporating botanicals and nanozymes is presented for antibiotic-free triple-synergistic therapy against polymicrobial wound infections. The membrane integrates berberine hydrochloride (BBH)-loaded hollow zinc-doped carbon nanocubes into polyvinyl alcohol (PVA) nanofibrous membranes. Upon low-intensity 808 nm NIR laser irradiation (0.3 W cm −2 ), the membrane achieved a controlled release of BBH and enhanced peroxidase-like (POD-like) enzyme activity. This NIR light-actuated botanical/photothermal therapy (PTT)/chemodynamic therapy (CDT) system demonstrated high efficacy in eradicating methicillin-resistant Staphylococcus aureus and Pseudomonas aeruginosa both in vitro and in vivo. Furthermore, it mitigated inflammation and promoted wound healing in polymicrobial infection wounds, highlighting its potential as a promising alternative to antibiotic-based therapies.

NIR-II luminescence and temperature sensing properties based on double perovskite La2MgTiO6: Yb3+/Er3+/Ho3+ phosphor

Xiaoyu Wu,Yang Yu,Kejie Li,Mingqin Huang,Zuoling Fu

The near-infrared-II (NIR-II) fluorescence thermometer based on rare earth-doped double perovskite is rarely. Herein, Er 3+ -Ho 3+ and Yb 3+ co-doped lead-free double perovskite La 2 MgTiO 6 (LMTO) phosphors were synthesized by a conventional high temperature solid phase method. The phase purity and morphology of the samples were analyzed using XRD and FE-SEM. The NIR-II luminescence properties of LMTO: Er 3+ -Ho 3+ -Yb 3+ phosphors were systematically investigated under laser irradiation at 980 nm. A NIR-II fluorescence thermometer was also designed based on non-thermal coupled energy levels originated from the 4 I 13/2 energy level of Er 3+ and the 5 I 6 energy level of Ho 3+ . The maximum value of relative sensitivity (S r ) was 0.54 % at 513 K. Compared to visible thermometers (LMTO: Er 3+ -Yb 3+ and LMTO: Ho 3+ -Yb 3+ ), the NIR-II thermometers offer better temperature measurement capability and penetration depth. All the experimental results show that the NIR-II thermometer with a wider range and higher sensitivity has been realized, which puts forward a new idea for the design and optimization of NIR-II thermometers.

NIR-II light-encoded 4D-printed magnetic shape memory composite for real-time reprogrammable soft actuator

Shushu Miao,Yue Xing,Xu Li,Bing Sun,Zheyuan Du,Hongshuo Cao,Pengfei Guo,Yincheng Chang,Yanhong Tian,Minghui Yao,Ke Chen,Dengbao Xiao,Xuejun Zhang,Biao Zhao,Kai Pan,Jiangman Sun,Xiubing Liang

For the intelligent 4D-printed actuators, the excellent performance, including quickly reversible spatial-shape transformation and locking, digital and precise shape manipulation in real-time, and remote actuation in special spaces or harsh environments, is significantly desirable but still challenging. Here, using a UV-curable system containing the shape memory polymer (SMP) and NdFeB particles, namely the magSMP composite, we fabricate a real-time reprogrammable soft actuator via high-resolution Digital Light Processing (DLP)-based 4D printing. The printed structure is composed of an array of physical binary magSMP composite elements (m-bits), analogous to digital bits. Owing to the NdFeB's photothermal effect, each m-bit can be independently and reversibly switched between unlocking or locking states (allowing or prohibiting responsive shape-morphing) in response to the on/off state of NIR-II light. Through projecting NIR-II light patterns for encoding a set of binary instructions onto 4D-printed actuators, the real-time light-programmed deformations are induced precisely under an actuation magnetic field due to the NdFeB's huge coercivity. Thus, the synergistic magnetic and light field-manipulated multimodal deformations of actuators, including mimosa shape changing, grasping, and wire guiding, are achieved. This study shines lights on the fabrication of soft structures of arbitrary sizes and provides their future perspectives in soft robot design.

Molecular Etching-Derived High-Brightness NIR-II Gold Nanoclusters for High-Resolution Bioimaging and Photothermal Therapy

Fangzheng Tian,Siyu Li,Xue Yang,Kai Fan,Mei Zhou,Duyang Gao,Dehong Hu,Shenghong Ju

Glutathione-capped gold nanoclusters (GSH-Au NCs) with second near-infrared (NIR-II, 1000–1700 nm) fluorescence have received increasing interest in biomedical applications. However, conventional aqueous synthesis of GSH-Au NCs results in low NIR-II absolute fluorescence quantum yield (≈0.27%) and limited renal clearance efficiency (≈70%), which restricts its clinical applications. This study reports a GSH-etching post-synthesis method to selectively convert nonfluorescent GSH-Au n NCs ( n > 25) into fluorescent GSH-Au 25 NCs (eGSH-Au NCs). This molecular etching process decreases the size of the NCs from 3.3 to 2.0 nm and afforded 37% increase in NIR-II absolute fluorescence quantum yield and 11.9% enhancement in renal clearance efficiency. The optimized eGSH-Au NCs facilitates whole cerebrovascular imaging through the scalp and skull, tumor vasculature imaging with a resolution of 40.8 µm, dynamic tumor perfusion imaging with a signal-to-background ratio of 10.5, and enhanced lymphatic vessel and lymph node imaging. Additionally, eGSH-Au NCs exhibit superior photothermal properties, achieving almost 100% tumor growth inhibition in Hepa1-6 tumor-bearing mice. This study presents a novel strategy to improve the bioimaging and therapeutic performance of NIR-II Au NCs, thus advancing their clinical translation.

Organic–Inorganic Hybrid One-Dimensional Photonic Crystals with Blue and NIR Dual Bandgaps for Light Protection

Minglu Zhang,Qi Yuan,Yanyan Guo,Saijin Song,Dongyu Wang,Junli Zhang,Hao-Yang Mi,Jian Han,Chuntai Liu,Changyu Shen

Planar 1D photonic crystals (1DPhCs), owing to their photonic bandgaps (PBGs) formed by unique structural interference, are widely utilized in light protection applications. Multifunctional coatings that integrate various light management functions are highly desired. In this study, we present the fabrication of dual-PBG 1DPhCs with high reflectance in both the blue and near-infrared (NIR) regions. The 1DPhCs were constructed through the alternating stacking of hollow SiO2 (HS) nanoparticles hybridized with poly(vinyl alcohol) (PVA), referred to as PHS, and TiO2 layers. The 1DPhC with 7 bilayers demonstrated remarkable reflectance of 98.2% and 93.4% in the blue and NIR spectra, respectively, with shielding efficiencies of 93.9% and 85.6%. Compared to bare PC, the temperature was observed to be 10.6 °C lower, achieving a thermal insulation efficiency of 44.8% in the 900 nm near-infrared range. Additionally, spectral fitting using the transfer matrix method (TMM) and optimization of process parameters, including PHS concentration and spin-coating speed, enabled the fabrication of 1DPhCs with tunable optical thickness ratios. The fabricated films demonstrated a smooth surface morphology with flat and uniform interfaces, as well as excellent environmental stability. These features make the 1DPhCs highly promising for applications in light protection and thermal management. This study provides a novel approach for the development of dual-PBG 1DPhCs with enhanced protective capabilities.

A hematoporphyrin and indocyanine green co-delivery system with NIR triggered-controllable photoactivities for photodynamic therapy

Ziying Li,Tingting Lv,Yingying Zhang,Liang Xu,Lu Zhang,Xiuying Wang,Haijun Chen,Yu Gao

Photodynamic therapy (PDT) was hindered by the poor water solubility and long-enduring photoactivities of photosensitizers . Here, we developed a novel controllable photodynamic agent CPHI based on β-cyclodextrin modified polyamidoamine (PAMAM) dendrimer (CP) co-delivery of indocyanine green (ICG) and hematoporphyrin (HP). The CPHI possessed appropriate particle sizes and zeta potentials , considerable photostability, and NIR-triggered release property. The fluorescence of HP could be quenched by ICG when HP and ICG were co-loaded inside the CP, and the quenching efficiency was related with the weight ratio of HP and ICG. When the feeding weight proportion of HP and ICG is 1/10, the fluorescence of HP was quenched by 87.3% in CPHI. Under 808 nm laser irradiation , the ICG in CP was degraded, and the fluorescence of HP could recover. The recovered HP was capable of generating reactive oxygen species (ROS) for PDT with 606 nm laser irradiation. The cytotoxicity and cell apoptosis studies showed that CPHI could effectively induce MCF-7 cells apoptosis with PDT effects under 808/660 nm laser. These results suggested that CPHI could be an efficient cancer therapeutic agent for controllable PDT with low dark toxicity.

Acidity-Triggered “Sticky Spotlight”: CCK2R-Targeted TME-Sensitive NIR Fluorescent Probes for Tumor Imaging In Vivo

Ruiqi Sun,Yuxin Wang,Wenhui Shi,Hongfu Zhang,Jianhua Liu,Weina He

Cancer which causes high mortality globally threatens public health seriously. There is an urgent need to develop tumor-specific near-infrared (NIR) imaging agents to achieve precise diagnosis and guide effective treatment. In recent years, imaging probes that respond to acidic environments such as endosomes, lysosomes, or acidic tumor microenvironments (TMEs) are being developed. However, because of their nonspecific internalization by both normal and tumor cells, resulting in a poor signal-to-noise ratio in diagnosis, these pH-sensitive probes fail to be applied to in vivo tumor imaging. To address this issue, a cholecystokinin-2 receptor (CCK2R)-targeted TME-sensitive NIR fluorescent probe R2SM was synthesized by coupling pH-sensitive heptamethine cyanine with a CCK2R ligand, minigastrin analogue 11 (MG11) for in vivo imaging, in which MG11 would target overexpressed CCK2Rs in gastrointestinal stromal tumors (GISTs). Cell uptake assay demonstrated that R2SM exhibited a high affinity for CCK2R, leading to receptor-mediated internalization and making probes finally accumulated in the lysosomes of tumor cells, which suggested in the tumor tissues, the probes were distributed in the extracellular acidic TME and intracellular lysosomes. With a pKa of 6.83, R2SM can be activated at the acidic TME (pH = 6.5–6.8) and lysosomes (pH = 4.5–5.0), exhibiting an apparent pH-dependent behavior and generating more intense fluorescence in these acidic environments. In vivo imaging showed that coupling of MG11 with a pH-sensitive NIR probe facilitated the accumulation of probe and enhanced the fluorescence in CCK2R-overexpressed HT-29 tumor cells. A high signal was observed in the tumor region within 0.5 h postinjection, indicating its potential application in intraoperative imaging. Fluorescence imaging of R2SM exhibited higher tumor-to-liver and tumor-to-kidney ratios (2.1:1 and 2.3:1, respectively), compared separately with the probes that are lipophilic, pH-insensitive, or MG11-free. In vitro and in vivo studies demonstrated that the synergistic effect of tumor targeting with pH sensitivity plays a vital role in the high signal-to-noise ratio of the NIR imaging probe. Moreover, different kinds of tumor-targeting vectors could be conjugated simultaneously with the NIR dye, which would further improve the receptor affinity and targeting efficiency.

Biomimetic gold nano-modulator for deep-tumor NIR-II photothermal immunotherapy via gaseous microenvironment remodeling strategy

Honglin Huang,Zhengxin Xie,Ningxi Li,Li Zeng,Qianyi Zeng,Ziman Yang,Jinyang Shen,Hong Yang,Yiyao Liu,Chunhui Wu

Introduction Effective immunotherapeutic treatment of solid tumors has been greatly challenged by the complex hostile tumor immunosuppressive microenvironment (TIME), which typically involves hypoxia and immunosuppression. Methods Herein, a multifunctional biomimetic gold nano-modulator (denoted as GNR-SNO@MMT ) was developed to realize the efficient second near-infrared (NIR-II) photothermal immunotherapy via tumor targeting and deep penetration, vascular normalization and immune reprogramming. NIR-II photothermal agent gold nanorods (GNR) were grafted with thermosensitive S-nitrosothiol (SNO) donors and camouflaged with the tumor-penetrating peptide tLyp-1-modified macrophage membrane (MM) to yield GNR-SNO@MMT . Results The engineered membrane coating increased the capacity for tumor inflammatory tropism and deep penetration, which aided GNR-SNO@MMT in ablating tumors together with NIR-II laser irradiation. Moreover, hyperthermia-stimulated nitric oxide (NO) release in situ acted as a gas immunomodulator to effectively enhance blood perfusion and reprogram the TIME via multiple functions (e.g., decreasing PD-L1, repolarizing tumor-associated macrophages, and revitalizing cytotoxic T cells). Ultimately, the inhibition rate against 4T1 mouse mammary tumor model mediated by GNR-SNO@MMT plus NIR-II laser was 94.7% together with 2.4-fold CD8 + T cells infiltrated into tumors than that of the untreated counterpart. Conclusions The engineered biomimetic nano-modulator of GNR-SNO@MMT provides an effective and novel photoimmunotherapy candidate against deep-sited solid tumors through immune reconfiguration via NO-involved nanomedicine and external NIR-II laser assistance. Biomimetic NIR-II fluorescent proteins created from chemogenic protein-seeking dyes for multicolor deep-tissue bioimaging

Xu Jiajun,Zhu Ningning,Du Yijing,Han Tianyang,Zheng Xue,Li Jia,Zhu Shoujun

Near-infrared-I/II fluorescent proteins (NIR-I/II FPs) are crucial for in vivo imaging, yet the current NIR-I/II FPs face challenges including scarcity, the requirement for chromophore maturation, and limited emission wavelengths (typically < 800 nm). Here, we utilize synthetic protein-seeking NIR-II dyes as chromophores, which covalently bind to tag proteins (e.g., human serum albumin, HSA) through a site-specific nucleophilic substitution reaction, thereby creating proof-of-concept biomimetic NIR-II FPs. This chemogenic protein-seeking strategy can be accomplished under gentle physiological conditions without catalysis. Proteomics analysis identifies specific binding site (Cys 477 on DIII). NIR-II FPs significantly enhance chromophore brightness and photostability, while improving biocompatibility, allowing for high-performance NIR-II lymphography and angiography. This strategy is universal and applicable in creating a wide range of spectrally separated NIR-I/II FPs for real-time visualization of multiple biological events. Overall, this straightforward biomimetic approach holds the potential to transform fluorescent protein-based bioimaging and enables in-situ albumin targeting to create NIR-I/II FPs for deep-tissue imaging in live organisms.
