This study selected three approved folate sources—folic acid (FA), L-5-methyltetrahydrofolate (MTFA), and calcium 5-methyltetrahydrofolate (CMTFA)—to explore their interaction mechanisms with soy protein isolate (SPI) through spectrofluorometric analysis and molecular docking simulations. We investigated how these interactions influence the structural and physicochemical stability of folates and SPI. Three folates spontaneously bound to SPI, forming complexes, resulting in a decrease of approximately 30 kJ·mol−1in Gibbs free energy and an association constant (Ka) of 105L·mol−1. The thermodynamic parameters and molecular docking study revealed the unique binding mechanisms of FA and MTFA with SPI. FA’s planar pteridine ring and conjugated double bonds facilitate hydrophobic interactions, whereas MTFA’s reduced ring structure and additional polar groups strengthen hydrogen bonding. Although the formation of SPI–folate complexes did not result in substantial alterations to the SPI structure, their binding has the potential to enhance both the physical and thermal stability of the protein by stabilizing its conformation. Notably, compared with free FA, the FA-SPI complexes significantly enhanced FA’s stability, exhibiting 71.1 ± 1.2% stability under light conditions after 9 days and 63.2 ± 2.6% stability in the dark after 60 days. In contrast, no similar effect was observed for MTFA. This discrepancy can be ascribed to the distinct degradation pathways of the Fa and MTFA molecules. This study offers both theoretical and experimental insights into the development of folate-loaded delivery systems utilizing SPI as a matrix.

Folates, a crucial B-group vitamin, serve as a significant functional food supplement. Nevertheless, considerable obstacles persist in improving folates stability in liquid products. In this study, folic acid (FA) and 5-methyltetrahydrofolate (MTFA), two approved sources of folates, were encapsulated with sodium caseinate (NaCas) to enhance their stability. The protective effect of NaCas on folate molecules was investigated using experimental and computational methods. Meanwhile, the influence of divalent calcium ion (Ca2+) on the properties of the NaCas-MTFA complex was examined to evaluate the potential application of calcium 5-methyltetrahydrofolate (CaMTFA). Fluorescence tests showed both folates had static quenching behavior and bound to NaCas with a binding constant of 104–105M−1. Hydrophobic interactions were crucial in NaCas-FA complex formation, while hydrogen bonding drove NaCas-MTFA binding. The encapsulation of caseinate notably slowed down the degradation of folates under both light and dark conditions. Moreover, the addition of a low concentration of Ca2+did not adversely impact the binding mechanism of the NaCas-MTFA complex or the degradation curve of MTFA. The results of this study could serve as a valuable resource for the utilization of caseinates in incorporating folates, specifically MTFA, in the creation of natural liquid dietary supplements.

Background Hepatocellular carcinoma (HCC) is the most common primary liver cancer and severely threatens human health. Since the prognosis of advanced HCC remains poor, there is an urgent need to develop new therapeutic approaches. Porphyrin metal-organic frameworks are a class of porous organic-inorganic hybrid functional materials with good biocompatibility. Methods Gadolinium-porphyrin metal-organic frameworks were used as a skeleton for folic acid (FA) conjugation to synthesize a novel type of nanoparticle, denoted as folic acid-nanoscale gadolinium-porphyrin metal-organic frameworks (FA-NPMOFs). The FA-NPMOFs were characterized using transmission electron microscopy, Fourier transform infrared spectroscopy and thermogravimetric-differential thermal analysis. The biotoxicity and imaging capability of the FA-NPMOFs were determined using HepG2 cells and embryonic and larval zebrafish. The delivery and photodynamic therapeutic effect of FA-NPMOFs were explored in transgenic zebrafish with doxycycline-induced HCC. Results FA-NPMOFs were spherical in structure with good dispersion and water solubility. They showed low biotoxicity, emitted bright red fluorescence, and exhibited an excellent magnetic resonance imaging capability, both in vitro and in vivo . Meanwhile, the FA-NPMOFs exhibited a strong affinity for folate receptor (FR)-expressing cells and were delivered to the tumor site in a targeted manner. Moreover, HCC tumor cells were eliminated following laser irradiation. Conclusion FA-NPMOFs can be used for dual-modality imaging and photodynamic therapy in HCC and show promise for use as a carrier in new therapies for HCC and other FR-positive tumors.

Sorafenib is a multitarget tyrosine kinase inhibitor for the first-line treatment of hepatocellular carcinoma (HCC). However, moderate therapeutic effects are common in the clinic. Nanotechnology has provided feasible strategies to solve this problem. In the current study, we coated sorafenib with human serum albumin (HSA) and grafted folic acid (FA) onto the nanoparticle surface (FA-HSA-SRF-NPs) to enhance tumor targeting. Our results showed that the new nanoparticles had a high loading efficiency with a small diameter and good stability. In vitro , the FA-HSA-SRF-NPs showed enhanced cytotoxicity against hepatocellular BEL-7402 cells and increased the safety for normal liver LO2 cells. In vivo , FA-HSA-SRF-NPs exhibited effective antitumor activity toward nude mice bearing xenograft tumors without systemic toxicity, and this antitumor activity manifested as the inhibition of tumor proliferation and suppression of tumor angiogenesis. In addition, FA-HSA-SRF-NPs notably enhanced the accumulation of sorafenib in the tumor tissues of mice and markedly increased the amount of sorafenib in systemic circulation in rats. This current study indicated that FA-HSA-SRF-NPs could increase the efficacy of sorafenib against liver tumors that highly express the folate receptor (FR) and possess good safety, thereby highlighting their potential clinical value.

Herein, distinctive nanomaterial-based mesoporous silica nanoparticles (MSNs) successively coupled with bovine serum albumin (BSA) and folic acid (FA) were engineered for targeting the release of epirubicin (EPI). The BSA molecule as a “gatekeeper” prevents untimely drug leakage until the BSA coating layer is biodegraded in response to GSH. FA promotes the specific intracellular delivery of the drug to high folate receptor (FR)-expressing cancer cells to improve the efficacy of chemotherapy. In vitro drug release profiles show that this drug carrier can avoid premature release of the drug under physiological conditions, and redox-responsive EPI release from EPI/MSN-FA with easy biodegradability is significantly accelerated in the presence of GSH due to the cleavage of the intramolecular disulfide bond; thus, a “redox-triggered release” of the drug in the simulated targeted tissue microenvironment is achieved. The folate-mediated cellular uptake measured by confocal laser scanning microscopy (CLSM) shows that the hybrid nanoparticles more likely enter the FR-positive (FR+) HepG2 cells than the FR-negative (FR−) PC-12 cells. Toxicity experiment also demonstrates the negligible toxicity of MSN-FA and an increase in the antitumor suppression of EPI/MSN-FA with an increase in the dosage. The results show that the strategy of combining mesoporous silica with protein molecules having stimuli-responsive biodegradability will offer new potential directions towards the “on-demand” drug release for pinpoint chemotherapy in cancer treatment.

Folic acid (FA) is the natural form of water-soluble vitamins widely found in most plants and animal products and its deficiency leads to several human body abnormalities. The advancements of metal nanoclusters are highly increasing due to their molecule-like optical properties and attractive applications. Because of increasingly demand of noble metal nanoclusters as sensing templates, different synthesis methods have been developed for facile synthesis of noble metal nanoclusters . Herein, red-emitting fluorescent bovine serum albumin (BSA)-capped Au–Ag bimetallic NCs are facilely synthesized through green one-pot synthetic approach. The effect of silver on the fluorescence properties of Au NCs was investigated and it was found that introduction of silver can enhance the fluorescence intensity . The fluorescence intensity of the as-prepared Au–Ag nanoclusters gets quenched in the presence of folic acid in an aqueous medium and it was used as ultrasensitive sensing probe for FA detection. The developed Au–Ag NCs-based sensing probe shows linear response in the wide range of 0–100 μM and the detection limit is as low as 0.47 nM. Its applicability has also been confirmed successfully in real human serum, urine and FA tablet samples. Due to the high stability, sensitivity and selectivity , the developed bimetallic cluster sensing system is highly promising to be applied in the pharmaceutical and clinical laboratories.

Although some detection methods for folic acid have been developed based on the conventional instruments, the rapid, simple and in-situ detection for folic acid was rarely reported. In this work, a “turn-on” and “color-changing” fluorescence sensor ( TBCS ) on detecting folic acid was designed and synthesized. TBCS emits the green fluorescence, which transferred to the strong blue fluorescence after binding folic acid. The high sensing selectivity was observed among various ions and biomolecules. The detection limitation of TBCS for folic acid was 4.32 × 10 −7 M. The sensing mechanism was proposed as that the ICT effectiveness of TBCS was interrupted by sensing folic acid. The application experiments on test paper, folic acid tablets and simulated sample displayed the good in-situ detecting ability of TBCS for folic acid, suggesting the good application prospect on rapid, simple and in-situ detecting folic acid in real environments.

Folic acid (FA) is an essential vitamin in humans, and thus, rapid, accurate, and sensitive methods for its quantification in different biological samples are needed. This work describes a novel, simple, and effective dual-emission fluorescence nanoprobe for FA detection and quantification. The probe was covalently linked to amino-modified orange quantum dots (QDs) and carboxyl-modified blue graphene quantum dots (GQDs). The resulting material exhibited two emission peaks at 401 and 605 nm upon excitation at 310 nm. The probe had good selectivity and sensitivity toward FA with an exceptionally low detection limit (LOD = 0.09 nM). This probe was effectively used to quantify FA in animal serum samples. The method has potential utility for FA analysis in different types of biological samples.

Acute kidney injury (AKI) is a common clinical problem with high morbidity and mortality. The discovery of ferroptosis has provided novel insights into the mechanisms underlying AKI and paves the way for developing ferroptosis-based approaches to treat AKI. Glycyrol (GC) is a representative coumarin compound isolated from licorice that demonstrates various pharmacological activities. However, its potential for a protective effect against kidney injury remains unknown. We hypothesized that GC might be able to protect against AKI via suppression of ferroptosis. This hypothesis was tested in a cell-culture model of RSL3-induced nephrocyte ferroptosis and a mouse model of folic acid-induced AKI. The results showed that GC exerted a significant protective effect against nephrocyte ferroptosis in vitro and was effective against folic acid-induced AKI in vivo, where it was mechanistically associated with suppressing HO-1-mediated heme degradation. Collectively, the findings of the present study support the hypothesis that GC holds considerable potential to be developed as a novel agent for treating ferroptosis-related AKI.

The aim of the present study was to determine the binding mechanisms between zein and folic acid in an ethanol – water solution. Fluorescence spectroscopy demonstrated that folic acid quenched the fluorescence intensity of zein by both dynamic and static fluorescence quenching mechanisms, with static quenching playing a prominent role. The binding constants (Ka) of the zein-folic acid complex at 292, 302, and 312 K were evaluated to be 5.97 × 105, 3.00 × 105 and 1.54 × 105 L/mol, respectively. The process of folic acid binding zein was driven by ∆H0 = –51.24 kJ/mol, ∆S0 = –64.90 J/mol•K. Van der Waals forces and hydrogen bonding primarily governed the formation of the zein-folic acid complex. Confirmation of the complex formation was obtained via UV-visible absorption spectroscopy, Fourier transform infrared spectroscopy, and circular dichroism spectroscopy, which also revealed alterations in the secondary structure of zein upon binding to folic acid. Measurements of particle size and zeta potential demonstrated an enhancement in the stability of the complex system. Scanning electron microscopy and transmission electron microscopy showed the complex to possess a spherical morphology, with significant changes in the zein’s morphology following its binding to folic acid. Additionally, zein increased the photostability of folic acid against UV light radiation. Molecular docking and molecular dynamics simulations were used to investigate the binding mechanisms involved further, and the results effectively elucidated the observed experimental phenomena. This research establishes a significant theoretical foundation for the utilization of zein as a system for delivering and safeguarding folic acid.

In this study, CuS@PDA nanoparticles were synthesized and used to create a novel tumor-targeting nanocomposite platform composed of copper sulfide@polydopamine–folic acid/doxorubicin (CuS@PDA–FA/DOX) for performing both photothermal and chemotherapeutic cancer treatment. The nanocomposite platform has ultrahigh loading levels (4.2 ± 0.2 mg mg−1) and a greater photothermal conversion efficiency (η = 42.7%) than CuS/PDA alone. The uptake of CuS@PDA–FA/DOX nanocomposites is much higher in MCF-7 cells than in A549 cells because MCF-7 cells have much higher folic acid receptors than A549. Under near infrared (NIR) irradiation, the CuS@PDA–FA/DOX system using a synergistic combination of photothermal therapy and chemotherapy yields a better therapeutic effect than either photothermal therapy or chemotherapy alone. The treatment is very effective with the cell viability is only 5.6 ± 1.4%.

Graphical Targeted drug delivery : Nanoscale metal–organic frameworks modified with folic acid (FA-NMOFs) have been prepared as potential drug carriers for targeted delivery to tumor cells (see figure). The anticancer drug 5-fluorouracil (5-FU) was encapsulated in the pores of the FA-NMOFs. The 5-FU-loaded FA-NMOFs showed excellent biocompatibility and were found to effectively kill tumor cells through the targeting effect of the FA-NMOFs. Nanoscale metal–organic frameworks (NMOFs) have proven to be a class of promising drug carriers as a result of their high porosity, crystalline nature with definite structure information, and potential for further functionality. However, MOF-based drug carriers with active tumor-targeting function have not been extensively researched until now. Here we show a strategy for constructing active tumor-targeted NMOF drug carriers by anchoring functional folic acid (FA) molecules onto the metal clusters of NMOFs. Two zirconium-based MOFs, MOF-808 and NH 2 -UiO-66, were chosen as models to reduce to the nanoscale for application as drug carriers, and then the terminal carboxylates of FA molecules were coordinated to Zr 6 clusters on the surfaces of the nanoparticles by substitution of the original formate or terminal -OH ligands. The successful modification with FA was confirmed by solid-state 13 C MAS NMR and UV/Vis spectroscopy and other characterization methods. Drug loading and controlled release behavior at different pH were determined by utilizing the anticancer drug 5-fluorouracil (5-FU) as the model drug. Confocal laser scanning microscopy measurements further demonstrated that 5-FU-loaded FA-NMOFs have excellent targeting ability through the efficient cellular uptake of FA-NMOFs. This work opens up a new avenue to the construction of active tumor-targeted NMOF-based drug carriers with potential for cancer therapies.
