Unlocking the potential of Komagataeibacter europaeus FMES: A novel source of bacterial cellulose from Monascus vinegar biofilm

Siyu Liu,Jiaqi Luo,Ying Shi,Qihe Chen

Various bacterial species synthesize bacterial cellulose (BC), each exhibiting unique metabolic pathways and environmental adaptations. Herein, the microbial profile of Monascus vinegar biofilm was analyzed for the first time using metagenome sequencing, and a BC-producing bacteria, Komagataeibacter europaeus FMES, was isolated from Monascus vinegar biofilm and identified as a novel strain. The complete genome of strain FMES was analyzed using whole genome sequencing, revealing carbon metabolism and BC biosynthesis pathways in detail. The biosynthesis of BC is regulated by the bacterial cellulose synthase ( bcs ) operon, and three bcs operons were identified in the genome. Additionally, the BC production of strain FMES was evaluated under static conditions using different carbon sources. The highest BC yield of 5.89 ± 0.38 g/L was achieved using 5% glycerol as the only carbon source. The addition of 3% ethanol enhanced the biosynthesis of BC by K. europaeus FMES. This work brings valuable insights into the isolation of BC-producing strains from fermented foods and the metabolic pathways of BC synthesis, which provides useful information for further biological research on Komagataeibacter spp., and the potential use of BC in medical wound care and sustainable packaging.

Characterization of active films based on chitosan/polyvinyl alcohol integrated with ginger essential oil-loaded bacterial cellulose and application in sea bass (Lateolabrax japonicas) packaging

Xuening Chen,Weiqing Lan,Jing Xie

The poor mechanical properties, low water-resistance, and limited antimicrobial activity of chitosan (CS)/polyvinyl alcohol (PVA) based film limited its application in aquatic product preservation. Herein, bacterial cellulose (BC) was used to load ginger essential oil (GEO). The effects of the addition of BC and different concentrations of GEO on the physicochemical and antimicrobial activities of films were systematically evaluated. Finally, the application of sea bass fillets was investigated. Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction analysis (XRD) analysis indicated dense networks were formed, which was verified by enhanced physical properties. The mechanical properties, barrier properties, and antimicrobial activities enhanced as GEO concentration increased. CPB 0.8 (0.8 % GEO) film had better tensile strength (TS) and barrier performance, improved the quality, and extended the shelf-life of sea bass for another 6 days at least. Overall, active films are potential packaging materials for aquatic products.

Effect of TEMPO-oxidized bacterial cellulose nanofibers stabilized Pickering emulsion of cinnamon essential oil on structure and properties of gelatin composite films

Dongmei Li,En-Jie Li,Li Li,Bo Li,Shiru Jia,Yanyan Xie,Cheng Zhong

Pure gelatin film often exhibits high hydrophilicity and a lack of antibacterial activity, hindering its practical application in the field of food preservation. To address these issues, we incorporated 2,2,6,6-tetramethylpiperidine-1-oxyl radical (TEMPO)-oxidized bacterial cellulose (TOBC) nanofibers stabilized cinnamon essential oil (CEO) Pickering emulsions into the gelatin matrix to develop active food packaging films. The study revealed that the good distribution of emulsion droplets in the film matrix. While with increasing Pickering emulsion proportion, the microstructures of composite films were more heterogeneous, showing some pores or cavities. In addition, the insertion of TOBC-stabilized CEO emulsions could improve the elongation at break (EAB), water-resistance, UV blocking ability, and antibacterial activity of film, but reduced its tensile strength (TS) and water vapor barrier properties (WVP). Notably, the film prepared with 4 % TOBC-stabilized CEO Pickering emulsion demonstrated enhanced preservation of strawberries. Overall, the as-prepared gelatin-based active composite films have considerable potential for food packaging.

Construction and Particulate Filtration Performance of Bacterial Cellulose-Derived Aerogels Optimized by Plant Polysaccharides

Han Han,Hetang Wang,Rujie Wang,Zhijie Duan,Xiaodong Jiang,Jiaqi Du

Air pollution has emerged as a global challenge, posing significant threats to public health and human well being. This issue has garnered substantial attention from researchers focused on developing environmentally friendly and sustainable materials. This study synthesizes bacterial cellulose (BC)-derived aerogels based on plant polysaccharides and BC using a directional freeze-drying technique, followed by surface modification with methyltrimethoxysilane (MTMS). The physicochemical characteristics of these BC-derived aerogels are thoroughly investigated to elucidate the interaction mechanisms between plant polysaccharides and bacterial cellulose at the molecular level, and their capabilities for particulate matter filtration are explored. The results demonstrate that incorporating plant polysaccharides and MTMS into BC aerogels results in synergistic mechanical properties characterized by a unique combination of softness and rigidity. Notably, sodium alginate shows the highest affinity for bacterial cellulose and MTMS, leading to optimal reinforcement effects. An oriented honeycomb structure forms internally within the aerogels, potentially reducing pressure drop. Furthermore, these aerogels exhibit a “fiber+pore” multi-filtration mechanism, achieving up to 97% filtration efficiency specifically for PM2.5 particles. These findings suggest bacterial cellulose-derived aerogels could be a sustainable alternative for mitigating air pollution.

Enhancing refrigerated chicken breasts preservation: Novel composite hydrogels incorporated with antimicrobial peptides, bacterial cellulose, and polyvinyl alcohol

Sha Si,Xiaoxia Huang,Qi Wang,Sivakuma Manickam,Dan Zhao,Yanan Liu

Microbial contamination annually leads to substantial food resource loss. Effective food packaging can mitigate food contamination and waste, yet conventional materials such as plastics often lack bacteriostatic activity. This study aimed to synthesise FengycinA-M3@bacterial cellulose@polyvinyl alcohol composite hydrogels via dual cross-linking with hydrogen and borate bonding, with the goal of enhancing antibacterial properties and prolonging the preservation period of refrigerated chicken breast. The composite hydrogel was subjected to comprehensive characterisation for structural, mechanical, water absorption, slow peptide release, antimicrobial capacity, biocompatibility, and chicken breast freshness preservation. The results showed that the composite hydrogel had a porous network structure and excellent gel elasticity and biocompatibility. It was effective in inhibiting Staphylococcus aureus and Escherichia coli , and prolonged the storage time of frozen chicken breast for up to 12 days. These findings emphasise the potential of hydrogel food packaging to prolong storage periods and its suitability for food industry applications due to ease of manufacture.

Amine-impregnated elastic carbon nanofiber aerogel templated by bacterial cellulose for CO2 adsorption and separation

Sifan Bao,Xudong Zheng,Zihuai Xu,Biao Ji,Zhouzhou Yang,Wei Sun,Jinfeng Mei,Jian Rong,Zhongyu Li

Carbon aerogel has become a promising electrode material for CO 2 capture and capacitor due to its large specific surface area, well-developed pore structure, adjustable porosity and ultra-high specific power. However, most of the carbon-based materials suffer from poor mechanical properties, low recycling efficiency, and low adsorption capacity, making it difficult to be put into industrial applications on a large scale. Bacterial cellulose (BC) has an ultra-fine reticular structure, a modulus of elasticity that is several to more than ten times that of typical plant fibers, and high tensile strength. In this paper, BC was treated by unidirectional freeze-drying method, and the aerogels with ordered honeycomb pore structure were constructed by unidirectional growth of ice crystals, and the pyrolytic chemical properties of BC were adjusted by using (NH 4 ) 2 SO 4 , which effectively prevented the shrinkage and deformation of materials during carbonization, and successfully prepared elastic nanocarbon fiber aerogels. At the same time, the introduction of (NH 4 ) 2 SO 4 made the material realize N doping in the carbonization process, providing more adsorption sites, which was conducive to the adsorption of CO 2 . Finally, the carbon fiber nanoaerogel was modified by TEPA to further improve its adsorption selectivity for CO 2 . The prepared elastic carbon nanofiber aerogel (CBCNT) has high CO 2 adsorption performance and high selectivity, and its CO 2 capture capacity is up to 4.88 mmol/g. At the same time, its capacitance also reached 246F/g.

Biosynthesis of positively charged bacterial cellulose hydrogel with antibacterial and anti-inflammatory function for efficient wound healing

Suriguga Meng,Qinggele Borjihan,Douxin Xiao,Yutian Wang,Mei Chen,Chunzu Cheng,Alideertu Dong

In bacterial cellulose (BC)-based living materials, the effective and permanent incorporation of bactericidal agents into BC remains a persistent challenge. In this study, midazole quaternary ammonium salt was grafted onto a dispersion of bacterial cellulose, which was subsequently directly added to the fermentation medium of BC-producing bacteria to obtain BC-based hydrogel materials (BC/BC-[PQVI]Br) with inherent antibacterial properties. The BC/BC-[PQVI]Br hydrogel prepared in this study exhibits favorable tensile properties, with a maximum tensile stress of 970 KPa and water retention for up to 6 h. Moreover, it demonstrates acceptable antibacterial activity against S. aureus (93 %) and E. coli (71 %), respectively. Additionally, the hydrogel displays a high cell survival rate of 98 % after contact with NIH 3T3 cells, indicating its non-cytotoxic nature. Furthermore, the mouse wound experiment confirms the excellent wound healing effect of the hydrogel. This research presents an innovative approach towards developing environmentally friendly active wound dressings with microbial-derived antibacterial functionality.

Bio-inspired chemistry for developing micro/nano structured nanofiber membranes for membrane distillation

Yan Guo,Qidi Wu,Hongyun Liu,Yumei Xian,Xun Zhou,Ying Wu,Peiqing Zhang,Zhenyu Li

Membrane distillation (MD) holds significant promise for effluent purification to mitigate the global water shortage. However, the existing petroleum-based MD membranes suffer from high costs, limited sustainability, and wetting issues. Herein, we employed biosynthetic bacterial cellulose (BC) as the substrate and successfully achieved a uniform and stable dispersion of ZnO nanoflowers onto the BC nanofibers through in-situ synthesis technology. After undergoing a fluorination process, a superhydrophobic membrane was prepared by the synergistic effect of micro/nano structures and low surface energy. The resulting membrane exhibited exceptional properties, including a remarkable contact angle of 151.7°, a substantial liquid entry pressure of 2.5 bar, and exhibiting structural integrity and high hydrophobicity even under harsh conditions. Furthermore, this strategy ensured exceptional membrane porosity (∼94.9 %), increased the permeation flux and reduced conductive heat transfer, thereby enhancing the thermal efficiency of the membrane. Furthermore, the membrane demonstrated effective bactericidal activity and self-cleaning capability, facilitating easy cleaning and extending service life. These properties endow the membrane with an impressive durability. After 96 h of continuous operation, the membrane maintained a stable permeation flux of 10.29 L m −2  h −1 and an extraordinary salt rejection rate exceeding 99.9 %. Notably, even with the addition of sodium dodecyl sulfate to the feed, the membrane exhibited stable permeation flux and outstanding antiwetting properties. In summary, the MD membrane fabricated using the bio-inspired chemistry demonstrated a promising potential for long-term applications in direct contact MD operations.

Bidirectional anisotropic bacterial cellulose/polyvinyl alcohol/MXene aerogel phase change composites for photothermal conversion enhancement

Liheng Zhu,Boyang Zou,Naici Bing,Huaqing Xie,Wei Yu

The issues of inherent low thermal conductivity , slow heat transport rate and easy leakage are critical challenges for accelerating solar-thermal energy harvesting and storage for organic solid-liquid phase change materials (PCMs). Aerogel carriers with thermal conductivity enhancement based on ice temple methods show favorable potential for PCMs solar thermal applications. Comparing with disordered and unidirectional freezing, bidirectional freezing can assemble efficiently in multiple dimensions and result in long-range ordered structures. In this paper, bacterial cellulose/polyvinyl alcohol/MXene aerogels were constructed by bidirectional freezing. The anisotropic PCMs composites were from vacuum impregnation PEG into aerogel networks. By controlling crystallization and growth of ice crystals through the dual temperature gradients , the PCMs composites obtained enhanced heat transfer efficiency in horizontal and vertical directions. Bidirectional aligned PCMs with PDMS wedge angle of 0° and 20° exhibit high thermal conductivity of 0.716 W m −1  K −1 and 0.768 W m −1  K −1 , which were 184% higher than that of PEG , and photothermal conversion efficiency of 55.17% and 76.91%. Remarkably, the PCMs composites with a 20° PDMS wedge angle also exhibited a high PCM loading capacity (96.3%) and a high enthalpy (157.7 J/g). The results show that bidirectional anisotropic PCMs composites have excellent overall performance and are expected to improve solar thermal application.

In Situ Anchored, Ultrasmall, Oxygen Vacancy-Rich TiO2−x on Carbonized Bacterial Cellulose for the Efficient Adsorption and Separation of Organic Pollutants

Man Zhou,Yanli Zhou,Minmin Ni,Yuzhe Zhang,Song Xu,Hao Ma,Jian Zhou,Jin Zhao,Liwei Lin,Zhongyu Li

Superior selective adsorption of organic dye is still a big challenge in the process of dye wastewater treatment. Meanwhile, low-price and environmentally friendly biomass-based adsorbents show huge potential in the fields of separation and purification. In this study, we adopted the “hydrolysis–calcination method” to develop a novel in situ anchoring strategy for ultrasmall TiO2−xon carbonized bacterial cellulose (CBC), which was derived from natural bacterial cellulose. Notably, 3D networks of porous CBC played a dual role for both providing hydrolytic sites and controlling the oxygen vacancies (Vo) of TiO2−x. As for the single-dye adsorption, the TiO2−x/CBC had a strong adsorption ability (101.4 mg/g) for removing methylene blue (MB), which was much higher than that of methyl orange (MO), malachite green (MG), rhodamine B (RhB), and tetracyclines (TC). Moreover, under the optimized carbonization temperature (Tc) of 300 °C, the TiO2−x/CBC-300 exhibited an outstanding separation efficiency of 97.07% for the MB/MO solution. Detailed analysis confirmed that Tcwas a key regulator for adjusting the Voconcentration, which directly influenced the surface charge density and, further, the separation efficiency of TiO2−x/CBC. Additionally, the used adsorbent could be easily regenerated from washing by ethanol. After 4 regenerations, the adsorption efficiency declined only by 6.9% after 20 min and 13.6% after 120 min adsorption, respectively. Ultimately, this oxygen vacancy-rich TiO2−x/BC system illuminated good prospects for mixed dye wastewater adsorption and separation.

Concentrated O/W Pickering emulsions stabilized by soy protein/cellulose nanofibrils: Influence of pH on the emulsification performance

Xingzhong Zhang,Xiaogang Luo,Yixiang Wang,Yan Li,Bin Li,Shilin Liu

In this work, concentrated Pickering emulsions had been prepared by using soybean protein isolate (SPI) and TEMPO-oxidized bacterial cellulose (TOBC) complex as solid stabilizers. The effects of solid particle content, pH and oil phase volume fraction on the morphology, stability and rheological behaviors of the emulsions had been investigated. It indicated that the emulsion droplets had uniform particle size distribution when the ratio of SPI/TOBC was controlled to 12.5:1 (wt/wt). The emulsification performance could be improved with the increase of solid complex particles content from 1.0% to 3.0%. Moreover, the pH played an important role in regulating the structure and properties of the emulsions. With microscopic observation, the emulsions at pH 7.0 had smaller droplet size (10–15 μm) than that (15–40 μm) of the emulsions at pH 3.0. While the emulsions at pH 3.0 maintained a low creaming index (CI) value than those at pH 7.0, and uniform cellular network structure could be seen by SEM observation. It suggested that the electrostatic complexation between soy proteins and anionic bacterial cellulose nanofibrils at pH 3.0 was more favorable for stabilizing emulsions than those at pH 7.0. This work would provide some guidance for regulating the properties of emulsions that stabilized by soy protein and cellulose nanofibrils.

Facile and Green Route to Fabricate Bacterial Cellulose Membrane with Superwettability for Oil–Water Separation

Huazheng Sai,Zhiqiang Jin,Yutong Wang,Rui Fu,Yaxiong Wang,Litong Ma

The separation of oil–water mixtures by membranes with hydrophilicity and underwater superoleophobicity is an effective method. However, the fabrication of these oil–water separation membranes usually needs a complex chemical treatment process. In this study, a facile and green method to fabricate bacterial cellulose membranes (BCMs) with superwettability through one-step filtration process without any further tedious and costly chemical modification for efficient oil–water separation is proposed. Owing to the ultrafine (20–80 nm in diameter) nanofiber structure and intrinsic hydrophilicity of bacterial cellulose (BC), BCMs with a 3D web-like structure show superhydrophilicity in air (water contact angles about 0°) and underwater superoleophobicity (oils contact angles >150°). During the oil–water separation process, water permeates through BCMs quickly while oils remain at the BCMs' upper side. Furthermore, all separation processes present high separation efficiency (>99.7%) and high fluxes. In addition, two different models are proposed to describe the formation and microstructure of BCMs and how it affect the separation process. Thus, this work provides a facile and environment benign route for obtaining excellent separation membranes for oil-polluted-water treatment.
