Catalytic hairpin assembly-based AIEgen/graphene oxide nanocomposite for fluorescence-enhanced and high-precision spatiotemporal imaging of microRNA in living cells

Yuchen Song,Changqing Mao,Wenjiao Zhang,Dongmei Deng,Huinan Chen,Pei Sun,Meiyin Liu,Chang Feng,Liqiang Luo

The low abundance, heterogeneous expression, and temporal changes of miRNA in different cellular locations pose significant challenges for both the detection sensitivity of miRNA liquid biopsy and intracellular imaging. In this work, we report an intelligently assembled biosensor based on catalytic hairpin assembly (CHA) and aggregation-induced emission (AIE), named as catalytic hairpin aggregation-induced emission (CHAIE), for the ultrasensitive detection and intracellular imaging of miRNA-155. To achieve such goal, tetraphenylethylene-N 3 (TPE-N 3 ) is used as AIE luminogen (AIEgen), while graphene oxide is introduced to quench the fluorescence. When the target miRNA is present, CHA reaction is triggered, causing the AIEgen to self-assemble with the hairpin DNA . This will restrict the intramolecular rotation of the AIEgen and produce a strong AIE fluorescence. Interestingly, CHAIE does not require any enzyme or expensive thermal cycling equipment, and therefore provides a rapid detection. Under optimal conditions, the proposed biosensor can determine miRNA in the concentration range from 2 pM to 200 nM within 30 min, with the detection limit of 0.42 pM. The proposed CHAIE biosensor in this work offers a low background signal and high sensitivity, making it applicable for highly precise spatiotemporal imaging of target miRNA in living cells.

AIEgen-based porous organic polymer-Ag nanoparticles hybrids with coreaction accelerator amplification and covalent rigidification-enhanced electrochemiluminescence for aflatoxin M1 detection

Jun-Qi Song,Qiu-Yi Peng,Mei-Ling Lu,Shu-Zhen Gao,Wen-Bin Liang,Shaoguang Sun,Ruo Yuan,Dong-Rong Xiao

Developing new high-performance electrochemiluminescence (ECL) emitters is a continuous hotspot. Herein, a novel AgNP-decorated porous organic polymer (AgNPs@ETTBC-BPyDAN-POP, BPyDAN = 2,2′-([2,2′-bipyridine]-5,5′-diyl)diacetonitrile) nanocomposite with coreaction accelerator amplification and covalent rigidification-enhanced ECL was prepared. AgNPs@ETTBC-BPyDAN-POP nanocomposite exhibited excellent ECL performance, not only because abundant AIE-active 4′,4″′,4″″′,4″″″′-(ethene-1,1,2,2-tetrayl)tetrakis(([1,1′-biphenyl]-4-carbaldehyde)) (ETTBC) luminogens were covalently assembled and rigidified into ETTBC-BPyDAN-POP network, which diminished radiationless transition, but also because AgNPs functioned as coreaction accelerators that expedited the generation of coreactant radicals, which in-situ react with adjacent ETTBC + intermediates, thereby producing more ETTBC ⁎ excited states for significantly enhancing the ECL emission. Considering the eminent ECL performance, AgNPs@ETTBC-BPyDAN-POP was employed as an ECL indicator to fabricate a supersensitive biosensor for detecting aflatoxin M1, showing a wide response range (5 fg/mL − 500 ng/mL) with a detection limit of 0.57 fg/mL. Overall, this study offered a promising dual-enhancement strategy to boost AIEgens' ECL emission, thereby providing significant inspiration to develop high-efficiency AIEgen-based ECL materials for fabricating hypersensitive ECL biosensors.

Molecular Structural Transformation from the D–A to D–D’ of an AIEgen for the Highly Sensitive Detection of Hydrogen Peroxide in Exhaled Breath Condensate

Qiaoru Zhang,Yu Wang,Tianyu Song,Mashooq Khan,Yanjun Gong,Yongxian Guo,Qiongzheng Hu

Hydrogen peroxide (H 2 O 2 ) plays a vital role in various physiological activities. Its efficient detection in exhaled breath condensate (EBC), a highly promising noninvasive diagnostic matrix, is of great significance for the early diagnosis of diseases. However, complex biological components present in EBC impede efficient H 2 O 2 detection. Herein, we synthesize an H 2 O 2 probe, TPEB , which demonstrates excellent sensing performance in phosphate-buffered saline (PBS) and EBC. TPEB exhibits turn-on fluorescence signal generated through H 2 O 2 -triggered hydrolysis, releasing TPEN , a compound with characteristic aggregation-induced emission properties. Density functional theory (DFT) and time-dependent DFT calculations reveal a molecular transformation from the donor–acceptor (D–A) structure of TPEB to the D–D′ structure of TPEN . Leveraging this transformation, the probe achieves a limit of detection (LOD) of approximately 0.1   μM in a DMSO/PBS buffer solution for H 2 O 2 , with excellent selectivity and stability. To enhance its portability and applicability, a probe film is fabricated by depositing TPEB nanoparticles onto a filter paper. This film exhibits stronger emission than TPEB in the DMSO/PBS buffer solution and achieves a lower LOD of 0.01   μM. The probe film enables effective H 2 O 2 detection in complex matrices, such as cell culture medium and EBC, demonstrating potential for practical point-of-care testing applications.

Multifunctional nanogel engineering with redox-responsive and AIEgen features for the targeted delivery of doxorubicin hydrochloride with enhanced antitumor efficiency and real-time intracellular imaging

Yuwei Ma,Huiyi Zhou,Fan Hu,Zhichao Pei,Yongqian Xu,Qi Shuai

Nanogels exhibit potential application values in drug delivery systems because of their tunable properties and biocompatibility. In this study, multifunctional hyaluronic-based nanogels (HNPs), all-trans retinoid acid (ATRA)/aggregation-induced emission luminogen (AIEgen) fluorophores (TPENH2)-grafted hyaluronic acid (HA) with disulfide bonds as linkers of (HA-ss-ATRA/TPENH2), were successfully developed for doxorubicin hydrochloride (DOX) delivery. Besides, the controls of HA-ATRA/TPENH2 were also developed for comparison. The HNPs with nanoscale particle sizes possessed an excellent DOX-loading capacity. As expected, the DOX-loaded HNPs exhibited the higher stability in a normal physiological environment (10 µM GSH), but rapidly disintegrated in the cancer microenvironment (20 mM GSH) according to an in vitro drug release study. The intracellular observation of HNPs by the fluorescence microscopy indicated that the self-fluorescent DOX-loaded HNPs with unique AIEgen characteristic transported DOX into the cancer cells and visibly accumulated within the cytoplasm. Importantly, the accumulation of DOX-loaded HNPs was largely increased due to the targeted reorganization of CD44 or LYCE-1 receptors by HA moieties on the surface of HNPs. Based on in vitro cytotoxicity analyses, the DOX-loaded HNPs displayed dramatically enhanced virulence to cancer cells compared to the controls and free DOX. This is the first study to achieve combined functionalities of targeted delivery, controllable release and real-time intracellular imaging on HA-base nanogels with enhanced antitumor efficiency. Therefore, the multifunctional HA-ss-ATRA/TPENH2 HNPs have good potential to develop a novel drug delivery platform for the targeted delivery and controlled release of DOX to achieve enhanced antitumor efficiency as well as real-time intracellular imaging.
