Controllable in situ fabrication of portable AuNP/mussel-inspired polydopamine molecularly imprinted SERS substrate for selective enrichment and recognition of phthalate plasticizers

Yuan-Yuan Yang,Yuan-Ting Li,Xue-Jian Li,Lei Zhang,Essy Kouadio Fodjo,Sheng Han

A bio-inspired polydopamine (PDA) molecularly imprinted substrate is fabricated by one-step oxidative self-polymerization of dopamine and template molecules in weakly alkaline medium on screen-printed electrode (SPE). Through the catechol and amine groups on PDA surface, Au nanoparticles (AuNPs) can be easily anchored and growth on PDA-MIP coating, thereby forming a three-dimensional (3D) architecture. By adjusting the reaction conditions, the particle size and distribution of PDA and AuNPs can be well controlled. Benefiting from the “hot spots” generated by AuNPs and recognition sites produced by MIP, the AuNP/PDA-MIP nanocomposite can serve as excellent SERS substrate enabling selective enrichment and identification of phthalate plasticizers (PAEs). Using this substrate, an enhancement factor (EF) up to 1.70 × 10 7 for dimethyl phthalate (DMP) is achieved with a detection limit as low as 1.0 × 10 −10 M. Furthermore, the rebinding kinetics study demonstrates that the portable MIP-based SERS substrate can promptly reach the adsorption equilibrium within 10 min. Thanks to its highly selectivity, the substrate exhibits low SERS interference for structural analogues benzyl butyl phthalate (BBP) and di(2-ethylhexyl) phthalate (DEHP). Besides, the straightforward SERS recognition of DMP in surface water, commercial bottled water and liquor samples are successfully realized using this substrate. This green and environmental-friendly fabrication method is promising for synthesis of highly performance sensing substrate, adsorbent or catalysts. Combining with portable Raman spectrometer, the fabricated MIP-based SERS substrate has exhibited a great potential as on-site device for environmental monitoring, food safety and homeland security.

A triply amplified electrochemical lead(II) sensor by using a DNAzyme and via formation of a DNA-gold nanoparticle network induced by a catalytic hairpin assembly

Song Xiaolei,Wang Yu,Liu Su,Zhang Xue,Wang Jingfeng,Wang Haiwang,Zhang Fenfen,Yu Jinghua,Huang Jiadong

An amplified electrochemical biosensing scheme is described for lead(II) ions. It is making use of DNAzyme-assisted target recycling and catalytic hairpin assembly (CHA). The hairpin strand (substrate probe for the Pb 2+ -based DNAzyme; referred to as SP) is composed of trigger probe (TP) and a capture probe 1 attached to gold nanoparticles (AuNP). In the presence of the enzyme probe that partially hybridizes with SP, the introduction of Pb 2+ triggers target recycling and drives the highly amplified translation of target Pb(II) to TP. The CHA reaction is further initiated by TP. The modified AuNP act as the connecting unit, and this leads to the formation of a 3D DNA-AuNP network on the electrode (which is the third amplification step). It can bind the positively charged redox mediator RuHex via electrostatic interaction for electrochemical detection. This biosensor has a low detection limit (95 pM) and any analytical range that covers the 100 pM to 5 μM Pb(II) concentration range. It is selective over other divalent metal ions. It was applied to the determination of Pb 2+ in spiked real-world samples. Schematic presentation of the electrochemical biosensor. The triply amplified electrochemical assay is based on the use of DNAzyme-assisted target recycling with catalytic hairpin assembly (CHA) reaction for sensitive and selective determination of lead ion (Pb 2+ ). Point-of-care testing device platform for the determination of creatinine on an enzyme@CS/PB/MXene@AuNP-based screen-printed carbon electrode

Li Yilong,Hang Yuteng,Gopali Rusha,Xu Xinxin,Chen Guanhua,Guan Xiaorong,Bao Ning,Liu Yang

Screen-printed carbon electrodes (SPCE) functionalized with MXene-based three-dimensional nanomaterials are reported for rapid determination of creatinine. Ti 3 C 2 T X MXene with in situ reduced AuNPs (MXene@AuNP) were used as a coreactant accelerator for efficient immobilization of enzymes. Creatinine could be oxidized by chitosan-embedded creatinine amidohydrolase, creatine amidinohydrolase, or sarcosine oxidase to generate H 2 O 2 , which could be electrochemically detected enhanced by Prussian blue (PB). The enzyme@CS/PB/MXene@AuNP/SPCE detected creatinine within the range 0.03–4.0 mM, with a limit of detection of 0.01 mM, with an average recovery of 96.8–103.7%. This indicates that the proposed biosensor is capable of detecting creatinine in a short amount of time (4 min) within a ± 5% percentage error, in contrast with the standard clinical colorimetric method. With this approach, reproducible and stable electrochemical responses could be achieved for determination of creatinine in serum, urine, or saliva. These results demonstrated its potential for deployment in resource-limited settings for early diagnosis and tracking the progression of chronic kidney disease (CKD). Graphical Abstract

An aptamer@AuNP-modified POSS–polyethylenimine hybrid affinity monolith with a high aptamer coverage density for sensitive and selective recognition of ochratoxin A

Xia Yu,Hongliang Song,Jing Huang,Yongxuan Chen,Ming Dai,Xucong Lin,Zenghong Xie

A novel hybrid affinity monolith with a high aptamer coverage density was developed via a highly efficient modification of aptamer@gold nanoparticles (Apt@AuNPs) on a well-controlled 3D skeletal POSS–polyethylenimine (PEI) matrix. The effects of amine reagents and the skeletal structures of the hybrid monoliths on the modification efficiency of Apt@AuNPs were studied, and the resultant Apt@AuNPs@POSS–PEI1200 monolith was proposed with a high coverage density of aptamer of up to 1413 pmol μL−1, which is 4–5 times more than the coverage density when modified with Apt@AuNPs that was reported previously. Characterization including morphology, spectra analysis, mechanical stability, binding capacity and specificity studies was also carried out. Significant specific recognition targeting of ochratoxin A (OTA) was achieved with a detection limit as low as 0.06 ng mL−1. High recoveries of OTA were achieved of 92.6 ± 1.3% to 94.7 ± 1.4% (n = 3), while the cross-reactivity towards analogues such as ochratoxin B (OTB) and aflatoxin B1 (AFB1) was weak. This highlights a facile approach to POSS-based affinity monoliths with a high aptamer coverage density for sensitive and selective recognition of a trace target (OTA).

A target-activated plasmon coupling surface-enhanced Raman scattering platform for the highly sensitive and reproducible detection of miRNA-21

Yanfang Cui,Jing Zheng,Wei Zhuang,Haiwang Wang

A novel surface-enhanced Raman scattering (SERS) platform has been developed for the highly sensitive detection of miRNA-21 by target activation depending on exonuclease-assisted signal amplification with a plasmon coupling phenomenon. In this system, a Raman dye-modified gold nanoparticle (AuNP) substrate can be covalently linked by the thiolated DNA track and DNA walker, which combine the protection probe at the 3′ terminal end to prevent reaction with the DNA track. In the presence of miRNA-21, it will combine the protect probe to the DNA walker depending on more complementary base pairing and release the DNA walker, which will combine with the adjacent DNA track. Depending on the exonuclease III (exo III) cleavage, the DNA walker can walk along the surface of AuNPs and produce the self-assembled hot spots between AuNPs, allowing a homogenous detection of the miRNA-21 with high sensitivity and reproducibility. Under optimized experimental conditions, the platform detected miRNA-21 with a wide range from 10 pM to 1.0 × 105 pM and displayed very low detection limit (4.0 pM). Hence, the novel strategy will become a promising tool for the SERS assay of miRNA-21 with high sensitivity and repeatability. Furthermore, this platform also owns great potential for cancer typing and clinical diagnostics.

An AuNP-labeled triple-helix molecular switch for ultrasensitive, on-site detection of tetracycline in milk via fiber-optic localized surface plasmon resonance biosensing

Xuqi Situ,Jiacong Li,Yeshun Zhang,Hui Yan

To meet the urgent demand for monitoring tetracycline residues in milk—a critical food for vulnerable groups a novel fiber-optic Localized Surface Plasmon Resonance (LSPR) biosensor was engineered using gold-nanoparticle-labeled DNA triple-helix switches. The sensor operates via a target-activated disassembly: tetracycline binding dissociates the immobilized triple-helix, releasing gold nanoparticles and inducing a measurable LSPR blue shift. This active displacement amplifies the signal via localized refractive-index changes, enabling rapid, label-free detection. The sensor achieved excellent performance, with a linear range of 5–100 ng/mL and a low detection limit of 0.28 ng/mL. When applied to real milk samples, it delivered excellent recovery rates of 96.75%–104.15% (RSD <5.1%) and showed no statistically significant difference ( p  > 0.05) compared to HPLC reference analysis. Combining high sensitivity with a modular, engineering-flexible design, this platform not only provides a practical solution for on-site tetracycline screening in food safety but also holds significant potential for adapting to other antibiotics and environmental targets, extending its utility beyond dairy monitoring.

Persistent luminescence nanorod based luminescence resonance energy transfer aptasensor for autofluorescence-free detection of mycotoxin

Yuan-Yuan Jiang,Xu Zhao,Li-Jian Chen,Cheng Yang,Xue-Bo Yin,Xiu-Ping Yan

Serious ochratoxin A (OTA) contamination necessitates the development of rapid, sensitive and selective analytical methods for its determination in food safety. Herein, we report a persistent luminescence resonance energy transfer (LRET) based aptasensor for the autofluorescence-free detection of OTA. OTA aptamer functionalized persistent luminescence nanorod (PLNR) Zn 2 GeO 4 :Mn 2+ and the aptamer complementary DNA modified gold nanoparticle (AuNP) were used as the donor and the acceptor, respectively. The developed LRET aptasensor integrated the advantages of the long-lasting persistent luminescence of PLNR, the high selectivity of aptamer and the low probe background of LRET sensors, allowing autofluorescence-free detection of OTA in biological samples with high sensitivity and selectivity. The developed LRET aptasensor gave an excellent linearity in the range of 0.01–10 ng mL −1 , the detection limit of 3 pg mL −1 and the precision of 2.7% (RSD, n  = 11) at 1 ng mL −1 level. The applicability of the developed aptasensor was demonstrated by analyzing beer samples for OTA with the recoveries of 92.3%–104%.

An ultrasensitive signal-on electrochemical aptasensor for ochratoxin A determination based on DNA controlled layer-by-layer assembly of dual gold nanoparticle conjugates

Wei Chen,Chao Yan,Lin Cheng,Li Yao,Feng Xue,Jianguo Xu

In this paper, a novel ultrasensitive signal-on electrochemical aptasensor has been proposed for Ochratoxin A (OTA) assay based on DNA controlled layer-by-layer assembly of dual gold nanoparticle (AuNP) conjugates. To construct the aptasensor, the 1 st AuNP conjugate was prepared by simultaneous immobilization of the capture probe 2 (CP2) and bridge probe (BP) onto the AuNPs. Then, OTA aptamer was loaded onto 1 st AuNPs by hybridization with CP2. The 1 st AuNP conjugate can be further immobilized onto the electrode by hybridization between BP and capture probe 1 (CP1), which was pre-immobilized on Au electrode. The 2 nd AuNP conjugate was prepared by immobilization of ferrocene (Fc) tagged SH-signal probe (SSP). Due to the recognition between aptamer on 1 st AuNP conjugate and OTA, CP2 was reformed in the ssDNA state, which can be utilized as the anchor for immobilization of 2 nd AuNP conjugate for electrochemical signal reporting. Because of the high surface-to-volume ratio and good conductivity of AuNPs, this dual AuNPs assembled nanoarchitecture finally lead to greatly improved abilities to load a large number of Fc molecules and significantly amply the electrochemical response even at a low target concentration. Both qualitative and quantitative analysis of OTA were thus realized by differential pulse voltammetry (DPV) signals, resulting in an excellent detection limit of 0.001 ppb and a wide dynamic range from 0.001 to 500 ppb over 6 orders of magnitude. Moreover, the real sample analysis towards OTA spiked wine samples was favorable, implying a great potential for practical applications. We envision that this unique dual AuNP conjugate assembly strategy would pave a new avenue for the development of versatile signal amplified electrochemical aptasensors.

Growth of Spherical Gold Satellites on the Surface of Au@Ag@SiO2 Core–Shell Nanostructures Used for an Ultrasensitive SERS Immunoassay of Alpha-Fetoprotein

Ying Yang,Jian Zhu,Jing Zhao,Guo-Jun Weng,Jian-Jun Li,Jun-Wu Zhao

The identification and detection of cancer biomarkers in early stages is an important issue for the therapy of cancer. However, most methods are time-consuming and have limited sensing sensitivity and specificity. In this work, we prepared a novel plasmonic multilayered core–shell–satellite nanostructure (Au@Ag@SiO2–AuNP) consisting of a gold nanosphere with a silver coating core (Au@Ag), an ultrathin continuous silica (SiO2) shell, and a high coverage of gold nanosphere (AuNP) satellites. The Au@Ag core is a prominent surface enhanced Raman scattering (SERS) platform, and the thin SiO2 layer exhibits a long-range plasmon coupling between the Au@Ag core to the AuNP satellites, further leading to enhanced Raman scattering. Meanwhile, the outer AuNP satellites have a high biocompatibility and long-term stability. Combining the above advantages, the well-designed metallic nanoassemblies would be a promising candidate for SERS-based applications in biochemistry. For specific detection of alpha-fetoprotein (AFP), we utilized the SERS-active core–shell–satellite nanostructures modified with AFP antibody as immune probes and nitrocellulose membrane (NC) stabilized captured anti-AFP antibodies as solid substrate. To improve the detection performance, we further systematically optimized the parameters, including the silver coating thickness of the Au@Ag core and the density and size of the satellite AuNPs. Under the optimized conditions, AFP could be detected by the SERS-based sandwich immunoassay with an ultralow detection limit of 0.3 fg/mL, and the method exhibited a wide linear response from 1 fg/mL to 1 ng/mL. The limit of detection (LOD) was considerably lower than conventional methods in the literature. This work relies on the unique Au@Ag@SiO2–AuNP nanostructures as the immune probe develops a new outlook for the application of multilayered nanoassemblies and demonstrates the great potential in early tumor marker detection.

An ultrasensitive colorimetric strategy for protein O-GlcNAcylation detection via copper deposition-enabled nonenzymatic signal amplification

Yingshuai Liu,Jin Xie,Zeying Zhang,Zhisong Lu

O-GlcNAcylation is a very important post-translational modification of nucleocytoplasmic proteins. Aberrant O-GlcNacylation has been found in a variety of human diseases including many types of cancers, diabetes, and Alzheimer's disease. However, analysis of protein O-GlcNacylation still remains a great challenge due to its low levels and labile nature. Herein, a highly sensitive and reliable colorimetric method is developed by use of wheat germ agglutinin (WGA) lectin for specific recognition of O-GlcNAc and gold nanoparticle (AuNP)-catalyzed copper deposition for signal amplification. In presence of the target molecule, AuNP is specifically introduced onto the sensing surface and then catalyzes the copper deposition. An Au@Cu core–shell structure is produced from the deposition. The obtained copper shell on AuNP is dissolved by FeCl3 due to the oxidation–reduction reaction between copper and Fe3+, resulting in Cu2+ and Fe2+, which subsequently form a red coordination complex in the presence of bathophenanthrolinedisulfonic acid disodium salt (BPT2−). Thus, the amount of the red complex is proportional to the level of O-GlcNAc. Qualitative O-GlcNAc detection is realized by direct readout with the naked eye, while the quantitative analysis is achieved by measurement of the absorbance using a UV-Vis spectrophotometer. With increasing time, more and more copper is deposited on the surface of AuNP, thus achieving significant signal amplification. Under optimal conditions, an ultralow limit of detection (LOD) down to 0.02 pg mL−1 is achieved with GlcNAc-conjugated BSA as a model analyte. The developed platform was also successfully applied to analyze O-GlcNAcylation levels in prostate cancer cell line PC-3 and normal cell line RWPE-1. The results obtained by the proposed method are in good agreement with the data from protein microarray assays, demonstrating its great potential for sensitive and reliable O-GlcNAcaylation detection in fundamental research and in cancer diagnosis.

Fluorescence ratio immunoassay for fumonisin B1 based on the oxidase characteristics of the growth of monodispersed 2-D MnO2 nanosheet on an individual gold nanoparticle (AuNP@MnO2)

Yu Yao,Li Yansong,Xu Ning,Zha Yonghong,Bai Xue,Liu Mingyuan,Zhou Yu,Liu Xiaolei

Fumonisin B1 (FB1) is one of the important mycotoxins posing health risks in the area of food safety. A sensitive fluorescence ratio immunoassay has been established for FB1 based on the growth of monodispersed 2-D MnO 2 nanosheet on an individual gold nanoparticle (AuNP@MnO 2 ). FB1 competed with the coated FB1-BSA to bind the FB1 monoclonal antibody. After a washing step, alkaline phosphatase-labeled goat anti-mouse IgG (ALP-IgG) with high catalytic activity was combined with FB1 monoclonal antibody. ALP reacts with ascorbic acid 2-phosphate (AAP) to produce ascorbic acid (AA), which decomposes AuNP@MnO 2 to dehydroascorbic acid (DHAA). O-Phenylenediamine dihydrochloride (OPD) is oxidized to yellow-fluorescent substrate of 2,3-diaminophenazine (DAP) (excitation, 423 nm; emission, 570 nm) by AuNP@MnO 2 . Meanwhile, OPD can also be reduced to blue fluorescent substrate of OPDred (excitation, 350 nm; emission, 430 nm) by DHAA. The content of FB1 can be determined by fluorescence ratio of blue/yellow. The limit of detection (LOD) of the fluorescence ratio immunoassay for FB1 was 0.06 ng mL −1 , and the linear range was from 0.25 to 60.00 ng mL −1 . The effectiveness of the assay was verified in real maize samples, and satisfactory recoveries were attained. The correlation coefficient of these results between the fluorescence ratio immunoassay and commercial ELISA kit was 0.9999. This method provides a sensitive and selective tool for the detection of FB1 in maize samples. Graphical Abstract

Development of dual strip biosensors based on hybridization chain reaction and microplate strategies for signal amplification of HBV-DNA detection

Hai-Bo Wang,Zi-Tao Zhong,Ting Zhang,Yuan-Di Zhao

Hepatitis B virus (HBV) can cause liver cancer, thus the detection of HBV-DNA is of great significance for liver cancer prevention. Since the concentration of HBV-DNA is often low, it is important to establish a highly sensitive detection method. In this study, microplate amplification and hybridization chain reaction (HCR) were used to improve the sensitive detection of HBV-DNA on dual strip biosensors. For the microplate-based amplification strategy on RS strip, the dual-DNA-AuNP probe could be immobilized on SA-Biotin-Anti-HBV-DNA-1 covered microplate through HBV-DNA. By adding dithiothreitol (DTT), a large amount of HS-RS-DNA on the dual-DNA-AuNP probe was dissociated. The indirect detection of HS-RS-DNA on RS strip could achieve the signal amplification of HBV-DNA. For the HCR-based strategy on HCR strip, HBV-DNA triggered the cascade reaction of H1-AuNP probe and H2-Biotin, a large amount of HCR product (AuNP-H1-H2-Biotin-AuNP-H1…) was produced. Thus, a lot of AuNP was fixed on the test zone of HCR strip to improve the HBV-DNA detection sensitivity. The detection limits by HCR strategy on HCR strip and microplate strategy on RS strip could reach to 5 pM and 0.1 nM, respectively, which were 200 and 10-fold higher than that of unamplified strategy, respectively. The recovery of HBV-DNA ranged from 97.2 to 102 % in the spiked human serum, presented a good accuracy for HBV-DNA detection in actual samples.
