Prebiotics such as oligosaccharides, fructans, and resistant starch (RS) stimulate the growth of beneficial bacteria in large bowel and modify the human gastrointestinal environment. In this study, compared with glucose (GLU) and high amylose maize starch (HAMS), the in vitro effects of LRS3 and P-LRS3 (RS3 and purified RS3 prepared from lotus seed starch) on the proliferation of bifidobacteria were assessed by assessing the changes in optical density (OD), pH values, short chain fatty acid (SCFA) production, and tolerance ability to gastrointestinal conditions. Significantly higher OD values were obtained from media containing LRS3 and P-LRS3, and especially in the medium containing P-LRS3, the OD value of which reached 1.36 when the concentration of the carbon source was 20 g L−1. Additionally, the lag phase of bifidobacteria was 8 h in the medium with LRS3 or P-LRS3, whereas it was 16 h in the medium with GLU or HAMS. What is more, a higher content of butyric acid was obtained in the P-LRS3 medium. Compared with GLU and HAMS media, bifidobacteria had a higher tolerance to gastrointestinal conditions in LRS3 and P-LRS3 media. It shows that lotus seed resistant starch, especially P-LRS3, could stimulate the growth of bifidobacteria. The rough surface of resistant starch and the SCFAs produced during fermentation might influence the proliferation of bifidobacteria.

Mutants E395D and S321D showed 1.6× and 1.3× higher catalytic efficiency than BlGBE. • S321D/E395D-modified pea starch had further reduced retrogradation and digestibility. • E395D/S321D increased DP 2 and 3 short branches, reduced R₁₀₄₇/₁₀₂₂ and crystallinity. • S321D-modified cake had further improved slowly digestible starch and reduced hardness. • S321D/E395D had enhanced substrate binding and stabilized transition-state by H-bonds. The 1,4-α-glucan branching enzyme found in Bifidobacterium longum can modify the physicochemical properties of starch by forming denser and shorter branches. To enhance its activity and modification function with regard to pea starch, ten mutants were engineered using site-directed mutagenesis based on multiple sequence alignment, molecular docking, and dynamic simulation. The mutants E395D, S321D and M509L exhibited specific activities 1.32-, 1.23- and 0.81-fold higher than the wild type (WT). The catalytic efficiencies of S321D and E395D were 18.36 and 14.00 mg −1 ·min −1 ·mL, respectively, which were significantly higher than the WT's efficiency of 11.19 mg −1 ·min −1 ·mL. Docking analysis revealed that the mutations reorganised the hydrogen-bonding and electrostatic microenvironment at the active site, producing mutation-specific effects on substrate binding and transition-state stabilisation. These locally altered polar and nucleophilic properties ultimately dictated the catalytic efficiency of each mutant. Compared with WT, E395D and S321D increased DP 2 and 3 short branches, reduced FT-IR R₁₀₄₇/₁₀₂₂ and XRD crystallinity at 200–800 U/g starch, which expanded the amorphous region of starch. After seven days' storage at 4 °C, the retrogradation enthalpy of S321D-modified starch was 0.63 J/g lower than that of WT-modified starch of 0.71 J/g. Glucose release from S321D-modified starch was significantly lower than from WT-modified starch at the level of small intestinal α-glucosidase. The slowly digestible fraction of S321D-modified starch was 38.51%, significantly higher than the 29.19% observed in WT-modified starch. Fresh and stored pea cake modified by S321D had significantly lower hardness and higher slowly digestible properties than the WT-modified equivalent. Background Recent studies suggest a role of gut microbiota in the development of neonatal hyperbilirubinemia, with bifidobacteria showing promise in alleviating symptoms. However, uncertainties persist regarding gut bifidobacterial species composition and their effects on bilirubin metabolism. Therefore, the study investigated the association between the gut microbiota and neonatal hyperbilirubinemia, assessing the potential and underlying mechanisms of Bifidobacterium in managing the condition. Results Compared to the high-risk group (requiring clinical intervention), low-risk neonates (designated as controls without therapeutic needed) demonstrated a higher abundance of breastfeeding-associated Bifidobacterium species, including Bifidobacterium longum subsp. infantis , B. bifidum , and B. breve . In an experimental neonatal hyperbilirubinemia rat model, these Bifidobacterium species’ protective effects were demonstrated by reducing serum bilirubin levels, maintaining growth and development, and improving neurobehavioral reflexes. These benefits are associated with improved liver and intestinal barrier functions and reduced enterohepatic circulation of bilirubin. Among the various treatment groups, B. longum subsp. infantis exhibited the most potent effect, followed by B. bifidum and B. breve . Intestinal metabolism analysis revealed that the levels of arachidonic acid and docosahexaenoic acid were increased and negatively correlated with bilirubin levels. These findings were further confirmed in the neonatal cohort study. Direct supplementation of these metabolites into the colon of neonatal rats improved disease phenotypes. In addition, both in vitro and in vivo experiments demonstrated the specific inhibitory effect of arachidonic acid and docosahexaenoic acid on β-glucuronidase enzyme activity. Notably, both in vitro intestinal fermentation models and genomic analyses demonstrated that the three Bifidobacterium species cannot synthesize these unsaturated fatty acids due to the lack of related genes. However, data suggested that these species might promote the accumulation of arachidonic acid and docosahexaenoic acid by regulating the gut microbiota’s structure and function. Moreover, these species show species-specific genomic distribution patterns, which might influence the gut microbiota's α-linolenic and linoleic acid metabolism pathways, thereby affecting bilirubin levels. Conclusions The results indicate that Bifidobacterium species associated with breastfeeding can prevent neonatal hyperbilirubinemia by regulating the gut microbiota–α-linolenic and linoleic acid metabolism–enterohepatic circulation axis, increasing gut metabolites arachidonic acid and docosahexaenoic acid, and reducing the enterohepatic circulation of bilirubin.

In this study, the commercial bacteria Danisco and Bifidobacterium lactis were used to ferment soy yogurt, and then the quality of yogurt and the number of active probiotics in yogurt during storage were investigated. The results showed that the total number of viable bacteria in soy yogurt increased first and then decreased, but all of them met the standard for the number of viable bacteria in probiotic foods. The content of protein, lipid, and total sugar in soy yogurt decreased gradually with the extension of storage time. The texture, water holding capacity, and rheological properties of soy yogurt were improved within 0–10 days, and there was no significant change after 15 days. However, brightness and whiteness of yogurt were significantly reduced. Based on realizing the reuse of soy whey, this study provided a theoretical basis for the research of the shelf life of soy yogurt. Practical Application This study developed a soy yogurt with good quality and provided a theoretical basis for the study of the shelf life of soy yogurt. In addition, some technical support was provided for the reuse of soy whey.

Inflammatory bowel disease (IBD) is a chronic intestinal disease characterized by microbiota disturbance and intestinal mucosal damage. The current study aimed to investigate the preventive effects of Bifidobacterium bifidum BD-1 (BD-1) against long-term IBD and possible mechanism by which it alters the gut microbiota, immune response, and mucosal barrier. Our study found that early treatment of BD-1 + Ceftri (ceftriaxone followed by BD-1) and BD-1 confers a certain protective effect against the occurrence of long-term Dextran sulfate sodium-induced colitis, which manifests as a decrease in inflammation scores and MPO activity levels, as well as a relatively intact intestinal epithelial structure. Moreover, compared to BD-1, Ceftri, and NS, early treatment with BD-1 + Ceftri promoted greater expression levels of mucosal barrier-related proteins [KI67, MUC2, ZO-1, secretory immunoglobulin A (slgA), Clauding-1, and Occludin], better local immune responses activation, and moderately better modulation of systemic immune responses during long-term colitis. This may be due to the fact that BD-1 + Ceftri can deliberately prolong the colonization time of some beneficial microbiota (e.g., Bifidobacterium ) and reduce the relative abundance of inflammation-related microbiota (e.g., Escherichia/Shigella and Ruminococcus ). Interestingly, we found that the changes in the gut barrier and immunity were already present immediately after early intervention with BD-1 + Ceftri, implying that early effects can persist with appropriate intervention. Furthermore, intervention with BD-1 alone in early life confers an anti-inflammatory effect to a certain degree in the long-term, which may be due to the interaction between BD-1 and the host’s native gut microbiota affecting intestinal metabolites. In conclusion, BD-1 was not as effective as BD-1 + Ceftri in early life, perhaps due to its failure to fully play the role of the strain itself under the influence of the host’s complex microbiota. Therefore, further research is needed to explore specific mechanisms for single strain and native microbiota or the combination between probiotics and antibiotics.

Sialylated immunoglobulin G (IgG) is an important immunoglobulin in breast milk, but its effect on adult gut microbiota is not yet clear due to digestion by pepsin. Based on our previous IgG protecting study, effects of sialylated IgG on gut microbiota were investigated by in vitro anaerobic fermentation in the present study. It was found that the addition of sialylated IgG could significantly promote the growth of Bifidobacterium. Meanwhile, three bifidobacterial species B. bifidum CCX 19061, Bembidion breve CCX 19041, and B. longum subsp. infantis CCX 19042 were isolated. Furthermore, B. breve CCX 19041 and B. longum subsp. infantis CCX 19042 showed co-culture growth property with B. bifidum CCX 19061 in a sialylated IgG-based medium, which was also supported by changes of free monosaccharides and N-glycan structure. These findings suggest that the increase of Bifidobacterium in vitro fermentation is attributed to the commensal relationship of the three bifidobacterial species by utilizing sugars released from sialylated IgG.

Atherosclerosis is a major cause of cardiovascular disease (CVD). The trimethylamine (TMA)-trimethylamine N-oxide (TMAO) pathway is a key crossover pathway highly associated with diet, gut microbiome, and atherosclerosis. The Bifidobacterium animalis subsp. lactis F1-3–2 ( Bif. animalis F1-3–2, No. CCTCCM2020832) was screened through in vitro and in vivo experiments in the early stage of this study with excellent lipid-lowering and anti-inflammatory function. By building an atherosclerosis model and focusing on TMAO, the specific mechanism of Bif. animalis F1-3–2 to improve atherosclerosis was explored. The study found that Bif. animalis F1-3–2 effectively improved the accumulation of aortic plaque in atherosclerotic mice. The strain improved lipid metabolism in serum and liver. It decreased the serum TMA and TMAO, regulated bile acid composition, participated in the farnesoid X receptor (FXR) pathway to improve lipid metabolism, and further reduced the aortic macrophage foam cell accumulation. In addition, the strain could improve the structure of the intestinal microbiome and reduce the proportion of Firmicutes and Bacteroidetes. The abundance of Turicibacter , Clostridium sensu stricto_1 , and Romboutsia was reduced at the genus level. The differential microbiota is highly correlated with bile acid metabolism, which is speculated to be involved in ameliorating atherosclerotic lipid metabolism disorders. Graphical abstract

Atopic dermatitis (AD) is a prevalent
early childhood inflammatory
skin disease frequently linked to gut dysbiosis. While Bifidobacterium longum subsp. infantis is a pioneer infant gut colonizer driving immune maturation, its
role in AD prevention remains unclear. Here, we investigated early
life intervention with B. longum subsp. infantis CCFM1269 using a 2,4-dinitrofluorobenzene
(DNFB)-induced murine AD model. Oral administration of CCFM1269 significantly
ameliorated AD symptoms, reducing skin/ear thickness, dermatitis scores,
and serum IgE. Mechanistically, CCFM1269 modulated the systemic Th1/Th2
balance by downregulating TSLP, IL-4, and IL-13, while upregulating
IFN-γ. Notably, CCFM1269 produced indole-3-lactic acid (ILA).
Via the gut–skin axis, ILA activated the aryl hydrocarbon receptor
(AHR) and its target CYP1A1 in skin tissues, strengthening the epidermal
barrier. Additionally, AD mitigation was associated with gut microbiota
reshaping. Ultimately, CCFM1269 alleviates AD primarily through the
ILA- AHR signaling pathway, highlighting its potential as a functional
food ingredient for early life AD prevention.

The aim of this study was to investigate the proliferation effect of fractionated lotus seed resistant starch (LRS3-20%, LRS3-30%) and the structural changes following fermentation. Fractionated lotus seed resistant starch, especially LRS3-20%, promoted the proliferation of these probiotics, and Bifidobacterium longum produced large quantities of acetic acid, while Lactobacillus delbrueckii subsp. bulgaricus produced lactic acid. Additionally, the probiotics with lotus seed resistant starch (LRS3) and LRS3-20% survived more successfully in simulated gastrointestinal solutions compared to glucose, LRS3-30% and high amylose maize starch, but LRS3 and LRS3-20% differed in their abilities to enhance the tolerance to acidic buffer, bile acid and simulated gastric juice. Furthermore, after fermentation, fractionated lotus seed resistant starch displayed a loose, rough surface. The crystalline and double helical structure increased as the amount of amorphous and sub-crystalline structure reduced following fermentation. Interestingly, structural changes were correlated with the original structures and utilisation by the probiotics.

BACKGROUND Cyclophosphamide (CTX), a widely used alkylating chemotherapeutic agent, causes severe systemic immunosuppression and gut microbiota dysbiosis through non-specific cytotoxicity, increasing infection risk and impairing chemotherapy outcomes. Safe and effective adjuvant therapies for chemotherapy-related immune injury remain limited. This study evaluated the protective effect of compound AB probiotics ( Lactobacillus acidophilus LA15 combined with Bifidobacterium animalis subsp. lactis Bi66) against CTX-induced immunosuppression in mice and explored correlations associated with the gut-immune axis. RESULTS CTX challenge induced immune organ atrophy, cytokine imbalance, impaired innate and adaptive immunity, intestinal barrier disruption, reduced short-chain fatty acid (SCFA) synthesis and gut dysbiosis. AB probiotic intervention alleviated these injuries dose-dependently: it restored thymus and spleen indexes, rebalanced serum cytokines, improved core immune functions, and repaired ileal mucosal pathological damage. Furthermore, probiotic treatment was accompanied by elevated fecal SCFA concentrations, upregulated tight junction gene expression, recovered aryl hydrocarbon receptor (AhR)/ERK/STAT3 transcription, enriched beneficial bacterial genera, and suppressed opportunistic pathogens. CONCLUSION Compound AB probiotics alleviate CTX-induced immunosuppression in mice, and this effect correlates with restored gut microecology, enhanced SCFA metabolism, repaired intestinal barrier, and activated AhR signaling. These findings provide evidence for developing probiotic preparations as adjuvant interventions against chemotherapy-related immunosuppression. © 2026 Society of Chemical Industry.

ABSTRACT Probiotics and synbiotics can mitigate the negative health consequences of early-life antibiotic exposure. This study aimed to determine whether supplementation with Bifidobacterium longum ssp. infantis 79 (B79) or synbiotics composed of B79 and 2′-fucosyllactose (2′-FL) could mitigate the negative impact of ceftriaxone exposure in early-life. We found that antibiotic-treated mice exhibited lower body weight, crypt depth, short-chain fatty acid content, and α-diversity indices at weaning, while they increased the relative abundance of opportunistic pathogens (such as Enterococcus and Staphylococcus ) and decreased the relative abundance of intestinal commensal bacteria. Supplementation with B79 and 2′-FL revived these antibiotic-induced negative effects and reduced the mRNA expression of IL-6, IL-12p40, and TNF-α in the spleen at weaning. Moreover, B79 and 2′-FL supplementation persistently improved crypt depth, propionic acid synthesis, and IgG and sIgA production and revived the gut microbiota structure and composition in adulthood. In conclusion, our study suggests that early-life supplementation with B79 alone or in combination with 2′-FL can mitigate ceftriaxone-induced negative effects on the gut microbiota and intestinal and immune development of mice, and these improvements can partially last into adulthood.

Purpose Early-life gut microbiota establishment critically influences long-term host health. This study investigated whether maternal Bifidobacterium bifidum BD-1 supplementation during late gestation, combined with delivery mode, affects offspring responses to DSS-induced acute colitis. Methods Pregnant mice received B. bifidum BD-1 (1×10^9 CFU/day), ceftriaxone (100 mg/kg), or saline by gavage from gestational day 14 to 21. Offspring were delivered vaginally (VD) or by cesarean section (CS). Acute DSS-induced colitis was induced with 3% dextran sulfate sodium at postnatal day 42. Results VD offspring showed relatively stable microbial succession and comparatively limited variation in DSS-induced colitis-related outcomes across maternal treatment groups. In CS offspring, maternal BD-1 supplementation was associated with enrichment of beneficial genera, maintenance of α-diversity, higher fecal butyrate and propionate levels, increased expression of tight junction-related genes, and lower histopathological inflammatory scores compared with NS-CS-DSS offspring. Conclusion Maternal B. bifidum BD-1 supplementation was associated with lower indices of acute DSS-induced colitis, with more pronounced effects observed in CS offspring, supporting its potential as a prenatal probiotic candidate requiring further validation.
