Photothermal effect produced in photothermal therapy (PTT) can not only eradicate tumors but also allow the promotion of biological and chemical reactions in tumor tissues, leading to the potential of photothermal-enhanced anti-tumor therapy with desirable outcomes. Biodegradable polydopamine (PDA) nanoparticles exhibit remarkable photothermal conversion efficiency and stability, making them ideal photothermal agents (PTAs) to achieve photothermal-enhanced anti-tumor therapy. The high affinity of PDA to metal ions provides a broad application prospect for the development of multi-functional nanoplatforms to achieve cancer therapy with high efficacy and specificity. In this work, we fabricated Cu(II)-doped mesoporous PDA nanoparticles with polyethylene glycol (PEG) modified (CMP-P), which not only possessed higher photothermal conversion efficacy than mesoporous PDA nanoparticles , but also presented distinctive self-enhanced chemodynamic (CDT) performance. CMP-P mediated CDT was predicated on catalyzing the Fenton-like reaction to generate hydroxyl radicals (·OH). Impressively, CMP-P further triggered glutathione (GSH) depletion intracellular to prevent the consumption of ·OH, and the photothermal effect further improved ·OH generation. Meanwhile, CMP-P exhibited favorable degradability, the process of which was elaborated intuitively by transmission electron microscope (TEM). CMP-P was then employed for doxorubicin (DOX) loading, and enabled the enhanced DOX release in-situ under the stimulation of photothermal effect and tumor micro-environment (TME). The lower IC 50 value of DOX/CMP-P (1.68 μg/mL) than the signal therapy suggested the superior synergistic effect of DOX/CMP-P. Meanwhile, in vivo experiments also revealed the considerably improved anti-tumor efficacy of synergistic therapy mediated by DOX/CMP-P.

Chemodynamic therapy (CDT) is a new cancer treatment that uses low-valence transition metal ions to catalyze intracellular Fenton/Fenton-like reactions, producing hydroxyl radicals (•OH) to eliminate cancer cells. However, achieving satisfactory therapeutic outcomes with CDT alone can be challenging. In this work, we reported an anticancer drug delivery DOX@HMDN-TQDs@PEI-PEG (DMTP) based on hollow mesoporous manganese dioxide nanoparticle (HMDN), which has the potential to improve cancer therapy by combining synergistic therapeutic and tumor magnetic resonance imaging (MRI) capability. After loading the anticancer drug doxorubicin hydrochloride (DOX), the pores of HMDN were sealed with polyethyleneimine (PEI) modified Ti 3 C 2 T x MXene quantum dots (TQDs) and finally modified with polyethylene glycol (PEG) to obtain the final system. HMDN depletes glutathione (GSH), which is harmful to CDT, in cancer cells, and generates Mn 2+ that can be used for both CDT and MRI. TQDs can also be used for highly effective photothermal therapy (PTT) that enhances CDT. The combination of DOX with chemotherapy can significantly inhibit tumor growth. In addition, DMTP combines the capability of tumor MRI as Mn 2+ is also a T 1 -weighted contrast agent . This multifunctional delivery system provides new ideas for research related to tumor diagnosis and treatment.

The simultaneous inhibition of tumor recurrence and promotion of wound healing are critical for patients undergoing melanoma surgery. However, the development of an effective yet straightforward platform that meets the comprehensive therapeutic requirements of clinical melanoma postoperative care still remains a significant challenge. Herein, a 2D semiconductive MnPSe 3 nanosheets (MPS NSs)-engineered sodium alginate (SA) hydrogel dressing (SA-MPS) is developed. The MPS NSs, a member of the ternary transition metal phosphorous chalcogenides (TMPCs), are successfully prepared through an ultrasonic-assisted liquid phase exfoliation approach from bulk MnPSe 3 . These nanosheets exhibit the integrative functions of sonosensitizers, Fenton-like catalysts, glutathione (GSH) scavengers, and phosphorus (P)/selenium (Se)-containing nutrients. In vitro and in vivo evaluations demonstrate that the SA-MPS hydrogel dressing possesses excellent biocompatibility and is effective for combined sonodynamic/chemodynamic therapy (SDT/CDT) of tumors, with amplified reactive oxygen species (ROS) production. Additionally, it promotes skin regeneration in postoperative wounds by supplying bioactive P/Se elements. Hence, such a multifunctional SA-MPS hydrogel dressing without complex integration offers great potential for suppressing postoperative melanoma recurrence while facilitating wound healing, making it a promising candidate for melanoma postoperative care.

As a typical tumor microenvironment-responsive therapy, chemodynamic therapy (CDT), producing hydroxyl radicals ( • OH) to eliminate tumor cells, has demonstrated great promise. Nevertheless, there are still major challenges: • OH generated from endogenous H 2 O 2 is usually insufficient; the CDT effect is strongly dependent on the pre-reaction with glutathione. Addressing the challenges, Au@MnSe 2 core–shell nanoagent for synergetic chemodynamic-photothermo-photocatalytic therapy combined with tetramodal imaging, including magnetic resonance imaging, computed tomography, photoacoustic, and infrared thermal imaging is reported. Distinct from the reported glutathione-depleting agents, Mn 2+ in MnSe 2 allows immediate generation of • OH, independent of pre-reaction. Meanwhile, Mn 3+ consumes glutathione by its conversion to Mn 2+ . The Au-MnSe 2 combination promotes photothermal conversion and photocatalytic reaction, resulting in largely enhanced • OH generation from endogenous H 2 O 2 and significant hyperthermia. Meanwhile, immune response is effectively activated: the intratumoral expression of programmed cell death-1 and proinflammatory cytokines increase to 4–7 folds; the cytotoxic and helper T lymphocytes cells in the tumor area increase to more than 2.5-folds; an evident, temporary systemic immunostimulatory effect is demonstrated. High tumor inhibition rate (≈97.3%) and greatly prolonged survival are obtained. This highly-integrated design coordinating three different therapies with four different imaging modals provide new possibilities for high-performance theranostic nanoagents.

Oral squamous cell carcinoma (OSCC) is the most common malignant tumor in the head and neck region, and its treatment is limited by hypoxia and inadequate oxygen supply. Continuous oxygen delivery combined with photodynamic therapy (PDT) is the key to addressing this issue. Here, a dual-enzyme activity sea urchin-like Au@Pt-Ce6-HN-1 nanoplatform was designed to serve as an "oxygen pump" to alleviate tumor hypoxia for synergistic photodynamic/photothermal therapy (PTT). In this design, the photosensitizer chlorin e6 (Ce6) is covalently linked to the Au@Pt nanozyme for PDT treatment. The Au@Pt nanozyme exhibits catalase-like activity, continuously decomposing H 2 O 2 in the tumor microenvironment to enhance O 2 levels, thereby achieving efficient PDT. Furthermore, Au@Pt can perform PTT and increase oxygen levels under NIR-II light to further promote PDT. The Au@Pt nanozyme also exhibits peroxidase-like activity, generating ·OH for chemodynamic therapy (CDT). Additionally, HN-1 guides the direction of "sniping" OSCC, and its high specificity benefits Au@Pt-Ce6-HN-1 at the tumor site. Au@Pt-Ce6-HN-1 exhibits bright fluorescence (FL), strong CT signal, and photothermal imaging capabilities, laying the foundation for subsequent guided PDT/PTT. This nanoplatform, which combines advantages such as continuous oxygen production, tumor targeting, and multimodal imaging, is expected to provide valuable insights into the treatment of OSCC. Statement of Significance Accurate clinical diagnosis and treatment of OSCC are challenging. We report a dual-enzyme activity sea urchin-like Au@Pt-Ce6-HN-1 nanoplatform, serving as an "oxygen pump" to guide photodynamic therapy (PDT) and photothermal therapy (PTT) for OSCC. This nanoplatform targets OSCC for preoperative CT diagnosis and offers fluorescence visualization for surgical navigation, demonstrating potential in clinical cancer detection and surgery guidance. This innovative approach addresses OSCC hypoxia and enhances treatment efficacy through continuous oxygen production, tumor targeting, and multimodal imaging, significantly improving patient outcomes in OSCC treatment.

Combining the internal force-driven chemodynamic therapy (CDT) and the external energy-triggered photodynamic therapy (PDT) holds great promise to achieve an advanced anticancer effect based on reactive oxygen species (ROS). However, the insufficient oxy-substrates supply in tumor microenvironment, like hydrogen peroxide (H 2 O 2 ) and oxygen (O 2 ), is the Achilles heel that greatly restricts the efficacy of this ROS-based treatment. Herein, the construction of a c opper p e roxide-based tumor pH-re s ponsive a utocatalytic nano r eactor (CESAR), via an albumin-mediated biomimetic mineralization strategy is described. The decoration of human serum albumin endows the nanoreactor good hydrophilicity and biocompatibility, which is highly desired for the metal-based materials. Upon exposure to acidic tumor microenvironment, CESAR presents a pH-triggered disintegration with Cu 2+ , H 2 O 2 and O 2 generated instantly. The generated H 2 O 2 complements the hyperoxide deficiency and initiates a localized Fenton-like reaction with the assistance of Cu 2+ for highly toxic hydroxyl radicals (•OH) production for improving CDT. The evolved O 2 gas enables hypoxia relief for enhanced Ce6-mediated PDT. This H 2 O 2 /O 2 self-supplying strategy significantly amplifies the tumor oxidative damage and gains an optimal treatment outcome, which offers a new paradigm for optimizing the tumor therapeutic options limited by oxide or hyperoxide deficiency, not only for CDT/PDT, but also other oxy-substrates involved strategies. Statement of significance The shortage of oxy-substrates in the tumor microenvironment remains a great challenge for ROS-based cancer therapy. Herein, we introduce human serum albumin as a scaffold to stabilize copper peroxide nanomaterials for constant production of H 2 O 2 and O 2 to enhance chemodynamic/photodynamic therapy. The tumor pH-triggered H 2 O 2 /O 2 production and Cu 2+ release are confirmed, assuring the strategy of a highly precise, effective way to destroy tumor without any side effects. This work lends new and exciting insights into the engineering design of autocatalytic oxy-substrates self-supply nanoreactor for overcoming the bottlenecks, like the oxy-substrates deficiency of CDT/PDT and the poor stability of metal peroxides, to achieve highly effective chemodynamic/photodynamic therapy.

Chemodynamic therapy (CDT) and photothermal therapy (PTT) have been widely studied as treatment alternatives to anbiotics to avoid the occurrence of bacterial resistance because of their negligible bacterial resistance and high antibacterial efficiencies. Herein, we developed an infection microenvironment-responsive nanogel (STF) to successfully achieve a PTT synergized CDT for bacterial infection. Through a one-step/pot method, STF nanotherapeutics consisting of Fe3+/sodium alginate (SA) based nanogels and Fe3+/tannic acid (TA) based metal–phenolic networks (MPNs) were conveniently prepared by the coordination-triggered simultaneous self-assembly of SA, TA, and Fe3+ ions. Afterward, their photothermal and catalytic performances were intensively investigated. Moreover, the sterilization effect of the STF nanogel was evaluated through in vitro and in vivo bactericidal assays. The STF nanogel exhibited a photothermal enhanced peroxidase-like activity because of the good photothermal effect of MPNs. In addition, its Fenton activity was further accelerated by an extra amount of Fe2+ derived from the degradation of Fe3+-chelating agents (SA) and reduction of TA. Together, the STF nanogel possessed a photothermal/Fe2+ dual-augmented catalytic property. In vitro and in vivo antibacterial assays proved its satisfactory bactericidal efficiency owing to PTT/Fe2+ dual-enhanced CDT. Our research findings suggested that the developed intelligent bactericidal nanogel has great potential to be used as an antibiotic alternative to avoid bacterial resistance in future clinical treatments.

Single-atom catalysts (SACs), characterized by atomically dispersed single metal atoms, provide superior antibacterial properties over common nanozyme materials due to their satisfactory catalytic activity. However, when used for the treatment of biofilm-infected wounds, SACs are still unable to effectively solve severe microbial infection due to the insufficient supply of reactive oxygen species (ROS). Herein, a new hybrid nanozymes consisting of Zn single-atom catalysts and copper peroxide (Zn SACs@CuO 2 ) were prepared to realize the upgrading of ROS generation and the combination of photothermal/chemodynamic (PTT/CDT) therapy for biofilm-infected wound. Specifically, CuO 2 exhibited not only excellent photothermal property, but also H 2 O 2 -producing and Fenton-like catalytic activities, which assisted Zn SACs to accelerate the ·OH conversion of endogenous and exogenous H 2 O 2 . Compared with single antimicrobial strategies of Zn SACs, Zn SACs@CuO 2 hybrid nanozymes exhibited stronger therapeutic effects of biofilm destruction, immune activation and inflammation improvement. In conclusion, this work developed a hybrid single atom nanozyme material with for efficient biofilm-infected wound healing.

Chemodynamic therapy (CDT) is an emerging strategy for cancer treatment based on Fenton chemistry, which can convert endogenous H2O2 into toxic ·OH. However, the limited endocytosis of passive CDT nanoagents with low penetrating capability resulted in unsatisfactory anticancer efficacy. Herein, we propose the successful fabrication of a self-propelled biodegradable nanomotor system based on hollow MnO2 nanoparticles with catalytic activity for active Fenton-like Mn2+ delivery and enhanced CDT. Compared with the passive counterparts, the significantly improved penetration of nanomotors with enhanced diffusion is demonstrated in both the 2D cell culture system and 3D tumor multicellular spheroids. After the intracellular uptake of nanomotors, toxic Fenton-like Mn2+ is massively produced by consuming overexpressed intracellular glutathione (GSH), which has a strong scavenging effect on ·OH, thereby leading to enhanced cancer CDT. The as-developed MnO2-based nanomotor system with enhanced penetration and endogenous GSH scavenging capability shows much promise as a potential platform for cancer treatment in the near future.

The nanoscale multidrug codelivery system for synergistic therapy is an effective strategy for tumor treatment. However, the low drug delivery efficiency and poor therapeutic effects limit its application. Here, based on the coordination effect of Artemisinin (Art), quercetin (Qc), and Fe3+, we had constructed a safe and efficient carrier-free hyaluronic acid (HA)-modified Art–Fe–Qc nanoparticles (AFQ@HA NPs) for enhanced chemotherapy/photothermal therapy (PTT)–chemodynamic therapy (CDT) synergistic therapy, which achieved an ultrahigh drug loading efficiency and a multifunction anticancer strategy. The results showed that high drug loading was achieved based on drug coordination self-assembly, with Art and Qc contents of 38.6 and 42.7%, respectively. At the same time, based on the Qc–Fe coordination molecular network, the system had excellent photothermal conversion performance with an efficiency of 57.3% and could effectively inhibit the expression of HSP70, achieving enhanced PTT. Further, under the stimulation of excessive H2O2 and glutathione (GSH) in the tumor microenvironment, the AFQ@HA NPs were continuously degraded, while releasing Art and Fe3+/Fe2+ to achieve iron ion-enhanced CDT. The results of in vitro and in vivo experiments showed that AFQ@HA NPs could achieve chemotherapy–PTT–CDT synergistic therapy in response to tumor microenvironment by passively targeting and actively targeting tumor cells with CD44, demonstrating its excellent targeted antitumor effects.

Cisplatin (cis-diammine-dichloro-platinum II, CDDP) is one of the most commonly used chemotherapeutic agents for the treatment of esophageal cancer, and its clinical application has been greatly limited due to non-specific administration, severe systemic toxicity, and reduced efficacy due to drug resistance. The special physicochemical properties of hydrogels enable them to achieve a sustained controlled release of CDDP and to maintain high local drug concentrations over a long period, but the problem of drug resistance still needs to be addressed. Herein, CS-BA-PVA, a pH-responsive hydrogel is used to co-load CDDP and divalent copper ion (Cu 2+ ) for constructing a drug delivery system CS-BA-PVA@Cu/CDDP to achieve controlled and sustained drug release. Once reaching the acidic tumor microenvironment (TME), CS-BA-PVA@Cu/CDDP specifically responds and disassembles to release CDDP for chemotherapy against cancer. Besides, the simultaneous release of Cu 2+ can deplete the intracellular glutathione (GSH) for increased toxicity of CDDP as well as catalyzing the decomposition of intra-tumoral H 2 O 2 into highly toxic •OH for chemodynamic therapy (CDT). Moreover, the substantially reduced GSH can also protect the yielded •OH from scavenging and thus greatly improve the •OH-based CDT effect. In addition to providing a new drug delivery system CS-BA-PVA@Cu/CDDP, this study is also expected to establish a new form of TME-responsive carrier for highly efficient tumor-specific GSH-depletion-enhanced synergistic chemotherapy/chemodynamic therapy.

Ferroptosis induced by iron-dependent accumulation of lipid peroxides (LPOs) has received increasing attention in cancer therapy, especially chemodynamic therapy (CDT). However, the quick annihilation of hydroxyl radicals (˙OH) severely restricts the ˙OH/LPO conversion efficiency, which has become one of the key factors that influences the therapeutic efficacy of ferroptosis-based CDT. Herein, we designed a ˙OH/LPO nano-converter with a high LPO generation efficiency via loading ferrocene (Fc), a green Fenton catalyst, in the phospholipid bilayer of liposome-PEG (Fc-Lp-PEG). Under catalysis with Fc, the over-expressed H2O2 in tumors can be decomposed to ˙OH. The generated ˙OH in situ reacts with unsaturated lipids on the liposome, and is converted into LPOs, which spread the lipid peroxidation chain reaction to the remote membranes of cells and organelles, triggering efficient cancer cell ferroptosis. Systematic in vitro and in vivo therapeutic outcomes showed the high tumor inhibition ratio (74.0%) and the low side effects of Fc-Lp-PEG on 4T1 tumor-bearing mice. This novel strategy for improving the ˙OH/LPO conversion efficiency might provide new insights for the clinical development of ferroptosis-based CDT.
