This study presents a novel multifunctional hydrogel synthesized by crosslinking Chlorella protein using 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide/n-hydroxysuccinimide, aimed at extending the shelf life of perishable fruits like cherries. Structural, rheological, and scanning electron microscopy analyses revealed that Chlorella protein hydrogels (CPH) possess excellent solid-like properties and a stable porous structure. The water-holding capacity improved significantly from 67.11 ± 0.72% to 96.53 ± 0.61% with increasing CP concentration (10–22.5%, w/v). Additionally, CPH decomposition temperatures were ~ 150 °C (5% weight loss), demonstrating good thermal stability. Due to the ionization of -COOH and -NH 2 groups, the CPH showed excellent pH sensitivity, with low dissolution rates in acidic environments (64.97%) and significantly higher rates in alkaline environments (448.50%). Furthermore, the CPH inhibited the penetration of Staphylococcus aureus and Escherichia coli , and exhibited good free radical scavenging abilities against DPPH (74.50%) and ABTS •+ (97.92%). In cherries preservation tests, CPH extended preservation time to 15 days compared to 5 days in the control group, effectively inhibiting decay, suggesting CPH is a promising choice for multifunctional fruit preservation. Perfluorobutanoic acid (PFBA), an emerging alternative to perfluorooctanoic acid (PFOA), has become increasingly prevalent in aquatic ecosystems, yet its ecotoxicological impacts remain poorly understood. This study investigated the aquatic toxicity of PFBA using the freshwater algae Chlorella vulgaris ( C . vulgaris ) as a model organism, employing a 96h pre-exposure assay to determine the median effective concentration followed by acute toxicity experiments analyzing multiple endpoints including growth, photosynthetic parameters, oxidative stress markers, and antioxidant enzyme activities. Computer simulation techniques were utilized to illustrate the underlying molecular mechanisms of PFBA toxicity. The results showed that the 96h-EC 50 value of PFBA was 154.88 mg/L, which is comparable to conventional per- and polyfluoroalkyl substances (PFAS). Acute toxicity experiments revealed a biphasic dose-response relationship to the algal growth with the hormetic effects at the lower concentrations (30.97–92.93 mg/L) but inhibition at the higher levels (123.91–185.86 mg/L) of PFBA. High dosages of PFBA significantly decreased the maximum photosynthetic yield (Fv/Fm) and relative electron transfer rate (rETR), while inducing oxidative stress and inhibiting superoxide dismutase (SOD) and catalase (CAT) activities. Future AlphaFold2 modeling and molecular docking simulations demonstrated the potential binding of PFBA to photosystem II D1 C-terminal processing protease (PSII D1 protein), SOD, and CAT. These findings reveal a complex toxicity mechanism of PFBA on C . vulgaris involving photosynthetic disruption, oxidative stress, and antioxidant system impairment, contributing to the understanding of short-chain PFAS alternative ecotoxicity in aquatic ecosystems.

This comparative study investigates the physicochemical, functional, and nutraceutical properties of Spirulina ( Arthrospira platensis ) and Chlorella sp. biomass, two of the most prevalent microalgae species. We assessed their proximate composition, pH, acidity, color, granulometry, microstructure (SEM), functional groups (FTIR), crystallinity (XRD), and thermal stability (GTA). Functional properties were thoroughly examined, including bulk density, water and oil holding capacities, and gelling, foaming, and emulsifying capabilities. Additionally, the study quantified the content of pigments, phenolic compounds , carotenoids , and flavonoids and evaluated antioxidant activity . Our findings indicate that Spirulina biomass exhibits higher protein and ash content, whereas Chlorella biomass surpasses phenolic compounds, carotenoids, and antioxidant activity. Notable differences were observed in their microstructure and thermal degradation patterns. Both biomasses demonstrated considerable potential as functional ingredients, owing to their superior water and oil retention, foaming, emulsifying, and gelling properties. This investigation underscores the distinct physicochemical and nutraceutical profiles of Spirulina and Chlorella biomass, highlighting their significant applicability in the food industry for developing novel value-added food products.

Bioconversion of food waste into value-added products is a promising way to tackle the global food waste management problem. In this study, a novel valorisation strategy for bioenergy and lutein production via microalgal fermentation was investigated. Significant amount of glucose was recovered from enzymatic hydrolysis of food waste. The resultant hydrolysate was then utilised as culture medium in mixotrophic cultivation of Chlorella sp. to obtain high levels of lipid and lutein, whose accumulation patterns were consistent with molecular analyses. The resultant algal lipid derived from microalgal biomass using food hydrolysate was at high quality in terms of biodiesel properties. Further, in semi-continuous fermentation, the average algal biomass was 6.1 g L −1 with 2.5 g L −1 lipid and 38.5 mg L −1 lutein using hydrolysate with an initial glucose concentration of 10 g L −1 . Meanwhile, the resultant algal biomass was 6.9 g L −1 with 1.8 g L −1 lipid and 63.0 mg L −1 lutein using hydrolysate with an initial glucose concentration of 20 g L −1 , which suggests food waste hydrolysate could trigger algal products preferences. The experimental results of this study suggested the potential of microalgae as a platform for bioconversion of food waste into high-value products, especially sustainable bioenergy.

Roxithromycin (ROX) has received increasing concern due to its large usage, ubiquitous detection in environment and high ecotoxicology risk. This study investigated the acute and chronic effects of ROX on the growth, chlorophyll, antioxidant enzymes, and malonaldehyde (MDA) content of Chlorella pyrenoidosa , as well as the removal mechanism of ROX during microalgae cultivation. The calculated 96 h median effective concentration of ROX on yield (E y C 50 ) and specific growth rate (E r C 50 ) of C. pyrenoidosa was 0.81 and 2.87 mg/L, respectively. After 96 h exposure, 1.0 ~ 2.0 mg/L of ROX significantly inhibited the synthesis of chlorophyll and promoted the activities of SOD and CAT ( p < 0.05). The MDA content increased with the ROX concentration increasing from 0.5 ~ 1.0 mg/L, and then decreased to 105.76% of the control exposure to 2.0 mg/L ROX, demonstrating the oxidative damage could be moderated by the upregulation of SOD and CAT activities. During the 21 d chronic exposure, low concentration of ROX (0.1 and 0.25 mg/L) showed no significant effect on the growth and chlorophyll content of algae during the first 14 d, but significantly inhibited the growth of algae and the synthesis of chlorophyll at 21 d ( p < 0.05 or p < 0.01). 1.0 mg/L ROX significantly inhibited the growth of microalgae during 3 ~ 21 d and the synthesis of chlorophyll at 7 ~ 21 d. High concentration and long-term exposure of low concentration of ROX caused the SOD and CAT activities and MDA content to increase, demonstrating a higher level of oxidative damage of microalgae. During the first 14 d, abiotic removal of ROX played a more important role, contributing about 12.21% ~ 21.37% of ROX removal. After 14 d, the biodegradation of ROX by C. pyrenoidosa gradually became a more important removal mechanism, contributing about 45.99% ~ 53.30% of ROX removal at 21 d. Bio-adsorption and bioaccumulation both played minor roles in the removal of ROX during algae cultivation.

Although the addition of triclosan (TCS) in consumer products has been strictly restricted, its continuous applications in hospitals and other medical facilities and its numerous residues still pose a potential risk to aquatic organisms and aquatic ecosystems. In this study, we investigated the growth, biochemical alterations, and physiological responses of Chlorella vulgaris exposed to different concentrations of TCS. The potential toxicity mechanisms associated with excessive production of reactive oxygen species (ROS) and disruption of photosynthetic system II (PSII) were also analyzed. The results indicated that the growth, cellular ultrastructure, and physiology of C. vulgaris were severely affected by TCS in a dose-effect dependent manner. TCS inhibited the growth of C. vulgaris , leading to mitochondria enlargement, the disordering of the arrangement of thylakoids, cell wall rupture, organelles loss, and the cytoplasm lysis. TCS induced severe oxidative damage characterized by ROS accumulation, elevated malondialdehyde (MDA), and up-regulation of antioxidant enzyme activities. Moreover, in TCS-induced algal cells, the main sites of ROS accumulation were chloroplasts, mitochondria, and cell membranes, with ROS accumulating most in the mitochondria. In addition, TCS caused damage to the reaction center (RC inactivation), donor side (OEC damage), and accepted side (electron transport from Q A to Q B ) of PSII in C. vulgaris , leading to inhibition of photosynthetic activity. These results could provide novel insights into the mechanisms of TCS-induced ROS accumulation and photosynthetic inhibition in C. vulgaris , which would contribute to a deep understanding of TCS toxicity on algae.

Microalgae, which accumulate considerable carbohydrates, are a potential source of glucose for biofuel fermentation. In this study, we investigated the enzymatic hydrolysis efficiency of wet microalgal biomass compared with freeze-dried and oven-dried biomasses, both with and without an acidic pretreatment. With the dilute sulfuric acid pretreatment followed by amy (α-amylase and amyloglucosidase) and cellulase hydrolysis, approximately 95.4% of the glucose was recovered; however, 88.5% was released by the pretreatment with 2% (w/v) sulfuric acid, which indicates the potential of the acids for direct saccharification process. There were no considerable differences in the glucose yields among the three kinds of materials. In the direct amy hydrolysis without any pretreatment, a 78.7% glucose yield was obtained, and the addition of cellulase had no significant effect on the hydrolysis to glucose. Compared with the oven-dried biomass, the wet biomass produced a substantially higher glucose yield, which is possibly because the cross-linked cells of the oven-dried biomass prevented the accessibility of the enzymes. According to the results, the fresh microalgal biomass without cell disruption can be directly used for enzymatic hydrolysis to produce glucose. The enzymatic hydrolysate of the wet microalgal biomass was successfully used for acetone–butanol–ethanol (ABE) fermentation, which produced 7.2 g/L of ABE, indicating the application potential of wet microalgae in the bioalcohol fuel fermentation process. Keywords:wet microalgae;pretreatment;enzymatic hydrolysis;glucose

Salinity stress in estuarine environments poses a significant challenge for microalgal survival and proliferation. The interaction between microalgae and bacteria shows promise in alleviating the detrimental impacts of salinity stress on microalgae. Our study investigates this interaction by co-cultivating Chlorella sorokiniana , a freshwater microalga, with a marine growth-promoting bacterium Pseudomonas gessardii , both of which were isolated from estuary. In this study, bacteria were encapsulated using sodium alginate microspheres to establish an isolated co-culture system, preventing direct exposure between microalgae and bacteria. We evaluated microalgal responses to different salinities (5 PSU, 15 PSU) and interaction modes (free-living, gel-encapsulated), focusing on growth, photosynthesis, cellular metabolism, and extracellular polymeric substances (EPS) properties. High salinity inhibited microalgal proliferation, while gel-fixed interaction boosted Chlorella growth rate by 50.7 %. Both attached and free-living bacteria restored Chlorella 's NPQ to normal levels under salt stress. Microalgae in the free-living interaction group exhibited a significantly lower respiratory rate compared to the pure algae group (−17.2 %). Increased salinity led to enhanced EPS polysaccharide secretion by microalgae, particularly in interaction groups (19.7 %). Both salt stress and interaction increased the proportion of aromatic proteins in microalgae's EPS, enhancing its stability by modulating EPS glycosidic bond C-O-C and protein vibrations. This alteration caused microalgal cells to aggregate, free-living bacteria co-culture group, and fixed co-culture group increasing by 427.5 %, 567.1 %, and 704.1 %, respectively. In gel-fixed bacteria groups, reduced neutral lipids don't accumulate starch instead, carbon redirects to cellular growth, aiding salt stress mitigation. These synergistic activities between salinity and bacterial interactions are vital in mitigating salinity stress, improving the resilience and growth of microalgae in saline conditions. Our research sheds light on the mechanisms of microalgal-bacterial interactions in coping with salt stress, offering insights into the response of estuarine microorganisms to global environmental changes and their ecological stability.

The combined effect of polystyrene (PS) particles and triphenyltin chloride (TPTCl) to the green algae Chlorella pyrenoido s a was studied. The 96 h IC 50 of TPTCl to the green algae C. pyrenoidosa was 30.64 μg/L. The toxicity of PS particles to C. pyrenoidosa was size-dependent, with the 96 h IC 50 at 9.10 mg/L for 0.55 μm PS but no toxicity observed for 5.0 μm PS. The exposure to 0.55 μm PS led to damage on structure of algal cells, which could in turn cause inhibition on photosynthesis and population growth of the green algae. TPTCl concentrations in test medium were lowered by 15–19% at presence of 0.55 μm PS particles, indicating a reduced bioavailability of TPTCl. In spite of this reduced bioavailability, the presence of PS increased the toxicity of TPTCl, which might be attributed to facilitated uptake of TPTCl by the green algae after the damage of cell structure. The overall results of the present study provided important information on the effect of PS on the bioavailability and toxicity of TPTCl to phytoplankton species.

Particulate emerging contaminants (PECs) pose increasing ecological risks due to their widespread occurrence and complex environmental behaviors, yet their heterogeneous toxic mechanisms remain poorly understood, especially under environmentally relevant conditions and concentration gradients. Here,Chlorella vulgariswas used as a model organism to systematically compare the effects of polystyrene nanoparticles (PSNPs), silver nanoparticles (AgNPs), and titanium dioxide nanoparticles (TiO2NPs) across environmentally relevant and elevated concentrations (100 μg/L and 10 mg/L). Distinct toxicity pathways were identified among PEC types. PSNPs primarily induced chronic interference via particle–cell interactions, heteroaggregation, sedimentation-driven shading, and extracellular polymeric substance (EPS) regulation, rather than ROS-dominated toxicity. In contrast, AgNPs exhibited transformation-driven toxicity, undergoing intracellular speciation into Ag2S, AgCl, and Ag+, which triggered oxidative stress, membrane damage, and lipid peroxidation. TiO2NPs showed relatively high bioavailability and persistent oxidative stress effects. These results demonstrate that PEC toxicity evolves with particle type and concentration. Importantly, oxidative stress alone is insufficient to capture PEC ecotoxicity, which also involves the long-term impacts on algal behavior, sedimentation dynamics, and energy metabolism. This study provides mechanistic insights into PEC-induced algal toxicity and supports the source-oriented management of particulate pollutants in aquatic environments, particularly in hotspot scenarios such as wastewater discharge and sediment resuspension.

Background: Patients with melanoma are confronted with postoperative challenges such as tumor relapse, impaired wound healing, infection, and hypertrophic scarring, with chronic wound inflammation potentially increasing the risk of tumor recurrence. Photoresponsive hydrogels have great potential for integrated postsurgical treatment of melanoma by embedding photosensitizers or photothermal agents in porous polymer networks, combining the advantages of hydrogels and phototherapy technologies. Methods: The photoresponsive PCCUR hydrogel was synthesized via a thermal induction method, incorporating polydopamine nanoparticles (PDA-NPs), Chlorella extract (CE), and curdlan (CUR). The physicochemical properties of PCCUR and its effects on skin cells and B16F10 cells were systematically evaluated through in vitro experiments. The antitumor recurrence efficacy of the PCCUR hydrogel was assessed via an incomplete melanoma resection model. Furthermore, the regulatory effects of PCCUR on infected wound healing and its inhibitory effects on hypertrophic scar formation were investigated under temporally controlled laser irradiation. Results: The photoresponsive PCCUR hydrogel possesses temperature sensitivity, injectable properties, optimal mechanical strength, and significant antimicrobial activity and is tailored to mitigate postsurgical issues through healing phase-specific laser irradiation adjustments. During the inflammatory phase, synergistic photodynamic therapy (PDT) and photothermal therapy (PTT) not only eliminate infections but also induce immunogenic cell death (ICD), releasing tumor antigens that bypass tumor heterogeneity and are captured by the in situ hydrogel. The antigen-captured hydrogel acts as an “antigen reservoir” and releases captured antigens to recruit dendritic cells (DCs) to stimulate an antitumor immune response. In the proliferation phase, the inherent properties of the hydrogel inhibit inflammation and facilitate cell proliferation and angiogenesis. The remodeling phase involves low-intensity laser-based phototherapy to modulate myofibroblast activity and collagen remodeling, preventing hypertrophic scarring. Conclusions: The dynamic photoresponsive properties of PCCUR by introducing a 3R paradigm (Remove-Residual tumor cells/bacteria; Remodel-Immune microenvironment; Repair-Skin tissue) provide a safe and efficient therapeutic approach tailored to the three stages of wound healing, making it a promising dressing option for the comprehensive management of postoperative wounds in melanoma patients.

In this study,Chlorella pyrenoidosawas cultured with different concentrations of Cr(III) and Cr(VI), and the changes in biomass, lipid content, protein content and total sugar content ofChlorella pyrenoidosaunder different culture conditions were detected. The effects of Cr(III) and Cr(VI) on the intracellular antioxidant system and related key enzymes of lipid synthesis were analyzed. The results showed that appropriate Cr(III) and Cr(VI) concentrations could makeChlorella pyrenoidosaregulate the contents of SOD, POD, ASA, CAT and GSH in cells, improve the enzyme activity of ACC and promote lipid synthesis. The aim was to investigate the mechanism of lipid synthesis by analyzing the distribution of Cr(III) and Cr(VI) in algal cells. The lipid content ofChlorella pyrenoidosareached its highest level at 50.43 wt% and 38.00 wt% when both Cr(III) and Cr(VI) concentrations in the culture environment were 0.5 mg/L. These values were 1.37 and 1.03 times higher than the control group, respectively. The addition of appropriate Cr(III) and Cr(VI) concentrations can increase ME enzyme activity and decrease PEPC enzyme activity, shorten the biomass accumulation cycle and prolong the lipid synthesis cycle.
