On the purpose of improving the surface wettability and tribological properties of Ti6Al4V alloy for artificial joints, we designed a novel ‘soft (hydrogel layer)–hard (porous Ti6Al4V alloy substrate)’ structure, which was fabricated by laser texturing, surface dopamine modification and poly(vinyl alcohol) (PVA) hydrogel casting. Characterization results revealed that the PVA hydrogel layer could be attached to Ti6Al4V alloy successfully, and the LTi–DA–PVA specimen obtained a hydrophilic surface with a static water contact of 32°. It is worth noting that compared to the pristine Ti6Al4V alloy, the friction coefficient of the LTi–DA–PVA specimen could be as low as 0.01, which is similar to that of natural cartilage. This result was attributed to the synergistic effect of the high load-bearing capacity of the lower Ti6Al4V substrate and the good biphasic lubrication of the upper PVA hydrogel layer. In addition, the introduction of dopamine could significantly improve the interfacial bonding performance between the substrate and the hydrogel layer. This work provides an effective method for improving the surface wettability and tribological properties of Ti6Al4V as artificial joints.

Thermal reversible Diels-Alder reaction (DA) is widely used to prolong the lifespan of polyurethane and cut down maintenance costs. However, it requires high temperature to repair damages with limited repair efficiency. In this study, a novel dual dynamic covalent bonds-based self-healing polyurethane (SPU-DA) with remarkable room temperature repair capability is fabricated by incorporating a custom-made chain extender containing disulfide bonds into DA bonds-based polyurethane. The resultant SPU-DA exhibits good comprehensive mechanical properties, and the tensile strength and elongation at break can reach 9.87 MPa and 636 %, respectively. Meanwhile, cracks emerged in SPU-DA can be repaired efficiently by the synergetic actions of the exchange of disulfide bonds, dissociation of hydrogen bonds, and the thermal reversible DA reaction. Furthermore, SPU-DA can be repaired multiply and the self-repair efficiency reaches 90.1 % at 120 °C and maintains a considerable 71 % even after three damage-repair cycles. More importantly, the as-prepared SPU-DA demonstrates the ability to heal at lower and room temperature. The repair efficiency of 41.3 % is achieved even at room temperature for 24 h without any additional treatment. Therefore, this research offers an ideal method to balance the rigidity provided by Diels-Alder bonds and the self-healing efficiency offered by the disulfide bonds. SPU-DA will be a potential functional material to be utilized in a wide range of engineering applications.

Background Perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) are persistent environmental contaminants of global concern. However, the soil ecotoxicity of emerging alternatives, such as hexafluoropropylene oxide dimer acid (HFPO-DA, a PFOA substitute), remains poorly understood. Objectives This study evaluated the ecotoxicological effects of a typical long-chain PFAS, perfluorooctanoic acid (PFOA), and its short-chain substitute, HFPO-DA, on diverse arylsulfatase (ARS) pools across eight soil types. The inhibitory mechanisms were further elucidated using integrated enzyme kinetics and molecular simulation approaches. Results The results showed that intracellular ARS (intra-ARS) dominated the total enzyme pool (54.44%–77.44%) and was positively correlated with microbial biomass carbon ( p < 0.05). The catalytic efficiency of intra-ARS was the most sensitive to PFAS exposure, with HFPO-DA exhibiting higher toxicity, as reflected by lower ED 10 values (the concentration causing a 10% reduction in intra-ARS catalytic efficiency), ranging from 2.06 to 36.33 mg kg −1 . Enzyme kinetics analyses indicated that both PFAS primarily caused competitive inhibition. Notably, HFPO-DA displayed a lower inhibition constant ( K ic ) than PFOA, suggesting a stronger binding affinity to the enzyme. Molecular docking further supported this finding, with binding energies of −5.8 and − 6.6 kcal mol −1 for PFOA and HFPO-DA, respectively, indicating that HFPO-DA forms a more stable complex with ARS. Principal Component Analysis (PCA) revealed that variability in the toxicity threshold (ED 10 ) was mainly driven by soil organic matter, total nitrogen, clay content, and pH. Based on these factors, a multivariate regression model was developed that accurately predicted toxicity thresholds (adjusted R 2 = 0.914 and 0.842), demonstrating strong predictive capability. Conclusion These findings provide a critical theoretical basis from the perspective of soil enzymatic function for establishing scientifically sound safety thresholds for PFAS across different soil types and for optimizing remediation strategies for contaminated soils.

Organic field-effect transistors (OFETs) represent a breakthrough biosensing technology owing to their exceptional sensitivity, mechanical flexibility, and cost-effective manufacturing. The simultaneous detection of dopamine (DA), ascorbic acid (AA), and uric acid (UA) holds critical importance in clinical diagnostics as these biomarkers provide vital information about neurological health, antioxidant status, and renal function. Conventional detection methods face significant challenges due to the overlapping oxidation potentials of these molecules in electrochemical measurements, resulting in poor selectivity and accuracy. Developing advanced sensing platforms capable of discriminating these biomarkers in complex biological samples remains an urgent requirement in modern healthcare diagnostics. This research successfully developed a molecularly engineered extended-gate OFET biosensor for simultaneous detection of DA, AA, and UA with exceptional performance. The functionalized sensing interface, incorporating cysteamine for DA recognition, carboxylated graphene oxide for AA catalysis, and p-AMTa for UA binding, achieved remarkable detection limits of 0.28 μM, 0.35 μM, and 0.41 μM respectively. Comprehensive cross-reactivity studies demonstrated excellent specificity with interference below 5.2% in mixed analyte solutions. Quantitative validation using standard addition method showed recovery rates of 97.2-103.8% and relative errors consistently under 4.2%, confirming high detection accuracy. The biosensor maintained outstanding operational stability over 30 detection cycles with RSD < 5.1%, demonstrating excellent reproducibility. This integrated platform represents a significant advancement in multiplexed biomarker detection, offering a reliable approach for complex sample analysis. This work provides a robust strategy for simultaneous electrochemical detection of multiple biomarkers, effectively addressing selectivity challenges in conventional biosensing. The developed platform demonstrates strong potential for clinical diagnostics and point-of-care testing applications, enabling rapid disease screening and health monitoring. This technology represents a significant step forward in the development of high-performance, cost-effective diagnostic tools for healthcare applications.

Objective: Hexafluoropropylene oxide dimer acid (HFPO-DA), also known as GenX, is widely used globally, raising concerns about its safety and public health implications. However, its toxicity mechanism remains unclear. The purpose of this study was to develop a reliable method for detecting HFPO-DA in mice and to investigate its absorption, distribution, and impact on hepatic lipid metabolism. Method: HFPO-DA levels were measured in the serum and eight tissues of C57BL/6J mice after oral administration using ultra-performance liquid chromatography–tandem mass spectrometry (UPLC-MS/MS). Lipid metabolites in the liver were also detected and analyzed. Results: HFPO-DA was rapidly absorbed into the bloodstream and widely distributed throughout all tested tissues. It penetrated the blood–brain barrier, with the highest concentration in the liver; however, long-term effects on the lungs also warrant attention. HFPO-DA disrupted liver lipid metabolism, leading to acylcarnitine accumulation while lowering triglycerides and cholesterol. Conclusion: This study on the pharmacokinetics and tissue distribution of HFPO-DA in mice following oral exposure revealed that HFPO-DA exacerbates liver injury by altering hepatic lipid metabolism. These findings provide theoretical support for toxicological studies on the emerging environmental pollutant HFPO-DA.

In general, atherosclerosis is considered to be a form of chronic inflammation. Dexamethasone has anti-inflammatory effects in atherosclerosis, but it was not considered for long-term administration on account of a poor pharmacokinetic profile and adverse side effects. Nanoparticles in which drugs can be dissolved, encapsulated, entrapped or chemically attached to the particle surface have abilities to incorporate dexamethasone and to be used as controlled or targeted drug delivery system. Long circulatory polymeric nanoparticles present as an assisting approach for controlled and targeted release of the encapsulated drug at the atherosclerotic site. Polymeric nanoparticles combined with ultrasound (US) are widely applied in cancer treatment due to their time applications, low cost, simplicity, and safety. However, there are few studies on atherosclerosis treatment using polymeric nanoparticles combined with US. In this study, targeted dexamethasone acetate (DA)-loaded poly (lactide-glycolide)-polyethylene glycol-cRGD (PLGA-PEG-cRGD) nanoparticles (DA-PLGA-PEG-cRGD NPs) were prepared by the emulsion-evaporation method using cRGD modified PLGA-PEG polymeric materials (PLGA-PEG-cRGD) prepared as the carrier. The average particle size of DA-PLGA-PEG-cRGD NPs was 221.6 ± 0.9 nm. Morphology of the nanoparticles was spherical and uniformly dispersed. In addition, the DA released profiles suggested that ultrasound could promote drug release from the nanocarriers and accelerate the rate of release. In vitro, the cellular uptake process of fluorescein isothiocyanate (FITC)@DA-PLGA-PEG-cRGD NPs combined with US into the damaged human umbilical vein endothelial cells (HUVECs) indicated that US promoted rapid intracellular uptake of[email protected]PLGA-PEG-cRGD NPs. The cell viability of DA-PLGA-PEG-cRGD NPs combined with US reached 91.9% ± 0.2%, which demonstrated that DA-PLGA-PEG-cRGD NPs combined with US had a positive therapeutic effect on damaged HUVECs. Overall, DA-PLGA-PEG-cRGD NPs in combination with US may provide a promising drug delivery system to enhance the therapeutic effects of these chemotherapeutics at the cellular level. Keywords:dexamethasone acetate;polymeric nanoparticles;ultrasound;atherosclerosis;polypeptide targeting

Simultaneous monitoring of key metabolites like dopamine, ascorbic acid, and uric acid is essential for early disease diagnosis and evaluating treatment. Electrochemical techniques are increasingly used for precise, point-of-care testing (POCT) of these metabolites. Herein, a sample pretreatment method called effervescent solid-phase extraction (ESPE) was proposed for efficient enrichment of trace analytes for electrochemical detection. In an ESPE process, effervescent tablets made of gold nanoparticle-decorated graphene oxide (Au/GO) were first self-dispersed in a test solution to promote the enrichment of analytes on the Au/GO adsorbents, followed by the addition of flocculant effervescent tablets to cause Au/GO sheets to form self-assembled aggregates, which then can be efficiently collected by the foam electrodes. The entire sample pretreatment process operates without external power and takes only 5 min. With the assistance of the ESPE method, our electrochemical sensors achieve an ultralow detection limit for dopamine, ascorbic acid, and uric acid of 80 pM, 1.8 nM, and 460 pM, respectively, which are two to three orders of magnitude lower than the results obtained by the drop casting technique. The enhancement mechanism of our approach is based on increasing the contact probability with analytes through dynamic dispersion of the Au/GO adsorbents, in contrast to the static diffusion mechanism relied on Brownian motion. We also show that combining the ESPE solution kit with a portable micro-electrochemical workstation can attain the same detection level as HPLC in real urine samples. The proposed ESPE approach holds great promise for POCT applications in 2D material-based biosensors.

In situ SiO 2 growth with DA-g-C 3 N 4 co-doping yields hybrid crosslinked PA layer. • Compound strategy mitigates DA-g-C 3 N 4 aggregation in PA layer. • Achieved simultaneous gains in Mg 2+ /Li + selectivity and permeability performance. • MgCl 2 rejection stayed >93% over 160 h, demonstrating excellent long-term stability. It is often difficult for thin-film composite (TFC) nanofiltration (NF) membranes to balance high separation selectivity and high permeability, and the contradiction between the two will lead to their overall performance constraints. In this study, a dual-phase regulation strategy is proposed, which adopts the in-situ sol method and the composite strategy of introducing two-dimensional materials for the preparation of organic-inorganic cross-linked polyamide (PA) NF membranes. A trimesoyl chloride (TMC) organic phase solution containing tetraethyl orthosilicate was first introduced onto the polyethersulfone (PES) substrate, followed by contact with a polyethyleneimine (PEI) aqueous phase solution containing dopamine-functionalized carbon nitride (DA-g-C 3 N 4 ), during which silicon dioxide (SiO 2 )/Si–O–Si domains were generated in situ within the PA layer. These domains restricted the free volume of the PA layer, narrowed the effective pore-size distribution, and suppressed nonselective defects, while the embedded DA-g-C 3 N 4 improved the compatibility between the nanofillers and the PA layer, thereby producing P-1Si-0.025CN membranes with an effective pore size of 0.4 nm. Compared with the P-NF membrane prepared by reverse interfacial polymerization of TMC and PEI directly on the PES substrate, Mg 2+ /Li + separation factor (SF) increased to 67.2 (Mg 2+ /Li + mass ratio is 20), salt solution flux was 27.1 LMH, separation and permeation capacity of mixed salt was enhanced, salt solution flux has increased by 4 times, and the SF has increased by 7 times, Mg 2+ /Li + ratio decreased from 20 in feed to 0.71 in permeation process. Moreover, it exhibited excellent separation performance in diluted salt lake brine, with an Mg 2+ rejection rate exceeding 98%, and it also exhibited high fouling resistance and long-term stability. Background Snake venom serine proteases (SVSPs) are critical toxic components that interfere with the hemostatic balance and serve as a key candidate library for anticoagulant drug development. Despite the diversity of SVSPs in Deinagkistrodon acutus venom, systematic studies of low-abundance SVSPs with thrombin-like activity profiles are limited. Hence, this research aimed to isolate and characterize a bioactive SVSP candidate from D. acutus venom using integrated multi-omics and to assess its antithrombotic effects and bleeding risk. Methods PacBio full-length transcriptome sequencing combined with label-free quantitative proteomics was used to screen SVSP candidates with matched transcription–translation associations. The target protein Da-29 was purified using gel filtration and reverse-phase chromatography, and its primary structure was confirmed using transcriptome sequencing and liquid chromatography–tandem mass spectrometry. In vitro enzymatic activity and fibrinogen hydrolysis specificity, as well as in vivo antithrombotic efficacy and bleeding risk, were evaluated using rodent thrombosis and bleeding models. Results Da-29 was observed to be a 260-amino-acid SVSP containing a conserved catalytic triad and an S1 substrate-binding pocket. It exhibited significant amidase and fibrinogen-clotting activities and selectively hydrolyzed fibrinogen Aα and Bβ chains. Furthermore, Da-29 exerted dose-associated antithrombotic effects in vivo , extending the coagulation time, reducing the fibrinogen/TXB2 levels, and upregulating 6-keto-PGF1α, without causing considerable bleeding risks at effective doses. Conclusion Da-29 exhibits antithrombotic activity associated with fibrinogen degradation and linked to concurrent alterations in TXB 2 and 6-keto-PGF1α levels, establishing its potential as an antithrombotic experimental lead. This integrated omics strategy can effectively identify low- abundance bioactive molecules from snake venom, offering a reproducible paradigm for the discovery of functional proteins in natural products.

Cholestasis is characterized by bile acid (BA) circulation disorders, which is usually related to damage of hepatocyte barrier function. Currently, patients with cholestasis face several obstacles in seeking diagnosis and therapy. Da-Huang-Xiao-Shi decoction (DHXSD) is an ancient classic formula that has been used clinically for cholestasis treatment. Nevertheless, the underlying biological activities and therapeutic mechanisms remain unclear. In this study, an alpha-naphthylisothiocyanate (ANIT)-induced cholestasis rat model was established to examine the anticholestatic effects of DHXSD using histopathological and molecular analyses. Transcriptomic analysis combined with 16S rRNA gene sequencing analysis was systematically applied to study the mechanism of action of DHXSD. Simultaneously, the effect of DHXSD on gut microbiota, short-chain fatty acids (SCFAs), and intestinal barrier function were evaluated based on the ANIT-induced cholestasis model in rats. The results showed that DHXSD effectively attenuated ANIT-induced cholestasis by reducing liver function indicators (alanine transaminase [ALT], P < 0.05; alkaline phosphatase [ALP], P < 0.05; total bile acid [TBA], P < 0.01; γ-glutamyl transpeptidase [GGT], P < 0.001) and levels of hepatotoxicity-related enzymes (P < 0.05), thus improving the recovery of histopathological injuries, and regulating levels of inflammatory cytokines (P < 0.05). In addition, 16S rRNA gene sequencing analysis combined with intestinal barrier function analysis revealed that the DHXSD significantly ameliorated ANIT-induced gut microbiota dysbiosis. Significantly altered genes in the model and treatment groups were screened using transcriptomic analysis. Sixty-eight genes and four microbial genera were simultaneously altered with opposing trends in variation after ANIT and DHXSD treatments. We built a framework for predicting targets and host-microbe interaction mechanisms, as well as identifying alternative treatment for cholestasis, which should be validated further for clinical application. In conclusion, DHXSD appears to be a promising agent for protection against liver injury.IMPORTANCE Cholestasis is a serious manifestation of liver diseases resulting in liver injury, fibrosis, and liver failure with limited therapies. To date, only ursodeoxycholic acid (UDCA) has been approved by the U.S. Food and Drug Administration for the treatment of cholestasis. However, approximately one-third of patients with cholestasis are unresponsive to UDCA. Therefore, it is urgent to search for appropriate therapeutic agents for restoring stoppage status of the bile components to treat cholestasis. In this study, we investigated how the microbiome and transcriptome data sets correlated with each other to clarify the role of microbiome alterations in host metabolism. In combination, this research offers potential molecular biomarkers that should be validated for more accurate diagnosis of cholestasis and the clinical utilisation of gut microbiota as a target for treatment.

Perfluorooctanoic acid (PFOA), a typical perfluoroalkyl group compound, has received worldwide attention due to its significant environmental toxicity. Following regulatory bans on the production and emission of PFOA, concerns have been raised about the potential health risks and the safety of novel perfluoroalkyl analogues. HFPO-DA (trade name Gen-X) and HFPO-TA are two perfluoroalkyl analogues known to be bioaccumulative, whose level of toxicity and whether they are safe alternatives to PFOA remain unclear. In the following study, the physiological and metabolic effects of exposure to PFOA and its novel analogues were explored in zebrafish using 1/3 LC 50 (PFOA 100 μM, Gen-X 200 μM, HFPO-TA 30 μM). At the same LC 50 toxicological effect, exposure to PFOA and HFPO-TA resulted in abnormal phenotypes such as spinal curvature, pericardial edema and aberrant body length, while Gen-X was little changed. Metabolically, PFOA, HFPO-TA and Gen-X all significantly increased total cholesterol in exposed zebrafish with PFOA and HFPO-TA also increasing total triglyceride levels. Transcriptome analysis showed that the number of differentially expressed genes in PFOA, Gen-X, and HFPO-TA treated conditions compared to control groups were 527, 572, and 3, 933, respectively. KEGG and GO analysis of differentially expressed genes revealed pathways and functions related to lipid metabolism as well as significant activation of the peroxisome proliferators-activated receptor (PPARs) pathway. Furthermore, RT-qPCR analysis identified significant dysregulation in the downstream target genes of PPARα, which is responsible for lipid oxidative catabolism, and the SREBP pathway, which is responsible for lipid synthesis. In conclusion, both perfluoroalkyl analogues HFPO-TA and Gen-X exhibit significant physiological and metabolic toxicity to aquatic organisms and their environmental accumulation should be closely regulated.
