Growth and metabolism regulation of cinnamic acid and its derivatives to Auxenochlorella pyrenoidosa

Min Li,Zhen-Fang Shang,Zhi-Jing Ni,Hui-Jun Zhao,Qian Lei,Zhao-Jun Wei

Auxenochlorella pyrenoidosa 's industrial utility is closely associated with the concentration of high-value products. This study focuses on the effects of cinnamic acid (CA) and its derivatives, which known as secondary metabolites in higher plants and effective plant growth regulators, on the growth and metabolic profiles of A. pyrenoidosa . It was discovered that CA and its derivatives significantly enhance the biomass of A. pyrenoidosa under conditions of 10 mg/L over 48 h or 100 mg/L over 120 h. Cells treated with 10 mg/L CA exhibited lower oxidative stress and maintained robust metabolic activity. Metabolic pathways, including those for carbohydrates, lipids, small peptides, and cofactors, were stimulated, leading to the accumulation of a wide array of bio-products, notably high-value products such as α-linolenic acid, cis-4,7,10,13,16,19-docosahexaenoic acid (DHA), nicotinamide adenine dinucleotide plus hydrogen (NADH), glutathione, and vitamins B2/B3. This study demonstrates that CA and its derivatives are good nutritional enhancers for cultivating A. pyrenoidosa , highlighting their potential for broad application in biotechnological industries.

LncRNA-H19 induces hepatic stellate cell activation via upregulating alcohol dehydrogenase III-mediated retinoic acid signals

Zhi-Min Wang,Si-Wei Xia,Tian Zhang,Zhen-Yi Wang,Xiang Yang,Jun Kai,Xu-Dong Cheng,Jiang-Juan Shao,Shan-Zhong Tan,An-Ping Chen,Shi-Jun Wang,Feng Zhang,Zi-Li Zhang,Shi-Zhong Zheng

Activation of hepatic stellate cells (HSCs) is a pivotal event in liver fibrosis, characterized by enhanced retinoic acid signals. Although up-regulated retinoic acid signal responds further to maintain HSC activation, the underlying molecular mechanisms are largely unknown. In this study, we sought to investigate the role of lncRNA-H19 in regulation of retinoic acid signals, and to further examine the underlying mechanism in this molecular context. We found that lncRNA-H19 upregulation could enhance retinoic acid signals to induce HSC activation, whereas lncRNA-H19 knockdown completely disturbed retinoic acid signals. Moreover, the activation of retinoic acid signals impaired the lncRNA-H19 knockdown mediated HSC inactivation. Interestingly, we also found that enhanced retinoic acid signals by lncRNA-H19 was associated with a coordinate increase in retinol metabolism during HSC activation. Increased retinol metabolism contributed to obvious lipid droplet consumption. Importantly, we identified that alcohol dehydrogenase III (ADH3) was essential for lncRNA-H19 to enhance retinoic acid signals. The inhibition of ADH3 completely abrogated the lncRNA-H19 mediated retinoic acid signals and HSC activation. Of note, we identified dihydroartemisinin (DHA) as a natural inhibitor for lncRNA-H19. Treatment with DHA significantly decreased the expression of lncRNA-H19, reduced the expression of ADH3, blocked retinoic acid signals, and in turn, inhibited HSC activation. Overall, these results provided novel implications to reveal the molecular mechanism of increased retinoic acid signals during HSC activation, and identify lncRNA-H19/ADH3 pathway as a potential target for the treatment of liver fibrosis.

Synergistic dual functionality of HIPPEs stabilized by lactoferrin-vanillic acid-astragalus polysaccharide complex: simultaneously enabling DHA delivery and 3D printing of dysphagia foods

Haifu Jia,Jiayue Yang,Zhishan Song,Xiangying Meng,Wei Zhang,Ling Guo,Qianyu Zhao,Yujun Jiang

HIPPEs were stabilized by LF-VA-APS ternary covalent complexes. • The emulsion with 77.5% oil content exhibited the highest environmental stability. • HIPPEs stabilized with LF-VA-APS complex exhibited superior rheological properties. • HIPPEs enhance the oxidative stability, bioavailability and cellular uptake of DHA. • HIPPEs stabilized with LF-VA-APS complex are suitable 3D printed dysphagia foods. The rapidly ageing global population has led to an increasingly severe prevalence of dysphagia and associated malnutrition, necessitating the development of innovative foods that integrate appropriate texture with high nutritional density. This study aims to develop high-internal-phase Pickering emulsions (HIPPEs) stabilized by a ternary covalent complex of lactoferrin (LF) – vanillic acid (VA) - Astragalus polysaccharide (APS), aiming to concurrently enhance the bioaccessibility and cellular uptake efficiency of docosahexaenoic acid (DHA), while exploring their potential in 3D-printed dysphagia foods. Results indicate that at an oil phase volume fraction of 77.5%, the HIPPEs stabilized by the LF-VA-APS complex exhibited optimal physicochemical and environmental stability. Compared to binary complexes, this ternary system significantly improved the oxidative stability and in vitro digestion and release profiles of DHA. Regarding 3D printing applications, the emulsion exhibited desirable shear-thinning behavior, a high storage modulus, and robust physical stability, thereby ensuring high printing fidelity and structural integrity. Furthermore, in accordance with the International Dysphagia Diet Standardization Initiative (IDDSI) framework, the emulsion samples were classified as Level 6 (soft & bite-sized) foods, fulfilling the criteria for safe swallowing. In summary, the multifunctional HIPPE system developed herein successfully overcomes the inherent trade-offs in high-internal-phase emulsions regarding nutritional fortification and texture control, offering novel strategies for the development of foods for dysphagia. Hot water pretreatment-induced significant metabolite changes in the sea cucumber Apostichopus japonicus

Peipei Yin,Airong Jia,Kirsten Heimann,Miansong Zhang,Xin Liu,Wei Zhang,Changheng Liu

Hot water pretreatment of sea cucumbers potentially changes nutritional benefits. This study aimed to quantify hot water pretreatment-induced changes in metabolite profiles of sea cucumber body walls. ICP-OES, GC-MS, and LC-MS analyses of untreated- (UT-BW), hot water-treated body walls (HW-BW) of Apostichopus japonicus , and the hot water extract (HW-E) determined significant losses of minerals (25–50% w/w), protein (~11% w/w), carbohydrate (33% w/w), saponins (~41% w/w), and spermidine (100%), a potential antipsychotic from hot water-treated samples. Multivariate comparisons of HW-BW with UT-BW and HW-BW with HW-E showed increases in amino acids and fatty acids, suggesting hot water-induced degradation or transformation or easier extraction of protein, lipid or other components. Presence of 80 to 88.5% of compounds in the HW-E and lower DHA, EPA and glycerophospholipids levels in HW-BW suggested extraction of these metabolites. These data indicate that novel processing technologies are required to preserve the full nutritional benefits of sea cucumbers.

Docosahexaenoic acid inhibits pheromone-responsive-plasmid-mediated conjugative transfer of antibiotic resistance genes in Enterococcus faecalis

Hongrui Zhou,Xiaobo Yang,Yutong Yang,Yuanyuan Niu,Jing Li,Xinyue Fu,Shang Wang,Bin Xue,Chenyu Li,Chen Zhao,Xi Zhang,Zhiqiang Shen,Jingfeng Wang,Zhigang Qiu

The rapid spread of antibiotic-resistance genes (ARGs) in Enterococcus faecalis ( E. faecalis ) poses a great challenge to human health and ecological and environmental safety. Therefore, it is important to control the spread of ARGs. In this study, we observed that the addition of 5 μg/mL docosahexaenoic acid (DHA) reduced the conjugative transfer of pCF10 plasmid by more than 95% in E. faecalis . DHA disturbed the pheromone transport by inhibiting the mRNA levels of the prgZ gene, causing the iCF10 pheromone to accumulate in the donor bacteria and bond to the PrgX receptor to form an inhibitory phase, which resulted in the down-regulation of the expression of genes related to conjugative transfer, inhibiting biofilm formation, reducing bacterial adhesion and thus inhibiting conjugative transfer. Collectively, DHA exhibited an admirable inhibitory effect on the transfer of ARGs in E. faecalis . This study provided a technical option to control the transfer of ARGs.

ROS-Responsive and Self-Catalytic Nanocarriers for a Combination of Chemotherapy and Reinforced Ferroptosis against Breast Cancer

Beibei Zhang,Hao Liu,Yifei Wang,Yong Zhang

Ferroptosis is an appealing cancer therapy strategy based on the H2O2-involved Fenton reaction to produce toxic •OH for lipid peroxidation. However, intracellular H2O2 is easily consumed and results in a deficient Fenton reaction. This obstacle can be overcome by traditional chemotherapeutic drugs for H2O2 supplements. Moreover, a recent work illustrated that dihydroartemisinin (DHA) could promote ferroptosis against tumoral cells, particularly in the presence of ferrous compounds. To achieve combined chemotherapy and ferroptosis, a nanocarrier (TKNPDHA-Fc) was constructed by using thioketal (TK)-bridged paclitaxel prodrug (PEG-TK-PTX) and ferrocene (Fc)-conjugated PEG-Fc, where DHA was encapsulated by a hydrophobic–hydrophobic interaction. Upon cellular uptake, TKNPDHA-Fc could facilitate PTX release through TK breakage under an excess H2O2 microenvironment. Owing to the loss of the hydrophobic PTX component, TKNPDHA-Fc underwent a rapid dissociation for improving DHA to act as a ferroptotic inducer along with Fe supplied from Fc. Moreover, both the chemotherapy-induced reactive oxygen species and the •OH produced from reinforced ferroptosis further stimulated the TK cleavage. The “self-catalytic” loop of TKNPDHA-Fc remarkably improved the antitumor performance in vivo via combined mechanisms, and its tumor inhibition rate reached 78.3%. This work highlights the contribution of ROS-responsive and self-catalytic nanoplatforms for enhancing the potential of combined chemotherapy and ferroptosis for cancer therapy in the future.

Tumor targeting with docosahexaenoic acid‑conjugated docetaxel for inhibiting lung cancer metastasis to bone

Shougang Jiang,Zhiguo Liu,Lei Wu,Yingjie Yuan,Yan Hu,Xingyao Zhang,Liang Wei,Yuangang Zu

Docetaxel (DTX) is currently used as a first‑ or second‑line drug treatment for patients with lung cancer, however, it is less effective for the treatment of patients with bone metastasis of lung cancer. This is primarily due to the fact that docetaxel is nonspecific. In the present study, docosahexaenoic acid (DHA) was selected as a tumor‑targeting ligand, and DHA‑conjugated DTX (DTX‑DHA) was prepared for inhibiting lung cancer metastasis to bone. The anti‑cancer activity assay revealed that DTX‑DHA exhibited a similar antitumor efficacy to DTX in vitro. The maximum tolerated dose of DTX‑DHA was increased compared with that of DTX. The present study results indicated that DTX‑DHA exhibited an improved inhibition efficacy of lung cancer metastasis to bone in comparison with DTX in vivo. Encouragingly, the mean survival time of the DTX‑DHA group (30.60 days) was increased compared with the DTX group (26.10 days; P<0.01). Furthermore, the results of cell migration and osteoclast‑induced formation assays suggested that DTX‑DHA inhibited lung cancer metastasis to bone primarily by affecting lung cancer cell migration. These results indicate that DTX‑DHA may exhibit a potential therapeutic effect against lung cancer metastasis to bone.

Microalgae oil from Schizochytrium sp. alleviates obesity and modulates gut microbiota in high-fat diet-fed mice

Liyuan Ran,Jinhui Yu,Rui Ma,Qing Yao,Mingjie Wang,Yuping Bi,Zichao Yu,Yingjie Wu

Omega-3 PUFAs rich in fish oil are believed to prevent obesity by improving lipid metabolism and regulating gut microbiota. Microalgae oil is considered as an alternative source of omega-3 PUFAs owing to diminishing fish resources. Schizochytrium microalgae oil (SMO), with a high DHA proportion, is a promising source for commercial DHA production. However, its weight-loss and gut microbiota-regulating properties are not well studied. Here we compared the obesity reducing effects of SMO, commercial fish oil (FO) and a weight-loss drug, Orlistat (OL), in a high-fat diet (HFD) induced obesity mouse model. We found that SMO is comparable to commercial FO and OL with regard to weight loss, and it even exhibits the weight-loss effects earlier than FO and OL. It can efficiently inhibit the expression of lipogenesis-related genes and induce the expression of lipolysis-related genes. Moreover, SMO has different gut microbiota modulating effects from those of FO and OL. It does not influence the diversity of bacterial community, but does increase the abundance of several beneficial SCFAs-producing bacteria and inhibits obesity-promoting Desulfovibrio and several pathogens. We also found that SMO recovers the HFD-disturbed metabolic capability of gut microbiota. It can increase the abundance of several metabolism-related pathways, such as those of amino acids, SCFAs and bile acid, and decrease the level of the LPS biosynthesis pathway, which probably contributes to an improvement of lipid metabolism and restoration of the colonic mucosal barrier impaired by HFD. Our data suggest that SMO can be used as a superior dietary supplement for alleviating obesity.

Production of Lipid and Xylo-oligosaccharides from Cellulose and Hemicellulose in Reed Sawdust

Jiao MeiZhen,Zhang Xinran,Wang Yiqin,Du Jian,Tao Yehan,Lv Yanna,Chen Ming,Lu Jie,Wang Haisong

Reed sawdust is a kind of paper mill waste with high cellulose and hemicellulose content. To promote the rational use of resources, it is essential to make full use of waste resources and transform them into new values. In this work, reed sawdust was pretreated with liquid hot water (LHW) at 170 °C for 30 min. A total of 39.00 g/L glucose was obtained after enzymatic saccharification of cellulose at 50 °C, 20 FPU/g- reed sawdust cellulase, 25% (w/v) reed sawdust, in 5 replenishments. When the fermentation was performed 96 h, the medium contained xylo-oligosaccharides (XOS) 11.74 g/L and biomass 15.21 g/L, in which lipid was 4.14 g/L. After spray drying, feed additives containing 29.17% XOS and 10.29% docosahexaenoic acid (DHA) can be prepared. In particular, the hemicellulose and cellulose in reed sawdust are creatively used at the same time without separation, which greatly reduces the cost of purification in traditional processes and provides a new way for the high-value transformation of sawdust resources.

New dual-modal electrochemical and photoelectrochemical aptasensing strategy for the sensitive detection of penicillin G based on an iron-doped covalent triazine framework

Miaoran Xu,Hao Dang,Mengfei Wang,Shuai Zhang,Qiaojuan Jia,Jia-Yue Tian,Minghua Wang,Miao Du

The sensitive determination of penicillin G (PG) in complex environments is essential for guaranteeing food safety and human health. Herein, a new dual-modal electrochemical (EC)–photoelectrochemical (PEC) aptasensing strategy has been proposed based on an Fe-doped covalent triazine framework (denoted as Fe-MA-Dha-CTF) synthesized by using 2,5-dihydroxyterephthalaldehyde (Dha) and melamine (MA) as building blocks and utilized to detect trace PG. Compared with the platform based on metal-free MA-Dha-CTF, the constructed Fe-MA-Dha-CTF demonstrated enhanced photoelectric conversion efficiency, superior electrochemical activity, and high bioaffinity toward the PG-targeted aptamer. The established Fe-MA-Dha-CTF-based dual-modal EC–PEC aptasensor exhibited ultralow limits of detection of 0.08 and 0.7 fg mL −1 as deduced by using EC and PEC techniques, respectively, within a wide PG concentration range of 1 fg mL −1 to 10 ng mL −1 . Moreover, the proposed EC–PEC aptasensor exhibited high selectivity, good stability, acceptable reproducibility, and great practicability in diverse real samples. The CTF-based dual-modal aptasensing approach presented here can extend the application of porous-organic frameworks in biosensing.

n-3 essential fatty acids in Nile tilapia, Oreochromis niloticus: Bioconverting LNA to DHA is relatively efficient and the LC-PUFA biosynthetic pathway is substrate limited in juvenile fish

Cuiying Chen,Wutai Guan,Qingmei Xie,Guoliang Chen,Xiaolei He,Haoliang Zhang,Weiyi Guo,Feng Chen,Yuwen Tan,Qing Pan

This study aimed to gain a better understanding of the long-chain (≥C 20 ) polyunsaturated fatty acid (LC-PUFA) biosynthetic metabolism, with a specific focus on the metabolic fate of linolenic acid (18:3n-3, LNA) and linoleic acid (18:2n-6, LA) and the relationship between these two substrates in the process of LC-PUFA biosynthesis in juvenile Nile tilapia , Oreochromis niloticus . Fish were fed four isoproteic (37.5%) and isolipidic (6.2%) diets formulated with fixed levels of LA (0.60%) and gradual levels of LNA (0.10, 0.63, 1.56 and 2.04% of dry weight, respectively) for 10 weeks. The whole-body fatty acid balance method was used to evaluate fish in vivo fatty acid metabolism . The results showed that most of dietary LNA was bioconverted to LC-PUFA when tilapia receiving not enough LNA for optimal growth. In contrast, when fish receiving sufficient dietary LNA, most of the LNA was β-oxidized, with only <10% LNA being bioconverted. The in vivo apparent △-5/-6 desaturase and elongase activities on n-3 and n-6 PUFA, respectively, exhibited a significant linear increase and decrease with the increasing dietary LNA levels, clearly indicating that for all of the enzyme activities , n-3 PUFA is a preferred substrate and the LC-PUFA biosynthetic pathway is substrate limited in Nile tilapia. Dietary higher inclusion of LNA could block or at least slow down n-6 LC-PUFA biosynthesis from LA, indicating that a direct substrate competition for accessing the Δ-6 desaturase and elongase exists between these two substrates. Furthermore, Nile tilapia were relatively efficient in bioconversion from LNA to DHA, and the DHA biosynthesis from EPA may be more direct or faster than the production of EPA from LNA. Despite of the active bioconversion of LNA up to DHA, this metabolic effort was still insufficient to compensate for the significant reduced dietary intake of n-3 LC-PUFA. No significant difference was observed in the gene expression of fatty acyl desaturase ( fads2 ) and fatty acid elongase ( elovl5 ) among all groups, which suggests that the total in vivo apparent enzyme activities are directly and positively affected by substrate availability rather than the extent of the enzyme gene expression.

Intelligent Nanoplatform with Multi Therapeutic Modalities for Synergistic Cancer Therapy

Linjie Shao,Taishun Hu,Xingyu Fan,Xiaozan Wu,Fangfang Zhou,Botao Chen,Songwen Tan,Hui Xu,Anqiang Pan,Shuquan Liang,Yongju He

Chemodynamic therapy (CDT) has attracted increasing attention in tumor treatment but is limited by insufficient endogenous H2O2. Moreover, it is challenging for monotherapy to achieve a satisfactory outcome due to tumor complexity. Herein, we developed an intelligent nanoplatform that could respond to a tumor microenvironment to induce efficient CDT without complete dependence on H2O2 and concomitantly generate chemotherapy and oncosis therapy (OT). The nanoplatform was constructed by a calcium- and iron-doped mesoporous silica nanoparticle (CFMSN) loaded with dihydroartemisinin (DHA). After entering into cancer cells, the nanoplatform could directly convert the intracellular H2O2 into toxic •OH due to the Fenton-like activity of CFMSN. Meanwhile, the acidic microenvironment and endogenous chelating molecules triggered Ca2+ and Fe3+ release from the nanoplatform, causing particle collapse with accompanying DHA release for chemotherapy. Simultaneously, the released Ca2+ induced intracellular Ca2+-overloading for OT, which was further enhanced by DHA, while the released Fe3+ was reduced to reactive Fe2+ by intracellular glutathione, guaranteeing efficient Fenton reaction-mediated CDT. Moreover, Fe2+ cleaved the peroxy bonds of DHA to generate C-centered radicals to further amplify CDT. Both in vitro and in vivo results confirmed that the nanoplatform exhibited excellent anticancer efficacy via the synergistic effect of multi therapeutic modalities, which is extremely promising for high-efficient cancer therapy.
