A Label-Free and Continuous Fluorescence Detection for L–DOPA Decarboxylase Activity Based on Supramolecular Tandem Assay

Siying Ren,Wen-Chao Geng,Xuexian Cui,Bian Wu,Zhe Zheng

Graphical We present a robust supramolecular tandem assay to monitor L-DOPA decarboxylase activity and screen enzyme inhibitors using cucurbit[7]uril and acridine orange reporter pair. L–DOPA decarboxylase (DDC) deficiency causes serious progressive neurodegenerative disorders. Developing a sensitive enzyme activity monitoring and inhibitors or activators screening approach is highly on demand for DDC deficiency diagnosis. Here, we report a simple and sensitive fluorescence supramolecular tandem assay based on reporter pair of cucurbit[7]uril and acridine orange for DDC activity monitoring continuously. Furthermore, this strategy demonstrates the potential of STA to screen inhibitors of DDC.

A mussel-bioinspired coating strategy to licence PCL osteoinductive activity for repairing bone defects

Wentao Shi,Zhijia Shen,Lu Bian,Yan Gao,Yiqing Wu,Feifei Zhao,Hong Tang,Xiaojie Lu

3,4-Dihydroxyphenylalanine (DOPA) is believed to play an essential role in the solidification and adhesion of liquid mussel adhesives; this thin polymer layer very tightly adheres to the surface of scaffolds, making it stable enough to prevent it from being peeled off. Fascinatingly, the deposition of DOPA on various types of surfaces via the self-polymerization of DOPA under alkaline conditions is a simple approach for creating biomimetic coatings. In this study, DOPA derivatives were used as monomers to form primary coatings on polycaprolactone (PCL) surfaces. After that, a second chemical crosslinking method was utilized to graft alendronate (AL) on the DOPA coatings via genipin (GP) crosslinking ( [email protected] ). X-ray photoelectron spectroscopy, Fourier transform infrared reflection and scanning electron microscopy analyses indicated that AL was distributed homogeneously on the surface of PCL. This new organic–inorganic hybridized coating could support EMSC growth and osteogenic differentiation. Additionally, animal studies were also performed to verify the function of [email protected] in promoting osteogenesis. The method is simple, cheap and highly efficient, which makes it very convenient to apply on a large scale.

Antioxidant N-acetylcysteine removing ROS: an antifouling strategy inspired by mussels

Jine Wang,Chen Zhang,Rui Zhao,Pei Wang,Meihua Jin,Jiujun Xu

Marine biofouling is a thorny issue that causes serious economic losses and adverse ecological impacts on marine ecosystems. Effective and promising antifouling strategies such as surface hydration, flow shear force, and lubricant injection have been developed to address this challenge. However, for the complex marine environment, they still appear inadequate. Mussels are a common fouling organism with strong surface adhesion ability. However, when hypoxia and the oxidative cross-linking reaction of 3,4-dihydroxy phenyl-L-alanine (DOPA) in the structure of adhesion proteins are disrupted, their adhesion ability will be greatly reduced. Inspired by this, we developed an effective antifouling strategy based on reactive oxygen species (ROS) scavenging using N-acetylcysteine (NAC) and evaluated its performance. As a ROS scavenger interfered with the oxidative cross-linking reaction of DOPA in an aqueous solution, the adhesion of DOPA was also affected on the surface of NAC functionalized polyvinyl chloride (PVC) (PVC-NAC). In addition, the colonization level of mussels and the adhesion rate of marine bacteria and benthic diatoms on PVC-NAC were low. The antifouling strategy proposed in this paper was eco-friendly and broad-spectrum, and may provide a new idea for solving marine biofouling and reducing the environmental and economic impacts of fouling organisms.

Microneedle patch based on molecular motor as a spatio-temporal controllable dosing strategy of L-DOPA for Parkinson’s disease

Xin Zhou,Bowen Li,Mingming Guo,Wenchang Peng,Deping Wang,Qinglu Guo,Shuchao Wang,Dong Ming,Bin Zheng

Controlling medication according to the condition has become a universal treatment regimen for Parkinson's disease (PD) worldwide. However, the general oral administration method is prone to a variety of digestive system discomfort, easy to be broken down by digestive juices and intestinal microorganisms, resulting in low absorption efficiency, and a serious delay in the efficacy of the drug. Here, we developed a microneedle patch (MNP) that could release drugs with controlled temporal and spatial precision triggered by near-infrared (NIR) light for the treatment of PD. The microneedles (MNs) took Gelatin-methacryloy (GelMA) as the matrix, and therapeutic drugs (L-DOPA) were stored in the mesopore of the upconversion micron-rods (UCMRs), dispersing in the GelMA. The molecular motor, as a poreblocker and gate switch, isomerizes under the excitation of ultraviolet and visible light from UCMRs for controlling drug release. This kind of MNP penetrates the epidermis in a painless, non-invasive, and non-infectious way. Under the exposure of NIR light, the L-DOPA in the MNs will be released as needed, and then it seeps into the blood and brain for relieving the symptoms of PD. In the treatment of Parkinson's mice model, this regimen significantly restored motor function, and further mechanistic studies revealed that it reduced neuroinflammation and dopaminergic neuronal death in the substantia nigra. Besides, the released L-DOPA directly entered the blood, which reduces the side effects on the gastrointestinal tract and improves the utilization rate of the drug. These attractive superiorities indicated that the MNP with controlled drug delivery system may be candidate strategies for the treatment of PD and can be used in various related biomedical fields.

The synthesis and tissue adhesiveness of temperature-sensitive hyperbranched poly(amino acid)s with functional side groups

Dedai Lu,Yongyong Zhang,Ting'e Li,Yunfei Li,Hongsen Wang,Zhiqiang Shen,Qiangbing Wei,Ziqiang Lei

The incorporation of L-3,4-dihydroxyphenylalanine (L-DOPA) residues in mussel-like biomimetic copolymers has drawn great attention due to their adhesive properties. In this work, a series of temperature-sensitive and hyperbranched poly(amino acid)s containing different functional side groups (catechol, guanidyl, mercapto and double bond) were designed and synthesized by the ring-opening polymerization of N-carboxy-α-amino acid anhydride (NCA), using a temperature-sensitive and multi-hydroxyl end pluronic L-31 as the initiator in this process. The tissue adhesive properties of these copolymers were evaluated by tensile strength tests on wet pork skin. We found that the topological structure, side groups of copolymers, adhesive temperature and cure time had influence on the wet adhesive strength, especially side groups. While only L-DOPA was incorporated into copolymeric chains, the wet adhesive strength was about 106 kPa. With guanidyl, mercapto and double bonds incorporated into copolymeric chains, an obvious increase in the wet adhesive strength was observed. Additionally, the wet adhesive strength improved by about 20 kPa at body temperature (37 °C) as compared to that at room temperature (25 °C). Meanwhile, we found that it has good biocompatibility and degradability through its cytotoxicity and degradation tests.

Expression of chiral molecular and supramolecular structure on enantioselective catalytic activity

Qingqing Sun,Baocheng Bao,Wenqian Dong,Yanchao Lyu,Mengyuan Wang,Zheng Xi,Jie Han,Rong Guo

Understanding the structure-function relationships encoded on chiral catalysts is important for investigating the fundamental principles of catalytic enantioselectivity . Herein, the synthesis and self-assembly of naphthalene substituted bis - l / d -histidine amphiphiles ( bis - l / d - NapHis) in DMF/water solution mixture is reported. The resulting supramolecular assemblies featuring well-defined P/M nanoribbons (NRs). With combination of the ( P/M )-NR and metal ion catalytic centers (M n + = Co 2+ , Cu 2+ , Fe 3+ ), the ( P )-NR-M n + as chiral supramolecular catalysts show catalytic preference to 3,4-dihydroxy- S -phenylalanine ( S -DOPA) oxidation while the ( M )-NR-M n + show enantioselective bias to R -DOPA oxidation. In contrast, their monomeric counterparts bis - l / d - NapHis-M n + display an inverse and dramatically lower catalytic selectivity in the R/S -DOPA oxidation. Among them, the Co 2+ -coordinated supramolecular nanostructures show the highest catalytic efficiency and enantioselectivity (select factor up to 2.70), while the Fe 3+ -coordinated monomeric ones show nearly racemic products. Analysis of the kinetic results suggests that the synergistic effect between metal ions and the chiral supramolecular NRs can significantly regulate the enantioselective catalytic activity, while the metal ion-mediated monomeric bis - l / d - NapHis were less active. The studies on association constants and activation energies reveal the difference in catalytic efficiency and enantioselectivity resulting from the different energy barriers and binding affinities existed between the chiral molecular/supramolecular structures and R/S -DOPA enantiomers . This work clarifies the correlation between chiral molecular/supramolecular structures and enantioselective catalytic activity, shedding new light on the rational design of chiral catalysts with outstanding enantioselectivity.

Mussel-inspired co-deposited L-DOPA/PEI/CNTs/PVDF membrane with hierarchically gradient pores for efficient oil/water emulsion separation

Bang Li,Juncai Ren,Jiayi Xie,Junfeng Wu,Yingchun Wang,Xiaogang Gu,Xiaoxian Hu,Xiang Guo,Zuwen Liu,Yanbin Yun,Chunli Li,Xiying Hao,Shasha Chen,Shiqiang Yin

The discharge of oily wastewater and industrial wastewater poses serious threats to aquatic ecosystems and human health, highlighting the urgent need for advanced antifouling membranes capable of efficient oil-in-water emulsion separation. Unlike conventional surface coating or single-component modification strategies, herein, we report a facile strategy to transform hydrophobic PVDF membranes into superhydrophilic ones with hierarchically gradient pores. This approach distinctively exploits mussel-inspired L-3,4-dihydroxyphenylalanine (L-DOPA), rather than dopamine, in combination with polyethyleneimine (PEI) co-deposition and carbon nanotubes (CNTs) nanostructuring, forming a highly stable, multifunctional hydrophilic network that anchors CNTs while synergistically regulating pore size, pore distribution, and surface wettability. As a result, the average pore size decreased from 0.4075 μm to 0.2883 μm, enhancing selective permeability. The optimized L-DOPA/PEI/CNTs/PVDF membrane exhibited an ultrahigh pure water flux of 5367.9 L m −2  h −1  bar −1 and achieved over 99 % rejection of diverse surfactant-stabilized emulsions. Moreover, the membrane demonstrated excellent antifouling, consistently maintaining ≥98 % oil rejection over seven cycles, attributed to the formation of a compact hydration layer. By integrating bio-inspired adhesion chemistry with nanostructured pore-gradient engineering, this study provides a scalable and environmentally relevant route for fabricating high-performance separation membranes, offering strong potential for sustainable oily wastewater treatment and resource recovery applications.

A Wearable Paper-Based Patch Integrating Osmotic Sweat Extraction and Differential Sensing for Continuous L-Dopa Monitoring in Parkinson's Disease

Zhiyang Chen,Yu Cai,Shanshan Zhang,Yue Zhou,Yiming Wei,Junhao Li,Rong Xu,Yunbo Gai,Xuesong Ye,Congcong Zhou,Bo Liang

Levodopa (L-Dopa) pharmacokinetic monitoring is crucial for optimizing therapy in Parkinson's disease (PD); however, existing sweat sensors are still limited by their reliance on active perspiration and poor specificity in complex biofluids. Here, we present a passive osmotic differential electrochemical (PODE) patch for continuous, non-invasive L-Dopa monitoring under resting conditions. The patch employs an ethylene glycol-loaded  polyacrylamide/polyvinyl alcohol (PAAm/PVA) hydrogel as a “chemical osmotic pump” to drive sweat transport without external power or physical exertion. For sensitive detection, a three-dimensional paper-based electrode is constructed via in situ growth of acrylamide-doped poly(3,4-ethylenedioxythiophene):poly(styrenesulfonate) (PEDOT:PSS) and gold nanoparticles (AuNPs). To eliminate interferences from uric acid (UA) and ascorbic acid (AA), a tyrosinase-enabled dual-channel differential sensing strategy is employed. The sensor exhibits a wide linear range and maintains high selectivity in complex artificial sweat. By seamlessly integrating passive sampling with interference-rejecting electrochemical detection, this flexible, low-cost patch offers a practical and user-friendly platform for personalized, at-home management of PD.

Horseradish peroxidase-catalyzed polymerization of l-DOPA for mono-/bi-enzyme immobilization and amperometric biosensing of H2O2 and uric acid

Mengzhen Dai,Ting Huang,Long Chao,Qingji Xie,Yueming Tan,Chao Chen,Wenhua Meng

Horseradish peroxidase (HRP)-catalyzed polymerization of l -DOPA (vs. dopamine) in the presence of H 2 O 2 (and uricase (UOx)) was exploited to immobilize mono-/bi-enzymes for hydroquinone-mediated amperometric biosensing of H 2 O 2 and uric acid (UA). The relevant polymeric biocomposites (PBCs) were prepared in phosphate buffer solution containing HRP and l -DOPA (or plus UOx) after adding H 2 O 2 . The mono-/bi-enzyme amperometric biosensors were prepared simply by casting some of the PBCs on Au-plated Au (Au plate /Au) electrodes, followed by coating with an outer-layer chitosan (CS) film for each. UV–vis spectrophotometry , scanning electron microscopy, cyclic voltammetry and electrochemical impedance spectroscopy were used for film characterization and/or process monitoring. The HRP immobilized by enzyme catalysis well preserved its bioactivity, as confirmed by UV–vis spectrophotometry . Under optimized conditions, the monoenzyme CS/HRP-poly( l -DOPA) (PD)/Au plate /Au electrode potentiostated at −0.1   V responded linearly to H 2 O 2 concentration from 0.001 to 1.25   mM with a sensitivity of 700   μA   mM −1   cm −2 and a limit of detection (LOD) of 0.1   μM, and the bienzyme CS/UOx–HRP–PD/Au plate /Au electrode at −0.1   V responded linearly to UA concentration from 0.001 to 0.4   mM with a sensitivity of 349   μA   mM −1   cm −2 and a LOD of 0.1   μM. The mono-/bi-enzyme biosensors based on biosynthesized PD performed better than many reported analogues and those based on similarly biosynthesized polydopamine.

Identification and enhanced activity of the tyrosine phenol-lyase from Mycobacterium tuberculosis

Hengrui Liu,Xiaorong Yang

L-DOPA (3,4-dihydroxyphenyl-L-alanine) is a key drug for the Parkinson's disease. The global demand of L-DOPA is continuously increasing. Tyrosine phenol-lyase (TPL) is the environmentally friendly biocatalyst for L-DOPA biosynthesis. In this study, a novel TPL from Mycobacterium tuberculosis (Mt-TPL) was identified and expressed in Escherichia coli . The characterization of Mt-TPL indicated that the optimal temperature and pH values were 40 °C and 9.0, respectively, and the half-lives (t 1/2 ) at 30 °C were 6.52 h. To further improve its catalytic performance, protein engineering of Mt-TPL was undertaken using a consensus sequence-based strategy. It was found that the I384V mutant exhibited the increase of 40.31 % in the enzymatic activity as compared with the wild-type enzyme. Molecular docking and molecular dynamics (MD) simulations revealed that the I384V mutation changed the local flexibility of enzyme and the orientation of substrate, and shortened the proton transfer distance between the generic acid Y71 and substrate pyrocatechol. Finally, the whole-cell catalysts with wild type and I384V Mt-TPL were used to product the L-DOPA, the L-DOPA production of 95.46 g/L for whole-cells with I384V Mt-TPL was achieved in a 5 L fermenter by the fed-batch, 38.6 % higher than that of the wild type enzyme.

A chiral sensor for recognition of DOPA enantiomers based on immobilization of β-cyclodextrin onto the carbon nanotube-ionic liquid nanocomposite

Chen Ya,Huang Yihan,Guo Dongmei,Chen Cui,Wang Qinghong,Fu Yingzi

A sensitive electrochemical method was explored for chiral recognition of 3,4-dihydroxyphenylalanine (DOPA) enantiomers based on immobilization of β -cyclodextrin ( β -CD) onto the nanocomposite comprising multiwalled carbon nanotubes (MWCNTs) and ionic liquid (IL) 1-butyl-3-methylimidazolium hexafluorophosphate (BIMIMPF 6 ) modified glassy carbon electrode ( β -CD/MWCNTs-IL/GCE). The combination of ionic liquid and carbon nanotubes could create unique nanomaterials which facilitated the electron transfer. In addition, β -CD was brought in to act as the chiral selector for DOPA enantiomers recognition. The immobilization process of β -CD/MWCNTs-IL/GCE was monitored by cyclic voltammetry, electrochemical impedance spectroscopy, and scanning electron microscopy. The chiral interface ( β -CD/MWCNTs-IL/GCE) was employed to discriminate DOPA enantiomers via differential pulse voltammetry. The results indicated that the proposed sensor exhibited a stronger electrochemical response toward D-DOPA over the linear range from 4.0 × 10 −3 to 4.0 × 10 −9  mol L −1 and the detection limit was 1.2 × 10 −9  mol L −1 (S/N = 3). This work provided an available platform for enantioselective recognition of DOPA enantiomers based on the nanocomposite.

Tyrosinase-catalyzed polymerization of L-DOPA (versus L-tyrosine and dopamine) to generate melanin-like biomaterials for immobilization of enzymes and amperometric biosensing

Mengzhen Dai,Ting Huang,Long Chao,Yueming Tan,Chao Chen,Wenhua Meng,Qingji Xie

Learning from nature emerges as one of the most promising ways to develop advanced functional materials and biodevices. Here, inspired by melanin formation, we report the tyrosinase (Tyr)-catalyzed polymerization of L-DOPA (versus L-tyrosine and dopamine) to immobilize enzymes for amperometric biosensing. The enzymatic polymerization is examined by UV-vis spectrophotometry, scanning electron microscopy and electrochemical methods. A poly(L-DOPA) (PD)-Tyr/glassy carbon electrode (GCE) prepared by casting an aqueous mixture of L-DOPA and Tyr on a GCE exhibits a linear cathodic amperometric response to catechol concentration from 0.4 to 57 μM (R2 = 0.997) with a sensitivity of 4.29 mA mM−1 cm−2 and a limit of detection (LOD) of 70 nM (S/N = 3). A PD-glucose oxidase (GOx)-Tyr/Pt electrode prepared by casting an aqueous mixture of L-DOPA, GOx and Tyr on a Pt electrode exhibits a linear anodic amperometric response to glucose concentration from 2 to 5700 μM (R2 = 0.998) with a sensitivity of 78.6 μA mM−1 cm−2 and a LOD of 0.1 μM (S/N = 3). The PD-based enzyme electrode shows better biosensing performance and higher bioactivity of the immobilized Tyr than those based on similarly biosynthesized poly(L-tyrosine) and polydopamine as well as well-established chitosan and Nafion systems, implying that the biosynthesized PD as a melanin-biomimetic material has promising application potential for biomacromolecular immobilization and biosensing.
