Fabrication of gelatin-EGCG-pectin ternary complex stabilized W/O/W double emulsions by ultrasonic emulsification: Physicochemical stability, rheological properties and structure

Xiang Huang,Rui Tu,Hongbo Song,Kai Dong,Fang Geng,Lei Chen,Qun Huang,Yingmei Wu

Double emulsions have very promising application prospects in the pharmaceutical, cosmetic, and food industries. Nevertheless, its usage in the food industry is restricted mainly due to its immense stability problems. Here, W 1 /O/W 2 double emulsion was prepared by two-step emulsification method using ultrasonic emulsification technology. The physicochemical stability, rheological properties, thermal stability, and microstructure of the double emulsion were investigated. The results revealed that W 1 /O/W 2 double emulsion with optimum stability was prepared on the parameters that the ratio of W 1 (gelatin)/O primary emulsion to the external aqueous phase (gelatin-EGCG-high methoxyl pectin ternary complex) was 6:4, ultrasonic power was 80 W, ultrasonic time was 6 min, and ternary complex concentration was 0.75%. The double emulsion produced under these conditions had the smallest particle size (1143 nm) and the maximum absolute zeta potential value (39.17 ± 0.50 mV). The results of full-featured stability analyzer manifested that the emulsion system no longer underwent phase separation. Confocal laser scanning microscopy (CLSM) successfully observed a uniformly dispersed double emulsion. Furthermore, dynamic rheological properties demonstrated that the cohesive strength of W 1 /O/W 2 double emulsion was strength than that of W 1 /O primary emulsion. Differential scanning calorimetry (DSC) analysis suggested that both remained stable throughout the temperature change, while the W 1 /O/W 2 double emulsion was more thermally stable. Hence, the gelatin-EGCG-high methoxyl pectin ternary complex can act as an effective emulsion stabilizer to stabilize the W 1 /O/W 2 double emulsion.

Fabrication of Zein-Lecithin-EGCG complex nanoparticles: Characterization, controlled release in simulated gastrointestinal digestion

Hujun Xie,Chengzhi Liu,Jian Gao,Jieyu Shi,Fangfang Ni,Xin Luo,Ying He,Gerui Ren,Zisheng Luo

The Zein-Lecithin-Epigallocatechin gallate (EGCG) complex nanoparticles were fabricated by anti-solvent coprecipitation method. The Zein-Lecithin (Z-L) nanocomplexes exhibited great encapsulation efficiency of 68.5% for EGCG, and the encapsulated EGCG still had good antioxidative capacity. The cumulative release of EGCG in simulated gastric fluid (SGF) and simulated intestinal fluid (SIF) were 19% and 92%, respectively, and the release was closest to Fick release in SGF and First release in SIF. Fluorescence spectroscopy (FL), Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD) experiments revealed that the EGCG was successfully encapsulated by Z-L nanocomplexes through electrostatic, hydrophobic and hydrogen bonding interactions. The Zein-Lecithin-EGCG complex nanoparticles exhibited excellent stability and great sustained-release performance, which will be the alternative for potential application in the food industry.

Enhancing the storage and gastrointestinal passage viability of probiotic powder (Lactobacillus Plantarum) through encapsulation with pickering high internal phase emulsions stabilized with WPI-EGCG covalent conjugate nanoparticles

Xin-Sheng Qin,Qun-Yu Gao,Zhi-Gang Luo

In this study, we demonstrated the role of Pickering high internal phase emulsions (HIPEs) stabilized with the whey protein isolate (WPI)/(-)-epigallocatechin-3-gallate (EGCG) covalent conjugates on the storage and GIT passage viability of probiotics powder ( Lactobacillus Plantarum ). At pH 9 value, the WPI-EGCG covalent conjugates by free-radical induction reaction formed hydroxyquinone or hydroxyquinone dimers with better wettability, and digestive enzyme inhibition effect and smaller particle size. Then, the WPI-EGCG covalent conjugates were used to stabilize the O/W type Pickering emulsion. The oil fraction ( ϕ ) addition led to an increase in the droplet size and modulus of Pickering emulsions toward to generate gel-like structure. On the other hand, at a constant ϕ value, the droplet size of these Pickering HIPEs reduced with increasing particle concentration ( c ) from 0.5% to 5%, indicating that high c can stabilize more oil droplets. Encapsulation of Lactobacillus Plantarum powder with the Pickering HIPEs was successful to enhance the viable cell count after 14 days storage and GIT digestion. The protective effect of Pickering emulsions was improved with the addition of the c and/or ϕ value. Therefore, Pickering HIPEs can be applied as a promising carrier for enhancing the viability of probiotic powder during storage and GIT digestion by reason of the high oil phase fraction and the adsorption of the WPI-EGCG covalent conjugates on the droplet interface.

EGCG protects against homocysteine-induced human umbilical vein endothelial cells apoptosis by modulating mitochondrial-dependent apoptotic signaling and PI3K/Akt/eNOS signaling pathways

Liu Shumin,Sun Zhengwu,Chu Peng,Li Hailong,Ahsan Anil,Zhou Ziru,Zhang Zonghui,Sun Bin,Wu Jingjun,Xi Yalin,Han Guozhu,Lin Yuan,Peng Jinyong,Tang Zeyao

Homocysteine (Hcy) induced vascular endothelial injury leads to the progression of endothelial dysfunction in atherosclerosis. Epigallocatechin gallate (EGCG), a natural dietary antioxidant, has been applied to protect against atherosclerosis. However, the underlying protective mechanism of EGCG has not been clarified. The present study investigated the mechanism of EGCG protected against Hcy-induced human umbilical vein endothelial cells (HUVECs) apoptosis. Methyl thiazolyl tetrazolium assay (MTT), transmission electron microscope, fluorescent staining, flow cytometry, western blot were used in this study. The study has demonstrated that EGCG suppressed Hcy-induced endothelial cell morphological changes and reactive oxygen species (ROS) generation. Moreover, EGCG dose-dependently prevented Hcy-induced HUVECs cytotoxicity and apoptotic biochemical changes such as reducing mitochondrial membrane potential (MMP), decreasing Bcl-2/Bax protein ratio and activating caspase-9 and 3. In addition, EGCG enhanced the protein ratio of p-Akt/Akt, endothelial nitric oxide synthase (eNOS) activation and nitric oxide (NO) formation in injured cells. In conclusion, the present study shows that EGCG prevents Hcy-induced HUVECs apoptosis via modulating mitochondrial apoptotic and PI3K/Akt/eNOS signaling pathways. Furthermore, the results indicate that EGCG is likely to represent a potential therapeutic strategy for atherosclerosis associated with Hyperhomocysteinemia (HHcy).

Fabrication of chitosan-coated epigallocatechin-3-gallate (EGCG)-hordein nanoparticles and their transcellular permeability in Caco-2/HT29 cocultures

Hongdong Song,Aijing He,Xiao Guan,Zhengyu Chen,Yize Bao,Kai Huang

Epigallocatechin-3-gallate (EGCG) has gained appreciable attention because of its health benefits. However, the poor permeability across the intestine limits its use. In this study, we have fabricated chitosan-coated EGCG-hordein nanoparticles (Cs-EHNs), with the aim to enhance the intestinal permeability of EGCG. Cs-EHNs were fabricated by layer-by-layer electrostatic stacking method, and its uptake and transcellular permeability were studied in the Caco-2/HT29 co-culture model. The constructed Cs-EHNs had the average diameter of 296 nm, polymer dispersity index (PDI) of 0.30, zeta potential of 59.6 mV, and showed a spherical morphology. Encapsulation efficiency of EGCG was 87.3%. The transcellular permeability experiments indicated that the apparent permeability coefficient (P app ) of Cs-EHNs was higher than that of free EGCG. Furthermore, the cellular uptake of Cs-EHNs was studied by specific endocytosis inhibitors, and results showed that the uptake mechanisms of Cs-EHNs were through caveolae-mediated endocytosis and macropinocytosis. This study demonstrated that encapsulation of EGCG using chitosan-coated hordein nanoparticles could be a promising approach to improve the absorption of EGCG.

Synthesis of novel EGCG-glucose conjugates and studies of their antioxidative properties for neuroprotections

Jiaqi Lu,Jinxia Chen,Chunyan Yang,Chen Xia,Junling Deng,Zuoya Xiang,Jian Chen,Yong Wu

Epigallocatechin-3-gallate (EGCG) is a polyphenol naturally sourced in green tea with extensive bioactivities, including antioxidant, anti-carcinogenic, anti-microbial, anti-obesity, and neuroprotection. However, EGCG is unstable in neutral or alkaline conditions and has poor lipid solubility , leading to inadequate systemic absorption, reduced pharmacokinetics , limited biodistribution , and first-pass metabolism. The aim of this study is to solve these problems, thus two novel EGCG-glucose conjugates , Glu-EGCG and 2Glu-EGCG possessing one and two glucose, respectively, were designed, synthesized, and characterized. The EGCG-glucose conjugates’ chemical stability, cytotoxicity and alleviation H 2 O 2 -induced antioxidative damage to neuronal cell and antioxidative neuroprotection activities were examined. Compared with their precursor EGCG, Glu-EGCG and 2Glu-EGCG are more stable, less toxic to PC12 and bEnd.3 cell, stronger ability to attenuate H 2 O 2 -induced injury to PC12 cells resulting higher cell viability and lower reactive oxygen species (ROS) in the cell. Animal study results further proved that Glu-EGCG and 2Glu-EGCG possess higher antioxidative activities than EGCG, through decreasing the ischemia area, inhibiting morphological changes and neuronal loss in brain tissues, and modulating the oxidative factor and antioxidant factor. In conclusion, 2Glu-EGCG shows stronger antioxidant activities than Glu-EGCG does. Both EGCG-glucose conjugates are promising brain target antioxidative neuroprotection agents.

Development of Pickering water-in-oil emulsions using a dual stabilization of candelilla wax and acylated EGCG derivatives to enhance the survival of probiotics (Lactobacillus plantarum) powder

Zhaojuan Shi,Jun Wu,Xingting Wang,Tongqiao Nie,Qingmei Zeng,Chuanxun Yuan,Risheng Jin

Probiotics have considerable interest due to their inseparable link to human health. However, probiotic products are seriously challenged during processing, preservation, and intake. Food-grade probiotic delivery systems need to be further explored as an effective way to enhance cell viability. In this study, water-in-oil (W/O) Pickering emulsions were fabricated by adding candelilla wax (CLW) as a network stabilizer based on acylated EGCG derivatives in the crystalline form as a Pickering stabilizer. The effects of acylated EGCG derivatives’ concentration, CLW concentration, and oil phase volume fraction on the droplet size distribution, microstructure, and physical stability of Pickering emulsions were explored. The presence of CLW reduced the particle size and improved the physical stability of acylated EGCG-based emulsions, and the effect was more positive with increasing concentration. The protective effect of emulsions with different oil phase volume fractions on Lactobacillus plantarum during freeze-thaw cycles, storage, and gastrointestinal digestion was evaluated. The outer-phase physical barrier of W/O emulsions co-stabilized with acylated EGCG derivatives and CLW facilitated the sensitivity of probiotics to ice crystal growth, temperature changes, acidic environments, and digestive enzymes. The emulsions formulated with 40% oil phase volume fractions allowed Lactobacillus plantarum to survive up to 7.75 log CFU g−1 in the harsh gastrointestinal environment. The results offer promising strategies for applying W/O emulsion probiotic delivery systems in food processing, storage, and oral administration.

Injectable and tissue adhesive EGCG-laden hyaluronic acid hydrogel depot for treating oxidative stress and inflammation

Zhichao He,Huitong Luo,Zetao Wang,Dafu Chen,Qi Feng,Xiaodong Cao

Oxidative stress and inflammation are common pathological mechanisms for the progression of tissue degeneration. Epigallocatechin-3-gallate (EGCG) features antioxidant and anti-inflammatory properties, which is a promising drug for the treatment of tissue degeneration. Herein, we utilize the phenylborate ester reaction of EGCG and phenylboronic acid (PBA) to fabricate an injectable and tissue adhesive EGCG-laden hydrogel depot (EGCG HYPOT), which can achieve anti-inflammatory and antioxidative effects via smart delivery of EGCG. Specifically, the phenylborate ester bonds, formed by EGCG and PBA-modified methacrylated hyaluronic acid (HAMA-PBA), endow EGCG HYPOT injectability, shape adaptation and efficient load of EGCG. After photo-crosslinking, EGCG HYPOT exhibits good mechanical properties, tissue adhesion and sustained acid-responsive release of EGCG. EGCG HYPOT can scavenge oxygen and nitrogen free radicals . Meanwhile, EGCG HYPOT can scavenge intracellular reactive oxygen species (ROS) and suppress the expression of pro-inflammatory factors. EGCG HYPOT may provide a new idea for alleviation of inflammatory disturbance.

Evaluation of the neuroprotective effect of EGCG: a potential mechanism of mitochondrial dysfunction and mitochondrial dynamics after subarachnoid hemorrhage

Ying Chen,Jianjun Chen,Xiaoxue Sun,Xiaoxiao Shi,Lei Wang,Liyong Huang,Wenke Zhou

(−)-Epigallocatechin-3-gallate (EGCG), the main bioactive component of tea catechins, exhibits broad-spectrum health efficacy against mitochondrial damage after subarachnoid hemorrhage (SAH). The mechanisms, however, are largely unknown. Here, the ability of EGCG to rescue mitochondrial dysfunction and mitochondrial dynamics following the inhibition of cell death was investigated by using in vitro and in vivo SAH models. EGCG blocked the cytosolic channel ([Ca2+])i influx via voltage-gated calcium channels (VGCCs), which induced mitochondrial dysfunction, including mitochondrial membrane potential depolarization and reactive oxygen species (ROS) release. As expected, EGCG ameliorated oxyhemoglobin (OxyHb)-induced impairment of mitochondrial dynamics by regulating the expression of Drp1, Fis1, OPA1, Mfn1, and Mfn2. As a result, EGCG restored the increases in fragmented mitochondria and the mtDNA copy number in the OxyHb group to almost the normal level after SAH. In addition, the normal autophagic flux induced by EGCG at both the initiation and formation stages regulated Atg5 and Beclin-1 after SAH for the timely elimination of damaged mitochondria. In the end, EGCG increased the neurological score by decreasing cell death through the cyt c-mediated intrinsic apoptotic pathway. The results revealed the mechanisms behind the neuroprotective effects of EGCG via inhibition of the overloaded [Ca2+]i-induced mitochondrial dysfunction and the imbalanced mitochondrial fusion and fission cycle. Therefore, the simultaneous inhibition and timely elimination of damaged mitochondria could determine the therapeutic effect of EGCG.

ROS scavenging and immunoregulative EGCG@Cerium complex loaded in antibacterial polyethylene glycol-chitosan hydrogel dressing for skin wound healing

Jing Ye,Qinghua Li,Yushan Zhang,Qi Su,Zujian Feng,Pingsheng Huang,Chuangnian Zhang,Yinglei Zhai,Weiwei Wang

The elevation of oxidative stress and inflammatory response after injury remains a substantial challenge that can deteriorate the wound microenvironment and compromise the success of wound healing. Herein, the assembly of naturally derived epigallocatechin-3-gallate (EGCG) and Cerium microscale complex ( [email protected] ) was prepared as reactive oxygen species (ROS) scavenger, which was further loaded in antibacterial hydrogels as wound dressing. [email protected] shows superior antioxidation capacity towards various ROS including free radical, O 2 − and H 2 O 2 through superoxide dismutase-like or catalase-mimicking catalytic activity . Importantly, [email protected] could provide mitochondrial protective effect against oxidative stress damages, reverse the polarization of M1 macrophages and reduce the secretion of pro-inflammatory cytokines. Furtherly, [email protected] was loaded into the PEG-chitosan hydrogel with dynamic, porous, injectable and antibacterial properties as wound dressing, which accelerated the regeneration of both epidermal layer and dermis, resulting in improved healing process of full-thickness skin wounds in vivo . Mechanistically, [email protected] re-shaped the detrimental tissue microenvironment and augmented the pro-reparative response through reducing ROS accumulation, alleviating inflammatory response, enhancing the M2 macrophage polarization and angiogenesis . Collectively, antioxidative and immunomodulatory metal-organic complex-loaded hydrogel is a promising multifunctional dressing for the repair and regeneration of cutaneous wounds without additional drugs, exogenous cytokines, or cells. Statement of significance (1) We reported an effective antioxidant through self-assembly coordination of EGCG and Cerium for managing the inflammatory microenvironment at the wound site, which not only showed high catalytic capacity towards multiple ROS, but also could provide mitochondrial protective effect against oxidative stress damage, reverse the polarization of M1 macrophages and downregulate pro-inflammatory cytokines. [email protected] was further loaded into porous and bactericidal PEG-chitosan (PEG-CS) hydrogel as a versatile wound dressing, which accelerated wound healing and angiogenesis. (2) The applicability of alleviating sustainable inflammation and regulating macrophage polarization through ROS scavenging is a promising strategy for tissue repair and regeneration without additional drugs, cytokines, or cells.

A whole-course-repair system based on ROS/glucose stimuli-responsive EGCG release and tunable mechanical property for efficient treatment of chronic periodontitis in diabetic rats

Qingchen Feng,Mei Zhang,Guanning Zhang,Hongxiang Mei,Chongying Su,Lisa Liu,Xiaoxia Wang,Ziqianhong Wan,Zhengyi Xu,Liangkui Hu,Yu Nie,Juan Li

Elevated glucose levels, multiple pro-inflammatory cytokines and the generation of excessive reactive oxygen species (ROS) are pivotal characteristics within the microenvironments of chronic periodontitis with diabetes mellitus (CPDM). Control of inflammation and modulation of immune system are required in the initial phase of CPDM treatment, while late severe periodontitis requires a suitable scaffold to promote osteogenesis, rebuild periodontal tissue and reduce alveolar bone resorption. Herein, a whole-course-repair system is introduced by an injectable hydrogel using phenylboronic acid functionalized oxidized sodium alginate (OSA–PBA) and carboxymethyl chitosan (CMC). Epigallocatechin-3-gallate (EGCG) was loaded to simultaneously adjust the mechanical property of the OSA–PBA/CMC + EGCG hydrogel (OPCE). This hydrogel has distinctive adaptability, injectability, and ROS/glucose-triggered release of EGCG, making it an ideal drug delivery carrier. As expected, OPCE hydrogel shows favourable antioxidant and anti-inflammatory properties, along with a regulatory influence on the phenotypic transition of macrophages, providing a favourable immune microenvironment. Apart from that, it provides a favourable mechanical support for osteoblast/osteoclast differentiation regulation at the late proliferation stage of periodontal regeneration. The practical therapeutic effects of OPCE hydrogels were also confirmed when applied for treating periodontitis in diabetic rats. In summary, OPCE hydrogel may be a promising whole-course-repair system for the treatment of CPDM.
