The protective effect of rutin on sciatic nerve injury in acrylamide-exposed rats and its mechanisms

Suqiu Dong,Chunmei Zhang,Yunjue Wang,Shuping Liu,Junhua Yang,Lixia Li,Yuxin Ma,Jing Liu

Rutin (Rut) is a flavonoid with pharmacological activities such as anti-inflammatory and antioxidant. Acrylamide (ACR) is a toxic substance widely found in human life that can induce neurotoxicity. Some studies have confirmed that neurotoxicity caused by ACR induces myelin damage, which in turn causes neurological dysfunction. Therefore, we established a rutin intervention model to investigate the protective effect of Rut on ACR-induced sciatic nerve injury in rats and its mechanism. The results showed that superoxide dismutase (SOD) activity and glutathione (GSH) content increased and lactate dehydrogenase (LDH) activity decreased in the middle and high dose groups of Rut compared with the ACR group, and the expression of Myelin basic protein (MBP), Extracellular-regulated kinase 1/2(ERK1/2), Phosphorylated extracellular regulated kinase 1/2 (P-ERK1/2), and Nuclear factor E−2-associated factor (Nrf2) was promoted in the Rut-protected group, which suggests that Rutin has a protective effect on ACR-induced sciatic nerve injury and that the mechanism of Rutin's protective effect is related to activation of the ERK1/2 pathway and alleviation of oxidative stress injury.

Janus Piezoelectric Fibrous Scaffolds with Ultrasound-Triggered Electroresponsive Drug Release for Dual-Lineage Induction and Immune Microenvironment Remodeling in Tendon-Bone Interface Regeneration

Chengzhi Liang,Fanrong Ai,Huajun Pan,Fengyang Cui,Hang Fu,PinKai Wang,Yudan Zhu,Weixiang Xiong,Dewei Qiu,Jun Tao

A rotator cuff tear (RCT) is a common sports injury. Current surgical interventions frequently result in suboptimal healing and fail to regenerate the complex, heterogeneous tendon–bone interface. Piezoelectric materials, which convert ultrasound-derived micromechanical energy into electrical signals, offer a potential means to overcome the limitations of conventional electrical stimulation therapies for musculoskeletal tissue repair. Moreover, active and controlled drug release has become increasingly essential in modern regenerative strategies. We developed a Janus bilayered fibrous scaffold (KZP@PS/PSPM) with dual-phase induction and immunomodulatory capabilities using electrospinning and oxidative polymerization. Under ultrasonic exposure, the underlying PSPM fibrous membrane transduces mechanical energy into electrical signals that stimulate osteogenic differentiation in bone marrow mesenchymal stem cells (BMSCs). Concurrently, the upper KZP@PS layer converts mechanical energy into electrical signals, activating the electrochemical reduction of polyaniline (PANI) and inducing a stress-responsive release of KZF, which secretes zinc ions (Zn 2 ⁺) and kartogenin (KGN) to promote immunomodulation and chondrogenesis. This process further enhances osteogenic and chondrogenic differentiation through upregulation of the ERK1 and PI3K–Akt signaling pathways. In a rotator cuff injury model, the KZP@PS/PSPM composite scaffold significantly reduced early inflammatory markers and markedly improved tendon-to-bone healing. By synergizing “ultrasound–electricity” with “electricity–drug release,” this Janus bilayer scaffold enables biphasic induction and immunomodulation, offering a novel strategy for addressing the complex challenges of tendon–bone interface regeneration.

Chondroitin sulfate/silk fibroin hydrogel incorporating graphene oxide quantum dots with photothermal-effect promotes type H vessel-related wound healing

Tanjun Deng,Wenli Lu,Xiaoxian Zhao,Haoyu Wang,Yumeng Zheng,Ao Zheng,Zhengyu Shen

Chronic wounds with bacterial infection present formidable clinical challenges. In this study, a versatile hydrogel dressing with antibacterial and angiogenic activity composite of silk fibroin (SF), chondroitin sulfate (CS), and graphene oxide quantum dots (GOQDs) is fabricated. GOQDs@SF/CS (GSC) hydrogel is rapidly formed through the enzyme catalytic action of horseradish peroxidase. With the incorporation of GOQDs both gelation speed and mechanical properties have been enhanced, and the photothermal characteristics of GOQDs in GSC hydrogel enabling bacterial killing through photothermal treatment (PTT) at ~51 °C. In vitro studies show that the GSC hydrogels demonstrate excellent antibacterial performance and induces type H vessel differentiation of endothelial cells via the activated ERK1/2 signaling pathway and upregulated SLIT3 expression. While in vivo results show that the hydrogel significantly promotes type H vessels formation, which related to the collagen deposition, epithelialization and, ultimately accelerating the regeneration of infected skin defects. Collectively, this multifunctional GSC hydrogel, with dual action of antibacterial efficacy and angiogenesis promotion, emerges as an innovative skin dressing with potential for advancing in infected wound healing.

Protective effects of Lactococcus lactis subsp. lactis HFY14 supplementation on the brain, intestines, and motor function of antibiotic-treated mice

Yang Yang,Zhao Yuanji,Lei Huan

Introduction: This study aimed to explore the anti-oxidative and anti-inflammatory properties of Lactococcus lactis subsp. lactis HFY14 (LLSLHFY14) and investigate its effects on the intestinal barrier, cranial nerve, and motor function in mice treated with antibiotics. Methods: Mice were administered an antibiotic mixture (neomycin 5 mg/mL, vancomycin 25 mg/mL, amphotericin B 0.1 mg/mL, ampicillin 10 mg/mL, metronidazole file 5 mg/mL, and lipopolysaccharide 1.5 μg/mL) intraperitoneally, and oxidative stress and inflammatory markers in the serum and brain tissues, and liver index were measured. H&E staining was performed to detect pathological alterations in brain tissues. The expression of intestinal-barrier-related genes and that of genes involved in inflammatory pathways in the brain were detected using polymerase chain reaction (PCR).Results: LLSLHFY14 administration extended the weight-loaded swimming and running times of mice and decreased the liver index. Moreover, the levels of malondialdehyde (MDA), interleukin-6 (IL-6), and tumor necrosis factor alpha (TNF-α) in the serum and brain tissue were reduced, whereas those of superoxide dismutase (SOD), glutathione (GSH), and interleukin-10 (IL-10) were elevated. Elevated brain expression of the protein kinase B (AKT)/cAMP-response element binding protein (CREB)/brain-derived neurotrophic factor (BDNF)/extracellular signal-regulated kinase 1 (ERK1) pathway, decreased brain expression of the IL-6 gene, and elevated cecum expression of zonula occludens-1 (ZO-1), occludin-1, and claudin-1 genes were noted. LLSLHFY14 supplementation significantly increased Bacteroidetes expression but decreased Firmicutes expression, thus increasing the Bacteroidetes/Firmicutes ratio.Discussion: Overall, LLSLHFY14 supplementation ameliorated antibiotic-induced oxidative stress and inflammation in the mouse central nervous system, intestinal barrier dysfunction, and increased motor function, thus confirming its potential application as probiotics.
