Intensive poultry farming has significantly increased the incidence of lipid metabolic disorders, severely compromising the economic benefits of poultry industry. Currently, gamma-aminobutyric acid (GABA) is primarily used to mitigate adverse effects of heat stress in poultry, while the effects and mechanisms of GABA on lipid metabolism disorders remain underexplored. Lactobacillus plantarum ( L. plantarum ) serves as a significant source of GABA and is widely used in the livestock industry. This study therefore examines the effects of postbiotic GABA and the GABA-producing probiotic L. plantarum 1-2-3 on abdominal adipose tissue of laying hens following corticosterone-induced stress. To this end, hens subjected to corticosterone subcutaneous injections (4 mg/kg of body weight) were respectively received GABA (100 mg/kg BW) or L. plantarum 1-2-3 (1 × 10⁹ CFU/day). Results demonstrated that both GABA and L. plantarum 1-2-3 alleviated corticosterone-induced lipid metabolism disorders and reduced adipocyte size in abdominal fat. Additionally, expression analyses of genes and proteins related to lipid metabolism (PPARγ, C/EBPα, CD36, LPL, ATGL, and HSL) further showed that GABA and L. plantarum 1-2-3 inhibited excessive deposition of abdominal lipids in laying hens by suppressing adipogenesis and lipogenesis, while promoting lipolysis. Moreover, GABA and L. plantarum 1-2-3 both mitigated lipid deposition-induced inflammation and oxidative damage by normalizing macrophage infiltration and improving antioxidative enzyme activities (GSH-Px, T-SOD, CAT). These findings demonstrate the efficacy of GABA and L. plantarum 1-2-3 in alleviating lipid metabolism disorders in the abdominal adipose tissue of laying hens, suggesting their promise as nutritional supplements for counteracting stress-induced metabolic dysfunction.

Cyromazine (CYR) is a widely used insect growth regulator. Its metabolite, melamine, is classified as a Group 2B carcinogen and has been associated with urinary tract lesions and tumors. While previous toxicological studies have largely focused on high-concentration exposures, the mechanisms of toxicity at environmentally relevant concentrations remain poorly understood. In this study, using Caenorhabditis elegans as a model, we demonstrated that CYR induces dose-dependent neurotoxicity, developmental impairment, reproductive toxicity, and oxidative stress. Transcriptomic analysis revealed disruptions in glutamate, dopamine, GABA, and glutathione metabolic pathways. CYR exposure caused damage to dopaminergic and glutamatergic neurons, decreased levels of several neurotransmitters, and downregulated key genes including dop-3 , tph-1 , and eat-4 , resulting in behavioral deficits such as impaired pharyngeal pumping, body bending, and head thrashing. Parental exposure to CYR significantly elevated reactive oxygen species (ROS) and lipofuscin accumulation, activated the glutathione-mediated antioxidant pathway, and disturbed enzymatic balance, indicating a central role for oxidative stress in CYR-induced neurotoxicity. By systematically elucidating the mechanism of ROS-mediated oxidative stress in neurobehavioral toxicity, this study provides a mechanistic foundation for understanding the broader toxicological profile of CYR, thereby offering critical insights for assessing its comprehensive health risks at environmental levels.

Gelsemium elegans ( G. elegans ) is an extremely poisonous plant that is widely distributed in southern China and southeastern Asia. G. elegans poisoning events occur frequently in southern China, and are therefore an urgent public health problem requiring multidisciplinary action. However, the toxic components and toxicological mechanisms remain unclear. Here, we describe a systematic investigation on the toxic components of G. elegans , resulting in the isolation and identification of 120 alkaloids. Based on acute toxicity screening, the structure–toxicity relationship of Gelsemium alkaloids was proposed for the first time. Moreover, gelsedine- and humantenine-type alkaloids were detected in the clinical blood sample, and were confirmed to be causative in the poisoning. The most toxic compound, gelsenicine ( 1 ), had selective inhibitory effects toward ventral respiratory group (VRG) neurons in the medulla, which is the main brain region controlling respiration in the central nervous system. Gelsenicine ( 1 ) strongly inhibited the firing of action potentials in VRG neurons through its ability to stimulate GABA A receptors, the main receptors involved in inhibitory neurotransmission. Application of GABA A receptor antagonists successively reversed action potential firing in gelsenicine ( 1 )-treated VRG neurons. Importantly, the GABA A receptor antagonists securinine and flumazenil significantly increased the survival of poisoned animals. Our findings provide insight into the components and mechanisms of G. elegans toxicity, and should assist the development of effective emergency treatments for G. elegans poisoning.

γ-Aminobutyric acid (GABA) has several physiological functions and broad application prospects. In this study, as a tool used for transcriptomics , RNA-seq sequencing was employed to determine the mechanism by which GABA slows browning and induces resistance against the growth of bacterial in fresh-cut apples. GABA altered the expression level of genes related to the synthesis of browning enzymes and phenolic substances in fresh-cut apples, which slowed the browning process during storage. Additionally, GABA promoted the gene expression of enzymes or proteins related to disease resistance, which resultantly induced disease resistance in fresh-cut apples. Notably, GABA maintained the integrity and stability of the cell wall composition and structure in apple tissues to reduce browning and induce disease resistance. Our findings revealed the mechanism by which GABA acts as a protective agent against browning and bacterial growth in for fresh-cut apples.

Stress is a well-established disruptor of reproductive function in poultry. Gamma-aminobutyric acid (GABA), a notable postbiotic derived from Lactobacillus plantarum ( L. plantarum ), has attracted increasing attention as a dietary supplement. While existing research predominantly focuses on GABA applications in broilers and other species, its effects on laying hens remain underexplored. This study investigated whether GABA and L. plantarum 1-2-3 supplementation mitigated corticosterone (CORT)-induced chronic stress in laying hens. Results demonstrated that both GABA and L. plantarum 1-2-3 effectively mitigated CORT-induced declines in laying performance and ovarian damage. They reduced ovarian oxidative stress, inflammation, and atretic follicles by enhancing antioxidant capacity and normalizing expression of inflammatory factors ( IL18 , LITAF , IL6 , IL1B) , and apoptosis-related factors ( BCL2 , BAX , CASP9 , CASP3 ). Furthermore, GABA and L. plantarum 1-2-3 markedly increased the serum reproductive hormone levels, as well as the transcription level of ovarian steroidogenic genes ( LHCGR, FSHR, StAR, CYP11A1 ), while modulated ovarian cholesterol metabolism via PPARα-mediated pathways. These findings elucidate the mechanisms by which GABA and L. plantarum 1-2-3 alleviate stress in laying hens, providing a theoretical and practical basis for their use as functional feed additives in stress-exposed flocks.

Parvalbumin interneurons belong to the major types of GABAergic interneurons. Although the distribution and pathological alterations of parvalbumin interneuron somata have been widely studied, the distribution and vulnerability of the neurites and fibers extending from parvalbumin interneurons have not been detailly interrogated. Through the Cre recombinase-reporter system, we visualized parvalbumin-positive fibers and thoroughly investigated their spatial distribution in the mouse brain. We found that parvalbumin fibers are widely distributed in the brain with specific morphological characteristics in different regions, among which the cortex and thalamus exhibited the most intense parvalbumin signals. In regions such as the striatum and optic tract, even long-range thick parvalbumin projections were detected. Furthermore, in mouse models of temporal lobe epilepsy and Parkinson’s disease, parvalbumin fibers suffered both massive and subtle morphological alterations. Our study provides an overview of parvalbumin fibers in the brain and emphasizes the potential pathological implications of parvalbumin fiber alterations.

To investigate the effect of fermented black barley on cooking oil fume (COF)-induced lung injury, male ICR mice were randomized into five groups: normal control (NC), fermented black barley treatment (NF), COF exposure (O), COF + fermented black barley treatment (OF) and COF + Lactobacillus treatment (OL). The exposure of mice to COF was performed for 5 min per day and 4 days per week for a total of 9 weeks, and the mice in the OF, NF and OL groups were administered fermented black barley or Lactobacillus continuously for 9 weeks (1 mL/100 g). Our results showed that the gamma-aminobutyric acid (GABA), total phenolic, and flavonoid contents significantly increased after fermentation ( P < 0.01). In addition, fermented black barley significantly increased SOD activity in the lung tissue, decreased the wet pulmonary coefficient, inhibited the reduction of microbial diversity and richness, and upregulated genes involved in cilium assembly and the cilium axoneme. These findings support the notion that fermented black barley can ameliorate COF-induced lung injury in mice.

In a rotenone-induced Parkinson’s disease (PD) rat model, behavioral investigation, pathological examination, inflammatory factor analysis, and mitochondrial structure and function investigation verified the anti-PD efficacy of nardosinone. A combined transcriptome and proteome analysis proposed that the anti-PD target of nardosinone is the slc38a2 gene and may involve the GABAergic synaptic pathway and cAMP-signaling pathway. Analysis of targeted slc38a2 knockout cells and expression of key enzyme-encoding genes in both pathways verified the target and pathways proposed by the ‘omics analysis. This further confirms that nardosinone can regulate the slc38a2 gene, a potential new target for the treatment of Parkinson's disease, and plays an anti-PD role through the GABAergic synaptic and cAMP pathways.

Ultrasound and slightly acidic electrolyzed water (SAEW) treatments have been widely applied to enrich γ-Aminobutyric Acid (GABA) during the germination of grains and legumes; however, the effects and underlying mechanisms of their combined treatment on GABA enrichment remain unclear. After 72 h of germination, this study investigated the impacts of different treatment methods—distilled water treatment (GM), ultrasound treatment (UGM), SAEW treatment (SGM), and combined treatment (USGM)—on key enzyme activities, protein structural characteristics, and the GABA metabolic pathway in germinated mung beans. The results indicated that the combined treatment of ultrasound and SAEW promoted GABA enrichment in germinated mung beans, with the GABA content reaching 6.25 ± 0.025 mg/g. Additionally, it significantly enhanced the activities of glutamate decarboxylase (GAD), diamine oxidase (DAO), and polyamine oxidase (PAO) to 0.359 ± 0.008 U/g, 0.554 ± 0.021 U/g, and 1.265 ± 0.036 U/g, respectively ( p < 0.05). Fourier-transform infrared spectroscopy and intrinsic fluorescence spectroscopy analyses revealed that the combined treatment significantly increased the content of α-helix and β-turn structures in the protein secondary structure ( p < 0.05), while reducing the proportions of β-sheet and random coil structures, and enhanced protein surface hydrophobicity. Untargeted metabolomics analysis further demonstrated that the combined treatment significantly upregulated the levels of amino acid-related compounds, with a strong positive correlation observed between Glu and GABA content. Moreover, it primarily facilitated GABA synthesis by modulating pathways such as arginine-proline metabolism, arginine biosynthesis, alanine, aspartate, and glutamic acid metabolism, and butanoate metabolism. The findings of this study provide theoretical support for elucidating the molecular mechanisms by which ultrasound and SAEW promote GABA enrichment in mung beans and offer new insights for developing functional mung bean foods rich in GABA..

Physical exercise has beneficial effect on anxiety disorders, but the underlying molecular mechanism remains largely unknown. Here, it is demonstrated that physical exercise can downregulate the S-nitrosylation of gephyrin (SNO-gephyrin) in the basolateral amygdala (BLA) to exert anxiolytic effects. It is found that the level of SNO-gephyrin is significantly increased in the BLA of high-anxiety rats and a downregulation of SNO-gephyrin at cysteines 212 and 284 produced anxiolytic effect. Mechanistically, inhibition of SNO-gephyrin by either Cys212 or Cys284 mutations increased the surface expression of GABA A R γ2 and the subsequent GABAergic neurotransmission, exerting anxiolytic effect in male rats. On the other side, overexpression of neuronal nitric oxide synthase in the BLA abolished the anxiolytic-like effects of physical exercise. This study reveals a key role of downregulating SNO-gephyrin in the anxiolytic effects of physical exercise, providing a new explanation for protein post-translational modifications in the brain after exercise.

As an emerging flame retardant, organic phosphate flame retardants have been extensively used worldwide. The aim of this study is to determine the effects of TnBP on neurobehavior of Caenorhabditis elegans ( C. elegans ) and its mechanisms. L1 larvae of wild-type nematodes (N2) were exposed to TnBP of 0, 0.1, 1, 10, and 20 mg/L for 72 hours. Then, we observed that the body length and body width were inhibited, the head swings were increased, the pump contractions and chemical trend index were reduced, the production of reactive oxygen species (ROS) was increased, and the expression of mitochondrial oxidative stress related genes ( mev-1 and gas-1 ) and P38 MAPK signal pathway-related genes ( pmk-1 , sek-1 , and nsy-1 ) was altered. After reporter gene strains BZ555, DA1240, and EG1285 were exposed to TnBP of 0, 0.1, 1, 10, and 20 mg/L for 72 hours, the synthesis of dopamine, glutamate, and Gamma-Amino Butyric Acid (GABA) was increased. In addition, the pmk-1 mutants (KU25) led to the sensitivity of C. elegans to TnBP in terms of head swings. The results showed that TnBP had harmful effects on the neurobehavior of C. elegans , oxidative stress might be one of the mechanisms of its neurotoxicity, and P38 MAPK signal pathway might play an important regulatory role in this process. The results revealed the potential adverse effects of TnBP on the neurobehavior of C. elegans .

Introduction:
In women of childbearing age with epilepsy, 40% experience the comorbidity of polycystic ovary syndrome (PCOS), which is marked by a higher prevalence of hyperandrogenism. Our recent clinical observations indicate the potential contribution of hyperandrogenism-induced PCOS to epilepsy susceptibility, and this study aimed to unravel the underlying factors that increase the susceptibility of females to epilepsy. Methods:
A letrozole-induced PCOS rat model was employed to simulate endogenous hyperandrogenism. Susceptibility was assessed through seizure kindling rates using pentetrazol and electroencephalogram recordings. Additionally, the role of androgens in epilepsy was verified through interventions using Diane-35. Results:
This study revealed that letrozole induced elevated testosterone levels and PCOS-related changes in female rats. PCOS rats, through pentetrazol-kindling, exhibited a reduced seizure threshold compared with controls. Elevated testosterone levels were observed in both the hippocampal and frontal brain tissues, accompanied by changes in circulation. Two weeks of Diane-35 intervention showed a tendency to alleviate these changes, modifying testosterone levels in both the plasma and brain tissue. Western blotting and immunohistochemistry revealed increased expression of GABA-A receptor in the hippocampus and decreased AMPA receptor expression in the frontal cortex, correlating with anti-epileptic status in PCOS rats. Conclusion:
This study delves into the impact of elevated androgen levels on seizure threshold, providing crucial insights into the underpinnings of the comorbidity between PCOS and epilepsy. These findings significantly contribute to the evolving field of epilepsy research, emphasizing the imperative consideration of hormonal influences for the development of targeted therapeutic interventions in individuals with epilepsy and PCOS
