Microstructure, mechanical properties, in vitro degradation behavior and cytocompatibility of biodegradable Zn-3Fe-HAP composites prepared by vacuum heating-press sintering

Ruifeng Shan,Runhua Yao,Hui Wang,Lin Liu,Yuyu Zhao,Xiaohong Yao,Yonghua Sun,Ruiqiang Hang

Zinc (Zn) is a promising biodegradable material, and have attracted widespread attention due to its befitting degradation rate compared to magnesium (Mg) and iron (Fe). However, the inferior mechanical strength of pure Zn limits its application for bone repair. Therefore, there is an urgent need to improve its mechanical properties , regulate its degradation rate, and endow it with optimal biocompatibility. The alloying of metallic elements and preparation of Zn-based composites can be a promising approach for this purpose. In this study, the pure Zn and Zn-3 wt%Fe-xHAP (x = 0, 5, 10, 15, 20 wt%) samples were fabricated by vacuum heating-press sintering. The microstructure, compressive mechanical property, in vitro degradation behavior, antibacterial property and in vitro cytocompatibility of different samples were investigated. We confirmed that the secondary phase Zn 13 Fe significantly improved the mechanical property, and micro-galvanic between Zn and Fe accelerated degradation of the samples. Compressive yield strength of all samples ranges from 85.65 MPa to 301.99 MPa, compressive elastic modulus varies between 8.26 GPa and 15.03 GPa, and corrosion rates are significantly improved which are about 0.005–3.065 mm/y. Moreover, Zn-3Fe-xHAP samples exhibited better biocompatibility and antibacterial property than pure Zn, especially the sample of Zn-3Fe-5HAP. In summary, biodegradable Zn-based composites prepared by vacuum heating-press sintering could be promising material for bone repair.

Structure evolution of MXene membranes mediated by ultralong HAP nanowires: from 2D plains to 3D mountains for ultrafast dye/salt separation

Yuanjia Zhao,Lishan Lin,Ang Lin,Fei-Fei Chen,Yan Yu

The development of high-performance nanofiltration membranes for dye/salt separation remains challenging due to the ubiquitous permeability–selectivity trade-off. Conventional MXene membranes suffer from structural compactness caused by irreversible nanosheet restacking, which severely limits their water permeability and separation efficiency. Herein, we propose a structural engineering strategy using ultralong hydroxyapatite (HAP) nanowires as nanoscale spacers to transform conventional 2D MXene laminates into 3D mountain-like architectures. The ultralong HAP nanowires (length: tens of micrometers; diameter: <100 nm) with ultrahigh aspect ratios (>1000) serve as effective structure mediators, creating abundant water transport channels while preventing MXene nanosheet restacking. Comprehensive characterization reveals that the optimized HAP/MXene membrane exhibits significantly expanded structural parameters, including a 98.31 % increase in surface roughness, 12.05 % rise in porosity, and 26.05 % enhancement in specific surface area compared to pristine MXene membranes. These structural advantages translate into exceptional separation performance: the membrane achieves an unprecedented water flux of 608.50 LMH/bar (1023.73 % improvement) while maintaining high dye rejection rates (>98 % for Congo Red) and minimal salt rejection (<15 % for various salts). Furthermore, the membrane demonstrates excellent stability in mixed dye/salt solutions, with consistent performance across different solute concentrations and long-term operation. This work not only overcomes the long-standing permeability–selectivity dilemma in MXene membranes but also establishes a generalizable paradigm for designing next-generation loose membranes through rational structural engineering.

CD44-Targeting Hydroxyapatite Nanoparticles (HAP) Induce Mitochondrial Dysfunction-Driven PANoptosis and Immunogenic Cell Death (ICD) via Ca Overload in Colorectal Cancer

Yao Xiao,Yuxuan Yang,Haosen Qiu,Jia Yang,Hao Liang,Zhaojun Jiang,Xiao Tong,Yongying Li,Qunfeng Huang,Jiaming Wu,Tian Lin,Jiang Yu,Min Liang

Colorectal cancer (CRC) remains therapeutically challenging due to high metastasis, recurrence, and immunotherapy resistance driven by tumor microenvironment-mediated immune evasion. Immunogenic cell death (ICD) offers a promising strategy to reshape the immune microenvironment, yet existing ICD inducers suffer from poor targeting efficiency and insufficient death signal release. Here, we constructed a calcium overload-based smart nanosystem, HA-HAP@CUR, to achieve highly efficient ICD induction via a triple-effect mechanism: hyaluronic acid (HA)-mediated CD44 targeting enables tumor-selective accumulation; pH-responsive hydroxyapatite (HAP) degradation releases Ca 2+ in the acidic tumor microenvironment; and curcumin (CUR) amplifies intracellular calcium overload by promoting endoplasmic reticulum Ca 2+ release, collectively establishing a positive feedback loop disrupting calcium homeostasis. Mechanistically, calcium overload induces mitochondrial membrane potential dissipation and sustained mPTP opening, triggering mitochondrial oxidative stress and energy metabolic disorders. This mitochondrial crisis concurrently activates caspase-3, GSDMD, and RIPK1, synergistically initiating apoptosis, pyroptosis, and necroptosis, ultimately converging into PANoptosis with potent immunostimulatory potential. This strategy, encompassing targeted accumulation, calcium storm activation, and multi-modal cell death synergy, provides a biologically precise approach to overcoming immunotherapy resistance in CRC.

Facile synthesis of metformin loaded Mn3O4-HAp magnetic hydroxyapatite nanocomposites for T1-magnetic resonance imaging guided targeted chemo-phototherapy in vitro

Asim Mushtaq,Xuehua Ma,Jabeen Farheen,Xiaoqing Lin,Muhammad Tayyab,M. Zubair Iqbal,Xiangdong Kong

Magnetic hydroxyapatite nanocomposites are promising candidates for various biomedical applications. However, the uncontrolled growth with the unfavorable size limits their widespread utilization in real-time applications. Herein, a facile method was utilized to fabricate uniform spherical-shaped nanocomposites of manganese oxide combined hydroxyapatite (Mn 3 O 4 -HAp) with an average size of about 28 nm. The structural characterization reveals the presence of Mn 3 O 4 in the crystalline phase, confined by HAp to give a composite structure. The biocompatibility and drug loading capability of Mn 3 O 4 -HAp nanocomposites are improved by surface modification using Food and Drug Administration-approved triblock copolymer Pluronic® F-127, followed by folic acid (FA) for targeting ability. Taking advantage of the surface properties of the polymer and the drug delivery potential of HAp, metformin was encapsulated in the prepared nanocomposites with a loading capacity of 98 %. The prepared nanocomposites indicate promising cell viability up to 87 % and an excellent drug release profile in an acidic atmosphere. Moreover, the magnetic component Mn 3 O 4 presents auspicious T 1 -Magnetic resonance imaging with notable r 1 relaxivity of 2.166 mM −1 s −1 . Further, the Mn 3 O 4 [email protected] nanocomposites demonstrate good cellular uptake and excellent reactive oxygen species (ROS) production under low-intensity UV irradiation , resulting in a remarkable photodynamic therapeutic effect. The results showed that the designed Mn 3 O 4 -HAp nanocomposites are promising targeted chemo-theranostic agents.

Multi-level design of a chitosan-based membrane: from sisal fiber reinforcement to surface micropatterning and hybrid hap for enhanced bone regeneration

Shuo Tang,Rui Lin,Yuqing Wang,Xinyuan Feng,Fang Shi,Yupeng Guo,Shu Huang,Xiang Hu,Liuyun Jiang

Overcoming the performance trade-offs among mechanical strength, bioactivity, and processability remains a pivotal challenge for Guided Bone Regeneration (GBR) membranes. Herein, we present a multi-level design strategy that systematically integrates material engineering across molecular, microstructural, and macroscopic scales to address this challenge. Specifically, at the macroscopic level, a sisal fiber-reinforced chitosan base layer provides robust structural support, achieving exceptional mechanical strength (39.48 MPa). At the molecular level, a hybrid hydroxyapatite co-functionalized with carboxymethyl cellulose, zoledronic acid, and phytic acid confers enhanced osteogenic activity and antioxidant capacity (87–95% DPPH scavenging). At the microstructural level, sophisticated surface micropatterning is constructed via a scalable process combining dynamic crosslinking and temperature-modulated phase separation, which actively guide cell behavior. This integrated design synergistically enhances osteogenic differentiation and mineralization, as evidenced by in vitro studies demonstrating the cooperative effects of its unique topology and sustained bioactive signals from the hybrid HAp. In a rat calvarial defect model, the membrane achieved exceptional bone regeneration (97.17 ± 1.7% BV/TV) at 8 weeks, markedly outperforming the control (64.04 ± 3.55%). This work presents a versatile platform highlighting the pivotal role of engineered carbohydrate polymers (chitosan and CMC) and plant-derived fibers in advanced bone biomaterials, offering a new design paradigm.

Improved bacteriostatic and in vitro cell activity of 3D printed silica-doped HAp scaffolds with tunable mechanical response and mass transport characteristic

Chengyu Zhang,Jimin Chen

Bone tissue defects caused by trauma, aging, or tumors necessitate advanced regenerative strategies. This study investigates silica-doped hydroxyapatite (HAp) scaffolds with Gyroid triply periodic minimal surface (TPMS) structures fabricated via digital light processing (DLP), combined with femtosecond laser (FS) surface modification to enhance mechanical, bioactive, and antibacterial properties. Finite element analysis (FEA) and computational fluid dynamics (CFD) revealed that the Gyroid structure exhibited superior mechanical isotropy (Zener ratio ≈ 1) and permeability (34.52×10 -9 m 2 ) compared to Schwarz and Fluorite lattices, facilitating efficient nutrient transport and cell migration. FS ablation induced hierarchical micro/nanostructures on silica-doped HAp (FSH), increasing surface area (121 ± 1.03 μm 2 ) and hydrophilicity (contact angle: 27.7°), which enhanced protein adsorption (88.0 ± 20.3 μg/mL) and osteoblast adhesion/proliferation. In vitro tests demonstrated FSH’s antibacterial efficacy against E. coli (82.83% inhibition) and robust bioactivity via hydroxyapatite layer formation in simulated body fluid. These findings highlight the potential of DLP-printed Gyroid scaffolds with FS-modified surfaces as a promising platform for bone tissue engineering, offering tailored mechanical properties, enhanced mass transfer, and multifunctional biological responses.

4-Hydroxyacetophenone is an Antipigmentation Reagent via Inhibiting Tryrosinase Activity

Qingquan Guo,Yudong Zhang,Chuanjin Tang,Qiuru Li,Fei Liu,Zhenming He,Chenyang Jiang,Jun Xu,Haishan Zhao

This study investigates the skin-whitening potential of 4-Hydroxyacetophenone (4-HAP), a compound commonly used as a preservative in cosmetics. Despite its widespread use, its whitening activity has not been extensively reported. Our preliminary research identified several commercial cosmetic formulations exhibiting whitening effects without known whitening agents. We hypothesized that 4-HAP might be a key ingredient. To test this, we employed a mushroom tyrosinase model, B16 mouse melanoma cells, and zebrafish to systematically evaluate the whitening activity of 4-HAP. Our findings demonstrate that 4-HAP is a potent tyrosinase inhibitor, significantly reducing melanin content in vitro and in vivo. Additionally, molecular docking and dynamics analyses revealed that the stable binding of 4-HAP to tyrosinase is crucial for its whitening effect. In formulations containing 2% 4-HAP, human skin models showed nearly complete recovery from UV-induced pigmentation after 4 weeks, while control groups exhibited persistent UV marks. These results confirm the efficacy of 4-HAP as a stable whitening agent in cosmetic formulations, highlighting its potential for further development in skin lightening products. This study has confirmed that 4-HAP holding the potential to become an effective whitening ingredient in the cosmetics field.

Photoelectric-Responsive Extracellular Matrix for Bone Engineering

Jieni Fu,Xiangmei Liu,Lei Tan,Zhenduo Cui,Yufeng Zheng,Yanqin Liang,Zhaoyang Li,Shengli Zhu,Kelvin Wai Kwok Yeung,Xiaobo Feng,Xianbao Wang,Shuilin Wu

Using noninvasive stimulation of cells to control cell fate and improve bone regeneration by optical stimulation can achieve the aim of precisely orchestrating biological activities. In this study, we create a fast and repeatable photoelectric-responsive microenvironment around an implant using a bismuth sulfide/hydroxyapatite (BS/HAp) film. The unexpected increase of photocurrent on the BS/HAp film under near-infrared (NIR) light is mainly due to the depletion of holes through PO43– from HAp and interfacial charge transfer by HAp compared with BS. The electrons activate the Na+ channel of mesenchymal stem cells (MSCs) and change the cell adhesion in the intermediate environment. The behavior of MSCs is tuned by changing the photoelectronic microenvironment. RNA sequencing reveals that when photoelectrons transfer to the cell membrane, sodium ions flux and the membrane potential depolarizes to change the cell shape. Meanwhile, calcium ions fluxed and FDE1 was upregulated. Furthermore, the TCF/LEF in the cell nucleus began transcription to regulate the downstream genes involved in osteogenic differentiation, which is performed through the Wnt/Ca2+ signaling pathway. This research has created a biological therapeutic strategy, which can achieve in vitro remotely, precisely, and noninvasively controlling cell differentiation behaviors by tuning the in vivo photoelectric microenvironment using NIR light.

Construction of Z-scheme Pt-TiO2/(Er3+:Y3Al5O12@Ta2O5)/HAP and its application in sonocatalytic degradation of prometryn and disinfection of Escherichia coli

Guanshu Li,Yuejiao Cao,Ludong Yi,Jun Wang,Youtao Song

BACKGROUND The recovery and reuse of sonocatalysts are the pivotal issues in the sonocatalytic degradation of organic pollutants. The use of some natural minerals as the carrier is not only beneficial to recycling of sonocatalysts, but also reduces the treatment costs of organic pollutants. Two kinds of hydroxyapatite (HAP), Ox Bone (OB)-HAP and synthesized (Syn)-HAP combine with the Pt-TiO 2 /(Er 3+ :Y 3 Al 5 O 12 @Ta 2 O 5 ) (P-TET) composite, obtaining two Z-scheme sonocatalysts, P-TET/OB-HAP and P-TET/Syn-HAP, respectively. The sonocatalytic activities of the prepared samples are investigated via degradation of prometryn and disinfection of Escherichia coli . RESULTS A series of characterizations showed that the Z-scheme P-TET/(OB or Syn)-HAP sonocatalysts were prepared successfully. To evaluate their sonocatalytic activities, some influencing factors such as ultrasonic irradiation time, inorganic oxidants, used times and trapping agents on the sonocatalytic degradation prometryn were determined. The best degradation ratio (80.31% based on N computing and 85.07% based on S atom computing) of prometryn could be obtained for 10 mmol L −1 K 2 S 2 O 8 , 1.0 g L −1 P-TET/OB-HAP sonocatalyst and 150 min ultrasonic irradiation. The excellent sonocatalytic disinfection ratio (88.65%) of E. coli could also be obtained under 40 min ultrasound irradiation. Moreover, a possible mechanism on sonocatalytic degradation of prometryn was illustrated. CONCLUSION The treated bovine bone HAP was directly used as a carrier to combine with P-TET to construct a novel Z-scheme P-TET/OB-HAP sonocatalyst. The high degradation ratio of prometryn and disinfection ratio of E. coli showed that P-TET/OB-HAP could be a promising sonocatalyst for the degradation of organic pollutants and the disinfection of bacteria. © 2020 Society of Chemical Industry

Amyloid-Mediated Nanoarchitectonics with Biomimetic Mineralization of Polyetheretherketone for Enhanced Osseointegration

Yingtao Gao,Yanyun Pang,Shuo Wei,Qian Han,Shuting Miao,Min Li,Juanhua Tian,Chengyu Fu,Zhengge Wang,Xu Zhang,Peng Yang,Yongchun Liu

Polyetheretherketone (PEEK), a widely used implant material, has attracted the attention of scientific researchers because of its bone-matched elastic modulus, radiolucency, and chemical resistance. However, the bioinert chemical properties of PEEK do not promote bone apposition once implanted. In this study, using a phase-transitioned lysozyme (PTL) nanofilm as a sandwiched layer, a robust hydroxyapatite (HAp) coating on PEEK (HAp@PTL@PEEK) is constructed. The PTL nanofilm shows strong adhesion to the PEEK surface and induces biomimetic mineralization to form a compact HAp coating on PEEK in simulated body fluids. This HAp coating not only shares a higher adhesion strength and better stability but can also be applied to implants with complex 3D structures. HAp@PTL@PEEK showed significantly enhanced osteogenic capacity when cultured with rat bone marrow mesenchymal stem cells by promoting initial cell adhesion, proliferation, and osteogenic differentiation in vitro. In vivo evaluations utilizing models of femoral condyle defects and skull defects confirm that the HAp coating substantially augments bone remodeling and osseointegration ability. Compared with the traditional method, our modified method is simpler, more environmentally friendly, and uses less hazardous components. Furthermore, the obtained HAp coating shares a higher adhesion strength to PEEK and a better osteogenic capacity. The study offers a novel method to improve the osseointegration of PEEK-based implants in biointerfaces and tissue engineering.

Development of Electrospun Composite Fibers in Multiscale Structure and Investigating the Performance on Proliferation and Osteogenic Differentiation of ADSCs

Liuyang Xuan,Ming Shi,Shuangshuang Ma,Feng Ye,Xuetao Shi,Ling Yan,Yan Li

Electrospun composite membranes in multiscale structures are developed for bone tissue engineering. Aligned polycaprolactone (PCL) fibers entrapping CA-HAp microparticles (containing CaCO 3 , hydroxyapatite, and casein in a hierarchical organization) are electrospun to find whether synergistic effects of fiber alignment and CA-HAp microparticles on improving osteogenic differentiation can be obtained. CA-HAp microparticles are in a spherical morphology of 1.42 ± 0.26 µm. Their presence increases fiber diameter and does not significantly affect fiber alignment. On all membranes, adipose derived stem cells (ADSCs) from humans spread very well. On a random group, cells distribute randomly and the presence of CA-HAp microparticles facilitates cell proliferation, especially for the one at CA-HAp/PCL 50 wt%; the one at CA-HAp/PCL 20 wt% shows significantly much higher alkaline phosphatase (ALP) activity (112.0% higher) than the pure PCL membrane. On aligned samples, cells align along fibers and expression of ALP is enhanced. However, at the same composition (CA-HAp/PCL 20 wt%), the random sample has much higher ALP activity than the aligned sample. The expressions of osteogenic marker genes are also evaluated. Combining the results and the applicability of membranes together, the random membrane at CA-HAp/PCL 20 wt% is the best candidate for bone tissue engineering.

A Biphasic Hydrogel with Self-Healing Properties and a Continuous Layer Structure for Potential Application in Osteochondral Defect Repair

Yue Jiang,Shanzhu Guo,Jingjing Jiao,Long Li

The treatment of osteochondral defects remains challenging due to the limited healing capacity of cartilage and the poor results of traditional methods. Inspired by the structure of natural articular cartilage, we have fabricated a biphasic osteochondral hydrogel scaffold using a Schiff base reaction and a free radical polymerization reaction. Carboxymethyl chitosan (CMCS), oxidized sodium alginate (OSA), and polyacrylamide (PAM) formed a hydrogel (COP) as the cartilage layer, while hydroxyapatite (HAp) was incorporated into the COP hydrogel to obtain a hydrogel (COPH) as an subchondral bone layer. At the same time, hydroxyapatite (HAp) was incorporated into the COP hydrogel to obtain a hydrogel (COPH) as an osteochondral sublayer, combining the two to obtain an integrated scaffold for osteochondral tissue engineering. Interlayer interpenetration through the continuity of the hydrogel substrate and good self-healing properties due to the dynamic imine bonding of the hydrogel resulted in enhanced interlayer bond strength. In addition, in vitro experiments have shown that the hydrogel exhibits good biocompatibility. It shows great potential for osteochondral tissue engineering applications. Keywords:biphasic scaffolds;self-healing hydrogel;osteochondral repair
