Quantum dot-modified aptamer probe for chemiluminescence detection of carcino-embryonic antigen using capillary electrophoresis

Zi-Ming Zhou,Zhe Feng,Jun Zhou,Bi-Yun Fang,Zhi-Ya Ma,Bo Liu,Yuan-Di Zhao,Xue-Bin Hu

A new method is proposed for determination of carcino-embryonic antigen (CEA) based on quantum dot (QD)-modified aptamer probe by capillary electrophoresis-chemiluminescence (CE-CL) detection system. An oligonucleotide sequence A contains two parts, one is CEA aptamer and the other is bridging segment. Horseradish peroxidase (HRP) conjugates with sequence A (HRP-DNA A ) and then mixes with QD labeling complementary bridging segment (QD-DNA B ), forming HRP-DNA A-B -QD probe. When CEA exists, the specific combination of the aptamer in sequence A and CEA can form CEA/HRP-DNA A-B -QD complex. After separation of CEA/HRP-DNA A-B -QD complex and HRP-DNA A-B -QD probe by capillary electrophoresis (CE), the chemiluminescence (CL) catalyzed by CEA/HRP-DNA A-B -QD complex can be detected without any interference, and the content of CEA can be estimated by the CL intensity. It has been proved that the separation efficiency of HRP-DNA A-B -QD and CEA/HRP-DNA A-B -QD complex is improved greatly after QD modification, then the interference issue resulted from free HRP-DNA A-B -QD probe is solved well; and the specific combination of HRP-DNA A-B -QD and CEA leads to a closer distance of HRP and QD, then chemiluminescent resonance energy transfer (CRET) occurs, which confirms the validity of the strategy. Results show that the CL intensity has a linear relationship with the concentration of CEA in the range from 0.0654 to 6.54   ng/mL, and the limit of detection is approximately 8   pg/mL ( S / N   =   3). This proposed method with high specificity has been applied for the accurate analysis of content of CEA in patient's serum.

Highly efficient catalytic hydrogen bond organic frameworks (HOFs)@HRP integrated with bowl-shaped organic SERS substrates for ultrasensitive detection of BPS

Yao Liu,Runzi Zhang,Xiaoyu Yang,Li Tang,Bing Shao,Yi He,Xiao Yang

With the growing public concern over environment and food safety, the detection of trace amounts of bisphenols S (BPS) becomes crucial. In this work, an ultrasensitive surface-enhanced Raman scattering (SERS) biosensor was developed based on hydrogen-bonded organic frameworks@horseradish peroxidase (HOFs@HRP) and hollow bowl-shaped SERS substrates (AFRNBs@AuNPs) for the detection of BPS. Due to the excellent biocompatibility and mild synthesis conditions, HOFs had been utilized as protective matrices for HRP, effectively preserving its enzymatic activity. Furthermore, hollow bowl-like morphology AFRNBs@AuNPs were successfully prepared as efficient SERS substrate. When BPS was present, the stronger affinity between BPS and its aptamer triggered a displacement reaction, allowing HOFs@HRP to bind with AFRNBs@AuNPs, thereby enabling the catalytic conversion of TMB into oxTMB and generating a detectable Raman signal. Accordingly, the biosensor exhibited a detection limit of 1.28 × 10 −12  M. This method pioneers a new approach for trace BPS detection, demonstrating broad application prospects.

Visual dual-mode aptasensor for non-small cell lung cancer exosome detection via HRP self-coupling enhanced oxidized iridium nanoparticle aggregation

Xu Xin,Zhang Ze,Shen Tong,Pan Hongzhi,Chang Dong

The main reason for the high mortality rate of non-small cell lung cancer is that patients are usually diagnosed at an advanced stage of the disease. Exosomes, small membrane vesicles secreted by normal cells or tumor cells, play a significant role in the progression of NSCLC. This study successfully optimized the preparation of artificial nanoenzymes self-coupling with horseradish peroxidase (IrO 2 NPs@HRP-AptCD63), without adding any ligand, demonstrating remarkable catalytic activity suitable for detecting the EGFR protein on the surface of NSCLC exosomes. When fused with the CD63 aptamer for identifying NSCLC exosomes, IrO 2 NPs@HRP showed enhanced catalytic activity in the 3,3′,5,5′-tetramethylbenzidine-H 2 O 2 oxidation–reduction system, thereby enhancing the colorimetric signal. This phenomenon can be distinguished by the naked eye and quantified using a UV–Vis spectrophotometer. Meanwhile, as the redox reaction occurs, the current signal of 3,3′,5,5′-tetramethylbenzidine-H 2 O 2 , acting as an electrolyte, changes. The developed aptasensor generates dual-mode signal outputs, firstly, to visually assess the samples for their positive or negative status, and subsequently employ more in-depth electrochemical or colorimetric analysis methods for a detailed quantitative analysis of suspected positive samples. The detection limits of electrochemical analysis and colorimetric analysis were 0.9 × 10 3  particles/mL and 0.14 × 10 3  particles/mL, respectively. Compared with traditional biomarkers such as CA125, this method exhibits exceptional specificity, capable of simultaneously distinguishing serum exosomes of healthy volunteers, COPD patients, and NSCLC patients, promoting exosome detection in mouse models for tumor monitoring. Additionally, it elucidates the changes in EGFR protein expression on the surface of serum exosomes throughout the developmental trajectory. Graphical abstract

A versatile and ultrasensitive molecularly imprinted electrochemiluminescence sensor with HRP-encapsulated liposome labeled by light-triggered click reaction for pesticide residues

Liu Guangyan,Li Shiyu,Jiang Zejun,Li Jianping

A novel approach for trace detection of fipronil with a molecularly imprinted electrochemiluminescence sensor (MIECLS) is proposed. The sensitivity is significantly improved via signal amplification of the enzymatic reaction of horseradish peroxidase (HRP) released from encapsulated liposomes which linked onto the template molecules after rebinding. The molecularly imprinted polymer membrane was prepared through the electropolymerization of monomers with fipronil as a template. After the elution of the template molecules, the analyte fipronil was reabsorbed into the cavities. HRP-encapsulated liposomes were linked to the target molecules by light-triggered click reaction. The higher the concentration of the target was, the more HRP-encapsulated liposomes were present on the molecularly imprinted polymer (MIP) sensor. Then, HRP was liberated from liposomes, and the catalytic degradation of hydrogen peroxide (H 2 O 2 ) by HRP occurs, which changed the electrochemiluminescence intensity of luminol significantly. The change of the ∆ ECL intensity was linearly proportional to the logarithm of the fipronil concentration ranging from 1.00 × 10 −14 to 1.00 × 10 −9  mol/L, and the detection limit was 7.77 × 10 −16  mol/L. The recoveries obtained ranged from 95.7 to 105.8% with RSD < 5%. The sensitivity of the detection was significantly improved, and the analysis process was simplified in that the incubation step required in the conventional method was avoided. The sensor proposed provides a feasible platform for ultra-trace amount determination. Graphical abstract

Exploration of the structural mechanism of hydrogen (H2)-promoted horseradish peroxidase (HRP) activity via multiple spectroscopic and molecular dynamics simulation techniques

Chuang Zhu,Fenfen Wei,Huibin Jiang,Zihan Lin,Lingyue Zhong,Yan Wu,Xiangjun Sun,Lihua Song

Horseradish peroxidase (HRP) is an enzyme that is widely used in various fields. In this study, the effects of molecular hydrogen (H 2 ) on the activity and structural characteristics of HRP were investigated by employing multiple spectroscopic techniques , atomic force microscopy (AFM) and molecular dynamics (MD) simulations. The results demonstrated that H 2 could enhance HRP activity, especially in 1.5 mg/L hydrogen-rich water (HRW). The structural analysis results showed that H 2 might alter HRP activity by affecting the active sites, secondary structure , hydrogen bonding network, C S groups, and morphological characteristics . The MD results also confirmed that H 2 could increase the Fe N bond distance in the active site, affect the secondary structure, and increase the number of hydrogen bonds . The MD results further suggested that H 2 could increase the number of salt bridges, and lengthen the S S bonds in HRP. This study primarily revealed the mechanism by which H 2 enhances the HRP activity, providing insight into the interactions between gas and macromolecular proteins. However, some of the results obtained via MD simulations still need to be verified experimentally. In addition, our study also provided a new convenient strategy to enhance enzyme activity .

Enzyme catalytic amplification of miRNA-155 detection with graphene quantum dot-based electrochemical biosensor

Tianxing Hu,Le Zhang,Wei Wen,Xiuhua Zhang,Shengfu Wang

A specific and sensitive method was developed for quantitative detection of miRNA by integrating horseradish peroxidase (HRP)-assisted catalytic reaction with a simple electrochemical RNA biosensor. The electrochemical biosensor was constructed by a double-stranded DNA structure . The structure was formed by the hybridization of thiol-tethered oligodeoxynucleotide probes (capture DNA), assembled on the gold electrode surface , with target DNA and aminated indicator probe (NH 2 -DNA). After the construction of the double-stranded DNA structure, the activated carboxyl groups of graphene quantum dots (GQDs) assembled on NH 2 -DNA. GQDs were used as a new platform for HRP immobilization through noncovalent assembly. HRP modified biosensor can effectively catalyze the hydrogen peroxide (H 2 O 2 )-mediated oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB), accompanied by a change from colorless to blue in solution color and an increased electrochemical current signal. Due to GQDs and enzyme catalysis, the proposed biosensor could sensitively detect miRNA-155 from 1   fM to 100   pM with a detection limit of 0.14   fM. High performance of the biosensor is attributed to the large surface-to-volume ratio, excellent compatibility of GQDs. For these advantages, the proposed method holds great potential for analysis of other interesting tumor makers.

A dual catalytic hairpin-based cyclic self-assembly reaction system for sensitive detection of miRNA21 using Zn@HRP snowflake metal-organic frameworks

Jiaxin Li,Yafei Fu,Mingjia Yuan,Jie Li,Haojing Zhang,Xuejiao Chen,Mengliu Yang,Ling Li

MicroRNAs are remarkably stable and detectable, making them ideal candidates for non-invasive diagnostics. The clinical relevance of fluid-specific miRNA profiling has implications for early disease detection, prognostic prediction, and therapeutic monitoring. A novel colorimetric biosensor for the highly sensitive detection of miRNA-21 is presented, integrating a dual catalytic hairpin-based cyclic self-assembly reaction system with Zn@HRP snowflake metal-organic frameworks (MOFs). This system leverages the unique snowflake structure and high catalytic activity of Zn@HRP MOF for signal amplification. Upon target miRNA-21 recognition, the system undergoes a cascade amplification process, leading to a significant increase in absorbance that is directly proportional to miRNA-21 concentration. The developed biosensor exhibits an ultra-low detection limit of 0.27 fM, demonstrating exceptional sensitivity, specificity, and anti-interference capabilities. Furthermore, the biosensor performs robustly in complex plasma samples, showing reliable recovery rates and excellent correlation with qRT-PCR results. This work not only provides a robust platform for precise miRNA detection but also offers significant potential for clinical diagnostics and biomarker profiling, paving the way for more accessible and reliable diagnostic tools in personalized medicine.

Sensitive detection of carcinoembryonic antigen (CEA) by a sandwich-type electrochemical immunosensor using MOF-Ce@HA/Ag-HRP-Ab2 as a nanoprobe

Wenjun Li,Chaoyun Ma,Yiju Song,Chenglin Hong,Xiuwen Qiao,Bincheng Yin

Sandwich-type electrochemical immunosensor was one of the main methods for detecting carcinoembryonic antigen (CEA). In this work, using Ce-MoF as the skeleton precursor, hyaluronic acid (HA) was coated on the surface of Ce-metal organic framework (Ce-MoF), which loaded with silver nanoparticles (Ag NPs) and horseradish peroxidase (HRP) to catalyze H2O2 and double amplified the current signal. Thus, a sensitive sandwich-type electrochemical immunosensor (Ce-MoF@ HA/Ag-HRP) was designed to detect carcinoembryonic antigen (CEA). The designed immunosensor used Au NPs to enhance the ability of attach more the first antibody (Ab1). This was due to Au NPs had good electrical conductivity and biocompatibility to accelerate electron transfer on the surface of the electrode. HA was riched in –COOH, –OH and had excellent biocompatibility, which can carry more Ag NPs to catalyze H2O2. Finally, the prepared sandwich-type electrochemical immunosensor had excellent biocompatibility and great catalytic performance. The immunosensor can be tested within 30 min and the logarithm of the current signal and CEA concentration showed a broad linear response range of 1 pg ml−1–80 ng ml−1, and the detection limit of CEA was 0.2 pg ml−1. More importantly, the proposed immunosensor had good reproducibility, selectivity, stability and without matrix effect. This confirmed that the proposed immunosensor had broad prospects in early clinical trials.

Co-immobilization of bienzyme HRP/GOx on highly stable hierarchically porous MOF with enhanced catalytic activity and stability: Kinetic and thermodynamic studies

Xia Gao,Huibin Pan,Siyu Tian,Le Su,Zhanlong Hu,Chengfang Qiao,Qian Liu,Chunsheng Zhou

Immobilization of enzymes onto solid carriers usually results in the reduction of enzyme catalytic activity . To solve this tricky problem, we proposed a novel strategy by accurately constructing the micro-mesoporous channel structure of highly stable hierarchically porous metal-organic framework (HP-Zr-MOF) for co-immobilization of horseradish peroxidase (HRP) and glucose oxidase (GOx). Notably, the precise construction and co-regionalization of HP-Zr-MOF with hierarchical structure can efficiently suppress enzyme leaching and significantly improve the catalytic efficiency, enzyme activity and the affinity for substrates of co-immobilized bienzyme reactor (GOx&HRP@HP-Zr-MOF). The K m value for GOx&HRP@HP-Zr-MOF was 0.2050 mM, which was 0.66-fold less than that of free GOx&HRP, while the K a value for GOx&HRP@HP-Zr-MOF was superior to that of free GOx&HRP, manifesting the affinity of GOx&HRP@HP-Zr-MOF for substrates strengthened. Importantly, the GOx&HRP@HP-Zr-MOF exhibited favorable stability and reusability, maintaining 97.4% of the original activity after 1 h incubation at 60 °C and over 90% initial activity after 30 cycles. Moreover, the GOx&HRP@HP-Zr-MOF was applied to the colorimetric detection of glucose, manifesting outstanding detection capability. Finally, the GOx&HRP@HP-Zr-MOF exhibited excellent catalytic efficiency for the degradation of 2,4-dichlorophenol, which indicated that the bio-catalytic material has potential practical application value in wastewater treatment .

Flow injection chemiluminescence immunoassay of microcystin-LR by using PEI-modified magnetic beads as capturer and HRP-functionalized silica nanoparticles as signal amplifier

Jusheng Lu,Wei Wei,Lihong Yin,Yuepu Pu,Songqin Liu

A rapid sandwiched immunoassay of microcystin-LR (MC-LR) in water is proposed with flow injection chemiluminescence detection. The magnetic beads (MBs) were first modified with polyethyleneimine (PEI) by acylamide bond between the carboxyl group on the surface of MBs and the primary amine group in PEI, followed by immobilizing of anti-MC-LR (Ab1) onto PEI with glutaraldehyde as linkage. The resulting Ab1 modified MBs captured the target MC-LR in water, reacted with the horseradish peroxidase and anti-MC-LR co-immobilized silica nanoparticles, and were detected with flow injection chemiluminescence. When using PEI/MBs as the carrier of anti-MC-LR, the CL signal was greatly enhanced up to 9-fold compared to that using MBs without PEI modification. The CL signal was further amplified 13-fold when Si/Ab2 was used as the signal probe. Under the optimal conditions, the present immunoassay exhibited a wide quantitative range from 0.02 to 200 μg L−1 with a detection limit of 0.006 μg L−1, which was much lower than the WHO provisional guideline limit of 1.0 μg L−1 for MC-LR in drinking water. The relative standard deviation was 4.8% and the recoveries for the spiked samples ranged from 84% to 115%, which indicated acceptable precision and accuracy for MC-LR. The present method is easier to perform and less time-consuming (the entire analysis process lasted about 40 minutes) and has been applied to the detection of MC-LR in different water samples successfully.

In situ assembly of porous Au-paper electrode and functionalization of magnetic silica nanoparticles with HRP via click chemistry for Microcystin-LR immunoassay

Shenguang Ge,Weiyan Liu,Lei Ge,Mei Yan,Jixian Yan,Jiadong Huang,Jinghua Yu

A simple, low-cost and sensitive origami electrochemical immunoassay-device was developed based on a novel gold nanoparticle modified porous paper working electrode (Au-PWE) for point-of-care testing. Azide-functionalized Au-PWE was prepared by the functionalization of Au-PWE with 1-azidoundecan-11-thiol. Alkyne end-terminated antibody was prepared with 4-pentynoic acid and antibody by the 1-ethyl-3-(3-(dimethylamino) propyl) carbodiimide hydrochloride and N-hydroxysuccinimide activation reaction . Alkyne-antibody was coupled to azido-Au-PWE by click reaction as a recognition element. Nearly monodispersed sphere-like silica-coated ferroferric oxide (Fe 3 O 4 @SiO 2 ) nanoparticles were prepared via the reverse microemulsion method. Azide-functionalized Fe 3 O 4 @SiO 2 was prepared by the functionalization of silica shell with 3-bromopropyltrichlorosilane followed by substitution with sodium azide . Alkyne-functionalized antibody and horse radish peroxidase were coupled to azide-functionalized Fe 3 O 4 @SiO 2 by click reaction as signal label. Horse radish peroxidase and ferroferric oxide could catalyze the oxidation of thionine in the presence of hydrogen peroxide. After the sandwich immunoreaction, the current was proportional to the logarithm of the Microcystin-LR. The linear regression equation was i (μA)=119.89+46.27   log   c MC-LR (μg/mL) in the range from 0.01 to 200   μg/mL. The limit of detection was 0.004   μg/mL. This immunoassay would provide a universal immunoassay method in environmental monitoring and public health.

Three-Dimensional Hierarchical HRP-MIL-100(Fe)@TiO2@Fe3O4 Janus Magnetic Micromotor as a Smart Active Platform for Detection and Degradation of Hydroquinone

Jie Yang,Jia Li,Xiaohui Yan,Yangsai Lyu,Ningning Xing,Ping Yang,Peng Song,Min Zuo

A novel multifunctional Janus magnetic micromotor was designed and constructed by using MIL-100(Fe)@TiO2@Fe3O4 multicore–shells modified with horseradish peroxidase (HRP) as a smart active platform to realize detection and degradation of hydroquinone (HQ). The obtained micromotor showed a unique three-dimensional (3D) hierarchical architecture with highly exposed active sites and could autonomously move at a speed of 140 ± 7.0 μm·s–1 by O2 bubbles generated from the catalytic decomposition of H2O2 fuel. Benefiting from the combination of active self-propulsive motion, high peroxidase-like activity, tuned heterojunctions with matching band structures, and a 3D hierarchical structure, an effective platform involving dynamically sensitive detection and quick removal of HQ from water was established by using the multifunctional HRP-integrated MIL-100(Fe)@TiO2@Fe3O4 Janus micromotor. The proposed multifunctional Janus magnetic micromotor had advantages of simple and feasible fabrication, sensitive detection and effective photo-Fenton degradation of HQ in a wide pH range of 4–7, and magnetic recycling, revealing potential for environmental remediation applications.
