A ratiometric fluorescent sensor based on molecularly imprinted multilevel mesoporous silica for highly sensitive detection of imidacloprid

Ziqi Wei,Weiyi Zhang,Shengjie Wang,Yiyi Han,Dongsheng Feng,Yingqing Ma,Bo Deng,Zixin Chen,Jialuo Mao,Fei Xu,Zhen Wang,Hui Cao

Herein, A ratiometric fluorescent sensor based on molecularly imprinted multilevel mesoporous silica (MIFP—SiCQDs@CdTe QDs) was designed for highly sensitive detection of imidacloprid (IDP) pesticide with long half-life in soil, plants, and water environments. In this sensor, CdTe QDs were anchored onto the multilevel mesoporous of MIFP—SiCQDs by post-imprinting modification as the analytical signal to improve the detection sensitivity of the conventional doping method (d-MIFP). The results showed that the synthesized MIFP—SiCQDs@CdTe QDs possessed both small mesopore (2–3 nm) and large mesopore (5–50 nm), which is favorable for the anchoring of CdTe QDs (about 2–3 nm in diameter) and the mass transfer of IDP into them. Due to the multilevel mesoporous structure and low background, the sensor responded well to the addition of IDP in the range of 5 ng mL −1 –0.5 μg mL −1 with a detection limit of 3.55 ng mL −1 . The MIFP—SiCQDs@CdTe QDs sensor was successfully applied to determine IDP levels in real samples with the average recovery ranging from 97.64 to 109.88% and RSD values ranging from 1.00 to 4.65%. Furthermore, the quenching mechanism of MIFP—SiCQDs@CdTe QDs by IDP was considered to be the dynamic quenching. These results proved that the construction of multilevel mesoporous and post-imprinting modification of the analytical signal is an effective method to improve the sensitivity of d-MIP.

Rapid screening of imidacloprid residue in grains and medicinal herbs: A newly designed hapten and monoclonal antibody

Yudan Wang,Jia’ an Qin,Jing Zhang,Ziyue Jin,Jiaoyang Luo,Meihua Yang

Three different imidacloprid hapten structures were designed to conjugate with proteins (bovine serum albumin, BSA; ovalbumin , OVA; keyhole limpet hemocyanin , KLH) for screening the optimal immunogen and coating antigen. Among these, an unreported antigen (hapten 6-KLH) was selected as the optimal immunogen and coating antigen. In addition, an imidacloprid-specific and high titer monoclonal antibody (IMIB7C3) was obtained by using the above-selected immunogen. A sensitive ic-ELISA (indirect competitive enzyme-linked immunosorbent assay) with a half-maximal inhibitory concentration (IC 50 ) of 1.3 ng mL −1 was established by using the IMIB7C3 antibody (only 1.2 ng per well) to detect the residues of imidacloprid in grains (wheat and maize) and different herbs ( Notoginseng radix et rhizoma, Dioscoreae rhizoma, Lonicerae japonicae flos, Astragali radix , Jujubae fructus ). The detection results of real samples by the developed immunoassay were confirmed by liquid chromatography-tandem mass spectrometry (LC–MS/MS), which proved the accuracy and reliability of the established ic-ELISA. These results indicate that the proposed ic-ELISA method is suitable for rapid and high-throughput detection of imidacloprid residues in agricultural products and medicinal herbs. Furthermore, a quantitative risk assessment was conducted for Lonicerae japonicae flos based on the detection results, which indicates an acceptable risk to human health after the intake of Lonicerae japonicae flos polluted by imidacloprid.

Layer-by-layer assembled decomposable nanocapsules for light responsive release of pesticide imidacloprid on Aphis craccivora Koch

Yiyun Gao,Wenjun Xiao,Chenhua Gu,Sheng Yu,Shengke Li,Aihua Zou

Background Conventional pesticide formulations are often griped about inefficiency due to the low biological uptake after spraying. Controlled release nanopesticides can precisely release pesticides in response to specific stimuli, thereby killing pests and pathogens with the least effective concentration. This study aims to develop nanocapsules-based photo decomposable nanopesticides for efficient pesticide control. Results The target nanopesticides were successfully fabricated by using layer-by-layer assembly of azobenzene grafted hyaluronic acid (Azo-HA) and PolyDADMAC, confirmed by UV–vis, DLS, Zeta potential and TEM measurements. Particle size and Zeta potential of the fabricated nanocapsules were 220 nm and + 46.1 mV, respectively, and the nanocapsules were found to maintain stable for up to 30 days. The optimized drug loading and encapsulation ratio of Imidacloprid (IMI) in IMI/Azo-HA@PolyDADMAC were 21.5% and 91.3%, respectively. Cumulative release of IMI from the nanopesticides was increased from ~50% to ~95% upon UV-light irradiation (365 nm). LC 50 value of the nanopesticides towards Aphis craccivora Koch was decreased from 2.22 mg/L to 0.55 mg/L upon UV-light irradiation. Conclusion The trans to cis transformation of Azo group in HA will decompose IMI/Azo-HA@PolyDADMAC nanopesticides upon UV irradiation, thus facilitated the IMI release, resulting in decreased concentration of pesticides for efficient pesticide control. Our work demonstrated the great potential of light-responsive nanocapsules as controlled release nanocarrier for efficient and eco-friendly pesticide-control in sustainable agriculture. Imidacloprid-Induced Ferroptosis-Associated Injury in Common Carp Hepatocytes: Potential Involvement of the miR-153c/SQSTM1 Axis

Huijie Chen,Yitong Wang,Zhenkai Yao,Jing Li,Peng Li,Lei Diao

The neonicotinoid insecticide imidacloprid (IMD), widely used in agriculture, poses potential risks to aquatic ecosystems, yet its hepatotoxic mechanisms in freshwater fish remain poorly understood. This study investigates the roles of ferroptosis, mitochondrial dysfunction, inflammation, and the miR-153c/SQSTM1-mediated ferritinophagy pathway in IMD-induced hepatotoxicity in carp ( Cyprinus carpio ) hepatocytes. Using the CCK-8 assay, 0.6 μM IMD was selected for subsequent experiments. IMD exposure triggered ferroptosis, evidenced by increased intracellular Fe 2+ accumulation, upregulated pro-ferroptotic gene and protein expression, and downregulated ferroptosis inhibitors (SLC7A11 and GPX4). Mitochondrial dysfunction was confirmed by reduced ATP content, decreased mtDNA levels, and a lowered NADPH/NADP + ratio, along with aberrant expression of mitochondrial fission (Fis1 and Drp1) and fusion (Mfn1 and TFAM) genes. Additionally, IMD induced oxidative stress (elevated ROS and MDA; decreased T-AOC, CAT, and SOD activities) and a pronounced inflammatory response with upregulated pro-inflammatory cytokines. Mechanistically, IMD downregulated miR-153c, with concomitant upregulation of SQSTM1 at both the mRNA and protein levels, and dual-luciferase assays validated SQSTM1 as a direct target of miR-153c. miR-153c overexpression attenuated IMD-induced ferritinophagy, ferroptosis, and inflammation via the miR-153c/SQSTM1 axis. Collectively, these findings demonstrate that IMD induces hepatotoxicity in carp by triggering ferroptosis, mitochondrial dysfunction, and inflammation through the miR-153c/SQSTM1-mediated ferritinophagy pathway, providing mechanistic insights and potential targets for mitigating IMD’s ecological risks.

Modularly integrated synergistic SERS strategy for ultra-trace detection of imidacloprid in fresh fruit and vegetable juices

Xinru Yu,Hongbin Pu,Da-Wen Sun

A reliable IMIPCs–AMoO 2 /C SERS analytical method was established. • Validated results were acceptable for IMI in fresh fruit and vegetable juices. • The method displayed good linear correlation, low LOD, trueness and precision. • The mechanisms of a modularly integrated synergistic strategy were explored. • Rapid detection of trace levels of IMI in the sample was achieved (10 min). Imidacloprid (IMI), a first-generation neonicotinoid insecticide, is taken up by plants and distributed to edible parts owing to its systemic transport properties. This leads to its residues in fruit and vegetable juices, posing a potential food safety risk. Herein, we report a modular integrated synergistic SERS strategy for reliable, efficient and selective detection of IMI. An IMI-imprinted polymer-pesticide carb (IMIPCs) cartridge was designed for recognition, purification, and enrichment. The AMoO 2 /C SERS substrate, optimised through annealing, aligns its bandgap with the excitation wavelength, while IMI adsorption induces band bending and triggers charge-transfer resonance, enabling signal amplification. Under optimised conditions, the method exhibits excellent linearity from 0.500 ng/mL to 50.0 ng/mL, a low detection limit of 0.0904 μg/kg, and high accuracy in spiked typical juice matrices (tomato, celery, apple, and mixed juices), with recoveries of 92.7%–98.1% (RSD%, 0.332%–2.50%) and no significant difference compared with the HPLC method at the 95% confidence level. Application to twenty real samples collected from the market, including fruit and vegetable juice samples such as kiwi, kale, banana, and orange juices and their mixtures, demonstrated good selectivity, reliability, and reproducibility, with residue levels detected in the ppb range. Overall, this LEGO-inspired SERS method provides a simple, efficient and highly selective analytical strategy for monitoring IMI residues in fruit and vegetable juices, and can serve as a supplementary screening tool for routine food safety monitoring of such juice products. Its applicability to complex high-fat or protein-rich matrices, such as milk-based or nut-based beverages, remains to be further validated through subsequent research. Imidacloprid dissipation, metabolism and accumulation in Agaricus bisporus fruits, casing soil and compost and dietary risk assessment

Qicai Zhang,Xianli Wang,Qinxiong Rao,Shanshan Chen,Weiguo Song

An improved understanding of imidacloprid (IMI) metabolism and accumulation in casing soil or compost-mushroom systems will help to optimise the safe use of IMI for agricultural pest control in Agaricus bisporus cultivation. In this study, the dissipation, metabolites and accumulation of IMI in casing soil or compost- A. bisporus systems were investigated. The results show that the IMI dissipation half-lives at doses of 10 and 50 mg kg −1 were 65 d and 59 d in casing soil and 6.6 d and 6.2 d in compost, respectively during the cultivation period. Three IMI metabolites were observed in casing soil during cultivation of mushrooms. Urea-imidacloprid (IMI-urea) was a major metabolite, accounting for more than 59%. In compost, the main metabolite in the first 5 days was IMI-urea, from 5 to 10 days olefin-imidacloprid (IMI-ole) and 6-chloronicotinic acid (6-CNA) were both the most prominent compounds, and after 10 days 6-CNA alone. At 50 mg kg −1 dose, IMI and the metabolites (IMI-urea, IMI-ole) were detected in the fruiting body with IMI applied to casing soil and only the metabolite 6-CNA was detected in fruiting body with IMI applied to compost. The bio concentration factors (BCFs) of IMI-ole and IMI-urea were higher than of IMI and 6-CNA, and the BCFs had no obvious relationship with log K ow . Furthermore, the potential dietary risk of IMI in A. bisporus was acceptable when application rates in casing soil or compost was up to 50 mg kg −1 . Our study supports a safe use of IMI as agricultural pest control in A. bisporus cultivation.

Uptake and translocation of imidacloprid via seed pathway and root pathway during early seedling growth of corn

Xiujun Tang,Fengjiao Hao,Huizhu Yuan,Xiaojing Yan,Daibin Yang,Alan G Taylor

BACKGROUND A systemic seed treatment can be taken up into shoot tissues during early corn seedling growth. However, the pathway that a systemic compound is taken up, either from seed or root uptake to shoot tissues is not fully understood. To study the single contributions of seed pathway and root pathway for the uptake and translocation of imidacloprid seed treatment, two methods were developed: A seed treatment method and a growing media/hydroponic solution delivery method. The seed treatment method employed a live and dead seed separated with plastic wrap to provide a barrier. The growing media/hydroponic solution delivery method quantified the capacity for uptake by seeds and roots. RESULTS The seed pathway transported 1.9–2.5 times more imidacloprid into shoot tissues when compared to the root pathway at the same dosage rate. The higher amount of imidacloprid taken up and translocated by the seed pathway was attributed to the fact that the corn seeds were in direct contact with high provided concentrations of imidacloprid. However, the root pathway showed 7.9–9.5 times higher capacity for transporting imidacloprid into shoot tissues when compared with the seed pathway. Whenever imidacloprid was taken up by seed or root, amounts of imidacloprid were measured in both tissues of seed and root. CONCLUSION The seed pathway transported more imidacloprid than the root pathway during early seedling growth since corn seeds were contacted by higher concentrations of imidacloprid. Both seed pathway and root pathway were occurring concurrently during early seedling growth of corn. © 2020 Society of Chemical Industry

Dual-Responsive Graphene Oxide/Poly(NIPAM-co-AA) Hydrogel as an Adsorbent for Rhodamine B and Imidacloprid

Guohong Yao,Sisi Li,Jun Xu,Hui Liu

A dual-responsive hydrogel based on graphene oxide (GO) was prepared by in situ radical copolymerization of N-isopropyl acrylamide (NIPAM) as a temperature-sensitive component and acrylic acid (AA) as a pH-sensitive unit. The resulting hydrogels (GO/poly(NIPAM-co-AA) (PNA) were systematically characterized by Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, Raman spectroscopy, thermogravimetric analysis, differential scanning calorimetry, and scanning electron microscopy. With the change of AA content, the lower critical solution temperature of hydrogels changed from 26.91 to 24.38 °C. The swelling ratio under alkaline condition was higher than that under acidic condition. The adsorption of rhodamine B (RB) followed Langmuir isotherm model and pseudo-second-order model, and nearly 93% of RB could be released within 10 h at pH 4.01 and 60 °C. With imidacloprid as the model drug, the higher cumulative release rate of the GO/PNA hydrogel was obviously observed at pH = 4.01 than at pH = 9.18. On the basis of their good adsorption–desorption, loading-releasing, and regeneration ability, the GO/PNA hydrogel as a multifunctional adsorbent might have broad application prospects in environmental protection and biomedical engineering.

Co/Mo2C-Embedded N-Doped Carbon Nanotubes Combined with Molecularly Imprinted Membranes for Selective Electrocatalytic Determination of Imidacloprid

Dongshi Feng,Jiangdong Dai,Yongsheng Yan,Chunxiang Li

Developing a catalyst with excellent electrical conductivity and catalytic performance for on-site testing of residual imidacloprid is significant and challenging. In situ growth of Mo2C nanodots on Co-induced N-doped carbon nanotubes (Co/Mo2C/N-CNT) was synthesized to construct a molecularly imprinted electrochemical sensor for the detection of imidacloprid. The results proved that the catalytic performance of Co/Mo2C/N-CNT for imidacloprid was over two times higher than those of Co/N-CNT and commercial CNT. This improvement was attributed to the formation of a heterostructure between Co species, Mo2C, and N-CNT, which facilitated highly exposed catalytic active sites. Additionally, the abundant Mo2C nano-dots promoted interfacial charge transfer to achieve optimal dynamics. The optimum preparation parameters of the catalysts were obtained by response surface methodology. By analyzing the relationship between different pH values and peak potential, as well as the influence of different scanning rates on peak potential, it was deduced that the possible electrocatalytic mechanism of imidacloprid involved the reduction of the nitro group to a hydroxylamine group and H2O. Under optimal conditions, the limit of detection (LOD) was 0.033 × 10−6mol·L−1(R2= 0.99698), and the linear range was 0.1 × 10−6~100 × 10−6mol·L−1. The application effect of the prepared sensor was evaluated by measuring the imidacloprid in two kinds of tea, indicating that the sensor possessed good sensitivity and selectivity, and was capable of meeting the requirements of on-site detection.

Degradation of the neonicotinoid insecticide imidacloprid by ozonation and the E-peroxone process: Mechanisms, transformation products, and toxicity

Yang Zhang,Lingwei Gao,Juhong Zhan,Erzhuo Zhao,Huijiao Wang,Chunrong Wang,Yujue Wang

In this study, degradation of the neonicotinoid insecticide imidacloprid (IMI) by ozonation and the E-peroxone process was investigated. Results show that IMI is an O 3 -refractory compound with second-order rate constants for the reaction with O 3 and • OH ( k O3,IMI and k •OH,IMI ) being 3.9 and 3.95 × 10 9 M −1 s −1 at pH 7, respectively. Due to its low O 3 reactivity, IMI was only abated by < 20 % in a selected groundwater during ozonation with typical ozone doses applied in water treatment (1–5 mg/L). In comparison, the E-peroxone process considerably enhanced IMI removal efficiency to > 80 % under similar reaction conditions. During both ozonation and the E-peroxone process, IMI was mainly abated by • OH oxidation. The reaction mechanism of IMI with • OH was investigated with quantum computations, which show that H-abstraction at H23 site was the most favorable pathway. Sixteen transformation products (TPs) generated from IMI degradation were identified using various chromatographic and mass-spectrometric techniques. The acute and chronic toxicities of IMI and its TPs to different aquatic organisms were evaluated with Microtox bioassay and the ECOSAR program, which indicate that some TPs (e.g., TP 2 –TP 5 and TP 7 –TP 9 ) are even more toxic than IMI to aquatic organisms (e.g., daphnia). The results of this study suggest that IMI along with its TPs should be sufficiently abated during water treatment, and the E-peroxone process provides an efficient option to upgrade conventional ozonation for enhanced IMI abatement.

Exploring the toxicity of biodegradable microplastics and imidacloprid to earthworms (Eisenia fetida) from morphological and gut microbial perspectives

Baikerouzi Baihetiyaer,Nan Jiang,Xianxu Li,Jie Song,Jun Wang,Xiaoteng Fan,Yajie Zuo,Xianqiang Yin

Biodegradable microplastics (BMPs) pose serious environmental problems to soil organisms, and their adsorption capacity might make pesticides more dangerous for soil organisms. Therefore, in this study, polylactic acid (PLA) BMPs and imidacloprid (IMI) were used as a representative of BMPs and pesticides, respectively. Eisenia fetida was used as a test animal to investigate the effects of environmentally relevant concentrations of single and compound contaminated PLA BMPs and IMI on mortality, growth, number of offspring, tissue damage, and gut microorganisms of E.fetida . Exposure to PLA BMPs treatment and PLA BMPs + IMI treatment resulted in a sustained increase in E.fetida mortality, reaching 16.7% and 26.7%, respectively. The growth inhibition rate of single treatments was significantly increased. The compound contamination had the greatest effect on E.fetida offspring compared to the control. PLA BMPs and IMI cause histological damage to E.fetida, with the compound treatment causing the most severe damage. Based on the results of 16S sequencing, the bacterial communities in E.fetida gut and soil treated to PLA BMPs and IMI were significantly different. PLA BMPs + IMI treatment suppresses the abundance and diversity of E.fetida gut microorganisms, disrupting the homeostasis of bacterial communities and causing immune and metabolic dysfunction. These findings highlight the more severe damage of combined PLA BMPs and IMI pollution to E.fetida , and help to assess the risk of earthworm exposure to environmentally relevant concentrations of PLA BMPs and IMI.

ZIF-8-wrapped AgNPs modified with β-cyclodextrin for sensitive detection of thiacloprid and imidacloprid by SERS technology

Bin Yang,Jia Liu,Xin Liu,Shiyue Niu,Fengming Zhang,Yuting Wang,Daqian Song,Shuyun Bi

The high efficient surface-enhanced Raman scatterring (SERS) methods to detect thiacloprid and imidacloprid were established using ZIF-8-wrapped Ag nanoparticles (AgNPs) modified with β -cyclodextrin ( β -CD). The substrate of ZIF-8/ β -CD@AgNPs was characterized by ultraviolet visible spectra (UV–vis), thermogravimetric analysis (TGA), X-ray diffraction (XRD), transmission electron microscopy (TEM) and scanning transmission electron microscopy (STEM). The interaction between the substrate and thiacloprid/imidacloprid was also explored. The optimum measurement conditions were obtained by response surface model based on single-factor experiments. Enhancement factors (EFs) of thiacloprid and imidacloprid were respectively 2.29 × 10 6 and 2.60 × 10 6 . A good linearity between the scattering intensity and the concentration of thiacloprid/imidacloprid within 3–1000 nmol L −1 /6–400 nmol L −1 was established. The interference experiments indicated that the methods had good selectivity . The SERS methods were successfully applied to detect thiacloprid and imidacloprid in several vegetables samples. The recoveries ranged from 95.5 % to 105 % (n = 5). The detection limits (LODs) (S/N = 3) for thiacloprid and imidacloprid were 1.50 and 0.83 nmol L −1 , respectively.
