Preparation and physicochemical properties of konjac glucomannan ibuprofen ester as a polysaccharide-drug conjugate

Huiting Kang,Lianxiong Guan,Kai An,Dating Tian

A novel drug-polysaccharide conjugate with konjac glucomannan (KGM) as a drug carrier was fabricated through the esterification of ibuprofen (IBU), an anti-inflammatory drug, with KGM. The influences of the reaction conditions, such as the amount of ibuprofen acryl chloride, reaction time, reaction temperature, and the amount of catalyst, on the degree of substitution were investigated. KGM ibuprofen ester (KGM-IBU) was characterized by Fourier transform infrared spectrometry (FTIR), X-ray diffraction (XRD), solid-state 13C NMR, scanning electron microscopy (SEM), transmission electron microscopy (TEM), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), and dynamic mechanical analysis (DMA). The hydrophobic structure of IBU in KGM-IBU was proven by the fluorescence emission spectra of pyrene. In addition, by using commercially available ibuprofen sustained-release capsules (IBU-SRC) as a control, the in vitro controlled release performance of KGM-IBU was evaluated. The cumulative release of IBU-SRC within 36 h was 94%, while that of KGM-IBU within 36 h was 77%. The results showed that KGM-IBU had better sustained-release performance without a burst release effect. The obtained products could be used as a potential biocompatible sustained-release drug delivery system.

The enhancement in antifungal efficacy of konjac glucomannan films with attapulgites immobilized of AgNPs

Xiaohan Chen,Daoming Li,Weifei Wang

The objective of this study was to enhance the water barrier properties and antifungal efficacy of KGM film by incorporating (attapulgites-Ag nanoparticles, APTs-AgNPs) composites. The X-ray photoelectron spectroscopy (XPS), Transmission electron microscopy (TEM), and Energy dispersive X-ray (EDS) analyses indicated that the successful fabrication of APTs-AgNPs composites. Water vapour permeability (WVP), moisture content (MC), water solubility (WS) and water absorption (WA) values decreased by 19.15%, 12.50%, 74.96% and 41.74% respectively, while the water contact angle (WCA) increased by 134.74%. These results demonstrated that the water barrier property of KGM/APTs-AgNPs films was superior to that of pure KGM films. The Rheological properties, fourier transform infrared (FTIR) spectrum, X-ray diffraction (XRD) patterns and Scanning electron microscopy (SEM) images results indicated that hydrogen bonds formed between homogeneous APTs-AgNPs (1.5 wt%) and KGM, which enhanced the water barrier properties of the KGM/APTs-AgNPs films. The KGM/APTs-AgNPs composite films possessed excellent oxygen permeability, Mechanical properties, UV barrier properties, opacity properties and thermostability. Moreover, KGM/APTs-AgNPs films slowly released Ag + and exhibited long-acting antifungal efficacy against Aspergillus niger , as the inhibition circle diameter was 20.70 ± 0.32 mm for 10 days. Therefore, KGM/APTs-AgNPs films could be a potential alternative to petroleum-based films.

Fabrication and cell imaging of konjac glucomannan-copper nanocluster conjugates with aggregation-induced emission

Lianxiong Guan,Wei Liu,Huiting Kang,Dating Tian

A positively charged konjac glucomannan- graft -poly-(2-methacryloyloxyethyl) trimethyl ammonium chloride (KGM- g -PDMC) was prepared by using ceric ammonium nitrate as the initiator in aqueous solution. Negatively charged copper nanoclusters (CuNCs) were prepared by a one-step reduction method using glutathione as the reductant. Then, the two precursors mentioned above were combined through electrostatic attraction to form a conjugate (KGM- g -PDMC/CuNCs) exhibiting aggregation-induced emission (AIE). The resultant products were characterized by Fourier transform infrared spectroscopy , zeta potential and particle size analysis , X-ray photoelectron spectrometry , transmission electron microscopy , and fluorescence spectrophotometry . The results showed that KGM- g -PDMC enhanced the AIE effect of CuNCs, with an approximate 4-fold increase in fluorescence intensity . A cytotoxicity analysis showed that the products possess good biocompatibility. Moreover, cell imaging indicated that the products have potential for application in the field of biomedicine.

Interaction between konjac glucomannan and tannic acid: Effect of molecular weight, pH and temperature

Yun He,Shishuai Wang,Jing Li,Hongshan Liang,Xianlin Wei,Dengfeng Peng,Zhengqiang Jiang,Bin Li

The objective of this study was to investigate the interaction between tannic acid (TA) and konjac glucomannan (KGM) with different molecular weight at different pHs,temperatures and urea concentration. The different molecular weight of KGM without change in primary structure were obtained by ultrasonic treatment. Turbidity titration showed that TA/KGM complexes could form at lower pH. The dynamic light scattering (DLS) and transmission electron microscopy (TEM) results indicated that the TA/KGM complexes assembled into large and uniform colloidal particles and would lead to precipitate with the increase of TA/KGM weight ratio. Isothermal titration calorimetry (ITC) results further revealed two-step binding behavior: the first stage corresponded to soluble complex formation, while the second stage represented aggregation between complexes. When the molecular increased, the transition of TA/KGM mass ratio between stage 1 and stage 2 changed from 3.5 to 3.0. KGM of higher molecular weight needed to bind more TA to move to the next stage. When the pH was 7 at 25 °C, the pH was 3.8 at 40 °C or urea addition, the ITC signal was completely abolished, the formation of complexes between KGM and TA was shown to be highly dependent on pH, temperature, urea concentration, suggesting the main driving force of TA-KGM complexes formation was hydrogen bond.

Bioinspired aerogel based on konjac glucomannan and functionalized carbon nanotube for controlled drug release

Lin Wang,Ruo-Jun Mu,Lizhuan Lin,Xiaohan Chen,Sisi Lin,Qianwen Ye,Jie Pang

In this study, a facile marine bioinspired surface modification approach for carboxyl-functionalized multiwalled carbon nanotube (CCNT) and enhanced interfacial adhesion with the konjac glucomannan (KGM) matrix were illustrated to develop aerogels. Combined with FT-IR, XRD, Raman, TGA, XPS and SEM results, it was indicated that functionalized CCNT (PCCNT) is a reinforcer through hydrogen bond interactions in the aerogel formation process, which could be the main reason for the enhancement. The swelling and vitro release behavior of KGM/PCCNT aerogels were studied under two conditions using the drug 5-fluorouracil (5-FU). The release amount of 5-FU incorporated into KGM/PCCNT4 aerogel was about 48% at pH 1.2 and 62% at pH 6.8 after11 h, respectively. The results showed that the release rate of 5-FU from the KGM/PCCNT4 aerogel using PCCNT could be effectively controlled, suggesting potential applications for it as a drug carrier in targeted delivery in the biomedical filed.

Preparation and evaluation of magnetic molecularly imprinted polymers based on konjac glucomannan for urea

Kang Huiting,An Kai,Guan Lianxiong,Tian Dating

Magnetic molecularly imprinted polymers (MIPs) for urea were prepared using konjac glucomannan (KGM) as the substrate, methacrylic acid as the graft comonomer, ethylene glycol dimethacrylate as the cross-linker, urea as the template molecule, and Fe 3 O 4 NPs as the magnetic ingredient. The resultant MIPs were characterized by Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, X-ray diffraction, transmission electron microscopy, thermogravimetric analysis, vibrating sample magnetometry, and Brunauer–Emmett–Teller spectroscopy. The MIPs possessed high superparamagnetism with core–shell structures, which could be easily separated from the mixture by an external magnetic field. The MIPs could specifically recognize urea, and the recognition capacity and adsorption performance remained almost unchanged after three cycles of adsorption and elution. The adsorption kinetics of MIPs on urea could be described by the second-order kinetic model. The maximum adsorption capacity of MIPs on urea was 16.80 mg/g, and the adsorption isotherms of MIPs on urea followed the Langmuir model. In addition, in vitro adsorption experiments through simulated body fluids showed that the MIPs had good adsorption capacity for urea. The product is expected to be used as a novel adsorption material for blood purification.

Preparation and characterization of deacetylated konjac glucomannan / pectin composite films crosslinked with calcium hydroxide

Huang Tuankang,Qin Youlin,Li Mingchao,Gao Shanjun,Shen Chunhui

Deacetylated konjac glucomannan (DKGM)/calcium pectin (Ca-PEC) composite films were prepared by calcium hydroxide to realize deacetylation of konjac glucomannan (KGM) and the crosslinking of pectin at the same time. Fourier transform infrared spectroscopy (FTIR) indicates that there are intermolecular hydrogen bond and crosslinking structure between DKGM and Ca-PEC. The physical properties of the composite films are characterized by mechanical properties, water vapor permeability (WVP), swelling degree (SD), water solubility (WS), thermogravimetry (TG) and Opacity. The results exhibit that deacetylation of KGM and crosslinking structure of pectin reduce the swelling degree and water solubility of the films, as well as improve its thermal stability and tensile strength. These are attributed to enhanced intermolecular interactions, more compact entanglement of molecular chains and more solid spatial network structures. When pectin content is 40%, the combination property of DKGM / Ca-PEC composite film is the best.

Size effect-inspired fabrication of konjac glucomannan/polycaprolactone fiber films for antibacterial food packaging

Wanmei Lin,Yongsheng Ni,Jie Pang

The exploration of new methods to produce food packaging with excellent physicochemical and antibacterial properties is of great scientific and technological interest. Here, we successfully prepare the food packaging films that are composed of konjac glucomannan (KGM), poly(ε-caprolactone) (PCL) and silver nanoparticles (AgNPs) via microfluidic spinning technology (MST). The obtained fiber films (average fiber diameter: 7.8 ± 0.2 μm) exhibit excellent antibacterial activities against S. aureus (34 ± 0.71 mm) and E. coli (39 ± 5.66 mm), which is ascribed to the good swelling of KGM in KGM/PCL/AgNPs fiber films (SD: 37.86 ± 6.87%). Fourier transform infrared (FT-IR) spectroscopy and X-ray diffraction (XRD) are employed to study the interactions between polymers. Thermogravimetric analysis (TGA), derivative thermogravimetry (DTG), water vapor permeability (WVP), and mechanical property measurements are conducted to evaluate the thermal properties, hydrophilicity and mechanical performances of the films. The results show that the films are thermal stable and relatively hydrophobic (WVP: 5.8 × 10 − 6  ± 1.44 g/(m·h·kPa), WCA: 101.0°) as well as have terrific elongation at break (EB: 223.59 ± 98.14%), which is beneficial for food packaging. This strategy provides a facile and green pathway for the construction of promising antibacterial food packaging.

Konjac glucomannan/polyvinyl alcohol nanofibers with enhanced skin healing properties by improving fibrinogen adsorption

Bo Yang,Yushan Chen,Zhiqiang Li,Pengfei Tang,Youhong Tang,Yaping Zhang,Xiaoqing Nie,Cheng Fang,Xiaodong Li,Hongping Zhang

Skin tissue engineering aims to develop the effective healing strategy to repair the wound by optimizing skin scaffold materials. During the skin wound healing process, fibrin plays an important role due to the specific blood coagulation effect. In this study, the outstanding fibrin capability of konjac glucomannan (KGM) is demonstrated by the molecular dynamics simulation and confirmed by the protein adsorption experiments. A series of konjac glucomannan/polyvinyl alcohol (KGM/PVA) composites with different ratio are fabricated and their role in enhancing the skin repair is tested by in vitro cell culture and in vivo study. The E ads (adsorption energy) between fibrin and KGM is about 30% larger than that between fibrin and PVA. The fibrinogen adsorption rates of PVA and KGM/PVA (5:5) composites can reach about 20% and 60%, respectively. The results show the blood adsorption capacity of KGM/PVA (5:5) composite can reach about 13 g/g. After 7 days of cell culture, the optical density values of 3T3 fibroblasts on KGM/PVA (5:5) composite could reach 0.8. The mechanical properties of the composites are also verified to meet the practical needs. Thus, we propose a potential wound dressing material strategy based on the materials design and the intrinsic properties of KGM.

Ultralight, hydrophobic, monolithic konjac glucomannan-silica composite aerogel with thermal insulation and mechanical properties

Jundong Zhu,Jiang Hu,Chongwen Jiang,Siyuan Liu,Ying Li

Konjac glucomannan (KGM) aerogel was prepared by a facile freeze-drying process, which was used as green sustainable biopolymer matrix to synthesis three-dimensional (3D) network interpenetrated KGM-SiO 2 aerogel with thermal insulation performance and high mechanical properties. Herein, we explored that the potential structure–performance relationship of KGM aerogel between the physical parameters, morphology and thermal insulation. Besides, the preparation conditions of KGM aerogel including concentration of KGM, Na 2 CO 3 -to-KGM mass ratio were investigated. Meanwhile, the KGM aerogel and KGM-SiO 2 aerogel were characterized using scanning electron microscopy, Fourier transform infrared spectrometer, Brunauer-Emmett-Teller method, thermogravimetric analysis and contact angle test. Results showed that the as-prepared KGM aerogel exhibited excellent thermal insulation performance (λ = 0.021 W m −1  K −1 ) and low bulk density (ρ = 0.030 g cm −3 ) when concentration of KGM was 2 wt% and Na 2 CO 3 -to-KGM mass ratio of 0.12. In order to improve the mechanical properties and high hydrophobicity of biopolymer aerogel, a novel of KGM-SiO 2 aerogel was successful fabricated by incorporating methylsilsesquioxane (MTES) derived SiO 2 into the KGM matrix via the freeze-drying method. The obtained aerogel exhibited remarkable compressive strength (δ max  = 1.65 MPa at 80% strain), high specific surface area of 416.1 m 2  g −1 , high hydrophobicity (θ = 146° for water) and low thermal conductivity of 0.032-0.039 W m −1  K −1 . Thus, the high-performance KGM-SiO 2 aerogel would be further expand the thermal insulation application in sustainable development and energy-saving building.

Yongsheng Ni,Wanmei Lin,Ruojun Mu,Chunhua Wu,Zheya Lin,Su Chen,Jie Pang

The exploration of methods to produce biomaterials with antibacterial properties in ultra-small scales is of great scientific and technological interest. Here, we reported a microfluidic spinning approach to prepare a novel film combined with konjac glucomannan (KGM), polyvinylpyrrolidone (PVP), and epigallocatechin gallate (EGCG). The film was characterized by scanning electron microscopy, Fourier transform infrared spectroscopy, X-ray diffraction, and thermogravimetric analysis. This film was transparent, orderly, thermally stable, and uniform in size, with an average width less than 1 μm. Also, the film exhibited excellent antibacterial efficiency (97.1% against E. coli , 99.7% against S. aureus , 97.3% against S. enterica and 99.9% against B. subtilis ) in our antibacterial test. In addition, the film promoted wound healing with the advanced development of neovascularization and hair follicles. This strategy provides a facile and green pathway for the construction of biomaterial films for medical applications.

Aggregation-induced emission of active nanoparticles based on konjac glucomannan: Fabrication, properties, and applications

Lianxiong Guan,Zhi Zeng,Rui Zhao,Sheng Hu,Wei Liu,Dating Tian

Recently, the design of aggregation-induced emission (AIE) materials with good water-solubility and biocompatibility has attracted much attention. In this work, amphiphilic AIE-active nanoparticles (KGM-TPE) were synthesized by a facile Schiff base condensation of oxidized konjac glucomannan with amino-tetraphenylethylene. The final KGM-TPE had good water dispersibility and could self-assemble into nanoparticles with average diameters ranging from 100 to 200 nm. A cytotoxicity test showed that the cell survival was approximately 90% even when the concentration of KGM-TPE reached 150 μg/mL, indicating that KGM-TPE possessed good biocompatibility. In addition, the excellent cell imaging and good anti-counterfeiting function results of KGM-TPE indicated that this material has great potential in the fields of biomedicine and anti-counterfeiting.
