The bionics of natural bone structures plays an essential role in the selection of materials for bone tissue engineering. Although the content of trace metallic elements in bone is low, they have significant effects on the process of bone growth and metabolism. Up to now, the applications of “green” heavy metals in bone regeneration are limited. Herein, in this study, we present a straightforward one-pot strategy for the synthesis of in situ bismuth-doped amorphous calcium phosphate nanocomposites (RBCP), effectively integrating the beneficial properties of each component. The characterization of these products can be readily optimized by adjusting reaction parameters. Our in vitro studies show that under coordination of each component, the RBCP biomaterial demonstrates distinguished biocompatibility and significantly accelerates vascular pattern formation within just 4 h by stimulating the expression of angiogenesis-related genes in human umbilical vein endothelial cells (HUVECs). In vivo experiments indicate that the incorporation of bismuth effectively enhances bone regeneration and osseointegration in a rat femur defect model. In conclusion, the as-prepared RBCP biomaterials hold promising prospects for treating segmental bone defects, owing to the facile, cost-effective, and eco-friendly preparation process, along with their remarkable capabilities.

Bone defects caused by trauma, tumor resection, and surgery present significant clinical challenges, often resulting in complications such as delayed union, nonunion, and even long-term functional impairment. Current treatments, including autografts and allografts, are limited by donor site morbidity, immune rejection, and pathogen transmission, highlighting the need for developing reliable synthetic alternatives. To address these challenges, we report a binary composite hydrogel combining gelatin methacryloyl (GelMA) and κ-Carrageenan, reinforced with calcium phosphate cements (CPC). GelMA ensures rapid gelation and biocompatibility, κ-carrageenan improves injectability, and CPC enhances mechanical strength and osteogenic activity, collectively creating a robust and versatile hydrogel system. Furthermore, the hydrogel’s injectable, adaptive, and self-healing characteristics enable it to conform to irregular bone defect sites, providing mechanical support and osteogenic stimulation. It also releases bioactive components to accelerate bone regeneration. With exceptional toughness and resilience, this hydrogel recovers its shape after deformation, positioning it as a promising candidate for clinical bone defect repair applications.

The structural and functional repair of critical-sized irregular bone defect is still considered a serious challenge. Most bone implant materials used in current clinic are not able to induce reconstruction of functional neural network, which is the key in bone regeneration via neural-osteogenic pathway. In this study, we aimed to manufacture a novel injectable double-crosslinked collagen/hyaluronic acid hydrogel loaded with nerve growth factor (NGF), which was capable of well filling irregularly shaped bone defects and inducing reconstruction of neural network. Hyaluronic acid (HA) was sequentially oxidized by sodium periodate and modified by methacrylic anhydride to prepare methacrylated oxidized HA (MOH) as a crosslinker. Subsequently, the Collagen-MOH@NGF (CMHN) hydrogel was established by Schiff base reaction and double-bond photocrosslinking. Our results demonstrated the excellent microporous structure, injectable, self-healing, in-situ rapid crosslinking properties of CMHN hydrogel along with suitable mechanics, rheology, swelling, degradation and biocompatibility. Significantly, the introduction of NGF endowed CMHN hydrogel with better biological properties than that without NGF. In vitro , CMHN hydrogel promoted DRG neuron growth and osteogenic differentiation of BMSCs mediated by NGF-calcitonin gene-related peptide (CGRP) axis. In vivo , CMHN hydrogel possessed a better bone repair effect than free NGF and blank hydrogel, with a long-lasting NGF-promoting neuro-osteogenic action within 8 w in a rat cranial critical defect model. Overall, the prepared injectable NGF-loaded collagen/HA dual network hydrogel is a promising material to facilitate functional and structural repair of complex bone defects.

Complex wound repair due to tumor recurrence and infection following tumor resection presents significant clinical challenges. In this study, a bifunctional nanocomposite immune hydrogel dressing, SerMA-LJC, is developed to address the issues associated with repairing infected damaged tissues and preventing tumor recurrence. Specifically, the immune dressing is composed of methacrylic anhydride-modified sericin (SerMA) and self-assembled nanoparticles (LJC) containing lonidamine (Lon), JQ1, and chlorine e6 (Ce6). In vitro and in vivo experiments demonstrate that the nanocomposite hydrogel dressing can trigger immunogenic cell death (ICD) and has a potent anti-tumor effect. Moreover, this dressing can mitigate the acidic microenvironment of tumor cells and suppress the overexpression of PD-L1 on the tumor cell surface, thereby altering the immunosuppressive tumor microenvironment and augmenting the anti-tumor immune response. Further, the RNA sequencing analysis revealed that the hydrogel dressing significantly impacts pathways associated with positive regulation of immune response, apoptotic process, and other relevant pathways, thus triggering a potent anti-tumor immune response. More importantly, the dressing generates a substantial amount of reactive oxygen species (ROS), which can effectively kill Staphylococcus aureus and promote infectious wound healing. In conclusion, this dual-function nanocomposite immune hydrogel dressing exhibits promise in preventing tumor recurrence and promoting infectious wound healing.

The degradability of hydrogels plays a pivotal role in bone regeneration, yet its precise effects on the bone repair process remain poorly understood. Traditional studies have been limited by the use of hydrogels with insufficient variation in degradation properties for thorough comparative analysis. Addressing this gap, our study introduces the development of matrix metalloproteinase (MMP)-responsive hydrogels engineered with a tunable degradation rate, specifically designed for bone regeneration applications. These innovative hydrogels are synthesized by integrating MMP-sensitive peptides, which exhibit chirality-transferred amino acids, with norbornene (NB)-modified 8-arm polyethylene glycol (PEG) macromers to form the hydrogel network. The degradation behavior of these hydrogels is manipulated through the chirality of the incorporated peptides, resulting in the classification into L, LD, and D hydrogels. Remarkably, the L hydrogel variant shows a significantly enhanced degradation rate, both in vitro and in vivo, which in turn fosters bone regeneration by promoting cell migration and upregulating osteogenic gene expression. This research highlights the fundamental role of hydrogel degradability in bone repair and lays the groundwork for the advancement of degradable hydrogel technologies for bone regeneration, offering new insights and potential for future biomaterials development.

Osteoarthritis (OA) is a degenerative joint disease characterized by cartilage degradation and limited effective treatment options for regeneration. To address this clinical challenge, an innovative injectable biomimetic hybrid nanofiber, namely hyaluronic acid-graft-poly(2-methacryloyloxyethyl phosphorylcholine)-chondrocyte affinity peptides (HA-PMPC-CAP), was designed and synthesized through a ultraviolet light (UV)-initiated reaction process for intervention of early osteoarthritis (OA). Comprehensive in vitro assessments and molecular docking experiments have confirmed its non-cytotoxicity, exceptional lubrication properties, favorable biocompatibility and high binding affinity to cartilage proteins. Utilizing an OA rat model, the therapeutic potential of HA-PMPC-CAP was explored through intra-articular injection, with a focus on its impact on cartilage regeneration. During an 8-week period, macroscopic, histological, immunohistochemical, and microscopic CT evaluations were conducted on the treated joints. The findings were remarkable: HA-PMPC-CAP established a durable lubricating layer on the surface of cartilage, significantly reduced cartilage deterioration, replenished glycosaminoglycan (GAG) levels, lowered the scores of Osteoarthritis Research Society International (OARSI), and curtailed the development of osteophytes, which was superior to the untreated OA control groups. These results position HA-PMPC-CAP as an effective biomimetic therapy, offering a novel strategy for cartilage regeneration and lubrication in the early stages of OA, which has promising implications for future clinical applications.

Hydrogel microparticles (HMPs) have many advantages for biomedical applications, particularly for minimally invasive therapy, for example, acute lung injury (ALI) that is characterized by high levels of reactive oxygen species (ROS) and pro-inflammatory mediators in the microenvironment. In this study, ROS-scavenging and pro-inflammatory cytokine-neutralizing HMPs were designed and prepared by using a membrane emulsification device. The HMPs were composed of double bond-modified hyaluronic acid and ROS-cleavable hyperbranched poly(acrylate-capped thioketone-containing ethylene glycol) (HBPAK) containing thioketal linkages and unsaturated double bonds. Surface-coating of inflammatory macrophage (M1) cell membranes was performed to obtain the membrane-coated HBPAK HMPs (mem HMPs) via electrostatic force. The mem HMPs exhibited strong ROS-scavenging and anti-inflammatory properties both in vitro and in vivo . After administered by inhalation in an ALI mouse model, the mem HMPs reduced neutrophil infiltration and tissue oxidative damage, thereby alleviating lung inflammation. Our results suggest that the mem HMPs could serve as a potential therapeutic platform for treating inflammatory diseases with high efficiency. Transplantation of beneficial bacteria to specific microbiota has been widely exploited to treat diseases by reshaping a healthy microbial structure. However, direct exposure of exogenous bacteria in vivo suffers from low bioavailability and infection risk. Here, we describe a noncontact microbiota transplantation system (NMTS) by core-shell microgel-enabled nonleakage envelopment. Bacteria are encapsulated into the core of core-shell microgels via two-step light-initiated emulsion polymerization of gelatin methacrylate. NMTS is versatile for biocontainment of diverse strains, showing near complete encapsulation and negligible influence on bacterial activity. As a proof-of-concept study on probiotic transplantation to the gut microbiota, NMTS demonstrates the shielding effect to protect sealed bacteria from intraluminal insults of low pH and bile acid, the toughness to prevent bacterial leakage during entire gastrointestinal passage and reduce infection risk, and the permeability to release beneficial metabolites and reconstruct a balanced intestinal microbial structure, proposing a contactless fashion for advanced microbiota transplantation.

This study explores the use of chicken egg white (EW), a rich source of natural proteins, to address challenges in wound healing management. Herein, a novel Zn2+-infused EW/GelMA (EW/Gel) hybrid hydrogel is developed, featuring an interpenetrating network (IPN) structure, where the first network consists of photo-cross-linked GelMA and the second network consists of Zn2+-infused EW (Zn-EW) through ion–protein binding. By optimizing the design and formulation, the resulting Zn-EW/Gel hydrogel exhibited enhanced mechanical stability and self-adhesive properties. In vitro experiments demonstrated that the combined effects of functional proteins and active ions within the Zn-EW/Gel hydrogel promote fibroblast proliferation and type I collagen expression, modulate the immune microenvironment, and enhance angiogenesis. The hydrogel also demonstrated excellent biocompatibility and bioactivity in vivo, showing strong promise for restoring the physiological properties of the damaged wound tissue.

Natural polymers-based hydrogels have been widely applied in bone tissue engineering due to their excellent biocompatibility and the outstanding ability of drug encapsulation. However, they have relatively low mechanical properties and lack of bioactivity. Hence, we developed a bioactive nanoparticle composite hydrogel by incorporating the laponite, which is an osteo-inductive inorganic nanoparticle. The incorporation of the nanoparticle significantly increased mechanical properties. In vitro evaluation indicated that the nanocomposite hydrogel was capable of good biocompatibility. Besides, the nanocomposite hydrogel was proved to have excellent osteogenic ability with up-regulated expression of osteogenic markers such as COL-I, Runx-2 and OCN. Furthermore, the in vivo study confirmed that the SFMA-LAP nanocomposite hydrogel could significantly promote new bone formation, providing a prospective strategy for bone tissue regeneration.

Hydrogel microspheres hold great promise as scaffolds for bone repair. Their hydrated matrix, biocompatibility, and functional properties make them an attractive choice in regenerative medicine. However, the irregularity of defect requires shape adaptability of the microspheres. Additionally, there is still room for improvement regarding the component of the microspheres to achieve sufficient bioactivity. Here, we prepare multi-bioactive microspheres composed of methacrylated silk fibroin (SFMA) via microfluidic electrospray. Magnesium ascorbyl phosphate (MAP) is encapsulated within the microspheres, whose sustained release facilitates angiogenesis and osteogenic differentiation. The microspheres are further coated with a polydopamine (PDA) layer, allowing them to assemble in situ into a scaffold that conforms to the non-uniform contours of bone defects. The photothermal conversion capability of PDA also provides mild photothermal stimulation to further promote bone regeneration. Based on the synergistic effects, our in vivo experiments demonstrated that the microsphere scaffold effectively promotes bone defect healing. Thus, this multi-bioactive scaffold offers a versatile strategy for bone repair with promising clinical potential.

Oral ulcers may be addressed with various biomaterials infused with medications or cytokines. However, many patients derive little advantage from these substances due to inadequate adhesion, brief retention in the mouth, and diminished drug effectiveness. Here, we introduce a hydrogel patch (MPG3) consisting of a biopolymer matrix with light-activated adhesion features infused with platinum-decorated melanin nanoparticles that possess healing-promoting abilities. Initially, through straightforward local application, an adhesive hydrogel barrier forms over mucosal imperfections upon exposure to ultraviolet light. Following this, MPG3 exhibits outstanding oxygen production and reactive oxygen species elimination properties. These capabilities contribute to regulating the wound microenvironment, thereby expediting the wound's transition from the inflammatory to the proliferative stage. In rat models of oral ulcers, the MPG3 patch hastens recovery by modulating the inflammatory milieu of the damaged tissue and fostering expedited re-epithelialization and blood vessel formation. Conclusively, this light-activated adhesive hydrogel shows promise for swift wound healing and could revolutionize therapeutic approaches for treating clinical oral ulcers, especially in diabetic patients.
